Abstract:Most insects must accumulate glycogen before entering diapause. Photoperiod influences diapause beyond the maternal generation in Cotesia vestalis (Haliday) (Hymenoptera: Braconidae). In this study, we measured glycogen content and the activity and messenger RNA (mRNA) expression of glycogen synthase and glycogen phosphorylase at different G0 (25 °C under light:dark photoperiods of 8L:16D, 12L:12D, and 16L:8D) and G1 life stages (13 °C under 8L:16D). Glycogen content in G0 and G1 increased with shorter light periods, except for G0 adult males, which showed no significant difference under the three photoperiods. Compared with those under 16 h, in those under 8 h light, glycogen synthase activity was significantly higher at all tested stages, except for G0 pupa and adult male, for which it was identical; mRNA expression was higher at G0 larva and prepupa, lower at pupa, and identical at rest stages. Glycogen phosphorylase activity was significantly higher for G1 egg, lower at G1 prepupa, and identical at all other stages; mRNA expression was higher at G0 and G1 larval stages, and similar at rest stages. These findings suggest that glycogen, being regulated by the two enzymes, may be a consequential factor in the transmission of maternal information and diapause induction of C. vestalis.

Abstract:Photoperiods influence diapause beyond the maternal generation in Cotesia vestalis (Haliday) (Hymenoptera: Braconidae). The sizes, 20-hydroxyecdysone (20E), and juvenile hormone III (JH III) contents in the G(1) diapausing prepupae, G(0) nondiapausing prepupae, adults, and G(1) eggs from C. vestalis cultures reared under three photoperiods (8: 16 hours light: dark, 12: 12 hours light: dark, and 16: 8 hours light: dark) were measured to investigate the hormones' roles on maternal information transmission and diapause regulation. Results showed that the body size of G(1) diapausing prepupae was significantly larger than that of G(0) nondiapausing prepupae. The sizes of G(0) prepupae, female abdomens, and G(1) eggs under eight-hour light were significantly larger than those under 16-hour light. The 20E content in diapausing prepupae was significantly lower than that in nondiapausing prepupae, whereas JH III content did not show significantly difference under the same photoperiod. The 20E content in maternal prepupae, mated females and G(1) eggs decreased as the light period was reduced. Juvenile hormone III contents in G(0)-mated females and G(1) eggs (after oviposition) under three photoperiods showed no significant differences. These findings suggest that 20E may play a critical role in diapause regulation of C. vestalis, and be used as a maternal environmental message to be transmitted to the next generation.
Evolutionary trade-offs in dormancy phenology

Abstract:Seasonal animal dormancy, hibernation or diapause, is widely interpreted as a physiological response for surviving energetic challenges during the harshest times of the year (the physiological constraint hypothesis). However, there are other mutually non-exclusive hypotheses to explain the timing of animal dormancy, that is, entry into and emergence from hibernation (i.e. dormancy phenology). Other survival advantages of dormancy that have been proposed are reduced risks of predation and competition (the “life-history” hypothesis), but comparative tests across animal species are not yet available. Under this hypothesis, dormancy phenology is influenced by a trade-off between reproductive advantages of being active and survival benefits of dormancy. Within a species, males and females differ in the amount of time and energy they invest in reproduction. Thus, the trade-off between reproduction and survival may be reflected by within-species sex differences in the phenology of dormancy. To examine this hypothesis, we used two complementary approaches: (i) a set of phylogenetic comparative analyses on mammals (mainly holarctic rodents), and (ii) a comparison between endotherm and ectotherm dormancy, via analyses of endotherms (including mainly holoarctic rodents) and the existing literature on ectotherms. Using the phylogenetic comparative method applied to more than 20 hibernating mammalian species, we found support for both hypotheses as explanations for the phenology of dormancy. In accordance with the life history hypotheses, sex differences in emergence and immergence were favored by the sex difference in reproductive effort. In addition, physiological constraint may influence the trade-off between survival and reproduction such that, low temperature and precipitation as well as smaller body mass influence sex differences in phenology. We also compiled initial evidence that ectotherm dormancy (invertebrates and reptiles) may be 1) less temperature dependent than previously thought and 2) associated with trade-offs consistent with the life history hypothesis. Dormancy in some endotherms and ectotherms show staggered phenology with respect to the growing season (earlier emergence and immergence than expected) which illustrates the selection pressure exerted by the trade-off between reproduction (earlier emergence than expected) and adult survival (earlier immergence than expected). Thus, dormancy during non-life-threatening periods that are unfavorable for reproduction may be more widespread than previously appreciated.

Abstract:ABSTRACT In seasonal environments, many animals, including insects, enter dormancy, where they are limited to a fixed energy budget. The inability to replenish energetic stores during these periods suggests insects should be constrained by pre-dormancy energy stores. Over the last century, the community of researchers working on survival during dormancy has operated under the strong assumption that energy limitation is a key fitness trait driving the evolution of seasonal strategies. That is, energy use has to be minimized during dormancy because insects otherwise run out of energy and die during dormancy, or are left with too little energy to complete development, reproductive maturation or other costly post-dormancy processes such as dispersal or nest building. But if energy is so strongly constrained during dormancy, how can some insects – even within the same species and population – be dormant in very warm environments or show prolonged dormancy for many successive years? In this Commentary, we discuss major assumptions regarding dormancy energetics and outline cases where insects appear to align with our assumptions and where they do not. We then highlight several research directions that could help link organismal energy use with landscape-level changes. Overall, the optimal energetic strategy during dormancy might not be to simply minimize metabolic rate, but instead to maintain a level that matches the demands of the specific life-history strategy. Given the influence of temperature on energy use rates of insects in winter, understanding dormancy energetic strategies is critical in order to determine the potential impacts of climate change on insects in seasonal environments.

Abstract:Abstract Many asexually-propagating marine invertebrates can survive extreme environmental conditions by developing dormant structures, i.e., morphologically simplified bodies that retain the capacity to completely regenerate a functional adult when conditions return to normal. Here, we examine the environmental, morphological, and molecular characteristics of dormancy in two distantly related clonal tunicate species: Polyandrocarpa zorritensis and Clavelina lepadiformis . In both species, we report that the dormant structures are able to withstand harsher temperature and salinity conditions compared to the adult, and are the dominant forms these species employ to survive the colder winter months. By finely controlling the entry and exit of dormancy in laboratory-reared individuals, we were able to select and characterize the morphology of dormant structures associated with their transcriptome dynamics. In both species, we identified putative stem and nutritive cells in structures that resemble the earliest stages of asexual propagation. By characterizing gene expression during dormancy and regeneration into the adult body plan (i.e., germination), we observed that genes which control dormancy and environmental sensing in other metazoans, notably HIF-α and insulin signaling genes, are also expressed in tunicate dormancy. Germination-related genes in these two species, such as the retinoic acid pathway, are also found in other unrelated clonal tunicates during asexual development. These results are suggestive of repeated exaptation of conserved eco-physiological and regeneration programs for the origin of novel dormancy-germination processes across distantly related animal taxa.
Prolonged pupal dormancy is associated with significant fitness cost for adults of Rhagoletis cerasi (Diptera: Tephritidae)

Abstract:In temperate areas, dormancy (diapause and/or quiescence) enables herbivorous insect species to persist and thrive by synchronizing growth and reproduction with the seasonal phenology of their host plants. Within-population variability in dormancy increases survival chances under unpredictable environmental changes. However, prolonged dormancy may be costly, incurring trade-offs in important adult fitness traits such as life span and reproduction. We used the European cherry fruit fly, Rhagoletis cerasi, a stenophagous, univoltine species that overwinters in the pupal stage for usually one or more years to test the hypotheses that prolonged dormancy of pupae has trade-offs with body size, survival and reproduction of the resulting adults. We used two geographically isolated populations of R. cerasi to compare the demographic traits of adults obtained from pupae subjected to one or two cycles of warm-cold periods (annual and prolonged dormancy respectively). Regardless of population, adults from pupae that experienced prolonged dormancy were larger than counterparts emerging within 1year. Prolonged dormancy did not affect adult longevity but both lifetime fecundity and oviposition were significantly decreased. Extension of the life cycle of some individuals in R. cerasi populations in association with prolonged dormancy is likely a bet-hedging strategy.
Dormancy promotes coexistence in fluctuating environments

Abstract:Dormancy allows organisms to survive hostile conditions and is hypothesized to enable species to coexist in fluctuating environments. Although determining how species avoid extinction is critical to understanding the dynamics of natural populations, experimental work exploring if and when dormancy rescues populations from extinction remains rare. We conducted an experiment, where we grew two species of nematode at three temperatures. Strains of Caenorhabditis elegans had mutations altering their propensity to enter a dormant stage and Caenorhabditis briggsae was a single strain with a wildtype background. We used those empirical results to parameterize a model and simulate competitive outcomes in fluctuating environments between the two species. We show that upregulating the dormancy pathway rescues populations that would otherwise go extinct, thereby increasing coexistence between competing species. By leveraging the genetic tools available from a model system, this study provides experimental confirmation that dormancy specifically facilitates species coexistence and thereby promotes diversity. This study system could be used more expansively to explore the role of dormancy in species interactions.

Complement resistance of human carcinoma cells depends on membrane regulatory proteins, protein kinases and sialic acid

Abstract:Nucleated cells employ several strategies to evade killing by homologous complement. We studied complement resistance in the human carcinoma cell lines (CA) T47D (mammary), SKOV3 (ovarian), and PC‐3 (prostate) with emphasis on the following mechanisms of defense: 1. Expression and shedding of the membrane complement regulatory proteins (mCRP) CD46, CD55 and CD59; 2. Resistance based on protein phosphorylation; 3. Cell surface expression of sialic acid residues; 4. Desensitization to complement upon exposure to sublytic complement doses. Anti‐mCRP antibody blocking experiments demonstrated that CD59 is the main mCRP protecting these CA from complement. Soluble CD59 was also found in supernates of PC‐3> SKOV3 > T47D cells. Second, inhibitors of PKC, PKA and MEK sensitized the CA to lysis, thus implicating these protein kinases in CA complement resistance. Third, removal of sialic acid residues with neuraminidase also sensitized CA to lysis. Finally, exposure of CA to sublytic doses of complement conferred on them enhanced resistance to lytic complement doses in a PKC‐dependent process. Combined treatment of CA with anti‐CD59 antibodies, PD98059 (a MEK inhibitor) and neuraminidase produced a large enhancement in CA sensitivity to complement. Our results show that CD59 and sialic acid residues present on the cell surface, and intracellular processes involving protein phosphorylation act additively to secure CA resistance to complement‐mediated lysis. Therefore, the effectiveness of antibody‐ and complement‐based cancer immunotherapy will markedly improve by suppression of the various complement resistance mechanisms.
Fetal Ductus Venosus Blood Flow Velocities Before and After Transfusion in Red-Cell Alloimmunized Pregnancies

Abstract:Objective: To compare fetal ductus venosus blood flow velocities in anemic fetuses and normal controls and to study the effect of intravascular transfusion on ductus venosus flow velocities. Methods: Fetal ductus venosus flow velocities were measured using pulsed Doppler ultrasound in 21 anemic fetuses immediately before and after intravascular transfusion and again the day after transfusion. The control group consisted of 21 normal fetuses matched for gestational age. Results: In the anemic fetuses, ductus venosus flow velocities were significantly higher than in controls. Transfusion initially resulted in even higher flow velocities. The following day, ductus venosus flow velocities decreased to values comparable to those in the control group. The ratio of peak to minimum velocity was higher in the anemic fetuses. Conclusion: Our finding of increased ductus venosus blood flow in anemic fetuses supports the theory that in fetal anemia, venous return and therefore cardiac preload is increased. High peak to minimum velocity ratio may reflect increased atrial pressure as a sign of imminent congestive heart failure. Because blood passing through the ductus venosus is directed into the left atrium, increased ductus venosus blood flow in the anemic fetus may be an essential compensatory mechanism to maintain oxygen supply to vital organs such as the heart and brain.
Optimal dormancy strategies in fluctuating environments given delays in phenotypic switching

Abstract:Organisms have evolved different mechanisms in response to periods of environmental stress, including dormancy - a reversible state of reduced metabolic activity. Transitions to and from dormancy can be random or induced by changes in environmental conditions. Prior theoretical work has shown that stochastic transitioning between active and dormant states at the individual level can maximize fitness at the population level. However, such theories of 'bet-hedging' strategies typically neglect certain physiological features of transitions to dormancy, including time lags to gain protective benefits. Here, we construct and analyze a dynamic model that couples stochastic changes in environmental state with the population dynamics of organisms that can initiate dormancy after an explicit time delay. Stochastic environments are simulated using a multi-state Markov chain through which the mean and variance of environmental residence time can be adjusted. In the absence of time lags (or in the limit of very short lags), we find that bet-hedging strategy transition probabilities scale inversely with the mean environmental residence times, consistent with prior theory. We also find that increasing delays in dormancy decreases optimal transitioning probabilities, an effect that can be influenced by the correlations of environmental noise. When environmental residence times - either good or bad - are uncorrelated, the maximum population level fitness is obtained given low levels of transitioning between active and dormant states. However when environmental residence times are correlated, optimal dormancy initiation and termination probabilities increase insofar as the mean environmental persistent time is longer than the delay to reach dormancy. We also find that bet hedging is no longer advantageous when delays to enter dormancy exceed the mean environmental residence times. Altogether, these results show how physiological limits to dormancy and environmental dynamics shape the evolutionary benefits and even viability of bet hedging strategies at population scales.

Abstract:Organisms from many distinct evolutionary lineages acquired the capacity to enter a dormant state in response to environmental conditions incompatible with maintaining normal life activities. Most studied organisms exhibit seasonal or annual episodes of dormancy, but numerous less studied organisms enter long-term dormancy, lasting decades or even centuries. Intriguingly, many planktonic animals produce encased embryos known as resting eggs or cysts that, like plant seeds, may remain dormant for decades. Herein, we studied a rotifer Brachionus plicatilis as a model planktonic species that forms encased dormant embryos via sexual reproduction and non-dormant embryos via asexual reproduction and raised the following questions: Which genes are expressed at which time points during embryogenesis? Abstract:Vegetative hyphae of Aspergillus niger rapidly converted caproic acid into 2-pentanone. More caproic acid was required for maximal ketone production at alkaline as compared to acidic pH values. Further increases in caproate concentrations at each pH value tested (4.5, 5.5, 6.5, 7.5, and 8.5) resulted in inhibition of ketone production and O2 uptake. At alkaline pH values (8.5 and 7.5), oxygen uptake above the endogenous level and the production of 2-pentanone were parallel. This relationship did not hold at acidic pH values. At these pH values, ketone production continued (pH 6.5) or attained a maximum (pH 5.5 and 4.5) at caproate concentrations at which oxygen uptake was inhibited below endogenous levels. These data indicate that endogenous oxygen uptake was not inhibited by caproate at alkaline pH values at concentrations which did inhibit caproate oxidation and 2-pentanone production. Conversely, at acidic pH values, endogenous oxygen uptake was vigorously inhibited by caproate at concentrations at which exogenous fatty acid oxidation and 2-pentanone production were less affected. Simon-Beevers plots of these data showed that the undissociated acid was the permeant form of caproic acid. The fatty anion appeared to be the active or inhibitory form of caproate within the cell. Vegetative hyphae of A. niger were poorly buffered. Once the hyphae were washed and resuspended in phosphate buffer, they were well buffered towards inhibitory concentrations of caproic acid. These findings suggest that the primary mechanism(s) by which caproate inhibits oxygen uptake and ketone formation does not involve a change in the intracellular pH.

Abstract:The phenological response is among the most important traits affecting a species' sensitivity to climate. In insects, strongly seasonal environments often select for a univoltine life cycle such that one seasonal extreme is avoided as an inactive stage. Through understanding the underlying mechanisms for univoltinism, and the consequences of its failure, we can better predict insect responses to climate change. Here we combine empirical data and simulation studies to investigate the consequences of an unusual diapause mechanism in a parthenogenetic matchstick grasshopper, Warramaba virgo, from arid southern Australia. Our field body temperature measurements indicate that this species is a thermoconformer and our laboratory studies of the thermal response of feeding rate imply strong constraints on winter activity. However, the species exhibits no obligate winter diapause, and eggs can develop in 1 month under constant temperatures approximating the mean soil temperature at the time of oviposition (summer). We show that diurnal temperature cycles exceeding a peak of 36 °C inhibit egg development in summer, and that this is sufficient to prevent autumnal hatching of eggs. Development is also strongly retarded below 24 °C. Microclimate-driven simulation studies of egg development show that these thermal responses provide robust maintenance of a univoltine life cycle, thereby resulting in survival of heat stress as an egg (due to limited developmental state) and avoidance of cold stress as a nymph and adult (due to overwintering in the soil as an egg).

Abstract:Winter diapause consists of cessation of development that allows individuals to survive unfavourable conditions. Winter diapause may bear various costs and questions have been raised about the evolutionary mechanisms maintaining facultative diapause. Here, we explored to what extent a facultative winter diapause affects life-history traits and the transcriptome in the damselfly Ischnura elegans, and whether these effects were latitude-specific. We collected adult females at central and high latitudes and raised their larvae in growth chambers. Larvae were split into a non-diapausing and post-winter (diapausing) cohort, were phenotyped and collected for a gene expression analysis. At the phenotypic level, we found no difference in survival between the two cohorts, and the post-winter cohort was larger and heavier than the non-winter cohort. These effects were mostly independent of the latitude of origin. At the transcriptomic level, wintering affected gene expression with a small fraction of genes significantly overlapping across latitudes, especially those related to morphogenesis. In conclusion, we found clear effects of diapause on the phenotype but little evidence for latitudinal-specific effects of diapause. Our results showed a shared transcriptomic basis underpinning diapause demonstrated, here, at the intraspecific level and supported the idea of evolutionary convergence of the response to diapause across organisms.

Abstract:Diapause is a survival strategy for freshwater crustaceans in the genus Daphnia that involves a genetically encoded inhibition of organism growth, development and reproduction. While the environmental triggers for Daphnia to produce diapause‐destined embryos and those that induce hatching are well documented, the influence of environmental challenges during different diapause sub‐phases remains unexplored. In this study, we exposed diapause‐destined embryos of Daphnia magna to different temperatures and light conditions during designated diapause sub‐phases. Our results underscore the importance of the maternal environment for the embryos that are in preparation for diapause. Moreover, we demonstrate a period of obligate developmental arrest during which reinitiation of development cannot be triggered. Additionally, we illustrate the embryos' ability to adjust hatching in response to environmental changes during the hatching process. This study reveals the inherent developmental pattern in Daphnia embryos as they progress through diapause and their adaptability to environmental changes.

Abstract:alpha-Synuclein (ASN) is a brain-enriched protein that functions as a molecular chaperone and regulator of the synaptic vesicle cycle. However, if ASN is overexpressed and in prefibrillar oligomeric forms it activates free radical formation and has been implicated in neurodegeneration. The nuclear target for the free radical cascade is poly (ADP-ribose) polymerase-1 (PARP-1), a DNA-binding enzyme and transcriptional regulator that decides on cell survival or death. Our previous data indicated that soluble oligomeric form of ASN significantly stimulated nitric oxide synthase (NOS) activity and by oxidative stress leads to mitochondria failure and cell death. The aim of this study was to investigate the effect of ASN on PARP-1 protein level and on its activity in the rat brain using radiochemical and immunochemical methods. It was found that ASN (10 microM) had no effect on PARP-1 protein level. However, ASN inhibited this enzyme activity by 35% in rat brain cortex and hippocampus investigated together and in striatum by 18%. An inhibitor of constitutive NOS isoform, N(G)-nitro-L-arginine (NNLA, 100 microM), partially prevented ASN-evoked PARP-1 inhibition. The NO pool liberated by ASN could be involved in the decrease of PARP-1 activity. The direct interaction between ASN and PARP-1 protein should be taken into consideration.
Stabilization of fluctuating population dynamics via the evolution of dormancy

Abstract:Dormancy is usually understood as a strategy for coping with extrinsically variable environments, but intrinsic population fluctuations also create conditions where dormancy is adaptive. By analyzing simple population models, we show that, very generally, population fluctuations favor the evolution of dormancy, but dormancy stabilizes population dynamics. This sets up a feedback loop that can enable the coexistence of alternative dormancy strategies. Over longer timescales, we show that evolution of dormancy to an evolutionary stable state can drive populations to the edge of stability, where dynamics are only weakly stabilized. We briefly consider how these conclusions are likely to apply in more complex community contexts. Our results suggest that chaos and high-amplitude population cycles are highly vulnerable to invasion and subsequent stabilization by dormancy, potentially explaining their rarity. At the same time, the propensity of ecological dynamics to fluctuate may be an underappreciated driver of the evolution of dormancy.

Abstract:Abstract In temperate environments most species of insects enter an arrested state of development, known as diapause, that enables them to survive the adverse environmental conditions associated with winter. Although diapause is restricted to a single life stage within species of insects, there are examples of insects that overwinter in the egg, larval, pupal and adult stages. Here we offer a targeted, non-systematic literature review examining how overwintering impacts subsequent reproduction in female insects. Several factors, including the lifestage at which insects overwinter, the type of energy investment strategy females use for breeding, elements of the winter environment, and contributions from male insects can influence trade-offs that female insects face between overwintering survival and post-diapause reproduction. Additionally, climate change and elements of the urban environment, including light pollution and higher temperatures in cities, can exacerbate or ameliorate trade-offs faced by reproducing female insects. Better understanding trade-offs between overwintering survival and reproduction in insects not only enhances our understanding of the underlying physiological mechanisms and ecological processes governing diapause and reproduction, but also provide opportunities to better manage insect pests and/or support beneficial insects.
