Abstract:Biotic stress deleteriously affects growth, development, and productivity in plants. Proline (Pro) plays a significant role in enhancing plant resistance to pathogen infection. However, its effects on reducing Lelliottia amnigena-induced oxidative stress in potato tubers remain unknown. The present study aims to evaluate the in vitro Pro treatment in potato tubers exposed to a newly emerging bacterium, L. amnigena. Sterilized healthy potato tubers were inoculated with 0.3 mL of L. amnigena suspension (3.69 × 107 CFU mL-1) 24 h before Pro (5.0 mM) application. The L. amnigena treatment significantly increased the contents of malondialdehyde (MDA) and hydrogen peroxide (H2O2) in the potato tubers by 80.6 and 85.6%, respectively, compared to the control. Application of proline (Pro) decreased MDA and H2O2 contents by 53.6 and 55.9%, respectively, compared to the control. Application of Pro to L. amnigena-stressed potato tubers increased the activities of NADPH oxidase (NOX), superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), polyphenol oxidase (PPO), phenylalanine ammonia-lyase (PAL), cinnamyl alcohol dehydrogenase (CAD), 4-coumaryl-CoA ligase (4CL) and cinnamate-4-hydroxylase (C4H) C4H by 94.2, 96.3, 97.3, 97.1, 96.6, 79.3, 96.4, 93.6, and 96.2%, respectively, compared to the control. In comparison to the control, the genes PAL, SOD, CAT, POD, and NOX were significantly increased in the Pro-treated tubers at 5.0 mM concentration. Tubers treated with Pro + L. amnigena increased the transcript levels of PAL, SOD, CAT, POD, and NOX by 2.3, 2.2, 2.3, 2.5, and 2.8-fold respectively, compared to the control. Our findings suggested that pretreatment of tubers with Pro might reduce lipid peroxidation and oxidative stress by enhancing enzymatic antioxidant activity and gene expression.
Isolation and in vitro screening of plant growth promoting bacteria from rhizosphere and root tissues of potato tuber (Solanum tuberosum L.)

Abstract:The accumulation of chemical fertilizers that harm the environment is one of the major Indonesian agricultural problems. However, it still has less effect on potato production and yield. The discovery and use of bacteria that have the potential as plant growth-promoting agents (PGPR) is a breakthrough that can help to increase growth to increase production, especially in potato plants. In this study, several bacteria successfully isolated from the rhizosphere and root tissue of potato plants (Solanum tuberosum L.) were isolated from potato farms in Plaosan Village. Several in vitro screenings were carried out to assess the functional activity of plant growth promoters, including the IAA (indole-3-acetic acid) production test, siderophore production test, ACC (1-aminocyclopropane-1-carboxylate) deaminase production test and phosphate dissolution test. Based on the screening results, five isolates were considered as the best inoculants, there are R1.3, R2.2, JR2.1, E1.2 and E1.2.1. All R1.3, R2.2, E1.2 and E1.2.1 isolates were known to have the ability to produce phytohormones IAA, ACC deaminase, and siderophores. In contrast, JR2.1 isolate was not known to have the ability to fix nitrogen and produce IAA, ACC deaminase and siderophores. These isolates could be used as potential biofertilizer inoculants and provide a step towards sustainable agriculture.

Abstract:Black scurf and stem canker caused by Rhizoctonia solani is a significant disease problem of potatoes. Currently, chemical methods are the primary means of controlling this pathogen. This study sought to explore an alternative approach by harnessing the biocontrol potential of a bacterial mix of Bacillus subtilis and Bacillus amyloliquefaciens against black scurf, and to determine their effect on rhizosphere microorganisms of soil microbiota. This study showed that these bacteria demonstrate antagonistic activity against Rhizoctonia solani. Reduced damage to potato plants during the growing season in Siberia was observed. The index of disease development decreased from 40.9% to 12.0%. The treatment of tubers with this mix of bacteria also led to a change in the composition of the rhizosphere microbiota (according to CFU, 16S and ITS sequencing). This effect was accompanied by a positive change in plant physiological parameters (spectrophotometric analysis). The concentration of chlorophyll in potatoes with the bacterial mix treatment increased by 1.3 fold (p ≤ 0.001), and of carotenoids by 1.2 fold (p ≤ 0.01) compared with the control. After bacterial mix treatment, the length of the aerial parts of plants was 1.3 fold higher (p ≤ 0.001), and the number of stems 1.4 fold higher (p ≤ 0.05). The yield of potatoes was increased by 8.2 t/ha, while the large tuber fraction was increased by 16% (p ≤ 0.05). The bacteria mix of Bacillus subtilis and Bacillus amyloliquefaciens suppressed the plant pathogenic fungus Rhizoctonia solani, and simultaneously enhanced the physiological parameters of potato plants. This treatment can be used to enhance the yield/quality of potato tubers under field conditions.

Abstract:Plant growth-promoting rhizobacteria (PGPR) represent a set of microorganisms that play significant role in improving plant growth and controlling the phytopathogens. Unpredictable performance after the application of PGPR has been observed when these were shifted from in-vitro to in-vivo conditions due to the prevalence of various abiotic stress conditions. During growing period, the potato crop is subjected to a combination of biotic and abiotic stresses. Rhizoctonia solani, a soil-borne plant pathogen, causes reduced vigor and yield of potato crop worldwide. In the current study, multi-stress-tolerant rhizobacterial strain, Bacillus subtilis PM32, was isolated from field-grown potato with various plant growth promoting (PGP) traits including zinc and potassium solubilization, biological nitrogen fixation, ammonia and siderophore, as well as extracellular enzyme productions (cellulase, catalase, amylase, protease, pectinase, and chitinase). The strain PM32 exhibited a distinct potential to support plant growth by demonstrating production of indole-3-acetic acid (102.6 μM/mL), ACC-deaminase activity (1.63 μM of α-ketobutyrate/h/mg protein), and exopolysaccharides (2.27 mg/mL). By retarding mycelial growth of R. solani the strain PM32 drastically reduced pathogenicity of R. solani. The strain PM32 also suppressed the pathogenic activity significantly by impeding mycelial expansion of R. solani with inhibition co-efficient of 49.87. The B. subtilis PM32 also depicted significant tolerance towards salt, heavy metal (Pb), heat and drought stress. PCR based amplification of ituC and acds genes coding for iturin and ACC-deaminase activity respectively indicated potential of strain PM32 for lipopeptides production and ACC deaminase enzyme activity. Results of both in-vitro and pot experiments under greenhouse conditions depicted the efficiency of B. subtilis PM32 as a promising bio-control agent for R. solani infection together with enhanced growth of potato plants as deciphered from biomass accumulation, chlorophyll a, b, and carotenoid contents. Therefore, it was envisioned that application of indigenous multi-stress tolerant PGPR may serve to induce biotic and abiotic stress tolerance in crops/plants for pathogen control and sustainable global food supply.

Multi-omics analysis reveals the effects of three application modes of plant growth promoting microbes biofertilizer on potato (Solanum tuberosum L.) growth under alkaline loess conditions

Abstract:Potato is an important crop due to its high contents of starch, protein, and various vitamins and minerals. Biofertilizers are composed of plant growth promoting microbes (PGPMs) which are essential for improving the growth and resistance of potato. However, little information has focused on the modes of inoculation of biofertilizers on plant growth and microecology. This study aims to reveal the response mechanism of the potato to three modes of inoculation of biofertilizers all containing PGPM Bacillus amyloliquefaciens EZ99, i.e. scattered mode of 5 kg/ha biofertilizer (M5), soaking seed tubers with dissolved 5 kg/ha biofertilizer (MZG), and scattered mode of 3 kg/ha biofertilizer + 2 kg/ha sucrose (MY34) in alkaline loess field through multi-omics analysis of transcriptome, metabolome and microbiome. The physiological result revealed that two application modes of equal amount of biofertilizer M5 and MZG significantly improved the growth and yield of potatoes. Furthermore, the transcriptome of potato exhibited sets of differentially expressed genes enriched in photosynthesis, sugar metabolism, and phenylpropanoid biosynthesis among the three modes, with the M5 mode exhibiting overall up-regulation of 828 genes. Based on the untargeted metabolomic analysis of potato tuber, M5 mode significantly accumulated sucrose, while MZG and MY34 mode significantly accumulated the stress metabolites euchrenone b6 and mannobiose, respectively. Besides, the microbial structure of potato rhizosphere showed that the diversity of bacteria and fungi was similar in all soils, but their abundances varied significantly. Specifically, beneficial Penicillium was enriched in M5 and MZG soils, whereas MY34 soil accumulated potential pathogens Plectosphaerella and saccharophilic Mortierella. Collectively, these e findings highlight that MZG is the most effective mode to promote potato growth and stimulate rhizosphere effect. The present study not only encourages sustainable agriculture through agroecological practices, but also provides broad prospects for the application of PGPM biofertilizer in staple foods.

Abstract:Potato soft rot and wilt are economically problematic diseases due to the lack of effective bactericides. Bacteriophages have been studied as a novel and environment-friendly alternative to control plant diseases. However, few experiments have been conducted to study the changes in plants and soil microbiomes after bacteriophage therapy. In this study, rhizosphere microbiomes were examined after potatoes were separately infected with three bacteria (Ralstonia solanacearum, Pectobacterium carotovorum, Pectobacterium atrosepticum) and subsequently treated with a single phage or a phage cocktail consisting of three phages each. Results showed that using the phage cocktails had better efficacy in reducing the disease incidence and disease symptoms' levels when compared to the application of a single phage under greenhouse conditions. At the same time, the rhizosphere microbiota in the soil was affected by the changes in micro-organisms' richness and counts. In conclusion, the explicit phage mixers have the potential to control plant pathogenic bacteria and cause changes in the rhizosphere bacteria, but not affect the beneficial rhizosphere microbes.
Regulation of antioxidant production, ion uptake and productivity in potato (Solanum tuberosum L.) plant inoculated with growth promoting salt tolerant Bacillus strains

Abstract:The exchangeable sodium (Na+) in salt affected soils is a major constraint in potassium (K+) availability to plants that disturb ion transport and inhibit plant growth, adversely. Salt tolerant plant growth promoting rhizobacteria (PGPR) may regulate the Na+/K+ efflux and increase K+ uptake by the plant from the soil. Therefore, a pot study was performed to examine the effect of salt tolerant PGPR Bacillus sp. alone and in consortium, on antioxidant enzyme activity, ion uptake and potato (Solanum tuberosum L.) tuber yield in normal and salt affected soils. We observed that Bacillus sp. (strains SR-2-1 and SR-2-1/1) solubilized insoluble phosphorous and produced indole-3-acetic acid while only SR-2-1/1 produced ACC deaminase in culture medium supplemented with various concentrations of NaCl (0-6%). In the pot experiment, the consortium treatment of strains was found to increase relative leaf water contents whereas decreased the electrolyte leakage and antioxidant enzyme activity both in normal and salt affected soils. Similarly, consortium treatment decreased Na+ whereas increased K+, Ca+2, K+/Na+ and Ca+2/Na+ in plant dry matter in both soils. It has been investigated that inoculation of PGPR significantly (p < 0.05) increased plant biomass, number of tubers per plant and tuber weight as compared to un-inoculated plants in both soils. In addition, PGPR inoculation enhanced auxin production in root exudates of young potato plants and bacterial population dynamics in both soils. Na+ ion regulation (R2 = 0.95) and tuber weight (R2 = 0.90) in salt affected soil were significantly correlated with auxin production in the rhizosphere. Results of this study conferred that consortium of Bacillus strains (SR-2-1, SR-2-1/1) enhanced auxin production in the rhizosphere of potato plants and that ultimately regulated antioxidant enzyme production and uptake of Na+, K+ and Ca+2 in potato plants resulted into a higher tuber yield in both normal and salt affected soils.

Abstract:Plant growth-promoting rhizobacteria (PGPR) can be used to increase plant growth rates in natural field conditions, as well as in micropropagation of plants in vitro and in substrate-free aero/hydroponic systems. This study demonstrates the ability of rhizobacteria Azospirillum baldaniorum Sp245 and Kocuria rosea T1Ks19 to stimulate the cultivation of potato microplants of the 'Nevsky' and 'Kondor' cultivars in in vitro culture, to increase the rate of adaptation of microplants to unsterile conditions of aeroponics ex vitro, and also to increase the productivity of minitubers. The conclusions obtained are based on the results of morphometric analysis of microplants, cytological analysis of stomata and biochemical assessment of the content of antioxidant enzymes (catalase, peroxidase and ascorbate peroxidase), as well as hydrogen peroxide and malondialdehyde in the leaves. The presence of bacteria on plant roots was monitored using enzyme immunoassay and CFU counting methods. It has been established that inoculation with the used combination of rhizobacteria stimulates the synthesis of antioxidant enzymes in microplants under stressful conditions, which leads to a decrease in the level of hydrogen peroxide. As a result, the adaptation efficiency, growth rates and productivity of micro-plants increase. It is recommended to inoculate potato microplants with A. baldaniorum Sp245 and K. rosea T1Ks19 strains at the stage of in vitro cultivation in the production of healthy seed material based on aeroponics in the potato seed production system.

Abstract:Potato is a major crop whose yield and quality are impacted by the pathogenic fungus Rhizoctonia solani. Here, a control approach based on the use of endophytic bacillus spores and bacterial consortia was assessed as an innovative solution to improve tuber productivity and health. Endophytic potato bacteria extracted from several organs (leaves, stems and roots) evaluated for their antagonistic activity against R. solani by the double culture technique and their ability to rot potato slices. Additionally, the ability of Bacillus strains to generate hydrolytic enzymes and produce lipopeptides (using ESI-MS/MS mass spectrometry) was assessed. Then, the effectiveness of selected bacterial strains, either individually or in consortium (after carrying out compatibility tests) in the suppression of R. solani was evaluated in the greenhouse. Six endophytes not affecting potato tubers and having an antagonistic effect against R. solani were identified: B. halotolerans SpS5, B. velezensis KnL15, B. aryabhattai FaR1, B. amyloliquefaciens LiR9, B. haynesii ReR10 which produce several types and combinations of lipopeptides belonging to different families (iturins, fengycins and surfactins) as well as hydrolytic enzymes (chitinase, protease and cellulase). The vegetative and sporulating strain SpS5, as well as the 5 other Bacillus and their Sps5-based consortia, were found to be very effective in increasing the yield of healthy, high-quality tubers and reducing disease severity. Based on the results mentioned above, a microbial fungicide from the spores of (i) B. halotolerans SpS5 for which we sequenced the whole genome or (ii) bacterial consortia could be developed to combat black scurf potato and other fungal infections.

Abstract:Adaptation ex vitro is strongly stressful for microplants. Plant-growth-promoting rhizobacteria (PGPR) help to increase the adaptation potential of microplants transplanted from test tubes into the natural environment. We investigated the mechanisms of antioxidant protection of PGPR-inoculated potato microclones adapting to ex vitro growth in an aeroponic system. Potato (Solanum tuberosum L. cv. Nevsky) microplants were inoculated in vitro with the bacteria Azospirillum baldaniorum Sp245 and Ochrobactrum cytisi IPA7.2. On days 1 and 7 of plant growth ex vitro, catalase and peroxidase activities in the leaves of inoculated plants were 1.5-fold higher than they were in non-inoculated plants. The activity of ascorbate peroxidase was reduced in both in vitro and ex vitro treatments, and this reduction was accompanied by a decrease in the leaf content of hydrogen peroxide and malondialdehyde. As a result, inoculation contributed to the regulation of the plant pro/antioxidant system, lowering the oxidative stress and leading to better plant survival ex vitro. This was evidenced by the higher values of measured morphological and physiological variables of the inoculated plants, as compared with the values in the control treatment. Thus, we have shown some PGPR-mediated mechanisms of potato plant protection from adverse environmental factors under aeroponic conditions.
Short impact on soil microbiome of a Bacillus amyloliquefaciens QST713 based product that correlates with higher potato yield across USA

Abstract:Potato ( Solanum tuberosum L. ) is considered one of the most widely consumed crops worldwide, due to its high yield and nutritional profile, climate change-related environmental threats and increasing food demand. This scenario highlights the need of sustainable agricultural practices to enhance potato productivity, while preserving and maintaining soil health. Plant growth-promoting bacteria (PGPB) stimulate crop production through biofertilization mechanisms with low environmental impact. For instance, PGPB promote biological nitrogen fixation, phosphate solubilization, production of phytohormones, and biocontrol processes. Hence, these microbes provide a promising solution for more productive and sustainable agriculture. In this study, the effects of Bacillus amyloliquefaciens QST713 based-product (MINUETTM, Bayer) were assessed in terms of yield, soil microbiome, potato peel and petiole nutrient profile as a promising PGPB in a wide range of potato cultivars across the United States of America. Depending on the location, potato yield and boron petiole content increased after biostimulant inoculation to maximum of 24% and 14%, respectively. Similarly, nutrient profile in potato peel was greatly improved depending on the location with a maximum of 73%, 62% and 36% for manganese, zinc and phosphorus. Notably, fungal composition was shifted in the treated group. Yield showed strong associations with specific microbial taxa, such as Pseudoarthrobacter, Ammoniphilus, Ideonella, Candidatus Berkiella, Dongia . Moreover, local networks strongly associated with yield, highlighting the important role of the native soil microbiome structure in indirectly maintaining soil health. Our results showed that treatment with B. amyloliquefaciens based product correlated with enhanced yield, with minor impacts on the soil microbiome diversity. Further studies are suggested to disentangle the underlying mechanisms of identified patterns and associations.

Abstract:About 200 Bacillus isolates were isolated from tomato and potato rhizosphere and examined for their antagonistic activities against Ralstonia solanacearum T-91, the causal agent of tomato bacterial wilt (TBW), in vitro and in vivo . Four strains, AM1, D16, D29 and H8, have shown high potential of antagonistic activity against the pathogen in laboratory and greenhouse experiments. In greenhouse, 81.1 to 89.0% reduction of disease incidence of TBW was recorded in treated tomato plants with 4 isolates, which also significantly (p > 0.05) increased plant height by 22.7 to 43.7% and dry weight by 47.93 to 91.55% compared with non-treated control. 16SrRNA gene sequence, the biochemical and physiological tests and fatty acid methyl esters analysis assigned strains AM1 and D29 as Bacillus amyloliquefaciens , while strains D16 and H8 as Bacillus subtilis and B. methylotrophicus , respectively. In addition, the 4 strains showed ability to inhibit growth of the three soil-borne fungi, produce indole-3- acetic acid, siderophores and also with exception of strain D16, the other 3 strains were capable of solubilizing phosphate. Therefore, these results suggest that out of 200 isolates, Bacillus stains AM1, D16, D29 and H8 support good antagonistic activity and could be applied as biocontrol agents against TBW under greenhouse conditions beside their potential to promote tomato plants growth. The objective of this study was to assess the potential of biofumigation using three Brassica crops including Brassica nap us, B. juncea and B. campestris against potato stem rot caused by S. sclerotiorum by in vitro tests. Both macerated and irradiated dried tissues were able to reduce radial growth and sclerotia formation of five pathogen isolates on PDA, but macerated live tissues were more effective. Compared with other tested crops, B. juncea showed more inhibitory effect against the pathogen. The volatile compounds produced from macerated tissues were identified using a gas chromatograph-mass spectrometer. The main identified compounds were methyl, allyl and butyl iso-thiocyanates. Different concentrations of these compounds inhibited mycelial growth of the pathogen in vitro when applied as the vapor of pure chemicals. A negative relationship was observed between chemicals concentrations and growth inhibition percentage. In this study, it became clear that the tissues of local Brassica crops release glucosinolates and have a good potential to be used against the pathogen in field examinations.
Bacillus velezensis Strain HN-Q-8 Induced Resistance to Alternaria solani and Stimulated Growth of Potato Plant

Abstract:Bacillus velezensis HN-Q-8, isolated in our previous study, has an antagonistic effect on Alternaria solani. After being pretreated with a fermentation liquid with HN-Q-8 bacterial cell suspensions, the potato leaves inoculated with A. solani displayed smaller lesion areas and less yellowing than the controls. Interestingly, the activity levels of superoxide dismutase, peroxidase, and catalase in potato seedlings were enhanced by the addition of the fermentation liquid with bacterial cells. Additionally, the overexpression of key genes related to induced resistance in the Jasmonate/Ethylene pathway was activated by the addition of the fermentation liquid, suggesting that the HN-Q-8 strain induced resistance to potato early blight. In addition, our laboratory and field experiments showed that the HN-Q-8 strain can promote potato seedling growth and significantly increase tuber yield. The root activity and chlorophyll content of potato seedlings were significantly increased along with the levels of indole acetic acid, gibberellic acid 3, and abscisic acid upon addition of the HN-Q-8 strain. The fermentation liquid with bacterial cells was more efficient in inducing disease resistance and promoting growth than bacterial cell suspensions alone or the fermentation liquid without bacterial cells. Thus, the B. velezensis HN-Q-8 strain is an effective bacterial biocontrol agent, augmenting the options available for potato cultivation.

THE EFFECT OF BACTERIAL BIOFERTILIZER (AZOSPIRILLUM BRASILENSE), COMPOST AND CHEMICAL FERTILIZER ON THE GROWTH AND YIELD OF POTATO PLANT (SOLANUM TUBEROSUM L.)

Abstract:The nitrogen-fixing bacteria were isolated from the (10) root samples of different plants using the semi-solid culture medium (NFB), where the isolated bacterial isolates were identified based on the culture and microscopic traits and biochemical tests. The identification results showed the presence of two isolates belonging to the Azospirillum brasilense and was given a symbol A2 and A5, then their ability to nitrogen fixation was tested in the laboratory, and it was observed that the isolate A5 was more able to nitrogen fixation by 11.78 mg N.kg-1. The most efficient isolate of nitrogen fixation was grown in the liquid culture medium (Nutrient.broth) several times to use a biological inoculant. Potato tubers were inoculated with the biological inoculant and Arabic gum, then they were planted in rows after adding organic and chemical fertilizers before planting, then the irrigation was done by the drip irrigation system. Finally, The results showed that the highest concentration of nitrogen% in potato tubers was for treatment (B1) Azospirillum brasilense with organic fertilizer by an average of 2.06 N% compared to the comparison treatment 0.40 N%. In addition, it achieved the highest concentration of available nitrogen in the soil by an average of 44.28 mg N.kg-1 soil compared to the comparison treatment that achieved 16.15 mg N.kg-1 soil. The treatment of Azospirillum brasilense with organic fertilizer (B1O1) also exceeded in the total yield of the potato plant and achieved an average of 33.03 Mg. Ha-1 compared to the comparative treatment of 10.71 Mg. Ha-1. These results confirm that the bacterial biofertilizer with organic and chemical fertilizer achieved the best results for some potato growth parameters and yield.

Abstract:Plant growth-promoting rhizobacteria are helpful soil microorganisms that are used extensively in the agricultural industry. In the current investigation, a total of 120 extremophilic bacterial isolates were successfully obtained from a variety of drastic soils. These bacterial isolates were tested to determine their effectiveness as plant growth promoters and biocontrol agents against the two most frequent soil-borne plant pathogenic fungi, Pythium ultimum, and Rhizocotnia solani. Results indicated that numerous isolates demonstrated antifungal activity against P. ultimum and R. solani, through the production of diffusible antifungal metabolites, cell-wall degrading enzymes (chitinase, cellulase), and siderophores. In addition, many of these extremophilic bacteria fixed atmospheric nitrogen, solubilized insoluble phosphate and zinc, and produced indole-3-acetic acid (IAA) and salicylic acid. The 16 S rRNA analysis revealed that the most potent isolates with multiple modes of action were identified as Bacillus halotolerans, Massilia alkalitolerans, and Bacillus aryabhattai. Under greenhouse conditions, these three isolates, either individually or in combination were used as microbial inoculants, and their effects on the development of tomato and maize were evaluated both with and without the presence of the two pathogens. Application of the three isolates as individual or combined treatments significantly (P < 0.05) reduced the growth of P. ultimum and R. solani in vivo, and significantly (P < 0.05) increased the percentage of survived seedlings as evidenced by the significant (P < 0.05) increase in plant resistance as measured by the in planta contents of total phenols, polyphenol oxidase, and peroxidase. Additionally, the treatment of any one of the three isolates, either individually or in combination, significantly (P < 0.05) improved the development of tomato and maize, as evidenced by a considerable increase in the amount of total nitrogen and dry weight yield. The utilization of these bacterial isolates presents a potentially fruitful alternative to the excessive use of potentially harmful agrochemicals.

Abstract:In sustainable agriculture, seeking eco-friendly methods to promote plant growth and improve crop productivity is a priority. Humic acid (HA) and plant growth promoting rhizobacteria (PGPR) are among the most effective methods that utilize natural biologically-active substances. The aim of the present study was to analyze the effect of the presence of HA on potato (Solanum tuberosum L.) inoculation with PGPR (Bacillus megatorium and Bacillus subtilis) when compared to control and recommended doses of NPK. Seed tubers treated by humic acid (200, 400, and 600 kg ha−1) and PGPR, separately or in combination, and NPK (50% and 100%) were planted into soil and untreated soil. Treatments were assessed for plant growth, classified tuber yields, quality, and mineral contents of potato tubers. There were highly significant increases in potato growth, tuber yields, and quality in PGPR and HA inoculated crops. Tuber size, weight, specific gravity, dry matter, starch, protein, and mineral contents (except Cu) were improved with PGPR treatments and further increased when administered with humic acids. Inoculation with PGPR mixed culture and 400 kg ha−1 HA increased total potato tuber yield by about 140% while conventional single treatment of 100% NPK fertilizer only led to an increase in potato production of 111% when compared to the control. The results demonstrated that this integrated approach has the potential to accelerate the transformation from conventional to sustainable potato production.

Abstract:The effect of wheat and maize rotations and the application of plant-animal compost on potato bacterial wilt and yield was investigated. The proposed method was tested in three separate locations, each two acres in size, with an untreated control. The first two were naturally infested, but the third was pathogen-free. Infested locations that had been treated experienced a significant reduction in disease incidence. The addition of a bio-fertilizer mixture ( Azotobacter sp., Azospirillum sp., Bacillus megaterium, and Bacillus circulans ) improved the compost's suppressive potential. The method increased soil organic matter (SOM), N, P, K, Ca +2 , and Mg +2 while decreasing Na + levels and increasing potato yield. The relative abundance of Ralstonia decreased while that of Arthrobacter , Streptomyces , and Nocardioides increased. The presence of Gracilibacillus , Cellvibrio , Bacillus , and Paenibacillus was associated with a decrease in Ralstonia , whereas the presence of Propionibacterium (a nitrogen-fixing bacteria) was associated with an increase in Ralstonia .

Abstract:Antifungal activities of plant-beneficial Bacillus have been widely studied in recent years. Numerous studies have studied the antifungal mechanisms of soluble non-volatile bioactive compounds such as lipopeptides and proteins produced by Bacillus against soil-borne diseases. However, the antagonistic mechanisms of volatile organic compounds (VOCs) from Bacillus against airborne phytopathogens are still largely unknown, and the function of Alternaria solani pathogenic genes has not been well identified. Here, we first isolated a Bacillus strain with strong antifungal activity and finally identified it as B. subtilis ZD01. Then, the antagonistic mechanisms of VOCs produced by strain ZD01, against A. solani, an airborne fungal pathogen that can cause early blight diseases of potato, were studied. We showed that VOCs produced by strain ZD01 can reduce the colony size and mycelial penetration and can cause serious morphological changes of A. solani. Scanning electron microscope (SEM) observation showed that VOCs released by ZD01 could cause more flaccid and gapped hyphae of A. solani. Also, we found that VOCs produced by ZD01 can inhibit the conidia germination and reduce the lesion areas and number of A. solani in vivo significantly. Meanwhile, based on gas chromatography/mass spectrometry (GC/MS) analysis, 29 volatile compounds produced by strain ZD01 were identified. Out of 29 identified VOCs, 9 VOCs showed complete growth inhibition activities against A. solani. Moreover, we identified two virulence-associated genes (slt2 and sod) in A. solani. slt2 is a key gene that regulates the mycelial growth, penetration, sporulation, and virulence in vivo in A. solani. In addition, sod plays a significant role in the SOD synthetic pathway in A. solani. Results from qRT-PCR showed that the transcriptional expression of these two genes was down-regulated after being treated by VOCs produced by ZD01. These results are useful for a better understanding of the biocontrol mechanism of Bacillus and offer a potential method for potato early blight disease control.
