Abstract:CONTEXT: Inflammation and cell differentiation lead to a number of severe diseases. In the recent years, various studies focused on the anti-inflammatory and anticancer activity of essential oils (EOs) of numerous plants, including different Pinus species.OBJECTIVE: The phytochemical composition, anti-inflammatory and cytotoxic activity of EOs from needles and twigs of Pinus heldreichii Christ (Pinaceae) and P. peuce Griseb., and from needles, twigs and cones of P. mugo Turra were determined.MATERIALS AND METHODS: For separation and identification of the EOs, gas chromatography/flame ion detector (GC/FID) and GC/mass spectrometry were performed. The amount of secreted IL-6 in a lipopolysaccharide (LPS)-stimulated macrophage model was quantified (concentration of oils: 0.0001-0.2%, 3 h incubation). Cytotoxicity on the cancer cell lines HeLa, CaCo-2 and MCF-7 were determined using a MTT (Thiazolyl Blue Tetrazolium Bromide) assay (concentration of oils: 0.001-0.1%, 24 h incubation).RESULTS: The most prominent members in the oils include: δ-3-carene, α-pinene and linalool-acetate (P. mugo); α-pinene, β-phellandrene and β-pinene (P. peuce); limonene, α-pinene and (E)-caryophyllene (P. heldreichii). EOs showed significant cytotoxic effects on cancer cell lines (IC50 0.007 to >0.1%), with a reduction in cell viability with up to 90% at a concentration of 0.1%, and anti-inflammatory activity (IC50 0.0008-0.02%) with a reduction of IL-6 secretion with up to 60% at a concentration of 0.01%.DISCUSSION AND CONCLUSION: The EOs of needles and twigs from P. peuce and P. heldreichii as well as of needles, twigs and cones of P. mugo can be considered as promising agents for anticancer and anti-inflammatory drugs.

Abstract:Background of the Study: Thuja orientalis L. is an indigenous medicinal plant belonging to family Cupressaceae found in Darjeeling Himalayan region, Dooars and Tarai region of India. The plant cone has been used traditionally as medicine to treat various diseases, like bronchitis, bacterial skin infection, osteoarthritis, trigeminal neuralgia. Aim: The aim of the present study is to evaluate the plant cone for phytochemical constinuents, and in vitro antioxidant, and antimicrobial properties. Place and Duration of Study: All the experiments were done in the Department of Biotechnology, University of North Bengal, Darjeeling, West Bengal, 734013, India. Methodology: Methanolic extract of T. orientalis cone was analyzed for phytochemicals by various biochemical methods. Antioxidant properties were analyzed by in vitro assays of DPPH, ABTS, NO and H2O2 scavenging. Antibacterial property was analyzed by agar well diffusion method and antifungal assay was monitored by radial growth bioassay. Results: Methanolic extract of T. orientalis cone contained flavonoid, phenol, saponin, tannin, terpenoid and alkaloid. The extract showed significant in-vitro antioxidant and antimicrobial activity. Conclusions: The study revealed that T. orientalis cone has potential as source of antioxidant, antibacterial and antifungal agents. Our further study is directed towards the isolation, and characterization of active compound from methanolic extract and evaluation of its potentiality against high blood sugar.
Exploration of Pharmacological Mechanism of Cinnamomum tamala Essential Oil in Treating Inflammation based on Network Pharmacology, Molecular Modelling, and Experimental Validation

Abstract:Background: Cinnamomum tamala (Buch.-Ham.) T.Nees & Eberm., also known as Indian Bay leaf, holds a distinctive position in complementary and alternative medicinal systems due to its anti-inflammatory properties. However, the active constituents and key molecular targets by which C. tamala essential oil (CTEO) exerts its anti-inflammatory action remain unclear. Objective: The present study used network pharmacology and experimental validation to investigate the mechanism of CTEO in the treatment of inflammation. Methods: GC-MS analysis was used to identify the constituents of CTEO. The key constituents and core targets of CTEO against inflammation were obtained by network pharmacology. The binding mechanism between the active compounds and inflammatory genes was ascertained by molecular docking and molecular dynamics simulation analysis. The pharmacological mechanism predicted by network pharmacology was verified in lipopolysaccharide-stimulated murine macrophage (RAW 264.7) cell lines. Results: Forty-nine constituents were identified by GC-MS analysis, with 44 constituents being drug-like candidates. A total of 549 compounds and 213 inflammation-related genes were obtained, revealing 68 overlapping genes between them. Compound target network analysis revealed cinnamaldehyde as the core bioactive compound with the highest degree score. PPI network analysis demonstrated Il-1β, TNF-α, IL8, IL6 and TLR4 as key hub anti-inflammatory targets. KEGG enrichment analysis revealed a Toll-like receptor signalling pathway as the principally regulated pathway associated with inflammation. A molecular docking study showed that cinnamaldehyde strongly interacted with the Il-1β, TNF-α and TLR-4 proteins. Molecular dynamics simulations and MMPBSA analysis revealed that these complexes are stable without much deviation and have better free energy values. In cellular experiments, CTEO showed no cytotoxic effects on RAW 264.7 murine macrophages. The cells treated with LPS exhibited significant reductions in NO, PGE2, IL-6, TNF-α, and IL-1β levels following treatment with CTEO. Additionally, CTEO treatment reduced the ROS levels and increased the antioxidant enzymes such as SOD, GSH, GPx and CAT. Immunofluorescence analysis revealed that CTEO inhibited LPS-stimulated NF-κB nuclear translocation. The mRNA expression of TLR4, MyD88 and TRAF6 in the CTEO group decreased significantly compared to the LPS-treated group. Conclusion: The current findings suggest that CTEO attenuates inflammation by regulating TLR4/MyD88/NF- κB signalling pathway.
Bioactivity of essential oils extracted from Cupressus macrocarpa branchlets and Corymbia citriodora leaves grown in Egypt

Abstract:BackgroundCupressus macrocarpa Hartw and Corymbia citriodora (Hook.) K.D. Hill & L.A.S. Johnson, widely grown in many subtropical areas, are used for commercial purposes, such as in perfumery, cosmetics, and room fresheners. Their potential as a source of antimicrobial compounds may be useful in different applications.MethodsThe chemical composition of essential oils (EOs) from C. macrocarpa branchlets and C. citriodora leaves was analyzed by using gas chromatography–mass spectrometry (GC/MS). Antibacterial and antifungal activities were assessed by the micro-dilution method to determine the minimum inhibitory concentrations (MICs), and minimum fungicidal concentrations (MFCs), and minimum bactericidal concentrations (MBCs). Further, the antioxidant capacity of the EOs was determined via 2,2′-diphenypicrylhydrazyl (DPPH) and β-carotene-linoleic acid assays.ResultsTerpinen-4-ol (23.7%), α-phellandrene (19.2%), α-citronellol (17.3%), and citronellal were the major constituents of EO from C. macrocarpa branchlets, and α-citronellal (56%), α-citronellol (14.7%), citronellol acetate (12.3%), isopulegol, and eucalyptol were the primary constituents of EO from C. citriodora leaves. Antibacterial activity with MIC values of EO from C. citriodora leaves was ranged from 0.06 mg/mL to 0.20 mg/mL, and MBC from 0.12 mg/mL against E. coli to 0.41 mg/mL. EO from C. macrocarpa branchlets showed less activity against bacterial strains. The MIC values against tested fungi of the EO from C. citriodora ranged from 0.11 to 0.52 mg/mL while for EO from C. macrocarpa from 0.29 to 3.21 mg/mL. The MIC and MFC values of EOs against P. funiculosum were lower than those obtained from Ketoconazole (KTZ) (0.20; 0.45; 0.29 and 0.53 mg/mL, respectively, vs 0.21 and 0.41 mg/mL. Antioxidant activity of the EO from C. citriodora was higher than that of the positive control but lower than that of the standard butylhydroxytoluene (BHT) (IC50 = 5.1 ± 0.1 μg/mL).ConclusionThe results indicate that the EO from Egyptian trees such as C. citriodora leaves may possesses strong bactericidal and fungicidal activities and can be used as an agrochemical for controlling plant pathogens and in human disease management which will add crop additive value.
Composition, in vitro Anti-inflammatory, Antioxidant and Antimicrobial Activities of Essential Oils from Leaf and Twig Parts of Cupressus cashmeriana

Abstract:The chemical composition and in vitro anti-inflammatory, antioxidant and antimicrobial activities of the essential oils isolated from the leaf and twig of Cupressus cashmeriana have been investigated. The essential oils were isolated using hydrodistillation in a Clevenger-type apparatus, and characterized by GC-FID and GC-MS, respectively. The leaf oil mainly consisted of alpha-pinene (21.8%), epizonarene (8.0%), sabinene (7.9%), limonene (7.6%), gamma-terpinene (7.0%), and allo-aromadendrene (7.0%); the twig oil was mostly carvacrol methyl ether (35.4%), manool (16.1%), carvacrol (14.2%), and (2Z,6E)-farnesol (6.9%). Twig oil, but not leaf oil, was able to reduce nitric oxide production by lipopolysaccharide-activated murine macrophages RAW 264.7 without reducing the cell viability. In addition, twig oil showed better antioxidant and antimicrobial activities than leaf oil. The major ingredient of the twig oil that was responsible for the anti-inflammatory, antioxidant and antimicrobial activities was carvacrol.
Analysis of chemical composition and biological efficiency of leaf essential oils isolated from seven species of Cinnamomum of the Western Ghats, India

Abstract:The chemical profiles of essential oils (EOs) of 7 Cinnamomum species ( C. sulphuratum , C. perotettii , C. verum , C. wightii , C. camphora , C. glanduliferum , and C. malabatrum ) composed from assorted parts of Western Ghats were analyzed. The antioxidant, antibacterial, and larvicidal potentialities of 7 essential oils were also investigated. Cinnamomum verum was observed to have the maximum yield (1.96%) of EO followed by C. malabatrum (1.71%). GC-MS analysis of EOs reported the highest amount of eugenol (38.04%), shyobunol (13.31%) in C. verum , whereas C. perrottetii identified 14–42 phytocompounds including terpenes, sesquiterpenes, monoterpenes, diterpenes, and phenylpropanoids, esters, aldehydes, and fatty acids. In the antibacterial activity, the higher zone of inhibition of C. malabatrum showed against S. pyogenes (28 ± 2.5 mm) and E. faecalis (27 ± 1.24 mm) followed by C. verum has shown notable inhibition zone against K. pneumoniae (26 ± 1.15 mm) and S. flexneri (24.33 ± 0.88 mm), with MIC value against C. verum , showed MIC value (1.17 ± 0.34 μg/mL) against S. flexneri, S. pyogenes, and K. pneumonia . All the Cinnamomum oil samples exhibited appreciable antioxidant radical scavenging activities with IC 50 values ranged 10.83–15.06 μg/mL. All the oil samples showed promising larvicidal activity against the late-third instar larvae of Aedes aegypti . Extending the present study with in vivo and in vitro animal studies will confirm the potentiality of Cinnamomum oils in the field of biomedicine to detect novel medicines.

Abstract:The complete amino acid sequence (72 amino acid residues) of a double-headed proteinase inhibitor from seeds of Vicia angustifolia L. var. segetalis Koch has been determined and compared with those of other double-headed inhibitors of known structure. Sequencing was performed by conventional methods with the aid of the fragments produced by reduction and S-carboxymethylation of the enzymatically modified inhibitors, and also using tryptic and chymotryptic peptides. The positions of the 14 half-cystine residues agreed among all the reported primary structures of the legume double-headed inhibitors. However, V. angustifolia inhibitor possessed extensive amino acid differences compared to the others. The phylogenetic relationship among these inhibitors was established using the unweighted pair-group method and revealed that the V. angustifolia inhibitor and the peanut inhibitor B-III had diverged at a relatively earlier stage compared to the other inhibitors.
Adaptive composition of services in context-aware ambient intelligent systems

Abstract:An environment of ambient intelligence or an ambient environment is an environment with intelligent applications able to adapt their behavior with the change of context. The ambient environments use many communicating objects; each one offers different and specific services. The majority of these services can perform only one specific task. The composition of services is then proposed to resolve the complex tasks. Multi agent systems underline the social capacities such as the communication, the cooperation, the resolution of conflict, the negotiation, the argumentation, and the emotion. They thus quickly became the principal paradigm in the ambient intelligence. In this context, we propose an adaptive multi agent system for automatic composition of services in an ambient environment. For each device is associated an agent. The set of the agents coordinate their competences taking into account the context in order to perform an activity. The system suggested is based on the Contract Net Protocol in which the agents exchange their proposals in the form of call for proposal. JADE platform (Java Agent DEvelopment framework) is used to implement our system.

Abstract:Introduction: Despite the increasing number of essential oils being reported on their potential therapeutic effects, some remain relatively unknown on their biological properties. That is the case of the essential oils obtained from copaiba ( Copaifera officinalis L.), wintergreen ( Gaultheria fragrantissima Wall.), everlasting ( Helichrysum italicum (Roth) G.Don) and clove ( Syzygium aromaticum (L.) Merr. & L.M.Perry), commonly labelled as being useful on the amelioration of conditions with an inflammatory background. Methods: To further broaden the current knowledge on the four essential oils, commercially available samples were approached on their effects upon a series of mediators that are involved on the inflammatory and oxidative response, both through in vitro cell-free and cell-based assays (5-lipoxygenase activity, lipid peroxidation, free radical and nitric oxide radical scavenging properties or tyrosinase inhibition). Results: The four oils proved to be active at some of the concentrations tested in most of the performed assays. Significant differences were found between the essential oils, S. aromaticum proving to tbe the most active, followed by G. fragrantissima against 5-lipoxygenase (5-LOX) and linoleic acid peroxidation, proving their potential use as antioxidants and anti-inflammatory agents. In fact, the IC 50 value of S. aromaticum in the 5-LOX assay was 62.30 μg mL −1 . Besides S. aromaticum efficiently scavenged superoxide radicals generated by xanthine/xanthine oxidase, displaying an IC 50 value of 135.26 μg mL −1 . The essential oil obtained from H. italicum exhibited a significant decrease in the nitric oxide levels on BV-2 cells, showing its potential as a cytoprotective agent against toxic damage. Copaiba oil ranked first as the most potent tyrosinase inhibitor, exhibiting an IC 50 98.22 μg mL −1 . Conclusion: More studies are needed to describe the essential oils properties, but these results confirm the potential of these essential oils as anti-inflammatory and antioxidant agents.

Abstract:The present study evaluated cytotoxic, anti-inflammatory, and anti-leishmanial properties of essential oil (EO) from fresh foliage of Eucalyptus camaldulensis Dehnh., and its major constituent compounds− 1,8-cineole, β -pinene and α -pinene. The chromatographic analysis of the hydrodistilled EO using gas chromatography-mass spectrometry (GC/MS) revealed the presence of 25 compounds which constituted 99.45% of EO. The EO comprised of 36% monoterpenoid hydrocarbons and 22.3% oxygenated monoterpenes. The most abundant constituent was 1,8-cineole (17.1%) followed by β -pinene (16.5%). In addition, significant quantities of β -eudesmol (10.9%), limonene (9.5%), and α -pinene (7.9%) were observed. Cell cytotoxicity concentration was observed to be highest in 1,8-cineole (CC 50 > 640 μg/mL), followed by α -pinene (CC 50 > 320 μg/mL), β -pinene (CC 50 > 160 μg/mL) and EO (CC 50 > 20 μg/mL). Investigation of anti-inflammatory potential by carrageenan-induced rat paw edema model revealed maximum inhibition in α -pinene (85.6% at 500 mg/kg body wt. and 81% at 250 mg/kg body wt.) after 3 h, which was comparable to indomethacin (positive control). Among the EO and its components, β -pinene exhibited maximum anti-leishmanial potential with IC 50 of 2.57 ± 0.15 mL. The study concluded that E. camaldulensis EO possesses significant anti-inflammatory and anti-leishmanial activity, thus suggesting the use of EO and its major constituents as a green drug that is a safer alternative to synthetic compounds.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Essential oils (EOs) are important active ingredients in Chinese herbal medicine. Cymbopogon citratus (DC.) Stapf, Cinnamomum cassia (L.) Presl, Salvia japonica Thunb., and Rosa rugosa Thunb. are used in local and traditional medicine to treat inflammation-related diseases. While the constituents and bioactivities of EOs of the above four Chinese herbals have been reported previously, but their anti-inflammatory and antioxidant activities have not been systematically evaluated.AIM OF THE STUDY: The purpose of this study was to research the chemical constituents, anti-inflammatory activities, and antioxidant activities of EOs of four Chinese herbal medicines.MATERIALS AND METHODS: Fresh plant materials were collected both in August 2018. The compositions of EOs were determined by the gas chromatography-mass spectrometry (GC-MS) method. TPA (12-O-tetradecylphophenol-13-acetate)-induced mouse ear model was evaluated the anti-inflammatory activities of EOs. The reduction of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), cyclooxygenase-2 (COX-2) and RelA protein of the nuclear factor kappa-B (NF-κB) in the signal pathway (P65) were measured. The antioxidant activities of EOs from four plants were determined by using 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging method.RESULTS: A total of 217 compounds were separated and identified from four EOs, mainly including trans-cinnamylaldehyde (68.75%), citronellal (38.16%), linalool (1.02-33.73%), geraniol (19.39%) and citronellol (17.18%). All four EOs reduced the damage of the ear tissue and had certain anti-inflammatory effects, and the EOs of C. citratus (CyCEOs) and S. japonica (SJEOs) had the best anti-inflammatory activities among others, even better than ibuprofen. The four EOs exhibited different DPPH radical-scavenging activities (IC50, 0.101-1.017%), where most of them were much lower than that of EOs of C. citratus (CiCEOs) (IC50, 0.101%) and EOs of R. rugosa (RREOs) (IC50, 0.142%).CONCLUSIONS: The main components determine diverse propertise of these four EOs, our results suggested that four EOs presented anti-inflammatory and antioxidant effect via reducing the expression of TNF-a, IL-6, COX-2, and NF-κB p65, which may provide a new approach for development of new anti-inflammatory drugs.

Abstract:Hedychium coronarium plant has attracted considerable attention from researchers due to its diverse phytochemical composition and potential therapeutic applications. The objective of this study was to evaluate the phytochemical profile and biological attributes of H. coronarium essential oils collected during different seasons in the Kumaun region of Uttarakhand. The essential oils were extracted from the rhizomes using hydrodistillation with a Clevenger‐type apparatus yielding between 0.07% to 0.38% (w/w). The major compounds identified by GC‐MS analysis exhibited seasonal variations and included 1,8‐cineole (26.6‐38.5%), coronarin E (11.9‐18.8%), α‐pinene (5.00‐14.9%), α‐terpineol (4.2‐9.3%), (E)‐β‐caryophyllene (1.2‐9.7%), and linalool (0.4‐2.9%). To illustrate the variations in oil composition, visualization techniques such as heat map and PCA were employed. The essential oils demonstrated potential biological activity in all tests conducted. The molecular modeling study indicated that the potential mechanism may be associated with acetylcholinesterase. Consequently, this study contributes to the development of novel natural pesticides.

Abstract:Ethnopharmacological relevance: The genus Hedychium of family Zingiberaceae comprises several perennial rhizomatous species widely used in perfumery and traditional folk medicine to treat diseases related to asthma, diarrhoea, nausea, stomach disorders, inflammation and tumours. Several species of Hedychium have remained under-explored with respect to their chemical composition and biological activities. Aim of the study: The current research aimed to explore the chemical composition and evaluate the antiproliferative and anti-inflammatory activities of rhizome essential oil from four Hedychium species (H. coccineum, H. gardnerianum, H. greenii and H. griffithianum). Materials and methods: Compound identification was accomplished using a Clarus 580 gas chromatography system in conjunction with mass spectrometry (GC-MS). The multivariate data statistics using chemometrics (PCA, PLS-DA, sPLS-DA) and cluster analysis (Dendrogram, Heat maps, K-means) were used to compare the similarity and relationship among Hedychium metabolomes. MTT assay was employed to visualize the antiproliferative property against MCF7, HepG2 and PC3 cancerous cell lines. The toxicity of essential oils was determined using 3T3-L1 non-tumorigenic/normal cells. Lipopolysaccharide (LPS)-induced RAW 264.7 cells were used to investigate the anti-inflammatory properties of Hedychium essential oils by measuring the production of nitric oxide (NO) using the Griess reagent method. Furthermore, the levels of prostaglandin (PGE2) and pro-inflammatory cytokines (TNF-α, IL-6, IL-1β) was assessed using the ELISA technique. Results: In total, 82 compounds were identified in four targeted species of Hedychium from which 1,8-cineole (52.71%), β-pinene (32.83%), α-pinene (19.62%), humulene epoxide II (19.86%) and humulene epoxide I (19.10%) were the major constituents. Monoterpenes (8.5-59.9%) and sesquiterpenes (2.87-54.11%) were the two class of compounds, found as the most prevalent in the extracted essential oils. The multivariate analysis classified the four Hedychium species into three different clusters. Hedychium essential oils exhibited potent antiproliferative activity against MCF7, HepG2 and PC3 cancer cell lines with IC50 values less than 150 μg/mL where H. gardnerianum exhibited the highest selective cytotoxicity against human breast and prostate adenocarcinoma cells with an IC50 value of 44.04 ± 1.07 μg/mL and 56.11 ± 1.44 μg/mL, respectively. The essential oils on normal (3T3-L1) cells displayed no toxicity with higher IC50 values thereby concluding as safe to use for normal human health without causing any side effects. Besides, the essential oils at 100 μg/mL concentration revealed remarkable anti-inflammatory activity in LPS-activated RAW 264.7 murine macrophages by inhibiting the production of inflammatory mediators, with H. greenii exhibiting the maximum anti-inflammation response by significantly suppressing the levels of NO (84%), PGE2 (87%), TNF-α (94.3%), IL-6 (95%) and IL-1β (85%) as compared to LPS treated group. Conclusion: The present findings revealed that the Hedychium species traditionally used in therapeutics could be a potential source of active compounds with antiproliferative and anti-inflammatory properties.

Abstract:Ethnopharmacological relevance Trachyspermum roxburghianum (DC.) H. Wolff, commonly known as 'Ajamoda,' is a neglected Indian spice highly used in Ayurveda and folklore remedies as an antimicrobial for chronic wounds and discharges, along with a multitude of other disease conditions. Aim of the study The aim of study was to explore chemical composition and to investigate the antioxidant, antimicrobial, analgesic, and wound healing activities of T. roxburghianum fruit essential oil from India. Materials and methods The phytochemical characterization of the oil was determined through standard qualitative procedures and gas chromatography-mass spectrometry (GC-MS) technique. The in vitro antioxidant aptitude was assessed by the scavenging of DPPH and ABTS radicals. The antimicrobial potential of the oil was investigated using the disc diffusion method, followed by the determination of minimum inhibitory concentration against Gram-positive and Gram-negative bacterial and fungal strains. The analgesic potential was evaluated using thermal and chemically induced pain models in Swiss albino mice. Wound healing was assessed in vivo , including the determination of wound contraction rates, histopathology, and hydroxyproline estimation, using the excision wound model in Swiss albino mice. Results GC-MS analysis identified 55 compounds, with major terpenoids, including thymol (13.8%), limonene (11.5%), and others. Substantial radical-scavenging activity was exhibited by T. roxburghianum fruit essential oil (TREO) (IC 50 94.41 ± 2.00 μg/mL in DPPH assay and 91.28 ± 1.94 μg/mL in ABTS assay). Microorganisms were inhibited with low MIC (2 μL/mL for the inhibition of Staphylococcus aureus and Bacillus subtilis ; 4 μL/mL against Salmonella typhi and 16 μL/mL against Candida albicans ). In cytotoxicity study, no cytotoxicity was observed on Monkey Normal Kidney Cell line (Vero). Significant antinociceptive effects were observed (25.47 ± 1.10 % of inhibition at 100 mg/kg and 44.31 ± 1.69 % of inhibition at 200 mg/kg). A remarkable rate for wound closure and epithelization, along with a marked increase in hydroxyproline content were observed for the oil during wound healing in mice. Conclusions The results suggested that oil could be utilized as a potential source of wound healing therapeutics.

Abstract:Ethnopharmacological relevance: Prinsepiautilis (PU) Royle, native to the Himalayan region, is a deciduous thorny shrub with numerous traditional uses of its roots, leaves and seeds for treatment of conditions such as rheumatic pain, joint pain, arthritis, and inflammation. Aim of the study: Keeping in mind the growing demand of products of natural origin as alternate medicine, the present study was undertaken to scientifically validate for the first time the traditional claims of healing pain and inflammation by evaluating the fatty oil isolated from the seeds using established in vitro and in vivo models. Materials and methods: PU Seeds were Soxhlet extracted using n-hexane and fatty oil was isolated. Chemical composition of the oil was established with the aid of Gas Chromatography-Flame Ionization Detection (GC-FID) and Gas Chromatography-Mass Spectrometry (GC-MS). The oil was then subjected to in vitro anti-inflammatory activity by following the established protocols of trypsin inhibitory and bovine serum albumin denaturation assays. The acute toxicity of the oil was also studied using OECD guidelines 423. The anti-inflammatory property of the oil was further evaluated using carrageenan-induced and formalin-induced edema in the rat paw. Moreover, hot plate latency and tail immersion assay were employed to evaluate analgesic activity of the oil. To establish the quality of the oil, various physicochemical properties were also studied. Results: GC-FID and GC-MS analysis of the oil revealed the presence of linoleic acid (59.06 ± 0.00%), oleic acid (28.11 ± 0.01%), palmitic acid (9.51 ± 0.01%) and stearic acid (3.32 ± 0.01%). In vitro trypsin inhibitory and bovine serum albumin denaturation assay revealed dose-dependent notable activity of the oil with IC50 value of 63.57 μg/mL and 518.14 μg/mL, respectively. The physico-chemical characterization demonstrated that the oil possesses a low acidity and a high oxidative stability index. The oil was found to be non-toxic and displayed effective anti-inflammatory activities with significant inhibition till 4 h in carrageenan-induced and formalin-induced rat paw edema at maximum tested dose of 200 mg/kg b.w. The oil also exhibited significant results in hot plate latency and tail immersion assay with positive effects showing up to 4 h after dose administration. Conclusion: These findings, besides supporting the traditional claims, suggest that P. utilis seed oil has potential therapeutic applications as a natural anti-inflammatory and analgesic agent. Further studies are warranted to explore its mechanisms of action and potential use in pharmaceutical and nutraceutical industries.

Abstract:Ethnopharmacological relevance: Hedychium coccineum rhizome is an anti-inflammatory ethnomedicine used to remedy inflammation-related swelling and bronchial asthma. Aim of the study: The study aimed to analyze the phytochemical constituents of H. coccineum rhizome essential oil (EO) and evaluate its in vitro and in vivo anti-inflammatory effects and underlying mechanisms. Materials and methods: Phytochemical constituents of H. coccineum rhizome EO were analyzed using GC-FID/MS. In RAW264.7 macrophages induced by LPS, blockade of PGE2, NO, IL-1β, IL-6, and TNF-α secretion by H. coccineum rhizome EO was measured, and then Western blot, qRT-PCR, and immunofluorescent staining were used to evaluate its underlying mechanisms. Moreover, we used the xylene-induced ear edema model for testing anti-inflammatory potential in vivo and examined auricular swelling as well as tissue and serum contents of IL-1β, IL-6, and TNF-α. Results: EO's main components were E-nerolidol (40.5%), borneol acetate (24.8%), spathulenol (4.5%), linalool (3.8%), elemol (3.5%), and borneol (3.4%). In RAW264.7 cells stimulated by LPS, EO downregulated the expression of pro-inflammatory enzyme (iNOS and COX-2) genes and proteins, thereby suppressing pro-inflammatory mediators (NO and PGE2) secretion. Simultaneously, it reduced TNF-α, IL-1β, and IL-6 release by downregulating their mRNA expression. Besides, H. coccineum EO attenuated LPS-stimulated activation of NF-κB (by reducing IκBα phosphorylation and degradation to inhibit NF-κB nuclear translocation) and MAPK (by downregulating JNK, p38, and ERK phosphorylation). In xylene-induced mouse ear edema, EO relieved auricular swelling and lowered serum and tissue levels of TNF-α, IL-1β, and IL-6. Conclusions: H. coccineum EO had powerful in vivo and in vitro anti-inflammatory effects by inhibiting MAPK and NF-κB activation. Hence, H. coccineum EO should have great potential for application in the pharmaceutical field as a novel anti-inflammatory agent.

Abstract:Pogostemon cablin Linn., commonly known as patchouli is an aromatic plant with immense use in aroma and perfumery. The purpose of this study was to examine and compare the chemical composition, antimicrobial activity, and various biological activities of Pogostemon cablin Linn flower and leaf essential oils. The compositional analysis was carried out using GC/MS, while standard biological experiments were used to determine the pharmacological potential of both the essential oils. Patchouli alcohol (27.52%, 44.52%) was found to be the major compound followed by caryophyllene (18.23%, 12.86%) for both flowers and leaves essential oil (EO) respectively. P. cablin flower (PCFEO) and leaf essential oil (PCLEO) exhibited weak to strong antimicrobial activities against selected microbial strains. The mitotic index for flower and leaf EO was 14.38% and 15.20% respectively. PCFEO and PCLEO had chromosomal aberrations of 9.0% and 9.60%, respectively. Both PCFEO and PCLEO had substantial antioxidant activity, with PCLEO exhibiting the highest antioxidant activity (IC 50 = 1.31 μg/mL). Highest anti-inflammatory activity was exhibited by PCLEO (IC 50 = 4.49 μg/mL). Similarly, the anti-diabetic ability of PCLEO (IC 50 = 14.19 μg/mL) was greater than the PCFEO (IC 50 = 15.60 μg/mL). The PCLEO (IC 50 = 7.42 μg/mL) and PCFEO (IC 50 = 12.67 μg/mL) both exhibited better anticholinesterase activity than galanthamine (IC 50 = 22.16 μg/mL). Better tyrosinase inhibitory activity was shown by PCLEO with IC 50 = 4.03 μg/mL than PCFEO IC 50 = 4.44 μg/mL. Apart from their antimicrobial activity, the PCLEO was found to outperform both PCFEO as well as the standards in their respective assays. Thus, the present study affirmed the wide application aspects of patchouli essential oil apart from its aromatic application.
