Abstract:Fusarium root rot (FRR) seriously affects the growth and productivity of A. chinensis. Therefore, protecting A. chinensis from FRR has become an important task, especially for increasing A. chinensis production. The purpose of this study was to screen FRR control strains from the A. chinensis rhizosphere soil. Eighty-four bacterial strains and seven fungal strains were isolated, and five strains were identified with high inhibitory effects against Fusarium oxysporum (FO): Trichoderma harzianum (MH), Bacillus amyloliquefaciens (CJ5, CJ7, and CJ8), and Bacillus subtilis (CJ9). All five strains had high antagonistic effects in vitro. Results showed that MH and CJ5, as biological control agents, had high control potential, with antagonistic rates of 86.01% and 82.78%, respectively. In the pot experiment, the growth levels of roots and stems of A. chinensis seedlings treated with MH+CJ were significantly higher than those of control plants. The total nitrogen, total phosphorus, total potassium, indoleacetic acid, and chlorophyll contents in A. chinensis leaves were also significantly increased. In the biocontrol test, the combined MH + CJ application significantly decreased the malondialdehyde content in A. chinensis roots and significantly increased the polyphenol oxidase, phenylalanine ammonolyase, and peroxidase ability, indicating a high biocontrol effect. In addition, the application of Bacillus spp. and T. harzianum increased the abundance and diversity of the soil fungal population, improved the soil microbial community structure, and significantly increased the abundance of beneficial strains, such as Holtermanniella and Metarhizium. The abundance of Fusarium, Volutella, and other pathogenic strains was significantly reduced, and the biocontrol potential of A. chinensis root rot was increased. Thus, Bacillus spp. and T. harzianum complex bacteria can be considered potential future biocontrol agents for FRR.
Efficacy and Potential Mechanism of Essential Oils of Three Labiatae Plants against the Pathogenic Fungi of Root Rot Disease in Atractylodes chinensis

Abstract:Atractylodes chinensis has a high medicinal value and is widely cultivated. However, root rot disease seriously affects the yield and quality of A. chinensis. To develop green and safe pesticides, the inhibitory effect of essential oils (EOs) of three Labiatae plants on the pathogenic fungi that causes root rot disease in Atractylodes chinensis was investigated. The results showed that the Origanum vulgare EO and Thymus mongolicus EO exhibited strong inhibitory effects on Fusarium oxysporum, Fusarium solani, and Fusarium redolens, with 100% inhibition rate. The low MIC values of EOs and their main components against the three pathogenic fungi indicated that all of them showed strong fungicidal effects. The MIC values of O. vulgare EO against F. oxysporum, F. solani, and F. redolens were 2.60 mg/mL, 3.13 mg/mL, and 1.56 mg/mL, respectively. Analyses using scanning electron microscopy (SEM) and transmission electron microscopy (TEM) showed that the O. vulgare EO severely damaged the cell wall and cell membrane of mycelial cells. The O. vulgare EO increased cell permeability, leading to a large leakage of cell contents (DNA and proteins). In addition, O. vulgare EO inhibited F. oxysporum by inducing ROS production and reducing the amount of intracellular GSH, leading to a large accumulation of ROS. This study showed that plant EOs have excellent fungicidal activity and can be used as novel natural and environmentally friendly pesticides for the control of root rot in A. chinensis.

Abstract:Fusarium avenaceum is the predominant pathogen associated with Angelica sinensis root rot, which results in mycotoxin contamination of Angelica, most prominently Enniatin B (ENN B). This study aimed to isolate bacteria capable of combating various phytopathogenic fungi and degrading ENN B to reduce Angelica root rot. Through co-culture with F. avenaceum, the bacterial strains YF and SY89 were isolated from Angelica rhizosphere soil for their antifungal activities. They were identified as Paenibacillus polymyxa and Bacillus tequilensis based on their morphological characteristics and phylogenetic trees constructed using the 16 S rDNA genes sequence. The strains YF and SY89 could produce antimicrobial substance such as surfactin, fengycin, iturin, polyketide synthases and non ribosomal polypeptide synthase detected by Polymerase Chain Reaction (PCR). In addition, strain P. polymyxa YF and B. tequilensis SY89 showed a prominent ability to inhibit synthesis and degrade ENN B in F. avenaceum suspensions and standard samples. The inhibition rate reached 61.93% and 77.64%, respectively, and the degradation rate reached 60.32% and 76.03%, respectively. Angelica pot experiments were conducted to further evaluate the strains YF and SY89 culture ability to promote plant growth and control Angelica root rot to assess its potential agricultural use. The results indicated that strains YF and SY89 could produce IAA and siderophores, which significantly promoted Angelica roots growth. In addition, strains YF and SY89 have the potential to increase the activity of resistant enzymes, thereby inhibiting F. avenaceum infection. The disease index of Angelica roots treated by strain YF and SY89 decreased by 65.38% and 61.54%, respectively. To further elucidate the antagonistic mechanism of YF and SY89, we examined their colonization pattern in the Angelica root using a green fluorescent protein (GFP) marker. The results indicated that YF and SY89 mainly colonized the root surface before migrating into the roots interior part. The present study demonstrated that P. polymyxa YF and B. tequilensis SY89 showed promising prospects for use as a biological control agent against Angelica root rot and ENN B inhibition under field conditions.

Abstract:Medicine plants such as Panax notoginseng (Sanqi) frequently experience replant failure owing to the accumulation of soilborne pathogens and autotoxins in continuous cropping soil. Bacillus sp. is a commonly used biocontrol agent to combat soilborne pathogens, but further research is needed to understand its impact on controlling Sanqi root rot and the underlying mechanisms. Here, we isolated and screened a Bacillus strain, B19, from Sanqi rhizosphere soil that exhibited potential for inhibiting Sanqi root rot pathogens. We then tested the strain's effect on the incidence of Sanqi root rot in continuous cropping soil, along with its motility/chemotaxis and biofilm formation ability (i.e., potential for root colonization). We also analyzed the strain's genomic characteristics, including potential secondary metabolite synthesis gene clusters. Our results showed that the strain B19 exhibited high inhibitory activity against fungal pathogens (F. solani and F. oxysporum) and could metabolize saponins as a sole carbon source. The application of B19 to continuous cropping soil significantly reduced the incidence of root rot in Sanqi seedlings. B19 was identified as B. velezensis through morphological characteristics, 16S rRNA and gyrA gene sequence analysis, and genome-level annotation. Notably, B19′s genome encodes many genes associated with functions such as motility/chemotaxis and biofilm formation, as well as 13 biosynthetic gene clusters that could encode secondary metabolites, including surfactin, fengycin, and siderophore. Additionally, B19 demonstrated the capability to produce volatile organic compounds that inhibit pathogenic fungi. The motility/chemotaxis and biofilm-forming ability of B19 were enhanced by saponins. Collectively, these properties could enhance B19′s biocontrol ability, thereby reducing the incidence of root rot.

Abstract:The aim of the present study was to screen native cultivable rhizobacteria with potential applications for biocontrol of root rot, which results in a serious and persistent disease problem for the pepper industry worldwide and is difficult to control through chemical and agricultural means. A total of 47 bacteria were isolated from pepper rhizosphere soil by spread plate and screened by dual culture assay. In in vitro dual culture assay, the strain DX1708 showed strong antagonistic activity against Fusarium oxysporum , F . solani , F . verticillioides , Phytophthora capsici and Rhizoctonia solani . Based on cultural and morphological characterization, and 16S rRNA gene and gyr A gene phylogenetic analysis strain DX1708 was identified as Bacillus atrophaeus . Strain DX1708 demonstrated many characteristics that are beneficial for plants, such as production of α -amylase, ammonia, cellulase, indole-3-acetic acid, pectinase, protease, siderophores, and phosphate solubilization. PCR-based detection of antimicrobial lipopeptides and peptides gene revealed the presence of ten genes in B . atrophaeus strain DX1708, which code for enzymes involved in production of bacilysin, fengycin, mycosubtilin, subtilosin and surfactin. In planta application of B . atrophaeus strain DX1708 significantly decreased root rot severity compared to untreated controls under greenhouse conditions. The strain DX1708 was tested in the field during three successive years (2018–2020). The results demonstrated a significant decrease in disease severity. These results suggest that B . atrophaeus strain DX1708 could control plant diseases via direct and indirect modes of action and can be used as a potential agent for biological control.

Antifungal Effects of Rhizospheric <i>Bacillus</i> Species Against Bayberry Twig Blight Pathogen <i>Pestalotiopsis versicolor</i>

Abstract:Bayberry is an attractive, nutritious, and popular fruit in China. The plant fungal pathogen Pestalotiopsis versicolor XJ27 is the causative agent of bayberry twig blight disease, which severely affects bayberry production. Traditional control techniques, such as chemical fungicides, are being used to control this disease; however, these techniques cause environmental and health hazards. In this study, we screened sweet potato rhizospheric bacteria with biocontrol potentials against P. versicolor XJ27, the bayberry twig blight pathogen. Ten isolates showed inhibition; Bacillus siamensis S3 and Bacillus tequilensis S5 showed the highest fungal growth inhibition. The antagonistic bacterial culture suspensions of S3 and S5 inhibited the mycelial growth by 82.9% and 76.2%, respectively. Their extracellular culture filtrates had mycelial growth inhibition of 86.8% and 82.2%, respectively. In detached leaf assay, the extracellular culture filtrates of S3 and S5 inhibited the size of the leaf lesion by 82.3% and 76.2%, respectively. SEM and TEM imaging showed a severe hyphal-damaged structure caused by the antagonistic bacteria. The fungal inhibition mechanisms might employ the hydrolytic enzymes and lipopeptides produced by the bacteria. Both the S3 and S5 have chitinase and protease activity; they produce a series of lipopeptides such as surfactin, iturin, and mycosubtilin. Therefore, we can suggest these bacteria as biocontrol agents for bayberry twig blight disease as an alternative to fungicides based upon their attributes of antifungal activity.

Abstract:Chemical agents in the rhizosphere soils of plants might have an influence on root-rot disease, which therefore might reveal the mechanism of root rot in Panax notoginseng (P. notoginseng). With this hypothesis the alterations of phenolic acids (PAs) in the rhizosphere soils of P. notoginseng after pathogen infection were determined. The effects of PAs on the growth of Fusarium oxysporum (F. oxysporum), a fungal pathogenic factor for P. notoginseng, as well as production of fusaric acid, a wilting agent for the plants, were also examined. The results indicate the presence of five PAs (ferulic acid, syringic acid, p-hydroxybenzoic acid, p-coumaric acid, and vanillic acid) in the rhizosphere soils of P. notoginseng, whose contents in the rhizosphere soils of healthy plants are higher than those of the diseased ones. Further we found that individual PA could inhibit the mycelium growth and spore production of F. oxysporum, but stimulate fusaric acid production as well, disclosing the double-edge sword role of PAs in the occurrence of root rot of P. notoginseng and paving the way for the intervention of P. notoginseng root rot via balancing PAs.

Abstract:Zanthoxylum armatum, a significant forest plant in southwestern China, is crucial for preserving soil and water resources. However, the presence of root rot disease has led to plant death, impacting the pepper sector. Effective control measures for this disease are still lacking. Rhizosphere microorganisms play a vital role in plant health by inhibiting plant pathogens and inducing plant resistance. This research aimed to isolate and characterize the pathogen responsible for root rot disease in Z. armatum. Comparative analysis of fungal and bacterial communities in the rhizosphere soil of healthy and diseased plants revealed Fusarium solani as the pathogenic fungus causing root rot disease. Diseased plants had a higher occurrence of Fusarium spp., while disease-free plants had a higher abundance of ecologically beneficial microbial communities that could potentially serve as biocontrol agents. Three bacterial strains (Bacillus subtilis, Bacillus amyloliquefaciens, and Bacillus siamensis) were identified as effective biocontrol agents, inhibiting the growth of the pathogenic fungus F. solani both in vivo and in vitro. This study deepens our understanding of the rhizosphere soil microbial community differences between diseased and healthy Z. armatum, providing potential biocontrol bacteria to enhance plant resistance against root rot disease.

Characterizing the Pathogenicity and Mycotoxin Production Capacity of Fusarium spp. Causing Root Rot of Angelica sinensis in China

Abstract:Root rot is a very destructive soil-borne disease, which severely affects the quality and yield of Angelica sinensis in major planting areas of Gansu Province, China. Twelve Fusarium strains were identified from root rot tissue and infected soil in the field by comparing each isolate strain internal transcribed spacer, translation elongation factor 1-α sequence and RNA polymerase second largest subunit gene with the sequences of known fungal species in the NCBI database. Of these isolates, four were F. acuminatum, followed by three F. solani, two F. oxysporum, and one each of F. equiseti, F. redolens, and F. avenaceum. Under greenhouse conditions, pathogenicity testing experiment was carried out using five strains: two F. acuminatum, one F. solani, one F. oxysporum, and one F. equiseti. Among them, the incidence of F. acuminatum-induced root rot on A. sinensis was 100%; hence, it was the most aggressive. Liquid chromatography was used to show that F. acuminatum could produce neosolaniol (NEO), deoxynivalenol, and T-2 toxins. Of these, the level of NEO produced by F. acuminatum was high compared with the other two toxins. By isolating Fusarium spp. and characterizing their toxin-producing capacity, this work provides new information for effectively preventing and controlling A. sinensis root rot in the field as well as improving the quality of its medicinal materials.

Abstract:We present a fast accelerated prox-level (FAPL) method for large scale ball constrained and unconstrained convex optimization. It achieves optimal iteration complexity in theory, and reduces computation time and increases accuracy significantly in practice. This is accomplished by reducing the number of sub-problems involved in most existing bundle level type methods, and the novel algorithm to solve the sub-problem exactly. Our numerical results on solving large-scale least square problems and total variation based image reconstruction have shown great advantages of the FAPL methods over several state-of-the-art firstorder methods.
Transcriptome Analysis Reveals the Biocontrol Mechanism of Endophytic Bacterium AM201, Rhodococcus sp., against Root Rot Disease of Atractylodes macrocephala

Abstract:Atractylodes macrocephala Koidz (AMK) is a perennial herb from the plant family Asteraceae (formerly Compositae). This herb is mainly distributed in mountainous wetlands in Zhejiang, Sichuan, Yunnan, and Hunan provinces of China. Its medicinal production and quality, however, are severely impacted by root rot disease. In our previous study, endophytic bacterium designated AM201 exerted a high biocontrol effect on the root rot disease of AMK. However, the molecular mechanisms underlying this effect remain unclear. In this study, the identity of strain AM201 as Rhodococcus sp. was determined through analysis of its morphology, physiological and biochemical characteristics, as well as 16S rDNA sequencing. Subsequently, we performed transcriptome sequencing and bioinformatics analysis to compare and analyze the transcriptome profiles of root tissues from two groups: AM201 (AMK seedlings inoculated with Fusarium solani [FS] and AM201) and FS (AMK seedlings inoculated with FS alone). We also conducted morphological, physiological, biochemical, and molecular identification analyses for the AM201 strain. We obtained 1,560 differentially expressed genes, including 187 upregulated genes and 1,373 downregulated genes. We screened six key genes ( GOLS2 , CIPK25 , ABI2 , egID , PG1 , and pgxB ) involved in the resistance of AM201 against AMK root rot disease. These genes play a critical role in reactive oxygen species (ROS) clearance, Ca 2+ signal transduction, abscisic acid signal inhibition, plant root growth, and plant cell wall defense. The strain AM201 was identified as Rhodococcus sp. based on its morphological characteristics, physiological and biochemical properties, and 16S rDNA sequencing results. The findings of this study could enable to prevent and control root rot disease in AMK and could offer theoretical guidance for the agricultural production of other medicinal herbs.

Abstract:Apple replant disease (ARD) is a significant factor restricting the healthy development of the apple industry. Biological control is an important and sustainable method for mitigating ARD. In this study, a strain of Paenibacillus polymyxa GRY-11 was isolated and screened from the rhizosphere soil of healthy apple trees in old apple orchards in Shandong Province, China, and the effects of strain GRY-11 on soil microbial community and ARD were studied. The result showed that P. polymyxa GRY-11 could effectively inhibit the growth of the main pathogenic fungi that caused ARD, and the inhibition rates of the strain against Fusarium moniliforme , Fusarium proliferatum , Fusarium solani , and Fusarium oxysporum were 80.00%, 71.60%, 75.00%, and 70.00%, respectively. In addition, the fermentation supernatant played an active role in suppressing the growth of pathogenic fungi. The results of the pot experiment showed that the bacterial fertilizer of the GRY-11 promoted the growth of Malus hupehensis seedlings, improved the activity of protective enzymes in plant roots, enhanced the soil enzyme content, and optimized the soil microbial environment. In general, the GRY-11 can be used as an effective microbial preparation to alleviate ARD. Our study offers novel perspectives for the prevention of ARD.

Arbuscular mycorrhizal fungi as a promising biomaterial for controlling root rot of Panax notoginseng

Abstract:Root rot is a main disease in Panax notoginseng , which is severe and difficult to control. Arbuscular mycorrhizal fungi (AMF) are known to effectively prevent plant diseases caused by soil-borne pathogens. However, the potential role of AMF as a biocontrol agent against root rot in P. notoginseng is still unclear. This study aims to investigate the efficacy of AMF, including Funneliformis mosseae, Rhizophagus intraradices and Claroideoglomus etunicatum , individually and in combination, for controlling root rot in P. notoginseng seedlings. The effect of different AMF on the indicators of P. notoginseng growth, superoxide dismutase, peroxidase activity, proline, malondialdehyde, soluble sugars and proteins were measured. The effects of different AMF on root rot resistance were comprehensively evaluated using the subordinate function method. The results showed that the incidence of root rot decreased from 75% to 12.5%–50.0% after inoculation with different AMF agents, and the disease index decreased from 22.92% to 6.25%–16.67%. Moreover, AMF significantly increased the growth parameters of P. notoginseng , such as plant height (6.35%–41.73%), stem diameter (12.98%–45.96%), fresh weight (6.28%–95.35%), and leaf area (8.89%–97.71%). Moreover, AMF also improved the content of chlorophyll (2.40%–50.67%) and rate of net photosynthetic (52.08%– 279.17%), enhancing superoxide dismutase (4.02%–114.89%) and peroxidase activity (85.60%–139.98%), as well as promoting the accumulation of soluble sugars (30.21%–90.00%), proteins (3.31%–65.81%), and proline (4.02%–114.89%) in the root and leaf, while reducing the malondialdehyde content (19.99%–139.98%). Particularly, C. etunicatum was found to be the most effective in promoting growth and disease resistance in P. notoginseng . These findings provide insights into the potential use of AMF as an effective biocontrol agent to manage root rot.

Abstract:Plant diseases lead to a significant decline in the output and quality of Chinese herbal medicines. Actinomycetes play a vital role in the rhizosphere ecosystem. This is especially true for Streptomyces, which have become a valuable biological control resource because of their advantages in producing various secondary metabolites with novel structures and remarkable biological activities. The purpose of this study was to isolate an effective antagonistic actinomycete against the pathogen of corm rot in saffron. An antagonistic actinomycete, CM253, was screened from the rhizosphere soil samples of Crocus sativus, by plate co-culture with four pathogenic fungi (Fusarium oxysporum, Fusarium solani, Penicillium citreosulfuratum, and Penicillium citrinum). CM253 inhibited the growth and development of F. oxysporum hyphae by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Furthermore, by analyzing the degrading enzyme, the growth-promoting performance, and the whole genome of strain CM253, it was identified as Streptomyces yangpuensis, which produces NH3, protease, glucanase, cellulase, IAA, and ACC deaminase. In addition, 24 secondary metabolite synthesis gene clusters were predicted in antiSMASH. We identified genes encoding 2,3-butanediol; methionine; isoprene (metH, mmuM, ispEFH, gcpE, idi, and ilvABCDEH); biofilm formation; and colonization (upp, rfbBC, efp, aftA, pssA, pilD, fliA, and dhaM). Above all, S. yangpuensis CM253 showed the potential for future development as a biocontrol agent.

Abstract:Rusty root rot is the most destructive disease in ginseng production. The bacterial strain FG14 isolated from the rhizosphere of Panax ginseng was characterised as Bacillus amyloliquefaciens. The inhibition rates of the FG14 bacterial suspension against the mycelial growth, spore production and spore germination of Ilyonectria robusta which causes ginseng rusty root rot were all more than 90% at a concentration of 108 cfu/mL. The sterile fermentation broth of strain FG14 also had significant inhibitory ability. The results of plantgrowth promotion showed that the fresh weight and root length of ginseng seedlings treated with 3 × 108 cfu/mL FG14 bacterial suspension were 4.49 times and 2.12 times greater than the seedlings treated with I. robusta. The soil enzyme activities of urease, phosphatase, invertase, and catalase of strain FG14 and the expression of seven lipopeptide biosynthesis genes were up-regulated, and antifungal metabolite production was stable. The diversity and abundance of bacterial and fungal communities in rhizosphere soil changed during different growth stages of ginseng after inoculation with strain FG14. The relative abundance of the Ilyonectria genus was significantly lower than the water control at the leaf spreading stage of P. ginseng. These results indicate the great potential of B. amyloliquefaciens FG14 for use as a biological agent for the green control of ginseng rusty root rot.

Abstract:Breast conserving surgery and adjuvant radiotherapy has equivalent oncological outcomes to mastectomy and is the standard of care for treatment of early‐stage invasive breast cancer. Auditing is an essential component of ongoing quality assurance and clinical governance. It also serves to identify patient and tumor factors that have prognostic and therapeutic implications. The aim of this paper is to report on the clinical audit of treatment outcomes for patients undergoing adjuvant radiation treatment for early breast cancer at the Northern Sydney Cancer Care Centre.

Abstract:Polygonatum cyrtonema (giant Chinese Solomon's seal) is a perennial herb belonging to the family Liliaceae, with significant medicinal and dietary value. However, a severe rhizome rot has led to a decline in the yield and quality of this species. In this study, a fungus was isolated from symptomatic rhizome samples and identified as Fusarium commune through molecular and morphological analyses. In pathogenicity tests, the identification of F. commune as the causative agent of rhizome rot was confirmed by fulfilling Koch's postulates. Biological characteristics indicated that the optimal growth conditions for F. commune were a temperature of 25°C, pH of 8, carbon source of soluble starch, nitrogen source of peptone and a photoperiod of 24 h light. In order to reduce the use of chemical fungicides and explore natural substances to control the disease, the sensitivity of F. commune to seven phytochemicals was studied. Four phytochemicals (honokiol, 2‐allylphenol, magnolol and carvacrol) showed inhibitory activity, with honokiol having a 50% of the maximal effect concentration (EC50) of 9.99 ± 0.14 mg/L. The results of pot experiment showed that the four phytochemicals had significant control efficacy on the rhizome rot of P. cyrtonema, and the highest control efficacy was observed with honokiol. These findings provide a scientific basis for the control of rhizome rot in P. cyrtonema.

Abstract:Root rot disease is a serious infection leading to production loss of Chinese wolfberry ( Lycium barbarum ). This study tested the potential for two bacterial biological control agents, Bacillus amyloliquefaciens HSB1 and FZB42, against five fungal pathogens that frequently cause root rot in Chinese wolfberry. Both HSB1 and FZB42 were found to inhibit fungal mycelial growth, in vitro and in planta , as well as to promote the growth of wolfberry seedlings. In fact, a biocontrol experiment showed efficiency of 100% with at least one treatment involving each biocontrol strain against Fusarium oxysporum . Metagenomic sequencing was used to assess bacterial community shifts in the wolfberry rhizosphere upon introduction of each biocontrol strain. Results showed that HSB1 and FZB42 differentially altered the abundances of different taxa present and positively influenced various functions of inherent wolfberry rhizosphere bacteria. This study highlights the application of biocontrol method in the suppression of fungal pathogens that cause root rot disease in wolfberry, which is useful for agricultural extension agents and commercial growers.
