Abstract:Rapamycin (RAP) is currently being developed as potential antibreast cancer drug. However, its poor solubility completely limits its use. The aim of this study was to develop polyethylene glycol-poly(lactide-co-glycolide) (PEG-PLGA)-based nanoparticles (NPs) to load RAP via microfluidics with an appropriate polyethylene glycol (PEG) content to enhance the bioavailability of RAP. Polydimethylsiloxane (PDMS) chips with a Y-shaped channel were designed to obtain RAP-loaded PEG-PLGA NPs (RAP-PEG-PLGA). The entrapment efficiency (EE) and drug loading (DL) as well as release profile of RAP-PEG-PLGA were evaluated, and their resistance to plasma albumin adsorption of NPs with different PEG contents was evaluated and compared. RAW264.7 and 4T1 cells were used to assess the antiphagocytic and anticancer cells effect of NPs, respectively. RAP-PEG-PLGA of around 124 nm in size were successfully prepared with the EE of 82.0% and DL of 12.3%, and sustained release for around 40 d. A PEG relative content of 10% within the PEG-PLGA molecule was shown superior in resisting protein adsorption. RAP-PEG-PLGA inhibited the growth of breast cancer cells when the concentration was over 10 μg/mL, and the inhibition efficiency was significantly higher than free RAP. Hence, the current RAP-PEG-PLGA could be a potential therapeutic system for breast cancer treatment.

Abstract:Poly(ethylene glycol)‐block‐poly(lactide) (PEG‐b‐PLA) micro‐ and nanoparticles (NPs) have been intensively investigated for applications in biomedicine, due to their inherent biocompatibility and biodegradability, which allows them to be used as sustained release systems. Current methods for preparing PEG‐b‐PLA NPs typically require two different steps that include polymer synthesis and NP assembly, with the necessary intermediate polymer purification and the use of a variety of organic solvents in the process. In order to facilitate the biomedical application of PEG‐b‐PLA NPs, it is of great interest to develop a strategy to formulate the NPs in a simplified manner. Here, we report a straightforward method to construct PEG‐b‐PLA NPs through a sequential two‐step process without intermediate work‐up, which involves synthesizing the polymer in a water‐miscible organic solvent that is, N,N‐dimethylformamide (DMF), followed by addition of water to the polymer solution. In this way, large NPs (~600 nm) were prepared. We comprehensively characterized the NPs using turbidity studies, dynamic light scattering (DLS), scanning electron microscopy (SEM), and transmission electron microscopy (TEM) techniques. We further demonstrated the ability of the NPs to encapsulate drugs, exemplified in the immunotherapeutic agent rapamycin, with relatively high encapsulation efficiency. In vitro drug release tests showed that rapamycin‐encapsulating NPs had comparable sustained‐release profiles at different pH conditions, highlighting the broad application window of our NP platform. Moreover, in vitro T cell suppression assays revealed that rapamycin‐loaded NPs exhibited similar inhibitory performance to free rapamycin on CD8+ cells at all rapamycin concentrations and on CD4+ cells at high and intermediate rapamycin concentrations, while the performance of the NPs was superior on CD4+ at low rapamycin concentration. Overall, this work provides a route for the scalable synthesis of biocompatible PEG‐b‐PLA NPs, which can be extended to other polymeric NPs, with potential in biomedical applications such as immunotherapy.

Microfluidic preparation of PLGA microspheres as cell carriers with sustainable Rapa release

Abstract:The current study, inspired by the immunosuppressive property of rapamycin (Rapa) and the benefit of microspheres both as drug delivery system and cell carriers, was designed to develop an efficient Rapa delivery system with tunable controllability to facilitate its local administration. A capillary-based two-phase microfluidic device was designed to prepare monodisperse poly(lactide-co-glycolide) (PLGA) microspheres to load Rapa (PLGA-Rapa-M). The physical and chemical properties of PLGA-Rapa-M were characterized, and the Rapa loading capacity and release profile were explored. Chondrocytes were chosen as a cell model to evaluate the adhesion and proliferation on these microspheres. Controllability over the microsphere properties was illustrated. The PLGA-Rapa-M is averagely 63.91 μm in size with a narrow size distribution and a CV of 2.44%. The encapsulation efficiency of Rapa within microspheres via the current microfluidics was around 98%, and Rapa loading could be easily varied with a maximum value of ∼20%. The PLGA-Rapa-M has a sustained Rapa release duration of ∼3 months. These microspheres could not only successfully be used for Rapa sustained release but also as cell carriers for cell therapy since they can support the attachment/proliferation of chondrocytes. Hence, improved therapeutic index could be expected by using the current developed Rapa-release system.

Abstract:Rapamycin is a triene macrolide antibiotic produced by Streptomyces hygroscopicus. Besides its wide application as an effective immunosuppressive agent, other important bioactivities have made rapamycin a potential drug lead for novel pharmaceutical development. However, the low titer of rapamycin in the original producer strain limits further industrialization efforts and restricts its use for other applications. Predicated on knowledge of the metabolic pathways related to rapamycin biosynthesis in S. hygroscopicus, we have rationally designed approaches to generate a rapamycin high producer strain of S. hygroscopicus HD-04-S. These have included alleviation of glucose repression, improved tolerance towards lysine and shikimic acid, and auxotrophy of tryptophan and phenylalanine through the application of stepwise UV mutagenesis. The resultant strain produced rapamycin at 450 mg/L in the shake flask scale. These fermentations were further scaled up in 120 and 20,000 L fermentors, respectively, at the pilot plant. Selected fermentation factors including agitation speed, pH, and on-line supplementation were systematically evaluated. A fed-batch strategy was established to maximize rapamycin production. With these efforts, an optimized fermentation process in the larger scale fermentor was developed. The final titer of rapamycin was 812 mg/L in the 120 L fermentor and 783 mg/L in the 20,000 L fermentor. This work highlights a high rapamycin producing strain derived by mutagenesis and subsequent screening, fermentation optimization of which has now made it feasible to produce rapamycin on an industrial scale by fermentation. Rapidly dissolving microneedles for the delivery of cubosome-like liquid crystalline nanoparticles with sustained release of rapamycin

Abstract:In this study, we developed a system for the transdermal delivery and controlled release of the hydrophobic immunosuppressive drug rapamycin, foreseeing an application in psoriasis treatment. To do so, rapamycin was encapsulated in phytantriol-based cubosome-like liquid crystalline nanoparticles stabilized with pluronic F127. The final mass percent composition of the lipid nanoparticles was 0.25% phytantriol, 0.1% pluronic F127, 4.75% ethanol and 94.9% water. These particles showed a rapamycin encapsulation efficiency above 95% and a sustained in vitrodrug release profile throughout 14 days. Subsequently the rapamycin-carrying particles were incorporated into rapidly dissolving microneedle patches composed of a polymeric matrix of poly(vinylpyrrolidone) and poly(vinyl alcohol). Confocal microscopy allowed to infer the preferential distribution of the cubosome-like particles at the tip and baseplate of the microneedles. The fabricated microneedles showed successful piercing and deposition of the loaded cubosome-like particles on a skin-mimicking agarose gel. Finally, the rapamycin-loaded cubosome-like particles showed antiproliferative activity in natural killer cells in vitro. The results here presented show the potential of the developed system to deliver cubosome-like particles into the skin and promote the sustained release of rapamycin in the context of immunomodulation.
Comparison of emulsion and spray methods for fabrication of rapamycin-loaded acetalated dextran microparticles

Abstract:Rapamycin (rapa), an immunosuppressive medication, has demonstrated considerable effectiveness in reducing organ transplant rejection and treating select autoimmune diseases. However, the standard oral administration of rapa results in poor bioavailability, broad biodistribution, and harmful off-target effects, necessitating improved drug delivery formulations. Polymeric microparticles (MPs) are one such solution and have demonstrated promise in pre-clinical studies to improve the therapeutic efficacy of rapa. Nevertheless, MP formulations are highly diverse, and fabrication method selection is a critical consideration in formulation design. Herein, we compared common fabrication processes for the development of rapa-loaded MPs. Using the biopolymer acetalated dextran (Ace-DEX), rapa-loaded MPs were fabricated by both emulsion (homogenization and sonication) and spray (electrospray and spray drying) methods, and resultant MPs were characterized for size, morphology, surface charge, and drug release kinetics. MPs were then screened in LPS-stimulated macrophages to gauge immunosuppressive efficacy relative to soluble drug. We determined that homogenized MPs possessed the most optimal combination of sizing, tunable drug release kinetics, and immunosuppressive efficacy, and we subsequently demonstrated that these characteristics were maintained across a range of potential rapa loadings. Further, we performed in vivo trafficking studies to evaluate depot kinetics and cellular uptake at the injection site after subcutaneous injection of homogenized MPs. We observed preferential MP uptake by dendritic cells at the depot, highlighting the potential for MPs to direct more targeted drug delivery. Our results emphasize the significance of fabrication method in modulating the efficacy of MP systems and inform improved formulation design for the delivery of rapa.
Subcutaneous nanotherapy repurposes the immunosuppressive mechanism of rapamycin to enhance allogeneic islet graft viability

Abstract:Standard oral rapamycin (that is, Rapamune) administration is plagued by poor bioavailability and broad biodistribution. Thus, this pleotropic mammalian target of rapamycin (mTOR) inhibitor has a narrow therapeutic window and numerous side effects and provides inadequate protection to transplanted cells and tissues. Furthermore, the hydrophobicity of rapamycin limits its use in parenteral formulations. Here, we demonstrate that subcutaneous delivery via poly(ethylene glycol)-b-poly(propylene sulfide) polymersome nanocarriers significantly alters rapamycin’s cellular biodistribution to repurpose its mechanism of action for tolerance, instead of immunosuppression, and minimize side effects. While oral rapamycin inhibits T cell proliferation directly, subcutaneously administered rapamycin-loaded polymersomes modulate antigen presenting cells in lieu of T cells, significantly improving maintenance of normoglycemia in a clinically relevant, major histocompatibility complex-mismatched, allogeneic, intraportal (liver) islet transplantation model. These results demonstrate the ability of a rationally designed nanocarrier to re-engineer the immunosuppressive mechanism of a drug by controlling cellular biodistribution.

[The lysozyme liberation test--a method for the demonstration of stimulation of human neutrophil granulocytes and monocytes].

Abstract:A method for proof of induced secretion of lysozyme as a marker of lysosomal enzymes is described. Secretory cells are neutrophil granulocytes and monocytes. The stimulation of the cells is affected by soluble inductors. We used phytohemagglutinin (PHA), Concanavalin A (Con A), purified protein derivates (PPD) and lipopolysaccharide (LPS). Myelin basic protein has a direct stabilizing action on the membrane of lysosomes and induces an increased secretory reaction of cells after irritation. That is why it is recommended to add this substance as a supplement to the culture medium of the test. The optimal incubation parameters for the lysozyme release assay are ascertained. The method is recommended to apply in the fundamental investigation, for testing of stimulating or antiphlogistic agents and in the clinical functional diagnosis.
Immunomodulatory potential of rapamycin-loaded mesoporous silica nanoparticles: pore size-dependent drug loading, release, and in vitro cellular responses

Abstract:Abstract Rapamycin is a potent immunosuppressive drug that has been recently proposed for a wide range of applications beyond its current clinical use. For some of these proposed applications, encapsulation in nanoparticles is key to ensure therapeutic efficacy and safety. In this work, we evaluate the effect of pore size on mesoporous silica nanoparticles (MSN) as rapamycin nanocarriers. The successful preparation of MSN with 4 different pore sizes was confirmed by dynamic light scattering, zeta potential, transmission electron microscopy and N 2 adsorption. In these materials, rapamycin loading was pore size-dependent, with smaller pore MSN exhibiting greater loading capacity. Release studies showed sustained drug release from all MSN types, with larger pore MSN presenting faster release kinetics. In vitro experiments using the murine dendritic cell (DC) line model DC2.4 showed that pore size influenced the biological performance of MSN. MSN with smaller pore sizes presented larger nanoparticle uptake by DC2.4 cells, but were also associated with slightly larger cytotoxicity. Further evaluation of DC2.4 cells incubated with rapamycin-loaded MSN also demonstrated a significant effect of MSN pore size on their immunological response. Notably, the combination of rapamycin-loaded MSN with an inflammatory stimulus (lipopolysaccharide, LPS) led to changes in the expression of DC activation markers (CD40 and CD83) and in the production of the proinflammatory cytokine TNF-α compared to LPS-treated DC without nanoparticles. Smaller-pored MSN induced more substantial reductions in CD40 expression while eliciting increased CD83 expression, indicating potential immunomodulatory effects. These findings highlight the critical role of MSN pore size in modulating rapamycin loading, release kinetics, cellular uptake, and subsequent immunomodulatory responses. Graphical abstract

Abstract:The outcome of scaffold-based stem cell transplantation remains unsatisfied due to the poor survival of transplanted cells. One of the major hurdles associated with the stem cell survival is the immune rejection, which can be effectively reduced by the use of immunosuppressant. However, ideal localized and sustained release of immunosuppressant is difficult to be realized, because it is arduous to hold the drug delivery system within scaffold for a long period of time. In the present study, the sustained release of immunosuppressant for the purpose of improving the survival of stem cells was successfully realized by a nanoparticle-anchoring hydrogel scaffold we developed. Methods: Poly (lactic-co-glycolic acid) (PLGA) nanoparticles were modified with RADA16 (RNPs), a self-assembling peptide, and then anchored to a RADA16 hydrogel (RNPs + Gel). The immobilization of RNPs in hydrogel was measured in vitro and in vivo, including the Brownian motion and cumulative leakage of RNPs and the in vivo retention of injected RNPs with hydrogel. Tacrolimus, as a typical immunosuppressant, was encapsulated in RNPs (T-RNPs) that were anchored to the hydrogel and its release behavior were studied. Endothelial progenitor cells (EPCs), as model stem cells, were cultured in the T-RNPs-anchoring hydrogel to test the immune-suppressing effect. The cytotoxicity of the scaffold against EPCs was also measured compared with free tacrolimus-loaded hydrogel. The therapeutic efficacy of the scaffold laden with EPCs on the hind limb ischemia was further evaluated in mice. Results: The Brownian motion and cumulative leakage of RNPs were significantly decreased compared with the un-modified nanoparticles (NPs). The in vivo retention of injected RNPs with hydrogel was obviously longer than that of NPs with hydrogel. The release of tacrolimus from T-RNPs + Gel could be sustained for 28 days. Compared with free tacrolimus-loaded hydrogel, the immune responses were significantly reduced and the survival of EPCs was greatly improved both in vitro and in vivo. The results of histological evaluation, including accumulation of immune cells and deposition of anti-graft antibodies, further revealed significantly lessened immune rejection in T-RNPs-anchoring hydrogel group compared with other groups. In pharmacodynamics study, the scaffold laden with EPCs was applied to treat hind limb ischemia in mice and significantly promoted the blood perfusion (~91 % versus ~36 % in control group). Conclusion: The nanoparticle-anchoring hydrogel scaffold is promising for localized immunosuppressant release, thereby can enhance the survival of transplanted cells and finally lead to successful tissue regeneration.
Development of a novel rapamycin loaded nano- into micro-formulation for treatment of lung inflammation

Abstract:Abstract It has recently emerged that drugs such as the mTOR inhibitor rapamycin (Rapa) may play a key role in the treatment of airway inflammation associated with lung diseases, such as chronic obstructive pulmonary disease, asthma, and cystic fibrosis. Nevertheless, Rapa clinical application is still prevented by its unfavorable chemical-physical properties, limited oral bioavailability, and adverse effects related to non-specific biodistribution. In this paper, the design and production of a novel formulation of Rapa based on nano into micro (NiM) particles are detailed. To achieve it, Rapa-loaded nanoparticles were produced by nanoprecipitation of an amphiphilic pegylated poly-ɛ-caprolactone/polyhydroxyethyl aspartamide graft copolymer. The obtained nanoparticles that showed a drug loading of 14.4 wt% (corresponding to an encapsulation efficiency of 82 wt%) did not interact with mucins and were able to release and protect Rapa from degradation in simulated lung and cell fluids. To allow their local administration to the lungs as a dry powder, particle engineering at micro-sized level was done by embedding nanoparticles into mannitol-based microparticles by spray drying. Obtained NiM particles had a mean diameter of about 2-μ, spherical shape and had good potential to be delivered to the lungs by a breath-activated dry powder inhalers. Rheological and turbidity experiments showed that these NiM particles can dissolve in lung simulated fluid and deliver the Rapa-loaded pegylated nanoparticles, which can diffuse through the mucus layer. T Power Subsystem Design and Early Mission Performance

Abstract:he power subsystem for the Near Earth Asteroid Rendezvous (NEAR) spacecraft is a direct energy transfer system with an Eagle Picher Industries Super nickel– cadmium battery and fixed solar panels populated with solar cells of gallium arsenide on germanium. Power is controlled by digital and analog sequential shunts. During the mission the distance from the Sun varies significantly, causing large variations in solar array power and temperature. To minimize solar array size, the solar array and power system electronics were designed to allow operation at voltages both greater than and less than the array’s maximum power voltage. The power system design emphasizes reliability with low weight and volume. Analyses verified subsystem capabilities over the varied mission environment, and early mission flight data indicate that performance is consistent with analysis models. (
Rapamycin nanoparticles improves drug bioavailability in PLAM treatment by interstitial injection

Abstract:Pulmonary lymphangiomyomatosis (PLAM) is a rare interstitial lung disease characterized by diffuse cystic changes caused by the destructive proliferation of smooth muscle-like cells or LAM cells. PLAM is more common in young women than other people, and a consensus is lacking regarding PLAM treatment. The clinical treatment of PLAM is currently dominated by rapamycin. By inhibiting the mTOR signaling pathway, rapamycin can inhibit and delay PLAM's occurrence and development. However, the application of rapamycin also has shortcomings, including the drug's low oral bioavailability and a high binding rate to hemoglobin, thus significantly decreasing the amount of drug distributed to the lungs.

Abstract:A kind of lipid nanoparticle containing rapamycin (LNP@rapa) is prepared. Following subcutaneous injection, LNP@rapa gains entry into lymph nodes via phagocytosis by immature dendritic cell (imDCs). Rapamycin's action involves the inhibition of imDC maturation by restraining the mammalian target of rapamycin (mTOR) pathway, culminating in costimulation blockade with T cells, and thus mitigating transplant rejection. The activation of naive T cells by mature dendritic cells (DCs) presenting allograft antigens marks a pivotal stage in triggering transplant rejection. A critical intervention in this process involves the administration of rapamycin, which disrupts the mTOR signaling pathway, thereby impeding DC maturation. Nevertheless, systemic administration of rapamycin faces challenges due to its limited bioavailability, non‐specific targeting, and notable side effects. To address these limitations, LNP@rapa (liposome‐encapsulated rapamycin) is developed, administered via subcutaneous injection. This formulation selectively targets lymph nodes, inhibiting DC maturation within these nodes and mitigating transplant rejection. This study validates the in vivo efficacy of LNP@rapa, demonstrating its ability to hinder DC maturation, reduce inflammatory cytokine secretion, and significantly prolong graft survival in two distinct mouse transplantation models. This study introduces an innovative strategy targeting lymph nodes to impede DC maturation, offering a promising approach to address transplant rejection.

Abstract:Impaired wound healing is a common complication of orthopedic surgery and poses a difficult challenge in the clinic. Fibroblasts are thought to play a significant role in wound healing, and can be positively affected by low concentrations of rapamycin; however, rapamycin is cytotoxic at higher concentrations. To address this issue, a RAPA/PLGA-PEG drug delivery system was constructed in this study to maintain low concentrations of rapamycin. The results showed that the nanoparticles were stable, had good sustained drug release properties and were able to reduce the toxicity of rapamycin to fibroblasts. These findings suggest that RAPA/PLGA-PEG nanoparticles can reduce the cytotoxicity of rapamycin and may be a potential clinical treatment for impaired wound healing.

Abstract:Objective To investigate the effects of rapamycin on the number and function of peripheral blood endothelial progenitor cells (EPCs). Methods Total mononuclear cells (MNCs) were isolated from peripheral blood by Ficoll density gradient centrifugation, and then the cells were plated on fibronectin-coated culture dishes. After 7-day culture, adherent cells were treated with rapamycin in a series of final concentrations of 1.0, 2.0, 5.0μg/ml for 6, 12, 24, and 48h. EPCs were identified as adherent cells double positive for DiLDL-uptake and lectin binding by direct fluorescent staining as demonstrated under a laser scanning confocal microscope. EPCs were further documented by demonstrating the expression of VEGFR-2, AC133 and CD34 with flow cytometry. EPCs proliferation and migration were assayed with MTT assay and modified Boyden chamber assay, respectively. EPCs adhesion assay was performed by replating them on fibronectin-coated dishes, and then adherent cells were counted. In vitro vasculogenesis activity was assayed by in vitro vasculogenesis kit. Results Incubation of isolated human MNCs with rapamycin resulted in a decrease in the number of EPCs, and rapamycin also decreased EPCs proliferative, migratory, adhesive and in vitro vasculogenesis capacity in both concentration and time dependent manners. Conclusion Rapamycin decreases the number, proliferative, migratory, adhesive and in vitro vasculogenesis capacity of EPCs.
Redv/Peptide Conjugated Rapamycin-Loaded Polymer Matrix for Endothelial Cells Selectivity

Abstract:Combination device containing drug release and HUVECs selectivity were constructed based on PEG copolymer. Polyethylene glycol methacrylate (PEGMA) -butyl methacrylate ( BMA) (PEGB) loading rapamycin was cast onto polyethylene terephthalate( PET) substrate as base layer, and then PEGMA-BMA-p-nitrophenyloxycarbonyl polyethylene glycol methacrylate(MEONP) (PEGBN) conjugated REDV peptide was further coated as top layer. In vitro profile of drug release indicates that rapamycin keeps a slow and sustain release implying that the top layer acts as a diffusion barrier. Cell behavior indicates that rapamycin remains its activity in inhibiting human umbilical vein endothelial cells(HUVECs) and human aortic smooth muscle cells(HASMCs) adhesion and proliferation. Although the eluted concentration of rapamycin decreased with drug eluting time, the polymeric coating keeps resisting nonspecific adsorption of HASMCs while REDV enhances HUVECs attachment specifically. This polymeric film with top layer promotes HUVECs competitive adhesion comparing with HASMCs. The immobilization of REDV onto rapamycin loaded PEG matrix could be an effective method to keep sustained inhibition of HASMCs and promote HUVECs competitive adhesion simultaneously.
Transcutaneous delivery of disease specific PI3K/Akt/mTOR inhibitor based hybrid nanoparticles in hydrogel system for the management of psoriasis: Insights from in vivo studies

Abstract:We have reported robust and scalable lipid polymeric conjugated hybrid nanoparticles comprised of phospholipid shell and polymeric core that combine the benefit of both polymeric and lipid nanoparticles. Rapamycin, a PI3K/Akt/mTORC1 inhibitor, was encapsulated inside the lipid-polymeric conjugated spherical-shaped hybrid nanoparticles (RPMN), having an encapsulation efficiency of ≈ 83% and particle size ≈ 277.6 nm. Further, RPMN was converted into the carpool-based hydrogel system (RPMNGel), which enhanced release kinetics, long-term stability and skin residence time. Specifically, in an in-vivo imiquimod-induced psoriatic model, RPMNGel showed high accumulation and deeper penetration inside the epidermis and slowly diffusing away inside the psoriatic skin without causing any side effect to normal skin. This leads to longer and sustained retention over more than three days without being affected by sweat, humidity or wiping due to adherence between the stratum corneum and epidermis. Similarly, the cumulative PASI score was also reduced from 10.25 to 1.75 on day 7 in the group treated with RPMNGel. Overall, RPMNGel has a potential role in treating and managing psoriasis.

Abstract:Pulmonary delivery of immune checkpoint inhibitors (ICIs) holds promise for increasing drug concentration in the lung and reducing off-target side effects compared to systemic administration. However, the development of an ideal carrier capable of efficiently encapsulating ICIs, maintaining stability during fabrication and nebulization, and facilitating transport through the mucus barrier remains a significant challenge. Herein, we developed such a carrier by synthesizing a responsive polyethylene glycol (PEG) nanogel through crosslinking four-arm PEG via copper-free click chemistry. This approach allows for the efficient in situ encapsulation of ICIs using biocompatible PEG, eliminating the need for potentially toxic catalysts or coupling reagents. The covalently crosslinked network ensures excellent colloidal stability of nanogel during nebulization. Importantly, the PEG nanogel greatly enhances the mucus penetration and lung accumulation of free ICIs. Upon reaching the tumors, the PEG nanogel undergoes dissolution triggered by the overexpressed matrix metalloproteinase-9, leading to the release of encapsulated ICIs. We demonstrated that pulmonary delivery of PEG nanogel greatly enhanced the therapeutic efficacy of ICIs and alleviated the potential toxicity associated with intravenously injected ICIs using a lung metastasis model in mice. Overall, this work presents a simple, safe, and effective PEG nanogel platform for the pulmonary delivery of ICIs.
Inhibitory Effect of Rapamycin-Loaded Nanoparticles on Proliferation of Vascular Smooth Muscle Cells

Abstract:Objective:To observe the inhibitory effect of rapamycin PLGA nanoparticles(RAPA-PLGA-NPs) on proliferation of vascular smooth muscle cells(VSMCs). Methods:Rat VSMCs were prepared by primary culture and identified by immunohistochemical staining for α-actin.Cell uptake of RAPA-PLGA-NPs was observed by using transmission electron microscopy(TEM).The effects of RAPA-PLGA-NPs on the growth and expression of the proteins downstream to mTOR(mammalian target of rapamycin) of VSMCs were determined by MTT assay and Western blot analysis,respectively.Meanwhile,the free rapamycin served as comparison. Results:Rat VSMCs were successfully cultured and identified.TEM results showed significant amount of RAPA-PLGA-NPs uptake into the cytoplasm of VSMCs.The MTT results showed that RAPA-PLGA-NPs(1,10 and 100 ng/mL) inhibited the proliferation of VSMCs in a concentration dependent manner(all P0.05),and the inhibitory effect of RAPA-PLGA-NPs at 1 ng/mL was similar to that of the free rapamycin at 10 ng/mL(P0.05).Western blot results showed that RAPA-PLGA-NPs of both concentrations(1 and 10 ng/mL) markedly reduced phosphorylation levels of S6K1 and 4E-BP1 of VSMCs,and both effects were more obvious than that of the free rapamycin(10 ng/mL). Conclusion:RAPA-PLGA-NPs are of enhanced permeability,can be rapidly taken up by cells,and have better biological effects than their free forms.
