Abstract:Metal-organic framework (MOF) nanoparticles with high porosity and greater tunability have emerged as new drug delivery vehicles. However, premature drug release still remains a challenge in the MOF delivery system. Here, we report an enzyme-responsive, polymer-coated MOF gatekeeper system using hyaluronic acid (HA) and PCN-224 nanoMOF. The external surface of nanoMOF can be stably covered by HA through multivalent coordination bonding between the Zr cluster and carboxylic acid of HA, which acts as a gatekeeper. HA allows selective accumulation of drug carriers in CD44 overexpressed cancer cells and enzyme-responsive drug release in the cancer cell environment. In particular, inherent characteristics of PCN-224, which is used as a drug carrier, facilitates the transfer of the drug to cancer cells more stably and allows photodynamic therapy. This HA-PCN system enables a dual chemo and photodynamic therapy to enhance the cancer therapy effect.

Abstract:The effect of myocardial ischemia and reperfusion on left ventricular interstitial 8-hydroxydeoxyguanosine (8-OH-dG), a possible biomarker for in vivo oxidative deoxyribonucleic acid damage, in anesthetized rats was investigated. A microdialysis probe was implanted. Levels of 8-OH-dG in microdialysates were analyzed via an on-line high performance liquid chromatography system equipped with an electrochemical detector. Myocardial ischemia for 10 or 20 min, induced by clamping of the left anterior descending coronary artery, did not affect 8-OH-dG levels. However, reperfusion following either 10-min or 20-min ischemia significantly increased 8-OH-dG levels in collected microdialysates. Reperfusion-induced increases in 8-OH-dG levels were more prominent in the 20 min ischemia group (as high as 3.5 fold relative to basal levels) than in the 10 min ischemia group as high as 2.0 fold relative to basal levels). In conclusion, we observed that left ventricular interstitial 8-OH-dG concentration increased following myocardial ischemia and reperfusion in anesthetized rats. These results suggest that 8-OH-dG might be a useful biomarker for oxidative damage following myocardial ischemia and reperfusion.

Abstract:The injectable self-healing polysaccharide hydrogel was prepared by the formation of Schiff base bonds between aldehyde-modified methylcellulose (MCCHO) and carboxymethyl chitosan (CMC). The copper sulfide nanoparticles (CuS NPs) and pH-sensitive doxorubicin-loaded zeolitic imidazolate frameworks nanoparticles (DOX@ZIF-8 NPs) were prepared and could be well-dispersed into the hydrogel system. The presence of CuS NPs can achieve photothermal therapy (PTT) for tumors under the irradiation of the near-infrared (NIR) laser. Moreover, CuS NPs can generate photodynamic effects under NIR irradiation, converting oxygen into toxic reactive oxygen species (ROS) and presenting efficient photodynamic therapy (PDT). The DOX@ZIF-8 NPs can be decomposed under an intracellular acidic environment and realize the controlled release of DOX. The injectable self-healing hydrogel loading CuS and DOX@ZIF-8 NPs can achieve synergistic photothermal-photodynamic-chemo therapy for tumors and will inspire the researchers to construct a platform from hydrogel combined with multifunctional nanomaterials to realize the effective multimodal therapy for tumor.
Coupling metal organic frameworks with molybdenum disulfide nanoflakes for targeted cancer theranostics

Abstract:The superior properties of metal organic frameworks (MOF) can provide great opportunities for merging functional nanoparticles to construct smart and versatile cancer theranostic agents. In this study, on the basis of non-mesoporous nanoparticles (molybdenum disulfide, MoS2), the structure of the MOF shell layer with an adjustable structure can be constructed through the natural coordination interaction between polydopamine (PDA) and iron ion, and the tumor cell target ligand was modified on the surface of the nanocomposite after loading the anticancer drug doxorubicin hydrochloride (DOX) to form a multifunctional cancer theranostics nanoplatform (DOX@MoS2-PMA). Benefiting from the excellent properties of MoS2 and MOF, the favorable photothermal properties and pH/near-infrared (NIR) laser-triggered DOX release behavior of composite nanoparticles were demonstrated. Its well-defined nanostructure, adequate colloidal stability, and satisfactory biocompatibility were further evidenced. Furthermore, the selective tumor cell targeting ability of DOX@MoS2-PMA can improve the cellular uptake efficacy and the photothermal-chemotherapy combination therapy can significantly enhance the killing effect on cancer cells both in vitro and in vivo. In addition, fluorescence imaging results show that nanoparticles can efficiently accumulate inside tumors. The photoacoustic (PA) and magnetic resonance (MR) imaging capabilities derived from different components of nanoparticles can perform better imaging effects. To the best of our knowledge, this is the first attempt to merge the performance of MoS2 with MOF for PA/MR dual-modality imaging-guided photothermal-chemotherapy combination therapy. Our work presented herein proves that MOF can be combined with non-mesoporous nanoparticles and exhibits excellent performance, thus opening a new avenue for endowing non-mesoporous nanoparticles with an efficient drug loading capacity and practical applications of MOFs in nanomedicine.
pH-Responsive injectable self-healing hydrogels loading Au nanoparticles-decorated bimetallic organic frameworks for synergistic sonodynamic-chemodynamic-starvation-chemo therapy of cancer

Abstract:A novel and efficient cancer therapy was developed using a smart hydrogel containing multifunctional bimetallic organic frameworks and anticancer drugs. The injectable self-healing hydrogel with pH-responsiveness was constructed through borate ester and imine bonds among dopamine-grafted sodium alginate (SADA), hydroxypropyl chitosan (HPCS) and 2-formylphenylboronic acid (2-FPBA). The Au nanoparticles-decorated Ti/Fe bimetallic organic framework tetragonal nanosheets (Au/TF-MOF TNS) were synthesized and incorporated into the hydrogel with the anticancer drugs doxorubicin (DOX). Upon intratumoral injection of nanocomposite hydrogel, the acidic tumor microenvironment triggered the cleavage of borate ester and imine bonds, causing the hydrogel to break down and accelerating the release of both Au/TF-MOF TNS and DOX. These Au/TF-MOF TNS functioned as nanozymes, producing hydroxyl radicals (·OH) for chemodynamic therapy (CDT), generating oxygen (O2) to support sonodynamic therapy (SDT), and depleting glucose for starvation therapy (ST). Additionally, the Au/TF-MOF TNS served as sonosensitizers, capable of converting O2 into singlet oxygen (1O2) upon ultrasound irradiation to achieve SDT. Therefore, this nanocomposite hydrogel system enabled synergistic sonodynamic-chemodynamic-starvation-chemo therapy (SDT-CDT-ST-CT) of cancer, presenting a promising platform for advanced cancer therapy strategies.

Abstract:Synergistic therapies of photodynamic therapy (PDT) and chemodynamic therapy (CDT) via metal-organic frameworks (MOF) for cancer treatment have recently attracted a lot of attention because of the limitations of insufficient reactive oxygen species (ROS) in single-modality approaches. However, few studies explored on the use of increased ROS synergized with chemotherapy (CT) to address the issue of inadequate anti-tumor efficacy in single-modality regimens. Here, the desired cascade nanoplatforms (noted as MOF(Cu)@Dox-PL NPs) were fabricated by a solvothermal method using tetrakis (4-carboxyphenyl) porphyrin (TCPP) and zirconyl(di)chloride octahydrate (ZrOCl 2 ·8H 2 O) as raw material, followed by Cu 2+ introduced into the porphyrin ring and doxorubicin (DOX) loaded into the nanoframework. In addition, the nanoparticles (NPs) were electrostatically and hydrophobically coated with phospholipid (PL) to improve the biocompatibility of the nanosystems. Singlet oxygen ( 1 O 2 ) was created by the MOF(Cu)@Dox-PL NPs to disturb intracellular redox equilibrium. The acidic microenvironment in cancer cells may cause the prior release of DOX, which encourages the production of hydrogen peroxide (H 2 O 2 ). And the doped Cu 2+ could deplete overexpressed reduced glutathione (GSH) to produce hydroxyl radicals (·OH) by catalyzing H 2 O 2 , further causing redox dyshomeostasis. In vivo experiments revealed that MOF(Cu)@Dox-PL nanosystem possessed good biosafety and a compelling therapeutic effect in 4T1 tumor-bearing mice. As a novel nanosystem, MOF(Cu)@Dox-PL NPs showed great potential in synergistic therapy based on redox dyshomeostasis for improving anti-tumor efficacy with high specificity.

Abstract:Developing nanomedicines with high-performance is a promising therapeutic strategy for cancer treatment. Herein, a multifunctional nanoplatform (CuS@COFs-BSA-FA/DOX) based on copper sulfide nanoparticles engineered covalent organic frameworks (CuS@COFs) has been established for synergistic photothermal therapy (PTT), chemotherapy and chemodynamic therapy (CDT). The high photothermal effect and excellent Fenton-like catalytic activity enabled CuS@COFs to generate a new minimally invasive PTT/CDT synergistic therapy. Moreover, with the pH-dependent fluorescence behavior and the inherited mesoporous structure, the nanocomposites can also be utilized as fluorescent probes for cancer cell imaging and drug-delivery carriers to encapsulate the chemotherapeutic agent, DOX. The densely coated PEI and BSA-FA layers on the surface of CuS@COFs not only increased the cancer-targeting efficiency but also prevented the nonspecific release of DOX from CuS@COFs. The acid tumor microenvironment and near-infrared light triggered the release of DOX for chemotherapy, which simultaneously augmented the CDT efficiency of CuS@COFs. Notably, the localized hyperthermia induced by PTT can further improve the CDT efficiency of the nanoplatform, leading to a synergistic PTT/chemotherapy/CDT effect. The nanoplatform possessed the capability of cancer cell-targeted imaging and achieved better therapeutic efficacy with negligible systemic toxicity both in vitro and in vivo. Our work opens up a powerful avenue for developing multifunctional COFs-based theranostic platform and shows strong potential in practical applications.

Abstract:Although the potential of metal-organic framework (MOF) nanoparticles as drug delivery systems (DDS) for cancer treatment has been established by numerous studies, their clinical applications are still limited due to relatively poor biocompatibility. We fabricated a multifunctional Cu-MOFs@Keratin DDS for loaded drug and chemodynamic therapy (CDT) against tumor cells. The Cu-MOFs core was prepared using a hydrothermal method, and then loaded with the anticancer drug DOX and wrapped in human hair keratin. The Cu-MOFs@Keratin was well characterized by transmission electron microscopy (TEM), fourier transform infrared spectroscopy (FTIR), dynamic light scattering (DLS), and X-ray photoelectron spectroscopy (XPS). Characterization and pharmacokinetic studies of Cu-MOFs@Keratin were performed in vitro and in vivo. The keratin shell reduced the cytotoxicity and potential leakage of Cu-MOFs to normal cells, and allowed the drug-loaded nanoparticles to accumulate in the tumor tissues through enhanced permeability and retention effect (EPR). The particles entered the tumor cells via endocytosis and disintegrated under the stimulation of intracellular environment, thereby releasing DOX in a controlled manner. In addition, the Cu-MOFs produced hydroxyl radicals (·OH) by consuming presence of high intracellular levels of glutathione (GSH) and H2O2, which decreased the viability of the tumor cells.
Mixed‐Metal MOF‐Derived Hollow Porous Nanocomposite for Trimodality Imaging Guided Reactive Oxygen Species‐Augmented Synergistic Therapy

Abstract:Here an excellent trimodality imaging-guided synergistic photothermal therapy (PTT)/photodynamic therapy (PDT)/chemodynamic therapy (CDT) is proposed. To this end, a mixed-metal Cu/Zn-metal-organic framework (MOF) is first assembled at room temperature on a nano-scale. Interestingly, heating the MOF results in a Cu+/2+-coexisting hollow porous structure. Subsequent heating treatment is used to integrate Mn2+ and MnO2 in the presence of manganese(II) acetylacetonate. The hollow composite achieves efficient loading of a photosensitizer, indocyanine green (ICG). Under laser irradiation, the aggregated ICG achieves photothermal imaging and PTT. Once released in the tumor site, ICG exhibits fluorescence imaging and PDT capacity. Cu+/Mn2+ ions perform Fenton-like reaction with H2O2 to produce cytotoxic •OH for the enhanced CDT. Cu2+/MnO2 scavenge glutathione to improve the reactive oxygen species-based therapy, while the formed Mn2+ ions enable "turn on" magnetic resonance imaging. Significantly, O2 is produced from the catalytic decomposition of endogenous H2O2 to improve ICG-mediated PDT. Moreover, photothermal-induced local hyperthermia accelerates •OH generation to enhance CDT. This synergistic drug-free antitumor strategy realizes high treatment efficacy and low side effects on normal tissues. Thus, this mixed-metal MOF is an efficient strategy to realize hollow structures for multi-function integration to improve therapeutic capacity.

Abstract:Simultaneous photodynamic/photothermal therapy (PDT/PTT) is a combination cancer treatment that combines the principles of both PDT and PTT to achieve an effective and comprehensive attack on cancer cells. However, both approaches require an external light source to generate reactive oxygen species (ROS) and thermal heat, which could potentially hinder the therapeutic efficacy due to the low tissue penetration of light in vivo. Chemodynamic therapy (CDT) is a type of therapy that uses the ROS generated by the chemical reaction between a prodrug and a catalyst to kill cancer cells. We developed a combination therapy of three modalities (PTT/PDT/CDT) using the metal-organic framework (MOF). The Cu2+-doped MOF PCN-224 nanostructures modified with platinum (Pt) cluster and folic acid (FA) provided excellent tumor targeting, photothermal conversion ability, and ROS generated by the Pt cluster and Cu2+ under 650 nm light irradiation. Moreover, this multimodel nanocomposite had an extremely low dark toxicity but excellent phototoxicity under the combination laser irradiation at 650 nm, both in vitro and in vivo. Therefore, our prepared folic acid-conjugated PCN-224/Pt/Cu2+ (FA-PPC) nanosphere could be applied as a very promising multimodal phototherapeutic agent for enhanced cancer therapy in future clinical applications.

Abstract:Tumors produce a hypoxic environment that greatly influences cancer treatment, and conventional chemotherapeutic drugs cannot selectively accumulate in the tumor region because of the lack of a tumor targeting mechanism, causing increased systemic toxicities and side effects. Hence, designing and developing new nanoplatforms that combine multimodal therapeutic regimens is essential to improve tumor therapeutic efficacy. Herein, we report the synthesis of ultrafine Cu nanoparticles loaded with a drug combination of cisplatin (Pt) and 1-methyl-d-tryptophan (1-MT) and externally coated with 5,10,15,20-tetrakis(4-carboxyphenyl)porphyrin (TCPP) photosensitizer, polydopamine (PDA) and CaO2 of MIL-101(Fe) as a new nanoplatform (Cu@MIL-101@PMTPC). The nanoplatform synergistically combined chemodynamic therapy (CDT), photodynamic therapy (PDT), and immunochemotherapy. The Fe3+ in MIL-101(Fe) and the surface Cu nanoparticles exhibited strong ability to consume intracellular glutathione (GSH), thereby generating a Fenton-like response in the tumor microenvironment (TME) with substantial peroxidase (POD)-like and superoxide dismutase (SOD)-like activities. In this design, we used the indoleamine 2,3-dioxygenase (IDO) inhibitor 1-MT to overcome chemotherapy-induced immune escape phenomena including enhanced CD8+ and CD4+ T cell expression, interferon-gamma (IFN-γ) and tumor necrosis factor-alpha (TNF-α) production, and accelerated immunogenic cell death. The targeted release of cisplatin loaded into Cu@MIL-101@PMTPC also reduced toxic side effects of chemotherapy. TCPP generated a large amount of singlet oxygen (1O2) upon specific laser irradiation to effectively kill tumor cells. CaO2 on the outer layer generated oxygen (O2) and hydrogen peroxide (H2O2) to ameliorate hypoxia in the tumor microenvironment, enhance the PDT effect, and provide a continuous supply of H2O2 for the Fenton-like reaction. Thus, this nanocarrier platform exhibited a powerful chemodynamic, photodynamic, and immunochemotherapeutic cascade, providing a new strategy for cancer treatment.

Abstract:Non-invasive cancer therapies, especially those based on reactive oxygen species, including photodynamic therapy (PDT), have aroused great interest. As an emerging photodynamic nanocarrier, metal-organic framework (MOF) based on porphyrin that can release reactive oxygen species (ROS) to destructive cancer cells. However, due to the inefficient production of ROS by photosensitizers and the over-expression of glutathione (GSH) in tumor microenvironment (TME), the therapeutic effect is not satisfactory. Therefore, we developed a multi-functional nanoparticle HN@Cu-MOF to enhance the efficacy of PDT. We combined chemical dynamic therapy (CDT) and Nitric oxide (NO) therapy by initiating the sensitization to PDT and cell apoptosis in the treatment of tumors. The Cu2+-doped MOF reacted with GSH to form Cu+, exhibited strong CDT ability to generate hydroxyl radicals (·OH). Cu-MOF was coated with the HN, which is hyaluronicacid (HA) modified by a nitric oxide donor. HN can target tumor cells that over-expressed CD44 receptor and consume GSH in the cells to release NO. Both cell experiments and in vivo experiments showed an excellent tumor inhibitory effect under the treatment. Overall, the HN@Cu-MOF nanoparticle integrated NO gas therapy and CDT with PDT, leading to significant enhancing the GSH consumption and a remarkable elevation of ROS production.

Abstract:Porphyrinic metal–organic frameworks (MOFs) had promising applications in the photodynamic therapy (PDT) and chemodynamic therapy (CDT) of cancers, but the insufficient reactive oxygen species restricted the efficacy. Herein, by coordinating Cu 2+ and coating ultrasmall gold nanoparticles (AuNPs), a bi-functional porphyrinic MOFs (PCN-224) nanoplatform (PCN-224(Cu)-Au) was designed, achieving the dual-enhancement of PDT/CDT. The results indicated that the Cu 2+ was doped in the core of porphyrin ring, and the singlet oxygen ( 1 O 2 ) generation increased from 79.9% to 85.7%, owing to the enhanced capture capability of photogenerated electrons by Cu 2+ . Using the glucose oxidase-like property of ultrasmall AuNPs, the produced hydrogen peroxide (H 2 O 2 ) promoted the Fenton-like reaction of Cu + , by which the hydroxyl radicals (OH) generation increased from 63.2% to 80.3%, exhibiting the enhanced CDT performance. Under the combination of enhanced PDT/CDT, the cell viability reduced to 9.1%, and the tumors disappeared completely, demonstrating the dual-enhancement effect of PCN-224(Cu)-Au nanoprobe.

Abstract:The emerging chemodynamic therapy (CDT) has received an extensive attention in recent years. However, the efficiency of CDT is influenced due to the limitation of H2O2 in tumor. In this study, we designed and synthesized a novel core-shell nanostructure, Cu-metal organic framework (Cu-MOF)/glucose oxidase (GOD)@hyaluronic acid (HA) (Cu-MOF/GOD@HA) for the purpose of improving CDT efficacy by increasing H2O2 concentration and cancer cell targeting. In this design, Cu-MOF act as a CDT agent and GOD carrier. Cu(II) in Cu-MOF are reduced to Cu(I) by GSH to obtain Cu(I)-MOF while GSH is depleted. The depletion of GSH reinforces the concentration of H2O2 in tumor to improve the efficiency of CDT. The resultant Cu(I)-MOF catalyze H2O2 to generate hydroxyl radicals (·OH) for CDT. GOD can catalyze glucose (Glu) to supply H2O2 for CDT enhancement. HA act as a targeting molecule to improve the targeting ability of Cu-MOF/GOD@HA to the tumor cells. In addition, after loading with GOD and coating with HA, the proportion of Cu(I) in Cu-MOF/GOD@HA is increased compared with the proportion of Cu(I) in Cu-MOF. This phenomenon may shorten the reactive time from Cu-MOF to Cu(I)-MOF. The CDT enhancement as a result of GOD and HA effects in Cu-MOF/GOD@HA was evidenced by in vitro cell and in vivo animal studies.

Active-transporting of charge-reversal Cu(II)-doped mesoporous silica nanoagents for antitumor chemo/chemodynamic therapy

Abstract:Fe-based Fenton agents can generate highly reactive and toxic hydroxyl radicals (·OH) in the tumor microenvironment (TME) for chemodynamic therapy (CDT) with high specificity. However, the low pH environment and insufficient endogenous hydrogen peroxide (H2O2) of the highly efficient Fenton reaction limits its practical application in clinic. Here, a Cu(II)-doped mesoporous silica nanoagent (Cu-MSN) with excellent dispersity was successfully developed. After loaded with doxorubicin (DOX) and ascorbate (AA), Cu-MSN@DA was coated with active targeting ligand folic acid (FA), dimethyl maleic an-hydride (DMMA) and carboxymethyl chitosan (CMC) to obtain an active transporting nanoagent (FCDC@Cu-MSN@DA) with tunable charge-reversal property, which is more adaptable to the pH value of TME than Fe-based Fenton agents, and can self-supply exogenous H2O2 by ascorbate to produce more toxic ·OH to trigger the apoptosis of cancer cells. Meanwhile, the high level of glutathione (GSH) in TME can reduce Cu(II) to Cu(I) by Fenton-like reaction, increasing the generation rate of ·OH and relieving tumor antioxidant ability. The supply of exogenous H2O2 significantly enhanced the synergistic effect of CDT by oxidative damage. Together with DOX-induced cell apoptosis, this novel nanoagent FCDC@Cu-MSN@DA can achieve maximum therapeutic efficacy, creating a new model of safe and effective tumor treatment with high specificity.

Abstract:Nowadays, the therapeutic efficiency of tumor can be significantly influenced by the limitation of endogenous tumor microenvironment (TME). An ideal solution for improving therapeutic efficiency is constructing TME-responsive and multifunctional nanoreactor. Herein, a TME-responsive nanoreactor has been constructed using novel dendritic mesoporous silica nanoparticles (DMSNs) as the cornerstone, followed by anchoring platinum-(IV) prodrugs, loading photosensitizer indocyanine green (ICG), and decorating Cu2+ carboxylate MOFs. The prepared nanoreactor provides a high loading efficiency for Pt prodrugs and ICG, while the externally decorated MOFs can prevent premature leakage in neutral physiological environment at the same time. However, after accumulation at the tumor site, the outer MOFs can be rapidly decomposed in the TME to release Cu2+, which achieves efficient chemodynamic therapy (CDT) through Cu2+-triggered Fenton-like reaction and GSH depletion ability. At the same time, the released Pt drugs can achieve efficient chemotherapy. Moreover, the near-infrared (NIR) can activate the photodynamic and photothermal therapy (PDT&PTT) ability of ICG, resulting in PTT-enhanced CDT. Meanwhile, the nanoreactor can exhibit photothermal and magnetic resonance imaging (MRI) ability for highly specific diagnosis of tumor. Overall, this study provides a paradigm of TME-responsive nanoreactor for collaboratively multimodal enhanced tumor therapy.
Mesoporous Rod‐Like Metal‐Organic Framework with Optimal Tumor Targeting Properties for Enhanced Activatable Photodynamic Therapy

Abstract:Nanoscale metal‐organic frameworks (nMOFs) have shown great potential in the highly effective delivery of exogenous drugs and imaging agents for biomedical applications, and rod‐like nanomaterials with superb loading capability have demonstrated broad versatility to tumor imaging and tumor‐targeted drug delivery. However, owing to difficulties in their fabrication, nMOFs with both rod‐like shape and mesoporosity for tumor imaging or tumor‐targeted cargo delivery are largely unexplored. Herein, a nanoscale rod‐like [{Cu2(ndc)2(dabco)]n (Cu‐MOF), a copper(II) nMOF, is successfully endowed with mesoporosity via defect engineering. Owing to the mesopores, the as‐synthesized defective rod‐like Cu‐MOF is able to load an aggregation‐induced‐emission (AIE) photosensitizer (PS) with inhibited sensitization at high loading weight percentage. In vitro and in vivo experiments demonstrate that the rod‐like Cu‐MOF can afford the AIE PS payload with enhanced tumor targeting specificity and penetration depth. After optimal tumor enrichment and cancer cell uptake, Cu(II) in the defective Cu‐MOF efficiently depletes intracellular glutathione and causes dissociation of the Cu‐MOF to release the PS payload and recover its efficient reactive oxygen species generation. Concomitantly, bright fluorescence is afforded by the AIE PS during delivery, contributing to image‐guided cancer‐cell‐activated photodynamic therapy with enhanced therapeutic effects.

Abstract:Chemodynamic therapy (CDT) can kill cancer cells by generating toxic agents within tumors through Fenton-/Fenton-like reaction, exhibiting high tumor specificity and low systemic toxicity. However, the therapeutic effect of CDT is usually limited by the overexpressed glutathione (GSH) and low catalytic efficiency in the tumor microenvironment. To deplete GSH and achieve chemo/photothermal/chemodynamic combination therapy, Cu2+, the photothermal material polydopamine (PDA), and chemotherapeutic agent DOX were used to construct a copper-doped MOF-based nanocomposite (2+/, DCZP). The Cu2+ could be reduced to Cu+ by GSH to achieve GSH depletion and CDT activation. Under laser irradiation, the heat generated by DCZP could be used for photothermal therapy and promote the Fenton-like reaction with the released DOX to achieve combination therapy. Extracellular experiments demonstrated that more than 50% of GSH could be consumed and the generated heat upon laser irradiation could promote •OH production. In vitro experiments showed that the DCZP combined therapy resulted in 49.4% of late cell apoptosis. More importantly, under the combination therapy of DCZP, the tumors of mice were almost ablated and the expression of tumor growth factors also could be suppressed. The work provided a promising strategy by constructing a GSH depletion and combination therapy nanocomposite for safe and efficient cancer therapy.

Abstract:In this study, a novel cascade Fe‐based MOFs nanodrug delivery system MIL‐101(Fe)‐DOX‐DHA@TCPP/GOx@PDA (MDDTG@P) is constructed for triple synergistic hepatocellular carcinoma therapy, in combination with chemodynamic, photodynamic, and starvation therapy, which provides a feasible strategy to promote synergistic modes of cancer therapy. The transformation of monotherapy into multimodal combined targeted therapy to fully exploit synergistic efficacy is of increasing interest in tumor treatment. In this work, a novel nanodrug‐carrying platform based on iron‐based MOFs, which is loaded with doxorubicin hydrochloride (DOX), dihydroartemisinin (DHA), and glucose oxidase (GOx), and concurrently covalently linked to the photosensitizer 5,10,15,20‐tetrakis(4‐carboxyphenyl)porphyrin (TCPP) in polydopamine (PDA)‐encapsulated MIL‐101(Fe) (denoted as MIL‐101(Fe)‐DOX‐DHA@TCPP/GOx@PDA, MDDTG@P), is successfully developed. Upon entering the tumor microenvironment, MDDTG@P catalyzes the hydrogen peroxide (H2O2) into hydroxyl radicals (·OH) and depletes glutathione (GSH); thus, exerting the role of chemodynamic therapy (CDT). The reduced Fe2+ can also activate DHA, further expanding CDT and promoting tumor cell apoptosis. The introduced GOx will rapidly consume glucose and oxygen (O2) in the tumor; while, replenishing H2O2 for Fenton reaction, starving the cancer cells; and thus, realizing starvation and chemodynamic therapy. In addition, the covalent linkage of TCPP endows MDDTG@P with good photodynamic therapeutic (PDT) properties. Therefore, this study develops a nanocarrier platform for triple synergistic chemodynamic/photodynamic/starvation therapy, which has promising applications in the efficient treatment of tumors.
