Abstract:With people's increasing awareness of healthy diet, the diverse health-promoting functions of ginseng have been widely recognized. As one of the key functional components, ginseng polysaccharides have attracted increasing research interest. Here, three purified polysaccharide fractions, GPS-1a, GPS-1b, and GPS-2, were obtained from the root extract of Panax ginseng C. A. Meyer. Structurally, GPS-1a and GPS-1b were both linked in a → 6)-α-D-Glcp-(1 → pattern but composed of glucose and galactose in molar ratios of 9.76:0.24 and 9.81:0.19. In contrast, GPS-2 was composed of glucose, galactose, arabinose, rhamnose, and galacturonic acid in a molar ratio of 1.82:1.94:0.79:0.52:4.93. The main backbone consisted of →4)-α-D-GalpA-(1→, →4)-α-D-GalpA-6OMe-(1→, →3, 4)-α-D-GalpA-(1→, →3)-α-L-Rhap-(1 → linages, and its branches are composed of →5)-α-L-Araf-(1→, →4)-β-D-Galp-(1→, →2)-β-D-Glcp-(1→, α-D-GalAp-(1→. Benefitting from this structural variance, GPS-2 exhibited the most significant immunoregulatory and radioprotective efficacies. Specifically, GPS-2 promoted TLR2, NF-κB, and TRAF6 protein expression levels, thereby significantly improving macrophage phagocytosis, splenic lymphocyte proliferation, and stimulation of NO, IL-1β, IL-6, and TNF-α secretion, which activated RAW264.7 and splenic lymphocytes. The following radioprotection activity tests unveiled that GPS-2 increased the organ index, number of peripheral blood cells, cellularity of splenocytes, and bone marrow cell numbers in irradiated mice. This investigation revealed the contribution of polysaccharide structure characteristics to the bioactive expression and elucidated the potential utilization of GPS-2 as a radioprotective agent or immunomodulator.

Abstract:Introduction: Polysaccharides are important components of Panax notoginseng that contribute to its immunomodulatory ability. This study aimed to isolate polysaccharides from notoginseng and investigate the structural feature and potential immunomodulatory activity. Methods: The polysaccharide was isolated from notoginseng by anion exchange and gel permeation chromatography. Its preliminary structure was characterized by Fourier transform infrared (FT-IR) spectroscopy, gas chromatography-mass spectrometry (GC-MS) and nuclear magnetic resonance (NMR) spectroscopy. The immunoregulatory function was further investigated in cyclophosphamide induced immunosuppressive mice, murine splenocytes and macrophages. Results: A novel homogeneous polysaccharide (PNPB1) was isolated from notoginseng with the molecular weight of 9.3 × 10 5 Da. Monosaccharide composition analysis indicated that PNPB1 consisted of Glc (88.2%), Gal (9.0%), Ara (2.4%) and trace GlcA, with the major backbone of (1→4)-linked α-Glcp, (1→6)-linked β-Glcp, and (1, 4→6)-linked β-Glcp. The polysaccharide was found to significantly enhance murine body weight, improve their thymus and spleen indices and increase the white blood cells (WBC). PNPB1 significantly enhanced splenic lymphocyte proliferation, NO and cytokine (TNF-α, IL-2, IL-10 and IFN-γ) production, as well as the phagocytosis and TLR2 expression of peritoneal macrophages, indicating potent immunoenhancement effect. Discussion: These findings provide a theoretical basis for elucidating the structure and immune activity of notoginseng polysaccharides.

Abstract:Panax ginseng C. A. Meyer is a dual-purpose plant for medicine and food, its polysaccharide is considered as an immune enhancer. Four polysaccharides, WGP-20-F, WGP-40-F, WGP-60-F and WGP-80-F were obtained from ginseng via water extraction and gradient ethanol precipitation with different molecular weights ( Mw ) of 1.720 × 10 6 , 1.434 × 10 6 , 4.225 × 10 4 and 1.520 × 10 4 Da, respectively. WGP-20-F and WGP-40-F which with higher Mw and a triple-helix structure are glucans composed of 4-ɑ-Glc p , do not show remarkable immunoregulatory effects. WGP-60-F and WGP-80-F are heteropolysaccharides mainly composed of 4-ɑ-Glc p and also contain t-ɑ-Ara f , 5-ɑ-Ara f and 3,5-ɑ-Ara f . They are spherical branched conformations without a triple-helix structure and can effectively increase the index of immune organs, lymphocyte proliferation, activate macrophages to regulate the immune system in mice and further enhance immune functions by improving delayed-type hypersensitivity reaction and antibody response. These results indicated that WGP-60-F and WGP-80-F could be used as potential immune enhancers, and gradient ethanol precipitation can be applied for the preparation of ginseng bioactive polysaccharide.

Abstract:A protein-bound polysaccharide (GSP-4) with a molecular weight of 8.3×105Da, was isolated from the water extract of the fruiting bodies of Ganoderma sinense. Chemical study revealed that this fraction was composed of mannose, glucose and galactose in the molar ratio of 4.7:27.1:1.0, with the sugar residues of t-, 1,3-, 1,4-, 1,6-, 1,3,4- and 1,3,6-linked Glcp, t-linked Galp, and 1,6-linked Manp. The immnomodulatory effects of GSP-4 were assessed using human peripheral blood mononuclear cells (PBMCs) and murine monocyte/macrophage cell line RAW 264.7. We found that GSP-4 could significantly stimulate the production of the immunomodulatory markers tumor necrosis factor α (TNF-α), interleukin (IL)-1β, IL-12, and granulocyte-macrophage colony-stimulating factor (GM-CSF) in PBMCs. This observation was further substantiated in RAW 264.7 cells, as indicated by the increase of nitric oxide (NO), TNF-α and IL-6 production. GSP-4 also enhanced the expression of inducible NO synthase mRNA in dose-dependent manner. Our current finding gives the first piece of evidence to support that GSP-4 possesses some promising immunomodulating effects and it could be a potential candidate to be further used in related cancer immunotherapy.
Ginsenoside Re and Notoginsenoside R1: Immunologic Adjuvants with Low Haemolytic Effect.

Abstract:The further purification of the total saponins from the roots of Panax notoginseng (BURK.) E H. CHEN by ordinary and reversed-phase silica-gel, as well as Sephadex LH-20 chromatography afforded two adjuvant active dammarane-type saponins, ginsenoside Re (1) and notoginsenoside R, (2). These two saponins were evaluated for haemolytic activities and adjuvant potentials on the cellular and humoral immune responses of ICR mice against ovalbumin (OVA). The concentrations inducing 50% of the maximum haemolysis (HD50), using 0.5% red blood cell suspensions, were 469.6 +/- 16.9 and 420.4 +/- 22.9 mu g/ml for 1 and 2, respectively. Compounds 1 and 2 significantly increased the concanavalin A (Con A)-, lipopolysaccharide (LPS)-, and OVA-induced splenocyte proliferation in the OVA-immunized mice (P < 0.05, P < 0.01, or P < 0.001). The OVA-specific IgG, IgG1, and IgG2b antibody titres in serum were also significantly enhanced by 1 and 2 compared with OVA control group (P < 0.05, P < 0.01, or P < 0.001). The results indicate that 1 and 2 showed a slight haemolytic activity and significant adjuvant effect on specific antibody and cellular immune response against OVA in mice, and that the type of the terminal sugar of the sugar chain at C(6) of protopanaxatriol could not only affect their haemolytic activities and adjuvant potentials, but have significant effects on the nature of the immune responses. The information about this structure-function relationship might be useful for developing semisynthetic dammarane-type saponin derivatives with immunological adjuvant activity.

Abstract:Ginsenoside Rh-4 (1), a saponin isolated from the roots of Panax notoginseng (BURK.) F. H. Compound 1 showed a slight haemolytic effect, its concentration inducing 50% of the maximum haemolysis (HD50 value) being 407 +/- 12 mu g/ml using a 0.5% suspension of red blood cells. Compound 1 significantly increased the concanavalin A (Con A)-, lipopolysaccharicle (LPS)-, and OVA-induced splenocyte proliferation in OVA-immunized mice especially at a dose of 25 mu g (P < 0.05, P < 0.01, or P < 0.001). The OVA-specific serum IgG, IgG1, and IgG2b antibody levels were also significantly enhanced by 1 at a dose of 25 mu g compared to the OVA control group (P < 0.05 or P < 0.01). Moreover, the enhancing effect of 1 on the OVA-specific IgG2b antibody responses to OVA in mice was more significant than that of Alum (Al(OH)(3) gel; P < 0.01). These results suggest that 1 could be safely used as adjuvant with low or non-haemolytic effect.

Abstract:Objective:to investigate the structure and in vitro immunomodulatory activity of polysaccharides from bamboo shavings (BSP).Methods:the structure was analyzed by the methods of methylation and nuclear magnetic resonance spectroscopy;the in vitro immunomodulatory activity was evaluated by the assays of mouse splenic lymphocyte proliferation,the phagocytosis of neutral red and NO secretion of RAW264.7 cell.Results:BSP was O-acetyl-(4-O-methylglucurono)-arabinoxylans consisting of a linear (l→4)-β-D-xylopyranosyl backbone decorated with branches at O-2 of 4-O-methylglucuronic acid units and acetyl groups or O-3 of α-L-arabinofuranosyl and acetyl groups.The structural element could be expressed as below:1,4-β-D-[2-O-Ac][3-O-Ac][4-O-Me-D-GlcpA-(1→2)][α-L-Araf-(1→3)]-xylp;BSP could evidently strengthen concanavalin A or lipopolysaccharide-induced splenic lymphocyte proliferation at 10-100μg/mL;BSP could significantly enhance the phagocytosis activity and NO production of RAW264.7 cell at 10-1 000 μg/mL in a dose-dependent manner.Conclusion:BSP exhibits excellent immunostimulatory activity in vitro,and can be considered as an ideal source of bio-active polysaccharides due to the abundance of its raw material.

Abstract:Protopanaxadiol saponins (Rg3, Rd, Rc, Rb1 and Rb2) and protopanaxatriol saponins (Rg1, Re and Rg2) isolated from the root of Panax ginseng C.A. Meyer were evaluated for their adjuvant effects on the immune responses to ovalbumin (OVA) in mice. BALB/c mice were subcutaneously injected twice at a 3-week interval with 10μg of ovalbumin or 10μg of OVA plus 50μg of ginsenosides Rg3, Rd, Rc, Rb1, Rb2, Rg1, Re or Rg2 or Quil A (n=5). Blood samples were collected for measuring specific total-IgG, IgG1 and IgG2a, and splenocytes were harvested for determining lymphocyte proliferation as well as IFN-γ and IL-5 production 2 weeks after the boosting. The results indicated that OVA-specific antibody responses were significantly higher in mice immunized with OVA co-administered with Rg1, Re, Rg2, Rg3 and Rb1 but not with Rd, Rc and Rb2 when compared with the control (immunized with OVA only). Significantly enhanced splenocyte proliferative responses to Con A, LPS and OVA as well as the production of both IL-5 and IFN-γ stimulated by OVA were also detected in mice immunized with OVA co-administered with Rg1 but not with Rb1, Re and Rg3. Of the ginsenosides studied, Rg1, Re, Rg2, Rg3 and Rb1 have more potent adjuvant properties than the others, indicating that they are the major constituents contributing to the adjuvant activities of total ginseng saponins. Varieties of ginsenosides in adjuvant activity might be attributed to the varieties of molecular conformations determined by the side sugar chains attaching to their dammarane skeleton.

Abstract:The current experiments were designed to explore the structural features and immunopotentiation activity of two homogeneous polysaccharides PCSPA and PCSPB prepared from Crocus sativus petals using DEAE-Sephadex A-50 and Sephadex G200 column chromatography. The structures of PCSPA and PCSPB were systematically characterized using extensive chemical and spectroscopic methods including colorimetry, HPGPC-RID, GC-MS, Smith degradations, methylation, solvolyticdesulfation, UV, FT-IR, NMR, SEM, and AFM. The average molecular weights of PCSPA and PCSPB were 1.98 x 10(6) and 2.53 x 10(6) Da, respectively. PCSPA consisted of Gal, Rha, Ara, and Xyl in the molar ratio of 16:5:7:3, while PCSPB were composed of Gal, Glc, Man, Rha, Ara, and Xyl with molar ratio of 16:2:7:19:15:16. Both polysaccharides contained sulfonic and acetyl groups. PCSPA and PCSPB significantly activated RAW264.7 cells by enhancing the phagocytic activity, up-regulating the expression of surface molecules, promoting the production and mRNA expression of cytokines and chemokines via MAPK and NF-kappa B pathway. Abstract:Spores of Ganoderma lucidum contain a large amount of bioactive substances and have a higher bioactivity than the fruit bodies of G. lucidum. However, ingredients from spores are less studied due to the difficulties in collecting the spores and breaking the rigid shell. In this study, a water-soluble polysaccharide named GSG was extracted from the spores of G. lucidum. GSG is characterized to be a branched glucan that contains several different kinds of linkages. It was an effective inducer of MAPKs- and Syk-dependent TNF-α and IL-6 secretion in murine resident peritoneal macrophages. Dectin-1 could recognize GSG and partially mediate its biological activities. Additionally, in vivo administration of GSG potentiated the Con A-induced proliferative response of splenocytes and induced anti-tumor activity against Lewis lung cancer in mice. Therefore, these results suggest that GSG is an effective immunomodulator and may be a promising adjuvant remedy for anti-tumor therapies.

Abstract:The “off” response of the human electroretinogram has been shown to resemble an inverted “on” response. The off response can only be obtained from a strongly light-adapted eye. The curve of response amplitude against length of stimulus flash passes through a maximum for flash durations of 12–20 msec, depending on the intensity of adaptation and stimulation. The curve of response amplitude against interval between two very short light flashes passes through a minimum for flash intervals of 15–25 msec under similar conditions of adaptation and stimulation. Algebraic addition of on and off responses or of two on responses has been shown to provide an adequate explanation of the two curves.
The Structural Characterization of a Polysaccharide from the Dried Root of Salvia miltiorrhiza and Its Use as a Vaccine Adjuvant to Induce Humoral and Cellular Immune Responses

Abstract:In order to supplement the research gap concerning Salvia miltiorrhiza polysaccharide extracted from Danshen in NMR analysis, and to clarify its immune enhancement effect as an adjuvant, we isolated and purified SMPD-2, which is composed of nine monosaccharides such as Ara, Gal, and Glc from Danshen. Its weight average molecular weight was 37.30 ± 0.096 KDa. The main chain was mainly composed of →4)-α-D-Galp-(1→, →3,6)-β-D-Glcp-(1→ and a small amount of α-L-Araf-(1→. After the subcutaneous injection of SMPD-2 as an adjuvant to OVA in mice, we found that it enhanced the immune response by activating DCs from lymph nodes, increasing OVA-specific antibody secretion, stimulating spleen lymphocyte activation, and showing good biosafety. In conclusion, SMPD-2 could be a promising candidate for an adjuvant.

Abstract:Purpose. The main purpose of this study is to prepare and characterize polysaccharides from Panax notoginseng, investigate their effects on immune system in vitro in order to find new immunostimulants for the prevention and supporting treatment of infection and immunodeficiency related diseases.Methods. Polysaccharides were extracted with aqueous solution, separated with column chromatography. Their anticomplementary activities were investigated by using human serum and antibody-sensitized sheep red blood cells. Interferon-γ and tumor necrosis factor inductive activities were studied by using isolated mouse spleen lymphocytes and peritoneal macrophages, respectively.Results. Four polysaccharides, homogeneous in gel-filtration chromatography, were prepared and designated PF3111, PF3112, PBGA11, and PBGA12. Component sugar analysis revealed that they are heteroglycans with MWs ranging from 37 kD to 760 kD, composed of glucose, galactose, arabinose, mannose, and xylose in different molar ratios. Fraction PBGA12 has the most anticomplementary activity which is mediated through both alternative and classical pathways. All the polysaccharides except PBGA11 induced the production of interferon-γ in the presence of concanavalin A. They induced the production of significant amount of TNF-α in cell cultures.Conclusions. The polysaccharides from P. notoginseng have immunostimulating activities invitro.

Abstract:OBJECTIVE To investigate the structural feature and immunomodulatory activity of the water-soluble polysaccharide DMP4a-1 from Dendrobium moniliforme (L.) Sw .METHODS The chemical structure of the polysaccharide DMP4a-1 was elucidated by periodate oxidation, HPGPC, TLC,GC,GC-MS and IR;TNF-α was quantified by sandwich ELISA, the proliferation of spleen lymphocyte was measured by CCK-8 reagent in vitro.RESULTS DMP4a-1 was composed of Glc-Man-Rha-Ara-Gal in a molar ratios of 2.873:2.850:1.762:1.279:1 with Mr 3 049. They were (1→4, 1→3, 1→6) linked with glycosidic residues, in a ratio of 1.194:2.430:1. DMP4a-l significantly stimulatd the secretion of TNF-α in macrophage and promoted the proliferarion of lymphocyte from spleen in vitro. CONCLUSION Polysaccharide DMP4a-1 consists of β-D-pyranyl glyeoside with (1→3)glycosidic bond, and markedly enhances immunological activity.

Abstract:Four protopanaxadiol-type saponins (PDS), ginsenosides-Rb1, -Rd, notoginsenosides-K, -R4 isolated from the roots of Panax notoginseng were evaluated for their haemolytic activities and adjuvant potentials on the cellular and humoral immune responses of ICR mice against ovalbumin (OVA). The effect of the substitution pattern of these PDS on their biological activities was investigated and structure–activity relationships were established. Among four PDS, the ranking of the haemolytic activity was K>R4>Rb1>Rd (P<0.01 or <0.001). Rd, Rb1, and K could significantly enhance mitogen- and OVA-induced splenocyte proliferation in the OVA-immunized mice (P<0.001), with the order in terms of stimulation index being Rd>Rb1>K>R4. OVA-specific IgG, IgG1, IgG2a and IgG2b antibody levels in the OVA-immunized mice were significantly enhanced by four PDS. Adjuvant potentials of Rd on antibody responses were higher than those of other three PDS. Meanwhile, Rd also significantly enhanced the production of the Th1 and Th2 cytokines in OVA-immunized mice (P<0.05 or <0.01). The structure–activity relationship studies suggested that the length of sugar side chains at position C-20 and the linkage of glucose moiety at position C-3 of protopanaxadiol could affect the haemolytic and adjuvant activities of PDS. The information about this structure/function relationship might be useful for developing semisynthetic tetracyclic triterpenoid saponin derivatives with immunological adjuvant activity, as well as a reference to the distribution of the functional groups composing the saponin molecule.

Abstract:The further purification of the total saponins from the roots of Panax notoginseng by using ordinary and reversed-phase silica-gel, as well as Sephadex LH-20 chromatography afford seven adjuvant active protopanaxatriol-type saponins (PTS), ginsenosides-Rh1 (Rh1),-Rh4 (Rh4),-Rg1 (Rg1),-Re (Re), notoginsenosides-R1 (R1),-R2 (R2),-U (U). These saponins were evaluated for their haemolytic activities and adjuvant potentials on the cellular and humoral immune responses of ICR mice against ovalbumin (OVA). The effect of the substitution pattern of these PTS on their biological activities was investigated and structure-activity relationships were established. Among seven PTS, the haemolytic activity of Rh1 was higher than that of other six compounds (p<0.001) The HD50 values of Rh4 and U were significantly bigger than those of R2, Rg1 and Re (p<0.05 or p<0.01). Seven PTS could significantly increase the concanavalin A (Con A)-, lipopolysaccharide (LPS)- and OVA-induced splenocyte proliferation in the OVA-immunized mice (p<0.01 or p<0.001). The OVA-specific IgG, IgG1, IgG2a and IgG2b antibody levels in serum were also significantly enhanced by seven PTS compared with OVA control group (p<0.01 or p<0.001). The structure-activity relationship studies suggested that the number, the length and the position of sugar side chains, and the type of glucosyl group in the structure of PTS could not only affect their haemolytic activities and adjuvant potentials, but have significant effects on the nature of the immune responses. The information about this structure/function relationship might be useful for developing semisynthetic tetracyclic triterpenoid saponin derivatives with immunological adjuvant activity, as well as a reference to the distribution of the functional groups composing the saponin molecule.
Characterization of the structure, anti-inflammatory activity and molecular docking of a neutral polysaccharide separated from American ginseng berries

Abstract:Aim: American ginseng berries, grown in the aerial parts and harvested in August, are a potentially valuable material. The aim of the study was to analyze the specific polysaccharides in American ginseng berries, and to demonstrate the anti-inflammation effect through in vitro and in vivo experiments and molecular docking. Methods: After deproteinization and dialysis, the extracted crude polysaccharide was separated and purified. The structure of the specific isolated polysaccharide was investigated by Fourier Transform infrared spectroscopy (FT-IR), GC-MS and nuclear magnetic resonance (NMR), and anti-inflammatory activity was evaluated using in vitro and in vivo models (Raw 264.7 cells and zebrafish). Molecular docking was used to analyze the binding capacity and interaction with cyclooxygenase-2 (COX-2). Results: A novel neutral polysaccharide fraction (AGBP-A) was isolated from American ginseng berries. The structural analysis demonstrated that AGBP-A had a weight-average molecular weight (Mw) of 122,988 Da with a dispersity index (Mw/Mn) value of 1.59 and was composed of arabinose and galactose with a core structure containing →6)-Gal-(1→ residues as the backbone and a branching substitution at the C3 position. The side-chains comprised of α-L-Ara-(1→, α-L-Ara-(1→, →5)-α-L-Ara-(1→, β-D-Gal-(1→. The results showed that it significantly decreased pro-inflammatory cytokines in the cell model. In a zebrafish model, AGBP-A reduced the massive recruitment of neutrophils to the caudal lateral line neuromast, suggesting the relief of inflammation. Molecular docking was used to analyze the combined capacity and interaction with COX-2. Conclusion: Our study indicated the potential efficacy of AGBP-A as a safe and valid natural anti-inflammatory component.

Abstract:The haemolytic activities and adjuvant potentials of Platycodon grandiflorum saponin (PGS) and its fractions on the cellular and humoral immune responses of ICR mice against ovalbumin (OVA) were evaluated. PGS was subjected to silica gel column chromatography to afford four fractions, and two fractions PGSC and PGSD selected for testing for activities because of containing dominant saponin peaks. PGS, PGSC, and PGSD showed a slight haemolytic effect, with their HD50 value being 37.91+/-2.24, 21.30+/-1.22, 37.58+/-1.86 microg/ml against 0.5% rabbit red blood cell, respectively. ICR mice were immunized subcutaneously with OVA 100 microg alone or with OVA 100 microg dissolved in saline containing Alum (200 microg), Quil A (10 microg), PGS (50, 100 or 200 microg), PGSC, or PGSD (25, 50 or 100 microg) on days 1 and 15. Two weeks later (day 28), concanavalin A (Con A)-, pokeweed (PWM)-, and OVA-stimulated splenocyte proliferation and OVA-specific antibodies in serum were measured. PGS and PGSC significantly enhanced the Con A-, PWM-, and OVA-induced splenocyte proliferation in OVA-immunized mice at three doses (P<0.01 or P<0.001). However, no significant differences (P>0.05) were observed among the OVA group, OVA/Alum group and OVA/PGSD group. OVA-specific IgG, IgG1, and IgG2b antibody levels in serum were significantly enhanced by PGS, PGSC, and PGSD compared with OVA control group (P<0.05, P<0.01, or P<0.001). Moreover, the adjuvant effects of PGSC (50 or 100 microg) on the OVA-specific IgG, IgG1, and IgG2b antibody responses to OVA in mice were more significant than those of Alum. In conclusion, PGS seem to be a promising balanced Th1 and Th2 directing immunological adjuvants which can enhance the immunogenicity of vaccine.
Structural Characteristics of Polysaccharide GP2a in Gardeniajasminoides and Its Immunomodulatory Effect on Macrophages

Abstract:Here, we elucidated the structural characteristics of a polysaccharide isolated from Gardenia jasminoides Ellis (labeled as GP2a) and its immunomodulatory activity. GP2a is an acidic polysaccharide with a molecular weight of 44.8 kDa, mostly comprising galacturonic acid. Methylation analysis revealed 4-GalpA (74.8%) to be the major sugar residue in GP2a. Nuclear magnetic resonance analysis indicated that its main chain comprised →4)-α-D-GalpA-6-OMe-(1→4)-α-D-GalpA-(1→ and →4)-α-D-GalpA-6-OMe-(1→2)-α-L-Rhap-(1→, with galactan and arabinans linked to the C-4 position of →2)-α-L-Rhap-(1→ residue as branched chains. Furthermore, GP2a showed no obvious toxicity to macrophages (RAW 264.7) while enhancing cell viability in a dose- and time-dependent manner. Compared with untreated cells, nitric oxide production and secretion of cytokines, such as tumor necrosis factor-α, interferon-γ, interleukin (IL)-1β, IL-6, and granulocyte macrophage colony stimulating factor, in GP2a-treated cells significantly increased after 48 h. At 300 μg/mL GP2a concentration, there was no significant difference in the cytokine levels in GP2a- and lipopolysaccharide-treated cells (the positive control). In summary, GP2a is a pectic polysaccharide with homogalacturonan and rhamnogalacturonan-I structural regions in the main chain. Based on its immunomodulatory effects in vitro, GP2a may have potential uses in functional food and medicine.
