Abstract:Lately, a great deal of attention is being paid to capsule coating, since the coat protects active pharmaceutical ingredients (APIs) from damage, as is in the case of tablet and pellet. However, moisture and heat sensitivity of gelatin shells make it challenging to coat capsules using the conventional aqueous coating techniques. In an effort to overcome this challenge, the present study aims to coat capsules using two different coating techniques: electrostatic dry powder coating (EDPC) and dip coating (DC). Both capsule coatings and free films were prepared by these two coating techniques, and the effects of coating formulations and processing conditions on the film quality were investigated. The corresponding drug in vitro release and mechanisms were characterized and compared. The results of dissolution tests demonstrated that the drug release behavior of both EDPC and DC coated capsules could be optimized to a sustained release of 24 h, following the Fick's diffusion law. The results of this study suggest that EDPC method is better than DC method for coating capsules, with respect to the higher production efficiency and better stability, indicating that this dry coating technology has promised in gelatin capsule coating applications.

Abstract:This paper presents a solvent evaporation method with silicone oil(PDMS) as the continuous phase for preparation of microcapsules.No surfactant was used in the microcapsule preparation process.Polyacrylonitrile(PAN),cellulose acetate(CA) and chitosan(CTS) microcapsules containing Aliquat 336(ALQ),tetraethylene glycol(TEG) and bovine serum albumin(BSA) have been prepared respectively.The prepared microcapsules are orbicular with very compact surfaces,and the mean diameters are around 10～100 μm.With increasing the viscosity of PDMS or decreasing the concentration of polymer solution,the mean diameters of microcapsules decrease.The loading ratio of microcapsules can be easily controlled by adding separation agents into PDMS.The loading ratio of ALQ/PAN,TEG/CA and BSA/CTS microcapsules can reach up to 0.43,0.38 and 0.08 g/g respectively.

Abstract:To encapsulate extractants by method of microencapsulation can solve the problems of phase dispersion, phase mixing, phase separation, the leakage of extractant, as well as complicated structure of equipment in the extraction process. In the present work polysulfone and polystyrene as polymer materials were used to encapsulate the extractants such as TBP, D2EHPA, TOA, Aliquat 336 etc by means of solvent evaporation. The influence of shell materials, dispersants, stirring speed and composition of the organic phase has been investigated. The experimental results showed that TBP and D2EHPA were successfully encapsulated with. polysulfone as the shell material while for the preparation of TOA and Aliquat 336 the shell material of polystyrene should be applied. The spherical shape and monodispersity of the microcapsules were related to the dispersion agent used in the O/W system. The mean diameter of the microcapsules decreased greatly with an increase of the stirring speed. Besides the stirring speed, the viscosity of polymer solution and the interfacial tension of working systems are the major factors that influence the size of the microcapsules. An optimal encapsulation efficiency of the microcapsules was reached when the content ratio of the shell material to the extractant in the polymer solution was optimized.

Abstract:This paper presents an improved solvent evaporation method with silicone oil ( PDMS) as the continuous phase for preparation of microcapsules to make more polymer solvents available. A microchannel device was used to produce emulsions instead of mechanical stirring to prepare the mono-dispersed microcapsules. Under the conditions of lower evaporation temperature and shorter evaporation time, uniform polyacrylonitrile (PAN) microcapsules containing Aliquat 336 (ALQ) have been successfully prepared. N,N-dimethylformamide (DMF) with lower evaporation ability was applied as the polymer solvent. The prepared microcapsules have rough surfaces and homogeneously internal structures. By changing the two-phase flow rate, the mean size of microcapsules can be easily controlled. When more ALQ was added in polymer solution, the loading ratio of microcapsules increased. The mass transfer performance and stability were determined by extraction of Cr (VI) ions from its aqueous solution. The mass transfer rate was fast enough. After three times of repeated extraction and stripping, the microcapsules kept almost the same extraction ability, which indicated that the microcapsules have very good stability.

Abstract:Objective: Casticin (Vitexicarpin) has shown immunoregulatory, antitumor, cytotoxicity, anti-inflammatory and analgesic properties. Application of the valuable bioactive compounds can be limited by their unpleasant taste, low bioavailability, volatilization of active compounds, sensitivity to the temperature, oxidation and UV light, as well as in vivo instability. The problem can be solved by coating the Casticin with a microencapsulation technique. The purpose of this research was to formulate the microcapsules of Casticin with solvent evaporation technique using Ethocel 10 cP. Methods: The microencapsulation process of Casticin was done by solvent evaporation technique (O/W: oil in water). The formula of Casticin microcapsules were designed into three formulas (Ethocel 10 cP: 10%, 15% and 20%). Microcapsules of Casticin were characterized for particle size, in terms of surface morphology by scanning electron microscope (SEM), encapsulation efficiency and release test. Results: In this research, the micoparticles containing Casticin has been developed by using ethyl cellulose (Ethocel 10 cP) as the polymer matrix. The results showed that high concentration of polymer (Ethocel 10 cP) used in microencapsulation resulted in better Casticin microcapsules in terms of physical characteristics. Particle size of microcapsules containing Casticin were in the range of 42.51 to 61.47 μm. Encapsulation efficiency (% EE) was categorized as good because the value were ≥ 80% to, which 91.57% to 96.24%. SEM picture of Casticin microcapsules revealed that the surface of microcapsule were a smooth surface and no pores of microcapsule were obtained. When Eudragit E100 used as a polymer, a rough and porous surface of microcapsule were obtained. Conclusion: It can be concluded that microcapsules of Casticin can be prepared by solvent evaporation method with a single emulsion system (O/W) using Ethocel 10 cP as polymer. Characterization of the microcapsules revealed that ethyl cellulose used on this method is applicable to produce microcapsules which stable in physical properties. A higher polymer concentration led to a more viscous solution, which delayed the polymer precipitation and resulted in a less porous polymer matrix with a slower drug release.

Abstract:The use of biodegradable polymers simplifies the development of therapeutic devices with regards to transient implants and three-dimensional platform suitable for tissue engineering. Further advances have also occurred in the controlled released mechanism of bioactive compounds encapsulated in biodegradable polymers. This application requires the understanding of the physicochemical properties of the polymeric materials and their inherent impact on the delivery of encapsulated bioactive. Hence, the objective of this study was to evaluate the effect of surfactant and sonication time on the bio-accessibility of lycopene encapsulated polymeric nanoparticles. The emulsion evaporation method was used to encapsulate lycopene in poly-lactic co-glycolic acid (PLGA) with surfactant concentration, sonication time and polymer concentration as independent variables. Physicochemical and morphological characteristics were measured with a zetasizer and SEM, while the encapsulation efficiency and controlled release kinetics with spectrophotometric, and the dialysis method was used to estimate bioaccessibility. The results have shown sonication time to have significantly (p < 0.05) influenced the encapsulation efficiency. Hence, the sonication time of 4 min yield an encapsulation efficiency of 78% and increased to 97% with increase sonication time (6 min). Increased sonication time had a decreasing effect on the hydrodynamic diameter and stability of the encapsulated nanoparticles. The slow release of lycopene was observed during the first 12 days, followed by a burst release of about 44% on the 13th day in-vitro. The study will have significant impact on the manufacturing of functional food with encapsulated ingredients and provide an understanding of their inherent control release mechanism in the GIT.
Effect of lipid and cellulose based matrix former on the release of highly soluble drug from extruded/spheronized, sintered and compacted pellets

Abstract:Background: The study was to develop an extended release (ER) encapsulated and compacted pellets of Atenolol using hydrophobic (wax based and polymeric based) and high viscosity grade hydrophilic matrix formers to control the release of this highly water soluble drug by extrusion/spheronization (ES). Atenolol is used for cardiovascular diseases and available as an immediate release (IR) tablet dosage form. The lipids, Carnauba wax (CW), Glyceryl monostearate (GMS) and cellulose based i.e. Hydroxypropyl methylcellulose (HPMC) and Ethyl cellulose (EC) were used in preparing Atenolol ER pellets. Thermal sintering and compaction techniques were also applied to control the burst release of Atenolol. Method: For this purpose, thirty-six trial formulations (F1-F36) were designed by Response Surface Methodology (RSM), using Design-Expert 10 software, keeping (HPMC K4M, K15 M & K100 M), (EC 7FP, 10FP & 100FP), waxes (GMS, & CW), their combinations, sintering temperature and duration, as input variables. Dissolution studies were performed in pH, 1.2, 4.5 and 6.8 dissolution media. Drug release kinetics using different models such as zero order, first order, Korsmeyer-Peppas, Hixon Crowell, Baker-Lonsdale and Higuchi kinetics were studied with the help of DDsolver, an excel based add-in program. Results: The formulations F35 and F36 showed compliance with Korsmeyer-Peppas Super case II transport model (R2 = 0.975-0.971) in dissolution medium pH 4.5. No drug excipient interaction observed by FTIR. Stereomicroscopy showed that sintered combination pellets, (F35), were highly spherical (AR = 1.061, and sphericity = 0.943). The cross-sectional SEM magnification (at 7000X) of F34 and F35 showed dense cross-linking. The results revealed that the optimized formulations were F35 (sintered pellets) and F36 (compacted pellets) effectively controlling the drug release for 12 h. Conclusion: Extended-release encapsulated, and compacted pellets were successfully prepared after the combination of lipids CW (10%) and GMS (20%) with EC (10FP 20% & 100FP 20%). Sintering and compaction, in addition, stabilized the system and controlled the initial burst release of the drug. Extended release (ER) Atenolol is an effective alternative of IR tablets in controlling hypertension and treating other cardiovascular diseases.

Abstract:Various microencapsulation techniques can result in significant differences in the properties of dried microcapsules. Microencapsulation is an effective approach to improve fish oil properties, including oxidisability and unpleasant flavour. In this study, β-carotene, lutein, zeaxanthin, and fish oil were co-encapsulated by microfluidic-jet spray drying (MFJSD), two-fluid nozzle spray drying (SD), and freeze-drying (FD), respectively. The aim of the current study is to understand the effect of different drying techniques on microcapsule properties. Whey protein isolate (WPI) and octenylsuccinic anhydride (OSA) modified starch were used as wall matrices in this study for encapsulating carotenoids and fish oil due to their strong emulsifying properties. Results showed the MFJSD microcapsules presented uniform particle size and regular morphological characteristics, while the SD and FD microcapsules presented a large distribution of particle size and irregular morphological characteristics. Compared to the SD and FD microcapsules, the MFJSD microcapsules possessed higher microencapsulation efficiency (94.0-95.1%), higher tapped density (0.373-0.652 g/cm3), and higher flowability (the Carr index of 16.0-30.0%). After a 4-week storage, the SD microcapsules showed the lower retention of carotenoids, as well as ω-3 LC-PUFAs than the FD and MFJSD microcapsules. After in vitro digestion trial, the differences in the digestion behaviours of the microcapsules mainly resulted from the different wall materials, but independent of drying methods. This study has provided an alternative way of delivering visual-beneficial compounds via a novel drying method, which is fundamentally essential in both areas of microencapsulation application and functional food development.

Abstract:In this study, two polyelectrolytes, chitosan and propyl methyl cellulose carbonate, were associated by crosslinking with tripolyphosphate (TPP), to prepare microspheres loaded with amlodipine besylate (Aml). The formulations were optimized by Box–Behnken experimental design, in which three factors, four responses were studied to check the effect of independent variables (concentration of TPP, pH of TPP solution, and pH of a polymer mixture) on responses: zeta potential, particle size, polydispersity index and entrapment efficiency. The optimal encapsulation efficiency was around 92.43%, the zeta potential on average 41.75 mV, and the minimum mean particle size was 310 nm. The effects of the parameters on changes in structure, crystallinity, and surface charges of polymers, by crosslinking in microparticles, were identified by FT-IR, XRD, and dynamic light scattering analyses. The encapsulation parameters and the environment media influenced the mechanism of release and the kinetics model, which was greater in the duodenal medium at pH 5.5 ( Q = 100%). The microencapsulation by nontoxic polymers in water is a good method of preparing amlodipine-loaded microspheres, which are intended for the oral route, with optimal dose release in the duodenal medium. This represents the target site for absorption of the drug, in the pulmonary sphere.

Abstract:Polymer microparticles used for drug encapsulation and delivery have various surface morphologies depending on the type of formulation ingredients and parameters of the manufacture process. This works aims at investigating the critical parameters governing the morphology of microparticles and to underline the influence of their surface state on the drug release. The classical fabrication process by the "emulsion-solvent evaporation" is addressed using poly(ɛ-caprolactone) as the polymer and methylene chloride as the volatile organic solvent. The typical surfactants poly(vinyl alcohol) and polysorbate 80 have been considered. Scanning electron microscopy observations showed the various surface morphologies mainly depending on the stirring rate, the viscosity of the oil phase and by the presence of inappropriate surfactants. Because of arrested coalescence during solvent evaporation, the evaporation of the organic solvent causing particles hardening is the most important parameter that controls the morphology. Indeed, slow evaporation allows partial coalescence of the soft particles swollen by the organic solvent, whereas the particles morphology is frozen rapidly upon fast evaporation, thus preventing damaged surface states. Moreover, an effective stabilizing system for the primary emulsion is also a determining factor to control the final morphology. The morphology of the particles has a definite influence on the drug delivery of cholecalciferol. The surface morphology should be taken into consideration in the design of polymer microparticles because it allows a control over the drug release kinetics.
Encapsulation of cinnamon leaf oil within chitosan: formulation and characterization

Abstract:Chitosan microcapsules containing cinnamon leaf oil were formed by ionotropic gelation of chitosan by crosslinking with sodium tripolyphosphate. The microencapsulation method for oil loading was optimized by varying the amounts of polymer, oil and cross linker. The effect of varying process parameters on the encapsulation efficience (EE), oil loading capacity (LC) and release rate was monitored. The formulated microcapsules were characterized using Scanning Electron Microscopy (SEM) and Fourier Transform Infrared Spectroscopy (FTIR). SEM imaged clear spherical shaped oil loaded microcapsules with a crimpy surface while empty capsules had a smooth texture.FTIR spectra evidence for the successful encapsulation of oil within wall material. EE increased with increasing oil load while it decreased with increasing polymer and cross linker concentration. LC increased with increasing amounts of oil and crosslinker and showed a decrease with increasing polymer amounts. Release rate of oil increased with increasing oil loading whereas it decreased with increasing polymer and crosslinker amounts. Varying the process parameters has a direct impact on the EE, LC and release rate. The optimum formulation for cinnamon oil microcapsules is polymer, 1 g : oil, 3g: cross linker, 0.5 g with an EE of 91%, and a LC of 38%.

Abstract:BACKGROUND: Enteric microencapsulation of the potential immunosuppressant TRAM-34 was investigated as a means of enhancing oral drug delivery and minimizing or eliminating hydrolysis of pyrazole-substituted triarylmethane to the respective alcohol.METHOD: TRAM-34 was successfully enteric microencapsulated by a coacervation method using the pH-sensitive Eudragit L 100 polymer. In this study, we utilized water-miscible organic solvents such as acetone and ethanol, which are considered safe class 3 solvents according to the ICH guideline. We deemed such an approach suitable for safe scale up and for enteric coating application to other compounds of a similar lipophilicity.RESULTS: The resulting microparticles were spherical and uniform with an average particle size of 460 microm at 15% theoretical loading. The encapsulation efficiency was 90 +/- 1.9% and the percentage yield was found to be 91.5 +/- 0.3%. The oral administration in rhesus macaques of TRAM-34-loaded enteric-coated microparticles illustrated six times enhancement in its oral bioavailability. However, the TRAM-34 plasma concentration was less than the therapeutic effective level.CONCLUSION: The low oral bioavailability, even after enteric coating, could be attributed to the compound's inherent absorption characteristics and high lipophilicity.

Abstract:Starch sodium octenyl succinate (SSOS) and maltodextrin (MD) are commonly used as wall materials to protect bioactive compounds, such as essential oils volatiles. In this work, Rimulus Cinamon (RC) and Angelica Sinenis (AS) essential oils (EOs) were encapsulated in (SSOS/MD) composite capsules via spray drying. The microcapsules were characterized by Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM) and thermogravimetric (TG). The particle size, Zeta potential, drug loading (DL), encapsulation efficiency (EE), stability and in vitro release of the EOs microcapsules were investigated. The average particle size of microcapsules and emulsion was 12.6 ± 1.09 μm and 0.22 ± 0.01 μm, respectively. The DL% and EE% of microcapsules were 16.3 ± 0.40% and 96.6 ± 1.17%. No phase separation was found in the emulsion within 7 d, and the creaming index was 0%, showing good stability. After 6 months of storage, the retention rate of cinnamaldehyde in the microcapsules was about 67.6%, which significantly improved the stability of EOs. Abstract:The objective of the current study was to develop and optimize a novel lyophilized liposomal formulation of anticancer agent carmustine, or bis-chloroethyl nitrosourea (BCNU) for prolonged release that could overcome the dose-dependent side effects and improve its bioavailability at the site of action. The optimization was done using a 32 factorial design approach wherein soya phosphatidylcholine (SPC) and cholesterol (CH) as independent variables. The optimized formulation (F4) exhibited high entrapment efficiency (81.57%) with an average vesicle size of 141.7 nm and a −22.6 mV Zeta potential. In-vitro drug release studies from all formulations revealed that the BCNU was released for up to 36 hours following the Higuchi matrix release model. The TEM, FTIR, DSC, PXRD, and SEM analyses confirm the formation of liposomes. BCNU-loaded nanoliposomal formulation demonstrated prolonged release, suggesting that it could be used to supplement cancer therapy efficiently with a reduction in dose-dependent side effects.

Abstract:Curcumin (CUR) has impressive pharmacologic properties, including cardioprotective, neuroprotective, antimicrobial, and anticancer activity. However, the pharmaceutical application of CUR is limited due to its poor aqueous solubility and low bioavailability. The development of novel formulations has attracted considerable attention to the idea of applying nanobiotechnology to improve the therapeutic efficacy of these challenging compounds. In this study, CUR-loaded lecithin–chitosan nanoparticles (CUR/LCSNPs) were developed and optimized by the concentration of chitosan, lecithin, and stirring speed by a 3-factorial Box-Behnken statistical design, resulting in an optimal concentration of chitosan (A) and lecithin (B) with a 1200 rpm stirring speed (C), with applied constraints of minimal average particle size (Y1), optimal zeta potential (Y2), and maximum entrapment efficiency (%EE) (Y3). The mean particle size of the checkpoint formulation ranged from 136.44 ± 1.74 nm to 267.94 ± 3.72, with a zeta potential of 18.5 ± 1.39 mV to 36.8 ± 3.24 mV and %EE of 69.84 ± 1.51% to 78.50 ± 2.11%. The mean particle size, zeta potential, %EE, and % cumulative drug release from the optimized formulation were 138.43 ± 2.09 nm, +18.98 ± 0.72 mV, 77.39 ± 1.70%, and 86.18 ± 1.5%, respectively. In vitro drug release followed the Korsmeyer–Peppas model with Fickian diffusion (n < 0.45). The optimized technique has proven successful, resulting in a nanoformulation that can be used for the high loading and controlled release of lipophilic drugs.

Abstract:OBJECTIVE To investigate how health plans manage chronic diseases. STUDY DESIGN Health plan medical directors were surveyed regarding the disease management (DM) practices of their plans. METHODS We took a stratified random sample of 65 plans, all members of the American Association of Health Plans. Forty-five plans responded. Results were weighted to be representative of the industry (including nonmember plans). Medical directors were asked to consider that they had a DM program only if 2 things were true: (1) A majority of a plan's enrollees could not be ineligible for a DM program for non-clinical reasons (eg, geographic location); and (2) a DM program had to have at least 6 of the 8 components of a DM program as defined by the Disease Management Association of America. RESULTS The 3 diseases most likely to be the focus of DM programs were diabetes, asthma, and congestive heart failure. For each of these diseases, at least one quarter of Americans were enrolled in plans offering a DM program. Medical directors perceived their DM programs to be highly effective in reducing mortality and morbidity and in improving the functional status of patients, and perceived them to be effective in lowering cost. The greatest challenge in implementing DM programs involves information technology. These results yield insights into the future of treatment of chronic disease in the United States. CONCLUSION Health plans have made a significant investment in programs to improve care for chronic illness. The almost universality of DM programs highlight the need for scholarly evaluations of their effectiveness and cost effectiveness.
Microencapsulation of caffeic acid and its release using a w/o/w double emulsion method: Assessment of formulation parameters

Abstract:Caffeic acid (CAF)has numerous health benefits mainly due to its antioxidant, antibacterial and fungicide properties. However, its incorporation in skin care products as anti-aging and the photoprotective agent is still limited due to its solubility and stability in oily matrices or solutions balanced with the skin pH. In this research, CAF–ethyl cellulose (EC) microparticles were produced by water-in-oil-water double emulsion solvent evaporation encapsulation technique using a biocompatible polymer, EC, as a coating material and a surfactant, polyvinyl alcohol, as a stabilizer of the double emulsion. The study assessed the influence of formulation parameters as the solubility of the polymer in organic solvents and the polymer concentration on microparticles final characteristics. CAF–EC microparticles were characterized by product yield, encapsulation efficiency, mean particle size, particle size distribution and polydispersity and imaged by scanning light microscopy. In vitro release profiles were obtained in water and octanol to mimic oily based and water-based matrices balanced with the skin pH. In vitro release kinetics studies were carried out to investigate the release pattern of CAF in simulated cosmetic formulations. Both the product yield and the encapsulation efficiency were found to be dependent on the solubility of the polymer in the organic phase. The product yield was mainly affected by operational factors such as the sticking and the agglomeration of the polymer to the walls and the magnet stirring during microparticles hardening and results from the encapsulation efficiency revealed that an increase of the polymer concentration led to an increase of the encapsulation efficiency. The usage of a water-soluble solvent contributed to a decrease in the mean particle size and reduction of polydispersity with higher polymer concentrations. The polymer concentration, the polymer solubility in the organic phase and the amount of CAF entrapped shown to affect the release in water, whereas the release in octanol was mainly independent of the amount of CAF entrapped in EC microparticles. The double emulsion solvent evaporation technique and the assessment of the selected formulation conditions have given significant and innovative insights on the microencapsulation of bioactive ingredients for cosmetics formulations.

Abstract:Many active ingredients in food products are destroyed by gastric acids and enzymes as they pass through the stomach, thereby reducing the desired effect of the target ingredient. In addition, traditional microcapsule preparation methods usually break up the dispersion by mechanical stirring or oscillation, which generates droplets of uneven size and inconsistent inclusion content, and mechanical external forces may cause denaturation and inactivation of active substances. This study aimed to use microfluidics to generate Oil-in-water-in-oil (O/W/O) droplet structures with good monodispersity and highly tunable properties, and to further prepare microcapsules with high encapsulation rate and targeted intestinal release by taking advantage of the cross-linking properties of alginate that reacts with Ca 2+ under acidic conditions. Finally, the particle size distribution, release effect, encapsulation effect and oxidative stability of the core-shell microcapsules were evaluated. In this study, two types of microcapsules, homogeneous and core-shell microcapsules, were prepared by selecting tea polyphenols and vitamin E as models for hydrophilic and lipophilic bioactive compounds, respectively. Thin shell microcapsules with a shell thickness of about 25 μm were prepared using PDMS chip conditioning. Large microcapsules with a particle size of 230 μm were generated by using capillary glass tube chips, which had an excellent antioxidant property with a retention rate of 88.66 ± 1.25% of VE after 20 days of storage at 4 °C, while all of the VE was oxidized in the bare environment. In addition, in simulated gastric fluid, the release of VE was only about 20%, while in artificial intestinal fluid, the release was close to 100%, so the microcapsules also had intestinal targeting properties.

Abstract:Liquid-filled hard gelatin capsules may have pertinent advantages both for therapeutic effect and extemporaneous preparations of medicines. Alpha lipoic acid is a substance used in medicines and dietary supplements and there is a need for creating an appropriate formulation which would be suitable for each individual patient or consumer. Based on its biopharmaceutical and physical chemical characteristics, eight different capsule formulations were designed and characterized. Silicon dioxide was added to form a semisolid content and prevent leakage. The formulation filled with alpha lipoic acid solution in polyethylene glycol 400 showed the best performance. Although the addition of silicon dioxide to the formulation with polyethylene glycol 400 led to a change in both flow character and viscosity, the release rate did not show a statistically significant decrease (more than 85% of content was released after 5 min testing). Applied technique is a simple and an appropriate approach for compounding and could be used for other substances with similar properties.
