Abstract:Detection of senescence-associated β-galactosidase (SA-β-gal) activity at an increased lysosomal pH is one of the most reliable approaches for probing senescent cells. Herein, we developed a novel near-infrared fluorescent probe (KSL0401) for sensing SA-β-gal activity and applying in visualization of aging. This probe was designed by connecting a cleavable D-galactose residue to the coumarin-dicyanoisophorone hybrid fluorophore KSL0401-OH via a β-glycosidic bond. The spectral properties of KSL0401 reaction with β-gal were measured in aqueous solution at pH 6.0, and a remarkable enhancement of fluorescence emission at 706 nm was observed, along with a visible colorimetric change. Probe KSL0401 was highly selective and sensitive to SA-β-gal, and displayed large Stokes shift (218 nm) and good biocompatibility. Moreover, it exhibited extremely rapid response to β-gal within 80 s, which surpassed most of the reported SA-β-gal probes. Additionally, KSL0401 also enabled to detect the endogenous SA-β-gal activity in living cells, and distinguish between senescent cells and non-senescent cells and qualify the degree of aging. These findings demonstrate that KSL0401 is a potential detection tool in the imaging research of aging.

Abstract:Real-time monitoring of cell apoptosis could provide valuable insights into early detection of therapy efficiency and evaluation of disease progression. In this work, we designed and synthesized a new live-cell-permeable, fluorescent light-up probe for real-time cell apoptosis imaging. The probe is comprised of a hydrophilic caspase-specific Asp-Glu-Val-Asp (DEVD) peptide and a hydrophobic tetraphenylethene (TPE) unit, a typical fluorogen with aggregation-induced emission characteristics. In aqueous solution, the probe is almost nonfluorescent but displays significant fluorescence enhancement in response to caspase-3/-7, which are activated in the apoptotic process and able to cleave the DEVD moieties. This fluorescence "turn-on" response is ascribed to aggregation of cleaved hydrophobic TPE residues, which restricts the intramolecular rotations of TPE phenyl rings and populates the radiative decay channels. The light-up nature of the probe allows real-time monitoring of caspase-3/-7 activities both in solutions and in living cells with a high signal-to-noise ratio. The probe provides a new opportunity to screen enzyme inhibitors and evaluate the apoptosis-associated drug efficacy.

Abstract:Senescence-associated diseases have severely diminished the quality of life and health of patients. However, a sensitive assay of these diseases remains limited due to a lack of straightforward methods. Considering that senescence-associated beta-galactosidase (SA-beta-Gal) is overexpressed in senescent cells, the detection of SA-beta-Gal in senescent cells and tissues might be a feasible strategy for the early diagnosis of SA diseases. In this study, a beta-galactosidase-activatable nanoprobe BOD-L-beta Gal-NPs was developed for the imaging of senescent cells and vasculature in atherosclerotic mice via real-time monitoring of beta-Gal. BOD-L-beta Gal-NPs was fabricated by encapsulating a newly designed NIR ratiometric probe BOD-L-beta Gal within a poly(lactic-co-glycolic) acid (PLGA) core. Nanoprobe BOD-L-beta Gal-NPs showed good accumulation in arteries, thus successfully visualizing senescent cells and vasculature in atherosclerotic mice by tail vein injection. Our findings indicated that nanoprobe BOD-L-beta Gal-NPs holds great potential for the early diagnosis and therapy of atherosclerosis and other aging-associated diseases.

Abstract:SIRT1 is an important protein that catalyzes the nicotinamide adenine dinucleotide (NAD)(+)-dependent deacetylation reaction, which is regarded as a novel target to treat metabolic disorders and aging-related diseases. However, there is lack of appropriate approach for SIRT1 modulator screening and bioimaging of SIRT1 in living cells. We designed, and synthesized a "turn-on" fluorescent probe by connecting a specifically recognized peptide to tetraphenylethene core. It exhibits excellent selectivity and sensitivity in homogeneous measurement of SIRT1 activity for screening both SIRT1 inhibitors and activators, 20(S)-ginsenoside Rg(3) and ophiopogonin D' were found to activate SIRT1. It was also successfully; applied to monitor SIRT1 modulation in the cardiomyocytes as well as in the wild,type and SIRT1(-/-) mesenchymal stem cells.

Abstract:Senescence is a driver of aging and various age‐related diseases. However, tracking and visualizing of senescence in vivo is very challenging. We have rationally developed a near‐infrared (NIR) fluorescent probe (XZ1208) that can be cleaved rapidly by ß‐galactosidase (ß‐Gal) and produces a strong fluorescence signal. XZ1208 achieved high specificity, sensitivity and a long‐term duration in labeling senescence without causing significant toxicities in multiple aging and senescence‐associated disease models. Senescent cells (SnCs) are implicated in aging and various age‐related pathologies. Targeting SnCs can treat age‐related diseases and extend health span. However, precisely tracking and visualizing of SnCs is still challenging, especially in in vivo environments. Here, we developed a near‐infrared (NIR) fluorescent probe (XZ1208) that targets β‐galactosidase (β‐Gal), a well‐accepted biomarker for cellular senescence. XZ1208 can be cleaved rapidly by β‐Gal and produces a strong fluorescence signal in SnCs. We demonstrated the high specificity and sensitivity of XZ1208 in labeling SnCs in naturally aged, total body irradiated (TBI), and progeroid mouse models. XZ1208 achieved a long‐term duration of over 6 days in labeling senescence without causing significant toxicities and accurately detected the senolytic effects of ABT263 on eliminating SnCs. Furthermore, XZ1208 was applied to monitor SnCs accumulated in fibrotic diseases and skin wound healing models. Overall, we developed a tissue‐infiltrating NIR probe and demonstrated its excellent performance in labeling SnCs in aging and senescence‐associated disease models, indicating great potential for application in aging studies and diagnosis of senescence‐associated diseases.

Abstract:Abstract Accumulation of senescent cells with age leads to tissue dysfunction and related diseases. Their detection in vivo still constitutes a challenge in aging research. We describe the generation of a fluorogenic probe (sulfonic-Cy7Gal) based on a galactose derivative, to serve as substrate for β-galactosidase, conjugated to a Cy7 fluorophore modified with sulfonic groups to enhance its ability to diffuse. When administered to male or female mice, β-galactosidase cleaves the O-glycosidic bond, releasing the fluorophore that is ultimately excreted by the kidneys and can be measured in urine. The intensity of the recovered fluorophore reliably reflects an experimentally controlled load of cellular senescence and correlates with age-associated anxiety during aging and senolytic treatment. Interestingly, our findings with the probe indicate that the effects of senolysis are temporary if the treatment is discontinued. Our strategy may serve as a basis for developing fluorogenic platforms designed for easy longitudinal monitoring of enzymatic activities in biofluids.

Abstract:Cellular senescence, a process that arrests the cell cycle, is a cellular response mechanism for various stresses and is implicated in aging and various age-related diseases. However, the understanding of senescence in living organisms is insufficient, largely due to the scarcity of sensitive tools for the detection of cellular senescence in vivo. Herein, we describe the development of a self-immobilizing near-infrared (NIR) fluorogenic probe that can be activated by senescence-associated β-galactosidase (SA-β-Gal), the most widely used senescence marker. The NIR signal is turned on only in the presence of SA-β-Gal, and the fluorescence signal is retained to the site of activation via in situ labeling, significantly enhancing the sensitivity of the probe. We demonstrate its efficient noninvasive imaging of senescence in mice xenograft models.

Abstract:Cellular senescence is involved in diverse physiological processes. Accumulation of senescent cells can lead to numerous age-related diseases. Therefore, it is of great significance to develop chemical tools to effectively detect and eliminate senescent cells. Till date, a dual functional probe that could detect and eliminate senescent cells has yet been accomplished. Herein, a β-gal-activated probe, MB-βgal, based on the methylene blue (MB) fluorophore, was designed to detect and eliminate senescent cells. In the absence of β-gal, the probe showed no fluorescence and its 1O2 production efficiency was suppressed simultaneously. On the other hand, MB-βgal could be specifically activated by the high level of β-gal in senescent cells, thus, releasing free MB with near-infrared (NIR) fluorescence and high 1O2 production efficiency under light irradiation. MB-βgal demonstrated a fast response, high sensitivity, and high selectivity in detecting β-gal in an aqueous solution and was further applied to visualization and ablation of senescent cells. Abstract:Currently, the process of problem analysis in product design involves multiple partners in a distributed environment. A tutoring approach to support knowledge modelling will be presented in this article, focusing on variables as a key component of knowledge. A set of Tutor Agents intends to aid distributed partners to coherently elicitate their know-how in order to use to analyse design problems. A granular Knowledge Base is proposed to standardise distributed knowledge, providing Product Life-Cycle (PLC) constraints (based on product knowledge) to construct optimisation models. Furthermore, a case will be presented in order to clarify the approach. The results will enable design teams to integrate constraints from PLC knowledge in order to optimise the evolution of design concepts.

Abstract:Senescence is happening in every corner of the living organisms. β-galactosidase (β-gal) is one of the most important biomarkers during senescence in both plant and mammalian cells. Most β-gal fluorescent probes were focused on bio-imaging, only a few probes were developed for the detection of β-gal in fruit, and the probes that could detect β-gal in both fruits and living cells were even less. Here, two β-gal probes (TNap-βGal and TBNap-βGal) were synthesized, which can not only image the increase of β-gal during both fruits softening and cellular senescence, but also prove that bananas are not suitable for storage in refrigerator and the subsequent accumulation of β-gal still in lysosome of mammalian cells. In addition, TNap-βGal was successfully applied to two-photon imaging of endogenous β-gal in both hDPMSCs and tissues of human dental pulp for the first time.

Abstract:Cell senescence is one of the hallmarks of aging known to negatively influence a healthy lifespan. Drugs able to kill senescent cells specifically in cell culture, termed senolytics, can reduce the senescent cell burden in vivo and extend healthspan. Multiple classes of senolytics have been identified to date including HSP90 inhibitors, Bcl-2 family inhibitors, piperlongumine, a FOXO4 inhibitory peptide and the combination of Dasatinib/Quercetin. Detection of SA-β-Gal at an increased lysosomal pH is one of the best characterized markers for the detection of senescent cells. Live cell measurements of senescence-associated β-galactosidase (SA-β-Gal) activity using the fluorescent substrate C12FDG in combination with the determination of the total cell number using a DNA intercalating Hoechst dye opens the possibility to screen for senotherapeutic drugs that either reduce overall SA-β-Gal activity by killing of senescent cells (senolytics) or by suppressing SA-β-Gal and other phenotypes of senescent cells (senomorphics). Use of a high content fluorescent image acquisition and analysis platform allows for the rapid, high throughput screening of drug libraries for effects on SA-β-Gal, cell morphology and cell number.

Abstract:Colorectal cancer was one of the major malignant tumors threatening human health and β-Gal was recognized as a principal biomarker for primary colorectal cancer. Thus, designing specific and efficient quantitative detection methods for measuring β-Gal enzyme activity was of great clinical test significance. Herein, an ultrasensitive detection method based on Turn-on fluorescence probe (CS-βGal) was reported for visualizing the detection of exogenous and endogenous β-galactosidase enzyme activity. The test method possessed a series of excellent performances, such as a significant fluorescence enhancement (about 11.3-fold), high selectivity as well as superior sensitivity. Furthermore, under the optimal experimental conditions, a relatively low limit of detection down to 0.024 U/mL was achieved for fluorescence titration experiment. It was thanks to the better biocompatibility and low cytotoxicity, CS-βGal had been triumphantly employed to visual detect endogenous and exogenous β-Gal concentration variations in living cells with noteworthy anti-interference performance. More biologically significant was the fact that the application of CS-βGal in BALB/c nude mice was also achieved successfully for monitoring endogenous β-Gal enzyme activity.

Abstract:Overexpression of β-galactosidase (β-gal) in tumor cells may serve as a valuable biomarker for the early diagnosis of some cancers (such as ovarian cancer). In addition, abnormal accumulation of β-gal is also considered an essential marker of cell senescence. Therefore, it is important to construct fluorescent probes with excellent fluorescence properties to visualize β-gal in biological systems. Here, we designed and screened a novel fluorescent probe XM for the detection of β-gal. Spectral data show that the probe has a good affinity (Km = 2.6 μM) for β-gal, large stokes shift (190 nm), fast response speed (stable within 20 min), and low detection limit (6.7 × 10−3 U/mL). Based on the above advantages, XM can not only detect β-gal content in cancer cells but also track the changes of β-gal content in zebrafish at different developmental period. We believe XM will become a powerful tool for the early cancer diagnosis and cellular senescence.

Abstract:β-galactosidase (β-gal) which is responsible for the hydrolysis of glycosidic bonds of lactose to galactose has been recognized as an important biomarker of cell or organism status especially cell senescence and primary ovarian cancer. Therefore, extensive efforts have been devoted to the visualization of β-gal. Herein, a fluorescence probe gal-HCA to monitor β-gal in living cells was prepared by attaching a D-galactose group to 2-hydroxy-4'-dimethylamino-chalcone (HCA), which intrinsically possessed both excited-state intramolecular proton transfer (ESIPT) and aggregation-induced emission (AIE) properties. The cleavage of gal-HCA triggered by β-gal induced the formation of HCA concomitantly with a significant bathochromic-shift in fluorescence from 532nm to 615nm. The probe exhibited high selectivity and sensitivity toward β-gal with a detection limit as low as 0.0137 U/mL. The confocal imaging results demonstrated the successful applications of gal-HCA to monitor the endocellular overexpressed β-gal in senescent cells and ovarian cancer cells. Therefore, we inferred that gal-HCA could play an illuminating role in the detection of β-gal associated disease and the design of related probes.

Abstract:In vivo measurement of abnormal β-galactosidase (β-gal) in living cells and tissues is urgently required for early diagnosis and treatment of primary ovarian cancer. Herein, a FRET-based ratiometric two-photon fluorescent probe FTR-βgal with a large emission shift, rapid response and low cytotoxicity was developed for sensitively detecting β-gal activity in vivo. FTR-βgal was designed with a naphthalene derivative as the energy donor and a rhodol derivative as the energy acceptor, which provided an efficient FRET strategy. Remarkably, FTR-βgal was successfully applied in TP imaging of β-gal with the ratio signal rapidly transform from 0.15 to 5.9 (∼40-fold) between 540 nm and 450 nm. Furthermore, FTR-βgal was used to visualize β-gal levels in tumor upon tissue penetration of 30–150 μm achieved. Therefore, FTR-βgal was applicable for clinical detection in living cells and tissues as a promising strategy for diagnosis of various associated cancers.

Abstract:The heterogeneous and complex phenotypes of cellular senescence necessitate a biomarker combination for the accurate detection of senescent cells from others. However, this raises the challenge of detecting multiple senescent biomarkers in the same live cell simultaneously. Herein we reported the strategy of biomarker combination triggered tandem activation for designing senescence-specific fluorogenic probes, which resulted in the development of the probe PGal-FA. The fluorescence of PGal-FA can only be activated by the sequential stimulation by the senescent biomarker combination of β-galactosidase (βGal) and formaldehyde (FA), with βGal activating the sensing ability of the probe toward FA. Facilitated by probe PGal-FA, the simultaneous detection of a biomarker combination in the same live cell was realized. We have demonstrated the improved selectivity of probe PGal-FA toward senescent cells compared to the traditional single-biomarker-based probe. Probe PGal-FA was also successfully used to detect senescent cells in bleomycin-induced pulmonary fibrosis tissues. We expect probe PGal-FA to be a reliable tool for the study on cellular senescence and envision that this probe design strategy may be expanded to other biological events to improve accuracy.

Abstract:A tailor‐made photosensitizer based on boron dipyrromethene conjugated with a galactose moiety through a self‐immolative linker has been designed and synthesized. It exhibits a selective response toward the senescence‐associated β‐galactosidase, rendering it an efficient theranostic behavior for detection and photodynamic elimination of senescent cells in vitro and in vivo. Senescence is a cellular response having physiological and reparative functions to preserve tissue homeostasis and suppress tumor growth. However, the accumulation of senescent cells would cause deleterious effects that lead to age‐related dysfunctions and cancer progression. Hence, selective detection and elimination of senescent cells are crucial yet remain a challenge. A β‐galactosidase (β‐gal)‐activated boron dipyrromethene (BODIPY)‐based photosensitizer (compound 1) is reported here that can selectively detect and eradicate senescent cells. It contains a galactose moiety connected to a pyridinium BODIPY via a self‐immolative nitrophenylene linker, of which the photoactivity is effectively quenched. Upon interactions with the senescence‐associated β‐gal, it undergoes enzymatic hydrolysis followed by self‐immolation, leading to the release of an activated BODIPY moiety by which the fluorescence emission and singlet oxygen generation are restored. The ability of 1 to detect and eliminate senescent cells is demonstrated in vitro and in vivo, using SK‐Mel‐103 tumor‐bearing mice treated with senescence‐inducing therapy. The results demonstrate that 1 can be selectively activated in senescent cells to trigger a robust senolytic effect upon irradiation. This study breaks new ground in the design and application of new senolytic agents based on photodynamic therapy.

An enzyme-activated two-photon ratiometric fluorescent probe with lysosome targetability for imaging β-glucuronidase in colon cancer cells and tissue

Abstract:Colon cancer is a malignant neoplasm with high mortality that has seriously threatened human life. Accumulating evidence reveals that the β-glucuronidase (GLU), a lysosomal exoglycosidase enzyme, plays important roles in the pathological progression of colon cancer. Unfortunately, understanding the pathological roles of GLU remains a challenge due to the lack of effective detection methods for visualization the fluctuations of GLU in tissues. In this paper, based on hydrolysis function of GLU, an enzyme-activated ratiometric two-photon (TP) fluorescent probe (RN-GLU) was designed. RN-GLU was synthesized by introducing a glucopyranuronic acid methyl ester as the recognition group and 1, 8-naphthalimide as a TP fluorophore. In the presence of GLU, the trigger group was removed made an ICT process occurred induced enhancement of fluorescence ratio (I553 nm/I441 nm, 214-fold). Probe RN-GLU displayed low detection limit (1.2 × 10-2 μg/L) and rapid detection to GLU in vitro through a ratiometric response mode. Meanwhile, RN-GLU exhibited high selectivity for GLU and showed nearly no response to other relevant biological species. The imaging results demonstrated that RN-GLU could be applied for ratiometric monitoring of endogenous GLU levels in HCT116 cells with good lysosome targetable ability. Due to its two-photon excitation, RN-GLU could monitor GLU in colon cancer tumor tissue with good penetration ability (imaging depth of 200 μm). RN-GLU could be developed as a potential method for evaluating and confirming the functions of GLU in colon cancer diagnosis and complex biosystem.
