Abstract:Objective. The development of electrode arrays able to reliably record brain electrical activity is a critical issue in brain machine interface (BMI) technology. In the present study we undertook a comprehensive physico-chemical, physiological, histological and immunohistochemical characterization of new single-walled carbon nanotubes (SWCNT)-based electrode arrays grafted onto medium-density polyethylene (MD-PE) films. Approach. The long-term electrical stability, flexibility, and biocompatibility of the SWCNT arrays were investigated in vivo in laboratory rats by two-months recording and analysis of subdural electrocorticogram (ECoG). Ex-vivo characterization of a thin flexible and single probe SWCNT/polymer electrode is also provided. Main results. The SWCNT arrays were able to capture high quality and very stable ECoG signals across 8 weeks. The histological and immunohistochemical analyses demonstrated that SWCNT arrays show promising biocompatibility properties and may be used in chronic conditions. The SWCNT-based arrays are flexible and stretchable, providing low electrode-tissue impedance, and, therefore, high compliance with the irregular topography of the cortical surface. Finally, reliable evoked synaptic local field potentials in rat brain slices were recorded using a special SWCNT-polymer-based flexible electrode. Significance. The results demonstrate that the SWCNT arrays grafted in MD-PE are suitable for manufacturing flexible devices for subdural ECoG recording and might represent promising candidates for long-term neural implants for epilepsy monitoring or neuroprosthetic BMI.

Soft and MRI Compatible Neural Electrodes from Carbon Nanotube Fibers.

Abstract:Soft and magnetic resonance imaging (MRI) compatible neural electrodes enable stable chronic electrophysiological measurements and anatomical or functional MRI studies of the entire brain without electrode interference with MRI images. These properties are important for many studies, ranging from a fundamental neurophysiological study of functional MRI signals to a chronic neuromodulatory effect investigation of therapeutic deep brain stimulation. Here we develop soft and MRI compatible neural electrodes using carbon nanotube (CNT) fibers with a diameter from 20 μm down to 5 μm. The CNT fiber electrodes demonstrate excellent interfacial electrochemical properties and greatly reduced MRI artifacts than PtIr electrodes under a 7.0 T MRI scanner. With a shuttle-assisted implantation strategy, we show that the soft CNT fiber electrodes can precisely target specific brain regions and record high-quality single-unit neural signals. Significantly, they are capable of continuously detecting and isolating single neuronal units from rats for up to 4-5 months without electrode repositioning, with greatly reduced brain inflammatory responses as compared to their stiff metal counterparts. In addition, we show that due to their high tensile strength, the CNT fiber electrodes can be retracted controllably postinsertion, which provides an effective and convenient way to do multidepth recording or potentially selecting cells with particular response properties. The chronic recording stability and MRI compatibility, together with their small size, provide the CNT fiber electrodes unique research capabilities for both basic and applied neuroscience studies.
Biocompatibility and magnetic resonance imaging characteristics of carbon nanotube yarn neural electrodes in a rat model

Abstract:Background: Implantation of deep brain stimulation (DBS) electrodes is a landmark therapy for movement disorders and some mental conditions. Compared to conventional platinum-iridium (Pt-Ir) electrodes, carbon nanotube yarns (CNTY) electrodes have improved stability and interface characteristics with less distortion during high field strength MRI. Sprague-Dawley rat models were used to examine thein vivo histological and imaging properties of biocompatible CNTY throughout the subacute period. Methods: Sprague-Dawley rats received CNTY (n = 16) or Pt-Ir control (n = 16) electrodes. Behavioral markers, body weight, and survival were recorded. Comparative histology (HE, NeuN, CD68, and GFAP) was performed at 1, 6, and 12 weeks post-implantation; 3.0T MRI was performed at 1 and 12 weeks. Results: Of 32 rats, 30 (15 per group) survived implantation without reduced activity, paralysis, or incapacity to feed. Following implantation, progressive decreases in macrophage activation and neuron-depleted margins surrounding electrodes were observed in both groups. Inflammatory marker expression (CD68) was significantly lower in rats with implanted CNTY electrodes compared to controls at all time points. CNTY electrodes also caused less inflammation and shallower depths of macrophage penetration and neural disruption relative to the interface. Artifacts and distortion were observed on MRI of Pt-Ir but not CNTY electrodes. Conclusions: CNTY electrodes exhibited reduced inflammatory margins compared to Pt-Ir electrodes throughout the subacute period, indicating reduced initial trauma, better overall biocompatibility, and reduced fibrous tissue formation. Coupled with less MRI distortion, CNTY electrodes may be useful alternatives when there is a need to monitor electrode placement by MRI.

Abstract:As a powerful tool for monitoring and modulating neural activities, implantable neural electrodes constitute the basis for a wide range of applications, including fundamental studies of brain circuits and functions, treatment of various neurological diseases, and realization of brain-machine interfaces. However, conventional neural electrodes have the issue of mechanical mismatch with soft neural tissues, which can result in tissue inflammation and gliosis, thus causing degradation of function over chronic implantation. Furthermore, implantable neural electrodes, especially depth electrodes, can only carry out limited data sampling within predefined anatomical regions, making it challenging to perform large-area brain mapping. With excellent electrical, mechanical, and chemical properties, carbonbased nanomaterials, including graphene and carbon nanotubes (CNTs), have been used as materials of implantable neural electrodes in recent years. Electrodes made from graphene and CNT fibers exhibit low electrochemical impedance, benefiting from the porous microstructure of the fibers. This enables a much smaller size of neural electrode. Together with the low Young's modulus of the fibers, this small size results in very soft electrodes. Soft neural electrodes made from graphene and CNT fibers show a much-reduced inflammatory response and enable stable chronic in vivo action potential recording for 4-5 months. Combining different modalities of neural interfacing, including electrophysiological measurement, optical imaging/stimulation, and magnetic resonance imaging (MRI), could leverage the spatial and temporal resolution advantages of different techniques, thus providing new insights into how neural circuits process information. Transparent neural electrode arrays made from graphene or CNTs enable simultaneous calcium imaging through the transparent electrodes, from which concurrent electrical recording is taken, thus providing complementary cellular information in addition to high-temporal-resolution electrical recording. Transparent neural electrodes from carbon-based nanomaterials can record well-defined neuronal response signals with negligible light-induced artifacts from cortical surfaces under optogenetic stimulation. Graphene and CNT-based materials were used to fabricate MRI-compatible neural electrodes with negligible artifacts under high field MRI. Simultaneous deep brain stimulation (DBS) and functional magnetic resonance imaging (fMRI) with graphene fiber electrodes in the subthalamic nucleus (STN) in Parkinsonian rats revealed robust blood oxygenation level dependent responses along the basal ganglia-thalamocortical network in a frequency-dependent manner, with responses from some regions not previously detectable. This review introduces the recent development and application of neural electrode technologies based on graphene and CNTs. We also discuss biological safety issues and challenges faced by neural electrodes made from carbon nanomaterials. The use of carbon-based nanomaterials for the fabrication of various soft and multi-modality compatible neural electrodes will provide a powerful platform for both fundamental and translational neuroscience research.
Stable, long-term single-neuronal recording from the rat spinal cord with flexible carbon nanotube fiber electrodes

Abstract:Objective. Flexible implantable electrodes enable months-long stable recording of single-unit signals from rat brains. Despite extensive efforts in the development of flexible probes for brain recording, thus far there are no conclusions on their application in long-term single neuronal recording from the spinal cord which is more mechanically active. To this end, we realized the chronic recording of single-unit signals from the spinal cord of freely-moving rats using flexible carbon nanotube fiber (CNTF) electrodes. Approach. We developed flexible CNTF electrodes for intraspinal recording. Continuous in vivo impedance monitoring and histology studies were conducted to explore the critical factors determining the longevity of the recording, as well as to illustrate the evolution of the electrode-tissue interface. Gait analysis were performed to evaluate the biosafety of the chronic intraspinal implantation of the CNTF electrodes. Main results. By increasing the insulation thickness of the CNTF electrodes, single-unit signals were continuously recorded from the spinal cord of freely-moving rats without electrode repositioning for 3-4 months. Single neuronal and local field potential activities in response to somatic mechanical stimulation were successfully recorded from the spinal dorsal horns. Histological data demonstrated the ability of the CNTF microelectrodes to form an improved intraspinal interfaces with greatly reduced gliosis compared to their stiff metal counterparts. Continuous impedance monitoring suggested that the longevity of the intraspinal recording with CNTF electrodes was determined by the insulation durability. Gait analysis showed that the chronic presence of the CNTF electrodes caused no noticeable locomotor deficits in rats. Significance. It was found that the chronic recording from the spinal cord faces more stringent requirements on the electrode structural durability than recording from the brain. The stable, long-term intraspinal recording provides unique capabilities for studying the physiological functions of the spinal cord relating to motor, sensation, and autonomic control in both health and disease.

Abstract:Implantable neural probes that are mechanically flexible yet robust are attractive candidates for achieving stable neural interfacing in the brain. Current flexible neural probes consist mainly of metal thin-film electrodes integrated on micrometer-thick polymer substrates, making it challenging to achieve electrode-tissue interfacing on the cellular scale. Here, we describe implantable neural probes that consist of robust carbon nanotube network embroidered graphene (CeG) films as free-standing recording microelectrodes. Our CeG film microelectrode arrays (CeG_MEAs) are ultraflexible yet mechanically robust, thus enabling cellular-scale electrode-tissue interfacing. Chronically implanted CeG_MEAs can stably track the activities of the same population of neurons over two months. Our results highlight the potential of ultraflexible and free-standing carbon nanofilms for stable neural interfacing in the brain.

Abstract:Implantable brain electrophysiology electrodes are valuable tools in both fundamental and applied neuroscience due to their ability to record neural activity with high spatiotemporal resolution from shallow and deep brain regions. Their use has been hindered, however, by the challenges in achieving chronically stable operations. Furthermore, implantable depth neural electrodes can only carry out limited data sampling within predefined anatomical regions, making it challenging to perform large-area brain mapping. Minimizing inflammatory responses and associated gliosis formation, and improving the durability and stability of the electrode insulation layers are critical to achieve long-term stable neural recording and stimulation. Combining electrophysiological measurements with simultaneous whole-brain imaging techniques, such as magnetic resonance imaging (MRI), provides a useful solution to alleviate the challenge in scalability of implantable depth electrodes. In recent years, various carbon-based materials have been used to fabricate flexible neural depth electrodes with reduced inflammatory responses and MRI-compatible electrodes, which allows structural and functional MRI mapping of the whole brain without obstructing any brain regions around the electrodes. Here, we conducted a systematic comparative evaluation on the electrochemical properties, mechanical properties, and MRI compatibility of different kinds of carbon-based fiber materials, including carbon nanotube fibers, graphene fibers, and carbon fibers. We also developed a strategy to improve the stability of the electrode insulation without sacrificing the flexibility of the implantable depth electrodes by sandwiching an inorganic barrier layer inside the polymer insulation film. These studies provide us with important insights into choosing the most suitable materials for next-generation implantable depth electrodes with unique capabilities for applications in both fundamental and translational neuroscience research.

Efficacy of Hyperopic Photorefractive Keratectomy Simultaneously Performed With Phototherapeutic Keratectomy for Decreasing Hyperopic Shift

Abstract:Purpose: To investigate the efficacy of hyperopic photorefractive keratectomy (HPRK) that was performed simultaneously with phototherapeutic keratectomy (PTK) with regard to decreasing hyperopic shift. Methods: A total of 63 eyes of 47 consecutive patients with granular corneal dystrophy (GCD) or band-shaped keratopathy (BSK) underwent PTK or PTK + HPRK. PTK alone was performed in 40 eyes (20 GCD and 20 BSK), and PTK + HPRK was performed in 23 eyes (15 GCD and 8 BSK). All patients underwent examinations including uncorrected distance visual acuity, corrected distance visual acuity, refractometry, and keratometry before and at 1, 3, and 6 months after PTK. Medical charts were reviewed retrospectively. Results: Superficial corneal opacity was successfully removed from all eyes, and uneventful reepithelialization was completed within 7 days after PTK or PTK + HPRK. The average uncorrected distance visual acuity was significantly better in the PTK + HPRK group than in the PTK group preoperatively and at 1, 3, and 6 months postoperatively. The corrected distance visual acuities were similar in both groups preoperatively and postoperatively. The hyperopic change in the PTK group was approximately 1.5 diopters, and that in the PTK + HPRK group was almost zero. There were significant differences in the average changes of the spherical equivalent from preoperation between the PTK and PTK + HPRK groups at all postoperative time points. Conclusions: Simultaneous performance of PTK and HPRK can effectively reduce hyperopic shift after PTK and is as safe as PTK alone. It is recommended that HPRK be added to PTK to treat eyes in which hyperopic shift is undesirable.

Abstract:A new form of deep brain stimulation (DBS) electrode was proposed that was made of carbon nanotube yarns (CNTYs). Electrode interface properties were examined using cyclic voltammetry (CV) and electrochemical impedance spectrum (EIS). The CNTY electrode interface exhibited large charge storage capacity (CSC) of 12.3 mC/cm(2) which increased to 98.6 mC/cm(2) after acid treatment, compared with 5.0 mC/cm(2) of Pt-Ir. Impedance spectrum of both untreated and treated CNTY electrodes showed that finite diffusion process occurred at the interface due to their porous structure and charge was delivered through capacitive mechanism. To evaluate stability electrical stimulus was exerted for up to 72 h and CV and EIS results of CNTY electrodes revealed little alteration. Therefore CNTY could make a good electrode material for DBS.
Mechanically Tissue‐Like and Highly Conductive Au Nanoparticles Embedded Elastomeric Fiber Electrodes of Brain–Machine Interfaces for Chronic In Vivo Brain Neural Recording

Abstract:A highly conductive, flexible, and biocompatible Au nanoparticles (AuNPs) embedded fiber electrode is developed with balanced electrical and mechanical properties. Compared to a conventional rigid probe, the AuNPs fiber probe can display high signal amplitude and low noise level. The fiber neural probe successfully records neural signals for four months in vivo with negligible immune responses. Implantable neural probes are a crucial part of brain–machine interfaces that serve as direct interacting routes between neural tissues and machines. The neural probes require both mechanical and electrical properties to acquire high‐quality signals from individual neurons with minimal tissue damage. However, overcoming the trade‐off between flexibility and electrical property is still challenging. Herein, a fiber neural probe, composed of core polymer and Au nanoparticles (AuNPs) on the outer shell, is fabricated by absorbing Au precursor following in situ chemical reduction with a variation of percolating and leaching time. The proposed fiber exhibits excellent electrical properties, with an electrical conductivity of 7.68 × 104 S m−1 and an impedance of 2.88 × 103 Ω at 1 kHz, as well as a Young's modulus of 170 kPa, which is comparable to that of brain tissue (≈100 kPa). Additionally, the AuNPs fiber neural probe demonstrates extremely stable in vivo electrophysiological signal recordings for four months with reduced foreign body responses at the tissue–probe interface. Furthermore, this innovative approach encourages a new paradigm of long‐term recording in the fields of neuroscience and engineering to better understand brain circuits, develop bioelectronic devices, and treat chronic disorders.

Polymer Composite with Carbon Nanofibers Aligned During Thermal Drawing As a Microelectrode for Chronic Neural Interfaces.

Abstract:Microelectrodes provide a direct pathway to investigate brain activities electrically from the external world, which has advanced our fundamental understanding of brain functions and has been utilized for rehabilitative applications as brain-machine interfaces. However, minimizing the tissue response and prolonging the functional durations of these devices remain challenging. Therefore, the development of next-generation microelectrodes as neural interfaces is actively progressing from traditional inorganic materials toward biocompatible and functional organic materials with a miniature footprint, good flexibility, and reasonable robustness. In this study, we developed a miniaturized all polymer-based neural probe with carbon nanofiber (CNF) composites as recording electrodes via the scalable thermal drawing process. We demonstrated that in situ CNF unidirectional alignment can be achieved during the thermal drawing, which contributes to a drastic improvement of electrical conductivity by 2 orders of magnitude compared to a conventional polymer electrode, while still maintaining the mechanical compliance with brain tissues. The resulting neural probe has a miniature footprint, including a recording site with a reduced size comparable to a single neuron and maintained impedance that was able to capture neural activities. Its stable functionality as a chronic implant has been demonstrated with the long-term reliable electrophysiological recording with single-spike resolution and the minimal tissue response over the extended period of implantation in wild-type mice. Technology developed here can be applied to basic chronic electrophysiological studies as well as clinical implementation for neuro-rehabilitative applications.

Abstract:Carbon nanotubes (CNTs) coatings have been shown over the past few years as a promising material for neural interface applications. In particular, in the field of nerve implants, CNTs have fundamental advantages due to their unique mechanical and electrical properties. In this study, carbon nanotubes multi-electrode arrays (CNT-modified-Au MEAs) were fabricated based on gold multi-electrode arrays (Au-MEAs). The electrochemical impedance spectra of CNT-modified-Au MEA and Au-MEA were compared employing equivalent circuit models. In comparison with Au-MEA (17 Ω), CNT-modified-Au MEA (8 Ω) lowered the overall impedance of the electrode at 1 kHz by 50%. The results showed that CNT-modified-Au MEAs have good properties such as low impedance, high stability and durability, as well as scratch resistance, which makes them appropriate for long-term application in neural interfaces.

Abstract:Abstract In this study, we report a flexible implantable 4-channel microelectrode probe coated with highly porous and robust nanocomposite of poly(3,4-ethylenedioxythiophene) (PEDOT) and carbon nanofiber (CNF) as a solid doping template for high-performance in vivo neuronal recording and stimulation. A simple yet well-controlled deposition strategy was developed via in situ electrochemical polymerization technique to create a porous network of PEDOT and CNFs on a flexible 4-channel gold microelectrode probe. Different morphological and electrochemical characterizations showed that they exhibit remarkable and superior electrochemical properties, yielding microelectrodes combining high surface area, low impedance (16.8 ± 2 MΩ.μm 2 at 1 kHz) and elevated charge injection capabilities (7.6 ± 1.3 mC/cm 2 ) that exceed those of pure and composite PEDOT layers. In addition, the PEDOT-CNF composite electrode exhibited extended biphasic charge cycle endurance, resulting in a negligible physical delamination or degradation for long periods of electrical stimulation. In vitro testing on mouse brain slices showed that they can record spontaneous oscillatory field potentials as well as single-unit action potentials and allow to safely deliver electrical stimulation for evoking field potentials. The combined superior electrical properties, durability and 3D microstructure topology of the PEDOT-CNF composite electrodes demonstrate outstanding potential for developing future neural surface interfacing applications.

Abstract:In physiological activities, the brain's electroencephalogram (EEG) signal and chemical concentration change are crucial for diagnosing and treating neurological disorders. Despite the advantages of flexible neural probes, such as their flexibility and biocompatibility, it remains a challenge to achieve in situ monitoring of electrophysiological and chemical signals on a small scale simultaneously. This study developed a new method to construct an efficient dual-sided multimodal integrated flexible neural probe, which combines a density electrode array for EEG recordings and an electrochemical sensor for detecting lactic acid. The EEG electrode array includes a 6-channel recording electrode array with each electrode 30 x 50 mu m in size, and the lactic acid sensor with overall contact is approximately 100 mu m wide. The EEG electrodes have an average impedance of 2.57 k Omega at 1 kHz and remained stable after immersing in NS (normal saline) for 3 months. The lactic acid sensor showed a sensitivity of 52.8 nA mM-1. The in vivo experiments demonstrated that the probe can reliably monitor electrophysiological signals. The probe is able to be implanted into the desired site with the help of a guide port. This flexible neural probe can provide more comprehensive insights into brain activity in the field of neuroscience and clinical practices.

Abstract:Flexible and biocompatible microwires are generally employed for biomedical implants in medicine, more specifically in neuroscience. These microwires used as electrodes need to be chemically inert and electrically conducting in order to record and stimulate the electrical activity of neurons in the nervous system. The electrodes available today are made of metals and silicon that lack the required flexibility resulting in a mechanical mismatch between soft tissues and rigid metals which induces faster biofouling and inconsistent performance for long-term implants. Biofouling triggered by inflammatory responses dramatically affects the performance of neural electrodes, resulting in decreased signal sensitivity and consistency over time. Thus, long-term clinical applications require electrically conducting flexible electrode materials with reduced dimensions and antibiofouling properties. These characteristics reduce the degree of inflammatory reactions and increase the lifetime of implanted neural electrodes. Carbon nanotubes (CNTs) are well known for their flexibility, electrical conductivity, and chemical inertness. This talk will report the fabrication of CNT fibers and subsequent coating using a biocompatible flexible polymer hydrogenated nitrile butadiene rubber (HNBR). Unlike the gold standard parylene-C coating that requires processing under vacuum, dissolved elastomer HNBR polymer can deposit uniform coating in solutions. Polymer dip-coating has been employed for uniform and continuous coating of meters-long CNT fibers of multiple diameters (26 to 66 mm) to evaluate the robustness of the approach. The polymer was able to provide uniform thickness and pinhole-free coating along the CNT fiber. Furthermore, the polymer has been shown to be biocompatible when tested with in vitro elusions and culturing neurons. The covalent functionalization of CNT microwire cross-section interfacing with the neurons using hydrophilic, anti-biofouling phosphorylcholine molecules displays reduced impedance and increased hydrophilicity. These CNT microwire assemblies are promising for the development of low-impedance neural electrodes with higher hydrophilicity and protein-fouling resistance to inhibit inflammatory responses.
Ultra-small carbon fiber electrode recording site optimization and improved in vivo chronic recording yield

Abstract:OBJECTIVE: Carbon fiber electrodes may enable better long-term brain implants, minimizing the tissue response commonly seen with silicon-based electrodes. The small diameter fiber may enable high-channel count brain-machine interfaces capable of reproducing dexterous movements. Past carbon fiber electrodes exhibited both high fidelity single unit recordings and a healthy neuronal population immediately adjacent to the recording site. However, the recording yield of our carbon fiber arrays chronically implanted in the brain typically hovered around 30%, for previously unknown reasons. In this paper we investigated fabrication process modifications aimed at increasing recording yield and longevity.APPROACH: We tested a new cutting method using a 532nm laser against traditional scissor methods for the creation of the electrode recording site. We verified the efficacy of improved recording sites with impedance measurements and in vivo array recording yield. Additionally, we tested potentially longer-lasting coating alternatives to PEDOT:pTS, including PtIr and oxygen plasma etching. New coatings were evaluated with accelerated soak testing and acute recording.MAIN RESULTS: We found that the laser created a consistent, sustainable 257 ± 13.8 µm2 electrode with low 1 kHz impedance (19 ± 4 kΩ with PEDOT:pTS) and low fiber-to-fiber variability. The PEDOT:pTS coated laser cut fibers were found to have high recording yield in acute (97% > 100 µV pp , N = 34 fibers) and chronic (84% > 100 µV pp , day 7; 71% > 100 µV pp , day 63, N = 45 fibers) settings. The laser cut recording sites were good platforms for the PtIr coating and oxygen plasma etching, slowing the increase in 1 kHz impedance compared to PEDOT:pTS in an accelerated soak test.SIGNIFICANCE: We have found that laser cut carbon fibers have a high recording yield that can be maintained for over two months in vivo and that alternative coatings perform better than PEDOT:pTS in accelerated aging tests. This work provides evidence to support carbon fiber arrays as a viable approach to high-density, clinically-feasible brain-machine interfaces.

Flexible Carbon Nanotubes Electrode for Neural Recording

Abstract:This paper demonstrates a novel flexible carbon nanotubes (CNTs) electrode array for neural recording. In this device, the CNTs electrode arrays are partially embedded into the flexible Parylene-C film using a batch microfabrication process. Through this fabrication process, the CNTs can be exposed to increase the total sensing area of an electrode. Thus, the flexible CNTs electrode of low impedance is realized. In application, the flexible CNTs electrode has been employed to record the neural signal of a crayfish nerve cord for in vitro recording. The measurements demonstrate the superior performance of the presented flexible CNTs electrode with low impedance (11.07 kohms at 1 kHz) and high peak-to-peak amplitude action potential (about 410 microV). In addition, the signal-to-noise ratio (SNR) of the presented flexible CNTs electrode is about 257, whereas the SNR of the reference (a pair of Teflon-coated silver wires) is only 79. The simultaneous recording of the flexible CNTs electrode array is also demonstrated. Moreover, the flexible CNTs electrode has been employed to successfully record the spontaneous spikes from the crayfish nerve cord. The amplitude of the spontaneous peak-to-peak response is about 25 microV.

Abstract:Though commonly used, metal electrodes are incompatible with brain tissues, often leading to injury and failure to achieve long-term implantation. Here we report a subdural neural interface of hydrogel functioning as an ionic conductor, and elastomer as a dielectric. We demonstrate that it incurs a far less glial reaction and less cerebrovascular destruction than a metal electrode. Using a cat model, the hydrogel electrode was able to record electrical signals comparably in quality to a metal electrode. The hydrogel-elastomer neural interface also readily facilitated multimodal functions. Both the hydrogel and elastomer are transparent, enabling in vivo optical microscopy. For imaging, cerebral vessels and calcium signals were imaged using two-photon microscopy. The new electrode is compatible with magnetic resonance imaging and does not cause artifact images. Such a new multimodal neural interface could represent immediate opportunity for use in broad areas of application in neuroscience research and clinical neurology.

Abstract:Implantable neural electrodes can record and regulate neural activities with high spatial resolution of single-neuron and high time resolution of sub-millisecond, which are the most extensive window in neuroscience research. However, the mechanical mismatch between conventional stiff electrodes and soft neural tissue can lead to inflammatory responses and degradation of signals in chronic recordings. Although remarkable breakthroughs have been made in sensing and regulation of neural signals, the long-term stability and chronic inflammatory response of the neural electrode-tissue interfaces still needs further development. In this review, we focus on the latest developments for the optimization of neural electrode-tissue interfaces, including electrode materials (graphene fiber-based and CNT fiber-based), electrode structures (flexible electrodes), nano-coatings and hydrogel-based neural interfaces. The parameters of impedance, charge injection limit, signal-to-noise ratio and neuron lost zone are used to evaluate the electrochemical performance of the devices, the recording performance of biosignals and the stability of the neural interfaces, respectively. These optimization methods can effectively improve the long-term stability and the chronic inflammatory response of neural interfaces during the recording and modulation of biosignals.

Abstract:Flexible implantable microelectrodes have been demonstrated to exhibit excellent biocompatibility for chronic neural activity recordings. However, the low bending strength of the commonly employed flexible materials presents a significant challenge for probe insertion into the brain. Traditional implantation methods for flexible electrodes generally require additional auxiliary materials or tools, which tend to have a much larger footprint than the probes themselves, greatly increasing the damage to neurons during insertion. Here we have proposed a stiffness-tunable polyimide probe for deep brain implantation, referred to as Neurotentacle, enabled by embedded microchannels in which the liquid pressure is controllable (from 0.1MPa to more than 2.0MPa). During the insertion phase into the brain, the neurotentacle can pose a high stiffness under elevated internal pressure to penetrate the brain tissues without the use of any additional materials or tools. Once the device has been successfully inserted, it can regain its flexibility by reducing the internal pressure. Importantly, the novel multilayer microfabrication process keeps the structural dimensions of the neurotentacle similar to those of a regular flexible probe. Therefore, the neurotentacle can produce an extremely low level of damage to brain tissue during its insertion phase, while extending its long-term biocompatibility and stability, which has been experimentally verified in histological evaluations conducted on both acute and chronic animal specimens. In addition, the chronically implanted neurotentacles enabled stable neural activity recordings in mice with an average spike yield of 96% and an average signal-to-noise ratio of 15.2. The proposed neurotentacle does not necessitate the use of complex devices and its insertion process is straightforward and highly controllable, thus rendering it an appealing technique for minimally invasive implantation and long-term neural recording of flexible electrodes.
