Abstract:Docosahexaenoic acid (DHA) is one ω-3 fatty acid that is essential for the development and function of the brain. However, a large number of clinical trials found that the DHA supplementation showed no advantage on mental and motor skill development in term infants. A strategy based on DHA nanoencapsulation (nano FO) using an edible plant protein, zein, mimicking the milk structure is applied for enhanced maternal and fetal absorptions of DHA to improve early brain development. The nano FO achieved increased absorption in GI tract, enhanced delivery to the maternal, fetal, and offspring brains, and reduced fatty acid accumulation in the fetal liver. In the behavior assessments, the nano FO diet showed enhanced learning and memory improvement compared to the normal FO diet. It indicated that zein nanoencapsulation is with high potential for drug and nutrient deliveries to brain and through placenta to fetus with no toxicity concern.
Study on the Digestive Characteristics of Short-and Medium-Chain Fatty Acid Structural Lipid and Its Rapid Intervention on Gut Microbes: in Vivo and in Vitro Studies

Abstract:Structuring is a feasible scheme to improve lipids' value. Here, S-BL and S-LBL (C4-C12 structural lipids) were obtained through enzymatic ester-ester transesterification or acidolysis interesterification using glyceryl tributyrate/glyceryl tridodecanoate and lauric acid/glyceryl tributyrate as raw materials, respectively. The digestive characteristics of S(BL & nbsp;)and S-LBL were investigated in vitro and in vivo, meanwhile, their effects on gut microbes were studied. Compared with their corresponding triglyceride physical mixture, S(BL & nbsp;)possessed an ideal butyric acid sustained-release effect in simulated stomach digestion. Moreover, the sustained-release effect of S-LBL on glycerol monolaurate (GML) was revealed both in vivo and in vitro, while this effect of the S-BL & nbsp;was obviously occurred in small intestine. S(BL & nbsp;)significantly increased the abundance of Bifidobacterium and S-LBL promoted the growth of Clostridiales within 24 h. Overall, both S-BL & nbsp;and S-LBL showed ideal sustained-release effects on GML rather than butyric acid, which may lead to positive changes in gut microbes.
Characterization of novel lactoferrin loaded capsules prepared with polyelectrolyte complexes.

Abstract:Novel capsules loaded with lactoferrin (LF) were prepared using polyelectrolyte complexes that were formed by water soluble chitosan (WSC), sodium cellulose sulfate (NaCS) and sodium polyphosphate (PPS). Normal chitosan (soluble in acidic conditions) was chosen as a control to prepare similar capsules with NaCS and PPS. 1H NMR and FTIR spectra analysis showed that WSC was in a form of chitosan hydrochloride which can be directly dissolved and protonated in acid-free water. SEM results showed that the capsules had a typical wall-capsule structure with a regular spherical shape and an average diameter of 1.97mm. TGA studies revealed that the thermal stability of the capsules were enhanced and the moisture content of the drug-free/loaded capsules were 6.3% and 3.2%. SDS-PAGE results showed that the primary structures of the processed LF in the capsules were unchanged. Drug loading (LE%) and encapsulation efficiency (EE%) analysis showed that the capsules had a higher LE% (45.6%) and EE% (70.7%) than that of the control. In vitro release studies showed that the capsules had a regular and sustainable release profiles in simulated colonic fluid. All of these results indicated that the capsules prepared could be used as a candidate protein drug carrier for colon.
Macronutrient content and fatty acid composition and their positional distribution in human breast milk from Zhejiang Province, China in different lactation periods

Abstract:Lactational changes in macronutrient content, lipid profile, fatty acid composition, and positional distribution of human breast milk were investigated in this study. A total of 378 milk samples of six different lactation periods, including 0-5, 6-14, 15-30, 31-90, 91-180, and 181-360 days, were collected cross-sectionally from healthy lactating women in Zhejiang, China. As lactation progressed from 0-5 to 15-30 days, the lipid content and the percentages of C10:0, C12:0, C14:0, C18:2n-6, and C18:3n-3 increased significantly, while the protein concentration and the proportions of phospholipids, cholesterols, C16:0, C18:1n-9, C24:1n-9, C20:4n-6, C22:4n-6, C22:5n-3, and C22:6n-3 decreased notably. When lactation was further extended to 181-360 days, the protein content continued to decrease, and the percentages of C12:0 and C14:0 continued to increase, whereas the levels of other tested nutrients remained stable. Although the triacylglycerol positional distributions of some fatty acids underwent significant lactational variations, C14:0, C16:0, C24:1n-9, C22:4n-6, C22:5n-3, and C22:6n-3 were located mainly at the sn-2 position, while C18:1n-9, C18:2n-6, and C18:3n-3 were primarily distributed at the sn-1,3 positions. Compared with human breast milk reported in Western countries, samples in our study demonstrated higher percentages of C18:2n-6, C18:3n-3, C20:4n-6, and C22:6n-3, but lower proportions of C12:0, C14:0, and C18:1n-9. The results from this study indicated a nutritional composition different from that of the Western countries and may provide useful data for the development of infant formulas closer to Chinese breast milk in terms of the fatty acid composition and its specified positional distribution on triglyceride structure.

Abstract:The great challenge in developing safe medications for placenta-derived diseases is to reduce or eliminate fetal drug exposure while still providing the necessary therapeutic effect. Rapid advances in nanotechnology have brought opportunities for the therapy of placenta-derived disease through accumulating the drug in the placenta while reducing its placental penetration. Among various nanocarriers, liposomes are regarded as an ideal type of carrier for placental drug delivery due to their biosafety and biodegradability. However, their placental retention effect with different particle sizes has not been studied. This research aimed to explore a suitable size of liposomes for placenta drug delivery. Cy 5 dye was chosen as a model molecule for tracing the distribution of three different-sized liposomes (similar to 80 nm, 200 nm, and 500 nm) in ICR pregnant mice. The stability, cytotoxicity, and cellular uptake study of Cy 5-loaded liposomes were performed. The in vivo fluorescence studies on ICR pregnant mice suggested that the particle size of liposomes was positively correlated with the degree of liposome aggregation in the placenta. The ratio of fluorescence in the placenta and fetus section (P/F value) was proposed to evaluate the placental retention effect of different-sized liposomes. The results showed that the liposomes with 500 nm had the highest P/F value and thus exhibited the strongest placental retention effect and the weakest placental penetration ability. Moreover, liquid chromatography-mass spectrometry analysis confirmed the reliability of the fluorescence section analysis in exploring the placental retention effect of nanovehicles. In general, this study introduced a simple and intuitive method to evaluate the placental retention effect of nanoplatforms and defined a suitable size of liposomes for placenta-derived disease drug delivery.

Abstract:Low bioavailability and stability of 7,8-dihydroxyflavone (7,8-DHF) hinder its potential application in prevention of humans' disease. 7,8-DHF loaded nanoliposomes decorated by original, crosslinked and glycosylated lactoferrin (LF) were fabricated. The mean particle size of spherical crosslinked and glycosylated LF coating liposomes were larger than uncoated liposomes with a low polydispersity index, and the former had higher encapsulation efficiency (Above 98%). Crosslinked and glycosylated LF coating liposomes had higher antioxidant activity, storage stability, slow-release ability, bioaccessibility and transepithelial transport compared to free 7,8-DHF and 7,8-DHF loaded liposome. Effective hydrogen bonding between phospholipid and 7,8-DHF, while electrostatic interaction and hydrogen bonding between LF proteins and phospholipid existed. The encapsulated 7,8-DHF was in an amorphous state rather than a crystalline form as detected by differential scanning calorimetry. Collectively, crosslinked or glycosylated LF coating liposomes are promising delivery systems for protecting and transporting 7,8-DHF or other relative bioactive components.
Comparative Study on the Digestion and Absorption Characteristics of N-3 LCPUFA-enriched Phospholipids in the Form of Liposomes and Emulsions

Abstract:Previous studies have reported that phospholipids rich in n-3 long-chain polyunsaturated fatty acids (n-3 LCPUFA) in the form of liposome exhibited superior bioactivities than other formulation. However, the digestion and absorption characteristics of n-3 LCPUFA-enriched phospholipids were still unclear, restricting the molecular mechanism analysis related to their distinctive activities. The aim of the present study was to compare the digestion and absorption characteristics of DHA/EPA-PC in the forms of liposome and emulsion. The fatty acid composition and lipid species in serum, intestinal wall and content of healthy mice were determined after oral administration with DHA/EPA-PC. Results showed that the peak value of serum DHA/EPA level in the liposome group was significantly higher than that of the emulsion group (p < 0.05), although the peak in the liposome group appeared at 3 h and the peak time was 2 h in the emulsion group. Lipidomics analysis indicated that the high levels of total PL and PL-DHA could be retained in serum for a substantial period after administration of the DHA/EPA-PC liposome, which might be attributed to that the DHA/EPA-PC in the form of liposomes was hydrolyzed slower by pancreatic phospholipase A2 than the emulsion form in small intestinal content. The obtained results might provide theoretical basis for the development and utilization of marine-derived phospholipids.

Abstract:Preterm infants, often characterized by lower birth weights and underdeveloped physiologies, necessitate specialized nutritional care. While breast milk stands as the ideal nutritional source, offering substantial energy through its fatty acid content to support the infants' growth and developmental needs, its usage might not always be feasible. Fatty acids in breast milk are critical for the development of these infants. In scenarios where breast milk is not an option, formula feeding becomes a necessary alternative. Thus, a comprehensive understanding of the fatty acid profiles in both breast milk and formulas is crucial for addressing the distinct nutritional requirements of preterm infants. This paper aims to summarize the effects of lipid composition, structure, and positioning in breast milk and formula on the growth and development of preterm infants. Furthermore, it explores recent advancements in the use of novel structural lipids in formulas, laying the groundwork for future innovations in formula design specifically catered to the needs of preterm infants.
Addition of Dairy Lipids and Probiotic Lactobacillus fermentum in Infant Formulas Modulates Proteolysis and Lipolysis With Moderate Consequences on Gut Physiology and Metabolism in Yucatan Piglets

Abstract:Breast milk is the gold standard in neonatal nutrition, but most infants are fed infant formulas in which lipids are usually of plant origin. The addition of dairy lipids and/or milk fat globule membrane extracts in formulas improves their composition with beneficial consequences on protein and lipid digestion. The probiotic Lactobacillus fermentum (Lf) was reported to reduce transit time in rat pups, which may also improve digestion. This study aimed to investigate the effects of the addition of dairy lipids in formulas, with or without Lf, on protein and lipid digestion and on gut physiology and metabolism. Piglets were suckled from postnatal days 2 to 28, with formulas containing either plant lipids (PL), a half-half mixture of plant and dairy lipids (DL), or this mixture supplemented with Lf (DL+Lf). At day 28, piglets were euthanized 90 min after their last feeding. Microstructure of digesta did not differ among formulas. Gastric proteolysis was increased ( P < 0.01) in DL and DL+Lf (21.9 ± 2.1 and 22.6 ± 1.3%, respectively) compared with PL (17.3 ± 0.6%) and the residual proportion of gastric intact caseins decreased ( p < 0.01) in DL+Lf (5.4 ± 2.5%) compared with PL and DL (10.6 ± 3.1% and 21.8 ± 6.8%, respectively). Peptide diversity in ileum and colon digesta was lower in PL compared to DL and DL+Lf. DL and DL+Lf displayed an increased ( p < 0.01) proportion of diacylglycerol/cholesterol in jejunum and ileum digesta compared to PL and tended ( p = 0.07) to have lower triglyceride/total lipid ratio in ileum DL+Lf (0.019 ± 0.003) as compared to PL (0.045 ± 0.011). The percentage of endocrine tissue and the number of islets in the pancreas were decreased ( p < 0.05) in DL+Lf compared with DL. DL+Lf displayed a beneficial effect on host defenses [increased goblet cell density in jejunum ( p < 0.05)] and a trophic effect [increased duodenal ( p = 0.09) and jejunal ( p < 0.05) weights]. Altogether, our results demonstrate that the addition of dairy lipids and probiotic Lf in infant formula modulated protein and lipid digestion, with consequences on lipid profile and with beneficial, although moderate, physiological effects.

Abstract:This study systematically explored the enhancement of mannosylerythritol lipid-A (MEL-A) on physicochemical stability, antioxidant activity, and bioavailability of bovine lactoferrin (BLF) emulsion under different pH conditions by spectroscopic techniques, molecular simulation, and simulated in vitro digestion model. The bovine lactoferrin-MEL-A (BLF-MEL-A) emulsions were prepared and characterized with the Fourier infrared, of which results showed that high concentration MEL-A (1.00 mg/mL) changed the secondary structure of pH-induced BLF and rendered an increase in β-sheet and random coil fractions. Based on the results of fluorescence spectrum and isothermal titration calorimetry, hydrogen bonding, van der Waals forces, and electrostatic force were the interaction forces of BLF and MEL-A, which were similar to the simulated data of molecular docking and molecular dynamics. BLF-MEL-A emulsion also exerted considerable antioxidant activities and had great potential for functional food development. In addition, MEL-A could improve the stability of BLF emulsion in simulated in vitro digestion test, which promoted the bioavailability of BLF. Therefore, this study will facilitate to explore the interaction between BLF and MEL-A and expand the application of MEL-A as a food additive in food industry.

Abstract:Human milk is uniquely optimized for the needs of the developing infant. Its composition is complex and dynamic, driven primarily by maternal genetics, and to a lesser extent by diet and environment. One important component that is gaining attention is the milk fat globule (MFG). The MFG is composed of a triglyceride-rich core surrounded by a tri-layer membrane, also known as the milk fat globule membrane (MFGM) that originates from mammary gland epithelia. The MFGM is enriched with glycerophospholipids, sphingolipids, cholesterol, and proteins, some of which are glycosylated, and are known to exert numerous biological roles. Mounting evidence suggests that the structure of the MFG and bioactive components of the MFGM may benefit the infant by aiding in the structural and functional maturation of the gut through the provision of essential nutrients and/or regulating various cellular events during infant growth and immune education. Further, antimicrobial peptides and surface carbohydrate moieties surrounding the MFG might have a pivotal role in shaping gut microbial populations, which in turn may promote protection against immune and inflammatory diseases early in life. This review seeks to: (1) understand the components of the MFG, as well as maternal factors including genetic and lifestyle factors that influence its characteristics; (2) examine the potential role of this milk component on the intestinal immune system; and (3) delineate the mechanistic roles of the MFG in infant intestinal maturation and establishment of the microbiota in the alimentary canal.

Review: early intervention services can be clinically beneficial for people with early psychosis

Abstract:reduced likelihood of relapse compared with standard care (2 randomised controlled trials (RCTs), RR 0.66, 95% CI 0.47 to 0.94) and reduced hospital admissions (3 RCTs, RR 0.67, 95% CI 0.54 to 0.83). Psychosis symptoms were also reduced (positive symptoms: 2 RCTs, standardised mean difference (SMD) −0.21, 95% CI −0.42 to −0.01; negative symptoms: 2 RCTs, SMD −0.39, 95% CI −0.57 to −0.20). Three trials (n=288) of family interventions were identifi ed. The intervention reduced risk of relapse and hospital admission combined compared with standard care (composite RR 0.50, 95% CI 0.32 to 0.80), but this effect was not observed with relapse and hospital admission as single end points. Four trials (n=620) of CBT-based interventions found a reduction in positive symptoms compared with usual care after 2-year post-treatment follow-up (SMD −0.60, 95% CI −0.79 to −0.41) but not at end of treatment. Negative symptoms also showed an improvement at 2-year follow-up only (SMD −0.45, 95% CI −0.80 to −0.09). There was no difference in rates of hospital admission or relapse between patients undergoing CBT and those receiving usual care. CONCLUSIONS Early intervention services, family intervention and CBT all showed some benefi ts for treatment for early psychosis when compared with standard care. Early intervention services seem to give the widest range of benefi ts by improving symptoms and reducing risk of hospital admission and relapse. These effects were observable by the end of treatment.
Comparative analysis of interfacial composition and structure of fat globules in human milk and infant formulas

Abstract:In this study, we analyzed and compared the fat globule interfacial compositions and structures in human milk and three types of infant formulas (IF1: supplied with MFGM, IF2: without MFGM and soy lecithin, IF3: supplied with soy lecithin). The results suggested that the interfacial protein composition of IF1 was comparatively closer to human milk, but still lacked certain bioactive MFGM proteins including XO, ADPH and PAS6/7. Considering the interfacial phospholipid, we observed 23, 31, and 29 phospholipid species that could be used to distinguish human milk and infant formulas (IF1, IF2 and IF3). We also found that phosphatidylinositol (PI) and phosphatidylserine (PS), which can absorb lipase, phosphatidylethanolamine (PE), and phosphatidylcholine (PC) and connect functional fatty acids, were lacking in infant formulas. Moreover, the infant formulas had a smaller average particle size of 0.38 μm and a thicker interfacial layer that interacted with the casein micelles. Even if IF1 included MFGM, fat globules structures like those found in human milk did not form, and the majority of the MFGM was still in the aqueous phase in free form. Overall, the comprehensive analysis in this study could be used to simulate or mimic human milk lipids at the supramolecular level.

Comprehensive lipidomic profiling of human milk from lactating women across varying lactation stages and gestational ages

Abstract:An untargeted lipidomic analysis was conducted to investigate the lipid composition of human milk across different lactation stages and gestational ages systematically. A total of 25 lipid subclasses and 934 lipid species as well as 90 free fatty acids were identified. Dynamic changes of the lipids throughout lactation and gestational phases were highlighted. In general, lactation stages introduced more variations in the lipid composition of human milk than gestational ages. Most lipids decreased as the milk progressed from the colostral stage to the mature stage, with some reaching a peak at the transitional stage. Significant variations in lipid composition across gestational ages were predominantly evident during early lactation period. In mature milks, most of the lipids exhibited no discernible statistical differences among gestational ages. This elucidation offers valuable insights and guidance for tailoring precise nutritional strategies for infants with diverse health needs.
A novel strategy to construct stable fat globules with all major milk fat globule membrane proteins to mimic breast milk fat emulsions at the protein level

Abstract:Milk fat globule membrane (MFGM) proteins have several biological functions and maintain the fat globule structure. However, the major MFGM protein compositions in simulated human milk emulsions are different from those in human milk due to the composition loss in the isolation process of MFGM materials. To overcome this limitation, we developed a novel strategy, namely, the solution enriched with MFGM was homogenized with cream separated from the milk rich in large-sized fat globules. The results of physicochemical properties and the interfacial protein coverage of the emulsions showed that the emulsions prepared by the new method had a smaller particle size, higher stability, and more interfacial protein coverage when the ratio of fat to protein was 1:3. In addition, proteome differences in interfacial proteins between the new emulsions and simulated infant formula emulsions were investigated, and the results revealed that the interface of the emulsions prepared by the new method contained all major MFGM proteins and unique GO annotations and KEGG pathways. However, only four MFGM proteins (XO, ADPH, PAS 6/7) were quantified at the interface of the emulsions prepared by the common method. Furthermore, the protein number and the total relative abundance of major MFGM proteins were approximately 2-fold and 475-fold higher at the interface of the emulsions prepared by the new method compared to the common method. Overall, the study modulated the interfacial protein composition of fat globules by screening the sources of lipid and homogenization methods and revealed its potential effect on processing stability and biological properties.

Abstract:The interfacial composition of fat globules can change the interfacial structure and further affect the digestive characteristics. In this study, the interaction between whey protein (WP), which is the main protein added in infant formula (IF), and milk fat globule membrane phospholipid (MPL) was investigated from the perspective of simulated fat globule structure, we further characterized the physicochemical properties and the in vitro digestive performance of the emulsions. The results showed that the interaction between MPL and WP changed the secondary and tertiary structures of WP, and formed a dense protective layer at the interface of the fat globule with strong interaction, which improved the emulsification property and physical stability of the emulsion. When MPL: WP was 5:10, the emulsions showed smaller particle size, higher absolute value of zeta-potential and more uniform and stable emulsion state. In addition, during the simulated in vitro digestion of infants, the degree of lipolysis and free fatty acid release content were significantly increased, which improved the fat digestion.

Abstract:The provision of essential and non-essential amino acids for breast-fed infants is the major function of milk proteins. In addition, breast-fed infants might benefit from bioactivities of milk proteins, which are exhibited in the intestine during the digestive phase and by absorption of intact proteins or derived peptides. For lactoferrin, osteopontin and milk fat globule membrane proteins/lipids, which have not until recently been included in substantial amounts in infant formulas, in vitro experiments and animal models provide a convincing base of evidence for bioactivities, which contribute to the protection of the infant from pathogens, improve nutrient absorption, support the development of the immune system and provide components for optimal neurodevelopment. Technologies have become available to obtain these compounds from cow´s milk and the bovine compounds also exhibit bioactivities in humans. Randomized clinical trials with experimental infant formulas incorporating lactoferrin, osteopontin, or milk fat globule membranes have already provided some evidence for clinical benefits. This review aims to compare findings from laboratory and animal experiments with outcomes of clinical studies. There is good justification from basic science and there are promising results from clinical studies for beneficial effects of lactoferrin, osteopontin and the milk fat globule membrane complex of proteins and lipids. Further studies should ideally be adequately powered to investigate effects on clinically relevant endpoints in healthy term infants.
Regulation of G Protein‐Coupled Receptor Kinase 2 in Brains of Opiate‐Treated Rats and Human Opiate Addicts

Abstract:Abstract: The effects of opiate drugs (heroin, morphine, and methadone) on the levels of G protein‐coupled receptor kinase 2 (GRK2) were studied in rat and human brain frontal cortices. The density of brain GRK2 was measured by immunoblot assays in acute and chronic opiate‐treated rats as well as in opiate‐dependent rats after spontaneous or naloxone‐precipitated withdrawal and in human opiate addicts who had died of an opiate overdose. In postmortem brains from human addicts, total GRK2 immunoreactivity was not changed significantly, but the level of the membrane‐associated kinase was modestly but significantly increased (12%) compared with matched controls. In rats treated chronically with morphine or methadone modest increases of the enzyme levels (only significant after methadone) were observed. Acute treatments with morphine and methadone induced dose‐ and time‐dependent increases (8–22%) in total GRK2 concentrations [higher increases were observed for the membrane‐associated enzyme (46%)]. Spontaneous and naloxone‐precipitated withdrawal after chronic morphine or methadone induced a marked up‐regulation in the levels of total GRK2 in the rat frontal cortex (18–25%). These results suggest that GRK2 is involved in the short‐term regulation of μ‐opioid receptors in vivo and that the activity of this regulatory kinase in brain could have a relevant role in opiate tolerance, dependence, and withdrawal.

Abstract:Human breastmilk, the ideal food for healthy infants, naturally contains a high concentration of medium-chain fatty acids (MCFAs, about 15% of total fatty acids). MCFAs are an important energy source for infants due to their unique digestive and metabolic properties. MCFA-enriched oils are widely used in an infant formula, especially the formula produced for preterm infants. Recently, there has been a growing interest in the triglyceride structure of MCFAs in human milk, their metabolism, and their effects on infant health. This study summarized the MCFA composition and structure in both human milk and infant formula. Recent studies on the nutritional effects of MCFAs on infant gut microbiota have been reviewed. Special attention was given to the MCFAs digestion and metabolism in the infants. This paper aims to provide insights into the optimization of formulations to fulfill infant nutritional requirements.
