Abstract:The estimation of the postmortem interval for skeletal remains is a crucial aspect of forensic anthropology. This paper illustrates the importance of radiocarbon analysis for establishing medico-legal significance and supporting forensic identification, through the analysis of three case studies for which the years of both birth and death were investigated. In Audresselles, Northern France, a partial skull was discovered with no contextual information or identity. Radiocarbon dating yielded an average calibrated calendar age of 4232 BCE (92.5% probability), indicating significant archaeological value but no forensic relevance. In the second case, skeletal remains were found in the flooded underground of a historical fort at Wimereux, Northern France, also with no identity. Radiocarbon dating based on the bomb-pulse curve indicated a calibrated date of death in 1962 CE (37.3% probability) or 1974-1975 CE (58.1% probability), both surpassing the French statute of limitations. Lastly, a skeleton with a suspected identity was discovered near Valenciennes, Northern France, and various biological tissues underwent radiocarbon dating. A bone sample suggested a calibrated date of death of 1998-2002 CE (84.6% probability), differing from a hair sample (2013-2018 CE, 83.3% probability) because of the slower bone tissue remodeling process. DNA analysis confirmed the person's identity, reported missing a decade prior to the discovery of the remains, following the alignment of the radiocarbon results with the individual's year of birth based on dental tissues and year of death. These case studies reveal that traditional radiocarbon dating and bomb-pulse dating are essential tools for estimating the postmortem interval, providing mutual benefits for archaeologists, forensic anthropologists, and the criminal justice system. Estimation of Age-at-Death Using Cortical Bone Histomorphometry of the Rib and Femur: A Validation Study on a British Population

Abstract:Histomorphometry constitutes a valuable tool for age estimation. Histological interpopulation variability has been shown to affect the accuracy of age estimation techniques and therefore validation studies are required to test the accuracy of the pre-existing methodologies. The present research constitutes a validation study of widely known histological methods on the sixth rib and the femoral midshaft of a 19th century British population originating from Blackburn, England. An evaluation of the histomorphometric features of eleven ribs and five femora was performed and used to test the accuracy of selected methods. Results indicated that osteon area and circularity were the only histomorphometric variables that presented significant interpopulation variability. Cho et al.'s method for the ribs and the average value produced using Kerley and Ubelaker's method for intact osteon and percentage of lamellar bone equations for femur were considered the only reliable markers for estimating the age on the Blackburn sample. In the case of old individuals, Goliath et al.'s method provided more satisfactory results. Overall, the present study provides evidence on the applicability of the aging histomorphometric methods on a British sample and highlights the limitations of applying histomorphometric methods developed on different reference populations than the one under investigation.
Influence of Propranolol on the Ventricular Depolarization Gradient

Abstract:Sensing of the ventricular depolarization gradient (VDG) has recently been used as The basis of a closed‐loop rate responsive pacemaker. Factors influencing this aspect of the evoked response have not been fully evaluated although previous reports have suggested that sympathetic stimulation and circulating catecholamines are primarily responsible for the observed changes during stress and exercise. In five patients (Table I), four males and one female (mean age 60.4 ± 20.1 years) implanted with the Prism pacemaker, the pacing response to exercise and tilting was assessed before and after the infusion of propranolol. There was an increase in the pacing rate in all patients during the infusion of the drug (mean 27 ± 12.9 beats/min) suggestive of a direct drug effect on the VDG. The rate control parameter (RCP) of the pacemaker, the numerical equivalent of the VDG, was significantly different after the administration of propranolol (P < 0.01). However, exercise performance and pacing rate behavior were not different after beta blockade. The pacing rate increase observed when tilting patients to the supine position was not altered by propranolol. Out date suggest that factors other than adrenergic stimulation may be of importance in affecting the ventricular evoked response and accordingly the rate adaptation of the Prism pacemaker.
Quantifying cortical bone in fragmentary archeological second metacarpals

Abstract:Objectives: Skeletal variation in cortical bone thickness is an indicator of bone quality and health in archeological populations. Second metacarpal radiogrammetry, which measures cortical thickness at the shaft midpoint, is traditionally used to evaluate bone loss in bioarcheological and some clinical contexts. However fragmentary elements are regularly omitted because the midpoint cannot be determined. This methodological limitation reduces sample sizes and biases them against individuals prone to fracture, such as older individuals with low bone mass. This study introduces a new technique for measuring cortical bone in second metacarpals, the "Region of Interest" (ROI) method, which quantifies bone in archeological remains with less-than-ideal preservation while accounting for cortical heterogeneity. Materials and methods: The ROI method was adapted from digital X-ray radiogrammetry (DXR), a clinical method used to estimate bone mineral density, and tested using second metacarpals from Middenbeemster, Netherlands, a 19th century known age and sex skeletal collection. The ROI method quantifies cortical bone area within a 1.9 cm-long, mid-diaphyseal region, standardized for body size differences using total area (CAIROI ). CAIROI values were compared to traditional radiogrammetric cortical indices (CI) to assess the method's ability to identify age-related bone loss. Results: CAIROI values have high intra- and interobserver replicability and are strongly and significantly correlated with CI values for both males (r[n = 39] = 0.906, p = 0.000) and females (r[n = 58] = 0.925, p = 0.000). Conclusion: The ROI method complements traditional radiogrammetry analyses and provides a reliable way to quantify cortical bone in incomplete second metacarpals, thereby maximizing sample sizes, allowing patterns in bone acquisition and loss to be more comprehensively depicted in archeological assemblages.

Abstract:Age-at-death estimation is an important issue in forensic medicine and anthropology. Initially, methods relied on morphological criteria, but with the advancement of radiology, new techniques such as morphological studies on multi-slice computed tomography (CT) reconstructions have emerged. Recent studies have shown promising results by investigating the correlation between age and bone mineral density (BMD). However, there is currently a lack of data on post-mortem CTs (PMCT) involving decomposed bodies, and limited information exists regarding changes in Hounsfield Units measurement in a post-mortem context. In light of these gaps, our study aimed to examine the relationship between age at death and pubic and ilium BMD using a sample of forensic bodies. We also aimed to determine whether post-mortem processes, such as putrefaction, could interfere with this correlation. Our retrospective analysis encompassed 637 PMCTs conducted before medicolegal autopsies at Tours University Hospital. Utilizing simple and multiple linear regressions, we investigated the correlation between age and pubic and ilium BMD, as well as the relationship between BMD and the radiologic alteration index (RAI), a scale employed to quantify the degree of putrefaction. Our findings indicate promising outcomes in age-at-death estimation using pubic and/or ilium BMD for bodies exhibiting no or moderate decomposition (RAI < 80), particularly among individuals under 40 years old. However, for highly decomposed corpses (RAI ≥ 80), the presence of gas infiltration significantly influences the BMD of both the ilium and pubis. Consequently, we advocate for the incorporation of the RAI score into the age estimation equation to enhance the accuracy of our results in such cases. Further investigation involving a larger cohort of decomposed bodies could facilitate refinement and validation of our method within this specific population.

Abstract:Claims of extreme survival of DNA have emphasized the need for reliable models of DNA degradation through time. By analysing mitochondrial DNA (mtDNA) from 158 radiocarbon-dated bones of the extinct New Zealand moa, we confirm empirically a long-hypothesized exponential decay relationship. The average DNA half-life within this geographically constrained fossil assemblage was estimated to be 521 years for a 242 bp mtDNA sequence, corresponding to a per nucleotide fragmentation rate (k) of 5.50 x 10(-6) per year. With an effective burial temperature of 13.18 degrees C, the rate is almost 400 times slower than predicted from published kinetic data of in vitro DNA depurination at pH 5. Although best described by an exponential model (R-2 = 0.39), considerable sample-to-sample variance in DNA preservation could not be accounted for by geologic age. This variation likely derives from differences in taphonomy and bone diagenesis, which have confounded previous, less spatially constrained attempts to study DNA decay kinetics. Lastly, by calculating DNA fragmentation rates on Illumina HiSeq data, we show that nuclear DNA has degraded at least twice as fast as mtDNA. These results provide a baseline for predicting long-term DNA survival in bone.
Age‐dependent change and intraskeletal variability in secondary osteons of elderly Australians

Abstract:This study evaluated age‐related histomorphometric changes (osteon population density (OPD), osteon area (On.Ar), and Haversian canal area (H.Ar)) in three different bones intra‐individually in a modern Australian sample. Femur and humerus histology data were adjusted with robusticity indices, and our findings demonstrate that bone size influences histomorphometry, and this could confound age‐at‐death estimations that have not been adjusted for robusticity. There is a need to fully understand intra‐skeletal variability within different populations to develop and improve age‐at‐death estimation methods. This study evaluates age‐related histomorphometric changes in three different bones intra‐individually in a modern Australian sample. Four female and 13 male elderly Australian adult donors (67–93 years) were examined for osteon population density (OPD), osteon area (On.Ar), and Haversian canal area (H.Ar) of secondary osteons to compare between femora, ribs, and humeri and assess against age. In the pooled sex sample, no statistically significant correlations were observed between age and each histological variable. In the males, OPD of the femur increased significantly with age, as did porosity in the rib. In the male humeri, OPD increased moderately with age, while H.Ar was decreased moderately with age. Intra‐bone comparisons showed that males had significantly higher osteon counts in their ribs compared to their femora, while their ribs showed statistically significantly less porosity than their humeri. When bone size was accounted for, by adjusting the femur and humerus histology data by robusticity indices, histology values were found to be similar between bones within the same individual. This is despite the upper and lower limbs receiving different ranges and types of biomechanical load. Our findings demonstrate that bone size influences histomorphometry, and this could confound age‐at‐death estimations that have not been adjusted for robusticity. Future studies would benefit from examining bone histomorphometry within a larger sample size and incorporating bone robusticity measures into histology analyses.

Abstract:Ancient genome analysis has become an indispensable tool in studies of human population history and evolution since the breakthrough of whole-genome sequencing technology. The problem remains, however, that ancient genomes cannot be analyzed without crushing non-small pieces of precious specimens; moreover, in many cases, there is insufficient DNA remaining in the pieces of sample to obtain whole-genome sequences. In previous studies, therefore, a couple of indicators (e.g. racemization ratios) have been proposed to estimate the endogenous DNA in ancient samples. However, these studies have used polymerase chain reaction (PCR) to test whether endogenous DNA remains, but this has proved inadequate because the success or failure of PCR amplification does not necessarily reflect the DNA remaining. To assess the amount of endogenous DNA, we use the ratios of reads generated by next-generation sequencing (NGS) mapped to the human reference genome sequence. We investigated 40 human remains excavated from three shell-mound sites of the late to final Jomon culture. The associations between the environmental/molecular factors and the mapping ratios (MRs) were examined. There were no significant associations between the environmental factors and MRs, or between the collagen residual ratios (CRRs) and the MRs. However, we found a significant association between CRRs in rib bones and MRs. The weight of bone required to measure residual collagen is much less than that required to obtain the DNA necessary for NGS analysis, and the process of measuring CRRs is always involved in dating. Hence, we propose the collagen in the ribs as a good indicator for successful ancient genome analyses.

Searching for alternative high DNA-yielding bone types for DNA analysis of aged skeletal remains

Abstract:The petrous bone contains significantly higher amounts of DNA than any other human bone. Because of highly destructive sampling and because it is not always part of the recovered remains, the need for alternative sources of DNA is important. To identify additional optimal bone types, petrous bones were compared to femurs, tali, and calcanei sampled from 66 adult skeletons from two distinct modern-era Christian cemeteries. An extraction method employing full demineralization was used to obtain DNA, real-time PCR quantification to ascertain DNA quantity and degradation, and a commercial forensic short tandem repeats (STR) PCR amplification kit to determine genetic profiles. Statistical analysis was performed to explore the differences in DNA yield, DNA degradation, and success of STR amplification. A systematic studies exploring intra-skeletal variability in DNA preservation including various excavation sites differing by time period and geographical position are rare, and the second part of the investigation was based on a comparison of both archaeological sites, which allowed us to compare the effect of different post-mortem intervals and environmental conditions on DNA preservation. The older burial site in Črnomelj was active between the 13th and 18th century, whereas the more recent Polje burial was in use from the 16th to 19th century, creating different temporal and geographical environments. Results for the Črnomelj burial site revealed that the petrous bone outperformed all other bone types studied, except the calcaneus. At the Polje archeological site calcanei, tali, and femurs yielded the same STR typing success as petrous bones. The results obtained highlight the importance of careful bone sample selection for DNA analysis of aged skeletal remains. In addition to petrous bones, calcanei were found to be an alternative source of DNA when older burial sites are investigated. When more recent burial sites are processed, calcanei, tali, and femurs should be sampled besides petrous bones, not only because they exhibited good performance, but also because of easier sampling and easier grinding in the case of trabecular bones. This study contributes valuable insights into the potential use of various skeletal types as a source of DNA for investigation of aged skeletal remains, and it offers practical implications for forensic and archaeological investigations.

Abstract:Correct age-at-death estimation in adult individuals is one of the challenges of forensic investigation. Forensic anthropology macroscopic techniques are non-invasive methods for this purpose. However, several methods need to be applied to accurately estimate age, and the difference between chronological and predictive age may still be around ±10 years. New research trends are focused on the inherent process of aging, which produces changes in tissues and organs at different biochemical levels. One of the oldest and most studied approaches in this field is aspartic acid racemization. The accuracy of this technique in age estimation has been widely demonstrated. However, only a few studies have assessed its accuracy in different populations. The aim of this research was to assess the accuracy of aspartic acid racemization in a Spanish sample and its applicability to forensic cases. Dentin from fifteen third molars from two Spanish populations (ages 19–70 years old) was isolated and D and L forms of aspartic acid were detected through GC/MS, according to a previous published protocol. D/L ratios were calculated and after the application of a regression analysis, a formula for age estimation was developed. The results were similar to previous studies, obtaining an R = 0.91 between racemization ratios and age and a mean absolute error (MAE) between chronological and predictive age of 5 years. These results were ratified by leave-one-out cross-validation, as well as the application of the formula to five teeth of a known age. Despite these promising results, this technique is not exempt from drawbacks; thus, further studies are required to apply this methodology to forensic cases and to combine it with forensic anthropology findings.

Microscopic characteristics of peri- and postmortem fracture surfaces

Abstract:This study investigated if microscopic surface features captured with a scanning electron microscope (SEM) effectively discriminate fracture timing. We hypothesized that microscopic fracture characteristics, including delamination, osteon pullout, and microcracks, may vary as bone elasticity decreases, elucidating perimortem and postmortem events more reliably than macroscopic analyses. Thirty-seven unembalmed, defleshed human femoral shafts from males (n=18) and females (n=2) aged 33-81 years were fractured at experimentally simulated postmortem intervals (PMIs) ranging from 1 to 60 warm weather days (250-40,600 ADH). A gravity convection oven was used to approximate tissue decomposition at 37 C and 27 C, and the resulting heat-time unit (accumulated degree hours, or ADH) was used to examine fractures in elastic/wet versus brittle/dry bone. The bones were fractured with a drop test frame using a three-point bending setup, sensors were used to calculate fracture energy, and high-speed photography documented fracture events. The following data were collected to relate fracture appearance to the biomechanical properties of bone: PMI (postmortem interval) length in ADH, temperature, humidity, collagen percentage, water loss, bone mineral density, cortical bone thickness, fracture energy, age, sex, cause of death, and microscopic fracture feature scores. SEM micrographs were collected from the primary tension zones of each fracture surface, and three microscopic fracture characteristics were scored from a region of interest in the center of the tension zone: percentage of delaminated osteons, percent osteon pullout, and number of microcracks. Multiple linear regression showed that microscopic fracture surface features are strong predictors of ADH (adjusted R-squared=0.67 for the 0 - 40,000 ADH samples; adjusted R-squared=0.92 for the 0-16,000 ADH samples). Osteon pullout is the single best predictor of ADH. Additionally, water loss is the primary driver of bone elasticity changes in low ADH samples, while collagen fibers appear to remain intact until later in the postmortem interval (approximately 40,000 ADH in this study). The results of this study indicate microscopic fracture surface analysis detects the biomechanical effects of decreased elasticity more reliably and with greater sensitivity than macroscopic analysis.
The Use of Informative Priors in Bayesian Modeling Age-at-death; a Quick Look at Chronological and Biological Age Changes in the Sacroiliac Joint in American Males

Abstract:The aim of this study is to examine how well different informative priors model age-at-death in Bayesian statistics, which will shed light on how the skeleton ages, particularly at the sacroiliac joint. Data from four samples were compared for their performance as informative priors for auricular surface age-at-death estimation: (1) American population from US Census data; (2) county data from the US Census data; (3) a local cemetery; and (4) a skeletal collection. The skeletal collection and cemetery are located within the county that was sampled. A Gompertz model was applied to compare survivorship across the four samples. Transition analysis parameters, coupled with the generated Gompertz parameters, were input into Bayes' theorem to generate highest posterior density ranges from posterior density functions. Transition analysis describes the age at which an individual transitions from one age phase to another. The result is age ranges that should describe the chronological age of 90% of the individuals who fall in a particular phase. Cumulative binomial tests indicate the method performed lower than 90% at capturing chronological age as assigned to a biological phase, despite wide age ranges at older ages. The samples performed similarly overall, despite small differences in survivorship. Collectively, these results show that as we age, the senescence pattern becomes more variable. More local samples performed better at describing the aging process than more general samples, which implies practitioners need to consider sample selection when using the literature to diagnose and work with patients with sacroiliac joint pain.

Abstract:Abstract Fracture risk increases with lower areal BMD (aBMD); however, aBMD-related estimate of risk may decrease with age. This may depend on technical limitations of 2-dimensional (2D) DXA which are reduced with 3D high-resolution peripheral quantitative computed tomography (HR-pQCT). Our aim was to examine whether the predictive utility of HR-pQCT measures with fracture varies with age. We analyzed associations of HR-pQCT measures at the distal radius and distal tibia with two outcomes: incident fractures and major osteoporotic fractures. We censored follow-up time at first fracture, death, last contact or 8 years after baseline. We estimated hazard ratios (HR) and 95%CI for the association between bone traits and fracture incidence across age quintiles. Among 6835 men and women (ages 40-96) with at least one valid baseline HR-pQCT scan who were followed prospectively for a median of 48.3 months, 681 sustained fractures. After adjustment for confounders, bone parameters at both the radius and tibia were associated with higher fracture risk. The estimated HRs for fracture did not vary significantly across age quintiles for any HR-pQCT parameter measured at either the radius or tibia. In this large cohort, the homogeneity of the associations between the HR-pQCT measures and fracture risk across age groups persisted for all fractures and for major osteoporotic fractures. The patterns were similar regardless of the HR-pQCT measure, the type of fracture, or the statistical models. The stability of the associations between HR-pQCT measures and fracture over a broad age range shows that bone deficits or low volumetric density remain major determinants of fracture risk regardless of age group. The lower risk for fractures across measures of aBMD in older adults in other studies may be related to factors which interfere with DXA but not with HR-pQCT measures.

Abstract:The use of diagenetic alterations in bone microstructure ('histotaphonomy') as indicators of funerary treatment in the past and for post-mortem interval calculations in forensic cases has received increasing attention in the last decade. Studies have used histological changes to conclude in-situ decomposition, mummification, infanticide and post-mortem interval. There has been very little attempt to experimentally validate the links between decomposition, depositional conditions, time-since-death and microscopic changes in human bone so that meaningful interpretations of archaeological and forensic observations can be made. Here, we address this problem experimentally using the largest sample of human remains from anatomical donors and the longest-term deposition framework to date. This study tests one key assumption of histotaphonomy; that putrefaction during the early stages of decay is reflected in bone microanatomy and composition. Seventeen human donors and six pigs were deposited on the surface in a known Australian environment and left to decompose between 463 and 1238 days. All remains underwent all stages of decomposition reaching skeletonisation. Rib and femur samples were analysed using conventional histological methods and scanning electron microscopy, by applying the Oxford Histological Index, and examining collagen birefringence, microcracking and re- and de mineralisation. Biomolecular changes of the femoral samples were analysed using Fourier-transform infrared (FTIR) spectroscopy. The results indicate that bioerosion in human bone does not occur due to putrefaction. There were no correlations between bone histology and the following variables: human vs pigs, season, primary vs secondary deposition, position, fresh vs frozen and time-since-deposition. Furthermore, no trends were observed between biomolecular changes and time-since-deposition. The study also shows that pigs cannot be used as substitutes for human remains for bone biodegradation research. This is the first, controlled, larger scale study of human remains providing a lack of support for a long-assumed relationship between putrefaction and bone histology bioerosion. Using bone degradation as an argument to prove putrefaction, in-situ decomposition and early taphonomic processes cannot be supported based on the experimental human data presented.
A Novel HR‐pQCT Image Registration Approach Reveals Sex‐Specific Changes in Cortical Bone Retraction With Aging

Abstract:During aging, changes in endosteal and periosteal boundaries of cortical bone occur that differ between men and women. We here develop a new procedure that uses high‐resolution peripheral quantitative CT (HR‐pQCT) imaging and 3D registration to identify such changes within the timescale of longitudinal studies. A first goal was to test the sensitivity of the approach. A second goal was to assess differences in periosteal/endosteal expansion over time between men and women.Rigid 3D registration was used to transform baseline and all follow‐up (FU) images to a common reference configuration for which the region consisting of complete slices (largest common height) was determined. Periosteal and endosteal contours were transformed to the reference position to determine the net periosteal and endosteal expansion distances. To test the sensitivity, images from a short‐term reproducibility study were used (15 female, 21‐47 years, scanned 3 times). To test differences between men and women, images from a subset of the Geneva Retirees Cohort were used (248 female, 61 male, average age 65 years, 3.5 and 7 years FU).The sensitivity study indicated a least significant change for detecting periosteal/endosteal expansion of 41/31 microns for the radius and 17/26 microns for the tibia. Results of the cohort study showed significant net endosteal retraction only in females at the radius and tibia after 3.5 years (38.0 and 38.4 microns, respectively) that further increased at 7 years FU (70.4 and 70.8 microns, respectively). No significant net periosteal changes were found for males or females at 7 years.The results demonstrate that it is possible to measure changes in endosteal contours in longitudinal studies within several years. For the investigated cohort, significant endosteal retraction was found in females but not in males. Whether these changes in cortical geometry are related to fracture risk remains to be investigated in larger cohorts.

Abstract:Radiology plays a crucial role in forensic anthropology for age estimation. However, most studies rely on morphological methods. This study aims to investigate the feasibility of using pubic bone mineral density (BMD) as a new age estimation method in the Chinese population. 468 pubic bone CT scans from living individuals in a Chinese hospital aged 18 to 87 years old were used to measure pubic BMD. The BMD of the bilateral pubic bone was measured using the Mimics software on cross-sectional CT images and the mean BMD of the bilateral pubic bone was also calculated. Regression analysis was performed to assess the correlation between pubic BMD and chronological age and to develop mathematical models for age estimation. We evaluated the accuracy of the best regression model using an independent validation sample by calculating the mean absolute error (MAE). Among all established models, the cubic regression model had the highest R 2 value in both genders, with R 2 = 0.550 for males and R 2 = 0.634 for females. The results of the best model test showed that the MAE for predicting age using pubic BMD was 8.66 years in males and 7.69 years in females. This study highlights the potential of pubic BMD as a useful objective indicator for adult age estimation and could be used as an alternative in forensic practice when other better indicators are lacking.

Assessing the usefulness of Raman spectroscopy and lipid analysis of decomposed human bones in forensic genetics and molecular taphonomy

Abstract:Bones are among the structures most likely to be recovered after death. However, the low quantity of preserved DNA and complex processing from sample to DNA profile make forensic DNA analysis of bones a challenging task. Raman spectroscopy and gas chromatography-mass spectrometry (GC/MS), have the potential to be useful as screening tools for DNA analysis and in decomposition studies. The objective of this research was to assess the usefulness of such molecular investigations. Femur samples collected from 50 decomposing human bodies were subjected to Raman spectroscopy and GC/MS. Assessment of nuclear DNA quantity and short tandem repeat (STR) genotyping efficiency were also performed. Raman parameters (crystallinity, carbonate-to-phosphate ratio, mineral-to-matrix ratio) and detected lipids were recorded. Background fluorescence proved problematic for Raman analysis of forensic bones. Regardless, it was not associated with less preserved DNA or less detected STR alleles. Fatty acids, hydrocarbons, and five types of fatty acid methyl esters (FAMEs) were detected. The main phosphate peak position in Raman spectra was significantly correlated with preserved DNA (p = 0.03713), while significantly more STR alleles were detected in bones containing methyl hexadecenoate (p = 0.04236). Detection of FAMEs in the bone matrix suggests a reaction between methanol produced by bacteria and free fatty acids, which are not associated with the level of preservation of endogenous DNA. The techniques assessed have shown to be useful in molecular taphonomy studies and forensic genetics.
Forensic age estimation of the knee by post-mortem DR, CT, and MR imaging: a comparative study

Abstract:It is believed by many that reference data for age estimation purposes must be imaging-modality specific. A study from our department has however proven otherwise. We therefore found it interesting to investigate this further by looking at the level of agreement between different imaging modalities. The aim of this study was to investigate the level of agreement between the three radiological modalities, computed tomography (CT), magnetic resonance imaging (MRI), and digital radiography (DR), in assessing the ossification of the epiphyses of the knee. A total of 34 deceased individuals of 10–25 years of age, brought in for a medicolegal autopsy at our department, were scanned by CT, MRI, and DR. The ossification stages of the three bones of the right knee, distal femoral, proximal tibial, and proximal fibular epiphysis were assessed using the established combined staging method by Schmeling et al. and Kellinghaus et al. Analysis of the results by Cohen's weighted kappa showed a good agreement between CT and DR ( K = 0.61–0.70), and MRI and DR ( K = 0.68–0.79) but only moderate agreement between CT and MRI ( K = 0.55–0.57). This leads us to conclude that different radiological images cannot be used interchangeably for age estimation purposes, so reference material needs to be imaging-modality specific. However, to make a more general conclusion research on a larger population is needed.

Abstract:A critical component of the biological profile involves accurate estimation of the age‐at‐death of the decedent(s). While bias, inaccuracy, and population specificity have plagued age estimation methods, these issues are minimized by Bayesian statistics. Our primary analysis generated ages of transition from the published Suchey‐Brooks pubic symphysis dataset and coupled them with informative priors derived from two modern American forensic samples (Forensic Data Bank [FDB] and Forensic Anthropology Database for Assessing Methods Accuracy [FADAMA]) to test the accuracy of Bayesian analysis against the original method. Exact binomial tests assessed the accuracy of the generated age ranges; realized accuracies and bias are reported for final age ranges at various coverages. In a second analysis, the Bayesian Suchey‐Brooks parameters and original, non‐Bayesian age ranges were also applied to FADAMA. In the primary analyses, the Bayesian approach improved age estimates over traditional ranges, especially for females. Highest posterior density ranges at 95% provided realized accuracies on a holdout sample between 93% and 96% with extremely low bias for most phases. We provide lookup tables with Bayesian age ranges for various coverages. In the second analysis, realized accuracies were slightly higher in the non‐Bayesian approach for both sexes (86%–92% vs. 83%–91%), due to lower precision and likely practitioner bias in aging forensic cases. The popularity of the Suchey‐Brooks pubic symphysis aging method in modern forensic casework necessitates the use of the Bayesian approach and we encourage practitioners to utilize the lookup tables for forensic casework in the United States.
