Abstract:Selenium nanoparticles (SeNPs) possess unique features with excellent bioavailability and bioactivity, but the poor stability limits its application. A combination of polysaccharides and SeNPs is an effective strategy to overcome the limitation. Herein, a heteropolysaccharide (SVL-3) with an average molecular weight of 2.428 × 104 Da was purified from the fruiting body residue of Sanghuangporus vaninii after soaking in sorghum wine, which was composed of fucose, galactose, glucose, fructose and 3-O-methyl-galactose. The main chain of SVL-3 was composed of →6)-α-3-MeO-Galp-(1→, →4)-α-D-Galp-(1→, →2,6)-β-D-Glcp-(1 → and →3)-α-D-Glcp-(1→, and the branched chain was composed of →4)-α-D-Xylp-(1 → and α-L-Fucp-(1→. For enhancing bioactivity of SVL-3 and stability of SeNPs, SVL-3-functionalized SeNPs (SVL-3-SeNPs) was prepared, which contained 45.31 % polysaccharide and 48.49 % selenium. SVL-3-SeNPs maintained an emphatic stability over 28 days at 4 °C and pH 6-8, and exhibited a higher cytotoxic effect on MCF-7 cells than SVL-3 and SeNPs. The inhibitory effect of SVL-3-SeNPs on the cancer cells may be associated with the mechanisms by inducing S-phase arrest, triggering apoptosis and elevating the protein levels of Cytochrome c, Caspases and cleaved caspases 3 and 9. These results indicated that SeNPs modified by S. vaninii polysaccharides can be utilized as a potential material for targeted antitumor drugs.

Abstract:Natural bioactive molecules have been widely used as stabilizers in the functional improvement of selenium nanoparticles (SeNPs) in recent years. In this study, Morchella sextelata polysaccharide (MSP) was introduced as a novel stabilizer for the synthesis of SeNPs based on the redox system of sodium selenite and ascorbic acid. The size, morphology, stability, and anti-cancer cell activities were respectively analyzed by various methods. The results showed that the synthesized SeNPs with MSP were 72.07 ± 0.53 nm in size, red in color, spherical in shape, and amorphous in nature. MSP-SeNPs showed high scavenging activity against DPPH and ABTS radicals. And, these MSP-SeNPs exhibited a significant anti-proliferation effect on human liver (HepG2) and cervical cancer (Hela) cells in vitro, while no significant cytotoxicity against normal human kidney cells (HK−2) was observed. Moreover, the mitochondria-dependent apoptosis pathway triggered by MSP-SeNPs in HepG2 cell was identified. The expression levels of p53, Bax, cytochrome c , caspase-3 and caspase-9 were all up-regulated in HepG2 cells after MSP-SeNPs treatment, while Bcl-2 expression was down-regulated. These results suggest that MSP-SeNPs have strong potential as the food supplement for application in cancer chemoprevention.

Abstract:Astragalus alcohol soluble polysaccharide (AASP) could present superior water solubility and antitumor activity with high concentration. Selenium nanoparticles (SeNPs) have received growing attention in various fields, but their unstable property increases the application difficulties. In the present study, functionalized nano-composites (AASP−SeNPs) were synthesized through SeNPs using AASP (average molecular weight of 2.1 × 103 Da) as a surface modifier, and the preliminary structural characteristics and inhibitory mechanism on liver cancer (HepG2) cells were investigated. Results showed that AASP−SeNPs prepared under a sodium selenite/AASP mass ratio of 1/20 (w/w) were uniformly spherical with a mean grain size of 49.80 nm and exhibited superior dispersivity and stability in water solution. Moreover, the composites could dose-dependently inhibit HepG2 cell proliferation and induce apoptosis through effectively regulating mitochondria-relevant indicators including ΔΨm depletion stimulation, intracellular ROS accumulation, Bax/Bcl-2 ratio improvement, and Cytochrome c liberation promotion. These results provide scientific references for future applications in functional food and drug industries.

Abstract:Combining selenium nanoparticles (SeNPs) with bioactive polysaccharides is one of the effective ways to overcome the shortcomings of SeNPs and polysaccharides and obtain novel antitumor drug candidates. In this study, a heteropolysaccharide (MTP70) with moderate antihepatoma activity was isolated from the stems of Marsdenia tenacissima (Roxb.) Wight et Arn. To further improve the antihepatoma activity of MTP70 and the application of SeNPs, a novel stable nanoparticle (MTP-SeNP) was designed and fabricated. MTP-SeNPs (Se content of 8.25 %) were characterized as monodisperse spherical nanoparticles (50 nm) with MTP70 wrapped on the surface of the SeNPs by the formation of CO⋯Se bonds and possessed high stability and good dispersion in water for almost a month. In addition, MTP-SeNPs showed higher inhibitory effect compared with MTP70. MTP-SeNPs could effectively inhibit the proliferation, invasion, and metastasis of HepG2 cells by inducing apoptosis and arresting the cell cycle at the S phase, which were closely related to the activation of the Bax/Bcl-2/Caspases and p21/Akt/Cyclin A2 signaling pathways. Our results provide a theoretical basis for further development and application of M. tenacissima polysaccharide, and show that MTP-SeNPs could be explored as a promising anti-hepatoma agent in the pharmaceutical and biomedical industries.

Abstract:In this study, the novel polysaccharides extracted from Stauntonia Brachyanthera pulp were used as stabilizers for the preparation of selenium nanoparticles (SBPPs-SeNPs). The preparation process, structural characterization, stability, and anti-hepatoma cell activity of SBPPs-SeNPs were studied in detail. The results showed that the SBPPs-SeNPs were symmetrical spheres with an average diameter of about 30–40 nm under optimal process parameters. SBPPs interacted with SeNPs via hydroxyl groups on the surface of SBPPs to form C – O –Se bonds. SBPPs-SeNPs exhibited good stability during the 21-day storage period at 4 °C, likely due to the interaction between SeNPs and the hydroxyl groups. Moreover, in vitro anti-tumor experiments demonstrated that SBPPs-SeNPs significantly inhibited the proliferation of HepG2 cells in a dose-dependent manner, and induced morphological changes, and cell cycle arrest in S phase. SBPPs-SeNPs also triggered apoptosis in HepG2 cells through the mitochondrial pathway. Our findings suggested that SBPPs-SeNPs have the potential to become novel anti-hepatoma agents.

Abstract:Nanoparticles have been found to possess unique advantages in many fields, especially in the field of cancer treatment. Herein, based on the unique physical and chemical properties of natural polysaccharides, the polysaccharide from the edible and medicinal fruits of Chaenomeles speciosa was prepared, and the complex nanoparticles constructed by combining C. speciosa polysaccharide with selenium have been successfully developed by a chemical method. Monodisperse spherical nanoparticles with the particle size of 80.5 nm were characterized by various methods, which exhibited ideal size distribution and prominent stability under physiological conditions and alkaline conditions. Cellular studies demonstrated the nanoparticles significantly inhibited the growth of MCF-7 cells with an IC 50 value of 8.37 ± 0.97 μ g/mL through inducing the apoptosis and arresting the cell circle at S phase. Moreover, the zebrafish assays confirmed the antitumor effects of the nanoparticles, which suppressed the proliferation and migration of tumor and blocked the angiogenesis of transgenic zebrafish. Collectively, the results suggested that the nanoparticles may be considered as a candidate agent to treat breast cancer. Abstract:Recently, selenium nanoparticles have been drawing attention worldwide, and it is crucial to increase the stability of nano-Se. Morinda officinalis polysaccharides (MOP) are the main active component in Morinda officinalis radix. However, their low activity has limited their application. A novel selenium nanoparticle (Se-MOP) was prepared to solve these problems using MOP as a dispersant. The zeta potential was measured to evaluate the stability, and UV and ATR-FTIR were used to investigate the binding type of selenium and MOP. The morphology was observed by the TEM method. Furthermore, the inhibitory effect on five selected cancer cells (HepG2, MCF-7, AGS, PC9, and HCT8) was evaluated, showing remarkable inhibition of all five cancer cells. The mechanism of inhibition was also investigated by cell circle assay, and it was found that Se-MOP could induce cell circle G0/G1 phase arrest. Immune-enhancing activities were evaluated by measuring the proliferation and cytokines of mouse spleen lymphocytes in vitro and quantitative RT-PCR. The results indicated that single stimulation of Se-MOP and synergistic stimulation with PHA or LPS increased immune capacity and improved immune by increasing the expression of cytokines.

Abstract:This wok investigated the effects of Cordyceps sinensis exopolysaccharide‐selenium nanoparticles (EPS-SeNPs), EPS-Se-1, EPS-Se-2, EPS-Se-3, and EPS-Se-4) with particle sizes (79–124 nm) and Se contents (20.11–40.80 μg/mg) on endocytosis and antitumor activity against human hepatocellular carcinoma (HepG2) cells and revealed the apoptosis-related mechanisms. EPS-SeNPs inhibited HepG2 cells proliferation in a dose and Se content-dependent manner by disrupting cell membrane and mitochondrial integrity, promoting reactive oxygen species production. EPS-SeNPs were endocytosed by HepG2 cells through a clathrin-mediated pathway and followed the quasi-first-order kinetics model, indicating physical adsorption played a dominant role in cellular uptake behavior of EPS-SeNPs. Notably, EPS-Se-3 with the lowest particle size (79 nm) showed the highest antitumor activity and the strongest ability to promote cell apoptosis. Western blotting results revealed that EPS-Se-3 increased expressions of Bax, Cytochrome c, cleaved caspase-9, cleaved caspase-3, Fas, p53, and cleaved caspase-8, while decreased the expressions of Bcl-2 and PARP, as contrast to that of control. Overall, EPS-SeNPs induced cell apoptosis through intrinsic mitochondria-mediated and extrinsic death receptor-mediated pathways.

Enhanced Growth Inhibition and Apoptosis Induction in Human Colon Carcinoma HT-29 Cells of Soluble Longan Polysaccharides with a Covalent Chemical Selenylation

Abstract:The selenylated polysaccharides chemically belong to the organic Se-conjugated macromolecules and have recently been attracting more and more attention due to their potential to promote body health or prevent cancers. Longan (Dimocarpus longan L.), as a subtropical fruit, contains soluble and non-digestible polysaccharides that are regarded with health care functions in the body. In this study, the longan polysaccharides (LP) were obtained via enzyme-assisted water extraction, and then chemically selenylated using a reaction system composed of HNO3–Na2SeO3 to yield two selenylated products, namely, SeLP1 and SeLP2, with Se contents of 1.46 and 4.79 g/kg, respectively. The anti-cancer effects of the three polysaccharide samples (LP, SeLP1, and SeLP2) were thus investigated using the human colon cancer HT-29 cells as the cell model. The results showed that SeLP1 and SeLP2 were more able than LP to inhibit cell growth, alter cell morphology, cause mitochondrial membrane potential loss, increase intracellular reactive oxygen and [Ca2+]i levels, and induce apoptosis via regulating the eight apoptosis-related genes and proteins including Bax, caspases-3/-8/-9, CHOP, cytochrome c, DR5, and Bcl-2. It was thereby proven that the selenylated polysaccharides could induce cell apoptosis via activating the death receptor, mitochondrial-dependent, and ER stress pathways. Collectively, both SeLP1 and SeLP2 showed higher activities than LP in HT-29 cells, while SeLP2 was consistently more active than SeLP1 in exerting these assessed anti-cancer effects on the cells. In conclusion, this chemical selenylation covalently introduced Se into the polysaccharide molecules and caused an enhancement in their anti-cancer functions in the cells, while higher selenylation extent was beneficial to the activity enhancement of the selenylated products.

A dandelion polysaccharide and its selenium nanoparticles: Structure features and evaluation of anti-tumor activity in zebrafish models

Abstract:In this study, an inulin fructan (TMP50-2) with moderate anti-tumor activity was obtained from dandelion. To further improve the anti-tumor activity of TMP50-2, a monodisperse and stable spherical nanoparticle (Tw-TMP-SeNP, 50 nm) was fabricated. Physico-chemical analysis revealed that TMP50-2 and Tween 80 were tightly wrapped on the surface of SeNPs by forming C–O⋯Se bonds or through hydrogen bonding interaction (O–H⋯Se). In vitro anti-tumor assay showed that Tw-TMP-SeNP treatment could significantly inhibit the proliferation of cancer cells (HepG2, A549, and HeLa) in a dose-dependent manner, while HepG2 cells were more susceptible to Tw-TMP-SeNP with an IC50 value of 46.8 μg/mL. The apoptosis induction of HepG2 cells by Tw-TMP-SeNP was evidenced by increasing the proportion of apoptotic cells ranging from 12.5% to 27.4%. Furthermore, in vivo zebrafish model confirmed the anti-tumor activity of Tw-TMP-SeNP by inhibiting the proliferation and migration of tumor cells as well as the angiogenesis of zebrafish embryos.

Abstract:An important micronutrient involved in immune response and antitumor is selenium. LMW-GFP, a polysaccharide extracted from Grifola frondosa seed bodies, has a relatively weak antitumor effect on BGC-823 and MFC cells in vitro , whereas selenium binding to LMW-GFP can significantly increase the in vitro antitumor activity of LMW-GFP. In this study, Se-LMW-GFP was prepared by the HNO 3 -Na 2 SeO 3 method, and the structures of LMW-GFP and Se-LMW-GFP were characterized by UV–visible spectroscopy of absorption, FTIR spectroscopy, and electron scanning microscopy, and these structural analyses showed that selenium was successfully complexed to LMW-GFP. The selenium content of Se-LMW-GFP was measured to be 2.08 % ± 0.08 % by ICP-MS. The anti-tumor activity of LMW-GFP before and after selenium modification was compared by cellular experiments, and the findings indicated that the anti-tumor activity of Se-LMW-GFP was considerably improved over that of LMW-GFP, and inhibited the proliferation of BGC-823 cells and MFC cells through a combination of the Fas/FasL-mediated exogenous death receptor pathway as well as the endogenous mitochondrial pathway. Our results suggest that Se-LMW-GFP not only has great potential for natural health food and anti-gastric cancer drug development but is also a good selenium supplement.

Novel walnut peptide-selenium hybrids with enhanced anticancer synergism: facile synthesis and mechanistic investigation of anticancer activity

Abstract:This contribution reports a facile synthesis of degreased walnut peptides (WP1)-functionalized selenium nanoparticles (SeNPs) hybrids with enhanced anticancer activity and a detailed mechanistic evaluation of its superior anticancer activity. Structural and chemical characterizations proved that SeNPs are effectively capped with WP1 via physical absorption, resulting in a stable hybrid structure with an average diameter of 89.22 nm. A panel of selected human cancer cell lines demonstrated high susceptibility toward WP1-SeNPs and displayed significantly reduced proliferative behavior. The as-synthesized WP1-SeNPs exhibited excellent selectivity between cancer cells and normal cells. The targeted induction of apoptosis in human breast adenocarcinoma cells (MCF-7) was confirmed by the accumulation of arrested S-phase cells, nuclear condensation, and DNA breakage. Careful investigations revealed that an extrinsic apoptotic pathway can be attributed to the cell apoptosis and the same was confirmed by activation of the Fas-associated with death domain protein and caspases 3, 8, and 9. In addition, it was also understood that intrinsic apoptotic pathways including reactive oxygen species generation, as well as the reduction in mitochondrial membrane potential, are also involved in the WP1-SeNP-induced apoptosis. This suggested the involvement of multiple apoptosis pathways in the anticancer activity. Our results indicated that WP1-SeNP hybrids with Se core encapsulated in a WP1 shell could be a highly effective method to achieve anticancer synergism. Moreover, the great potential exhibited by WP1-SeNPs could make them an ideal candidate as a chemotherapeutic agent for human cancers, especially for breast cancer.
The Chemical Profiling and Anticancer Potential of Functional Polysaccharides from Flos Sophorae Immaturus

Abstract:Polysaccharides from Flos Sophorae Immaturus (FSI) are one of its pharmacological compounds that can perform effective activities. Aiming to extract the most effective polysaccharides against hepatocellular carcinoma (HCC), the polysaccharides were separated from FSI through ultrasonic microwave extraction, and the first comparison was carried out on the characterization of the structure and its cytotoxic properties on HCC SMMC 7721 cells of undeproteinized purified polysaccharides (PFSI-1) and papain-deproteinized polysaccharides (PFSI-2) from FSI. The findings indicated that PFSI-1 and PFSI-2 had characteristic absorption peaks of polysaccharides; PFSI-1 contained three monosaccharides and PFSI-2 contained ten; and SEM, AFM, and NMR were consistent with the verification of IR polysaccharide characteristics, suggesting probable additional latent activities. The pharmacotoxic effects of both PFSI-1 and PFSI-2 on SMMC 7721 cells (p < 0.05), attenuated the migration ability of SMMC 7721 cells (p < 0.05) and promoted apoptosis (p < 0.05), with an increase in G0/G1-phase cells and decrease in S-phase cells in the PFSI-1 as well as a decrease in G0/G1-phase cells, increase in S-phase cells, and decrease in apoptosis in the PFSI-2 (p < 0.05). The significant cytotoxic effect of PFSI-2 on SMMC 7721 cells (p 0.05) could indicate its potential as a new drug for the treatment of HCC.

Abstract:Selenium-enriched polysaccharides have been gaining great attention for their antitumor activity in recent years. In this study, a novel selenium polysaccharide fraction (Se-POP-3) produced by Pleurotus ostreatus was characterized and its antitumor activity explored at cellular level. Results showed that Se-POP-3 has 25.9 μg/g of selenium, an average molecular weight of 16,106 Da, and is mainly composed of mannose, glucose and galactose with a molar ratio of 1.7:49.6:2.4. Spectra analysis revealed Se-POP-3 as a pyranopolysaccharide linked by α-glycoside bonds in the main chain, and selenium may occur in the form of COSe and SeO. A single sphere of Se-POP-3 has 50–60 nm in aqueous solution, even though it can agglomerate to form larger spherical structures. In vitro experiments with cancer and normal cell lines showed that Se-POP-3 can induce apoptosis and inhibit migration of cancer cells. Potential anticancer mechanism is that Se-POP-3 can disrupt the Bax/Bcl-2 protein ratio and inhibit the epithelial-to-mesenchymal transition (EMT) in cancer cells. Se-POP-3 showed no significant effect on the growth of normal cell lines. Se-POP-3 showed great potential as a broad-spectrum antitumor agent and dietary supplement.

Abstract:The development and application of new selenium-enriched polysaccharides has become a critical topic in recent years. In this study, a natural selenium-enriched polysaccharide fraction (Se-POP-21) produced by Pleurotus ostreatus was purified, characterized, and investigated the antioxidant and antitumor activities in vitro. The Se-POP-21 was mainly composed of mannose, glucose, galactose and arabinose, with a molar ratio of 18.01:2.40:26.15:7.34, of which molecular weight was 15,888 Da and the selenium content was 5.31 μg/g. Spectral analysis demonstrated that Se-POP-21 represented a non-triple helix pyranopolysaccharide and selenium occurred in the form of C-O-Se and SeO. Molecular size and morphology studies showed that Se-POP-21 exhibited a spherical shape with a particle size distribution between 100 and 200 nm, even though Se-POP-21 aggregates were also found with a size between 500 and 600 nm. In addition, Se-POP-21 showed strong scavenging capacity to DPPH and hydroxyl radical. More, cell experiments showed that Se-POP-21 could reduce viability of A549, SKOV3, HepG2 and MCF-7 cells, induce apoptosis and inhibit metastasis of A549 cells. A potential mechanism was that Se-POP-21 inhibited the epithelial-to-mesenchymal transition of cancer cells. Se-POP-21 featured no significant effect on normal cells. Se-POP-21 showed great potential to develop into a natural antioxidant or low-toxic antitumor drug.

Anti-hepatoma immunotherapy of Pholiota adiposa polysaccharide-coated selenium nanoparticles by reversing M2-like tumor-associated macrophage polarization

Abstract:Targeting macrophages to regulate the tumor microenvironment is a promising strategy for treating cancer. This study developed a stable nano drug (PAP-SeNPs) using Se nanoparticles (SeNPs) and the Pholiota adiposa polysaccharide component (PAP-1a) and reported their physical stability, M2-like macrophages targeting efficacy and anti-hepatoma immunotherapy potential, as well as their molecular mechanisms. Furthermore, the zero-valent and well-dispersed spherical PAP-SeNPs were also successfully synthesized with an average size of 55.84 nm and a negative ζ-potential of -51.45 mV. Moreover, it was observed that the prepared PAP-SeNPs were stable for 28 days at 4 °C. Intravital imaging highlighted that PAP-SeNPs had the dual effect of targeting desirable immune organs and tumors. In vitro analyses showed that the PAP-SeNPs polarized M2-like macrophages towards the M1 phenotype to induce hepatoma cell death, triggered by the time-dependent lysosomal endocytosis in macrophages. Mechanistically, PAP-SeNPs significantly activated the Tlr4/Myd88/NF-κB axis to transform tumor-promoting macrophages into tumor-inhibiting macrophages and successfully initiated antitumor immunotherapy. Furthermore, PAP-SeNPs also enhanced CD3+CD4+ T cells and CD3+CD8+ T cells, thereby further stimulating anti-hepatoma immune responses. These results suggest that the developed PAP-SeNPs is a promising immunostimulant that can assist hepatoma therapy.
Mitochondrion-targeted selenium nanoparticles stabilized by Sargassum fusiforme polysaccharides increase reactive oxygen species-mediated antitumour activity

Abstract:Authors prepared a nanoselenium particle stabilized with Sargassum fusiforme polysaccharide (SFPS-Tw-SeNPs) and confirmed that it could effectively inhibit the proliferation of A549 lung cancer cells in vitro. The aim of this study was to investigate its anti-lung cancer effect in vitro and in vivo and its possible mechanism. In cell experiments, AO/EB staining revealed that SFPS-Tw-SeNPs could induce the apoptosis of A549 cells and produce red fluorescence by inserting into DNA through damaged cell membranes, increasing the production of reactive oxygen species (ROS). SFPS-Tw-SeNPs that is loaded with coumarin-6 entered the cells in a concentration-dependent and time-dependent manner, acting on the mitochondria, reducing the mitochondrial membrane potential, increasing the Bax/Bcl-2 ratio, and increasing the expression of Cleaved-Caspase 3, Cleaved-Caspase 9, Cleaved-PARP and Cytochrome C-induced apoptosis in cells. In addition, the SFPS-Tw-SeNPs blocked the PI3K/AKT signalling pathway, downregulated the expression of Cyclin-A and CDK2, upregulated the expression of P21, and arrested the cell in the G1 phase. In animal experiments, SFPS-Tw-SeNPs treatment significantly inhibited the growth of A549 tumour xenografts but did not significantly negatively affect the body of the animals. Overall, SFPS-Tw-SeNPs have the potential to be developed as a pharmaceutical drug to prevent and treat non-small cell lung cancer.
Construction and anti-pancreatic cancer activity of selenium nanoparticles stabilized by Prunella vulgaris polysaccharide

Abstract:Selenium nanoparticles (SeNPs), as a potential cancer therapeutic agent, have attracted extensive attention due to their high anticancer activity and low toxicity. Polysaccharides could be the modifiers and stabilizers to improve the stability and dispersibility of SeNPs in aqueous solution. This study aimed to investigate the physicochemical characterization, stability, and anti-pancreatic cancer cell activities of SeNPs stabilized by a heteroxylan PVP3-1 extracted from the clusters of Prunella vulgaris Linn. Our results showed that PVP3-1 with Mw of 154 kDa was composed of →4)-β-D-Xyl p (1→, →2, 4)-β-D-Xyl p (1→, t-α-L-Ara f (1→ and 4-MeO-α-D-Glc p A(1→. Red, zero-valent, and uniform spherical SeNPs with an average diameter of about 60 nm and high stability in aqueous solution were constructed successfully by polysaccharide PVP3-1. Anti-pancreatic cancer cell activity assays showed that PVP3-1-SeNPs could inhibit the proliferation and migration of pancreatic cancer cells in vitro . Furthermore, PVP3-1-SeNPs induced apoptosis and autophagy of pancreatic cancer cells through inhibiting mTOR signaling pathway. In conclusion, these results indicated that PVP3-1-SeNPs could be potential anti-tumor nanoparticles for treating pancreatic cancer.
