Abstract:Olea europaea L. is the most valuable species of the Olea type, and its products offer a wide range of therapeutical uses. The olive tree has been extensively studied for its nourishing qualities, and the "Mediterranean diet", which includes virgin olive oil as a key dietary component, is strongly associated with a reduced risk of cardiovascular disease and various malignancies. Olive leaves, a by-product in the olive harvesting process, are valued as a resource for developing novel phytomedicines. For this purpose, two ethanolic extracts obtained from Olivae folium from Spain (OFS) and Greece (OFG) were investigated. Our findings contribute to a wider characterization of olive leaves. Both extracts displayed important amounts of phenolic compounds and pentacyclic triterpenes, OFG having higher concentrations of both polyphenols, such as oleuropein and lutein, as well as triterpenes, such as oleanolic acid and maslinic acid. The antioxidant capacity is similar for the two extracts, albeit slightly higher for OFG, possibly due to metal polyphenol complexes with antioxidant activity. The extracts elicited an antimicrobial effect at higher doses, especially against Gram-positive bacteria, such as Streptococcus pyogenes. The extract with lower inorganic content and higher content of polyphenols and triterpenic acids induced a strong anti-radical capacity, a selective cytotoxic effect, as well as antimigratory potential on A375 melanoma cells and antiangiogenic potential on the CAM. No irritability and a good tolerability were noted after evaluating the extracts on the in vivo Hen's Egg Test-Chorioallantoic Membrane (HET-CAM). Therefore, the present data are suggestive for the possible use of the two types of olive leaf products as high-antioxidant extracts, potentially impacting the healthcare system through their use as antimicrobial agents and as anticancer and anti-invasion treatments for melanoma.
Multiple antimelanoma potential of dry olive leaf extract

Abstract:Various constituents of the olive tree (Olea europaea) have been traditionally used in the treatment of infection, inflammation, prevention of chronic diseases, cardiovascular disorders and cancer. The anticancer potential of dry olive leaf extract (DOLE) represents the net effect of multilevel interactions between different biologically active compounds from the extract, cancer cells and conventional therapy. In this context, it was of primary interest to evaluate the influence of DOLE on progression of the highly malignant, immuno- and chemoresistant type of skin cancer-melanoma. DOLE significantly inhibited proliferation and subsequently restricted clonogenicity of the B16 mouse melanoma cell line in vitro. Moreover, late phase tumor treatment with DOLE significantly reduced tumor volume in a syngeneic strain of mice. DOLE-treated B16 cells were blocked in the G(0) /G(1) phase of the cell cycle, underwent early apoptosis and died by late necrosis. At the molecular level, the dying process started as caspase dependent, but finalized as caspase independent. In concordance, overexpression of antiapoptotic members of the Bcl-2 family, Bcl-2 and Bcl-XL, and diminished expression of their natural antagonists, Bim and p53, were observed. Despite molecular suppression of the proapoptotic process, DOLE successfully promoted cell death mainly through disruption of cell membrane integrity and late caspase-independent fragmentation of genetic material. Taken together, the results of this study indicate that DOLE possesses strong antimelanoma potential. When DOLE was applied in combination with different chemotherapeutics, various outcomes, including synergy and antagonism, were observed. This requires caution in the use of the extract as a supplementary antitumor therapeutic.

Abstract:The food products derived from Olea europaea are a fundamental part of the Mediterranean diet, and their health-promoting effects are well known. In this study, we analyzed the phytochemical characteristics, the redox state modulatory activity, and the cytotoxic effect of an olive leaf aqueous extract enriched by macroporous resin on different tumor and normal cell lines (LNCaP, PC3, HFF-1). HPLC-DAD analysis, the Folin–Ciocalteu and aluminum chloride methods confirmed the qualitatively and quantitatively high content of phenolic compounds (130.02 ± 2.3 mg GAE/g extract), and a DPPH assay (IC50 = 100.00 ± 1.8 μg/mL), the related antioxidant activity. The biological investigation showed a significant cytotoxic effect, highlighted by an MTT test and the evident cellular morphological changes, on two prostate cancer cell lines. Remarkably, the extract was practically non-toxic on HFF-1 at the concentrations (100, 150, 300 μg/mL) and exposure times tested. Hence, the results are selective for tumor cells. The underlying cytotoxicity was associated with the decrease in ROS production (55% PC3, 42% LNCaP) and the increase in RSH levels (>50% PC3) and an LDH release assay (50% PC3, 40% LNCaP, established necrosis as the main cell death mechanism.

Abstract:Twenty-four patients with ulnohumeral dislocation associated with radial head fracture were studied two to 35 years after injury. On the basis of an objective functional grading score that included elements of pain, motion, strength, and stability, results were excellent in three (12%), good in 15 (62%), and fair in six (25%). The best results were obtained in patients with Mason Type 2 injuries treated by closed reduction without fracture excision and with early complete radial head excision for a Type 3 fracture. Late instability was not observed in any of the 24 patients. Prolonged immobilization (greater than four weeks) was associated with poor results. Ectopic ossification occurred in only one patient who had surgical treatment at eight days after injury; the grading score was only fair. These observations demonstrate that the injury should be treated with early reduction of the ulnohumeral joint and treatment of the radial head fracture according to its type. Immobilization for more than four weeks should be avoided. The prognosis is better than what has been thought previously.

Abstract:Despite continuous improvements in diagnostic cardiology techniques, echocardiography remains the cornerstone for noninvasive cardiovascular assessment of the pregnant woman with heart disease or suspected cardiac abnormality. Reversible physiological cardiac remodeling of pregnancy associated with changes in valve patency or transvalvular flow pattern can be suitably assessed by the complementary use of quantitative pulsed‐ and continuous Doppler and qualitative color Doppler technology. These techniques are also useful for a better understanding of the pathophysiology of the hemodynamic consequences of fixed valve stenosis during pregnancy with respect to the labile nature of gradients resulting from variable loading conditions as occurs during pregnancy. Recent and specific areas of clinical use including cardiac output estimation, contrast echocardiography minimizing radioscopy during cardiac catheterization, and transesophageal echocardiography for selection of patients with mitral stenosis suitable for percutaneous catheter valvotomy illustrate the large spectrum of capabilities of this versatile method during pregnancy.

Abstract:ANESTHESIOLOGY is a discipline that is uniquely placed to contribute to the preeminent scientific quest of this century–namely the understanding of the neurologic mechanisms that underpin the phenomena of consciousness. The operating room is the crucible in which any putative theory should be tested. While acknowledging that there are arguments as to whether rapid eye movement sleep is really a conscious state, a simple practical approach to the definition of consciousness would include the idea of perception of the outside world. Perception involves some sort of convolution of external sensory input with the internal state of the brain. In this issue of the ANESTHESIOLOGY, an article by Hudetz et al. describes an ostensibly simple experiment in which they stimulated rats with flashes of light and measured how desflurane modified the resultant patterns of electrical response in the visual cortex. The authors used extracellular electrodes to measure the rate of firing of neuronal action potentials (for reasons of methodological purity, the authors use the term units rather than neurons). Consonant with previous studies, they showed that increasing concentrations of desflurane reduced the unstimulated background neuronal activity. However there were more specific effects on the flash-evoked activity. Desflurane had little effect on the early neuronal responses (within 100 ms of the light stimulus) in the primary visual cortex. In contrast, the late activity (100 ms to 2,000 ms) was progressively and profoundly depressed with increasing concentrations of the anesthetic vapor. These results imply that volatile anesthetics do not inhibit the raw sensory stimulus from getting directly to the cortex, but rather profoundly inhibit the ability of the cortex to respond to and perceive this stimulus. In the presence of more than 0.7 minimum alveolar concentration of desflurane, the cortical effects of the visual stimuli simply die out and are gone after 100 ms. During general anesthesia, the rat cortex receives the input, but it is not able to integrate the information into any sort of primitive rodent cognition—i.e., to say to itself: “A flash of light! Perhaps I’d better run for cover . . .” This study contributes to a new wave of ideas and experiments that are looking at the nuts and bolts of the neuronal processes required for perception. Many of these ideas have been summarized in a detailed yet readable article by Alkire et al. published in Science. From this literature, I will briefly mention two experiments that provide support for Hudetz’s results. In awake epileptic patients, Quian Quiroga et al. recorded action potentials from the neurons in the hippocampus and medial temporal lobe in response to the patients seeing various pictures of things and wellknown people. They found patterns of transient activation in neuronal firing that looked very similar in time course (starting at about 200 ms and lasting up to 2,000 ms) to the delayed baseline patterns reported in Hudetz’s article. In Quian Quiroga’s study, the activations were achieved by more subtle cognitive events—the most famous being the neuronal “recognition” of pictures of the movie and television personality Jennifer Aniston. It appears that this subset of neurons was encoding an abstract representation of Jennifer Aniston because the neurons could be activated either by a number of entirely different pictures of Jennifer Aniston or even by a slide with no picture at all but only the words “Jennifer Aniston.” Also, these neurons were not activated by pictures of other subjects, not even other young blond women. By various statistical arguments, it can be assumed that perception required activation of around 0.2% of the total number of neurons in the medial temporal lobe. This implies the existence of a subnetwork of neurons (analogous to the so-called Hebbian neuronal assembly) that–given the right context–dance together in response to the Jennifer Aniston stimulus. Unfortunately, these investigators did not report on how this activation of the Jennifer Aniston neurons might have been obtunded during induction of general anesthesia for subsequent epilepsy surgery. However, I suspect that we would have seen ablation of firing patterns very similar to that reported in Hudetz’s current study. The second study pertains to the question of whether the long-distance spread of information across the cortex is inhibited by sleep and general anesthesia. Using a completely undifferentiated direct stimulus (transcranial magnetic stimulation) Massimini et al. stimulated a section of the premotor parietal cortex and measured the resultant spread of cortical electrical activity. In agreement with numerous functional magnetic resonance imaging studies, they showed that the spread of evoked activity in the awake state was widespread in space (moving in a stereotypical sequence over most of the cortex) and prolonged in time. In contrast, the initial activation was more intense during slow wave sleep, but it then failed to spread further in space and died out quickly in time (approximately 100 ms). Similar experiments in patients under general anesthesia are underway, and a patent has been taken out–presumably with the view to developing another monitor of anesthesia. Abstract:According to many reports, phenolic compounds isolated from olive leaves have very good biological activities, especially antimicrobial. Presently, the resistance of microorganisms to antibiotics is greater than ever. Therefore, there are numerous recent papers about alternative solutions for inhibiting their influence on human health. Olive leaf is studied as an important source of antimicrobials with low cost and used in medicine. Numerous publications on involving green technologies for isolation of active compounds from olive leaves have appeared over the past few decades. The present review reports on current knowledge of the most isolated phenolic compounds from olive leaf extract as well as methods for their isolation and characterization. This paper uses recent research findings with a wide range of study models to describe the antimicrobial potential of phenolic compounds. It also describes the vast range of information about methods for determination of antimicrobial potential focusing on effects on different microbes. Additionally, it serves to highlight the role of olive leaf extract as an antioxidants and presents methods for determination of antioxidant potential. Furthermore, it provides an overview of presence of enzymes. The significance of olive leaves as industrial and agricultural waste is emphasized by means of explaining their availability, therapeutic and nutritional effects, and research conducted on this field.

Optimization of Bioactive Phenolics Extraction and Cosmeceutical Activity of Eco-Friendly Polypropylene-Glycol–Lactic-Acid-Based Extracts of Olive Leaf

Abstract:Olive leaf is a rich source of phenolic compounds with numerous activities related to skin health and appearance. In this study, a green extraction method was developed using eco-friendly solvents: polypropylene glycol (PPG), lactic acid (LA), and water. The optimal extraction conditions were established, including solvent, extraction time, technique (magnetic stirrer vs. ultrasound-assisted extraction), and herbal material/solvent ratio. The composition of the solvent mixture was optimized using a mixture design. The content of phenolic compounds, including oleuropein and verbascoside, was determined using high-performance liquid chromatography (HPLC) and spectrophotometric methods. Using different extraction conditions, three extracts were prepared and their phytochemical compositions and antioxidant and skin-related bioactivities were investigated. The extracts were excellent inhibitors of elastase, collagenase, tyrosinase, and lipoxygenase. The best activity was shown by the extract richest in phenolics and prepared using magnetic-stirrer-assisted extraction for 20 min, with 0.8 g of herbal material extracted in 10 mL of PPG/LA/water mixture (28.6/63.6/7.8, w/w/w), closely followed by the extract prepared using the same extraction conditions but with 0.42 g of herbal material. The investigated PPG/LA/water mixtures contributed to the overall enzyme-inhibitory activity of the extracts. The prepared extracts were appropriate for direct use in cosmetic products, thus saving the time and energy consumption necessary for the evaporation of conventional solvents.

Abstract:Olive leaves benefits are less well known, despite having potential health benefits that could make them a valuable addition to a daily diet, compared to olive oil, which is well known for the many benefits it provides. The antioxidant capacity of olive leaves extract (OLE) is higher than that of the most known powerful antioxidants, such as vitamins C and E or pure hydroxytyrosol. The aim of this study was first to maximize the bioactive compounds in extra virgin olive oil (EVOO) by enriching it with olive leaves; the EVOO acts as an extraction solvent, and a study of the macerating period was carried out to select the best macerating period to improve the stabilization of the bioactive compounds in the olive oil matrix. Once the best macerating period was selected (7 days), OLE was gamma-irradiated with a dose of 25 kGy to assess its oxidative stability. To this end, UV–visible spectroscopy was used to perform stability and quality analysis of the OLE during a macerating period of 21 days and after irradiation. The results of this study show an extremely significant (p < 0.0001) improvement in the quality parameters (K232, K270, and ΔK) and in total carotenoids and chlorophylls, assessed by K470 and K670, among all the macerating periods of OLE compared to the control group. Regarding the irradiated OLE compared to the irradiated EVOO, there was an extremely significant (p < 0.0001) difference in ΔK. In addition, there was a significant decrease in total carotenoids and chlorophylls in the irradiated EVOO compared to the irradiated OLE. In conclusion, olive leaves extract in olive oil could be used as a free radical scavenger in food irradiation and tissue sterilization to alleviate the detrimental effects of ionizing radiation.

Abstract:Olea europaea L. fruit is a peculiar vegetal matrix containing high levels of fatty acids (98-99% of the total weight of extra-virgin olive oil, EVOO) and low quantities (1-2%) of phenolics, phytosterols, tocopherols, and squalene. Among these minor components, phenolics are relevant molecules for human health. This review is focused on their beneficial activity, in particular of hydroxytyrosol (HT), oleuropein (OLE), oleocanthal (OLC), and lignans found in EVOO, olive oil by-products and leaves. Specifically, the cardioprotective properties of the Mediterranean diet (MD) related to olive oil consumption, and the biological activities of polyphenols recovered from olive oil by-products and leaves were described. Recent European projects such as EPIC (European Prospective Investigation into Cancer and Nutrition) and EPICOR (long-term follow-up of antithrombotic management patterns in acute coronary syndrome patients) have demonstrated the functional and preventive activities of EVOO showing the relation both between cancer and nutrition and between consumption of EVOO, vegetables, and fruit and the incidence of coronary heart disease. The data reported in this review demonstrate that EVOO, one of the pillars of the MD, is the main product of Olea europaea L. fruits; leaves and by-products are secondary but precious products from which bioactive compounds can be recovered by green technologies and reused for food, agronomic, nutraceutical, and biomedical applications according to the circular economy strategy.
Olive Leaf Extracts from Three Italian Olive Cultivars Exposed to Drought Stress Differentially Protect Cells against Oxidative Stress

Abstract:Olive leaves are an abundant by-product of olive oil production. Olive leaf extracts (OLEs) are rich in polyphenols, which can be used for health benefits. As polyphenols are the main antioxidant molecules in plants, plants typically increase their polyphenol content when exposed to drought stress. However, the phenolic profile of OLEs can vary in relation to the origin and variety of the plant material. In this work, olive leaf extracts from three different Italian olive cultivars (Giarraffa, Leccino, and Maurino) both exposed and not exposed to drought stress were studied in terms of antioxidant properties and profile, intestinal permeation, and protection against oxidative stress of human umbilical vein endothelial cells (HUVECs), since HUVECs are considered a model to study a wide range of diseases. OLEs from stressed Maurino and Giarraffa plants showed the highest increase in antioxidant capacity compared to controls. The phenolic profile of Maurino' was mainly increased by water deficit, with a large increase in the compounds oleuropein and luteolin-7-O-rutinoside. All tested extracts exposed to a water deficit protected HUVECs against oxidative stress by reducing ROS production, and this effect was more pronounced in OLEs from Giarraffa and Maurino exposed to drought stress compared to all other extracts. Finally, OLE from the stressed Giarraffa group showed a higher apparent permeability of antioxidant molecules than that of Maurino.

Abstract:The aim of this study was to characterise phenolic compounds of olive flower obtained from Olive tree cultivar Chemlali and to investigate their anticancer effect on MCF-7 cells. Phenolic characterisation was determined using LC/MS-MS. Cytotoxicity of the extract was determined using MTT. Biochemical markers of apoptosis were evaluated by immunoblotting. Our results showed that olive flower contained significant amounts of phenolic compounds mainly flavonoids, secoiridoids and simple phenols. Furthermore, the phenolic extract exerted a significant reduction in MCF-7 cell viability (EC50 values equal to 220.8 μg/ml). Western blot analysis revealed the presence of the cleaved forms of Parp-1. The DAPI staining analysis demonstrated a significant reduction in the number of cells and a considerable change in the morphology of the treated cells. In conclusion, Olea europaea. L flower contained great amounts of different bio-phenols able to reduce the proliferative activity of breast cancer MCF-7 cells by the induction of apoptosis.

Olive Pomace Phenolic Compounds Stability and Safety Evaluation: From Raw Material to Future Ophthalmic Applications

Abstract:Nowadays, increasing interest in olive pomace (OP) valorization aims to improve olive’s industry sustainability. Interestingly, several studies propose a high-value application for OP extracts containing its main phenolic compounds, hydroxytyrosol and oleuropein, as therapy for ocular surface diseases. In this work, the stability and accessibility of OP total phenolic and flavonoid content, main representative compounds, and antioxidant activity were assessed under different pretreatment conditions. Among them, lyophilization and supercritical CO2 extraction were found to increase significantly most responses measured in the produced extracts. Two selected extracts (CONV and OPT3) were obtained by different techniques (conventional and pressurized liquid extraction); Their aqueous solutions were characterized by HPLC-DAD-MS/MS. Additionally, their safety and stability were evaluated according to EMA requirements towards their approval as ophthalmic products: their genotoxic effect on ocular surface cells and their 6-months storage stability at 4 different temperature/moisture conditions (CPMP/ICH/2736/99), together with pure hydroxytyrosol and oleuropein solutions. The concentration of hydroxytyrosol and oleuropein in pure or extract solutions was tracked, and possible degradation products were putatively identified by HPLC-DAD-MS/MS. Hydroxytyrosol and oleuropein had different stability as standard or extract solutions, with oleuropein also showing different degradation profile. All compounds/extracts were safe for ophthalmic use at the concentrations tested.

Abstract:Polysaccharide-rich materials were extracted from the alcohol-insoluble solids of Olea europaea l. **leaves. Structural characteristics were determined by colorimetric techniques, FT-IR, GC–MS, SEC/MALS/VD/DRI, and NMR ( 1 H, 13 C). The extract and its main macromolecular components were characterized to assess their ability toward antioxidant, α-amylase inhibition, and antiproliferative activities. Results revealed that the ultrasound olive leave extract comprises polysaccharides with uronic acid, galactose, arabinose, and glucose in molar percentages of 11.7%, 11.3%, 7.5%, and 4.9% respectively, constituting 41% of the total mass. In addition, polyphenols (21%) and proteins (9%) are associated with these polysaccharides. Further, the extract showed noticeable ORAC and free radical scavenging abilities, in addition to high in vitro antiproliferative activity against Caco-2 colon carcinoma cell lines. Similarly, the extract exhibited a strong, uncompetitive inhibition of α-amylase by 75% in the presence of the extract with 0.75 μg/mL of concentration. This research concludes that ultrasound extraction method can be used for the extraction of polysaccharide–polyphenol–protein complexes. These conjugates exhibit the potential for combined biological activities resulting from a synergistic effect of its compounds, making them promising ingredients for the development of functional food.

Abstract:Abstract In this study, Box–Behnken experimental design was employed to optimize Ultrasound‐assisted extraction (UAE) method to obtain oleuropein‐rich extract from olive leaf. The effect of three parameters, ethanol concentration (30%–70%), amplitude (30%–50%), and ultrasonication time (5–15 min) on oleuropein content were evaluated through experimental design. The total phenolic content, antioxidant capacity, DNA damage scavenging activity, aroma profile, and antimicrobial activity of olive leaf were also investigated. The optimum extraction condition was determined as 30% amplitude, 70% ethanol, and 5 min extraction time. The amount of oleuropein was found to be 42.621 mg/g dry leaves at the optimum condition. The total phenolic content of the optimized extract was found as 45.13 mg GAE/g dry leaves while the antioxidant capacity of the optimized extract was 216 μmol Trolox/g in DPPH and 213.19 μmol Trolox/g in ABTS method. Twenty‐three volatile compounds were identified in extracts, mainly aldehydes and alcohols. The extracts showed high antimicrobial activity against selected bacterial strains. DNA damage protecting activity of extracts on plasmid DNA was clearly demonstrated. UAE has an important effect on the phenolic content and antioxidant capacity of olive leaves. Moreover, oleuropein‐rich extract can have potential as a therapeutic agent depending on its antimicrobial and DNA‐protecting activity.
Gastrointestinal digestion of olive leaf waste from olive mill production chain in a simulator of the gastrointestinal tract

Abstract:Background: Currently, olive leaves are considered waste, although their high phenolic content makes them a source of antioxidants that could be used directly. The aim of this work was to study the behavior of phenolic compounds contained in olive leaf from the olive mill production chain during their gastrointestinal digestion. Results: Phenolic compounds in the gastric digestion analyzed by high-performance liquid chromatography-diode array detection-mass spectrometry increased by 58% to 314.5% compared to the meal, while in the intestinal stage they ranged from 1.87 to 9.04 times higher. An increase of between 187% and 903% in bioaccessibility of phenolic compounds in the intestinal phase was observed, except for verbascoside. Furthermore, such compounds were fully bioavailable, except for apigenin-7-O-glucoside, which showed a bioavailability of 56%. Conclusion: The current study showed that the concentration of oleuropein, apigegin-7-O-glucoside, hydroxytyrosol hexoside and hydroxytyrosol contained in olive leaf continued to be extracted during the gastrointestinal digestion process. Furthermore, the results obtained with respect to their bioaccessibility and bioavailability suggest a good disposition to pass into the bloodstream where they could exert beneficial effects. Therefore, these results are promising for olive leaf becoming a consumable by-product that could be directly ingested through a simple infusion. © 2024 The Authors. Abstract:High concentrations of advanced glycation end products (AGEs) have been linked to diseases, including diabetic complications. The pathophysiological effects of AGEs are mainly due to oxidative stress and inflammatory processes. Among the proteins most affected by glycation are albumin, the most abundant circulating protein, and collagen, which has a long biological half-life and is abundant in the extracellular matrix. The potential cellular damage caused by AGEs underscores the importance of identifying and developing natural AGE inhibitors. Indeed, despite initial promise, many synthetic inhibitors have been withdrawn from clinical trials due to issues such as cytotoxicity and poor pharmacokinetics. In contrast, natural products have shown significant potential in inhibiting AGE formation. Olea europaea L. leaves, rich in bioactive compounds like oleuropein and triterpenoids, have attracted scientific interest, emphasizing the potential of olive leaf extracts in health applications. This study investigates the anti-glycation properties of two polyphenol-rich extracts (OPA40 and OPA70) and a triterpene-enriched extract (TTP70) from olive leaves. Using in vitro protein glycation methods with bovine serum albumin (BSA)–glucose and gelatin–glucose systems, this study assesses AGE formation inhibition by these extracts through native polyacrylamide gel electrophoresis (N-PAGE) and autofluorescence detection. OPA40 and OPA70 exhibited strong, dose-dependent anti-glycation effects. These effects were corroborated by electrophoresis and further supported by similar results in a gelatin–glucose system. Additionally, TTP70 showed moderate anti-glycation activity, with a synergistic effect of its components. The results support the real possibility of using olive leaf bioproducts in ameliorating diabetic complications, contributing to sustainable bio-economy practices.

Abstract:Background: Olive tree (Olea europaea, Oleaceae) leaves have been widely used in traditional herbal medicine to prevent and treat various diseases especially in Mediterranean countries. They contain several potentially bioactive compounds that may have hypoglycemic and hypolipidemic properties. Summary: The literature has recently been attempting to define the relationship between olive leaf (Olea europaea L. folium) polyphenols and a number of health problems. Oleuropein, the basic phenolic compound of olive leaf and its extract, is responsible for the characteristic bitter taste and unique aroma of olive fruits. Furthermore, it is shown that oleuropein and its hydrolyzed products have many beneficial effects on human health because of its antioxidant characters. A number of studies report that olive leaf has potentially positive effects on the parameters related to diabetes and cardiovascular diseases by various mechanisms. Besides, toxicity studies suggest that olive leaf is generally safe even at high doses. Key messages: Although current results obtained until today seem promising, the studies in this subject are usually on cell culture and animal trials. Moreover, mostly the extract forms of olive leaves are used in the studies. More randomized controlled human clinical trials with extensive toxicity studies are needed to evaluate potential health effects and safety.

Abstract:Olive trees not only produce olives but also generate a substantial amount of waste and by-products, including leaves, pomace (the solid remains after pressing olives for oil), and wastewater from the olive oil-making process. The waste products, particularly the leaves, contain bioactive compounds, especially phenolic compounds, known for their health benefits, such as high antioxidant potential and the ability to reduce inflammation. These compounds have shown promise in preventing and treating cancer. This review, based on in vitro evidence, provides a detailed description and discussion of the mechanisms through which these compounds from olive leaves can prevent development, the ways they might act against cancer cells, and their potential to increase the sensitivity of tumor cells to conventional anticancer therapy. The possible synergistic effects of these compounds suggest that olive leaf extracts may offer a promising approach for cancer treatment, compared with isolated compounds, thus providing novel possibilities for cancer therapy.

Abstract:Mechanical forces generated by contractile cells allow the cells to sense their environment and to interact with other cells. By locally pulling on their environment, cells can sense and respond to mechanical features such as the local stress (or strain), the shape of a cellular domain, and the surrounding rigidity; at the same time, they also modify the mechanical state of the system. This creates a mechanical feedback loop that can result in self-polarization of cells. In this paper, we present a quantitative mechanical model that predicts the self-polarization of cells in spheroidally shaped domains, comprising contractile cells and an elastic matrix, that are embedded in a three-dimensional, cell-free gel. The theory is based on a generalization of the known results for passive inclusions in solids to include the effects of cell activity. We use the active cellular susceptibility tensor presented by Zemel [Phys. Rev. Lett. 97, 128103 (2006)] to calculate the polarization response and hence the elastic stress field developed by the cells in the cellular domain. The cell polarization is analyzed as a function of the shape and the elastic moduli of the cellular domain compared with the cell-free surrounding material. Consistent with experiment, our theory predicts the development of a stronger contractile force for cells in a gel that is surrounded by a large, cell-free material whose elastic modulus is stiffer than that of the gel that contains the cells. This provides a quantitative explanation of the differences in the development of cellular forces as observed in free and fixed gels. In the case of an asymmetrically shaped (spheroidal) domain of cells, we show that the anisotropic elastic field within the domain leads to a spontaneous self-polarization of the cells along the long axis of the domain.
