Abstract:Plant-based enzymatic extraction of collagen from tannery rawhide trimmings can play an important role in the utilization of tannery solid waste, providing economically viable and sustainable solutions. This study employed bromelain enzyme derived from pineapple ( Ananas comosus ) peel waste for collagen extraction and compared the procedure with conventional acetic acid extraction. The recovery of bromelain enzyme at 30–70% ethanol precipitation exhibited a 2.21-fold purification and a 98% yield. Response surface methodology (RSM) analysis revealed that bromelain enzyme-extracted collagen (BEC) yielded 82.20% at a hydrolysis time of 18.39 h, a mixing ratio of 22.38, and a temperature of 39.32 °C. In contrast, a lower yield of 66.94% was observed in the case of acetic acid-extracted collagen (AEC) at a hydrolysis time of 18.66 h, a mixing ratio of 19.41, and a temperature of 27.09 °C. Fourier transform infrared spectroscopy (FTIR) confirmed the successful extraction of both AEC and BEC. Thermogravimetric analysis (TGA) indicated the higher thermal stability of BEC compared to AEC. X-ray Diffraction (XRD) displayed characteristic peaks of collagen in the range of 2θ of 15–25°, affirming that BEC (sharp peak at 19.49°) is more crystalline than AEC (broadening peak at 19.88°). Elemental analysis revealed a higher nitrogen content (6.78%) in BEC, indicating a higher protein content than that of AEC (6.31%). Furthermore, antimicrobial activity, cytotoxicity evaluation, and heavy metal analysis revealed the potential applications of collagen in cosmetics, biomaterials, and tissue engineering.

Abstract:Effective treatment and high-valued utilization of chrome leather scraps have always been desired by all tanneries, which is of great significance for green and sustainable development of leather industry. In this paper, we first devised a green and effective approach to obtain collagen hydrolyzed extract (CHE) from chrome leather scraps, and then the obtained CHE was used in the granulation of pastoral soil to prepare CHE-Soil granular fertilizer. The results indicated that chrome leather scraps were effectively dechromed with sodium oxalate and then hydrolyzed by a high-pressure hydrothermal method without any chemical to obtain high-purity CHE. When the 4.5 wt%similar to 12.5 wt% CHE solution was introduced into the granulation process, the granulation rate of pastoral soil was up to 89.8%similar to 95.6%. However, the effective granulation rate was decreased due to the increase in large granule size (>5 mm). CHE-Soil granular fertilizer prepared by adding more CHE exhibited better surface roundness and more excellent crush resistance. Moreover, this type of granular fertilizer possessed excellent water retention and slow-released nutrient properties. Pine leaf peony cultivation experiments further showed that the presence of CHE-Soil granular fertilizer rapidly promoted plant growth and effectively increased the harvest of plants. This study indicated that CHE obtained from chrome leather scraps could be effectively used as a nutritious additive with adhesiveness to prepare the granular fertilizer for plant growth, which provides a novel idea for designing valorization methods of leather waste.
Sustainable leather tanning: Enhanced properties and pollution reduction through crude protease enzyme treatment

Abstract:In this study, proteolytic bacteria, particularly Pseudomonas aeruginosa strain SM4 (OQ349573), were isolated from tannery solid waste dumping yard soil and employed to produce extracellular protease enzymes. The bacteria exhibited optimal growth after 30 h of incubation at 37 °C and pH 7. Under conditions of 55 °C, pH 8, and a substrate concentration of 2 %, the crude enzyme displayed its highest activity at 105 UmL −1 . Notably, the produced crude enzyme showed no discernible inhibitory effects on detergents, metal salts, or organic solvents. Application of the crude protease at concentrations of 3 % and 2 % in chrome tanning of goatskins (GS) and cowhides (CH), respectively, yielded significant reductions of 35 % and 30 % in chromium and other post-tanning chemicals compared to conventional processes. Despite the 30 to 35 % reduction in tanning and post-tanning chemicals, the uptake of chrome and associated chemicals by crust leather was higher than observed in conventional processes. Chromium content analysis of the effluent revealed an 81 % reduction during piloting in real industrial operations, accompanied by reductions of about 46 % in BOD and COD pollution loads. The finished leather obtained from the enzymatic process exhibited superior mechanical properties, including higher tensile strengths (210 and 195 kg cm - 2 ), stitch tear (92 and 165 kg cm −1 ), grain crack load (28 and 32 kg), and distension (73 and 62 mm) for GS and CH, respectively, surpassing or closely aligning with standard values and those obtained in conventional processes.

Abstract:<h3 class="Heading">Purpose</h3><p class="Para">Hide is a by-product of meat production and is mostly used for leather production. Collagen is the main protein in mammalian skin, connective tissue and cartilage and presents an opportunity for value addition to waste hide off-cuttings by extracting collagen. Three different extraction methods were applied to five different hide sources. The hide sources differed with respect to the animal's age, sex, diet and environment and influenced collagen extractables yield, and therefore the economic benefit of extraction.</p><h3 class="Heading">Methods</h3><p class="Para">Acid-solubilisation (AS), acid-enzyme solubilisation (AES1) and modified acid-enzyme solubilisation (AES2) were used to extract collagen from bull, calf, cow, face-pieces and ox-hides.</p><h3 class="Heading">Results</h3><p class="Para">The highest dry collagen content was from cow hides using the AES2 method (75.13%), followed closely by bull hides at 74.45%. On the other hand, the lowest collagen content was from cow hides (3.80%) with the AS extraction method and the AS method proved to be inefficient for collagen extraction from bull, cow, face-piece and ox-hide sources. Analysis concluded that all the samples were of Type I collagen with α, β, and γ chains.</p><h3 class="Heading">Conclusions</h3><p class="Para">Waste bovine hide off-cuttings can be used to extract high value product of collagen. Abstract:Skin fleshing wastes from tanneries are generated during leather processing when flesh in the limed hide or skin is removed with the sharp knives. These skin fleshing wastes from tanneries contain highest protein content and currently being wasted into the dumping sites or in open areas, consequently creating the solid waste disposal problem for tanners. In this study skin fleshing wastes were first hydrolyzed with alkaline mixture of sodium hydroxide and magnesium oxide for the recovery of protein fraction. This isolated protein was further treated with soluble starch solution in distilled water and acrylic acid solution in distilled water with the small addition of initiator such as sodium metabisulphite and potassium persulphate. The reaction was completed in 2h and finally whitish colored polymeric retanning product was prepared. This retanning product was subjected for application in leather processing at (5%, 10% and 15%). Prepared leathers were tested with standard physical tests and compared with reference leather processed with commercial retanning product. The most significant results in leather were obtained by applying 10-15% of prepared retanning product. Data from physical testing was quantified with statistical analysis and it was found significant.

Enzyme Preparation Characteristics and New Technical Solution Development for Bioconversion of Collagen-Containing Raw Materials

Abstract:One of the modern challenges facing the food and processing industry is the rational and resource saving technology development for the secondary resources use including collagen containing raw materials. Raw materials bioconversion is a promising solution method for the problem. A man run the research on the enzyme preparation use as hydrolysates of food raw materials including for the meat processing industry; studied the technological solutions appeared in the scientific and technical literature; identified the strengths and weaknesses of hydrolysis methods. Based on the studied data, the authors developed technological approach to the bioconversion of the broiler chicken skin. The developed approach consists in the technological preparation of collagen containing raw materials, its grinding, processing with an enzyme preparation from secondary fish raw materials for 15 minutes at a temperature of (3 ± 2)°C in a refrigerating chamber followed by filtration, regrinding and drying at a temperature of (41 ± 2) °C. According to the results of the conducted studies, the enzyme preparation is capable of collagen compound hydrolyzing, the proteolytic activity is 2.9 U/g. The enzyme preparation is a light-yellow substance with a faint specific aroma, a liquid, slightly viscous consistency. The mass fraction of dry substances in the enzyme preparation is 44%, the mass fraction of protein is 15%. Broiler chicken skin processing with an enzyme preparation resulted in a fine-grained free-running powder of light-yellow color with a mass fraction of 81.1% protein, 14.2% fat, 4.7% moisture, and 64.4% collagen. The ways of using the obtained collagen fermentolysate have been determined. The developed technology of using broiler chicken skin treated with an enzyme preparation is a new modern technical solution in the bioconversion of collagen containing raw materials. A man can exploit it in the food technology of meat pates and minced meat.

Abstract:Preparing pure collagen fiber woven materials involves a series of complex unit operations by conventional process with sodium sulfide and calcium hydroxide system, resulting in high cost and serious environmental pollution. So, a green and high-efficiency ecological processing technology was designed by using alkaline protease to remove hair and open collagen fiber-bundles after pre-swelling bovine hides with alkali. In this work, the effect of hairs removal, hydrolysis of non-collagen components and penetration mechanism of alkaline protease are studied, and the properties of cross-linked pure collagen materials and the pollutants load of wastewater from the alkaline protease process after alkali pre-swelling are analyzed and evaluated to compare with those of conventional one. As a result, the hairs of bovine hides were removed completely and the proteoglycans in hides were hydrolyzed thoroughly in 30 h by alkaline protease process after alkali pre-swelling. The fibers fullness, fleeciness and grain surface smooth of cross-linked pure collagen material from the alkaline protease process after NaOH pre-swelling are better than those of the Ca(OH)2 pre-swelling and conventional processes. Especially, the chemical oxygen demand (COD), ammonia nitrogen (NH3-N), total solids (TS) and discharged wastewater (WD) of the pollutant loads from the alkaline protease process after NaOH pre-swelling are 58%, 76.75%, 76.03% and 61.07% lower respectively than those of conventional one. This work is hopeful to provide a cleaner and simplifier method for manufacturing the ecological pure collagen based woven materials.

Abstract:Fish processing waste is a prospective source of collagen and a cost-effective environmental pollutant. The skin of the purple-spotted bigeye snapper (Priacanthus tayenus) was extracted utilising various acid soluble collagens (ASC) including acetic acid (AAC), lactic acid (LAC), citric acid (CAC) and pepsin soluble collagens (PSC). In this study, PSC (6.65%) had the highest collagen yield, followed by AAC (5.79%), CAC (4.15%), and LAC (3.19%). The maximum temperatures (Tmax) denaturation of AAC, LAC, CAC, and PSC were 31.4, 31.7, 31.5, and 33.2 °C, respectively. UV-VIS absorption spectra showed all extracted collagens had a range of absorbance at 230 nm, due to the presence of glycine, proline, hydroxyproline, and triple-helical collagen. Additionally, they exhibited amide A, B, amide I, II, and III peaks. SDS–PAGE identified all extracted collagens as type I. The PSC had a significantly higher (p < 0.05) hydroxyproline content than acidic extraction 66.3 ± 1.03 (mg/g sample). Furthermore, all samples were extremely soluble in acetic conditions at pH 5, and all collagen was soluble in NaCl up to 3% (w/v). Therefore, PSC was the best treatment since it did not impact collagen triple helical and acetic acid yielded the most collagen in ASC extraction. Overall, the analysis revealed that fish skin waste might be used as an alternate source of collagen in diverse applications, particularly in food applications.

Abstract:Researchers around the world are in continuous endeavor to develop environment friendly tanning agent due to adverse effect of conventionally used chromium during leather tanning. Recent trend of tanning is thus heading to chrome-free and greener chemical processing options. Vegetable tanning is an exoteric leather processing technique because of their lower pollution load on the environment. Considering the inadequacy and high costing for the commercialized tannins, development of alternative tannins from locally available plants and their characterizations are important. In this research, the stem barks of Trema Orientalis (L.) were extracted at different temperatures employing water solvent with or without additives (sodium hydroxide or sodium sulphite) to attain phenolic-rich extractives. To ascertain appropriateness as a vegetable tanning agent, the obtained extracts were thereafter characterized in respect to yield, total phenolic content, tannin content and molecular structure. The extraction yield for all extracts improves and the quality remains nearly unchanged with temperature rising. The elevated concentration of chemical additives enhances the extraction yield but lessens the quality of extracts. In terms of extraction yield and the quality of extracts the best condition for extraction was discerned at 80°C with water solvent. This water extract has a decent extent of phenolic and tannin content of 266.13 mg Gallic acid equivalent/ gm of dry extract and 30.12 % respectively. The final extract exhibits excellent leather retanning tendency comparable to the commercial quebracho tannins.

Abstract:The tannery is increasingly focusing on environmentally friendly processes, emphasizing the concept of clean production to minimize environmental impact. This study investigated the eco-friendly application of α-amylase/neutral protease system (ANS) in a single-step process for unhairing, fiber opening, and bating, providing an effective alternative to the conventional complex chemical-based lime-sulfide system (LSS). A mathematical model was established using response surface methodology (RSM) to optimize the operational conditions, yielding a 0.3 wt.% α-amylase/0.5 wt.% neutral protease solution at 28.4°C for 16.6 h. The effects on unhairing and fiber opening were evaluated using scanning electron microscopy (SEM), while the effect on bating was accessed by the removal of carbohydrate and proteoglycan. The obtained leathers had comparable physical properties to conventional counterparts, with enhanced hydrothermal shrinkage temperature and softness. In terms of environment benefits, the optimized ANS could significantly decrease more than 90% of the chemical oxygen demand (COD), NH3-N, and Cl-, respectively, along with a 73.91% reduction in total solids (TS). Cost analysis indicated direct savings of a significant cost reduction of 30.98% of ANS compared to LSS, as well as additional indirect cost reductions through improved production efficiency and streamlined wastewater treatment processes. The use of electricity and water was reduced by 44.6% and 93.9%, respectively, which could reduce the greenhouse gas emissions by 44.6%. This study highlighted the potential of α-amylase/neutral protease as a sustainable and cost-effective alternative in leather manufacturing, that aligns with environmentally friendly practices.

Abstract:The disposal of large amounts of skin waste resulting from the blue shark fishing industry presents several industrial and environmental waste management concerns. In addition, these marine subproducts are interesting sources of collagen, a fibrous protein that shows high social and economic interest in a broad range of biomedical, pharmaceutical, and cosmetic applications. However, blue shark wasted skins are a poorly explored matrix for this purpose, and conventional collagen recovery methodologies involve several pre-treatment steps, long extraction times and low temperatures. This work presents a new green and sustainable collagen extraction approach using a natural deep eutectic solvent composed of citric acid:xylitol:water at a 1:1:10 molar ratio, and the chemical characterization of the extracted collagen by discontinuous electrophoresis, thermogravimetric analysis, Fourier transformed infrared spectroscopy and circular dichroism. The extracted material was a pure type I collagen, and the novel approach presented an extraction yield 2.5 times higher than the conventional one, without pre-treatment of raw material and reducing the procedure time from 96 to 1 h. Furthermore, the in vitro cytotoxicity evaluation, performed with a mouse fibroblasts cell line, has proven the biocompatibility of the extracted material. Overall, the obtained results demonstrate a simple, quick, cheap and environmentally sustainable process to obtain marine collagen with promising properties for biomedical and cosmetic applications.

Abstract:As the collagen protein market increasingly grows, it becomes a priority to explore a economic, stable , and environmentally-friendly extraction method to reali ze the high-value transformation of pigskin. In thi s paper, the structure of collagen protein is introduced first. Then five methods, which extract collagen protein f rom pigskin or a kind of leather industry waste-chrome shavings, are reviewed. These methods include the salting out me thod, alkali method, acid method, enzyme method and hybrid metho d. They are contrasted in several ways, and thus pr ovide evidence for better extracting collagen protein.

Abstract:Acid soluble (ASC) and pepsin soluble (PSC) collagens were extracted from the skin, bone and muscle of a trash fish, leather jacket (Odonus niger) by three different extraction methods. Method I gave 46–50% yield for ASC, Method II gave 49–58% yield for both ASC and PSC and Method III gave 64–71% yield for PSC. The addition of pepsin had increased the yield by 30–45%. The yields of collagen from skin and bone were higher than muscle. SDS-PAGE pattern revealed that skin and bone collagen as Type I collagen with a typical (α1)2α2 chains and muscle collagen as Type V collagen with a typical α1α3α2 chains. Td values of bone and muscle collagen were high (30–32 °C) compared to skin collagen (27–28 °C). The higher imino acids (190 residues/1,000 residues) were found responsible for the higher Td values. The trash fish, leather jacket can therefore be exploited effectively for collagen as it has got good thermal properties for pharmaceutical and biomedical applications.

Abstract:Collagen has a great demand and fish skin is a safe alternative source of collagen. Pterygoplichthys pardalis is an endemic, freshwater fish with no economic benefit. The study was conducted to extract crude collagen from the skin of P. pardalis using a simple method and identify the functional properties of its hydrolysates. Acid and pepsin soluble collagens were extracted from skin using citric acid (CA) and ethylene diamine tetraacetic acid (EDTA). Selected crude collagens were hydrolysed using pepsin, protease and trypsin enzymes (1:100). Extracted crude collagens and best hydrolysates were selected using 8 and 15% SDS-PAGE. Hydrolysates were analysed for antioxidant, Fe(II) chelating and antibacterial activities. CA treatment showed significantly (P 0.05) different. Hydrolysates showed significantly (P<0.05) higher functional properties than crude collagen in both treatments. These results conclude that collagen hydrolysates from P. pardalis have good antioxidant, metal chelating and antibacterial properties.

Abstract:Background: To our knowledge, the effect of magnesium supplementation on blood pressure (BP) in individuals with preclinical or noncommunicable diseases has not been previously investigated in a meta-analysis, and the findings from randomized controlled trials (RCTs) have been inconsistent.Objective: We sought to determine the pooled effect of magnesium supplementation on BP in participants with preclinical or noncommunicable diseases.Design: We identified RCTs that were published in English before May 2017 that examined the effect of magnesium supplementation on BP in individuals with preclinical or noncommunicable diseases through PubMed, ScienceDirect, Cochrane, clinicaltrials.gov, SpringerLink, and Google Scholar databases as well as the reference lists from identified relevant articles. Random- and fixed-effects models were used to estimate the pooled standardized mean differences (SMDs) with 95% CIs in changes in BP from baseline to the end of the trial in both systolic blood pressure (SBP) and diastolic blood pressure (DBP) between the magnesium-supplementation group and the control group.Results: Eleven RCTs that included 543 participants with follow-up periods that ranged from 1 to 6 mo (mean: 3.6 mo) were included in this meta-analysis. The dose of elemental magnesium that was used in the trials ranged from 365 to 450 mg/d. All studies reported BP at baseline and the end of the trial. The weighted overall effects indicated that the magnesium-supplementation group had a significantly greater reduction in both SBP (SMD: -0.20; 95% CI: -0.37, -0.03) and DBP (SMD: -0.27; 95% CI: -0.52, -0.03) than did the control group. Magnesium supplementation resulted in a mean reduction of 4.18 mm Hg in SBP and 2.27 mm Hg in DBP.Conclusion: The pooled results suggest that magnesium supplementation significantly lowers BP in individuals with insulin resistance, prediabetes, or other noncommunicable chronic diseases.
Preparation of immobilized pepsin for extraction of collagen from bovine hide

Abstract:In the extraction of collagens from mammalian tissues, the free pepsin used in the acid–pepsin extraction system is hard to recycle, and there is a risk of enzyme protein contamination in the extracted collagen products, which limits their applications. To solve this problem, an immobilized pepsin was successfully prepared via the covalent crosslinking of glutaraldehyde using a 3-aminopropyltriethoxysilane (APTES) surface modified silica clay as the support. The immobilized pepsin was applied for the extraction of collagen from bovine hide. The optimal immobilization process involves incubating pepsin with an initial concentration of 35 mg mL −1 and glutaraldehyde with 5% activated APTES modified silica clay at 25 °C for 60 min, by which the loading amount of pepsin was 220 mg g −1 and the activity of the immobilized pepsin was 4.2 U mg −1 . The collagen extracted using acetic acid and the immobilized pepsin method retained its complete triple helix structure. This research thus details an effective separation method using pepsin for extraction of collagen via an acetic acid–enzyme method, where the extracted collagen may be a candidate for use in biomaterial applications.

Extraction of Collagen from Bovine Tannery Solid Waste Preserving Original Conformation via Radical Initiation and Hydrogen Bond Reformation

Abstract:Tannery solid waste (TSW) contains 70-90% collagen by weight. The rapid extraction of collagen while preserving its original and unique triple-helical conformation proves challenging. In this paper, an ionic liquid-synergistic dissolution and regeneration (IL-SDR) method was developed based on TBAH/DMSO/H2O ternary (TDH) solvent for the rapid recovery of collagen from TSW under mild conditions. A solubility of 10.25% TSW and a collagen recovery yield of 51.2% were achieved in 30 minutes at room temperature. The original triplet helical conformation and the molecular weight of collagen product were remained, confirmed via FT-IR, circular dichroism, and matrix-assisted laser desorption/ionization time-of-flight analysis. SEM verified that dense and uniform pores could be formed within collagen by adjusting DMSO content, cytotoxicity and moisture retention tests demonstrated that the collagen produced via IL-SDR process is comparable to natural collagen in cosmetic and skincare applications. Moreover, TDH solvent demonstrated excellent reproducibility across five cycles, achieving consistent solubility varying from 9.71% to 11.04%. EPR and 1H NMR disclosed that the OH- in TBAH induced the generation of DMSO• free radicals in TDH solvent, and these radicals were indispensable for the dissolution of TSW. DMSO content was positively correlated with the concentration of free radicals. A synergistic dissolution mechanism was proposed: DMSO• free radicals preferentially cleaved the terminal peptide chains, causing the collagen molecules to be exposed to DMSO. Subsequently, DMSO disrupted the original hydrogen bond network within the collagen molecules. Then, TBAH and DMSO formed hydrogen bonds with the collagen molecules, stabilizing the dissolved collagen and preventing reformation of hydrogen bond network between collagen molecules. This work provided an avenue for low-temperature and rapid dissolution of TSW, and revealed an in-depth, atypical dissolution mechanism of collagen-based materials during IL-SDR process.

Optimization of Three Phase Partitioning Purification of Papain from Carica papaya Latex by Response Surface Methodology

Abstract:Three phase partitioning (TPP), a novel and non-chromatographic method was effectively applied for the extraction and recovery of papain from Carica papaya latex. The effect of change in ammonium sulfate concentration, crude extract to solvent ratio and pH on extraction efficiency was studied, and these parameters were optimized by response surface methodology (RSM) using Box-Behnken Design. The protease was found to be concentrated more at the aqueous phase (AP) with 51.08% ammonium sulfate precipitation, crude extract to tert-butanol ratio of 1:0.78 (v/v), and pH 6.05 resulting 9.242 purification fold and 159.806% activity recovery. Compared to the intermediate phase (IP), the AP demonstrated higher recovery and purification fold. Purified papain showed significantly (p<0.05) greater activity at a temperature of 60°C and pH of 6.0. It also demonstrated stability within the temperature range of 40 to 70°C and pH range of 6.0 to 8.0. Enzyme retained maximum milk clotting activity when stored under freezing (-20°C) for 3 weeks. This study showed that the TPP can be used to extract and purify papain from papaya latex economically and it can also be employed as a vegetable coagulant for cheesemaking.
