Abstract:In grassland ecosystems, large herbivorous animal grazing activity and increasing nitrogen deposition strongly alters microbial community structure and function. Understanding the effects of grazing and nitrogen addition on the spatial heterogeneity in soil microbial community structure, enzymatic activities and the underlying mechanisms are crucial for making better predictions of soil organic matter dynamics and nutrient cycling.We examined the spatial heterogeneity of soil microbial community structure and enzymatic activity associated with changes in soil microclimate, soil characteristics, plant biomass and soil nutrient responses to grazing and nitrogen addition using a manipulative experiment with control (CK), grazing (G), nitrogen addition (N) and grazing plus nitrogen addition (NG) treatments in a Leymus chinensis meadow steppe, in northeastern China.The results demonstrated that soil microbial community structure and enzymatic activities showed a high level of spatial dependence [C/(C + C0)≥0.9] in the CK plot. G, N and NG treatments not only reduced the spatial variability of soil microbial community structure and enzymatic activities, but also reshaped the spatial links between enzymes activities and microbial community structure. Litter biomass, soil temperature and soil nutrients (soil dissolved inorganic nitrogen or soil dissolved organic carbon) explained 21-27% of the spatial variability of soil microbial community structure in the CK treatment and pH was the strongest driver for the spatial variability of soil enzymatic activities. Meanwhile, the homogenization in soil water content induced by the N addition treatment was a determinant of the reduction in spatial heterogeneity of the microbial community structure. The combination of soil physicochemical properties (bulk density, soil pH and soil dissolved inorganic nitrogen), soil temperature and root biomass explained 32-43% of the spatial variability of the microbial community structure in the G treatment, and N and G treatments had additive effects on the spatial heterogeneity of total PLFAs by homogenizing root biomass. Plant biomass and microbial community structure were the major drivers for the spatial heterogeneity of enzymatic activities under G, N and NG. In NG, the change in spatial variability of enzymatic activities was dominated by N addition. Regardless of grazing, N addition facilitated the spatial correlation between microbial community structure and enzyme activities.Overall, our results revealed the drivers of soil microbial community structure and enzymatic activities spatial pattern shift due to grazing and N addition, highlighting the role that spatial variability in soil microbial community structure and enzymatic activities has on the L. chinensis meadow steppe.

Abstract:Abstract As a grassland management strategy in the typical steppe, livestock grazing can affect soil microbes and ecosystem functions via vegetation removal, trampling and manure deposition. However, the effects of grazing and its main consequences (e.g. the shifts in plant functional types and alterations in soil properties) on soil bacterial and fungal community composition remain unresolved. Taking advantage of a 5‐year grazing exclosure experiment that tested different grazing intensities [control: no grazing (G0); light: 0.75 sheep ha −1 (G1); moderate: 1.50 sheep ha −1 (G2); heavy: 2.25 sheep ha −1 (G3); and overgrazing: 3.00 sheep ha −1 (G4)], we examined grazing‐related changes in plant communities, soil chemistry and soil microbial communities. Grazing intensity affected the diversity and composition of soil bacterial and fungal communities via changes in plant species and soil nutrient contents. Increased grazing intensity reduced soil organic carbon (OC), suppressed the dominant bacterial phyla Proteobacteria and Bacteriodetes, and increased the abundance of oligotrophic bacteria in subordinate phyla (e.g. Verrucomicroba and Chloroflexi). Increased grazing intensity also promoted the dominant fungal phylum Ascomycetes because of the poor‐nutrient soil environment. The decreased soil carbon‐to‐nitrogen (C/N) ratio in the G2 treatment was associated with an effect on the fungal community composition. Structural equation modelling demonstrated that responses of the bacterial community to grazing intensity were related to changes in soil OC and soil pH, whereas the combination of C 4 plant community biomass and soil pH drove changes in the fungal community. Our study provides novel evidence that grazing affects plant functional group and soil chemistry through the soil pH and C/N ratio, which in turn affects bacterial and fungal communities. Highlights Heavy and overgrazing suppressed bacterial phylum Proteobacteria and increased fungal phylum Ascomycete . Soil OC was the predominant driver of changes in bacterial richness and community composition. C 4 plant community biomass modulated fungal richness and community composition.

Abstract:Herbivore grazing has major effects on soil nutrient dynamics in a variety of grassland ecosystems. Previous studies have examined how large herbivores as a group affect nutrient cycling, but little information is available on how assemblages of different herbivore species may influence nutrient cycling, and whether herbivore assemblage effects are influenced by plant community characteristics (e.g. composition, diversity) of the grazed grassland. We conducted a 5‐year, replicated grazing experiment to test the effects of different large herbivore assemblages (cattle grazing, sheep grazing, combined cattle and sheep grazing, no grazing) under moderate grazing intensity on soil nitrogen (N) mineralization rate in two types of grassland communities (high forbs/high diversity and low forbs/low diversity) in meadow steppe habitat of northeast China. Moreover, we examined two distinctly different pathways that herbivores could influence soil N availability: directly through urine and dung deposition and indirectly by shifting grassland species composition (i.e. the grass: forb ratio), thereby the quality of plant litter available to soil decomposers. We found that grazer effects on soil N availability (indexed with anion and cation adsorption strips) depended on herbivore assemblage, and the herbivore assemblage effects varied in the two types of grasslands. In one type of grassland characterized by low diversity, grazing by each of the herbivore assemblages enhanced soil N availability compared to the ungrazed plots, and mixed species (cattle and sheep) grazing had a greater effect than single species grazing. In high diversity grassland, single species herbivore grazing significantly increased soil N availability, but mixed grazing had no effect. Mixed linear modelling revealed that soil N availability was facilitated primarily by excreta additions to the soil and secondarily by the abundance of grasses. Synthesis. Grazers increased soil N availability directly by adding readily accessible N in urine and dung to the soil. Herbivores indirectly influenced soil N availability by altering the plant composition (grass: forb cover). Both mechanisms contributed to the variation in how different herbivore assemblages affected soil N availability in the two grassland types. Abstract:Grazing causes changes in microbiome metabolic pathways affecting plant growth and soil physicochemical properties . However, how grazing intensity affects microbial processes is poorly understood. In semiarid steppe grassland in northern China, shotgun metagenome sequencing was used to investigate variations in soil carbon (C) and nitrogen (N) cycling-related genes after six years of the following grazing intensities: G0, control, no grazing; G1, 170 sheep days ha −1 year −1 ; G2, 340 sheep days ha −1 year −1 ; and G3, 510 sheep days ha −1 year −1 . Taxa and functions of the soil microbiome associated with the C cycle decreased with increasing grazing intensity. Abundances of genes involved in C fixation and organic matter decomposition were altered in grazed sites, which could effects on vegetation decomposition and soil dissolved organic carbon (DOC) content. Compared with the control, the abundances of nitrification genes were higher in G1, but the abundances of N reduction and denitrification genes were lower, suggesting that light grazing promoted nitrification, inhibited denitrification, and increased soil NO 3 − content. Q-PCR further revealed that the copies of genes responsible for carbon fixation ( cbbL ) and denitrification ( norB ) decreased with increasing grazing intensity. The highest copy numbers of the nitrification genes AOA and AOB were in G1, whereas copy numbers of the denitrification gene nirK were the lowest. A multivariate regression tree indicated that changes in C fixation genes were linked to changes in soil DOC content, whereas soil NO 3 − content was linked with nitrification and denitrification under grazing. Thus, genes associated with C fixation and the N cycle affected how C fixation and N storage influenced soil physicochemical properties under grazing. The findings indicate that grazing intensity affected C and N metabolism. Proper grassland management regimes (e.g., G1) are beneficial to the balances between ecological protection of grasslands and plant production in the semiarid steppe.

Abstract:Nitrifying microbes are of critical importance in regulating efficient nitrogen (N) cycling, which plays a crucial role in plant productivity and maintaining soil sustainability. Long-term different intensities of grazing can strongly influence the microbial communities, while our understanding of the complex nitrifying community in the grazed grassland soil environment is still limited. To investigate whether and how long-term grazing with different intensities influence soil nitrifying communities, high-throughput sequencing and quantitative PCR analyses were performed on soil samples from permanent grassland soils under four grazing intensities: 0 (G0), 1.5 (G1), 6 (G2) and 9 (G3) sheepha−1. Results showed that the G3 treatment significantly reduced the soil nutrient content and increased the soil bulk density, changes that are not sustainable in the long run. The G1 treatment, on the other hand, significantly increased the soil nutrient content and would improve soil fertility. Some functional microbes were specifically enriched after long term grazing, like Nitrospirae (phylum) to Nitrospira (class) in the G2 samples and Chromatiales (order) to Nitrosococcus (genus) in the G3 soils. The numerically dominant Nitrosococcus watsonii lineage of ammonia oxidizing bacteria (AOB) was observed in this grassland soil. The redundancy analysis (RDA) together with the structural equation modeling (SEM) analysis showed that grazing intensity was important in mediating the distribution of soil microorganisms and affected nitrifying communities by impacting soil physicochemical characteristics (e.g., bulk density, NH4+-N). These results showed the shifts of nitrifying communities across different grazing intensities, and could aid in the determination of an optimal grazing intensity for these grazed grassland soils.

Abstract:Ungulate grazing is known to play a crucial role in regulating nutrient cycling and controlling plant community structure and productivity in grassland ecosystems. However, little is known about the effects of grazing intensities on soil bacterial community structure and diversity, particularly at the long-term scale. In this study, we measured plant biomass and diversity, soil characteristics and bacterial community structure, and diversity in a 16-year field experiment that had four grazing intensity treatments (non-grazed, CK; low-intensity grazing (LG), moderate-intensity grazing (MG), and high-intensity grazing (HG)) in an Inner Mongolian typical grassland. The CK, LG, MG, and HG sites were grazed by 0.00, 1.33, 4.00, and 6.67 sheep ha −1 , respectively. Bacterial community structure and diversity under grazing intensity treatments were assessed with PCR amplification of DNAs extracted from soils and denaturing gradient gel electrophoresis (DGGE) separation. The results showed that the CK soil had higher moisture, organic C, NH 4 + –N, and NO 3 − –N concentrations than grazed soils, and the HG treatment had the lowest plant biomass and diversity across all the treatments. Principal component analysis of DGGE patterns showed that the LG and MG treatments were different from the CK and HG treatments. In addition, soil bacterial diversities in the LG and MG treatments were significantly higher than those in the other treatments. The relationships between environmental variables and soil bacterial community structure were assessed using redundancy analysis, and we found that soil moisture content, Artemisia frigida biomass, and pH were the best indicator of the changes in soil bacterial community structure among all the treatments. Overall, our results indicated that intermediate grazing intensities (LG and MG) increased soil bacterial diversities, and along with previous studies in this area, we suggested the MG treatment was the most suitable management practice in the Inner Mongolian steppe, not only supporting greater livestock amounts but also harboring greater bacterial diversity.

Alpha-diversity in the bacterial community was strongly correlated with soil organic content (SOC) and soil water content, while the alpha-diversity of the fungi depended on the SOC and pH of the soil. Grazing treatments LG, HG and OG caused strong shifts in bacterial and fungal community composition. Heavy grazing (HG and OG) significantly increased the relative abundances of Chloroflexi, Gemmatimonadetes, and Firmicutes bacteria, while light grazing (LG) significantly decreased the relative abundance of Actinobacteria. Grazing intensities HG and OG increased the relative abundances of certain fungi (e.g., Ascomycota). Co-occurrence network analysis indicated that bacterial communities had a more complex network than fungal communities. A multivariate regression tree demonstrated that the bacterial community responded to grazing via changes in the biomass of perennial plant species and SOC, whereas the SOC and pH value altered the fungal community composition.ConclusionsOur findings indicate that different grazing intensities can initiate different changes in the soil microbiome; sustainable grazing intensity over decades facilitates the recovery of primary productivity and ecosystem functions in a desert steppe.

Electronic Prescribing Usability: Reduction of Mental Workload and Prescribing Errors Among Community Physicians.

Abstract:BACKGROUND Medical errors are common in hospitals, and research is always needed to find ways of reducing these. This study attempts to address three gaps in this field. First, the factors leading to the reduction of mental workload and its relationship with the reduction of prescribing errors by improving electronic prescribing (e-prescribing) usability have not been empirically examined before. Second, the past research in the field of e-prescribing usability lacks robust theoretical models. Third, there are no existing studies to examine the direct influences of user interface consistency and error prevention with the reduction of mental workload and prescribing errors. MATERIALS AND METHODS A quantitative survey method was used to collect data from 188 community physicians. The partial least squares path modeling technique was applied to analyze the data. RESULTS Prescribing errors were reduced by improving the information quality, user interface consistency, system ease of use, and mental workload reduction. Mental workload is reduced by ease of use, error prevention, and consistency. No significant relationships between prescribing error reduction with error prevention and also between information quality with mental workload reduction were found. CONCLUSIONS The designers of e-prescribing should improve the error prevention and consistency of the system and make it easy to use if they wish for the system to reduce users' mental workload. They should also improve the system information quality, ease of use, and consistency if they claim that their system reduces physicians' prescribing errors. The system should also reduce users' mental workload to meet this objective.
Vertebrate herbivore-induced changes in plants and soils: linkages to ecosystem functioning in a semi-arid steppe

Abstract:Large grazing herbivores have been reported to determine the structure and function of grassland ecosystems. However, the ecological linkages between structure and functioning components have yet been thoroughly explored. Here, we test the hypothesis of the impact of grazing on soil nematode community (e.g. structure and composition) and linkages to ecosystem functioning (e.g. soil N mineralization and ANPP) via changes in pathways of plant community, soil nutrients and soil environment using a field experiment maintained for 5 years with seven levels of grazing intensity in the Inner Mongolian grassland. A structural equation model (SEM) with nematode abundances as response variables showed that plant‐feeding and fungal‐feeding nematodes were driven by changes in the plant community, and bacterial‐feeding nematodes were affected by soil abiotic nutrients and environment, while omnivorous + carnivorous nematodes were altered by soil environment and bacterial‐feeding nematodes. This indicates that the top‐down control by grazing leads to bottom‐up control in the soil food web. We found that grazing affected the ecosystem functioning via different pathways. Grazing effects soil N mineralization by changing plant community, soil nutrients, soil environment and nematodes community structure, while it affects ANPP by altering soil N mineralization and soil environment. Our findings could provide a better understanding of the responses of plants and soils to grazing and the linkages between structure and functioning of above‐ground and below‐ground in the semi‐arid steppe. Abstract:Nutrient addition can alter plant–soil‐microbe interactions. However, knowledge remains limited about how the soil biotic and abiotic properties drive changes in plant community composition and diversity after nitrogen (N) addition. In this study, four N addition rates (N0, N3, N6, and N9: 0, 3, 6, and 9 g N m −2 yr−1) were applied to a grassland community on the Loess Plateau (China) to evaluate the response of plant community biomass, evenness, and composition to N addition. Overall, N addition significantly increased plant shoot and root biomass by 27.75%–31.37% and 39.60%–65.42% compared with N0, but no significant difference was observed among N3, N6, and N9. N addition significantly decreased shoot/root biomass ratio by 7.17%–21.84%, and plant community evenness by 3.80%–8.80% compared with N0, particularly under N3 and N6 treatments. Furthermore, N addition enhanced the abundance of bacteria and fungi, but their composition was not altered. Responses of plant biomass and evenness to N addition were correlated to changes in NO3−‐N, soil total phosphorus, and the abundance of bacteria and fungi; additionally, the composition was primarily affected by the microbial abundance. Our research indicates that N addition can significantly alter plant community composition, particularly at low addition rates, and such alteration may be more affected by the soil microbial abundance. The findings of this study have important implications for the sustainable management of grassland ecosystems.

Abstract:In many grassland ecosystems, nitrogen (N) and phosphorus (P) are added to improve plant productivity, and the aboveground plant biomass is mowed and stored as hay for the bullamacow. Nutrient addition and mowing affect the biodiversity and ecosystem functioning, and most of the previous studies have primarily focused on their effects on macro-organisms, neglecting the responses of soil microbial communities. In this study, we examined the changes in three community attributes (abundance, richness, and composition) of the entire bacterial kingdom and 16 dominant bacterial phyla/classes in response to mowing, N addition, P addition, and their combinations, by conducting a 5-year experiment in a steppe ecosystem in Inner Mongolia, China. Overall, N addition had a greater effect than mowing and P addition on most of these bacterial groups, as indicated by changes in the abundance, richness and composition in response to these treatments. N addition affected these soil bacterial groups primarily through reducing soil pH and increasing available N content. Meanwhile, the 16 bacterial phyla/classes responded differentially to these experimental treatments, with Acidobacteria, Acidimicrobidae, Deltaproteobacteria, and Gammaproteobacteria being the most sensitive. The changes in the abundance, richness, and composition of various bacterial groups could imply some potential shift in their ecosystem functions. Furthermore, the important role of decreased soil pH caused by N addition in affecting soil bacterial communities suggests the importance of restoring acidified soil to maintain soil bacterial diversity.

Abstract:Soil microorganisms promote material transformation and energy flow in the entire ecological environment and play a key role in the stability and development of grassland ecosystems. Studies on the impacts of grazing on the soil microbial community and the establishment of a reasonable grazing intensity are crucial to improve our knowledge of the mechanisms underlying grassland degradation and to accurately assess the influence of grazing management on grassland functions and the nutrient cycle. Based on the grassland grazing control experimental platform, we compared the structure and diversity characteristics of soil microbial communities under six grazing intensities (0.00, 0.23, 0.34, 0.46, 0.69, and 0.92 AU ha−1) (1 AU = 500 kg of adult cattle) on the Hulunbuir Leymus chinensis meadow steppe. The results showed that soil microbial biomass carbon (MBC) and nitrogen (MBN) decreased with increasing soil depth. The 0–10 cm soil layer of G0.34 had the highest MBC and MBN, and the G0.92 treatment had the lowest MBC and MBN. Heavy grazing significantly decreased the MBC and MBN contents in the soil surface layer. The soil bacterial diversity under light grazing treatment (0.23 AU ha−1) was higher than that under heavy grazing, and the fungal diversity under the no-grazing treatment was higher than that under the grazing treatment. Overgrazing reduced the bacterial species in the soil. The plant belowground biomass significantly (p = 0.039) influenced the bacterial community structure, and the soil pH (p = 0.032), total nitrogen (p = 0.011), and litter (p = 0.007) significantly influenced the fungal community. The effects of grazing on microbial communities were primarily driven by vegetation productivity, litter mass, and soil geophysical and chemical characteristics. This study deepened our understanding of the impacts of grazing practices on soil microbial communities on the meadow steppe, suggesting that moderate-disturbance grazing can promote the sustainable development of grassland vegetation-soil microorganisms.

Abstract:Due to the region’s social economy and historical culture, rough grazing has led to unresolved grassland-based ecological problems in Northern China. Soil microorganisms are essential structural and functional components of underground ecosystems, and the effects of various grazing intensities on the physicochemical properties and bacterial communities of soil are unclear. A stocking density regulation experiment was carried out in the desert steppe of the Inner Mongolia Autonomous Region. In the study area, four grazing intensities were set, namely, the enclosure control group (CK), light grazing, moderate grazing, and heavy grazing. Field investigations and 16S rRNA sequencing were used to compare and analyze the characteristics of soil bacterial community structures and their correlations with soil nutrient factors under different grazing intensities. The experiment showed the following results: (1) The Shannon, Simpson, and Pielou indices of the light grazing group were significantly higher than those of the CK (p < 0.05), and the indices of the moderate and heavy grazing groups were lower than those of the CK, but the difference was not significant, and there were no significant differences in the Chao1 index between each group. (2) Acidobacteria, Actinobacteria, Proteobacteria, and Chloroflexi were the main bacterial phyla. (3) With the exception of soil organic matter and available potassium, which had significant negative correlations with the Shannon index (p < 0.05), other soil factors had no significant correlation with the soil bacterial diversity. (4) The contents of soil organic matter, total phosphorus, alkali-hydrolyzed nitrogen, available phosphorus, and available potassium influenced the differences between soil bacterial communities under different grazing intensities.

Abstract:AbstractSoil microorganisms play key roles in terrestrial biogeochemical cycles and ecosystem functions. However, few studies address how long-term nitrogen (N) addition gradients impact soil bacterial and fungal diversity and community composition simultaneously. Here, we investigated soil bacterial and fungal diversity and community composition based on a long-term (17 years) N addition gradient experiment (six levels: 0, 2, 4, 8, 16, 32 gN m−2 year−1) in temperate grassland, using the high-throughput Illumina MiSeq sequencing. Results showed that both soil bacterial and fungal alpha diversity responded nonlinearly to the N input gradient and reduced drastically when the N addition rate reached 32 g N m−2 year−1. The relative abundance of soil bacterial phyla Proteobacteria increased and Acidobacteria decreased significantly with increasing N level. In addition, the relative abundance of bacterial functional groups associated with aerobic ammonia oxidation, aerobic nitrite oxidation, nitrification, respiration of sulfate and sulfur compounds, and chitinolysis significantly decreased under the highest N addition treatment. For soil fungi, the relative abundance of Ascomycota increased linearly along the N enrichment gradient. These results suggest that changes in soil microbial community composition under elevated N do not always support the copiotrophic-oligotrophic hypothesis, and some certain functional bacteria would not simply be controlled by soil nutrients. Further analysis illustrated that reduced soil pH under N addition was the main factor driving variations in soil microbial diversity and community structure in this grassland. Our findings highlight the consistently nonlinear responses of soil bacterial and fungal diversity to increasing N input and the significant effects of soil acidification on soil microbial communities, which can be helpful for the prediction of underground ecosystem processes in light of future rising N deposition.

Grazing alters the soil nematode communities in grasslands: A meta-analysis

Abstract:Grazing causes great disturbances in grassland ecosystems and may change the abundance, diversity, and ecological function of soil biota. Because of their important role in nutrient cycling and as good environmental indicators, nematodes are very representative soil organisms. However, the mechanisms by which grazing intensity, livestock type, duration, and environmental factors (e.g., climate and edaphic factors) affect soil nematodes remain poorly understood. In this study, we collected 1964 paired observations all over the world from 53 studies to clarify the grazing response patterns of soil nematodes and their potential mechanisms. Overall, grazing significantly decreased the abundance of bacterial-feeding (BF) nematodes (-16.54%) and omnivorous-predatory (OP) nematodes (-36.81%), and decreased nematode community diversity indices (Shannon-Weiner index: -4.33%, evenness index: -9.22%, species richness: -5.35%), but had no effect on ecological indices under a global regional scale. The response of soil nematodes to grazing varied by grazing intensity, animals, and duration. Heavy grazing decreased OP nematode abundance, but had no effect on the abundance of other trophic groups, or on diversity or ecological indices. Grazing by small animals had stronger effects than that by large animals and mixed-size animals on BF, fungal-feeding (FF), plant-feeding (PF) and OP nematodes, the Shannon-Wiener index, and the species richness index. The abundance of FF and OP nematodes influenced significantly under short-term grazing. The evenness index decreased significantly under long-term grazing (>10 years). Climate and edaphic factors impacted the effects of grazing on nematode abundance, diversity, and ecological indices. When resources (i.e., rain, heat, and soil nutrients) were abundant, the negative effects of grazing on nematodes were reduced; under sufficiently abundant resources, grazing even had positive effects on soil nematode communities. Thus, the influence of grazing on soil nematode communities is resource-dependent. Our study provides decision makers with grazing strategies based on the resource abundance. Resource-poor areas should have less grazing, while resource-rich areas should have more grazing to conserve soil biodiversity and maintain soil health.

Abstract:To investigate the effects of stocking rates on nitrification activity and active nitrifying communities in a typical steppe grazing system, we conducted a laboratory incubation study using soil from a 10-year-old grassland gradient grazing experiment with sheep. A combination of molecular methods, such as DNA-based stable-isotope probing (DNA-SIP), real-time quantitative PCR, and high-throughput sequencing, was used to identify changes of nitrification activity and active nitrifying communities under different stocking rates (0 (SR0), 3 (SR3), 6 (SR6), and 9 (SR9) sheep per ha). The nitrification activity of soils was significantly increased by light grazing (SR3), while it was significantly decreased by heavy grazing (SR9). Nitrososphaera viennensis lineage of ammonia-oxidizing archaea (AOA) functionally predominated over ammonia-oxidizing bacteria (AOB) in nitrification in the SR3 soil, while the Nitrosospira cluster 3 of AOB was the major player in the SR9 soil. Therefore, stocking rates altered the distribution of active nitrifying communities by affecting soil chemical and physical conditions.
Response of microbial communities to the changes in grazing intensity and season in a typical steppe

Abstract:Livestock grazing is an influencing factor playing a key role in shaping the plant community, microbial community, and soil properties in grassland ecosystems. Northern China's Loess Plateau has been used for livestock grazing for centuries and is a vulnerable ecosystem. In this study, the fates of bacterial and fungal communities of the typical steppe of the Loess Plateau were investigated under increasing grazing intensities practiced in summer and winter seasons. The results revealed changes in soil physiochemical properties, plant community properties, and microbial diversity in response to alterations in the grazing intensity. The alpha diversity of microbial communities (including bacteria and fungi) exhibited an uneven trend during summer grazing due to an increase in grazing intensity, but it decreased during winter grazing; however, the observed changes were not significant. The beta diversity of the bacterial community was highly influenced by grazing intensity, the summer community clustered near nongrazing, and the winter community presented significantly different results. The beta diversity of the fungal community was not influenced by grazing intensity or season. Grazing induced the growth of fast-growing bacteria (such as Actinobacteria and Firmicutes) and saprophytic fungi and a reduction in overall pathogenic traits. Co-occurrence network analysis and a structural equation model revealed changes in soil and plant properties (such as soil nitrogen level, soil organic carbon level, aboveground biomass, and litter biomass), with an increase in grazing intensity contributing to alterations in bacterial and fungal diversities. This finding demonstrates that grazing intensity can directly affect soil microbes and play an indirect role by modifying soil nutrients and reducing plant biomass, which eventually contributes to changes in microbial communities. Overall, implementing low grazing intensity is suggested for maintaining the microbial community structure the same as that of the native microbiome (ungrazed) in the steppe ecosystems.

Abstract:<p>Livestock grazing often alters aboveground and belowground communities of grasslands and their mediated carbon (C) and nitrogen (N) cycling processes at the local scale. Yet, few have examined whether grazing‐induced changes in soil food webs and their ecosystem functions can be extrapolated to a regional scale. We investigated how large herbivore grazing affects soil micro‐food webs (microbes and nematodes) and ecosystem functions (soil C and N mineralization), using paired grazed and ungrazed plots at 10 locations across the Mongolian Plateau. Our results showed that grazing not only affected plant variables (e.g., biomass and C and N concentrations), but also altered soil substrates (e.g., C and N contents) and soil environment (e.g., soil pH and bulk density). Grazing had strong bottom‐up effects on soil micro‐food webs, leading to more pronounced decreases at higher trophic levels (nematodes) than at lower trophic levels (microbes). Structural equation modelling showed that changes in plant biomass and soil environment dominated grazing effects on microbes, while nematodes were mainly influenced by changes in plant biomass and soil C and N contents; the grazing effects, however, differed greatly among functional groups in the soil micro‐food webs. Grazing reduced soil C and N mineralization rates via changes in plant biomass, soil C and N contents, and soil environment across grasslands on the Mongolian Plateau. Spearman's rank correlation analysis also showed that grazing reduced the correlations between functional groups in soil micro‐food webs and then weakened the correlation between soil micro‐food webs and soil C and N mineralization. These results suggest that changes in soil micro‐food webs resulting from livestock grazing are poor predictors of soil C and N processes at regional scale, and that the relationships between soil food webs and ecosystem functions depend on spatial scales and land‐use changes.</p>

Abstract:Grazing by herbivores plays a key role in grassland ecosystems, but its effects on soil fauna biodiversity are not clear in alpine meadow ecosystems, especially on the Tibetan plateau. We investigated the effects of long-term grazing disturbance of varying intensities (low disturbed site, medially disturbed site, and seriously disturbed site) on ecosystem biodiversity and functional process using soil nematode communities. Soil physicochemical characteristics and plant communities were also measured to explore the relationships between soil nematode communities structure and key environment variable. Soil samples were collected in May, June, August, and October, 2013. The abundance of total nematode communities, plant feeders and bacterivores increased with the grazing intensity increase, and predators decreased with grazing intensity increase. Low disturbed site has the lowest abundance of fungivores. Grazing disturbance has no significant influence on abundance of omnivores (P > 0.05). Species richness was highest on seriously disturbed site and lowest on low disturbed site. MI and SI decreased as grazing intensity increase. The highest NCR was found in low disturbed site and the lowest in medially disturbed site. The highest EI was found in medially disturbed site and the lowest in low disturbed site. Principal component analysis (PCA) showed that nematode communities could be separated by three different grazing disturbance intensity sites. Redundancy analysis (RDA) showed that Helicotylenchus, Acrobeloides and Labronema were positively correlated with higher plant communities diversity. Filenchus was positively correlated with forbs biomass. Some omnivores such as Eudorylaimus and Aporcelaimus was positively correlated with soil moisture and rich nutrient resources, meanwhile, Enchodelus and Coomansus was positively correlated with biomass of sedges and grasses. Anaplectus and Prismatolaimus significantly correlated with soil moisture, nutrient resources and biomass of sedges and grasses. This study revealed that grazing herbivores can change composition, structure and diversity of soil nematode communities, and then reflect radical shift in below-ground soil faunal biodiversity and processes in alpine meadow on the Tibetan plateau.
