Abstract:Panax quinquefolium L. (American Ginseng, AG) is an herb characteristic for regions of North America and Asia. Due to its beneficial properties it has been extensively investigated for decades. Nowadays, it is one of the most commonly applied medical herbs worldwide. Active compounds of AG are ginsenosides, saponins of the glycosides group that are abundant in roots, leaves, stem, and fruits of the plant. Ginsenosides are suggested to be primarily responsible for health-beneficial effects of AG. AG acts on the nervous system; it was reported to improve the cognitive function in a mouse model of Alzheimer’s disease, display anxiolytic activity, and neuroprotective effects against neuronal damage resulting from ischemic stroke in animals, demonstrate anxiolytic activity, and induce neuroprotective effects against neuronal damage in ischemic stroke in animals. Administration of AG leads to inhibition of hypertrophy in heart failure by regulation of reactive oxygen species (ROS) in mice as well as depletion of cardiac contractile function in rats. It also has an anti-diabetic and anti-obesity potential as it increases insulin sensitivity and inhibits formation of adipose tissue. AG displays anti-cancer effect by induction of apoptosis of cancer cells and reducing local inflammation. It exerts antimicrobial effects against several pathogenic strains of bacteria. Therefore, AG presents a high potential to induce beneficial health effects in humans and should be further explored to formulate precise nutritional recommendations, as well as to assess its value in prevention and therapy of some disorders, including cancer.

Abstract:American ginseng (Panax quinquefolius) has gained recognition as a medicinal and functional food homologous product with several pharmaceutical, nutritional, and industrial applications. However, ginsenosides biosynthesis key regulators, spatiotemporal distribution characteristics, and factors influencing ginsenosides is largely unknown, which lead to the cultivated American ginseng faces the unstable quality and chemical extraction challenge. Here, this review presents an overview of the pharmacological effects, biosynthesis and spatiotemporal distribution of ginsenosides, with emphasis on the impacts of biotic and abiotic factors on ginsenosides in American ginseng. Modern pharmacological studies have demonstrated that American ginseng has neuroprotective, cardioprotective, anti-tumor, anti-diabetes and obesity effects. Additionally, the most genes involved ginsenoside biosynthesis in the upstream processes have been identified, while the downstream regulators(OSCs, CYP450 and UGTs) require further investigation. Additionally, limited knowledge exists regarding the molecular mechanisms of the impact of biotic and abiotic factors on ginsenosides. Notably, the nonmedical parts of American ginseng, particularly the flowers, fibrous roots, and leaves, exhibited higher ginsenoside content than that in the main roots and accounts for a considerable amount of weight in the whole plant, representing promising resources for ginsenosides. Finally, the prospects of molecular breeding and metabolic engineering based on multi-omics to improve the unstable quality of cultivated American ginseng and shortage of ginsenosides was proposed. This review highlights current deficiencies in American ginseng research and proposes solutions to address these limitations, providing a guide for future investigations into American ginseng ginsenosides.

Characterization of the structure, anti-inflammatory activity and molecular docking of a neutral polysaccharide separated from American ginseng berries

Abstract:Aim: American ginseng berries, grown in the aerial parts and harvested in August, are a potentially valuable material. The aim of the study was to analyze the specific polysaccharides in American ginseng berries, and to demonstrate the anti-inflammation effect through in vitro and in vivo experiments and molecular docking. Methods: After deproteinization and dialysis, the extracted crude polysaccharide was separated and purified. The structure of the specific isolated polysaccharide was investigated by Fourier Transform infrared spectroscopy (FT-IR), GC-MS and nuclear magnetic resonance (NMR), and anti-inflammatory activity was evaluated using in vitro and in vivo models (Raw 264.7 cells and zebrafish). Molecular docking was used to analyze the binding capacity and interaction with cyclooxygenase-2 (COX-2). Results: A novel neutral polysaccharide fraction (AGBP-A) was isolated from American ginseng berries. The structural analysis demonstrated that AGBP-A had a weight-average molecular weight (Mw) of 122,988 Da with a dispersity index (Mw/Mn) value of 1.59 and was composed of arabinose and galactose with a core structure containing →6)-Gal-(1→ residues as the backbone and a branching substitution at the C3 position. The side-chains comprised of α-L-Ara-(1→, α-L-Ara-(1→, →5)-α-L-Ara-(1→, β-D-Gal-(1→. The results showed that it significantly decreased pro-inflammatory cytokines in the cell model. In a zebrafish model, AGBP-A reduced the massive recruitment of neutrophils to the caudal lateral line neuromast, suggesting the relief of inflammation. Molecular docking was used to analyze the combined capacity and interaction with COX-2. Conclusion: Our study indicated the potential efficacy of AGBP-A as a safe and valid natural anti-inflammatory component.

Abstract:Panax ginseng C.A.Meyer is a perennial herb of the Araliaceae family.As the use of traditional Chinese medicine as a new resource food supply becomes more and more popular and sales of P.ginseng are increasing worldwide.The therapeutic potential of P.ginseng has been studied extensively,and ginsenosides,the principal active ingredients of P.ginseng,are shown to be involved in diverse biological and pharmacological activities.Currently, there are several commercial extract preparations such as G115 (marketed as Ginsana),which is the standardized extracts of the roots of P.ginseng and contains 4% ginsenosides.The modern action mechanism of P.ginseng had not been known until ginsenosides were isolated in 1963.Much effort has since been focused on evaluating the function and elucidating the molecular mechanism of each ginsenoside.A variety of publications on P.ginseng and ginsenosides has been growing exponentially since 1975.This article reviews the important biological and pharmacological activities of triterpenoids isolated from P.ginseng over the last few years.These include safety assessment,and central nervous and cardio-cerebral vascular regulation,immunomodulating, endocrine system and anti-stress, anti-inflammation, cytotoxicity against tumor cells and anticancer, antidiabetic and antiobesity,lowering blood lipid,hepatoprotection,anti-osteoporosis,anti-fatigue,as well as anti-aging activities and so on, which might greatly contribute to provide the scientific basis for Evidence-Based Medicine in clarifying the effective substance basis of P.ginseng.

Abstract:Neutral polysaccharides are one of the active components in the American ginseng roots. Recently, the studies about American ginseng polysaccharides are mainly centered on acidic polysaccharides, while there is relatively limited research that explores neutral polysaccharides. In this study a novel neutral polysaccharide (PPQN) was isolated from American ginseng roots and its structure and anti-inflammatory activity were investigated. The average molecular weight (M-w) of PPQN was 3.1 kDa. Monosaccharide components' analysis indicated that PPQN was composed of glucose (Glc) and galactose (Gal) in a molar ratio of 1:1.15. Secretions of nitric oxide (NO), TNF-alpha, IL-6, and IL-1 beta after lipopolysaccharide (LPS) stimulation were detected in PPQN pre-treatment RAW264.7 macrophages. PPQN (150 mu g/mL) exhibited more effective inhibition of TNF-alpha, IL-1 beta, and IL-6 secretions, followed by NO production with respective values of 40.5%, 41.1%, 34.4%, and 11.1% suppression. These results indicated that PPQN may have therapeutic implications in treatment of inflammation and inflammatory-related diseases. (C) 2014 Elsevier B.V. All rights reserved.

Abstract:American ginseng, a precious classic herbal medicine, is used extensively in China for life prolongation purpose. This study aimed to elucidate the structure and anti-inflammatory activity of a neutral polysaccharide isolated from American ginseng (AGP-A). Nuclear magnetic resonance in conjunction with gas chromatography-mass spectrometry were used to analyze AGP-A's structure, whereas Raw264.7 cell and zebrafish models were employed to assess its anti-inflammatory activity. According to the results, AGP-A has a molecular weight of 5561 Da and is primarily consisted of glucose. Additionally, linear α-(1 → 4)-glucans with α-D-Glcp-(1 → 6)-α-Glcp-(1→ residues linked to the backbone at C-6 formed the backbone of AGP-A. Furthermore, AGP-A significantly decreased pro-inflammatory cytokines (IL-1β, IL-6 and TNF-α) in Raw264.7 cell model. AGP-A in zebrafish model significantly lower the massive recruitment of neutrophils to the neuromast of the caudal lateral line. Inflammation may be relieved by the AGP-A component in American ginseng based on these results. In conclusion, our study shows the structural characterization, remarkable anti-inflammatory properties of AGP-A and its potential curative efficacy as a safe, valid natural anti-inflammatory medicine.

Abstract:Ginseng (Panax ginseng C. A. Meyer) is known as a valuable traditional Chinese herb for tens of centuries. Previous pharmacological and phytochemical investigations indicate that polysaccharides are a main biologically active ginseng component that possesses different bioactivities such as antitumor, immunoregulation, antioxidation, hepatoprotection, and anti-inflammation. Work is needed to summarize the full aspects of these polysaccharides systematically to boost their further investigation and application. In this review, studies on the extraction, purification, characterization, and bioactivity are introduced in detail. Moreover, the structure–bioactivity relationship of ginseng polysaccharides is explored and discussed. The uronic acids richer in ginseng polysaccharides tend to have higher biological effect, and the α-(1→4)-GalpA and α-(1→4)-Galp in the main chain are essential for the anti-inflammatory and antiviral effects. We believe our work proposes a helpful bibliography for further innovation and promotes these natural products in functional food.

Abstract:Asian ginseng (Panax ginseng) and American ginseng (Panax quinquefolius L.) are the two most recognized ginseng botanicals. It is believed that the ginseng saponins called ginsenosides are the major active constituents in both ginsengs. Although American ginseng is not as extensively studied as Asian ginseng, it is one of the best selling herbs in the U.S., and has garnered increasing attention from scientists in recent years. In this article, after a brief introduction of the distribution and cultivation of American ginseng, we discuss chemical analysis of saponins from these two ginsengs, i.e., their similarities and differences. Subsequently, we review pharmacological effects of the saponins, including the effects on the cardiovascular system, immune system, and central nervous system as well as the antidiabetes and anti-cancer effects. These investigations were mainly derived from American ginseng studies. We also discuss evidence suggesting that chemical modifications of ginseng saponins would be a valuable approach to develop novel compounds in drug discovery.

Abstract:Background Panax quinquefolius L, widely recognized for its valuable contributions to medicine, has aroused considerable attention globally. Different from the extensive research has been dedicated to the root of P. quinquefolius , its berry has received relatively scant focus. Given its promising medicinal properties, this study was focused on the structural characterizations and anti-inflammatory potential of acidic polysaccharides from the P. quinquefolius berry. Materials and methods P. quinquefolius berry was extracted with hot water, precipitated by alcohol, separated by DEAE-52-cellulose column to give a series of fractions. One of these fractions was further purified via Sephadex G-200 column to give three fractions. Then, the main fraction named as AGBP-A3 was characterized by methylation analysis, NMR spectroscopy, etc. Its anti-inflammatory activity was assessed by RAW 264.7 cell model, zebrafish model and molecular docking. Results The main chain comprised of α -L-Rhap, α -D-GalAp and β -D-Galp, while the branch consisted mainly of α -L-Araf, β -D-Glcp, α -D-GalAp, β -D-Galp. The RAW264.7 cell assay results showed that the inhibition rates against IL-6 and IL-1 β secretion at the concentration of 625 ng/mL were 24.83%, 11.84%, while the inhibition rate against IL-10 secretion was 70.17% at the concentration of 312 ng/mL. In the zebrafish assay, the migrating neutrophils were significantly reduced in number, and their migration to inflammatory tissues was inhibited. Molecular docking predictions correlated well with the results of the anti-inflammatory assay. Conclusion The present study demonstrated the structure of acidic polysaccharides of P. quinquefolius berry and their effect on inflammation, providing a reference for screening anti-inflammatory drugs.

Abstract:Panax notoginseng and Panax quinquefolium are important economic plants that utilize dried roots for medicinal and food dual purposes; there is still insufficient research of their stems and leaves, which also contain triterpenoid saponins. The extraction process was developed with a total saponin content of 12.30 ± 0.34% and 12.19 ± 0.64% for P. notoginseng leaves (PNL) and P. quinquefolium leaves (PQL) extracts, respectively. PNL and PQL saponin extracts showed good antioxidant, antihypertensive, hypoglycemic, and anti-inflammatory properties in vitro and RAW264.7 cells. A total of 699 metabolites were identified in PNL and PQL saponin extracts, with the majority being triterpenoid saponins, flavonoids and amino acids. Fourteen ginsenosides, 18 flavonoids or alkaloids, and 16 amino acids were enriched in both saponin extracts. Overall, the utilization of saponins from medicinal plants PNL and PQL has been developed to facilitate systematic research in the functional food and natural product industries.
Free Radical Scavenging Effect and Immunomodulatory Activity of Total Saponins Extract of Ginseng Fibrous Roots

Abstract:Ginseng (Panax ginseng C.A. Mey) is known for its rich saponin compounds and tonic effects. To better utilize the medicinal value of ginseng, this study investigated the extraction process, components, free radical scavenging ability, and immunomodulatory activity of total saponins of ginseng fibrous roots. The response surface methodology was employed to optimize the extraction process of total saponins, and Q-Orbitrap high-resolution liquid chromatography–mass spectrometry (LC-MS) was used to identify the chemical constituents in the total saponins extract of ginseng fibrous roots (GRS). The results showed that the optimal extraction process was achieved with an ethanol concentration of 68%, a material–solvent ratio of 1:25 mL/g, and an extraction time of 20 min, yielding a total saponin content of 6.34% under these conditions. The extract contained four terpenoid compounds and four polyphenolic compounds. GRS exhibited considerable scavenging activity against DPPH and ABTS radicals, with IC50 values of 0.893 and 0.210 mg/mL, respectively. Moreover, GRS restored immune suppression in mice by increasing white blood cell, red blood cell, and neutrophil counts, and improving the lymphocyte. It also promoted immune system recovery, as evidenced by elevated serum levels of IL-2, IFN-γ, TNF-α, and IL-1β in mice. GRS is a natural compound with promising potential for developing antioxidants and immunomodulatory foods.

Abstract:Polysaccharides from Panax ginseng C. A. Meyer (P. ginseng) are the main active component of P. ginseng and exhibit significant intestinal anti-inflammatory activity. However, the therapeutic mechanism of the ginseng polysaccharide is unclear, and this hinders the application for medicine or functional food. In this study, a polysaccharide was isolated from P. ginseng (GP). The primary structure and morphology of the GP were studied by HPLC, FT-IR spectroscopy, and scanning electron microscopy (SEM). Further, its intestinal anti-inflammatory activity and its mechanism of function were evaluated in experimental systems using DSS-induced rats, fecal microbiota transplantation (FMT), and LPS-stimulated HT-29 cells. Results showed that GP modulated the structure of gut microbiota and restored mTOR-dependent autophagic dysfunction. Consequently, active autophagy suppressed inflammation through the inhibition of NF-κB, oxidative stress, and the release of cytokines. Therefore, our research provides a rationale for future investigations into the relationship between microbiota and autophagy and revealed the therapeutic potential of GP for inflammatory bowel disease.

Abstract:Ginsenosides are the major active constituents in both white and red American ginseng (AG), and their pharmacological effects on central nervous, cardiovascular, and endocrine systems have been well elucidated. However, the anti-Propionibacterium acnes (P. acnes) activity of them is still unknown. In this study, 5 ginsenosides enriched fractions were obtained from the total ginsenosides extract of AG roots (TAG) by resin adsorption and heat-transformation. Resin purification decreased the total polyphenol (TP) content in the fractions. However, heat treatment increased the TP content and induced extensive conversion of the polar ginsenosides (Rg1, Re, Rb1, Rc, Rb2, Rd, and Gyp XVII) to less polar compounds (Rg2, Rg3, Rg6/F4, Rs3, and Rg5/Rk1). Only the heat-treated fraction from 65% methanol elution of the HP-20 column (HPHF) showed antibacterial activity against P. acnes (ATCC11827, MIC, 128 μg/mL; ATCC6919 MIC, 64 μg/mL), S. epidermidis (MIC, 4100 μg/mL) and S. aureus (MIC, 15000 μg/mL). In the anti-inflammatory activity assay, 10 μg/mL of HPHF significantly (P < 0.05) inhibited NO production in LPS-stimulated RAW264.7 cells. These results suggest that less polar ginsenosides enriched fraction HPHF obtained from AG might be useful to develop new types of antibacterial substances and new skin care cosmetics for acne prevention and therapy.

Abstract:The panax genus is a widely used medicinal plant with good biological activity. As one of the main active components of the Panax genus, polysaccharides have various pharmacological effects. This review summarizes the latest research reports on ginseng, American ginseng, and Panax notoginseng polysaccharides and compares the differences in extraction, isolation and purification, structural characteristics, and biological activities. The current research mainly focuses on ginseng polysaccharides, and the process of extraction, isolation, and structure analysis of each polysaccharide is roughly the same. Modern pharmacological studies have shown that these polysaccharides have antioxidants, antitumor, immunomodulatory, antidiabetic, intestinal protection, skin repair, and other biological activities. This review provides new insights into the differences between the three kinds of ginseng polysaccharides which will help to further study the medicinal value of ginseng in traditional Chinese medicine.

Abstract:Inflammation and oxidative stress lead to various acute or chronic diseases, including pneumonia, liver and kidney injury, cardiovascular and cerebrovascular diseases, metabolic diseases, and cancer. Ginseng is a well-known and widely used ethnic medicine in Asian countries, and ginsenoside Rg3 is a saponin isolated from Panax ginseng C. A. Meyer, Panax notoginseng, or Panax quinquefolius L. This compound has a wide range of pharmacological properties, including antioxidant and anti-inflammatory activities, which have been evaluated in disease models of inflammation and oxidative stress. Rg3 can attenuate lung inflammation, prevent liver and kidney function damage, mitigate neuroinflammation, prevent cerebral and myocardial ischemia–reperfusion injury, and improve hypertension and diabetes symptoms. The multitarget, multipathway mechanisms of action of Rg3 have been gradually deciphered. This review summarizes the existing knowledge on the anti-inflammatory and antioxidant effects and underlying molecular mechanisms of ginsenoside Rg3, suggesting that ginsenoside Rg3 may be a promising candidate drug for the treatment of diseases with inflammatory and oxidative stress conditions.

Abstract:The major commercial ginsengs are Panax ginseng Meyer (Korean ginseng), P. quinquifolium L. (American ginseng), and P. notoginseng (Burk.) FH Chen (Notoginseng). P. ginseng is the most commonly used as an adaptogenic agent and has been shown to enhance physical performance, promote vitality, increase resistance to stress and aging, and have immunomodulatory activity. These ginsengs contain saponins, which can be classified as dammarane-type, ocotillol-type and oleanane-type oligoglycosides, and polysaccharides as main constituents. Dammarane ginsenosides are transformed into compounds such as the ginsenosides Rg3, Rg5, and Rk1 by steaming and heating and are metabolized into metabolites such as compound K, ginsenoside Rh1, protoand panaxatriol by intestinal microflora. These metabolites are nonpolar, pharmacologically active and easily absorbed from the gastrointestinal tract. However, the activities metabolizing these constituents into bioactive compounds differ significantly among individuals because all individuals possess characteristic indigenous strains of intestinal bacteria. To overcome this difference, ginsengs fermented with enzymes or microbes have been developed.

[Non-cerebrovascular complication in chirotherapy manipulation of the cervical vertebrae].

Abstract:PURPOSE Chirotherapy is a popular and successful management option for reversible functional disorders of the cervical spine. Though rarely observed, complications do occur, mainly involving the cerebrovascular system. By means of the here described case and a literature survey, we aim to highlight non-cerebrovascular complications of chirotherapeutic cervical spine manipulation. RESULTS A 43-year-old male initially consulted an ENT specialist, suffering from tinnitus aurium and loss of hearing ability. His hearing significantly increased after intravenous drug therapy, but the tinnitus remained. During chiropractic manipulation of the cervical spine by an orthopaedic surgeon for the tinnitus, the patient described severe neck pain following a clearly audible clicking sound. Scans of the cervical spine prior to and after manipulation showed an intracapsular/intraosseus oedema of the facet joints C2/C3 with lesion of the nerve root C3, most probably induced by chirotherapy. CONCLUSION Although complications after chiropractic manipulation are extremely rare, treatment of the spine, especially the cervical spine, is not wholly harmless. An adequate history taking followed by clinical and radiographic patient evaluation is necessary to keep the risk of iatrogenic trauma at a minimum. Above all, the chiropractic manipulation of the cervical spine belongs in the hands of a qualified and experienced medical practitioner.
Anti-Inflammatory Activity of Vacuum Distillate from Panax ginseng Root on LPS-Induced RAW264.7 Cells

Abstract:Panax ginseng has been widely applied as an important herb in traditional medicine to treat numerous human disorders. However, the inflammatory regulation effect of P. ginseng distillate (GSD) has not yet been fully assessed. To determine whether GSD can ameliorate inflammatory processes, a GSD was prepared using the vacuum distillation process for the first time, and the regulation effect on lipopolysaccharide-induced macrophages was assessed. The results showed that GSD effectively inhibited nitric oxide (NO) formation and activation of inducible nitric oxide synthase (iNOS) mRNA in murine macrophage cell, but not cyclooxygenase-2 production. The mRNA expression pattern of tumor necrosis factor alpha and IL-6 were also reduced by GSD. Furthermore, we confirmed that GSD exerted its anti-inflammatory effects by downregulating c-Jun NH2-terminal kinase (JNK) phosphorylation, the extracellular signal-regulated kinase phosphorylation, and signaling pathway of nuclear factor kappa B (NF-κB). Our findings revealed that the inflammatory regulation activity of GSD could be induced by iNOS and NO formation inhibition mediated by regulation of nuclear factor kappa B and p38/JNK MAPK pathways.

Pain Sensitivity and Pain Reactivity of Pericranial Muscles in Migraine and Tension-Type Headache

Abstract:We investigated whether experimentally determined, suprathreshold pain sensitivity of pericranial musculature in patients with tension-type headache differs from that of migraine patients or from that of healthy subjects. Furthermore, we looked to see whether differences could be found in the effects of experimental pain induction on EMG activity of pericranial musculature and whether subgroups could be discovered with higher and lower pericranial pain sensitivity within the three diagnostic groups in terms of neurophysiological, psychological and clinical variables. In 20 patients with tension-type headache, 23 patients with migraine without aura, and 29 healthy individuals experimental pain was induced in the temporal muscle by mechanical pressure; pain sensitivity in the entire metrically subdivided suprathreshold pain sensitivity range was measured. Surface EMG activity of pericranial muscles was determined before, during and after experimental pain induction. In addition, headache characteristics as well as personality and mood states were determined and recorded in a standardized fashion. There were no significant differences in pain sensitivity of pericranial musculature between the three groups. Patients with tension-type headache showed significantly higher EMG scores during suprathreshold pain stimulation than did migraine patients. EMG scores of healthy subjects fell between these two groups. With respect to pericranial tenderness significant differences in clinical, neurophysiological and psychological variables were found only between subgroups within the group of patients with tension-type headache. The results indicate that significant differences in the examined groups are found not in pain perception but in the processing or reaction to experimental headache stimuli. In patients with tension-type headache subgroups evolve based on pericranial pain sensitivity with quantitatively and/or qualitatively impaired reactions; for this reason diagnostic grouping according to the IHS classification seems to be pathophysiologically relevant. The intraindividual phasic comparison of pain reactions appears to be more important than the absolute interindividual tonic comparison.
