Abstract:Thirty-five crossbred Angus heifers (initial BW = 359.5 ± 7.1 kg) were randomly assigned to a 2 × 2 factorial design to evaluate effects of vitamin and mineral supplementation [VMSUP; supplemented (VTM) vs. unsupplemented (NoVTM)] and different rates of gain [GAIN; low gain (LG), 0.28 kg/d, vs. moderate gain (MG), 0.79 kg/d] during the first 83 d of gestation on dam hormone and metabolic status, fetal tissue and organ mass, and concentration of glucose and fructose in fetal fluids. The VMSUP was initiated 71 to 148 d before artificial insemination (AI), allowing time for mineral status of heifers to be altered in advance of breeding. At AI heifers were assigned their GAIN treatment. Heifers received treatments until the time of ovariohysterectomy (d 83 ± 0.27 after AI). Throughout the experiment, serum samples were collected and analyzed for non-esterified fatty acids (NEFA), progesterone (P4), insulin, and insulin-like growth factor 1 (IGF-1). At ovariohysterectomy, gravid reproductive tracts were collected, measurements were taken, samples of allantoic (ALF) and amniotic (AMF) fluids were collected, and fetuses were dissected. By design, MG had greater ADG compared to LG (0.85 vs. 0.34 ± 0.04 kg/d, respectively; p < 0.01). Concentrations of NEFA were greater for LG than MG (p = 0.04) and were affected by a VMSUP × day interaction (p < 0.01), with greater concentrations for NoVTM on d 83. Insulin was greater for NoVTM than VTM (p = 0.01). A GAIN × day interaction (p < 0.01) was observed for IGF-1, with greater concentrations for MG on d 83. At d 83, P4 concentrations were greater for MG than LG (GAIN × day, p < 0.01), and MG had greater (p < 0.01) corpus luteum weights versus LG. Even though fetal BW was not affected (p ≥ 0.27), MG fetuses had heavier (p = 0.01) femurs than LG, and VTM fetuses had heavier (p = 0.05) livers than those from NoVTM. Additionally, fetal liver as a percentage of BW was greater in fetuses from VTM (P = 0.05; 3.96 ± 0.06% BW) than NoVTM (3.79 ± 0.06% BW), and from LG (p = 0.04; 3.96 ± 0.06% BW) than MG (3.78 ± 0.06% BW). A VMSUP × GAIN interaction was observed for fetal small intestinal weight (p = 0.03), with VTM-MG being heavier than VTM-LG. Therefore, replacement heifer nutrition during early gestation can alter the development of organs that are relevant for future offspring performance. These data imply that compensatory mechanisms are in place in the developing conceptus that can alter the growth rate of key metabolic organs possibly in an attempt to increase or decrease energy utilization.

Effect of repeated trace mineral injections on beef heifer development and reproductive performance

Abstract:To determine the effects of repeated trace mineral injections on heifer development and reproductive performance, commercial Angus heifers (n = 290; 199 ± 34.3 kg; 221 ± 22 d of age) were utilized in a completely randomized design. Heifers were stratified by body weight (BW) and were administered an injectable trace mineral (MM; Multimin 90) or saline (CON) given subcutaneously, post-weaning at 221, 319, 401, and 521 ± 22 d of age. Throughout development, heifers grazed endophyte-infected fescue, red clover pastures and were supplemented with corn distillers grains (2.7 kg per heifer per day) and given access to free choice inorganic minerals. Heifer BW and body condition scores (BCS) were collected at trial initiation and 4- to 7-wk intervals thereafter. Hair coat scores (HCS) and respiration rates (n = 30 heifers per treatment) were collected at 269, 310, and 361 ± 22 d of age. Blood and liver samples were collected at trial initiation and estrous synchronization from 30 heifers per treatment to determine trace mineral status. At 319, 372, and 421 ± 22 d of age, antral follicle count and ovarian size were determined via ultrasonography. Two blood samples from all heifers were collected 10 d apart, concurrent with ultrasound dates, for cyclicity determination. Estrous synchronization was initiated, and reproductive tract scores (RTS) were collected at 421 ± 22 d of age, and heifers were bred via artificial insemination (AI) at 430 ± 22 d of age. Heifer BW, BCS, and HCS did not differ (P ≥ 0.12) throughout development, except at 268 ± 22 d of age when BCS was greater (P = 0.03) for MM than CON heifers. Respiratory rates were greater (P = 0.05) for MM than CON heifers at 269 ± 22 d of age but did not differ (P ≥ 0.66) at 310 and 361 ± 22 d of age. Plasma Mn and Zn concentrations did not differ (P ≥ 0.54). However, MM heifers had greater (P ≤ 0.01) plasma and liver concentrations of Cu and Se compared to CON. Interestingly, MM decreased (P = 0.02) liver Zn concentrations compared to CON, and there was no difference (P = 0.60) in liver Mn. Antral follicle count and ovarian size did not differ (P ≥ 0.51) due to treatment. Throughout development, number of heifers cycling was lesser (P < 0.01) for MM than CON heifers. However, there was no difference (P ≥ 0.19) in RTS, AI pregnancy rates, or overall pregnancy rates. Supplementing an injectable trace mineral increased heifer Cu and Se status; however, no effect was noted on ovarian development or pregnancy rates.

Abstract:Abstract Despite the increased appreciation of the involvement of maternal nutrition in fetal development, its effects on jejunum programming remain largely unexplored. Here, we investigated the impact of maternal vitamin and mineral supplementation throughout gestation on the differential gene expression and underlying biological pathways in the jejunum of neonatal calves. For this, beginning 60 d before breeding and continuing throughout gestation, crossbred Angus heifers (n = 14) were fed either a basal diet (CON; n = 7) or the basal diet plus 113 g•heifer-1•d-1 of VTM supplement (VTM; n = 7). All heifers were gestating female calves, which were euthanized 30 h after birth. Samples of jejunum mucosa were collected, stored at -80°C, then subjected to total RNA isolation and RNA sequencing. After quality control, the reads were mapped using STAR aligner and differentially expressed genes (DEGs) were identified using DESeq2. Maternal VTM supplementation did not affect calf organ mass, including jejunum weight. However, high-resolution respirometry analysis revealed greater mitochondrial efficiency in jejunum of calves from VTM supplemented dams. Additionally, we identified 528 DEGs between the VTM and CON calves (P-value ≤ 0.05 and |log2FC| > 0.5). Among them, the genes IGFBP2 and IGFBP6, which are key genes in insulin pathway and involved in regulation of insulin-like growth factors (IGF), IGF transport, and uptake, were upregulated in the VTM group. Additional pathways were identified using ShinyGO, which included the pancreatic secretion pathway, over-represented by genes such as PLCB1, CCK, and SLC26A3. The gene PLCB1 has been associated with feed efficiency, growth, and carcass traits in multiple species. Likewise, the SLC26A3 gene encodes a key intestinal chloride anion exchanger associated with enhanced feed efficiency in cattle. Genes encoding signaling molecules involved with immune response, T cell activation, and defense response were also among the DEGs upregulated in the VTM calves, including the TNFSF18 and TNFRSF4 genes. Our findings suggest that maternal prenatal vitamin and mineral supplementation throughout gestation affects the expression of genes involved in the jejunum function of offspring at a neonatal time point. The long-term consequences of these changes, however, warrant further investigation.

Abstract:Our study objectives were to evaluate the effects of divergent rates of body weight (BW) gain during early gestation in beef heifers on F0 performance, metabolic and endocrine status, colostrum immunoglobulins, and subsequent F1 calf characteristics, growth performance, concentrations of hormones and metabolites, and response to vaccination. Angus-based heifers (n = 100; BW = 369 ± 2.5 kg) were adapted to individual feeding for 14 d and bred using artificial insemination with female-sexed semen. Heifers were ranked by BW and assigned to either a basal diet targeting 0.28 kg/d gain (low [LG], n = 50) or the basal diet plus an energy/protein supplement targeting 0.79 kg/d gain (moderate gain [MG], n = 50) until day 84 of gestation. Dam BW and blood samples were collected at 6 time points during gestation; body composition was evaluated on days -10 and 84; and fetal measurements were taken on days 42, 63, and 84. At calving (LG, n = 23; MG, n = 23), dam and calf BW were recorded; and colostrum, calf body measurements, and blood samples were collected. Cow-calf pairs were managed on a common diet from calving to weaning, followed by a common postnatal development period for all F1 female offspring. Growth performance, hormone and metabolite profiles, feeding behavior, and reproductive performance were assessed from birth to prebreeding in F1 heifers. Offspring were vaccinated against respiratory disease and bovine viral diarrhea pathogens on days 62.3 ± 4.13 and 220.3 ± 4.13 postcalving. By design, MG dams were heavier (P < 0.0001) than LG on day 84, and the BW advantage persisted until subsequent weaning of F1 calves. Concentrations of serum IGF-1 and glucose were increased throughout gestation (P < 0.001) in MG dams, whereas concentrations of NEFA were decreased (P < 0.001) in LG dams. Calves from MG dams were 2.14 kg heavier (P = 0.03) and had larger chest circumference (P = 0.04) at birth compared with LG cohorts. Heifers from MG dams continued to have greater (P ≤ 0.03) BW gain and feed efficiency during the development period, but no differences were observed (P ≥ 0.13) in body composition, concentrations of hormones and metabolites, feeding behavior, puberty attainment, and response to vaccination in F1 offspring. Hence, early gestation rate of gain impacted BW and concentrations of glucose and IGF-1 throughout gestation in the F0 dam, resulting in altered F1 calf BW and measurements at birth and increased gain and efficiency during the development period.

Abstract:Bacterial communities play major roles in rumen and uterine function toward optimal animal performance and may be affected by changes occurring during heifer development such as nutritional supplementation for optimal growth and the attainment of puberty. The effect of different levels of protein supplementation on ruminal and uterine bacterial communities following weaning was examined through first breeding of heifers. Angus heifers (n = 39) were blocked by initial body weight (BW) and randomly assigned to one of three 163-day (d) crude protein (CP) supplementation diets including control (10% CP, n = 14), 20% CP (n = 11), or 40% CP (n = 14) treatment groups. Growth and development were monitored by body weight, with blood progesterone concentration determined every 14 d to determine pubertal status. Uterine flush and rumen fluid were collected on d 56, 112, and 163 relative to the start of supplementation. Bacterial DNA was extracted from fluid samples, the V1–V3 hypervariable region of the 16S rRNA gene was amplified, and amplicons were sequenced then processed in R 4.1. Statistical analyses were performed in SAS 9.4 with a GLIMMIX procedure utilizing fixed effects of protein, month, pubertal status, and interactions, with random effects including BW, interaction of BW and protein, and heifer within the interaction, and repeated measures of day. In the uterus, pubertal status and day of supplementation affected the observed amplicon sequence variants (ASVs) and led to clustering of samples in a principal coordinate analysis (PCoA; P < 0.05), but no effect of protein supplementation was observed. Ruminal samples clustered in PCoA ( P = 0.001), and observed ASVs were impacted over time ( P < 0.0001), but no effect of protein supplementation was detected. In contrast, protein supplementation, pubertal status, and day of supplementation affected the abundance of multiple phyla and genera in the uterus and rumen ( P < 0.05). Temporal and pubertal status effects on the heifer’s uterine bacterial communities potentially indicate a maturing uterine microbiome. Protein supplementation did not impact microbial diversity measures but did affect the abundance of individual bacterial phyla and genera that may provide future opportunities to manipulate bacterial community composition and maximize productivity.
One-carbon metabolite supplementation to nutrient-restricted beef heifers affects placental vascularity during early pregnancy

Abstract:We hypothesized that restricted maternal nutrition and supplementation of one-carbon metabolites (OCM; methionine, folate, choline, and vitamin B12) would affect placental vascular development during early pregnancy. A total of 43 cows were bred, and 32 heifers successfully became pregnant with female calves, leading to the formation of four treatment groups: CON - OCM (n = 8), CON + OCM (n = 7), RES - OCM (n = 9), and RES + OCM (n = 8). The experimental design was a 2 × 2 factorial, with main factors of dietary intake affecting average daily gain: control (CON; 0.6 kg/d ADG) and restricted (RES; -0.23 kg/d ADG); and OCM supplementation (+OCM) in which the heifers were supplemented with rumen-protected methionine (7.4 g/d) and choline (44.4 g/d) and received weekly injections of 320 mg of folate and 20 mg of vitamin B12, or received no supplementation (-OCM; corn carrier and saline injections). Heifers were individually fed and randomly assigned to treatment at breeding (day 0). Placentomes were collected on day 63 of gestation (0.225 of gestation). Fluorescent staining with CD31 and CD34 combined with image analysis was used to determine the vascularity of the placenta. Images were analyzed for capillary area density (CAD) and capillary number density (CND). Areas evaluated included fetal placental cotyledon (COT), maternal placental caruncle (CAR), whole placentome (CAR + COT), intercotyledonary fetal membranes (ICOT, or chorioallantois), intercaruncular endometrium (ICAR), and endometrial glands (EG). Data were analyzed with the GLM procedure of SAS, with heifer as the experimental unit and significance at P ≤ 0.05 and a tendency at P > 0.05 and P < 0.10. Though no gain × OCM interactions existed (P ≥ 0.10), OCM supplementation increased (P = 0.01) CAD of EG, whereas nutrient restriction tended (P < 0.10) to increase CAD of ICOT and CND of COT. Additionally, there was a gain × OCM interaction (P < 0.05) for CAD within the placentome and ICAR, such that RES reduced and supplementation of RES with OCM restored CAD. These results indicate that maternal rate of gain and OCM supplementation affected placental vascularization (capillary area and number density), which could affect placental function and thus the efficiency of nutrient transfer to the fetus during early gestation.

Abstract:Abstract Trace minerals are known to play important roles in early embryo development. The study objective was to determine effects of trace mineral source on heifer reproductive performance. Beef heifers (n = 129) were randomly assigned to one of two treatments. From weaning through breeding, all heifers were individually fed a basal diet supplemented with cobalt (Co), copper (Cu), manganese (Mn), and zinc (Zn) either from organic sources (COMP; Cu, Mn, and Zn amino acid complexes and Co glucoheptonate; Availa-4, Zinpro Corporation, Eden Prairie, MN) or inorganic sources (INORG; Cu, Mn, and Zn hydroxychlorides; Intellibond C, M, and Z, Micronutrients, Indianapolis, IN) and Co as CoSO4. Blood samples and a reproductive tract score (RTS) were collected to determine pubertal status. All animals were synchronized and artificially inseminated. Pregnancy status was determined by lymphocyte gene expression, circulating concentrations of pregnancy-associated glycoproteins (PAGs), and by transrectal ultrasonography after artificial insemination. Embryonic loss was defined as when a previously pregnant animal was subsequently diagnosed not pregnant. Data were analyzed using the MIXED procedure in SAS. Puberty (P = 0.44), pelvic area (P = 0.74), RTS (P = 0.49), and estrus expression (P = 0.82) were not influenced by treatment. There was no effect of treatment (P = 0.37) or treatment by time (P = 0.19) on pregnancy, but there was a tendency (P = 0.13) for decreased embryonic loss among COMP heifers (27 ± 6%) compared to INORG heifers (38 ± 6%). There was a treatment by pregnancy status by time interaction (P < 0.01) on circulating PAG concentrations with PAG concentrations tending (P = 0.08) to be greater on day 25 among heifers in the COMP treatment compared to heifers in the INORG group. In summary, source of trace mineral did not affect puberty, RTS, pelvic area, or overall pregnancy success, but feeding complexed trace minerals tended to increase circulating PAG concentrations and embryo survival.

Abstract:This study evaluated the effects of maternal supplementation of a Bacillus-based direct-fed microbial (DFM) on the physiology and growth performance of Bos indicus-influenced cow-calf pairs. On day 0 (~139 d before expected calving date), 72 fall-calving, Brangus crossbred beef heifers (20 to 22 mo of age) pregnant with first offspring were stratified by their initial body weight (BW; 431 ± 31 kg) and body condition score (BCS; 6.0 ± 0.36; scale 1 to 9), and randomly allocated into 1 of 12 bahiagrass pastures (1 ha and six heifers per pasture). Treatments were randomly assigned to pastures (six pastures per treatment) and consisted of heifers supplemented with 1 kg/d of soybean hulls (dry matter, DM) that was added (BAC) or not (CON) with DFM containing Bacillus subtilis and B. licheniformis (Bovacillus; Chr. Hansen A/S, Hørsholm, Denmark). Treatments were provided from days 0 to 242 (139 ± 4 d prepartum to 104 ± 4 d postpartum). Calves were weaned on day 242 (96 ± 30 d of age) and then allocated into 1 of 16 drylot pens and fed the same concentrate at 3.25% of BW (DM) until day 319. Maternal treatment effects were not detected (P ≥ 0.29) for herbage allowance and forage chemical composition. Heifer BCS on days 39 and 63 tended (P ≤ 0.09) to be greater for BAC vs. CON heifers, whereas heifer BCS on day 91 was greater (P = 0.01) for BAC vs. CON heifers. Heifer BCS did not differ (P ≥ 0.20) between treatments on days 179 and 242. Plasma glucose concentration did not differ from days 0 to 63 (P ≥ 0.14) but were greater (P < 0.01) on day 179 and tended (P = 0.09) to be greater on day 242 for BAC vs. CON heifers. Calf BW at birth, ADG from birth to weaning, and BW at weaning did not differ (P ≥ 0.19) between treatments, but calf BW at drylot exit (day 319) was greater (P = 0.05) for BAC vs. CON calves. Maternal treatment effects were not detected (P ≥ 0.42) for calf serum concentration of IgG at birth and postvaccination plasma concentrations of glucose, cortisol, and haptoglobin. Serum titers against bovine respiratory syncytial virus (BRSV) were greater (P = 0.04) for BAC vs. CON calves on day 287, whereas seroconversion against parainfluenza-3 virus (PI-3) was greater (P < 0.01) for BAC vs. CON calves on day 271. Thus, maternal supplementation of a Bacillus-based DFM increased prepartum BCS gain and postpartum plasma glucose concentration of heifers and led to positive carryover effects on postweaning BW gain and humoral immune response in their offspring.

Abstract:The objective of this study was to determine the dose-dependent response of one-carbon metabolite (OCM: methionine, choline, folate, and vitamin B12) supplementation on heifer dry matter intake on fixed gain, organ mass, hematology, cytokine concentration, pancreatic and jejunal enzyme activity, and muscle hydrogen peroxide production. Angus heifers (n = 30; body weight [BW] = 392.6 ± 12.6 kg) were individually fed and assigned to one of five treatments: 0XNEG: total mixed ration (TMR) and saline injections at days 0 and 7 of the estrous cycle, 0XPOS: TMR, rumen-protected methionine (MET) fed at 0.08% of the diet dry matter, rumen-protected choline (CHOL) fed at 60 g/d, and saline injections at days 0 and 7, 0.5X: TMR, MET, CHOL, 5-mg B12, and 80-mg folate injections at days 0 and 7, 1X: TMR, MET CHOL, 10-mg vitamin B12, and 160-mg folate at days 0 and 7, and 2X: TMR, MET, CHOL, 20-mg vitamin B12, and 320-mg folate at days 0 and 7. All heifers were estrus synchronized but not bred, and blood samples were collected on days 0, 7, and at slaughter (day 14) during which tissues were collected. By design, heifer ADG did not differ (P = 0.96). Spleen weight and uterine weight were affected cubically (P = 0.03) decreasing from 0XPOS to 0.5X. Ovarian weight decreased linearly (P < 0.01) with increasing folate and B12 injection. Hemoglobin and hematocrit percentage were decreased (P < 0.01) in the 0.5X treatment compared with all other treatments. Plasma glucose, histotroph protein, and pancreatic α-amylase were decreased (P ≤ 0.04) in the 0.5X treatment. Heifers on the 2X treatment had greater pancreatic α-amylase compared with 0XNEG and 0.5X treatment. Interleukin-6 in plasma tended (P = 0.08) to be greater in the 0XPOS heifers compared with all other treatments. Lastly, 0XPOS-treated heifers had reduced (P ≤ 0.07) hydrogen peroxide production in muscle compared with 0XNEG heifers. These data imply that while certain doses of OCM do not improve whole animal physiology, OCM supplementation doses that disrupt one-carbon metabolism, such as that of the 0.5X treatment, can induce a negative systemic response that results in negative effects in both the dam and the conceptus during early gestation. Therefore, it is necessary to simultaneously establish an optimal OCM dose that increases circulating concentrations for use by the dam and the conceptus, while avoiding potential negative side effects of a disruptive OCM, to evaluate the long-term impacts of OCM supplementation of offspring programming.

Abstract:Replacement heifer development is one of the most critical components in beef production. The composition of the ideal uterine environment could maximize fertility and reproductive efficiency. Our hypothesis was that protein supplementation would affect the uterine environment of beef heifers without inhibiting development or reproduction. To test the effects of dietary supplementation on these outcomes, a randomized complete block design with repeated measures was implemented. Angus heifers (n = 60) were blocked by body weight (BW) and randomly assigned to one of three supplemental protein treatment groups (10% (CON), 20% (P20), and 40% (P40)). Mixed model ANOVAs were used to determine whether protein supplementation treatments, time, and the interaction or protein supplementation, semen exposure, and the interaction influenced uterine luminal fluid (ULF) and pregnancy outcomes. Amino acids (AAs) were impacted (p < 0.001), specifically, the essential AAs: Arg, Iso, Leu, Val, His, Lys, Met, Phe, Trp. Protein supplementation influenced multiple AAs post-insemination: Arg (p = 0.03), CC (p = 0.05), 1-MH (p = 0.001), and Orn (p = 0.03). In conclusion, protein supplementation did not affect the reproductive development via puberty attainment or the timing of conception even with alterations in growth. However, uterine AA concentrations did change throughout development and protein supplementation influenced ULF d 14 post-insemination, which may affect the conception rates.

Abstract:To investigate effects of inorganic or complexed trace mineral source (zinc, copper, manganese, and cobalt) on receiving period performance and morbidity, crossbred beef heifer calves (n = 287) arriving on three delivery dates were used in a 42-d receiving trial. Heifers were processed after arrival, stratified by day -1 body weights (BW) and allocated randomly to eight pens (11 to 13 heifers/pen, 24 pens total). Within truckload, pens were assigned randomly to dietary treatment (n = 12 pens/treatment). Heifers were housed on 0.42-ha grass paddocks, provided ad libitum bermudagrass hay and provided dietary treatments in grain supplements fed daily. Treatments consisted of supplemental zinc (360 mg/d), copper (125 mg/d), manganese (200 mg/d), and cobalt (12 mg/d) from complexed (Zinpro Availa 4, Zinpro Corp. Eden Prairie, MN) or inorganic sources (sulfates). Heifers were observed daily for clinical bovine respiratory disease (BRD). If presenting BRD symptoms and rectal temperature ≥ 40 °C, heifers were deemed morbid and treated with antibiotics. Six heifers/pen were bled to determine serum haptoglobin concentrations on days 0, 14, and 28. Liver biopsies were taken on day 5 ± 2 and 43 ± 1 from three calves selected randomly from each pen for mineral status comparisons. Statistical analyses were performed using the MIXED, GLIMMIX, and repeated measures procedures of SAS 9.4 with truckload as a random effect and pen within truckload specified as subject. There tended to be a treatment by day interaction for BW (P = 0.07). Heifer BW did not differ on day 0 (P = 0.82) and day 14 (P = 0.36), but heifers fed complexed trace minerals had greater BW on day 28 (P = 0.04) and day 42 (P = 0.05). Overall average daily gains were greater for heifers fed complexed trace minerals (P = 0.05; 0.78 vs. 0.70 kg, SE = 0.03). Heifers supplemented with inorganic trace minerals had greater BRD incidence (P = 0.03; 58 vs. 46%, SE = 3.6). Haptoglobin concentrations decreased throughout the trial (P < 0.001), and heifers fed complexed trace minerals tended to have a decrease in haptoglobin concentrations (P = 0.07). The source of trace mineral supplementation had no effect (P ≥ 0.20) on liver mineral concentrations and there were no treatment × day interactions (P ≥ 0.35). In conclusion, supplementing diets for the first 42 d after arrival with complexed trace mineral sources improved heifer performance as compared to heifers supplemented with inorganic trace minerals.

Abstract:The dairy cattle production rises worldwide, with genetically selected animals. Consequently, they present high nutritional requirements, especially during the transition period. The aim of this study was to evaluate the effects of parenteral supplementation of a mineral blend composed by selenium, copper, magnesium, phosphorus, and potassium on hematological, biochemical, immunological, and antioxidant status in transition period dairy cows and newborn calves. The Holstein cows were divided into the control group (A, n = 3) and treated group (B, n = 4), which were supplemented twice with an intramuscularly dose of 10 mL/animal, at approximately 20 days prior to the parturition, and a subsequent dose at the parturition. Total blood was collected from cows on days 20 and 5 antepartum, and 2 and 7 postpartum, and from calves, at the birth. We observed a decreased in concentrations of thiobarbituric acid reactive substances (TBARSs) for cows fed mineral blend during the transition period (P < 0.05), as well as a greater number of total leukocyte, total protein and globulin levels, and reduction of hemoglobin levels (P < 0.05). The catalase activity was increased in supplemented cows and in its calves (P < 0.05). Therefore, based on these evidences, the parenteral supplementation with mineral provides beneficial effects in pregnant cows and during the transition period, as well as in their calves.

Abstract:Objective: To investigate the effects of low-dose trace mineral proteinates on reproductive performance, mineral status, milk immunoglobulin contents and fecal mineral excretion of sows. Methods: Eighty crossbred sows (LandracexLarge White) were randomly allocated to two groups in a 135-day trail, from breeding through 21 d postpartum. The two treatments were inorganic trace minerals (ITM): a basal diet+inorganic iron (Fe), copper (Cu), manganese (Mn), and zinc (Zn) at 90, 15, 25 and 90 mg/kg, respectively and organic trace minerals (OTM): a basal diet+proteinates of Fe, Cu, Mn, and Zn at 72, 12, 20, and 72 mg/kg, respectively. Results: Compared with ITM, OTM significantly increased (p<0.05) the number of piglets with birthweight >1 kg, the litter weaning weight, and milk Fe, Cu contents. No significant differences (p>0.05) were observed on sow hair mineral contents or immunoglobulin M (IgM), IgG, and IgA contents in colostrum and milk. In comparsion to ITM, OTM decreased fecal Fe, Cu, Mn, and Zn contents of gestating sows (p<0.01) and Fe, Mn, and Zn in lactating sows (p<0.01). Conclusion: These results indicate that low-dose mineral proteinates can increase the number of piglets with birthweight >1 kg, the litter weaning weight and certain milk mineral concentrations while reducing fecal mineral excretion.

Abstract:This study examined the impact of maternal protein supplementation during mid-gestation on offspring, considering potential sex-related effects. Forty-three pregnant purebred Tabapuã beef cows (20 female and 23 male fetuses) were collectively managed in a pasture until 100 d of gestation. From 100 to 200 d of gestation, they were randomly assigned to the restricted group [(RES) - basal diet (75% corn silage + 25% sugar cane bagasse + mineral mixture); n = 24] or control group [(CON) - same basal diet + based-plant supplement [40% of crude protein, 3.5 g/kg of body weight (BW); n = 19]. From 200 d of gestation until parturition, all cows were equally fed corn silage and mineral mixture. During the cow-calf phase, cows and their calves were maintained in a pasture area. After weaning, calves were individually housed and evaluated during the backgrounding (255 to 320 d), growing 1 (321 to 381 d), and growing 2 (382 to 445 d) phases. Offspring's blood samples were collected at 210 and 445 d of age. Samples of skeletal muscle tissue were collected through biopsies at 7, 30, and 445 d of age. Muscle tissue samples were subjected to reverse-transcription quantitative polymerase chain reaction analysis. Prenatal treatment and offspring's sex (when pertinent) were considered fixed effects. The significance level was set at 5%. At mid-gestation, cows supplemented with protein reached 98% and 92% of their protein and energy requirements, while nonsupplemented cows attained only 30% and 50% of these requirements, respectively. The RES offspring were lighter at birth (27 vs. 31 kg), weaning (197 vs. 214 kg), and 445 d of age (398 vs. 429 kg) (P ≤ 0.05). The CON calves had greater (P < 0.05) morphometric measurements overall. The CON offspring had ~26% greater muscle fiber area (P ≤ 0.01). There was a trend (P = 0.06) for a greater Mechanistic target of rapamycin kinase mRNA expression in the Longissimus thoracis in the CON group at 7 d of age. The Myogenic differentiation 1 expression was greater (P = 0.02) in RES-females. Upregulation of Carnitine palmitoyltransferase 2 was observed in RES offspring at 445 d (P = 0.04). Expression of Fatty acid binding protein 4 (P < 0.001), Peroxisome proliferator-activated receptor gamma (P < 0.001), and Stearoyl-Coenzyme A desaturase (P < 0.001) was upregulated in CON-females. Therefore, protein supplementation during gestation enhances offspring growth and promotes favorable responses to lipogenesis, particularly in females.

Abstract:Oxidative stress may affect new born calves due to high stress suffered around birth. We hypothesized that maternal supplementation with micronutrients and vitamins in late gestation enhance the neonatal calf's antioxidant system, decreasing the occurrence and duration of diarrhea, and improving growth from birth through weaning. To test this hypothesis, 80 multiparous cows were cluster-assigned to treatment groups. Treated group (TG) cows received mineral and vitamin supplementation while control group (CG) cows received saline solution. Feed intake and fecal score were measured daily until the ninth week. Weight and body measurements were registered weekly, and blood samples were collected from postpartum cows and calves after birth and at 7, 14, and 63 days of life. Although CG calves had greater fecal scores (p = 0.01), diarrhea characteristics did not differ. Calves in the TG showed greater starter intake (p = 0.04). Feed efficiency showed a trend with treatment-age interaction (p = 0.06). Calves in the CG had wider hips in the first week (p = 0.03), but not by the ninth week. Total antioxidant status, thiobarbituric acid reactive substances, and haptoglobin did not differ between treatment groups. Serum metabolites showed no differences. Supplementation did not impact calf antioxidant system or growth in the first two months.

Abstract:We examined the hypothesis that maternal nutrition and day of gestation would affect the concentrations of AAs and hexoses in bovine utero-placental fluids and maternal serum from days 16 to 50 of gestation. Forty-nine cross-bred Angus heifers were bred via artificial insemination and fed a control diet (CON = 100% of requirements for growth) or a restricted diet (RES = 60% of CON) and ovariohysterectomized on days 16, 34, or 50 of gestation; nonpregnant controls were not bred and ovariohysterectomized on day 16 of the synchronized estrous cycle. The resulting design was a completely randomized design with a 2 × 3 factorial + 1 arrangement of treatments. Maternal serum, histotroph, allantoic fluid, and amniotic fluid were collected at time of ovariohysterectomy. Samples were then analyzed for concentrations of AAs and intermediary metabolites: alanine (Ala), arginine, asparagine (Asn), aspartate (Asp), citrulline, cysteine, glutamine, glutamate (Glu), glycine (Gly), histidine, isoleucine, leucine (Leu), lysine, methionine (Met), ornithine, phenylalanine (Phe), proline (Pro), serine (Ser), threonine (Thr), tryptophan, tyrosine (Tyr), and valine (Val). The concentrations of Gly, Ser, and Thr in maternal serum were greater (P ≤ 0.05) in CON compared with RES. Furthermore, day of gestation affected (P ≤ 0.05) concentrations of Asn, Glu, Phe, Thr, and Tyr in maternal serum. Status of maternal nutrition affected the Asp concentration of histotroph where RES was greater (P = 0.02) than CON. In histotroph, Ala, Leu, Met, and Val concentrations were greater (P ≤ 0.05) on day 50 compared with day 16. Additionally, Glu and Pro concentrations in histotroph were greater (P < 0.01) on days 34 and 50 compared with day 16. A day × treatment interaction was observed for the concentration of Val in allantoic fluid where day 34 CON was greater (P = 0.05) than all other days and nutritional treatments. In addition, the concentration of Gln in amniotic fluid experienced a day × treatment interaction where day 34 RES was greater (P ≤ 0.05) than day 34 CON, which was greater (P ≤ 0.05) than day 50 CON and RES. These data support our hypothesis that day of gestation and maternal nutrition affect the concentrations of various neutral and acidic AA in beef heifer utero-placental fluids and maternal serum from days 16 to 50 of gestation.

Abstract:It is well established that one of the pathology‘ reason in calves is the disturbances of prenatal development as a result of vitamin and mineral abstinence. The aim of the study was to research the influence of vitamin‐mineral bolus’ supplementation with prolong action to the ration of cows on the calves’ protein metabolism. The studies were carried out in one of the breeding farms of the Leningrad region. Holstein cows were used aged from 3 to 5 years and their calves. 5 groups were formed (n=5 by each). The cows of the 1st experimental group have received two boluses All – mineral plus (the compound: Mg, Zn, Cu, Co, Se, I, Vitamin A, D3, E, Holland Animal Care, Holland), 2d experimental – two boluses Uno Biotin (the compound: Mg, Zn, Cu, Co, Se, I, Vitamin A, D3, E, Holland Animal Care, Holland), 3d experimental group ‐ one Cattle bolus with Iodine (the compound: Cu, Co, Se, I, Telsol Limited, United Kingdom), 4th experimental group – one Cattle Bullet (the compound: Mg, Zn, Cu, Co, Se, I, Vitamin A, D3, E, Holland Animal Care, Holland). The 5th group was control with ordinary ration vitamin‐mineral diet. All cows of experimental groups have got boluses one time which was 60 days before calving. The prolongation of all boluses was 120 days after receiving except bolus All‐mineral plus. The prolongation of their was 180 days. The compound of these boluses includes manganese, zinc, copper, selenium, cobalt, iodine, Vitamin A, D3, E, H. We used the clinical examination of calves and biochemical analysis of calves blood samples on the 3d‐5th days after calving. Serum was analyzed for total protein, albumins and globulins. We were used Student's t‐tests to study differences between groups. Thus, the clinically health animals were nineteen calves of experimental groups and second calves if control group. The body mass of newborn calves was 35‐40 kg. The calves were stood up during first two hours after calving and had a healthy sucking reflex. The temperature of their was fluctuated within 38.5‐39.30°С. The results of biochemical analysis showed that the level of total protein was within the physiological norm in the bloods’samples of 1st, 3d, 4th and 5 th calves groups. The level of total protein was statistically increased in the calves of 1st group in 1.4 times (р<0,05) compared with data of calves in the control group. The value of albumins of calves was decreased in 1.07 times (р<0,05) in 2d group and in 1.4 times in control group (18,19±3,37g/l). We were observed the insignificant increasing trend of albumins’level in calves of 1st,2d,3d and 4th groups compared with data of control group. The difference of globulins’level between calves of 1st, 4th experimental and control group was 1.3 times (р<0,05). The value of globulin in the calves of 2d group was 35,64±5,93 g/l and 3d group ‐ 34,45±7,93 g/l. So, we can assume that the vitamin‐mineral supplementation of cow‐mothers during dry time has a positive influence on the synthesis of the total protein and its fraction ratio.

Effects of maternal nutrient restriction followed by realimentation during early and midgestation on beef cows. I. Maternal performance and organ weights at different stages of gestation

Abstract:The objectives were to evaluate the effects of nutrient restriction during early to midgestation followed by realimentation on maternal performance and organ mass in pregnant beef cows. On d 30 of pregnancy, multiparous, nonlactating cows (initial BW = 620.5 ± 11.3 kg and BCS = 5.1 ± 0.1) were assigned to 1 of 3 dietary treatments: control (CON; 100% NRC; n = 18) and restricted (RES; 60% NRC; n = 30). On d 85, cows were slaughtered (CON, n = 6; R, n = 6), remained on control (CC; n = 12) and restricted (RR; n = 12), or were realimented to control (RC; n = 11). On d 140, cows were slaughtered (CC, n = 6; RR, n = 6; RC, n = 5), remained on control (CCC, n = 6; RCC, n = 5), or were realimented to control (RRC, n = 6). On d 254, all remaining cows were slaughtered. Cows were weighed before slaughter and all maternal organs were dissected and weighed. The diet consisted of grass hay to meet 100 or 60% NEm recommendations for fetal growth and to meet or exceed recommendations for other nutrients. At d 85 slaughters, BW and empty BW (EBW) were not affected (P ≥ 0.84) by maternal nutrition. However back fat was decreased (P = 0.05) in RES vs. CON cows. Large intestine and abomasum mass were increased (P ≤ 0.05) in RES cows vs. CON. At d 140, BW was decreased (P = 0.05) and EBW tended to be decreased (P = 0.10) in RRC cows vs. CCC and RCC being intermediate. Liver mass was decreased (P = 0.02) in RR vs. CC with RC being intermediate. Ruminal mass was decreased (P = 0.003) in RR vs. CC and RC cows. At d 254, BW and EBW were similar (P ≥ 0.78) across treatments. We observed partial changes in maternal weight and organ masses due to different lengths of maternal nutrient restriction followed by realimentation. It appears that the dam undergoes some adaptations during an early to midgestation nutrient restriction and becomes more efficient in the utilization of nutrients after being realimented and as gestation advances.

Abstract:Abstract A total of 69 sows (DNA Line 200 × 400) and their progeny were used to determine if feeding a combination of vitamin D3 and 25(OH)D3 influences neonatal and sow vitamin D status, muscle fiber morphometrics at birth and weaning, and subsequent growth performance. Within 3 d of breeding, sows were allotted to one of three dietary treatments fortified with 1,500 IU/kg vitamin D3 (CON), 500 IU/kg vitamin D3 + 25 μg/kg 25(OH)D3 (DL), or 1,500 IU/kg vitamin D3 + 50 μg/kg 25(OH)D3 (DH). When pigs were sacrificed at birth, there were no treatment effects for all fiber morphometric measures (P > 0.170), except primary fiber number and the ratio of secondary to primary muscle fibers (P < 0.016). Pigs from CON fed sows had fewer primary fibers than pigs from sows fed the DH treatment (P = 0.014), with pigs from sows fed DL treatment not differing from either (P > 0.104). Pigs from CON and DL fed sows had a greater secondary to primary muscle fiber ratio compared to pigs from DH sows (P < 0.022) but did not differ from each other (P = 0.994). There were treatment × time interactions for all sow and pig serum metabolites (P < 0.001). Therefore, treatment means were compared within the time period. At all time periods, sow serum 25(OH)D3 concentrations differed for all treatments with the magnitude of difference largest at weaning (P < 0.011), where serum 25(OH)D3 concentration was always the greatest when sows were fed the DH diet. At birth, piglets from DH fed sows had greater serum 25(OH)D3 concentrations than piglets from sows fed the DL treatment (P = 0.003), with piglets from sows fed CON treatment not differing from either (P > 0.061). At weaning, serum concentrations of 25(OH)D3 in piglets from all sow treatments were different (P < 0.001), with the greatest concentration in piglets from DH sows, followed by CON, and followed by DL. There were no treatment × time interactions for any of the metabolites measured in milk and no treatment or time main effects for 24,25(OH)2D3 concentration (P > 0.068). Colostrum collected within 12 h of parturition contained less (P = 0.001) 25(OH)D3 than milk collected on day 21 of lactation. Regardless of time, concentrations of 25(OH)D3 in milk were different (P < 0.030), with the largest 25(OH)D3 concentration from DH fed sows, followed by DL, and then CON. In conclusion, combining vitamin D3 and 25(OH)D3 in the maternal diet improves the vitamin D status of the dam and progeny and it increases primary muscle fiber number at birth.
Mechanism of vertebral fracture in the F/FB-111 ejection experience.

Abstract:A review of the accident investigation reports of all non-fatal F/FB-111 ejections which occurred from 19 October 1967 to 26 March 1980 was conducted. The available spinal radiographs of the ejectees were also reviewed. The overall rate of vertebral compression fractures among the survived ejectees in a properly functioning module is 29.5% (23 of 78). The general mechanism of vertebral injury is a combined mechanism involving both axial compression and flexion. There is no evidence to indicate a hyperextension injury mechanism during retraction, nor is there evidence to support the presumed efficacy of the crossed-arms bracing procedure recommended for landing impact in modifying the vertebral injury rate. It appears that a significant reduction in the rate of vertebral fractures among F/FB-111 ejectees will require decreasing the acceleration stresses imposed on the crewmembers during landing impact.
