Abstract:Abstract Pulsed electrical stimulation (PES) is known to affect cellular activities. We previously found PES to human dermal fibroblasts (HFs) promoted platelet-derived growth factor subunit A (PDGFA) gene expression, which enhanced proliferation. In this study, we investigated PES effects on fibroblast collagen production and differentiation into myofibroblasts. HFs were electrically stimulated at 4,800 Hz and 5 V for 60 min. Imatinib, a specific inhibitor of PDGF receptors, was treated before PES. Six hours after PES, PDGFA, α-smooth muscle actin (α-SMA) and collagen type I α1 chain gene expressions were upregulated in PES group. Imatinib suppressed the promoted expression except for PDGFA. Immunofluorescence staining and ELISA showed the production of α-SMA and collagen I was enhanced in PES group but suppressed in PES + imatinib group at 48 h after PES. Therefore, PES promotes the production of α-SMA and collagen I in fibroblasts, which is triggered by PDGFA that is upregulated early after PES.

Effects of Pulsed Dye Laser on the Procollagen Mrna Expression and Proliferation of Cultured Fibroblasts in Vitro

Abstract:Cultured fibroblasts were exposed to 585-nm flashlamp-pumped pulsed dye laser(fluence 3 J/cm~2,5 J/cm~2,7 J/cm~2,spot size 7 mm,pulse duration 450 μs) followed by 24 h incubation.The proliferation of fibroblasts was measured by MTT methods and the expression of the procollagen mRNA and SMADs was investigated by real time PCR method.The results showed that 585 nm flashlamp-pumped pulsed dye laser inhibited the proliferation of fibroblasts after exposure to fluence of 5 J/cm~2 and 7 J/cm~2(P0.05).The mRNA expression of procollagen Iα1 and SMAD2,3,4,7 was significantly down-regulated after being treated with 585 nm flashlamp-pumped pulsed dye laser at fluence level of 5 J/cm~2(P0.01,or P0.05).The corresponding fluence level for significant down-regulation of mRNA expression of procollage III α_1 was 6 J/cm~2 and 7 J/cm~2(P0.05).It is concluded that sufficiently high level fluence of 585 nm flashlamp-pumped pulsed dye laser may inhibit the proliferation of fibroblasts in dose-dependent manner in vitro and down-regulate procollagen mRNA expression.

Abstract:The collagen present in the dermis of the skin is a fibrous protein that fills the gaps between cells and helps maintain tissue flexibility. Effectively increasing the collagen present in the skin is an important goal for cosmetic research. Recent research has shown that soybean peptide (SP) has anti-fatigue activity, antioxidant activity, and the ability to increase type I collagen, while collagen peptide (CP) has the ability to enhance corneal moisture content and viscoelasticity, as well as to increase levels of hyaluronic acid synthesizing enzymes in human skin. Little documented research, however, has been conducted on collagen formation in relation to these peptides. Therefore, this research applied SP and CP with molecular weights primarily around 500 and preparations containing both SP and CP to normal human dermal fibroblasts together with magnesium ascorbyl phosphate (VC-PMg), and used real-time PCR to determine the gene expression of type I collagen (COL1A1), which contributes to collagen synthesis, and Smad7, which contribute to collagen breakdown. In addition, enzyme linked immuno sorbent assay (ELISA) was used to measure collagen content in the media. COL1A1 gene expression at 24 h after sample addition showed higher tendency in all samples and increased with time at 4, 8 and 24 h after addition. Smad7 gene expression was not substantially different at 4 h after addition. matrix metalloproteinase-1 gene expression was higher following SP addition, but was lower after the addition of CP and SP+CP. Medium collagen content was higher in all samples and increased with time at 8 h after addition. Collagen levels were higher when SP and CP were added together.

Abstract:Objective:To study the effects of asiaticoside (AD) and madecassoside (MD) on the cell proliferation and the type Ⅰ and type Ⅲ collagen synthesis in cultured human skin fibroblast (HSF) in vitro.Methods:The effect of AD and MD was determined on prolifera- tion of cultured fibroblast by XTT method,and the protein content of procollagen Ⅰ of aminoterminal propeptide (P Ⅰ NP) and procol- lagen Ⅲ of aminoterminal propeptide (P Ⅲ NP) in the culture medium of treated groups and control groups were measured by radioim- munoassay.Results:(1) AD and MD did not have any cytotoxicity at the concentration of 200μg/ml.(2) Compared with control group,the increases of cell proliferation and an obvious stimulation of P Ⅰ NP and P Ⅲ NP protein secretion were showed in the fibrob- last treated with both AD and MD (P0.05).(3) The pharmacological action of AD and MD presented a dose-dependent manner. (4) AD showed a stronger biological activity than MD for the stimulation of target cells proliferation and the P Ⅰ NP excretion in the cultured fibroblasts and medium at the concentration range of 10μg/ml and 150μg/ml (P0.05).Conclusion:Both AD and MD exhibit a significant biology activities on stimulation of the cell proliferation and the type Ⅰ and Ⅲ collagen synthesis in cultured fibrob- lasts in vitro.
Oral Ingestion of AP Collagen Peptide Leads to Systemic Absorption of Gly-Pro-Hyp, Alleviating H2O2-Induced Dermal Fibroblast Aging

Abstract:Collagen-derived dipeptides and tripeptides have various physiological activities. In this study, we compared the plasma kinetics of free Hyp, peptide-derived Hyp, Pro-Hyp, cyclo(Pro-Hyp), Hyp-Gly, Gly-Pro-Hyp, and Gly-Pro-Ala after ingestion of four different collagen samples: AP collagen peptide (APCP), general collagen peptide, collagen, and APCP and γ-aminobutyric acid (GABA) combination. Each peptide was measured by high-performance liquid chromatography and triple quadrupole mass spectrometer. We found that, among all the peptides that were analyzed, only Gly-Pro-Hyp was significantly increased after ingestion of APCP compared with that of general collagen peptides and collagen. In addition, ingestion of the APCP and GABA combination improved the absorption efficiency of Gly-Pro-Ala. Finally, we reveal that Gly-Pro-Hyp was effective for preventing H2O2-induced reduction in extracellular matrix (ECM)-related genes, COL1A, elastin, and fibronectin, in dermal fibroblasts. Taken together, APCP significantly enhances the absorption of Gly-Pro-Hyp, which might act as an ECM-associated signaling factor in dermal fibroblasts, and the APCP and GABA combination promotes Gly-Pro-Ala absorption. Abstract:Collagen-based products are found in different pharmaceuticals, medicine, food, and cosmetics products for a wide variety of applications. However, its use to prevent or improve the health of skin is growing dizzyingly. Therefore, this study investigated whether collagen peptides could induce fibroblast and keratinocyte proliferation and activation beyond reducing an inflammatory response induced by lipopolysaccharide (LPS). Human skin fibroblasts (CCD-1072Sk) and human keratinocytes (hKT-nh-skp-KT0026) were seeded at a concentration of 5 × 104 cells/mL. LPS (10 ng/mL) and three doses of collagen peptides (2.5 mg/mL, 5 mg/mL, 10 mg/mL) were used. The readout parameters were cell proliferation; expression of inducible nitric oxide synthase (iNOS); expression of pro-collagen-1α by fibroblasts; and secretion of interleukin-1β (IL-1β), interleukin-6 (IL-6), interleukin-8 (IL-8), tumor necrosis factor α (TNF-α), transforming growth factor β (TGF-β), and vascular endothelial growth factor (VEGF) by both cell types. The results demonstrated that all doses of collagen supplementation induced increased proliferation of both human fibroblasts (p < 0.01) and human keratinocytes (p < 0.001), while only the dose of 10 mg/mL induced an increased expression of pro-collagen-1α by fibroblasts. Similarly, only the dose of 10 mg/mL reduced LPS-induced iNOS expression in fibroblasts (p < 0.05) and keratinocytes (p < 0.01). In addition, collagen supplementation reduced the LPS-induced IL-1β (p < 0.05), IL-6 (p < 0.001), IL-8 (p < 0.01), and TNF-α (p < 0.05), and increased the TGF-β and VEGF expression in fibroblasts. Furthermore, collagen supplementation reduced the LPS-induced IL-1β (p < 0.01), IL-6 (p < 0.01), IL-8 (p < 0.01), and TNF-α (p < 0.001), and increased the TGF-β (p < 0.05) and VEGF (p < 0.05) expression in keratinocytes. In conclusion, collagen peptides were found to induce fibroblast and keratinocyte proliferation and pro-collagen-1α expression, involving increased expression of TGF-β and VEGF, as well as the suppression of an inflammatory response induced by LPS.
Collagen from Iris squid grafted with polyethylene glycol and collagen peptides promote the proliferation of fibroblast through PI3K/AKT and Ras/RAF/MAPK signaling pathways

Abstract:Collagens from marine sources have been used widely in food, cosmetics and tissue engineering application due to their excellent functional and biological properties. In the present study, a novel protein, collagen from iris squid skin (SSC) was characterized, grafted with polyethylene-glycol (PEG) and Acid-Green 20 (AG) and was investigated the molecular signaling pathways in L-929 fibroblast cells along with their structural peptide analogs. SDS-PAGE and IR spectrum of SSC analysis showed the typical structure of type I collagen. The fibroblast proliferation was evaluated for SSC, SSC grafted PEG (SSC-PEG) and their structural analogs including Gly-Pro-Leu-Gly-Leu-Leu (PEP1), Gly-Pro-Leu-Gly-Leu-Leu-Gly-Phe-Leu (PEP2), Gly-Pro-Leu-Gly-Leu-Leu-Gly-Phe-Leu-Gly-Pro-Leu (PEP3) and Gly-Pro-Leu-Gly-Leu-Leu-Gly-Phe-Leu-Gly-Pro-Leu-Gly-Leu-Ser (PEP4). The optimal concentration of SSC and its derivative was 0.07 μ mol/L. The fibroblast growth-promoting factors were promoted by all the treatment groups by accelerating the PI3K/AKT and Ras/RAF/MAPK signaling pathways in L-929 cells, and inhibiting the secretion of apoptotic factors. Compared to the control group, mRNA and protein expression of AKT in the PI3K/AKT and Ras in Ras/RAF/MAPK signaling pathway were accelerated significantly by PEP4, respectively, while the Bax value was significantly lower (P < 0.01). The promoting effect of PEP1, PEP2, PEP3 and PEP4 on L-929 cells was closely related to the length of the peptides. Therefore, this study disclosed that PEP1, PEP2, PEP3 and PEP4 were novel analogs that greatly promote the proliferation of L-929 cells through PI3K/AKT and Ras/RAF/MAPK signaling pathways.

Abstract:A method to functionalize collagen-based biomaterials with free amine groups was established in an attempt to improve their potential for tethering of bioactive molecules. Collagen sponges were incorporated with amine-terminated multifunctional polyethylene glycol (PEG) derivatives after N-(3-dimethylaminopropyl)-N'-ethylcarbodiimide and N-hydroxysuccinimide (EDC/NHS) cross-linking. The extent of the incorporation of different amounts and different numbers of active moieties of amine-terminated PEG systems into the collagen scaffolds was evaluated using ninhydrin assay, Fourier transform infrared spectrophotometry (FTIR), collagenase degradation assay, denaturation temperature measurements, and in vitro cell studies. A 3% 8-arm amine-terminated PEG was found to be the minimum required effective concentration to functionalize EDC/NHS stabilized collagen scaffolds. EDC/NHS stabilized scaffolds treated with 3% 8-arm amine-terminated PEG exhibited significantly improved denaturation temperature and resistance to collagenase degradation over non-cross-linked scaffolds (p < 0.002). Biological evaluation using 3T3 cells demonstrated that the produced scaffolds facilitated maintenance of the cells' morphology, metabolic activity, and ability to proliferate in vitro. Overall, our results indicate that amine-terminated PEG systems can be used as means to enhance the functionality of collagenous structures.

Abstract:Objective Poly‐L‐Lactic Acid (PLLA) is a synthetic polymer which possesses biocompatible and biodegradable properties, and is widely used in the clinical filler material. This study focuses on the potential role of PLLA on the collagen production of dermal fibroblasts and its mechanism. Patterns of relapse and survival following radical mastectomy. Analysis of 716 consecutive patients

Abstract:The records of 716 consecutive patients with breast cancer randomized from January 1964 to January 1968 between radical and extended radical mastectomy not followed by postoperative radiation therapy were reviewed. Relapse and survival rates were related to different patterns with the intent to define, on clinical basis, the high‐risk groups in whom systemic adjuvant treatment can be attempted. The 10‐year relapse rate was 52.9% for the whole series. In patients with negative axillary nodes (N‐) this finding was 27.9% compared to 75.5% for patients with positive nodes (N+ 1–3: 66.5%, N+>3: 83.6%). The relapse rate was also affected by concomitant involvement of internal mammary nodes (IMN): N‐ IMN+: 60%, N+ IMN+: 96.7%. The corresponding 10‐year survival rates were as follows: 59.7% (total), 81.9% (N‐), 39.6% (N+), 53.7% (1–3), 25.6% (>3), 45.8% (N‐ IMN+), 20.5% (N+ IMN+). In N+ patients relapse and survival rates were directly proportional to the size of primary tumor, while this trend was not present in N‐ patients. Location of primary tumor as well as menopausal status showed no direct effect on relapse and survival. In both nodal groups the sites of first relapse were documented to occur preferentially in distant organs and tissues.
Molecular analysis and clinical aspects of four patients with Chédiak‐Higashi syndrome (CHS)

Abstract:To the Editor : Chédiak-Higashi syndrome (CHS) is a rare autosomal recessive disorder characterized by mutations in the CHS1 gene that consists of 51 coding exons and encodes for a 429-kD protein involved in intracellular trafficking (1–4). The disease was first mentioned by Beguez-Cesar (5), albeit the syndrome is named after Chédiak and Higashi, who first described its characteristic diagnostic findings (6, 7). More than two thirds of patients with CHS have the childhood form of the disease and experience at least one life-threatening ‘accelerated phase’, which corresponds to the clinical picture of hemophagocytic lymphohistiocytosis (HLH) (8). Without early stem cell transplantation most patients with CHS die in their first decade of life due to accelerated phases or severe infections (9). There are milder forms of CHS, the ‘adolescent’ and ‘adult’ form. These children survive until adulthood without experiencing an ‘accelerated phase’ and present with mostly neurological dysfunctions. We here describe the clinical and molecular characteristics of four patients with CHS. For polymerase chain reaction (PCR) amplification, genomic DNA was isolated from peripheral blood using standard methods. After PCR at an annealing temperature between 56 and 60◦C the products were extracted by agarose gel electrophoresis, purified, directly sequenced using BigDye Terminator v1.1 (Applied Biosystems, Weiterstadt, Germany), and run on ABI PRISM 310 or 3730 DNA analyzers. The exons containing a mutation in patients 1 and 3 were amplified in their parents as well. Primer sequences are available on request. The DNA mutation numbering was based on the numbering of (10). Patient 1 showed a homozygous 1 bp insertion in exon 7 (c.575insT). His parents showed equivalent heterozygous mutations. From birth on he suffered from recurrent skin infections and bruised easily. His hair was light blond, the eyes blue. After a cardiogenic shock due to ventricular tachycardia after diphtheria, pertussis, tetanus (DPT) immunization at the age of 4 months, the boy developed a severe pneumonia with atelectasis and a lung abscess at the age of 4 years. After surgical and antibiotic therapy he recovered. At this time the analysis of a blood smear led to the diagnosis of CHS. After a second pneumonia at the age of 6 years he received a long-term medication with antibiotics and antimycotics. Under this therapy severe infections no longer occurred. At present the patient is 17 years old, has a global development delay, but is in good physical condition. No accelerated phase has been observed until now. Patient 2 showed a homozygous 1 bp deletion in exon 17 (c.5004delA). His dark-grey silvery shining hair and a very pale skin colour were already noticed at birth. At the age of 1 year he presented with high fever, lymphadenopathy, pancytopenia and massive hepatosplenomegaly. CHS was diagnosed by bone marrow cytology. The accelerated phase was treated by immunosuppressive therapy with complete remission. Several mild infections followed. At the age of 2 years the boy presented with a second accelerated phase. He received a successful bone marrow transplantation (BMT) at the age of 3 years. His psychomotoric development within the following 2 years was normal. He is now 11 years old. Patient 3 showed a homozygous 1 bp deletion in exon 20 (c.5519 delC). His parents showed equivalent heterozygous mutations. The patient’s hair and skin were slightly grey and he had a partial ocular albinism. He first presented at the age of 3 years with an otitis media and mastoiditis without response to antibiotic therapy and slow recovery. Abstract:Pyropia yezoensis (P. yezoensis) is a marine algae that exhibits antioxidant, anti-inflammatory, antitumor and anti-aging activities. In this study, we investigated the effects of the P. yezoensis peptide, PYP1‑5, on collagen synthesis in the human dermal fibroblast cell line Hs27. Skin aging is related to reduced collagen production and the activities of multiple enzymes, including matrix metalloproteinases (MMPs), which degrade collagen structure in the dermis, and tissue inhibitor of tissue inhibitor of metalloproteinases (TIMPs), which inhibit the action of MMPs. While collagen synthesis is associated with a number of signaling pathways, we examined the increased collagen synthesis via the upregulation of the transforming growth factor-β (TGF-β)/Smad signaling pathway. Using MTS assay, we found that PYP1‑5 did not affect cell viability. Moreover, we confirmed that PYP1‑5 increased type 1 collagen expression using enzyme-linked immunosorbent assay (ELISA), western blot analysis and quantitative PCR. In addition, we identified changes in various enzymes, as well as the mechanisms behind the PYP1‑5-induced collagen synthesis. PYP1‑5 decreased the MMP-1 protein and mRNA levels, and increased the TIMP-1 and TIMP-2 protein and mRNA levels. In addition, PYP1‑5 activated the TGF-β/Smad signaling pathway, which increased TGF-β1, p-Smad2 and p-Smad3 expression, while inhibiting Smad7, an inhibitor of the TGF-β/Smad pathway. Furthermore, PYP1‑5 upregulated transcription factor specificity protein 1 (Sp1) expression, which is reportedly involved in type 1 collagen expression. These findings indicate that PYP1‑5 activates the TGF-β/Smad signaling pathway, which subsequently induces collagen synthesis in Hs27 cells.

Fibroblasts proliferated in collagen gel without adding Pro-Hyp and Hyp-Gly and did not respond to addition of Pro-Hyp and Hyp-Gly, which raises doubts about conclusions from prior research. Unexpectedly high levels of hydroxyprolyl peptides, including Pro-Hyp, however, were present in the FBS (approximately 100 μM), and also in other commercially available forms of FBS (70–80 μM). After removal of low molecular weight (LMW, < 6000 Da) compounds from the FBS by size exclusion chromatography, Pro-Hyp and Hyp-Gly again triggered growth of fibroblasts attached on collagen and increased the number of fibroblasts migrated from mouse skin. These results indicate the presence of bioactive hydroxyprolyl peptides in commercially available FBS, which can mask effects of Pro-Hyp and Hyp-Gly supplementation; our work confirms that Pro-Hyp and Hyp-Gly do play crucial roles in proliferation of fibroblasts.

Abstract:Prolyl-hydroxyproline (Pro-Hyp) levels increase in human blood on ingesting collagen hydrolysate and trigger proliferation of primary cultured fibroblasts attached on collagen. Such proliferative effects have been associated with the beneficial effects of collagen hydrolysate ingestion including enhancement of wound healing. However, some fibroblast cell lines do not respond to Pro-Hyp. In this study, we focused on the mesenchymal stem cell marker p75 neurotrophin receptor (p75NTR), because p75NTR-positive cells are increased at wound healing sites. Most fibroblasts that had migrated from the dissected mouse skin for 1–4 days expressed p75NTR. After prolonged cultivation, however, the ratio of p75NTR-positive fibroblasts dropped. The p75NTR-positive fibroblasts specifically incorporated fluorescein isothiocyanate (FITC)-labeled Pro-Hyp and Pro-Hyp significantly increased the number of p75NTR-positive fibroblasts grown on collagen gel. These results suggest that Pro-Hyp could enhance wound healing by stimulating p75NTR-positive fibroblasts at wound healing sites, while it has little effect on p75NTR-negative resident fibroblasts in normal tissues.

Abstract:BACKGROUND:Lower fluence of 585-nm flashlamp-pumped pulsed dye laser has been successfully used as a nonablative technique in the treatment of wrinkles. The objective of this study was to evaluate the effect of the pulsed dye laser (585 nm) on the production of collagen and the mRNA expression of collagen related gene in fibroblasts in vitro. METHODS:Cultured fibroblasts were treated with a 585-nm flashlamp-pumped pulsed dye laser (fluence 3 J/cm(2), 4 J/cm(2), spot size 7 mm, pulse duration 450 micros). The production of collagen and the mRNA expression of transforming growth factor (TGF)-beta1, SMAD2, SMAD3, SMAD4, SMAD7 and type I procollagen alpha1, alpha2 in fibroblasts were investigated by colorimetry or real time polymerase chain reaction. RESULTS:The production of collagen was significantly up-regulated after treatment with a 585-nm flashlamp-pumped pulsed dye laser with a fluence of 3 J/cm(2) (P < 0.001). The mRNA expression of TGF-beta1, SMAD2, SMAD3, SMAD4, SMAD7 and procollagen I was significantly up-regulated after treatment with a 585-nm flashlamp-pumped pulsed dye laser with a fluence of 3 J/cm(2) (P < 0.001). No significant difference of mRNA expression of SMAD2, SMAD3, SMAD4, SMAD7 and type I procollagen was found between controls and fibroblasts treated with pulsed dye laser with a fluence of 4 J/cm(2) (P > 0.05). CONCLUSIONS:Lower fluence (3 J/cm(2)) pulsed dye laser increased the collagen production in fibroblasts by up-regulating TGF-beta1, SMAD2, SMAD3, SMAD4, SMAD7 and type I procollagen mRNA expression. These may be the reason it can be effectively used in the treatment of wrinkles.

Abstract:Background: Several human studies have demonstrated occurrence of two major collagen peptides, prolyl-hydroxyproline (Pro-Hyp) and hydroxyprolyl-glycine (Hyp-Gly), in human peripheral blood. Some in vitro studies have demonstrated that Pro-Hyp and Hyp-Gly exert chemotaxis on dermal fibroblasts and enhance cell proliferation. Additionally, Pro-Hyp enhances the production of hyaluronic acid by dermal fibroblasts. These findings suggest that the amounts of Pro-Hyp and Hyp-Gly in blood are important factors to show the efficacy of collagen hydrolysates on skin health. Results: We conducted a randomised double-blind placebo-controlled clinical trial of ingestion of two types of collagen hydrolysates, which are composed of different amounts of the bioactive dipeptides Pro-Hyp and Hyp-Gly, to investigate their effects on the improvement of skin conditions. Improvement in skin conditions, such as skin moisture, elasticity, wrinkles, and roughness, were compared with a placebo group at baseline, and 4 and 8 weeks after the start of the trial. In addition, the safety of dietary supplementation with these peptides was evaluated by blood test. Collagen hydrolysate with a higher content of bioactive collagen peptides (H-CP) showed significant and more improvement than the collagen hydrolysate with a lower content of bioactive collagen peptides (L-CP) and the placebo, in facial skin moisture, elasticity (R2), wrinkles and roughness, compared with the placebo group. In addition, there were no adverse events during the trial. Conclusion: This study demonstrated that the use of the collagen hydrolysate with a higher content of Pro-Hyp and Hyp-Gly led to more improvement in facial skin conditions, including facial skin moisture, elasticity, wrinkles and roughness. © 2016 Society of Chemical Industry.

Abstract:A role for substance P has been proposed in musculoskeletal fibrosis, with effects mediated through transforming growth factor beta (TGFβ). We examined the in vitro effects of substance P on proliferation, collagen secretion, and collagen deposition in rat primary dermal fibroblasts cultured in medium containing 10% fetal bovine serum, with or without TGFβ. In six-day cultures, substance P increased cell proliferation at concentrations from 0.0002 to 100 nM. TGFβ increased proliferation at concentrations from 0.0002 to 2 pg/mL, although higher concentrations inhibited proliferation. Substance P treatment alone at concentrations of 100, 0.2, and 0.00002 nM did not increase collagen deposition per cell, yet when combined with TGFβ (5 ng/mL), increased collagen deposition compared to TGFβ treatment alone. Substance P treatment (100 nM) also increased smooth muscle actin (SMA) expression at 72 h of culture at a level similar to 5 ng/mL of TGFβ; only TGFβ increased SMA at 48 h of culture. Thus, substance P may play a role in potentiating matrix deposition in vivo when combined with TGFβ, although this potentiation may be dependent on the concentration of each factor. Treatments targeting substance P may be a viable strategy for treating fibrosis where both substance P and TGFβ play roles.

Abstract:Compound collagen peptide powder (CCPP) improves skin photoaging by reducing oxidative stress and activating the TGF β1/Smad signaling pathway to increase ECM synthesis in fibroblasts. Fish collagen peptide (FCP) has been extensively investigated as a natural product that can combat photoaging; however, its efficacy is limited by its singular composition. Compound collagen peptide powder (CCPP) is a novel functional food formulation that exhibits photoprotective properties and comprises FCP and a blend of natural botanical ingredients. The objective of this study was to investigate the efficacy of CCPP and its molecular mechanism. CCPP had a low molecular weight, facilitating its efficient absorption, and was abundant in amino acids, total polyphenols, and total flavonoids. The results of in vivo studies demonstrated that CCPP exhibited significant efficacy in reducing skin wrinkles, enhancing the contents of water and oil in the skin, and ameliorating histopathological alterations in mice. The results of in vitro studies demonstrated that CCPP effectively mitigated photoaging in human skin fibroblasts by attenuating oxidative stress and promoting extracellular matrix (ECM) synthesis. Moreover, we clearly demonstrated that the TGF β1/Smad pathway was involved in the promotion of ECM synthesis and cell proliferation by CCPP in human skin fibroblasts. These findings suggest that, compared with single collagen, CCPP has a more comprehensive range of antiphotoaging properties.
