Abstract:Droplet microfluidics generates and manipulates discrete droplets through immiscible multiphase flows inside microchannels. Due to its remarkable advantages, droplet microfluidics bears significant value in an extremely wide range of area. In this review, we provide a comprehensive and in-depth insight into droplet microfluidics, covering fundamental research from microfluidic chip fabrication and droplet generation to the applications of droplets in bio(chemical) analysis and materials generation. The purpose of this review is to convey the fundamentals of droplet microfluidics, a critical analysis on its current status and challenges, and opinions on its future development. We believe this review will promote communications among biology, chemistry, physics, and materials science.

Abstract:Droplet-based microfluidics offer great opportunities for applications in various fields, such as diagnostics, food sciences, and drug discovery. A droplet provides an isolated environment for performing a single reaction within a microscale-volume sample, allowing for a fast reaction with a high sensitivity, high throughput, and low risk of cross-contamination. Owing to several remarkable features, droplet-based microfluidic techniques have been intensively studied. In this review, we discuss the impact of droplet microfluidics, particularly focusing on drug screening and development. In addition, we surveyed various methods of device fabrication and droplet generation/manipulation. We further highlight some promising studies covering drug synthesis and delivery that were updated within the last 5 years. This review provides researchers with a quick guide that includes the most up-to-date and relevant information on the latest scientific findings on the development of droplet-based microfluidics in the pharmaceutical field.

Abstract:The advent of droplet‐based microfluidics provides vast opportunities for various applications including micro‐chemical reaction engineering, material synthesis, and biological detection. In this review, it is clarified the underlying working principles of droplet‐based microfluidic devices, assess the achievements and challenges in this field, and discuss the materials and fabrication methods for microdevices manufacturing. Finally, scaling‐up strategies for different specific applications are summarized. Compared with the conventional emulsification method, droplets generated within microfluidic devices exhibit distinct advantages such as precise control of fluids, exceptional monodispersity, uniform morphology, flexible manipulation, and narrow size distribution. These inherent benefits, including intrinsic safety, excellent heat and mass transfer capabilities, and large surface‐to‐volume ratio, have led to the widespread applications of droplet‐based microfluidics across diverse fields, encompassing chemical engineering, particle synthesis, biological detection, diagnostics, emulsion preparation, and pharmaceuticals. However, despite its promising potential for versatile applications, the practical utilization of this technology in commercial and industrial is extremely limited to the inherently low production rates achievable within a single microchannel. Over the past two decades, droplet‐based microfluidics has evolved significantly, considerably transitioning from a proof‐of‐concept stage to industrialization. And now there is a growing trend towards translating academic research into commercial and industrial applications, primarily driven by the burgeoning demands of various fields. This paper comprehensively reviews recent advancements in droplet‐based microfluidics, covering the fundamental working principles and the critical aspect of scale‐up integration from working principles to scale‐up integration. Based on the existing scale‐up strategies, the paper also outlines the future research directions, identifies the potential opportunities, and addresses the typical unsolved challenges.

Abstract:Micro-nanofluidic technology is widely used in food safety testing, drug screening, new material synthesis and bioengineering. Droplet microfluidic technology is an important branch of micro-nanofluidic technology. The microdroplet technology can produce high-throughput monodisperse droplets. The droplet technology overcomes many problems of continuous flow, such as small sample reagent volume, no cross-contamination, and rapid chemical reaction. So microdroplets have an important position in the fields of single cell analysis, gene sequencing, and real-time diagnosis. This review focuses on the different methods and applications of microdroplet generation in microfluidics. The droplet generation methods are passive and active methods. The passive method does not require external force, while the active method requires external force such as external electric field, magnetic field, acoustic field, and laser field. It is predicted that the quantitative generation of droplets on demand in microfluidics will be the important direction for future research. This review provides new ideas for the applications of the quantitative generation of microdroplets on demand in microfluidics.

Abstract:Advances in microbiology rely on innovations in technology. Droplet microfluidics, as a versatile and powerful technique that allows high-throughput generation and manipulation of subnanoliter volume droplets, has become an indispensable tool shifting experimental paradigms in microbiology. Droplet microfluidics has opened new avenues to various microbiological research, from resolving single-cell heterogeneity to investigating spatiotemporal dynamics of microbial communities, from precise quantitation of microbiota to systematic decipherment of microbial interactions, and from isolating rare and uncultured microbes to improving genetic engineered strains. In this review, we present recent advances of droplet microfluidics in various fields of microbiology: i) microbial cultivation, ii) microorganism detection and characterization, iii) antibiotic susceptibility testing, iv) microbial interactions, v) microbial biotechnology. We also provide our perspectives on the challenges and future directions for droplet microfluidic-based microbiology research.

Abstract:Abstract Droplet-based microfluidic systems have been employed to manipulate discrete fluid volumes with immiscible phases. Creating the fluid droplets at microscale has led to a paradigm shift in mixing, sorting, encapsulation, sensing, and designing high throughput devices for biomedical applications. Droplet microfluidics has opened many opportunities in microparticle synthesis, molecular detection, diagnostics, drug delivery, and cell biology. In the present review, we first introduce standard methods for droplet generation (i.e. passive and active methods) and discuss the latest examples of emulsification and particle synthesis approaches enabled by microfluidic platforms. Then, the applications of droplet-based microfluidics in different biomedical applications are detailed. Finally, a general overview of the latest trends along with the perspectives and future potentials in the field are provided.

Abstract:The microfluidic method primainly utilizes two incompatible liquids as continuous phase and dispersed phase respectively. It controls the formation of droplets by managing the microchannel structure and the flow rate ratio of the two phases. Droplet-based microfluidics is a rapidly expanding interdisciplinary research field encompassing physics, biochemistry, and Microsystems engineering. Droplet microfluidics offer a diverse and practical toolset that enables chemical and biological experiments to be conducted at high speeds and with greater efficiency compared to traditional instruments. The applications of droplet-based microfluidics are vast, including areas such as drug delivery, owing to its compatibility with numerous chemical and biological reagents and its ability to carry out various operations. This technology has been extensively researched due to its promising features. In this review, we delve into the materials used in droplet generation-based microfluidic devices, manufacturing techniques, methods for droplet generation in channels, and, finally, we summarize the applications of droplet generation-based microfluidics in drug delivery vectors, encompassing nanoparticles, microspheres, microcapsules, and hydrogel particles. We also discuss the challenges and future prospects of this technology across a wide array of applications.

Abstract:This review aims to introduce the droplet-based microfluidics which has been growing rapidly in recent years. Some of the basic droplet operations are summarized focusing on fluid flow pattern,mass transfer and reaction both inside droplets and at interface of droplets. Some of typical examples of existing as well as potential applications of droplet-based microreactors in various fields are also discussed. The developed technologies and techniques provide means to precisely control droplet volumes and accurately manipulate behavior of individual droplet(e.g.,generation of droplets, coalescence and fission,mixing and reactions inside the droplets). As a result,each individual droplet acts as a small batch reactor under confined conditions. Accordingly,these small flowing batch reactors together form a new type of novel continuous-flow reactors. Besides the usual advantages brought by microfluidics,e.g.,intensified mass and heat transfer and easy scale-up,droplet-based microfluidics have unique features,such as ability to avoid cross contamination,rapid mixing of fluids inside the droplets,independent control of each droplet and high throughput. These features allow them to be widely used in many fields,such as preparation of functional materials,chemical synthesis, biochemical engineering,and so on.

Abstract:Table of contents droplet microfluidics for molecular and single‐cell analysis. Droplet microfluidics can be applied to the analysis of numerous types of samples and lead to more accurate diagnoses and therapies. During the last decade, molecular diagnostics has become increasingly important, especially during the SARS‐CoV‐2 pandemic. Advances in droplet‐based microfluidics, which divides samples into thousands of microdroplets, are responsible for high‐throughput data provision, increased analysis sensitivity, and improved performance to overcome challenges in controlling diseases and early diagnosis. Different targets, such as nucleic acids, proteins, or single cells, can be detected and analyzed in terms of their characteristics in a much more extensive and precise manner with the integration of droplet microfluidics into various molecular diagnostic techniques. In this review, we explore recent advances in the development of droplet microfluidics‐based molecular diagnostic techniques and future perspectives in the field.

Abstract:Separation and detection are ubiquitous in our daily life and they are two of the most important steps toward practical biomedical diagnostics and industrial applications. A deep understanding of working principles and examples of separation and detection enables a plethora of applications from blood test and air/water quality monitoring to food safety and biosecurity; none of which are irrelevant to public health. Microfluidics can separate and detect various particles/aerosols as well as cells/viruses in a cost-effective and easy-to-operate manner. There are a number of papers reviewing microfluidic separation and detection, but to the best of our knowledge, the two topics are normally reviewed separately. In fact, these two themes are closely related with each other from the perspectives of public health: understanding separation or sorting technique will lead to the development of new detection methods, thereby providing new paths to guide the separation routes. Therefore, the purpose of this review paper is two-fold: reporting the latest developments in the application of microfluidics for separation and outlining the emerging research in microfluidic detection. The dominating microfluidics-based passive separation methods and detection methods are discussed, along with the future perspectives and challenges being discussed. Our work inspires novel development of separation and detection methods for the benefits of public health.

Abstract:Instrument miniaturization is one of the critical issues to improve sensitivity, speed, throughput, and to reduce the cost of analysis. Microfluidics possesses the ability to handle small sample amounts, with minimal concerns related to sample loss and cross-contamination, problems typical for standard fluidic manipulations. Moreover, the native properties of microfluidics provide the potential for high-density, parallel sample processing, and high-throughput analysis. Recently, the coupling of microfluidic devices to mass spectrometry, especially electrospray ionization (ESI) and matrix-assisted laser desorption/ionization (MALDI), has attracted an increasing interest and produced tremendous achievements. The interfaces between microfluidics and mass spectrometry are one of the primary focused problems. In this review, we summarize the latest achievements since 2008 in the field of the technologies and applications in the combining of microfluidics with ESI-MS and MALDI-MS. The integration of several analytical functions on a microfluidic device such as sample pretreatment and separations before sample introduction into the mass spectrometer is also discussed.

Abstract:Cells are the most basic structural and functional units of living organisms. Studies of cell growth, differentiation, apoptosis, and cell-cell interactions can help scientists understand the mysteries of living systems. However, there is considerable heterogeneity among cells. Great differences between individuals can be found even within the same cell cluster. Cell heterogeneity can only be clearly expressed and distinguished at the level of single cells. The development of droplet microfluidics technology opens up a new chapter for single-cell analysis. Microfluidic chips can produce many nanoscale monodisperse droplets, which can be used as small isolated micro-laboratories for various high-throughput, precise single-cell analyses. Moreover, gel droplets with good biocompatibility can be used in single-cell cultures and coupled with biomolecules for various downstream analyses of cellular metabolites. The droplets are also maneuverable; through physical and chemical forces, droplets can be divided, fused, and sorted to realize single-cell screening and other related studies. This review describes the channel design, droplet generation, and control technology of droplet microfluidics and gives a detailed overview of the application of droplet microfluidics in single-cell culture, single-cell screening, single-cell detection, and other aspects. Moreover, we provide a recent review of the application of droplet microfluidics in tumor single-cell immunoassays, describe in detail the advantages of microfluidics in tumor research, and predict the development of droplet microfluidics at the single-cell level.

Abstract:The mainstream of microfluidic chip research has transformed from the platform construction and method development to a wide range of applications. As a typical multiphase micro-functional unit, the droplet can be used as an independent microreactor with a volume ranging from pL to nL and formation rate up to thousands of droplets per second. It has the advantages of restricted diffusion, accelerated mixing, high heat transfer, effective mass transfer and so on. Droplet-based microfluidic technology has been emerged as a powerful tool to carry out high throughput screening and droplet manipulation research. Thus cell analysis, especially single cell analysis, and cell manipulation in droplets are easy and effective to be implemented with the assistance of a series of analytical methods. In this Chapter, we have briefly introduced an overview of droplet generation and corresponding principle by microfluidic chip. Besides, some usual methods coupled with droplet analysis have also been presented such as fluorescence analysis, mass spectrometry, capillary electrophoresis and others. Then we have mainly discussed the progress of droplets in cell analysis of recent decades. Finally, a summary and possible predication of droplet-based microfluidic chip are made at the end of this chapter.

Abstract:Even though droplet microfluidics has been developed since the early 1980s, the number of applications that have resulted in commercial products is still relatively small. This is partly due to an ongoing maturation and integration of existing methods, but possibly also because of the emergence of new techniques, whose potential has not been fully realized. This review summarizes the currently existing techniques for manipulating droplets in two-phase flow microfluidics. Specifically, very recent developments like the use of acoustic waves, magnetic fields, surface energy wells, and electrostatic traps and rails are discussed. The physical principles are explained, and (potential) advantages and drawbacks of different methods in the sense of versatility, flexibility, tunability and durability are discussed, where possible, per technique and per droplet operation: generation, transport, sorting, coalescence and splitting.

Abstract:The ability to manipulate and sort droplets is a fundamental issue in droplet-based microfluidics. Various lab-on-a-chip applications can only be realized if droplets are systematically categorized and sorted. These micron-sized droplets act as ideal reactors which compartmentalize different biological and chemical reagents. Array processing of these droplets hinges on the competence of the sorting and integration into the fluidic system. Recent technological advances only allow droplets to be actively sorted at the rate of kilohertz or less. In this review, we present state-of-the-art technologies which are implemented to efficiently sort droplets. We classify the concepts according to the type of energy implemented into the system. We also discuss various key issues and provide insights into various systems.

Abstract:In this review, we present recent advancements and novel developments in fluidic systems for applied analytical purposes in chemistry, biochemistry, and life science in general that employ and reflect the full benefits of microfluidics. A staggering rise in publications related to integrated, all-in-one microfluidic chips capable of separation, reaction, and detection have been observed, all of which realise the principal of micro total analysis systems or lab-on-a-chip. These integrated chips actively adopt the scaling law concepts, utilising the highly developed fabrication techniques. Their aim is to multi-functionalise and fully automate devices believed to assist the future advancements of point-of-care, clinical, and medical diagnostics.

Abstract:Exosomes are lipid-bilayer enclosed vesicles with diameters of 30–150 nm, which play a pivotal role in cell communication by transporting their cargoes such as proteins, lipids, and genetic materials. In recent years, exosomes have been under intense investigation, as they show great promise in numerous areas, especially as bio-markers in liquid biopsies. However, due to the high heterogeneity and the nano size of exosomes, the separation of exosomes is not easy. This review will deliver an outline of the conventional methods and the microfluidic-based technologies for exosome separation. Particular attention is devoted to microfluidic devices, highlighting the efficiency of exosome isolation by these methods. Additionally, this review will introduce advances made in the integrated microfluidics technologies that enable the separation and analysis of exosomes.

Abstract:We describe the first realization of liquid chromatographic separation in a droplet-based microfluidic system and develop a novel mode for microchip-based chromatography named as droplet-array liquid-liquid chromatography. In this system, two arrays of picoliter-scale droplets immobilized on both sidewalls of a microchannel with droplet trapping technique served as the stationary phase in chromatographic separation, while the other immiscible phase flowing in the microchannel served as the mobile phase. The chromatographic separation was achieved on the basis of multiple extraction and elution of analytes between the droplet array stationary phase and the mobile phase. The proof-of-concept study of the droplet-array LC system was performed in the separation of fluoranthene and benzo[b]fluoranthene. Under the optimum conditions, the two analytes were separated within 26 min with separation efficiencies of 112 mu m and 119 mu m plate height, respectively. The advantages of the present system include simple structure, low driving pressure, and relatively high sample capacity. It can also provide a useful platform for LC theory study and educational purposes by allowing the researchers and students to directly "see" the continuous extraction and elution process of a chromatographic separation.

Abstract:Liquid-liquid extraction plays an important role in separation and purification of substance. Miniaturized extraction processes based on microflow have received increasing attention for application in fields such as small-scale purification, fine chemical synthesis, and the rapid analysis of chemical and biological samples. To outline the microflow extraction from its more basic form to its utilizations, this review begins by introducing the progress of research into fluidic dynamics and mass transfer on the microscale. Typical miniaturized extractors and phase separators are then presented with analysis of their advantages. Finally, typical application of microflow extraction are demonstrated, which show great potential for the miniaturization of separation processes.
