Abstract:Amide, as a ubiquitous functional group, is essential in various aspects of chemistry and biology. Although the history of studying amide is rich and fruitful, the synthetic application of amide is very limited due to the inertness of amide C–N bond. Recently,significant advances have been achieved towards the nickel-mediated C–N activation of amides. This approach allows a facile generation of acyl-nickel intermediates, and a number of unique transformations have been designed and realized based on the amide C–N bond activation. Focused on the catalytic transformation, this review summarizes and categorizes the recent advances on the synthetic applications of Ni-mediated C–N bond activation of amides.

Abstract:Ni‐catalyzed activation of C(sp2)‐H bond remains an elusive challenge. Herein, we realized a Ni‐catalyzed selective activation of C(sp2)‐H bond of formamides, which underwent addition reaction with internal alkynes to provide α,β‐unsaturated amide compounds in high yields. The protocol was featured by the cheap and nontoxic catalyst system. Preliminary mechanistic studies shed light on the reaction pathways.

Abstract:Here, we reported for the first time a mechanistically distinctive cobalt-catalyzed Markovnikov-type sequential semihydrogenation/hydrohydrazidation of aliphatic terminal alkynes in one pot. A cobalt hydride species was employed as two roles for both a unique metal-catalyzed Markovnikov-type insertion of the aliphatic terminal alkynes and then metal-catalyzed hydrogen atom transfer of alkenes. This operationally simple protocol exhibits excellent functional group tolerance and step economy. The hydrazone products could be easily transferred to various valuable amine derivatives.

Abstract:Highly regio- and enantioselective intermolecular hydroamination of alkenes is a challenging process potentially leading to valuable chiral amines. Hydroamination of alkenes via metal-catalyzed hydrogen atom transfer (HAT) with good regioselectivity and functional group tolerance has been reported, however, high enantioselectivity has not been achieved due to the lack of suitable ligands. Here we report a ligand-promoted cobalt-catalyzed Markovnikov-type selective radical hydroamination of alkenes with diazo compounds. This operationally simple protocol uses unsymmetric NNN-tridentate (UNT) ligand, readily available alkenes and hydrosilanes to construct hydrazones with good functional group tolerance. The hydrazones can undergo nitrogen-nitrogen bond cleavage smoothly to deliver valuable amine derivatives. Additionally, asymmetric intermolecular hydroamination of unactivated aliphatic terminal alkenes using chiral N-imidazolinylphenyl 8-aminoquinoline (IPAQ) ligands has also been achieved to afford chiral amine derivatives with good enantioselectivities.

Abstract:An enantioselective NiH-catalyzed hydroalkynylation of α,β-unsaturated amides is reported. A wide range of α,β-unsaturated amides and bromoalkynes are suitable substrates for this reaction, which proceeds under mild conditions, allowing access to a variety of enantioenriched chiral β-alkynyl amides. β,γ-Unsaturated amides can also be used as substrates and undergo the β-selective hydroalkynylation with a slightly lower enantiomeric excess.

Abstract:Functionalization of C-H bonds has emerged as a powerful strategy for converting inert, nonfunctional C-H bonds into their reactive counterparts. A wide range of C-H bond functionalization reactions has become possible by the catalysis of metals, typically from the second row of transition metals. First-row transition metals can also catalyze C-H functionalization, and they have the merits of greater earth-abundance, lower cost and better environmental friendliness in comparison to their second-row counterparts. C-H bond alkylation is a particularly important C-H functionalization reaction due to its chemical significance and its applications in natural product synthesis. This review covers Ni-catalyzed C-H bond alkylation reactions using alkyl halides and olefins as alkyl sources.

Abstract:This review highlights the most recent reports in three powerful and ever-growing fields of borrowing hydrogen, acceptorless dehydrogenative coupling, and base-mediated hydride transfer strategies; which pave the way for generating reactive intermediates via shuttling hydrogen (or hydride) between starting materials without any need for an external hydrogen source to easily construct more complex structures. There is a thorough focus on diversifying the utility of alcohols for C/N-alkylation leading to the synthesis of branched ketones, alcohols, amines, indols, and 6-membered nitrogen-containing heterocycles such as pyridines and pyrimidines, various transformations with the focus on C-C and C-N bond-forming reactions via metal-based catalysis or metal-free approaches in this context to give a global overview in this area.

Abstract:The development of photocatalytic intermolecular hydroamination reaction between N‐aminated dihydropyridines and unactivated alkenes is reported. Metal‐free co‐catalysts, rhodamine 6G and thiophenol, in presence of visible light are used to initiate the process. The transformation shows a broad substrate scope, both alkenes and amidyl radical can act as coupling partners. The radical strategy provides excellent anti‐Markovnikov selectivity and regioselectivity in diene substrates.

Catalytic asymmetric hydrofunctionalization of simple alkenes is challenging but still highly sought after. Abstract:Transition-metal-catalyzed remote hydrofunctionalization of alkenes remains an efficient but challenging protocol in chemical synthesis. Herein, we reported a ligand-controlled, directing-group-assisted strategy to promote NiH-catalyzed site-selective (α, β, or γ) hydroamination of unactivated alkenes with weakly coordinating amide groups. The key to success lies in the employment of proper yet commercially available bidentate nitrogen-containing ligands, which enables delivery of 1,1-, 1,2-, and 1,3-diamines with good-to-excellent regioselectivity starting from the same substrates. A broad range of O-benzoylhydroxylamine electrophiles with different functional groups can be installed via Ni migration or nonmigration. Moreover, these predicable and positionally selective protocols provide a method for the enantioselective synthesis of highly valued 1,2-diamines (via remote aliphatic C–H amination) and 1,3-diamines.

Abstract:While enantioselective hydrofluorination methods for activated alkenes represent a notable advance, the resultant enantiomeric excesses remain largely moderate, indicating the necessity for enhancements in precision, efficiency, and scope. We have recently developed an innovative nickel hydride catalytic system that enables regio- and enantioselective C–F bond formation with unactivated alkenes. Abstract:The hydroalkylation reaction of olefins with alkanes is a highly desirable synthetic transformation for the construction of C(sp3)-C(sp3) bonds. However, such transformation has proven to be challenging for unactivated olefins, particularly when the substrates are lack of directing groups or acidic C(sp3)-H bonds. Herein, we address this challenge by merging NiH-catalyzed radical relay strategy with a HAT (hydrogen atom transfer) process. In this catalytic system, a nucleophilic alkyl radical is generated from a C(sp3)-H compound in the presence of a HAT promotor, which couples with an alkyl metallic intermediate generated from the olefin substrate with a NiH catalyst to form the C(sp3)-C(sp3) bond. Notably, this approach represents the first case of transition-metal hydride-catalyzed hydroalkylation of unactivated olefins by employing C(sp3)-H compounds as the alkyl sources. Starting from easily available materials, the reaction not only demonstrates wide func-tional group compatibility but also provides hydroalkylation products with regiodivergency and excellent enantioselectivity through effective catalyst control under mild conditions.

Abstract:Transition metal-catalyzed remote hydrofunctional-ization of alkenes is an efficient method to realize C(sp3)-H functionalization. However, remote hydrofunctionalization of alkenes with unactivated amines and alcohols has not been successfully developed to date. Herein, we report an efficient nickel-catalyzed remote hydroamination and hydroetherification of alkenes with unactivated amines and alcohols, accessing a series of gem-diamine and N,O-acetal derivatives in good to high yields (up to 93%) and exclusive regioselectivities. The mechanistic investigations and DFT computations indicated that the use of 2-iodo-2-methylpropane (tBuI) as both a mild hydride source and radical precursor was essential to afford the remote functionalized products. This research work provides an efficient method to install an amino or alkoxyl at the C(sp3)-H position that is far from the double bond of alkenes.

Abstract:Compounds rich in sp3-hybridized carbons are desirable in drug discovery. Nickel-catalyzed hydrocarbonation of alkenes is a potentially efficient method to synthesize these compounds. By using abundant, readily available, and stable alkenes as pro-nucleophiles, these reactions can have broad scope and high functional group tolerance. However, this methodology is still in an early stage of development, as the first efficient examples were reported only in 2016. Herein, we summarize the progress of this emerging field, with an emphasis on enantioselective reactions. We highlight major developments, critically discuss a wide range of possible mechanisms, and offer our perspective of the state and challenges of the field. We hope this Perspective will stimulate future works in this area, making the methodology widely applicable in organic synthesis.
