Abstract:Cadmium (Cd) has become one of the major contaminants in aquatic ecosystems. The microalgae Dunaliella salina is known as a cadmium (Cd) accumulator and shows a high level of tolerance to Cd, but improved Cd tolerance and relevant mechanisms are still required for investigation. In this study, two mutant strains (M1 and M2) of D. salina with high Cd resistance were obtained by chemical mutagenesis. In the normal medium, there were no differences between the two mutant strains and the wild-type strain in biochemical and physiological indicators. However, the two mutant strains showed high Cd resistance, rapid growth rate, and good Cd accumulation capacity in the medium containing high levels of Cd compared with the wild strain, which may be involved in increases in protein content, accumulation of carotenoids and chlorophylls, and glutathione reductase (GR) activity. We identified 829, 587, and 740 differentially expressed genes (DEGs) from the Cd/Control, Cd + M1/Cd, and Cd + M2/Cd sample groups by transcriptome analysis, respectively. The main DEGs were found in the ribosome, DNA replication, photosynthesis-antenna, and photosynthesis pathways after KEGG enrichment. There were 39 DEGs involved in the photosynthesis and photosynthesis-antenna pathways, of which 28 DEGs were down-regulated by Cd in the wild-type strain but remained relatively unchanged in the mutant strains. Under Cd exposure, the mutant strains showed higher mRNA levels of most DEGs involved in the ribosome and DNA replication pathways than the wild strain. Taken together, the present study will help us understand the mechanism of Cd tolerance in microalgae, and provide candidate genes and key pathways for phytoremediation.

Abstract:The investigation of heavy metal wastewater treatment utilizing microalgae adsorption has been extensively demonstrated. However, the response mechanism based on metabolomics to analyze the time-series changes of microalgae under Cd stress has not been described in detail. In this study, SEM/TEM demonstrated that Cd accumulated on the cell surface of microalgae and was bioconcentrated in the cytoplasm, vesicles, and chloroplasts. Carbonyl/quinone/ketone/carboxyl groups (O C O), membrane polysaccharides ( OH), and phospholipids ( PO) were involved in the interaction of Cd ions, and the chlorophyll content underwent a process of decreasing in the early stage (1.62 mg/g at 48 h) and recovering to the normal level in the late stage, and the contents of MDA, GSH, and SOD were all increased (29.7 nmol/g, 0.23 mg/g, and 30.01 u/10 6 cells) and then gradually returned to the steady state. The results of EPS content and fluorescent labeling showed that Cd induced the overexpression and synthesis of extracellular polysaccharides and proteins, which is one of the defense mechanisms participating in the reduction of cellular damage by complexed Cd. Metabolomics results indicated that the malate synthesis pathway was activated after Cd-20 h, and the microalgal cells began to shift the metabolic pathway to storage lipid or polysaccharide biosynthesis. In the Calvin cycle, the expression of D-Sedoheptulose 7-phosphate in Cd-20 h_vs_ck and Cd-72 h_vs_Cd-20 h firstly declined and then increased, and the photosynthesis system was suppressed at the beginning, and then gradually returned to normal to maintain the successful development of the dark reaction. The results of time series analysis revealed that the response of microalgae to Cd was categorized into fast response and slow response to regulate cell adsorption and growth metabolism.

Abstract:Anthropogenic activities have significantly increased the influx of carbon dioxide and metals into the marine environment. Combining ocean acidification (OA) and metal pollution may lead to unforeseen biological and ecological consequences. Several studies have shown that OA reduces cadmium (Cd) toxicity in marine diatoms. Although these studies have shed light on the physiological and transcriptomic responses of diatoms exposed to Cd, many aspects of the mechanisms underlying the reduced metal accumulation in diatoms remain unknown. This study aims to address this unresolved question by comparing Cd subcellular distribution, antioxidant enzyme activity, relative expression of metal transporters, surface potential, surface composition, and transmembrane potential in the diatom Phaeodactylum tricornutum grown under ambient and 1200 μatm p CO 2 conditions. Our findings reveal that diatoms grown in acidified seawater exhibit higher surface potential and higher plasma membrane depolarization. These changes and the competing effects of increased H + concentration result in a blunted response of P. tricornutum to the Cd challenge. Consequently, this study offers a new explanation for mitigating Cd toxicity by marine diatoms adapted to OA. Environmental Implication Several studies have shown that ocean acidification (OA) reduces metal accumulation or mitigates Cd toxicity in marine diatoms. However, the underlying mechanisms behind these phenomena remain unknown. Our study found that diatoms grown in acidified seawater exhibited higher surface potentials and plasma membrane depolarization. These physiological changes coupled with the competing effect of H + on Cd uptake explain the attenuation of Cd toxicity in marine diatoms under OA conditions.

Abstract:Cadmium (Cd) is one of the most toxic pollutants found in aquatic ecosystems. Although gene expression in algae exposed to Cd has been studied at the transcriptional level, little is known about Cd impacts at the translational level. Ribosome profiling is a novel translatomics method that can directly monitor RNA translation in vivo. Here, we analyzed the translatome of the green alga Chlamydomonas reinhardtii following treatment with Cd to identify the cellular and physiological responses to Cd stress. Interestingly, we found that the cell morphology and cell wall structure were altered, and starch and high-electron-density particles accumulated in the cytoplasm. Several ATP-binding cassette transporters that responded to Cd exposure were identified. Redox homeostasis was adjusted to adapt to Cd toxicity, and GDP-L-galactose phosphorylase (VTC2), glutathione peroxidase (GPX5), and ascorbate were found to play important roles in maintaining reactive oxygen species homeostasis. Moreover, we found that the key enzyme of flavonoid metabolism, i.e., hydroxyisoflavone reductase (IFR1), is also involved in the detoxification of Cd. Thus, in this study, translatome and physiological analyses provided a complete picture of the molecular mechanisms of green algae cell responses to Cd.
Transcriptomic profile of oyster Crassostrea gigas hemocyte after short-term cadmium exposure and bacteria stimulation

Abstract:Oyster Crassostrea gigas, is considered as a useful environmental indicator since it is widely distributed along the intertidal zone whereby it tends to accumulate cadmium and is always exposed to various pathogen agents. However, its molecular responses to both cadmium and pathogen stimulation remain unclear. In the present study, transcriptome data of hemocytes from oysters were analyzed to reveal specific molecular responses of oyster to cadmium or cadmium/bacteria stimulation. A total of 21591, 22872 and 20107 genes were detected in the BLANK, Cd24h and Cd/Bac24h group, respectively. Among them, there were 685 differentially expressed genes collected in the comparison of Cd24h versus BLANK. GO analysis of these genes found that sixteen terms into the Molecular Function category displayed transporter activities, and were all over-enrichment by cadmium exposure, whereas twelve terms into Biological Process category involved mainly in metabolic process of the various cellular components and two terms into Cellular Component category were all under-enrichment. The 330 immune responsive genes were shared by two gene lists of CdBac24h versus BLANK and CdBac24h versus Cd24h, and seven out of thirty terms in GO analysis were related to the immune process. Further annotation of these genes from the KEGG database revealed fourteen pathways, including two nervous system related pathways, arachidonic acid pathway, four immune pathways, MAPK cascade and other four cell signaling pathways, and two energy related pathways. Twenty-two differentially expressed genes were identified to responsive to both cadmium exposure and bacteria stimulation, but in different expression patterns, suggesting that bilateral responsive genes, such as alkaline phosphatase and sodium and chloride-dependent glycine transporter gene, could be candidate biomarkers for early warning of cadmium pollution. The present results collectively indicated that a profound neuro-endocrine-immune regulatory network was activated in response to cadmium and bacteria stimulation in oyster C. gigas, and the expression pattern of some cadmium responsive genes may be either reversed or strengthened by bacteria stimulation. The results provide knowledge on the transcriptomic response profile of oyster after short-term cadmium exposure and bacteria stimulation, which would be useful for future studies on stress response mechanism of mollusc, and some cadmium-bacteria responsive genes may be explored as potential biomarkers for monitoring marine pollution.
Enzyme-Linked Immunosorbent Assay for Human Leukocyte Antigen Antibody Detection and Urine Protein Test Recommended for Follow-Up Monitoring After Renal Transplantation

Abstract:Background. Although the usefulness of posttransplant human leukocyte antigen (HLA) antibody monitoring has been demonstrated, detailed recommendations have not been worked out in its frequency, the type of patients and methods to be used. Enzyme-linked immunosorbent assay is a simple and cost-efficient assay. The urine protein test that reflects renal dysfunction is performed everywhere. We assessed the clinical value of HLA antibody and urine protein monitoring after renal transplantation. Methods. Serum samples were consecutively collected from outpatients (n=323) in 2004 and in 2006. Because 18 had graft failure and 8 died with functioning graft for 2 years, 297 paired sera were tested for HLA antibody using enzyme-linked immunosorbent assay. Urine protein was determined to be positive when the dipstick protein reaction was±or over (20 mg/dL). Results. Total 297 patients were divided according to the change of HLA antibody status. Only patients with all of (i) de novo HLA antibody production, (ii) continuous detection from peripheral blood, and (iii) positive urine protein test had a significantly higher serum creatinine than the others and demonstrated rapid deterioration of Cr (&Dgr;Cr 1.26 mg/dL during 2 years). Negative change of HLA antibody stopped the increase of serum creatinine. Conclusion. The status of HLA antibody and urine protein provides useful information on graft prognosis. Although the tempo of graft injury is relatively slow, a yearly routine HLA antibody test for all patients and the attempt to reduce HLA antibody to negative levels is recommended, when HLA antibody is newly detected and urine protein test is positive.

Abstract:Cadmium (Cd) is one of the most toxic metals in the environment and exerts deleterious effects on plant growth and production. Duckweed has been reported as a promising candidate for Cd phytoremediation. In this study, the growth, Cd enrichment, and antioxidant enzyme activity of duckweed were investigated. We found that both high-Cd-tolerance duckweed (HCD) and low-Cd-tolerance duckweed (LCD) strains exposed to Cd were hyper-enriched with Cd. To further explore the underlying molecular mechanisms, a genome-wide transcriptome analysis was performed. The results showed that the growth rate, chlorophyll content, and antioxidant enzyme activities of duckweed were significantly affected by Cd stress and differed between the two strains. In the genome-wide transcriptome analysis, the RNA-seq library generated 544,347,670 clean reads, and 1608 and 2045 differentially expressed genes were identified between HCD and LCD, respectively. The antioxidant system was significantly expressed during ribosomal biosynthesis in HCD but not in LCD. Fatty acid metabolism and ethanol production were significantly increased in LCD. Alpha-linolenic acid metabolism likely plays an important role in Cd detoxification in duckweed. These findings contribute to the understanding of Cd tolerance mechanisms in hyperaccumulator plants and lay the foundation for future phytoremediation studies.

Cadmium induced oxidative stress, endoplasmic reticulum (ER) stress and apoptosis with compensative responses towards the up-regulation of ribosome, protein processing in the ER, and protein export pathways in the liver of zebrafish

Abstract:The present study identified that exposure to 5, 10, and 20 µg/L Cd for 48 days reduced growth, increased Cd accumulation and levels of reactive oxygen species (ROS) and lipid peroxidation, and induced ER stress and cellular apoptosis in the liver in a dose-dependent manner. However, the survival rate was not affected by Cd. The increased production of ROS might result from reduced catalase (CAT) and copper/zinc-superoxide dismutase (Cu/Zn-SOD) activities, which might trigger ER stress pathways and subsequently induce apoptotic responses, ultimately leading to growth inhibition. Transcriptomic analyses indicated that the differentially expressed genes (DEGs) involved in metabolic pathways were significantly enriched and dysregulated by Cd, suggesting that metabolic disturbances may contribute to Cd toxicity. However, there were increases in glutathione peroxidase (GPX) activity, protein levels of metallothioneins (MTs) and heat shock protein 70 (HSP70), and mRNA levels of sod1, cat, gpx, mt2, and hsp70. Furthermore, DEGs related to ribosome, protein processing in the ER, and protein export pathways were significantly enriched and up-regulated by Cd. These increases may be compensatory responses following oxidative stress, ER stress, and apoptosis to resist negative effects. Taken together, we demonstrated that environmentally relevant levels of Cd induced adaptive responses with compensatory mechanisms in fish, which may help to maintain fish survival at the cost of growth.

Abstract:Introduction Cadmium (Cd) is a toxic heavy metal that severely threatens safe food production. Zhe-Maidong, a well-known Chinese traditional herbal medicine, is susceptible to Cd stress. However, the characteristics of Cd transformation and migration, as well as the regulatory system for genes conferring Cd accumulation of Zhe-Maidong, remains an essential issue to be addressed. Methods Zhe-Maidong seedling growth in Cd-contaminated and uncontaminated soil was conducted for 90 days. The Cd concentration was determined by inductively coupled plasma-mass spectrometry, and the Cd 2+ fluorescence probe detected Cd distributions. The root transcriptome of Zhe-Maidong was then evaluated using various Cd stress hydroponic treatments designated Cd-0, Cd-M, and Cd-H. Results and discussion The enrichment factor (EF) value in the root was four times that of the leaves, indicating that the root has a high ability to absorb and accumulate Cd. The Cd 2+ were mainly distributed in the root hair and the epidermis in both roots and leaves, revealing that the epidermal cells of roots may collect Cd 2+ and also have an outstanding role in Cd 2+ uptake. A total of 50 DEGs involved in Cd translocation and accumulation were identified. Among these DEGs, ANN, ABCC2/4, HMA1- 5, and CCX gene expression were positively correlated with EF-root, EF-leaf, EF-total, Cd-leaf, Cd-root, and Cd-plant, indicating their role in Cd transport and accumulation under Cd-stress. These data could be helpful in uncovering the Cd accumulation characteristics in Zhe-Maidong, as well as provide a bioinformatic foundation for investigations on finding gene functions and the screening of candidate genes related to Cd accumulation.

Abstract:Introduction: Cadmium (Cd) is a significant threat environmental pollutant in the marine ecological environment offshore. The macroalga, Gracilariopsis lemaneiformis , of significant economic value, is widely cultivated along China's coastline. Yet, little is known about the molecular mechanisms underlying Cd tolerance in macroalga. Methods: Here, we examined the transcriptome of G. lemaneiformis exposed to Cd to identify the responses to Cd stress. Results and discussion: Our findings revealed that Cd led to the retardation of growth rate in G. lemaneiformis , accompanied by a notable reduction in the content of photosynthetic pigments and a decrease in the expression of genes associated with the photosynthetic system and nitrogen metabolism. When exposed to Cd, there was a rapid increase in Cd levels through the upregulation of genes encoding GlZIP6 and GlIRT1. Additionally, the expression of Cd efflux transporters, GlZIP1 and GlABCG22, and the ABCC7 transporter for compartmentation to the vacuole, was induced to mitigate Cd toxicity. Cd also activated crucial genes involved in the ABA biosynthesis and enhanced ABA content, thereby inducing ABA signaling pathway. Furthermore, exogenous ABA reduced the growth inhibition of G. lemaneiformis under Cd stress. Redox homeostasis was adjusted to adapt to Cd toxicity, with thioredoxin, glutaredoxin cycle and ascorbate-glutathione cycle identified as playing significant in maintaining reactive oxygen species homeostasis. Moreover, transcription factors such as several MYBs, signal transmission factors G protein and heat shock proteins (sHSPs, HSP 40, HSP 90, HSP101) were involved in the detoxification of Cd. Collectively, this study provided a comprehensive understanding of the molecular mechanisms underpinning the of responses of macroalga G. lemaneiformis to Cd exposure.

Integrated transcriptomics and metabolomics reveal key metabolic pathway responses in Pistia stratiotes under Cd stress

Abstract:Cadmium (Cd) can interfere with plant gene expression, change the content of metabolites and affect plant growth. In this study, untargeted metabolomics (LC-MS) and RNA-Seq sequencing were performed on root tissues of Pistia stratiotes exposed to Cd stress. The results showed that cadmium stress affected the accumulation and transport of cadmium in plants and increased the content of soluble sugar, the activities of ascorbate peroxidase (APX), and peroxidase (POD) by 34.89%, 41.45%, and 6.71% on average, and decreased the activity of superoxide dismutase (SOD) by 51.51% on average. At the same time, the contents of carotenoid, chlorophyll a, and chlorophyll b decreased by 29.52%, 20.11%, and 13.14%, respectively, Thus affecting the growth and development of plants. Metabolomic analysis showed that Cd stress affected eight metabolic pathways, involving 27 differentially expressed metabolites, mainly including unsaturated fatty acids, amino acids (phenylalanine), nucleotides, sulfur compounds, and flavonoids. By transcriptome analysis, a total of 3107 differentially expressed genes (DEGs, 2666 up-regulated genes, and 441 down-regulated genes) were identified, which were mainly involved in four pathways, among which glutathione metabolism and lignin biosynthesis were the key metabolic pathways. In conclusion, this study reveals the metabolic and transcriptional response mechanisms of P. stratiotes to Cd stress through multi-omics, providing the theoretical basis for the phytoremediation of water contaminated by Cd.

Altered Life History Traits and Transcripts of Molting- and Reproduction-Related Genes by Cadmium in Daphnia Magna.

Abstract:Cadmium (Cd) is a non-essential element and can be toxic to aquatic organisms at low concentrations. Despite its well-known toxicity to Daphnia magna, the effects of Cd on physiological parameters (heart rate and thoracic limb activity) and molting- and reproduction-related genes are relatively understudied. In this study, D. magna were exposed to 0 (control), 25, 50 and 75 μg L−1 of Cd for 7 d and 21 d to determine the toxicity of Cd. The results showed that the Cd body burden in D. magna was significantly increased with elevated Cd concentrations, up to 13.4 μg Cd/g dry weight (dw) after exposure to 75 μg L−1 for 21 d. After 21 d of exposure, the body length and body weight of D. magna were significantly decreased in all Cd treatments compared to the control. The heart rate and thoracic limb activity were reduced by 4.3–11.7 and 5.0–10.3%, respectively. The levels of malondialdehyde (MDA) were increased by ~24–37% and the activity of catalase (CAT) was inhibited by ~50% compared to the control. The reproductive parameters (i.e., size of the first brood, the total number of offspring per female and the number of offspring per brood) were remarkably reduced, causing adverse effects on the population dynamics. In addition, the transcripts of genes (cyp314, cyp18a1, ecra, usp, hr3, cut, cht and cht3) related to the molting of D. magna were altered, whereas the transcripts of genes (vtg1, vtg2 and vmo1) related to reproduction were down-regulated. This study helps better understand the effects of Cd at different biological levels.

A total of 50 DEGs involved in Cd translocation and accumulation were identified. Among these DEGs, ANN, ABCC2/4, HMA1- 5, and CCX gene expression were positively correlated with EF-root, EF-leaf, EF-total, Cd-leaf, Cd-root, and Cd-plant, indicating their role in Cd transport and accumulation under Cd-stress. These data could be helpful in uncovering the Cd accumulation characteristics in Zhe-Maidong, as well as provide a bioinformatic foundation for investigations on finding gene functions and the screening of candidate genes related to Cd accumulation.

Abstract:Cadmium ion (Cd2+) is a highly toxic metal in water, even at low concentrations. Microalgae are a promising material for heavy metal remediation. The present study investigated the effects of Cd2+ on growth, photosynthesis, antioxidant enzyme activities, cell morphology, and Cd2+ adsorption and accumulation capacity of the freshwater green alga Scenedesmus obliquus. Experiments were conducted by exposing S. obliquus to varying concentrations of Cd2+ for 96 h, assessing its tolerance and removal capacity towards Cd2+. The results showed that higher concentrations of Cd2+ (>0.5 mg L−1) reduced pigment content, inhibited algal growth and electron transfer in photosynthesis, and led to morphological changes such as mitochondrial disappearance and chloroplast deformation. In this process, S. obliquus counteracted Cd2+ toxicity by enhancing antioxidant enzyme activities, accumulating starch and high-density granules, and secreting extracellular polymeric substances. When the initial Cd2+ concentration was less than or equal to 0.5 mg L−1, S. obliquus was able to efficiently remove over 95% of Cd2+ from the environment through biosorption and bioaccumulation. However, when the initial Cd2+ concentration exceeded 0.5 mg L−1, the removal efficiency decreased slightly to about 70%, with biosorption accounting for more than 60% of this process, emerging as the predominant mechanism for Cd2+ removal. Fourier transform infrared correlation spectroscopy analysis indicated that the carboxyl and amino groups of the cell wall were the key factors in removing Cd2+. In conclusion, S. obliquus has considerable potential for the remediation of aquatic environments with Cd2+, providing algal resources for developing new microalgae-based bioremediation techniques for heavy metals.

Abstract:Introduction: Marine metal contamination caused by deep-sea mining activities has elicited great concern from both social and scientific communities. Among the various metals deep-sea organisms might encounter, cadmium (Cd) is a widely detected metal that in very small amounts is nonetheless capable of severe toxicity. Yet due to both remoteness and technical challenges, insights into the effects of metal exposure resulting from mining activities upon deep-sea organisms are limited. Methods: Here, we investigated Cd's toxicological effects on deep-sea mussels of Gigantidas platifrons exposed to 100 or 1000 g/L of Cd for 7 days; an integrated approach was used that incorporated proteomics and metabolomics along with traditional approaches (metal concentrations, metal subcellular distribution, and anti-oxidative and immune-related biochemical indexes). Results and Discussion: Results showed that Cd exposure caused significant Cd's accumulation in mussel gills and redistribution of Cd among subcellular compartments, with cellular debris being the primary binding site. Although anti-oxidative enzymes activities (superoxide dismutase and catalase) were not significantly altered in mussel gills of both exposed groups, the markedly increased level of glutathione S-transferase detected via proteomic technique clearly evinced that deep-sea mussels suffered from oxidative stress under Cd exposure. Besides, altered activities of acid phosphatase and alkaline phosphatase assayed by traditional methods along with the predominant presence of largely altered immune-related proteins detected by proteomic data strongly revealed an immune response of deep-sea mussels elicited by Cd. In addition, results of proteomics combined with those of non-targeted metabolomics demonstrated that Cd could exert toxicity by disrupting cytoskeleton structure, ion homeostasis, and primary metabolisms of energy, lipid, and nucleotide in deep-sea mussels. As demonstrated in this study, proteomics and metabolomics can be used in tandem to provide valuable insights into the molecular mechanisms of deep-sea organisms' response to Cd exposure and for helping to discover potential biomarkers for application during deep-sea mining assessments.

Abstract:The ultra-high Cd polluted environment is a special habitat in nature. Analysis of the biological adaptation and resistance mechanism of Auxenochlorella protothecoides UTEX234 to ultra-high Cd stress would offer some inspiring understanding on Cd detoxification mechanism and help discovering highly active bioremediation agents. In this study, integrated analyses of the transcriptome, multi-physiological and biochemical data and fatty acid profilings of UTEX2341 were performed for the first time. It was found that exogenous Ca ions could alleviate Cd stress. Manganese-dependent superoxide dismutase and peroxidase also participated in intracellular detoxification. And non-enzymatic antioxidants rather than one specific enzymatic antioxidant were suggested to be used as “core antioxidants”, which witnessed better performance in Cd detoxification. In addition, Cd stress improved sixteen alkane value and biofuel yield and quality.

Abstract:Cadmium (Cd) pollution is a widespread threat to marine life, and ongoing ocean acidification (OA) is predicted to impact bio-toxicity of Cd compounds. However, the cascading effects of changed Cd toxicity to marine primary producers are not well characterized. Here, we studied the impact of OA on Cd toxicity responses in a globally important diatom Phaeodactylum tricornutum under both ambient and elevated pCO2 conditions. We found that increased pCO2 alleviated the impact of additive Cd toxicity on P. tricornutum not only under controlled indoor experiments but also in outdoor mesocosm experiments that reflect more natural growth conditions. Transcriptome analysis suggested that genes involved in Cd efflux and phytochelatin production were up-regulated and genes involved in Cd influx were down-regulated in long-term selected lineages under elevated pCO2. We further found a significant reduction of Cd transfer across trophic level, when the scallop Argopecten irradians was fed with Cd-exposed P. tricornutum previously cultured under elevated pCO2. Our results indicate that after long-term selection of P. tricornutum exposed to future OA conditions (i.e. elevated pCO2), the diatom alters its Cd detoxification strategy, which could have broader impacts on the bio-geochemical cycle of Cd in the marine ecosystem.

Abstract:Cadmium (Cd) has become a severe issue in relatively low concentration and attracts expert attention due to its toxicity, accumulation, and biomagnification in living organisms. Cd does not have a biological role and causes serious health issues. Therefore, Cd pollutants should be reduced and removed from the environment. Microalgae have great potential for Cd absorption for waste treatment since they are more environmentally friendly than existing treatment methods and have strong metal sorption selectivity. This study evaluated the tolerance and ability of the microalga Tetratostichococcus sp. P1 to remove Cd ions under acidic conditions and reveal mechanisms based on transcriptomics analysis. The results showed that Tetratostichococcus sp. P1 had a high Cd tolerance that survived under the presence of Cd up to 100 μM, and IC 50 , the half-maximal inhibitory concentration value, was 57.0 μM, calculated from the change in growth rate based on the chlorophyll content. Long-term Cd exposure affected the algal morphology and photosynthetic pigments of the alga. Tetratostichococcus sp. P1 removed Cd with a maximum uptake of 1.55 mg g −1 dry weight. Transcriptomic analysis revealed the upregulation of the expression of genes related to metal binding, such as metallothionein. Group A, Group B transporters and glutathione, were also found upregulated. While the downregulation of the genes were related to photosynthesis, mitochondria electron transport, ABC-2 transporter, polysaccharide metabolic process, and cell division. This research is the first study on heavy metal bioremediation using Tetratostichococcus sp. P1 and provides a new potential microalga strain for heavy metal removal in wastewater. Abstract:Copper (Cu) and zinc (Zn) often discharge simultaneously from industrial and agricultural sectors and cause stress to aquatic biota. Although microalgae have been extensively investigated for their responses to Cu or Zn exposure, how they cope with the mixtures of two metals, especially at transcriptomic level, remains largely unknown. In this study, Chlamydomonas reinhardtii was exposed to environmentally relevant concentrations of two metals. It was found that Zn promoted the entry of Cu into the algal cells. With the increase of combined toxicity, extracellular polymeric substances (EPS) and cell wall functional groups immobilized significant amounts of Cu and Zn. Furthermore, C. reinhardtii adjusted resistance strategies internally, including starch consumption and synthesis of chlorophyll and lipids. Upon high level of Cu and Zn coexistence, synergistic effects were observed in lipid peroxidation and catalase (CAT) activity. Under 1.05 mg/L Cu + 0.87 mg/L Zn, 256 differentially expressed genes (DEGs) were mainly involved in oxidative phosphorylation, ribosome, nitrogen metabolism; while 4294 DEGs induced by 4.21 mg/L Cu + 3.48 mg/L Zn were mainly related to photosynthesis, citric acid cycle, etc. Together, this study revealed a more comprehensive understanding of mechanisms of Cu/Zn detoxification in C. reinhardtii, emphasizing critical roles of photosynthetic carbon sequestration and energy metabolism in the metal resistance.
