Abstract:The objective of the article was to study the effects of ultrasound (US) and 80 ppm nisin-assisted US (US + nisin) on the inactivation of microorganisms and the nutritional quality of fresh carrot juice. Fresh carrot juice was exposed to HT (60 °C, 10 min), US, and US + nisin at 271 W and 30 °C for 10 min since the treatments had a similar microbial inactivation effect. Results showed that nisin exhibited an additional inactivation effect on the total aerobic bacteria (TAB) with US, while having no obvious effect on the yeast and mold (YM). The TAB was increased by 2.56, 2.20, and 4.28 log10 cycles, while the YM was increased by 4.67, 4.75, and 6.84 log10 cycles in carrot juice treated by US, US + nisin, and HT, respectively, after 20 days' storage at 4 °C, indicating that US + nisin had a better post-inhibition effect on TAB than US and HT. Moreover, no significant changes occurred in the total suspended solids (TSS), viscosity, turbidity, or color, while carotenoid content was increased by 22.4%, 34.6%, and 21.4% in the carrot juice treated by US, US + nisin, and HT, respectively. In conclusion, the study ascertained that US + nisin is able to ensure both the microbial safety and the quality of carrot juice more effectively than HT alone.

Investigation on the inactivation effect and mechanism of Listeria monocytogenes in fresh-cut cucumber during storage by ultrasound combined with sodium hypochlorite

Abstract:Fresh agricultural products are frequently contaminated with Listeria monocytogenes (L. monocytogenes), which threatens consumer health. The mechanism of the inhibitory effect of ultrasound and sodium hypochlorite (US-NaClO) on L. monocytogenes on fresh-cut cucumber remains poorly understood. Therefore, the bactericidal ability and mechanism of US-NaClO treatment on L. monocytogenes were studied on fresh-cut cucumber during storage using various approaches such as determination of intracellular material leakage, scanning electron microscopy, flow cytometry, and expression analysis of virulence genes. The results showed that the number of L. monocytogenes on fresh-cut cucumber was significantly reduced after ultrasound treatment for 5 min in combined with 75 ppm sodium hypochlorite treatment(P < 0.05). The US-NaClO treatment affected cell morphology, impaired cell membrane integrity, increased cell membrane permeability, and reduced the concentration of K+, inorganic phosphate, ATP, proteins, and DNA in bacterial cells, leading to the inactivation of microorganisms. In addition, the US-NaClO treatment downregulated expression of the virulence genes actA, hly, inlA, mpl, pclA, and plcB, thus decreasing the pathogenicity of bacteria. It can avoid contamination by pathogenic bacteria during the production of fresh-cut cucumber, while providing safety assurance for production.
Combined Effect of High Pressure and Slightly Acidic Electrolyzed Water on Sterilization of Fresh-cut Fruits and Vegetables

Abstract:Due to the operation of cutting,fresh-cut fruits and vegetables are susceptible to microbial infection.In order to extend the shelf life of fresh-cut fruits and vegetables,the effects of the concentration and amount of available chlorine of slightly acidic electrolyzed water (SAEW) and the pressure level and holding time of high pressure (HP) on sterilization of fresh-cut carrots were studied through single factor experiment.Consequently,the combination treatment conditions were as follows:the concentration of available chlorine of slightly acidic electrolyzed water was 30 mg/L and the amount of available chlorine of slightly acidic electrolyzed water was 200 mL;the pressure level ranged from 100 MPa to 400 MPa,holding time was 5 min and the total processing time was 15 min.Fresh-cut carrots and apples were treated by the different combined treatments,and both brain heart infusion agar (BHIA) and vilolet red bile agar (VRBA) were used for the detection of E.coli.With the aim to explore the inactivation effects of combination treatment,during the detection,the slightly acidic electrolyzed water was chosen instead of HP sterilized water to do further test.The result shows that combined treatment can improve the efficiency of inactivation,but the enhancement was not significant at lower pressure.At 400 MPa,HP treatment combined with SAEW displayed the most significant enhancement of inactivation efficiency,where E.coli could not be detected on fresh-cut carrot,but some E.coli was still detected on the fresh-cut apples in BHIA.When comparing the results in BHIA and VRBA,it can be seen that the SAEW showed significant lethal effect on E.coli,while HP showed both lethal and sublethal effect on E.coli.

Abstract:The influence of ultrasound combined with sodium hypochlorite (US-NaClO) treatment on microorganisms and quality of fresh-cut cucumber during storage were investigated. Ultrasound (400 W, 40 kHz, US: 5, 10 and 15 min) and sodium hypochlorite (NaClO: 50, 75, 100 ppm) were used to treat fresh-cut cucumber in a single or combined treatment and stored at 4 °C for 8 days and analyzed for texture, color and flavor. The results showed that US-NaClO treatment had a synergistic effect on the inhibition of microorganisms during storage. It could significantly reduce (p < 0.05) the number of microorganisms by 1.73 to 2.17 log CFU/g. In addition, US-NaClO treatment reduced the accumulation of malondialdehyde (MDA) during storage (4.42 nmol/g) and water mobility, and maintained the integrity of the cell membrane, delayed the increase of weight loss (3.21%), reduced water loss, thus slowing down the decline of firmness (9.20%) of fresh-cut cucumber during storage. The degradation of chlorophyll (6.41%) was reduced to maintain the color of freshly cut cucumbers. At the same time, US-NaClO could maintain the content of aldehydes, the main aromatic substance of cucumber, and reduced the content of alcohols and ketones during storage. Combined with the electronic nose results, it could maintain the cucumber flavor at the end of the storage period and reduce the odor produced by microorganisms. Overall, US-NaClO was helpful to inhibit the growth of microorganisms during storage, improve the quality of fresh-cut cucumber.

Abstract:The present study investigated influences of ultraviolet light C (UV-C; 2 kJ m−2) irradiation and high-oxygen modified atmosphere packaging (MAP; 80 % O2, 10 % CO2 and 10 % N2) on physiological quality, microbial growth and lignification metabolism of fresh-cut carrots. After 15-d cold storage, comparing with either treatment alone, the combination of UV-C irradiation and high-oxygen MAP (abbr. UV-C + MAP) more obviously inhibited firmness, weight loss, ascorbic acid, total carotenoid and γ-aminobutyric acid declines, reduced respiration and ethylene production rates as well as delayed bacterial growth. UV-C + MAP more strongly restrained whiteness index, total phenolic, lignin and malondialdehyde increases. Furthermore, UV-C + MAP treatment more efficiently retarded lignin synthesis by suppressing phenolic metabolism-related enzyme (phenylalanine ammonialyase, PAL; polyphenoloxidase, PPO; peroxidase, POD) activities and their gene (DcPAL; DcPPO; DcPOD) expressions. Above results indicated that UV-C + MAP exhibited synergistic effects in retaining physiological quality, delayed senescence process, reducing microbial growth, alleviating lignification degree, and lessened surface whitening. Therefore, UV-C + MAP could maintain overall quality and extend shelf-life during storage of fresh-cut carrots.

Abstract:Influence of the combined effect of ultra-sonication (US) and high-voltage cold plasma treatment (HVCP) on the quality parameters of fresh carrot juice has been studied. During the treatment of ultra-sonication, carrot juice was subjected to a 0.5 inch probe for 3 min by adjusting the pulse duration 5 s on and off at 20 kHz frequency, amplitude level 80%. The ultrasound intensity was measured by using a thermocouple and was 46 Wcm−2. The temperature was maintained at 10 °C by an automatic control unit. During the treatment of HVCP, carrot juice was then subjected to dielectric barrier discharge (DBD) plasma discharge at 70 kV voltage for 4 min. Significant increases were observed when HVCP treated carrot juice was tested against total carotenoids, lycopene, and lutein when compared to the control treatments. Moreover, this increase was raised to its highest in all pigments, chlorogenic acid, sugar contents, and mineral profile, as the results of ultra-sonication when combined with high voltage atmospheric cold plasma (US-HVCP). Whereas, a significant decreased was observed in Mg, total plate count, yeast, and mold after US-HVCP treatment. Furthermore, results indicated that the combined effect of US-HVCP treatment has improved the quality and led to a higher concentration of lycopene, lutein, chlorogenic acid, and mineral compounds (Na, K, and P). Therefore, the findings of the current study suggested that US-HVCP treatment is a novel combined technique that could provide better quality and more stability during the processing of carrot juice with better physicochemical properties and bio-available nutrients, so this novel processing technique could serve as an alternative to traditional processes.
The effect of high-power ultrasound on the quality of carrot juice

Abstract:The effect of high-power ultrasound treatment on enzymes' activity, physicochemical attributes (total soluble solids, pH, viscosity, turbidity, particle size distribution and colour) and carotenoids' content of carrot juice was investigated. The treatments were carried out at 20 kHz (0.95, 2.38, 3.80 W/ml power) in an ice bath for 2, 4, 6, 8, 10 min. The polyphenol oxidase and pectin methylesterase activity were decreased by 43.90 and 37.95% at 3.80 W/ml power and 10 min exposure time, respectively. With the increase of power and time, the effect of high-power ultrasound on the inactivation of enzymes was getting stronger. However, high-power ultrasound had no inactivation effect on peroxidase activity under all treatment conditions. The visual colour differences were not obvious after high-power ultrasound. The pH, total soluble solids and particle size distribution of carrot juice were not significantly affected (p > 0.05) under all treatment conditions, while turbidity was increased and carotenoids' content was decreased. The viscosity of carrot juice was decreased by 1.27% at 0.95 W/ml power and 8 min, while it was increased by 2.29% at 2.38 W/ml power and 8 min. The value of viscosity was negatively correlated with the activity of pectin methylesterase (Pearson's r = -0.481, p < 0.05). According to these results, we could conclude that the optimal treatment condition was 3.80 W/ml for 10 min. Overall, high-power ultrasound treatment inhibited browning, maintained taste and nutritional value and improved stability of carrot juice. Therefore, this technology could well be an option for processing of carrot juice and laid the theoretical foundation for the production of carrot juice and carrot compound beverage.

Abstract:The presence of Salmonella in nature poses a significant and unacceptable threat to the human public health domain. In this study, the antibacterial effect and mechanism of ultrasound (US) combined with Litsea cubeba essential oil nanoemulsion (LEON) on Salmonella. LEON + US treatment has a significant bactericidal effect on Salmonella. Reactive oxygen species (ROS), malondialdehyde (MDA) detection, N-phenyl-l-naphthylamine (NPN) uptake and nucleic acid release assays showed that LEON + US exacerbated cell membrane lipid peroxidation and increased the permeability of the cell membrane. The results of field emission scanning electron microscopy (FESEM), transmission electron microscopy (TEM) showed that LEON + US treatment was able to alter cell morphology. It can be observed by flow cytometry (FCM) that LEON + US treatment can cause cell apoptosis. In addition, bacterial counts of cherry tomatoes treated with LEON (0.08 μL/mL) + US (345 W/cm2) for 9 min were reduced by 6.50 ± 0.20 log CFU/mL. This study demonstrates that LEON + US treatment can be an effective way to improve the safety of fruits and vegetables in the food industry.

Abstract:This study investigated the antibacterial effect of ultrasound (US) combined with Litsea cubeba essential oil nanoemulsion (LEON) on Salmonella Typhimurium in kiwifruit juice and effect on the quality and sensory properties of kiwifruit juice. In this study, LEON prepared by ultrasonic emulsification method had a good particle size distribution and high stability. The US+LEON treatment significantly (P < 0.05) improved antibacterial efficacy, compared to the control, and would not destroy the nutritional components containing ascorbic acid, flavonoids, total phenol and total soluble solids. Meanwhile, US+LEON treatment enhanced 2, 2-diphenyl-1-picrylhydrazyl (DPPH), 2, 2'-azino-bis-(3-ethylbenzothiazoline-6 sulfonic acid) (ABTS) radical scavenging capacity and ferric ion reducing antioxidant power (FRAP). In terms of sensory properties, US and LEON had a significant (P < 0.05) effect on the odor and overall morphology of kiwifruit juice. The enhance of antibacterial efficacy and the retention of nutrients by combined treatments shows that US+LEON is a promising antibacterial method that will provide new ideas for the processing and safety of fruit juices, and the US parameters and LEON concentration should be adjusted to reduce the effect on food sensory properties in future studies.
Comparison of High- and Low- Frequency Thermosonication and Carvacrol Treatments of Carrot Juice: Microbial Inactivation and Quality Retention

Abstract:The current study evaluated the comparative effects of carvacrol (CRV) with low- (24 kHz, TS) or high- (861 kHz, HTS) frequency thermosonication on microbial inactivation and quality parameters of carrot juice. The combinations of mild heat, ultrasound and carvacrol proved to be synergistic against aerobic bacteria (AB), yeast and molds (Y&M) and were effective for improving quality characteristics of juice. The optimal condition was CRV+TS (13.3 W/mL, 2.6 µM, 52 °C, 40 min) and CRV+HTS (13.3 W/mL, 2.6 µM, 52 °C, 40 min), under these treatments, AB and Y&M were inactivated by 2.74±0.18 and 2.28±0.21 log, 4.71±0.00 and 2.40±0.09 log, respectively. Compared to CRV+TS at 52 °C, CRV+HTS could extend the shelf life of juice by 25 days at 6 °C, with lower microbial amount and higher sedimentation stability. Overall, this study demonstrated the potentials of the combined approaches based on thermosonication and carvacrol to produce juice with the desired microbiological safety, enhanced shelf life and excellent quality parameters.
Study on Combined Effects of Acidification and Sonication on Selected Quality Attributes of Carrot Juice During Storage

Abstract:This study evaluated the combined effects of acid blanching and sonication treatments on selected quality parameters of carrot juice stored at 4 degrees C for 18 days. Carrots were blanched in acidified water (40g/L citric acid) at 100 degrees C for 4 min and the juice was then extracted. Sonication of the juice was done at an amplitude level of 70% and a frequency of 20 kHz for 2 min at 15 degrees C, keeping the pulse duration of 5 Sec on and 5 Sec off. As results, the combined treatment of acidification and sonication of carrot juice showed a significant decrease in pH and increase (P < 0.05) in acidity which remained stable during storage period. No significant changes were observed in degrees Brix. Color values (L*, a*, b*) and non enzymatic browning (NEB) influenced significantly in acidified and sonicated carrot juice during storage period. Maximum stability of total phenol, total antioxidant capacity, cloud value and ascorbic acid were also observed in the combined treatment of acidification and sonication. The findings of this study indicated that the combined treatments of acidification and sonication may successfully be utilized for the production of high quality carrot juice with improved stability of total phenol, total antioxidant capacity, cloud value and ascorbic acid during 18 days of storage.
Control Measures of Pathogenic Microorganisms and Shelf-Life Extension of Fresh-Cut Vegetables

Abstract:We investigated the combined effect of using slightly acidic electrolyzed water (SAEW), ultrasounds (US), and ultraviolet-C light-emitting diodes (UV-C LED; 275 nm) for decreasing pathogenic Escherichia coli and Staphylococcus aureus (SEA) in fresh-cut vegetables, including carrots, celery, paprika, and cabbage. Survival of pathogenic E. coli and SEA and quality properties of fresh-cut vegetables at 5 and 15 °C for 7 days were also investigated. When combined treatment (SAEW + US + UV-C LED) was applied to fresh-cut vegetables for 3 min, its microbial reduction effect was significantly higher (0.97~2.17 log CFU/g) than a single treatment (p < 0.05). Overall, the reduction effect was more significant for SEA than for pathogenic E. coli. At 5 °C, SAEW + US and SAEW + US + UV-C LED treatments reduced populations of pathogenic E. coli and SEA in all vegetables. At 15 °C, SAEW + US + UV-C LED treatment inhibited the growth of both pathogens in carrot and celery and extended the shelf life of fresh-cut vegetables by preventing color changes in all vegetables. Although the effects of treatments varied depending on the characteristics of the vegetables and pathogens, UV-C LED can be suggested as a new hurdle technology in fresh-cut vegetable industry.

Sterilization of black carrot juice by thermosonication: Compound changes revealed by NMR and GC–MS metabolomics methods

Abstract:The effect of themosonication treatment on the quality of black carrot juice was studied by metabolomics approach. In this study, thermosonic sterilization (TS), ultrasonic sterilization (US), and heat sterilization at 60°C, 80°C, and 90°C were used for the bactericidal treatment of black carrot juice. The processed samples were analyzed in terms of physicochemical, biological activity, microbial, and metabolomics approaches. The main differential metabolites were identified after treatment. Among these, the content of the important chemical component anthocyanins increased by 10.64% and 6.82% in TS and US, respectively. The TS treatment showed better retention of most of the active substances such as carbohydrates, malic acid, alanine, sterols, lutein, and terpenes compared to the other treatments. Furthermore, the TS treatment reduced the ultrasonic power and time for inactivating microorganisms and increased the DPPH radical scavenging activity by 11.31% compared to the control. The experimental results showed that the black carrot juice after TS treatment with 60% ultrasonic power at 60°C for 5 min sterilization was more favorable for the retention of nutrients and volatiles, improving the quality, safety, and economic potential of black carrot juice. Practical applications This work for the first‐time applied TS to black carrot juice. The effect of TS treatment on the quality of black carrot juice was studied and compared with ultrasonic and conventional heat treatment by metabolomics approach. TS could significantly reduce US treatment time from 25 min (20°C) to 5 min (60°C) for the sterilization of juice. TS has a better effect on the retention of nutrients and flavor substances in the juice. The study is beneficial to the application of TS in purple carrot juice sterilization, also the application of 1H‐NMR and GC–MS metabolomics approach in food processing.

Abstract:The purpose of this study was to quantify the effectiveness of ultrasound treatment combined with controlled atmosphere storage (UCA) in preserving quality of cucumbers (Cucumis sativus L.) compared with the controlled atmosphere storage (CA) alone. The combination of ultrasound (especially at 200 W for 10 min) with CA (5% O-2 + 2% CO2 + 93% N-2) storage was most effective in preserving the fresh-like quality of cucumbers. The treatment reduced the loss in mass, firmness, soluble solids content and color, maintained cell wall integrity, and limited the redistribution of water in cucumbers. UCA significantly better preserved the flavor volatiles and retained the taste characteristics. This method also better preserved the intermolecular hydrogen and carbonyl bonding in aldehyde and ketone compounds of cucumbers which helped them to preserved longer. These results suggested that UCA can be a much effective preservation method than CA alone for fruits and vegetables.

Abstract:The inhibition of Escherichia coli ATCC 25922 (E. coli), Staphylococcus aureus ATCC6538 (S. aureus), Salmonella Enteritidis ATCC 13076 (S. Enteritidis), and Listeria monocytogenes DSM12464 (L. monocytogenes) is one of the main aims of the food industry. This study was the first in which the use of ultrasound and microwave power were applied to optimize the values of the bioactive components, amino acids, and mineral compositions of tangerine juice and to inhibit Escherichia coli, Staphylococcus aureus, Salmonella Enteritidis, and Listeria monocytogenes. The response surface methodology (RSM) was used to describe the inactivation kinetics, and the effects of ultrasound treatment time (X1: 12–20 min), ultrasound amplitude (X2:60–100%), microwave treatment time (X3: 30–40 s), and microwave power (X4:200–700 W). The optimum parameters applied to a 5-log reduction in E. coli were determined as ultrasound (12 min, 60%) and microwave (34 s, 700 W). The optimum condition ultrasound–microwave treatment was highly effective in tangerine juice, achieving up to 5.27, 5.12, and 7.19 log reductions for S. aureus, S. Enteritidis, and L. monocytogenes, respectively. Ultrasound–microwave treatment increased the total phenolic compounds and total amino acids. While Cu, K, Mg, and Na contents were increased, Fe and Ca contents were lower in the UM-TJ (ultrasound–microwave-treated tangerine juice) sample. In this case, significant differences were detected in the color values of ultrasound–microwave-treated tangerine juice (UM-TJ) (p < 0.05). The results of this study showed that ultrasound–microwave treatment is a potential alternative processing and preservation technique for tangerine juice, resulting in no significant quality depreciation.

Abstract:Chitosan and organic acids are natural preservatives with proven antimicrobial efficacies. This study investigated the effects of chitosan coating combined with organic acids on inhibiting Listeria monocytogenes in ready-to-eat (RTE) shrimps during storage at 4°C. Cooked shrimps inoculated with L. monocytogenes were coated with 0.5 and 1 % chitosan solutions in combination with 2 % organic acids (acetic, citric, lactic, and malic acids) for 5 min and then stored at 4°C for 16 days. Bacterial counts, pH, and sensory properties were analyzed every 4 days. The results indicated that the antimicrobial effects of chitosan were significantly enhanced (P < 0.05) when it was combined with 2 % acetic, citric, lactic, or malic acid. Chitosan at 1 % combined with 2 % acetic acid was the most effective treatment, which caused a 5.38-log CFU/g bacterial reduction after 16 days in comparison with the controls. Transmission electron microscopy further confirmed the enhanced antimicrobial effects of the combination of chitosan and organic acids. Such combined treatments also maintained the sensory properties of RTE shrimps. The use of chitosan coating combined with organic acids significantly lowered (P < 0.05) the pH values of RTE shrimps compared with the use of chitosan alone. Therefore, we conclude that chitosan coating combined with acetic acid could be a promising antimicrobial method to prevent the proliferation of L. monocytogenes in RTE shrimps with extended shelf life.

Abstract:SummaryThe combined effects of ultrasound (US) and high hydrostatic pressure (HHP) on enzymes, micro‐organisms, colouring pigments, total polyphenols, ascorbic acid and physicochemical properties of carrot juice were investigated. Carrot juice was treated with sonication (20 kHz, 70% amplitude) and HHP (250, 350, 450 MPa) at room temperature for 10 min. For comparison, fresh carrots were water blanched (WB) at 100 °C for 4 min and carrot juice was then extracted. As results, highest improvements in colouring pigments were observed in WB and US‐HHP450 treatments, which also completely inactivated micro‐organisms. The highest reduction in enzymes was observed in WB treatment followed by US‐HHP450. Total polyphenol contents increased significantly in US, HHP and US‐HHP treatments with the highest increase in US‐HHP450, but it decreased significantly in WB treatment. Ascorbic acid contents significantly decreased in WB and HHP450, while increased in US and US‐HHP treatments. Results indicate that US‐HHP450 treatment improves the quality of carrot juice and serves as a good alternative for blanching treatment.
