Abstract:Soil salinity is a common environmental stress factor that limits agricultural production worldwide. Plants have evolved different strategies to achieve salt tolerance. miR393 has been identified as closely related to biotic and abiotic stresses, and targets F-box genes that encode auxin receptors. The miR393-TIR1/AFB2/AFB3 regulatory module was discovered to have multiple functions that manipulate the auxin response. This study focused on miR393 and one of its targets, TIR1, and found that they played potential roles in response to salt stress. Our results showed that overexpression of a miR393-resistant TIR1 gene (mTIR1) in Arabidopsis clearly enhanced salt stress tolerance, which led to a higher germination rate, less water loss, reduced inhibition of root elongation, delayed senescence, decreased death rate and stabilized Chl content. These plants accumulated more proline and anthocyanin, and displayed enhanced osmotic stress tolerance. The expression of some salt stress-related genes was altered, and sodium content can be reduced in these plants under salt stress. We proposed that highly increased auxin signaling by overexpression of mTIR1 may trigger auxin-mediated downstream pathways to enhance plant salt stress resistance by osmoregulation and increased Na+ exclusion.
Overexpression of AtHsp90.2, AtHsp90.5 and AtHsp90.7 in Arabidopsis Thaliana Enhances Plant Sensitivity to Salt and Drought Stresses

Abstract:Three AtHsp90 isoforms, cytosolic AtHsp90.2, chloroplast-located AtHsp90.5, and endoplasmic reticulum (ER)-located AtHsp90.7, were characterized by constitutive overexpressing their genes in Arabidopsis thaliana . Both types of the transgenic plants overexpressing cytosolic and organellar AtHsp90s showed reduced tolerance to salt and drought stresses with lower germination rates and fresh weights, but improved tolerance to high concentration of Ca 2+ comparing with the wild type plants. Transcriptional analysis of ABA-responsive genes, RD29A , RD22 and KIN2 under salt and drought stresses, indicated that the induction expression of these genes was delayed by constitutive overexpression of cytosolic AtHsp90.2, but was hardly affected by that of organellar AtHsp90.5 and AtHsp90.7. These results implied that Arabidopsis different cellular compartments-located Hsp90s in Arabidopsis might be involved in abiotic stresses by different functional mechanisms, probably through ABA-dependent or Ca 2+ pathways, and proper homeostasis of Hsp90 was critical for cellular stress response and/or tolerance in plants.
TaSAUR78 enhances multiple abiotic stress tolerance by regulating the interacting gene TaVDAC1

Abstract:SMALL AUXIN-UP RNAs (SAURs) regulated by abiotic stress play multiple functions in plants. However, the functions of SAURs in abiotic stress are largely unknown. In this study, we cloned a novel SAUR gene, TaSAUR78, from wheat, and we found that TaSAUR78 interacted with TaVDAC1 (voltage-dependent anion channel). Salt stress decreased expression of TaSAUR78 and increased expression of TaVDAC1. Overexpression of TaSAUR78 enhanced tolerance to salt, drought, and freezing stresses in transgenic Arabidopsis and reduced the accumulation of reactive oxygen species (ROS) under salt stress. Overexpression of TaVDAC1 enhanced tolerance to salt stress, while decreased tolerance to drought and low temperature stresses in transgenic Arabidopsis. TaVDAC1 overexpression increased the accumulation of ROS in plants. These results suggested that TaSAUR78 improved plant tolerance to abiotic stresses by regulating TaVDAC1. This study generated valuable information on the functions of TaSAUR78 and TaVDAC1 in multiple abiotic stresses, which may facilitate the deployment of these genes to enhance crop tolerance to abiotic stresses in the future.
A Member of the 14-3-3 Gene Family in Brachypodium Distachyon, BdGF14d, Confers Salt Tolerance in Transgenic Tobacco Plants

Abstract:Plant 14-3-3 proteins are involved in diverse biological processes, but for the model monocotyledonous species, Brachypodium distachyon, their roles in abiotic stress tolerance are not well understood. In this study, a total of eight Bd14-3-3 genes were identified from B. distachyon and these were designated respectively as BdGF14a-BdGF14g. The qRT-PCR analyses of 3-month-old plants of B. distachyon showed that these genes were all expressed in the stems, leaves, and spikelets. By contrast, most of the plants had relatively lower transcriptional levels in their roots, except for the BdGF14g gene. The different expression profiles of the Bd14-3-3s under various stress treatments, and the diverse interaction patterns between Bd14-3-3s and BdAREB/ABFs, suggested that these gene products probably had a range of functions in the stress responses. The NaCl-induced Bd14-3-3 gene, BdGF14d, was selected for overexpression in tobacco. BdGF14d was found to be localized throughout the cell and it conferred enhanced tolerance to salt in the transgenic plants. Lowered contents of malondialdehyde, H2O2, and Na+, and lower relative electronic conductance (Rec%), yet greater activities of catalase and peroxidase, were observed in the overexpressing plants. Higher photosynthetic rate, transpiration rate, stomatal conductance, and water use efficiency were measured in the transgenic lines. Following abscisic acid (ABA) or NaCl treatment, stomatal aperture in leaves of the BdGF14d-overexpression plants was significantly lower than in leaves of the wild type (WT) controls. The stress-related marker genes involved in the ABA signaling pathway, the reactive oxygen species (ROS)-scavenging system, and the ion transporters were all up-regulated in the BdGF14d-overexpressing plants as compared with WT. Taken together, these results demonstrate that the Bd14-3-3 genes play important roles in abiotic stress tolerance. The ABA signaling pathway, the ROS-scavenging system, and ion transporters were all involved in enhancing the tolerance to salt stress in the BdGF14d-overexpression plants.

Abstract:Aquaporin (AQP) proteins have been shown to transport water and other small molecules through biological membranes, which is crucial for plants to combat salt stress. However, the precise role of AQP genes in salt stress response is not completely understood in plants. In this study, a PIP1 subgroup AQP gene, designated TaAQP8, was cloned and characterized from wheat. Transient expression of TaAQP8-green fluorescent protein (GFP) fusion protein revealed its localization in the plasma membrane. TaAQP8 exhibited water channel activity in Xenopus laevis oocytes. TaAQP8 transcript was induced by NaCl, ethylene and H2O2. Further investigation showed that up-regulation of TaAQP8 under salt stress involves ethylene and H2O2 signaling, with ethylene causing a positive effect and H2O2 acting as a negative factor. Overexpression of TaAQP8 in tobacco increased root elongation compared with controls under salt stress. The roots of transgenic plants also retained a high K+/Na+ ratio and Ca2+ content, but reduced H2O2 accumulation by an enhancement of superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) activities under salt stress. Further investigation showed that whole seedlings from transgenic lines displayed higher SOD, CAT and POD activities, increased NtSOD and NtCAT transcript levels, and decreased H2O2 accumulation and membrane injury under salt stress. Taken together, our results demonstrate that TaAQP8 confers salt stress tolerance not only by retaining high a K+/Na+ ratio and Ca2+ content, but also by reducing H2O2 accumulation and membrane damage by enhancing the antioxidant system.
GhUBC10‐2 mediates GhGSTU17 degradation to regulate salt tolerance in cotton (Gossypium hirsutum)

Abstract:Ubiquitin‐conjugating enzyme (UBC) is a crucial component of the ubiquitin‐proteasome system, which contributes to plant growth and development. While some UBCs have been identified as potential regulators of abiotic stress responses, the underlying mechanisms of this regulation remain poorly understood. Here, we report a cotton (Gossypium hirsutum) UBC gene, GhUBC10‐2, which negatively regulates the salt stress response. We found that the gain of function of GhUBC10‐2 in both Arabidopsis (Arabidopsis thaliana) and cotton leads to reduced salinity tolerance. Additionally, GhUBC10‐2 interacts with glutathione S‐transferase (GST) U17 (GhGSTU17), forming a heterodimeric complex that promotes GhGSTU17 degradation. Intriguingly, GhUBC10‐2 can be self‐polyubiquitinated, suggesting that it possesses E3‐independent activity. Our findings provide new insights into the PTM of plant GST‐mediated salt response pathways. Furthermore, we found that the WRKY transcription factor GhWRKY13 binds to the GhUBC10‐2 promoter and suppresses its expression under salt conditions. Collectively, our study unveils a regulatory module encompassing GhWRKY13–GhUBC10‐2–GhGSTU17, which orchestrates the modulation of reactive oxygen species homeostasis to enhance salt tolerance.

Stress-Induced Expression of an Activated Form of Atbzip17 Provides Protection from Salt Stress in Arabidopsis

Abstract:Membrane-associated basic-leucine zipper (bZIP) transcription factors that reside in the endoplasmic reticulum represent a newly described class of plant stress sensor/transducers. The bZIP factors are anchored in endoplasmic reticulum (ER) membranes with their C-terminal tails facing the ER lumen and their N terminii, which contain transcriptional components, facing the cytosol. In response to stress, cytosolic components of the transcription factors are released by proteolysis and move to the nucleus where they promote the up-regulation of stress response genes. One such stress sensor/transducer in Arabidopsis is AtbZIP17, which is activated in response to salt stress. With the aim of enhancing salt tolerance, a constitutively activated form of AtbZIP17, truncated before its membrane-anchor domain, was introduced into transgenic plants. When placed under the control of a constitutive promoter, the activated form of AtbZIP17 up-regulated stress response genes under unstressed conditions, but caused a substantial delay in plant development. When the activated form of AtbZIP17 was placed under the control of stress-inducible promoter, development was normal under unstressed conditions. Under salt stress conditions, the stress-inducible expression of the activated AtbZIP17 enhanced salt tolerance as demonstrated by chlorophyll bleaching and seedling survival assays.
Overexpression of a Cytosolic Abiotic Stress Responsive Universal Stress Protein (SbUSP) Mitigates Salt and Osmotic Stress in Transgenic Tobacco Plants

Abstract:The universal stress protein (USP) is a ubiquitous protein and plays an indispensable role in plant abiotic stress tolerance. The genome of Salicornia brachiata contains two homologs of intron less SbUSP gene which encodes for salt and osmotic responsive USP. In vivo localization reveals that SbUSP is a membrane bound cytosolic protein. The role of the gene was functionally validated by developing transgenic tobacco and compared with control [wild-type (WT) and vector control (VC)] plants under different abiotic stress condition. Transgenic lines (T1) exhibited higher chlorophyll, relative water, proline, total sugar, reducing sugar, free amino acids, polyphenol contents, osmotic potential, membrane stability, and lower electrolyte leakage and lipid peroxidation (malondialdehyde content) under stress treatments than control (WT and VC) plants. Lower accumulation of H2O2 and [Formula: see text] radicals was also detected in transgenic lines compared to control plants under stress conditions. Present study confers that overexpression of the SbUSP gene enhances plant growth, alleviates ROS buildup, maintains ion homeostasis and improves the physiological status of the plant under salt and osmotic stresses. Principal component analysis exhibited a statistical distinction of plant response to salinity stress, and a significant response was observed for transgenic lines under stress, which provides stress endurance to the plant. A possible signaling role is proposed that some downstream genes may get activated by abiotic stress responsive cytosolic SbUSP, which leads to the protection of cell from oxidative damages. The study unveils that ectopic expression of the gene mitigates salt or osmotic stress by scavenging ROS and modulating the physiological process of the plant.

Abstract:Group A protein phosphatases 2Cs (PP2Cs) are essential components of abscisic acid (ABA) signaling in Arabidopsis; however, the function of group F2 subfamily PP2Cs is currently less known. In this study, TaPP2C1 which belongs to group F2 was isolated and characterized from wheat. Expression of the TaPP2C1-GFP fusion protein suggested its ubiquitous localization within a cell. TaPP2C1 expression was downregulated by abscisic acid (ABA) and NaCl treatments, but upregulated by H2O2 treatment. Overexpression of TaPP2C1 in tobacco resulted in reduced ABA sensitivity and increased salt resistance of transgenic seedlings. Additionally, physiological analyses showed that improved resistance to salt stress conferred by TaPP2C1 is due to the reduced reactive oxygen species (ROS) accumulation, the improved antioxidant system, and the increased transcription of genes in the ABA-independent pathway. Finally, transgenic tobacco showed increased resistance to oxidative stress by maintaining a more effective antioxidant system. Taken together, these results demonstrated that TaPP2C1 negatively regulates ABA signaling, but positively regulates salt resistance. TaPP2C1 confers salt resistance through activating the antioxidant system and ABA-independent gene transcription process.
UBIQUITIN-SPECIFIC PROTEASE16 modulates salt tolerance in arabidopsis by regulating Na+/H+ antiport activity and serine hydroxymethyltransferase stability

Abstract:Protein ubiquitination is a reversible process catalyzed by ubiquitin ligases and ubiquitin-specific proteases (UBPs). We report the identification and characterization of UBP16 in Arabidopsis thaliana. UBP16 is a functional ubiquitin-specific protease and its enzyme activity is required for salt tolerance. Plants lacking UBP16 were hypersensitive to salt stress and accumulated more sodium and less potassium. UBP16 positively regulated plasma membrane Na+/H+ antiport activity. Through yeast two-hybrid screening, we identified a putative target of UBP16, SERINE HYDROXYMETHYLTRANSFERASE1 (SHM1), which has previously been reported to be involved in photorespiration and salt tolerance in Arabidopsis. We found that SHM1 is degraded in a 26S proteasome-dependent process, and UBP16 stabilizes SHM1 by removing the conjugated ubiquitin. Ser hydroxymethyltransferase activity is lower in the ubp16 mutant than in the wild type but higher than in the shm1 mutant. During salt stress, UBP16 and SHM1 function in preventing cell death and reducing reactive oxygen species accumulation, activities that are correlated with increasing Na+/H+ antiport activity. Overexpression of SHM1 in the ubp16 mutant partially rescues its salt-sensitive phenotype. Taken together, our results suggest that UBP16 is involved in salt tolerance in Arabidopsis by modulating sodium transport activity and repressing cell death at least partially through modulating SMH1stability and activity.
Transcriptional regulation and functional analysis of Nicotiana tabacum under salt and ABA stress

Abstract:Soil salinization is an important factor that restricts crop quality and yield and causes an enormous toll to human beings. Salt stress and abscisic acid (ABA) stress will occur in the process of soil salinization. In this study, transcriptome sequencing of tobacco leaves under salt and ABA stress in order to further study the resistance mechanism of tobacco. Compared with controlled groups, 1654 and 3306 DEGs were obtained in salt and ABA stress, respectively. The genes function enrichment analysis showed that the up-regulated genes in salt stress were mainly concentrated in transcription factor WRKY family and PAR1 resistance gene family, while the up-regulated genes were mainly concentrated on bHLH transcription factor, Kunitz-type protease inhibitor, dehydrin (Xero1) gene and CAT (Catalase) family protein genes in ABA stress. Tobacco MAPK cascade triggered stress response through up-regulation of gene expression in signal transduction. The expression products of these up-regulated genes can improve the abiotic stress resistance of plants. These results have an important implication for further understanding the mechanism of salinity tolerance in plants.

Abstract:BACKGROUND:NAC (NAM, ATAF, and CUC) transcription factors play important roles in plant biological processes, including phytohormone homeostasis, plant development, and in responses to various environmental stresses.METHODS:TaNAC29 was introduced into Arabidopsis using the Agrobacterium tumefaciens-mediated floral dipping method. TaNAC29-overexpression plants were subjected to salt and drought stresses for examining gene functions. To investigate tolerant mechanisms involved in the salt and drought responses, expression of related marker genes analyses were conducted, and related physiological indices were also measured. Expressions of genes were analyzed by quantitative real-time polymerase chain reaction (qRT-PCR).RESULTS:A novel NAC transcription factor gene, designated TaNAC29, was isolated from bread wheat (Triticum aestivum). Sequence alignment suggested that TaNAC29 might be located on chromosome 2BS. TaNAC29 was localized to the nucleus in wheat protoplasts, and proved to have transcriptional activation activities in yeast. TaNAC29 was expressed at a higher level in the leaves, and expression levels were much higher in senescent leaves, indicating that TaNAC29 might be involved in the senescence process. TaNAC29 transcripts were increased following treatments with salt, PEG6000, H2O2, and abscisic acid (ABA). To examine TaNAC29 function, transgenic Arabidopsis plants overexpressing TaNAC29 were generated. Germination and root length assays of transgenic plants demonstrated that TaNAC29 overexpression plants had enhanced tolerances to high salinity and dehydration, and exhibited an ABA-hypersensitive response. When grown in the greenhouse, TaNAC29-overexpression plants showed the same tolerance response to salt and drought stresses at both the vegetative and reproductive period, and had delayed bolting and flowering in the reproductive period. Moreover, TaNAC29 overexpression plants accumulated lesser malondialdehyde (MDA), H2O2, while had higher superoxide dismutase (SOD) and catalase (CAT) activities under high salinity and/or dehydration stress.CONCLUSIONS:Our results demonstrate that TaNAC29 plays important roles in the senescence process and response to salt and drought stresses. ABA signal pathway and antioxidant enzyme systems are involved in TaNAC29-mediated stress tolerance mechanisms.

Abstract:Main conclusion TaGF14b enhances tolerance to multiple stresses through ABA signaling pathway by altering physiological and biochemical processes, including ROS-scavenging system, stomatal closure, compatible osmolytes, and stress-related gene expressions in tobaccos. The 14-3-3 proteins are involved in plant growth, development, and in responding to abiotic stresses. However, the precise functions of 14-3-3s in responding to drought and salt stresses remained unclear, especially in wheat. In this study, a 14 - 3 - 3 gene from wheat, designated TaGF14b , was cloned and characterized. TaGF14b was upregulated by polyethylene glycol 6000, sodium chloride, hydrogen peroxide, and abscisic acid (ABA) treatments. Ectopic expression of TaGF14b in tobacco conferred enhanced tolerance to drought and salt stresses. Transgenic tobaccos had longer root, better growth status, and higher relative water content, survival rate, photosynthetic rate, and water use efficiency than control plants under drought and salt stresses. The contribution of TaGF14b to drought and salt tolerance relies on the regulations of ABA biosynthesis and ABA signaling, as well as stomatal closure and stress-related gene expressions. Moreover, TaGF14b expression could significantly enhance the reactive oxygen species (ROS) scavenging system to ameliorate oxidative damage to cells. In addition, TaGF14b increased tolerance to osmotic stress evoked by drought and salinity through modifying water conservation and compatible osmolytes in plants. In conclusion, TaGF14b enhances tolerance to multiple abiotic stresses through the ABA signaling pathway in transgenic tobaccos by altering physiological and biochemical processes.

Abstract:Plants have developed robust signaling mechanisms to cope with environmental stresses. In a recent article in the Plant Journal,1 we reported on a salt stress signaling pathway in Arabidopsis, which is mechanically similar to endoplasmic reticulum (ER) stress responses described in mammalian systems. We found that a type II membrane-bound transcription factor (AtbZIP17) is a salt stress signal transducer and a subtilisin-like serine protease (AtS1P) is a key component of the protein processing machinery in the signaling pathway. AtbZIP17 is normally located in the ER, but in response to salt stress, the membrane-bound transcription factor is proteolytically processed in an AtS1P-dependent step, and the cytosolic component of AtbZIP17 is translocated to the nucleus. We have demonstrated in an in vitro assay that AtS1P directly cleaves AtbZIP17, an initial step in processing. Our study also identified several downstream target genes in the salt signaling pathway. In this addendum, we propose a testable model to describe the action of AtS1P and AtbZIP17 in salt stress signaling and we also present an outlook for future work to test this model.

Abstract:The objectives of this work were to determine the changes in the expression of neuroendocrine markers in Leydig cell by oestradiol treatment, and to determine whether testosterone is able to recover partially the effects of hormonal suppression induced by oestradiol. Adult male rats were injected daily with either 50 microg of oestradiol or oestradiol plus testosterone propionate (25 mg every 3 days) for 15 days. The animals were sacrificed and testicles were dissected and processed by routine histological protocols. FSH and LH serum levels were determined by radioimmunoassay. The visualization of antigens was achieved by the streptavidin-peroxidase immunohistochemical method. Antibodies against chromogranin A (CrA), S-100 protein (S-100), P substance (PS), synaptofisin (SYN), neurofilament protein (NF), gliofibrillary acidic protein (GFAP) and neuron specific enolase (NSE) were used. The mean LH and FSH serum concentrations were consistently suppressed with hormonal treatments. Intermediate filaments (NF and GFAP) showed no difference in their expression. The expression of S-100, NSE and SYN was significantly lower in both hormone-treated groups. In oestradiol-treated rats, the immunoreactivity of CrA and SP decreased significantly but was restored after testosterone supplementation. Although the nature and functions of many of these substances in Leydig cells remain unknown, these results are consistent with the hypothesis that the expression of some neuroendocrine markers is hormonally controlled.
Quantitative Proteomics Analysis Reveals That the Nuclear Cap-Binding Complex Proteins Arabidopsis CBP20 and CBP80 Modulate the Salt Stress Response.

Abstract:The cap-binding proteins CBP20 and CBP80 have well-established roles in RNA metabolism and plant growth and development. Although these proteins are thought to be involved in the plant's response to environmental stress, their functions in this process are unclear. Here we demonstrated that Arabidopsis cbp20 and cbp80 null mutants had abnormal leaves and flowers and exhibited increased sensitivity to salt stress. The aberrant phenotypes were more pronounced in the cbp20/80 double mutant. Quantification by iTRAQ (isobaric tags for relative and absolute quantification) identified 77 differentially expressed proteins in the cbp20 and cbp80 lines compared with the wild-type Col-0 under salt stress conditions. Most of these differentially expressed proteins were synergistically expressed in cbp20 and cbp80, suggesting that CBP20 and CBP80 have synergistic roles during the salt stress response. Biochemical analysis demonstrated that CBP20 and CBP80 physically interacted with each other. Further analysis revealed that CBP20/80 regulated the splicing of genes involved in proline and sugar metabolism and that the epigenetic and post-translational modifications of these genes were involved in salt stress tolerance. Our data suggest a link between CBP20/80-dependent protein ubiquitination/sumoylation and the salt stress response.
Over-Expression of AtEXPB2 Alleviates Salt Stress Damage in Transgenic Tobacco Plants

Abstract:Expansin protein is a component of the cell wall that is generally accepted as a key regulator of the cell wall extension during plant growth and stress response. Expansins loosen the cell wall in a pH-dependent manner. To investigate the role of AtEXPB2 , an Arabidopsis expansin gene, its mRNA expression level was analyzed in response to salt stress in overexpressed tobacco ( Nicotiana tabacum L). This over-expression of AtEXPB2 in transgenic lines was studied under salt stress in order to evaluate their resistance to salt stress conditions. In the findings of the current study, however, transgenic plants' cellular characteristics were probably altered because of the changes in some morphological traits like capsule seed weight, capsule weight, and root length; this alteration did not have any unfavorable effects on transgenic plant phenotypes. Transgenic lines were better than wild-type plant in terms of some morphological parameters, such as capsule number, capsule weight, total seed weight, and capsule seed number. In other words, seed yield was higher in transgenic lines than those in the control. Overexpression of AtEXPB2 conferred tolerance to salt stress by enhancing the activity of antioxidant enzymes, proline content, RWC, and chlorophyll, as well as reducing electrolyte leakage index (ELI). In addition, relative expression of AtEXPB2 and H + -ATPase gene increased in transgenic plants under salinity stress. Overall, these results confirmed the role of expansin proteins in plant stress tolerance.

Abstract:Understanding pulmonary pathophysiology has increasing implications for imaging modalities. Although it was sufficient to perform high-resolution computed tomography in the past, the impetus now is on providing quantitative and functional lung data. Magnetic resonance imaging, which was traditionally difficult to perform in the lungs, has developed into a promising technology. One of the main areas of interest is the use of hyperpolarized noble gases, such as 3-He and 129-Xe, which enable high-definition lung imaging that includes information on lung microstructure. It is possible to obtain three-dimensional information on essential pulmonary processes, such as ventilation, oxygen uptake, and spirometry, which offers new insight into lung pathophysiology. This article focuses on the novel aspects of hyperpolarized 3-He magnetic resonance imaging.

Abstract:Salt stress is a major abiotic stress affecting plant growth and development. Identification and characterization of genes, especially for transcription factors (TFs), can provide us a comprehensive understanding of the regulatory mechanisms of transcriptional pathways in response to salt stress. Here, we first showed that nuclear-localized transcription factor nuclear factor-Y subunit C1 (NF-YC1) was required for the salt tolerance response in Arabidopsis. Loss of function of NF-YC1 resulted in a salt-sensitive phenotype together with increased sodium (Na) content in roots and shoots, while the overexpression line could complement this phenotype. Subsequent transcriptome analysis revealed differentially expressed genes (DEGs) in WT plants and nf-yc1 mutant, independent of salt treatment. Further analysis identified two positive downstream target genes AOX3 and BGLU47, involved in the cyanide-resistant alternative respiration and lignin biosynthesis, respectively, and a negative downstream target gene, MLS, which is related to malate synthase, as being involved in these regulatory cascades controlling the salt tolerance, and these results were further confirmed by CHIP-qPCR in vivo. In conclusion, several novel pathways have been elucidated, demonstrating that NF-YC1-dependent transcriptional regulation in salt-treated Arabidopsis thaliana.

Abstract:Heavy metals are common pollutants of the coastal saline area and Salicornia brachiata an extreme halophyte is frequently exposed to various abiotic stresses including heavy metals. The SbMT-2 gene was cloned and transformed to tobacco for the functional validation. Transgenic tobacco lines (L2, L4, L6 and L13) showed significantly enhanced salt (NaCl), osmotic (PEG) and metals (Zn++, Cu++ and Cd++) tolerance compared to WT plants. Transgenic lines did not show any morphological variation and had enhanced growth parameters viz. shoot length, root length, fresh weight and dry weight. High seed germination percentage, chlorophyll content, relative water content, electrolytic leakage and membrane stability index confirmed that transgenic lines performed better under salt (NaCl), osmotic (PEG) and metals (Zn++, Cu++ and Cd++) stress conditions compared to WT plants. Proline, H2O2 and lipid peroxidation (MDA) analyses suggested the role of SbMT-2 in cellular homeostasis and H2O2 detoxification. Furthermore in vivo localization of H2O2 and O2-; and elevated expression of key antioxidant enzyme encoding genes, SOD, POD and APX evident the possible role of SbMT-2 in ROS scavenging/detoxification mechanism. Transgenic lines showed accumulation of Cu++ and Cd++ in root while Zn++ in stem under stress condition. Under control (unstressed) condition, Zn++ was accumulated more in root but accumulation of Zn++ in stem under stress condition suggested that SbMT-2 may involve in the selective translocation of Zn++ from root to stem. This observation was further supported by the up-regulation of zinc transporter encoding genes NtZIP1 and NtHMA-A under metal ion stress condition. The study suggested that SbMT-2 modulates ROS scavenging and is a potential candidate to be used for phytoremediation and imparting stress tolerance.
