Verification of AKT and CDK5 Gene and RNA Interference Combined with Irradiation to Mediate Fertility Changes in Plutella xylostella (Linnaeus)

Abstract:Plutella xylostella (Linnaeus) mainly damages cruciferous crops and causes huge economic losses. Presently, chemical pesticides dominate its control, but prolonged use has led to the development of high resistance. In contrast, the sterile insect technique provides a preventive and control method to avoid the development of resistance. We discovered two genes related to the reproduction of Plutella xylostella and investigated the efficacy of combining irradiation with RNA interference for pest management. The results demonstrate that after injecting PxAKT and PxCDK5, there was a significant decrease of 28.06% and 25.64% in egg production, and a decrease of 19.09% and 15.35% in the hatching rate compared to the control. The ratio of eupyrene sperm bundles to apyrene sperm bundles also decreased. PxAKT and PxCDK5 were identified as pivotal genes influencing male reproductive processes. We established a dose-response relationship for irradiation (0–200 Gy and 200–400 Gy) and derived the irradiation dose equivalent to RNA interference targeting PxAKT and PxCDK5. Combining RNA interference with low-dose irradiation achieved a sub-sterile effect on Plutella xylostella, surpassing either irradiation or RNA interference alone. This study enhances our understanding of the genes associated with the reproduction of Plutella xylostella and proposes a novel approach for pest management by combining irradiation and RNA interference.

Abstract:The sterile insect technique (SIT) is widely used for the inundative release of sterile mass-reared males to control lepidopteran pests. SIT based on X-ray irradiation is an eco-friendly alternative to chemical control. However, its use in Ephestia elutella , a stored tobacco pest currently controlled with insecticides, is poorly explored. This study aims to investigate the effects of X-ray irradiation on E. elutella to determine the optimal sterilizing dose and processing developmental stage for improving SIT application. The pupal stage was most suitable for irradiation that was more tolerant than the other insect stages including eggs, larvae, and adults. Subsequently, male pupae were irradiated with X-ray doses of 0, 50, 100, 150, 200, 250, and 300 Gy and mated with unirradiated females. Their emergence, longevity, egg number, egg hatch rate, developmental duration, survival rate, induced sterility, and male mating competitiveness were evaluated. The results suggest that a dose of 200 Gy can be applied to effectively induce sterility in male pupae, after which induced sterility and male mating competitiveness can be balanced by increasing the release ratio (sterile:normal). When the release ratio was 15:1, it was found that 71.91% of the wild population could be suppressed. The results of this study show that the SIT based on X-ray irradiation can be successfully used to manage E. elutella, improves our understanding of the biological effects of the SIT, and expands its future application to the control of other pests.

Abstract:Irradiation can be used as an insect pest management technique to reduce post-harvest yield losses. It causes major physiological changes, impairing insect development and leading to mortality. This technique is used to control the melon fly Zeugodacus cucurbitae , a major pest of Cucurbitaceae in Asia. Here, we applied irradiation to melon fly eggs, and the larvae emerged from irradiated eggs were used to conduct comparative transcriptomics and thereby identify key genes involved in the development and survival. We found 561 upregulated and 532 downregulated genes in irradiated flies compared to non-irradiated flies. We also observed abnormal small-body phenotypes in irradiated flies. By screening the 532 downregulated genes, we selected eight candidate genes putatively involved in development based in described functions in public databases and in the literature. We first established the expression profile of each candidate gene. Using RNA interference (RNAi), we individually knocked down each gene in third instar larvae and measured the effects on development. The knockdown of ImpE2 ecdysone-inducible gene controlling life stage transitions–led to major body size reductions in both pupae and adults. The knockdown of the tyrosine-protein kinase-like tok (Tpk-tok) caused severe body damage to larvae, characterized by swollen and black body parts. Adults subject to knockdown of the eclosion hormone ( Eh_1 ) failed to shed their old cuticle which remained attached to their bodies. However, no obvious developmental defects were observed following the knockdown of the heat shock protein 67B1-like (Hsp67), the insulin receptor ( Insr ), the serine/threonine-protein kinase Nek4 (Nek4), the tyrosine-protein kinase transmembrane receptor Ror ( Ror_1 ) and the probable insulin-like peptide 1 ( Insp_1 ). We argue that irradiation can be successfully used not only as a pest management technique but also for the screening of essential developmental genes in insects via comparative transcriptomics. Our results demonstrate that ImpE2 and Eh_1 are essential for the development of melon fly and could therefore be promising candidates for the development of RNAi-based pest control strategies.

Abstract:The sterile insect technique (SIT) is widely used to control Lepidopteran pests by inducing inherited sterility. The noctuid moth Xestia c-nigrum is a polyphagous pest whose subterranean larvae severely injure cereals and some vegetables. The goals of this study were to assess the impact of X-ray irradiation on the development and survival of X. c-nigrum and use the data to select suitable sterilizing doses for potential future use in pest management. Batches of male pupae were exposed to 0 (control), 10, 30, 50, 100, 200, 300, or 400 Gy of X-rays, approximately 24 h before adult emergence. Exposure of late-stage pupae to 10–200 Gy of radiation had no significant effect on adult emergence, but all doses (10–400 Gy) reduced adult longevity, the number of spermatophores in mated females, and the number of eggs laid per female in the irradiated parental generation compared with the controls. Exposure to 10 and 30 Gy had no significant effects in the F1 generation on 1) the rate of egg hatch, 2) the duration of larval or pupal development, or 3) adult longevity. However, exposure to 50 Gy reduced the rate of egg hatch in the F1 generation, and when male pupae were exposed to 100 Gy only 1% of the F1 eggs hatched. Also at 100 Gy, the developmental durations of larvae and pupae were significantly prolonged, and longevity of adult moths was reduced. There were no significant differences between the control group and any treatments in 1) the sex ratio of the F1 adults, 2) the duration of F1 pre-oviposition or oviposition periods, or 3) the number of eggs laid per F1 female. Our findings indicate that a dose of 100 Gy can effectively slow pest development and reduce larval survival in the F1 generation. In addition, F1 adults from lines treated with 100 Gy were able to mate and lay eggs, but all F2 eggs failed to hatch. Our results suggest that use of X-ray irradiation has potential to control this polyphagous pest at the regional level.

Abstract:Glyphodes pyloalis Walker (Lepidoptera: Pyralididae) is a common pest in sericulture and has developed resistance to different insecticides. However, the mechanisms involved in insecticide resistance of G. pyloalis are poorly understood. Here, we present the first whole-transcriptome analysis of differential expression genes in insecticide-resistant and susceptible G. pyloalis. Clustering and enrichment analysis of DEGs revealed several biological pathways and enriched Gene Ontology terms were related to detoxification or insecticide resistance. Genes involved in insecticide metabolic processes, including cytochrome P450, glutathione S-transferases and carboxylesterase, were identified in the larval midgut of G. pyloalis. Among them, CYP324A19, CYP304F17, CYP6AW1, CYP6AB10, GSTs5, and AChE-like were significantly increased after propoxur treatment, while CYP324A19, CCE001c, and AChE-like were significantly induced by phoxim, suggesting that these genes were involved in insecticide metabolism. Furthermore, the sequence variation analysis identified 21 single nucleotide polymorphisms within CYP9A20, CYP6AB47, and CYP6AW1. Our findings reveal many candidate genes related to insecticide resistance of G. pyloalis. These results provide novel insights into insecticide resistance and facilitate the development of insecticides with greater specificity to G. pyloalis.
Comparative Virulence of Zoophthora Radicans Isolates from Different Hosts Against Plutella Xylostella Larvae

Abstract:Four Zoophthora radicans (Entomophthorales) isolates from different hosts were bioassayed against the second instar larvae of the diamondback moth, Plutella xylostella in the laboratory. Each isolate included eight dosages resulting from spore showers at different times. On day 8 after exposure to a spore shower for inoculation, a P. xylostella derived isolate, ARSEF1100, caused a cumulative mortality of 2 38%-97 44% at 0 53-319 32 conidia/mm 2 with all cadavers exhibiting typical Zoophthora syndrome. Two isolates derived from Empoasca, ARSEF2699 and F99101, killed 2 38%-50 00% and 2 38%-57 89% at 1 56-314 84 and 1 84-484 08 conidia/mm 2, respectively, whereas the isolate ARSEF1342 from Pieris brassicae, killed only 6 52%-13 63% at 3 54-633 40 conidia/mm 2. Typical Zoophthora syndrome was observed only in approximately one third of cadavers killed by the latter three isolates. Fitting the data to a time dose mortality model generated the estimates of parameters for time and dose effects of each isolate. The parameter estimates for dose effect (β) was 1 89 for ARSEF1100, 1 48 for F99101, 1 23 for ARSEF2699, and 0 37 for ARSEF1342, which are significantly different. The LD 50 values estimated for ARSEF1100 were 231 68,113 08, 71 41, 40 87 and 35 30 conidia/mm 2 on days 4-8 after exposure, respectively. The corresponding estimates were 1 344 43,922 39,555 58,410 06 and 397 07 conidia/mm 2 for ARSEF2699, and 666 86,451 64,413 82,350 65 and 332 57 conidia/mm 2 for F99101 The virulence for ARSEF1342 was too weak for its LD 50 to be estimated. The isolate ARSEF1100 was the most virulent to P. xylostella, and can be considered as a microbial agent for control of the pest.
Side effects of X-ray irradiation on flight ability of Cydia pomonella moth

Abstract:Background: The sterile insect technique (SIT) has proven to be an effective approach in managing the population of major invasive pests. Our previous studies showed that irradiation of Cydia pomonella males at a dosage of 366 Gy X-rays resulted in complete sterility. However, the mating competitiveness of sterilized males is significantly compromised, which can be attributed to a decline in their ability to fly. Results: In this study, we examined the flight patterns of both male and female adults of C. pomonella. The results revealed significant variations in the average flight speed of both genders at different stages of maturity, with females displaying longer flight duration and covering greater distances. Effect of irradiation on the flight performance of 3-day-old male moths was further evaluated, as they demonstrated the longest flight distance. The findings indicated a significant decrease in flight distance, duration, and average speed, due to wing deformities caused by irradiation, which also limited the dispersal distance of moths in orchards, as indicated by the mark-and-recapture assay. Reverse-transcription quantitative polymerase chain reaction analysis revealed a down-regulation of flight-related genes such as Flightin, myosin heavy chain, and Distal-less following radiation exposure. Conclusion: These findings demonstrate that X-ray irradiation at a radiation dose of 366 Gy has a detrimental effect on the flight ability of male C. pomonella adults. These insights not only contribute to a better understanding of how radiation sterilization diminishes the mating competitiveness of male moths, but also aid in the development and improvement of SIT practices for the effective control of C. pomonella. © 2023 Society of Chemical Industry.

Abstract:Spodoptera litura is an omnivorous pest that has spread globally. Because irradiation sterilization technology has a great potential for control of S. litura , the effect of 25–150 Gy doses of X-rays on pupal survival, flight and reproductive variables of adult moths were analyzed in this research. The X-ray irradiation with the dose of 25–150 Gy significantly affected the reproductive ability of females. Irradiating male pupae with 25–150 Gy doses of X-rays had no effect on mating, life span, or flight ability of adult moths, but significantly reduced survival and fecundity of their offspring, and the sterility rate of the F 1 generation was 52.65%–99.9%. The results of logistic curve fitting showed that the sterility impact was 84% at the most appropriate irradiation dose (71.26 Gy). The sterility control was 91% in an indoor mating competition experiment when the release ratio of irradiated males (75 Gy) to nonirradiated males reached 12.6:1. The effects of X-ray irradiation doses on biological variables of S. litura and the most effective release ratio determined here provide a theoretical foundation for using radiation sterilization technology to control S. litura .

Abstract:The sterile insect technique (SIT) is considered an environmentally friendly, autocidal control tactic to manage insect pests. This work dealt with the improvement of quality management of the Indian meal moth Plodia interpunctella (Hübner) for enhanced efficacy of the SIT. The results indicated that egg hatching of irradiated mature eggs of P. interpunctella was higher than that of younger eggs, indicating that mature eggs were significantly more tolerant than younger eggs. Moreover, our data revealed that a dose of 500 Gy completely prevented pupal formation in irradiated young and mature larvae. Crosses between irradiated and non-irradiated adults resulted in considerable variations in fecundity. The mating competitiveness index (CI) value was higher for a ratio of 5:1:1 (sterile male, fertile male, and fertile female, respectively) as compared with the ratio 1:1:1 for the irradiated individuals of all life stages. Low temperature (5 °C) maintenance of irradiated pupae significantly affected adult emergence. Using cylinders to assess flight ability, we found that the flight performance of adults that were developed from cold treated irradiated pupae was influenced by cylinder diameter, cylinder height and the number of hours the insects were in the cylinders. The percentage of DNA damage of the reproductive organs of adults developed from cold treated pupae that were irradiated with 100 and 150 Gy varied significantly. The results of this study should be used to implement pilot-scale field tests aiming at a sterile- to-fertile male ratio of 5 to 1.

Abstract:Plutella xylostella is one of the most destructive pests for cruciferous vegetables, and is adaptability to different environmental stressors. However, we still know little about the molecular mechanisms of how P. xylostella adapt to thermal stress. Here, the comparative transcriptome analysis was conducted from the samples of control (27 °C, CK) and heat treatment (40 °C, 40 T) P. xylostella. The results showed 1253 genes were differentially expressed, with 624 and 629 genes up- and down-regulated respectively. The annotation analysis demonstrated that "Energy production and conversion", "Protein processing in endoplasmic reticulum", "Peroxisome" and "Tyrosine metabolism" pathways were significantly enriched. Additionally, we found the expression levels of heat shock protein genes (Hsps), cuticle related genes and mitochondrial genes were significantly up-regulated in 40 T insects, suggesting their vital roles in improving adaption to heat stress. Importantly, the SOD activity and MDA content of P. xylostella were both identified to be increased under high temperature stress, indicating the elevated antioxidant reactions might be involved in response to heat stress. In conclusion, the present study offered us an overview of gene expression changes after 40 °C treatments, and found some critical pathways and genes of P. xylostella might play the critical roles in resisting heat stress.
Effect of Irradiation on Reproduction of Female Spodoptera litura (Fabr.) (Lepidoptera: Noctuidae) in Relation to the Inherited Sterility Technique

Abstract:Radiobiological investigations on the reproductive behavior of female Spodoptera litura (Fabr.) were conducted with the aim of determining the suitable radio-sterilizing dose for females in order to release them along with sub-sterile males for effective implementation of the Inherited Sterility technique against this pest. Calling and copulation duration significantly increased, while mating success, oviposition, fertility and longevity significantly decreased with increasing radiation dose (100–200 Gy) compared to control. In view of the effect of irradiation on mating behavior and reproductive viability of female S. litura, 130 Gy was identified as a suitable radio-sterilization dose. Further molecular studies were conducted to corroborate this dose for female sterilization, along with a higher dose of 200 Gy in order to validate the gradational response of ionizing radiation. GC-MS analysis indicated decreased sex pheromone titer at 130 Gy, which was more pronounced at 200 Gy. Pheromone-associated genes, PBAN and PBAN-R showed decreased expression at 130 Gy, and were drastically reduced at 200 Gy. The fertility-related Vg gene also showed a negative correlation with radiation exposure. Based on these radiation responses of female S. litura, 130 Gy might be considered a suitable dose for complete female sterility and its inclusion in sterile insect programs against S. litura.

Experimental autoimmune thyroiditis: modulation of the disease level in high and low responder mice by thymosin.

Abstract:Experimental autoimmune thyroiditis (EAT) in mice is characterized by production of anti-thyroglobulin autoantibodies and lymphoid infiltration of the thyroid gland. The pathogenesis of EAT is genetically controlled, but its mechanism is not yet clear. To investigate the mode of the expression of genetic control in the development of the disease, we attempted to modulate immune responses with thymosin fraction 5 (T-5) in mice which are either high or low responders to thyroglobulin. Severe thyroid lesions in high responder mice appear 2-3 weeks after immunization, while low responders develop very mild or no lesions. High responder mice have also higher antibody titer than the low responder strain. T-5 administered in five daily injections before or simultaneously with immunization, was strongly suppressive to EAT development in the high responder strain while there was no effect on thyroiditis level in low responder mice. T-5 also decreased the severity of thyroiditis in high responder mice when injected 2-4 weeks after immunization. In low responder mice, however, the same treatment increased the thyroiditis level. The effects of late injection of T-5 were dose-dependent. In all experiments, the antibody titres were not affected by T-5 treatment. The results demonstrate the ability of T-5 to modulate development and intensity of EAT. The contrasting effects of T-5 in high and low responder mice illustrate the relationship between the effects of T-5 and host immune status prior to study, suggesting that the level of immunoregulatory cell activity may differ in these two strains.
Identification and transcriptional profiling of UV-A-responsive genes in Bemisia tabaci

Abstract:Ultraviolet-A (UV-A) radiation directly impacts the growth and spread of Bemisia tabaci. However, the mechanistic pathways of this phenomenon remain unknown. We analyzed B. tabaci transcriptome data after exposure to UV-A radiation for 6 h. The 453 genes were identified whose expression were significantly altered in response to the stress induced by UV-A irradiation. Forty genes were up-regulated, while 413 genes were down-regulated. Enrichment analysis using GO, KEGG, and Genomes databases revealed that the DEGs play key roles in antioxidation and detoxification, protein turnover, metabolic, developmental processes, and immunological response. Among the gene families involved in detoxification, shock, and development, down-regulated DEGs in transcriptional factor gene families were significantly greater than those up-regulated DEGs. Our findings demonstrated that exposure to UV-A stress can suppress immunity and affect the growth and biological parameters of B. tabaci by altering gene regulation. These results suggest a potential utility of UV-A stress in managing B. tabaci under greenhouse conditions.

Abstract:Background: Plutella xylostella has become a notorious pest of cruciferous crops all over the world. Delta-endotoxins of Bacillus thuringiensis are widely used insecticidal proteins for controlling P. xylostella. However, the interaction mechanism of B. thuringiensis with the immune system of P. xylostella, at the genomic level, is still unclear. This study explored the immune response of P. xylostella to B. thuringiensis, at different time intervals, 6 h, 12 h, 18 h, 24 h, and 36 h, by using RNA-Sequencing (RNA-Seq) and RT-qPCR. Results: In total, 167 immunity-related genes were identified and placed into different families, including pattern recognition receptors (PRRs), signal modulators, immune pathways (Toll, IMD, and JAK/STAT), and immune effectors. It is worth mentioning that the analyses of the differentially expressed immunity-related genes revealed that most of the differentially expressed genes (DEGs) (87, 56, 76, 67, and 73 genes) were downregulated in P. xylostella following B. thuringiensis oral infection at 6 h, 12 h, 18 h, 24 h, and 36 h. Interestingly, our RNA-Seq analysis also revealed reduced expression of antimicrobial peptides, that play a vital role in the humoral immune system of P. xylostella. Conclusion: This study demonstrates that B. thuringiensis plays a novel role in controlling P. xylostella, by suppressing the immune system.

Abstract:Introduction: Plumbagin is an important phytochemical and has been reported to exhibit potent larvicidal activity against several insect pests, However, the insecticidal mechanism of plumbagin against pests is still poorly understood. This study aimed to investigate the insecticidal activities of plumbagin and the underlying molecular mechanisms against a devastating agricultural pest, the fall armyworm Spodoptera frugiperda . Methods: The effects of plumbagin on S. frugiperda larval development and the activities of two detoxification enzymes were initially examined. Next, transcriptomic changes in S. frugiperda after plumbagin treatment were investigated. Furthermore, RNA-seq results were validated by qPCR. Results: Plumbagin exhibited a high larvicidal activity against the second and third instar larvae of S. frugiperda with 72 h LC 50 of 0.573 and 2.676 mg/g, respectively. The activities of the two detoxification enzymes carboxylesterase and P450 were significantly increased after 1.5 mg/g plumbagin treatment. Furthermore, RNA-seq analysis provided a comprehensive overview of complex transcriptomic changes in S. frugiperda larvae in response to 1.5 mg/g plumbagin exposure, and revealed that plumbagin treatment led to aberrant expression of a large number of genes related to nutrient and energy metabolism, humoral immune response, insect cuticle protein, chitin-binding proteins, chitin synthesis and degradation, insect hormone, and xenobiotic detoxification. The qPCR results further validated the reproducibility and reliability of the transcriptomic data. Discussion: Our findings provide a valuable insight into understanding the insecticidal mechanism of the phytochemical plumbagin.

Mechanistic effects of microwave radiation on pupal emergence in the leafminer fly, Liriomyza trifolii

Abstract:Liriomyza trifolii is a significant pest of vegetable and ornamental crops across the globe. Microwave radiation has been used for controlling pests in stored products; however, there are few reports on the use of microwaves for eradicating agricultural pests such as L. trifolii, and its effects on pests at the molecular level is unclear. In this study, we show that microwave radiation inhibited the emergence of L. trifolii pupae. Transcriptomic studies of L. trifolii indicated significant enrichment of differentially expressed genes (DEGs) in 'post-translational modification, protein turnover, chaperones', 'sensory perception of pain/transcription repressor complex/zinc ion binding' and 'insulin signaling pathway' when analyzed with the Clusters of Orthologous Groups, Gene Ontology and the Kyoto Encyclopedia of Genes and Genomes databases, respectively. The top DEGs were related to reproduction, immunity and development and were significantly expressed after microwave radiation. Interestingly, there was no significant difference in the expression of genes encoding heat shock proteins or antioxidant enzymes in L. trifolii treated with microwave radiation as compared to the untreated control. The expression of DEGs encoding cuticular protein and protein takeout were silenced by RNA interference, and the results showed that knockdown of these two DEGs reduced the survival of L. trifolii exposed to microwave radiation. The results of this study help elucidate the molecular response of L. trifolii exposed to microwave radiation and provide novel ideas for control.

Abstract:Introduction: Heavy ion beam is a novel approach for crop mutagenesis with the advantage of high energy transfer line density and low repair effect after injury, however, little investigation on the biological effect on plant was performed. 50 Gy irradiation significantly stimulated the growth of Arabidopsis seedlings, as indicated by an increase in root and biomass, while 200 Gy irradiation significantly inhibited the growth of seedlings, causing a visible decrease in plant growth. Methods: The Arabidopsis seeds were irradiated by 12C6+. Monte Carlo simulations were used to calculate the damage to seeds and particle trajectories by ion implantation. The seed epidermis received SEM detection and changes in its organic composition were detected using FTIR. Evidence of ROS and antioxidant systems were analyzed. RNA-seq and qPCR were used to detect changes in seedling transcript levels. Results and discussion: Monte Carlo simulations revealed that high-dose irradiation causes various damage. Evidence of ROS and antioxidant systems implies that the emergence of phenotypes in plant cells may be associated with oxidative stress. Transcriptomic analysis of the seedlings demonstrated that 170 DEGs were present in the 50 Gy and 200 Gy groups and GO enrichment indicated that they were mainly associated with stress resistance and cell wall homeostasis. Further GO enrichment of DEGs unique to 50 Gy and 200 Gy revealed 58 50Gy-exclusive DEGs were enriched in response to oxidative stress and jasmonic acid entries, while 435 200 Gy-exclusive DEGs were enriched in relation to oxidative stress, organic cyclic compounds, and salicylic acid. This investigation advances our insight into the biological effects of heavy ion irradiation and the underlying mechanisms.

Transcriptome Analysis and Screening for Potential Target Genes for RNAi-Mediated Pest Control of the Beet Armyworm, Spodoptera exigua.

Abstract:The beet armyworm, Spodoptera exigua (Hübner), is a serious pest worldwide that causes significant losses in crops. Unfortunately, genetic resources for the beet armyworm is extremely scarce. To improve these resources we sequenced the transcriptome of S. exigua representing all stages including eggs, 1(st) to 5(th) instar larvae, pupae, male and female adults using the Illumina Solexa platform. We assembled the transcriptome with Trinity that yielded 31,414 contigs. Of these contigs, 18,592 were annotated as protein coding genes by Blast searches against the NCBI nr database. It has been shown that knockdown of important insect genes by dsRNAs or siRNAs is a feasible mechanism to control insect pests. The first key step towards developing an efficient RNAi-mediated pest control technique is to find suitable target genes. To screen for effective target genes in the beet armyworm, we selected nine candidate genes. The sequences of these genes were amplified using the RACE strategy. Then, siRNAs were designed and chemically synthesized. We injected 2 µl siRNA (2 µg/µl) into the 4(th) instar larvae to knock down the respective target genes. The mRNA abundance of target genes decreased to different levels (∼20-94.3%) after injection of siRNAs. Knockdown of eight genes including chitinase7, PGCP, chitinase1, ATPase, tubulin1, arf2, tubulin2 and arf1 caused a significantly high level of mortality compared to the negative control (P<0.05). About 80% of the surviving insects in the siRNA-treated group of five genes (PGCP, chitinase1, tubulin1, tubulin2 and helicase) showed retarded development. In chitinase1-siRNA and chitinase7-siRNA administered groups, 12.5% survivors exhibited "half-ecdysis". In arf1-siRNA and arf2-siRNA groups, the body color of 15% became black 48 h after injections. In summary, the transcriptome could be a valuable genetic resource for identification of genes in S. exigua and this study provided putative targets for RNAi pest control.
