Abstract:The aim of this experiment was to investigate the effects of the partial substitution of fish meal with soybean products and chicken meal on the growth performance, antioxidant capacity and intestinal microbiota of Penaeus monodon. A total of 450 healthy, consistent shrimp were randomly divided into five groups, with three replicates per group and 30 shrimp per replicate. The proportion of fish meal substituted with soybean products and chicken meal in the five feed groups was 0% (FM), 40% (40SC), 60% (60SC), 80% (80SC) and 100% (100SC). The experiment lasted for 8 weeks. The results showed that, compared to the FM group, the 40SC and 60SC groups had a decrease in WG and SR, but there was no significant difference (p > 0.05). In contrast, compared to the FM group, the FCR in the 100SC group was significantly increased (p < 0.05), while there was no significant difference among the FM and 40SC, 60SC and 80SC groups (p > 0.05). Compared to the FM group, the ACP in the 80SC and 100SC groups significantly increased (p < 0.05), while the 40SC and 60SC groups had no significant difference (p > 0.05). The AKP in the 100SC group was significantly higher than that in the FM group (p < 0.05), while there were no significant differences among the other four groups (p > 0.05). There were no significant differences in T-AOC and T-SOD among all the treatment groups (p > 0.05). The next-generation sequencing of the intestinal microbiota showed that Proteobacteria was the most abundant phylum in the five groups, accounting for 37.67%, 66%, 40%, 40% and 43.33%, respectively. Compared to the FM group, the Fusobacteriota in the other four groups decreased significantly (p < 0.05). The functional prediction of FAPROTAX indicated that no functional components were observed which are harmful to the body. Considering the effects on growth performance, antioxidant capacity and intestinal microbiota, it is feasible to use soybean products and chicken meal to replace 60% of fish meal in the feed of P. monodon.
A mixed animal and plant protein source replacing fishmeal affects bile acid metabolism and apoptosis in largemouth bass (Micropterus salmoides)

Abstract:Chicken meal, shrimp meal, blood meal, and soybean protein concentrate (SPC) are common alternatives to fishmeal. This study used them to prepare three diets with different levels of fishmeal (FM48, FM40, FM32) for largemouth bass (Micropterus salmoides). The results found no significant difference in the growth performance of largemouth bass fed different diets. Mixed protein increased the total cholesterol (T-CHO) content in plasma, and reduced the total superoxide dismutase (T-SOD) activity in plasma and liver. Targeted metabolomics analysis found that the low fishmeal diets affected the cholesterol and bile acid metabolism of largemouth bass. Mixed protein inhibited cyp7a1 and enhanced hmgcr and pparγ mRNA levels, as well as enhanced the expression levels of FXR in the liver. The fish fed FM32 diet showed inhibited fxr, rxrα and cyp7a1 mRNA levels in the intestine. The results of TUNEL fluorescence staining showed that mixed protein induced apoptosis in largemouth bass. The caspase 3 and caspase 9 mRNA levels in the fish fed FM40 and FM32 diet significantly increased, as well as the expression levels of CASPASE 3. The experiment also found that it could induce oxidative stress and endoplasmic reticulum stress. In conclusion, replacement of fishmeal with mixed animal and plant protein diets did not affect the growth performance, but the health and bile acid metabolism of largemouth bass was affected when the fishmeal level was reduced to 32 %.
Effects of Supplementation Coated Lysine and Methionine in Mixture Protein Diets on Growth Performance, Digestibility and Serum Biochemical Indices of Juvenile Black Sea Bream, Acanthopagrus Schlegelii

Abstract:Four experimental diets were formulated to evaluate the effect of coated lysine (Lys) and methionine (Met) supplemented in mixture protein diets of juvenile black sea bream, Acanthopagrus schlegelii. Fish meal (FM) was the main protein source in control diet, and 21.2% FM was replaced by the mixture of soybean protein concentrate, meat and bone meal, and monosodium glutamate protein in replacement diets with coating Lys and Met were suppled at none (0%, 0%), low (1.7%, 1.1%) and high (5.0%, 3.4%) level (designated as NLM, LLM and HLM diet, respectively). Each diet was fed to triplicate groups of 20 fish (initial mean weight, 4.13 +/- 0.16 g) twice a day to apparent satiation for 8 weeks, in a flow-through system. Results showed that amino acid supplementation significantly improved weight gain and specific growth rate in these mixture protein diets groups, and the fish fed with HLM diet showed comparable growth with the control group. Feed efficiency ratio (FER), protein efficiency ratio (PER), and crude protein and Met content in dorsal muscle were significantly affected by diet amino acid level. The highest apparent digestibility coefficients appeared in HLM group but the values showed insignificant difference with the control group. Serum activities of aspertate aminotransferase and alanine aminotransferase in control group were lower than those of in other treatments. Fish fed with NLM and LLM diets had higher triglycerides concentration than those of fed the HLM and FM diets. Furthermore, the feed cost of the various feed treatments were evaluated, and the three test diets were all lower than the control diet. The present study indicated that 21.2% FM in black sea bream diet can be successfully replaced by mixture protein with sufficient coated Lys (5.0%) and Met (3.4%) supplemented.

Abstract:The effect of fish meal (FM) substitution with fermented soybean meal (FSBM) in the diets of the carnivorous marine fish, black sea bream, Acanthopagrus schlegelii, was investigated. An 8-wk feeding trial was conducted with black sea bream (11.82 +/- 0.32 g; mean initial weight) in indoor flow-through fiberglass tanks (25 fish per tank). Six isonitrogenous and isoenergetic diets were formulated, in which FM was replaced by FSBM at 0% (control diet), 10% (FSBM10), 20% (FSBM20), 30% (FSBM30), 40% (FSBM40), or 50% (FSBM50), respectively. Each diet was fed to triplicate groups of fish twice daily to apparent satiation. The results showed that there was no difference in survival of black sea bream during the feeding trial. Fish fed the FSBM10 or FSBM20 diet showed comparable growth performance compared with fish fed the control diet (P > 0.05), whereas more than 30% replacement of FM adversely affected weight gain and specific growth rate (P < 0.05). Feed intake was significantly lower for fish fed the FSBM50 diet compared with fish fed the control diet. Feed conversion ratio (FCR) tended to increase with increasing dietary FSBM with the poorest FCR observed for fish fed the FSBM50 diet. Protein efficiency ratio and protein productive values showed similar patterns. Apparent digestibility of nutrients significantly decreased with increasing dietary FSBM level. With the exception of protein content, no significant differences in whole body and dorsal muscle composition were observed in fish fed the various diets. Fish fed the FSBM50 diet had significantly lower intraperitoneal ratio than fish fed the control or FSBM10 diet. Hepatosomatic index and condition factor were unaffected by dietary treatments. This study showed that up to 20% of dietary FM protein could be replaced by FSBM protein in the diets of juvenile black sea bream.

Abstract:The present study was aimed to explore the influence of graded replacing fish meal (0.00%, 14.29%, 28.57%, 42.86%, and 57.14%) with spray dried chicken plasma (SDCP) on growth performance, feed utilization, and antioxidant capacity of largemouth bass. Triplicate groups of fish (initial weight: 10.05 ± 0.05 g) were fed experimental diets twice daily for 8 weeks. Results indicated that specific growth rate and feed intake of fish were not remarkably influenced with the replacement of fish meal. Meanwhile, no significant difference was observed in feed intake and feed efficiency ratio among treatments. The inclusion of SDCP significantly elevated crude protein content of muscle, liver and whole fish body, which resulted in the increased protein retention rate. Meanwhile, the lipid retention rate was also significantly increased with the replacement of fish meal. The replacement of 57.14% fish meal with SDCP significantly reduced the content of lipid peroxidation product malondialdehyde. Meanwhile, the total antioxidant capacity and catalase activity was significantly higher in fish fed the diet with replacement of 57.14% fish meal compared to the control group. In above, 57.14% fish meal could be substituted by SDCP in diets for largemouth bass under this experimental condition.

Abstract:Corn gluten meal (CGM), pea protein isolate (PPI) and their mixture (CPP, 1:1 ratio) were evaluated as fishmeal (FM) alternatives in black sea bream (Acanthopagrus schlegelii) juveniles (9.02 +/- 0.12 g). A FM diet was designed as control, and other six diets had 20% and 40% FM protein replaced by CGM, PPI and CPP, respectively, with the supplementation of crystalline methionine, arginine and lysine. After the 8-week feeding trial, significantly lower weight gain was found in fish fed the 40% CGM diet (p < .05), whereas other treatments had no statistical difference (p > .05). The values of feed efficiency ratio and feed intake, as well as proximate compositions of muscle and whole body, were not significantly influenced in all treatments. Apparent digestibility coefficients (ADCs) of dry matter, crude protein, threonine, valine, methionine, isoleucine, leucine, phenylalanine and lysine were significantly influenced by protein source. Fish fed the 40% CGM diet had significantly higher content of essential amino acid in muscle than that of the 20% PPI diet. Fish fed the 40% CGM diet had significantly lower content of serum cholesterol than other treatments (p < .05). Elevated serum superoxide dismutase activity was found in fish fed the 40% plant protein diets. In conclusion, between 20% and 40% FM protein could be replaced by CGM, while about 40% FM protein derived from PPI and CPP could be applied in black sea bream.
Effects of Isoenergetic Diets with Varying Protein and Lipid Levels on the Growth, Feed Utilization, Metabolic Enzymes Activities, Antioxidative Status and Serum Biochemical Parameters of Black Sea Bream (acanthopagrus Schlegelii)

Abstract:Six isoenergetic (20.9 kJ g(-1)) diets were fed to triplicate groups of 20 black sea bream, Acanthopagrus schlegelii (initial weight 11.20 +/- 0.20 g) to apparent satiation for eight weeks. The dietary protein (P) levels (28.85, 31.21, 34.15, 37.46, 40.22 and 42.87%) were combined with lipid (L) levels (26.32, 24.54, 22.73, 20.94, 19.13 and 17.35%), respectively, with the protein: energy (P/E) ratios ranging from 13.71 to 20.60 mg kJ(-1). The weight gain and specific growth rate of the experimental fish increased with increasing dietary P/E ratios up to fish fed the P37/L21 diet (P/E ratio 17.88 mg kJ(-1)), beyond which growth performance decreased (P < .05). Feed intake and feeding rate showed trends similar to the growth performance. Fish fed the P34/L23 diet had the highest values of viscerasomatic index and intraperitoneal fat ratio. The protein efficiency ratio, protein productive value and lipid contents of whole-body and muscle decreased with increasing dietary P/E ratios, whereas whole-body protein content had an inverse trend (P < .05). The fish fed P29/L26 and P31/L32 diets showed the highest contents of serum triglyceride and malondialdehyde, whereas fish fed P40/L19 and P43/L17 diets had the highest activities of liver alanine aminotransferase and aspartate aminotransferase (P < .05). Effective protein sparing effect of lipid occurred in the low protein diets. Based on quadratic regression analysis, the optimal protein requirement of black sea bream is estimated to be 37.10% (dietary energy level 20.9 kJ g(-1)) which is lower than previously reported.

Abstract:Five isonitrogenous and isocaloric diets [containing 54, 30, 15, 10, and 5% fishmeal crude-protein (CP), dry matter (DM) basis] were prepared by replacing fishmeal with poultry by-product meal plus soybean meal to feed juvenile largemouth bass (LMB, with an initial mean body weight of 4.9 g) for 8 weeks. All diets contained 54% CP and 13% lipids. There were four tanks of fish per treatment group (15 fish/tank). The fish were fed twice daily with the same feed intake (g/fish) in all the dietary groups. Results indicated that the inclusion of 15% fishmeal protein in the diet is sufficient for LMB growth. However, some of the fish that were fed diets containing ≤ 15% fishmeal CP had black skin syndrome (characterized by skin darkening and retinal degeneration, as well as intestinal and liver atrophies and structural abnormalities). The concentrations of taurine, methionine, threonine and histidine in serum were reduced (P < 0.05) in fish fed the diets containing 5, 10 and 15% fishmeal CP, compared with the 30 and 54% fishmeal CP diets. Interestingly, the concentrations of tyrosine and tryptophan in serum were higher in fish fed diets with ≤ 15% fishmeal CP than those in the 54% fishmeal CP group. These results indicated that 15% fishmeal CP in the diet containing poultry by-product meal and soybean meal was sufficient for the maximum growth and feed efficiency in LMB but inadequate for their intestinal, skin, eye, and liver health. A reduction in dietary methionine and taurine content and the possible presence of antinutritional factors in the fishmeal replacements diets containing high inclusion levels of soybean meal may contribute to black skin syndrome in LMB. We recommend that the diets of juvenile LMB contain 30% fishmeal CP (DM basis).

Abstract:The potential of three different protein resources (pea protein isolate, PPI; pea protein concentrate, PPC; enzyme treated poultry protein, ETPP) as fish meal (FM) alternative protein in diets for juvenile black sea bream, Acanthopagrus schlegelii. (initial average weight 7.90 +/- 0.13g) was evaluated. Seven isonitrogenous and isoenergetic diets were formulated to replace FM at 0% (T0, control diet), 8% (designated as T1-T3) and 16% (designated as T4-T6) using PPI, PPC and ETPP respectively. Each diet was randomly assigned to triplicate groups of 25 juvenile fish for 8weeks. At the end of the feeding period, survival rate was not significantly affected by dietary treatments. Growth performance in T6 (16% ETPP) group was significantly inferior to T0 group, however, weight gain and specific growth rate in other treatments showed no significant differences (0.05). Mean feed intake, feed efficiency ratio and protein efficiency ratio were also poorer in fish fed in T6 than those of fish fed with the control diet respectively. Apparent digestibility coefficients (ADCs) of dry matter and crude protein for fish fed ETPP diets were significant lower than those of fish fed with the control diet, whereas ADCs of lipid were unaffected by dietary treatments. ADC's of dietary Leu, Ile, His and Lys was also significantly influenced. There were no marked variations in proximate compositions of dorsal muscle. With regard to plasma characteristics, significant difference was observed in triacylglycerol content. Ammonia concentration in plasma tended to increase in alternative protein diets as substitution level increased. There were significant differences in aspartate aminotransferase activities among groups, but alanine aminotransferase levels were unaffected by treatments. In conclusion, the present study demonstrated that PPI and PPC were potential protein sources for using in juvenile black sea bream diet. However, the substitution level of FM by ETPP should be limited within 16%.

Abstract:In China, aquaculture costs have increased because of the increase in fish meal (FM) prices. Plant proteins, such as soybean meal, have the potential to replace FM partially and thus reduce the cost of aquatic feed. In this study, soybean meal (SBM) was fermented using compound microorganisms (lactic acid bacteria, yeast, and Bacillus). Fermented soybean meal (FSBM) replaced FM in the diet. The effect of replacing FM on largemouth bass was comprehensively evaluated at three levels: macro (growth performance), microbial (bacterial diversity and metabolic), and gene (key gene expression) levels. The results showed that FSBM increased the crude protein content by 7.45% and decreased the phytic acid concentration by 48.66% compared with original SBM. Compared with the control, the weight gain rate and feed conversion ratio showed no significant difference (p > 0.05) when the replacement amount of FM was less than 30%. Compared with the 50% FSBM treatment, the 10% and 30% treatments showed more steatosis in the liver slices. Although the replacement of FSBM with less than 30% did not have a significant (p < 0.05) negative impact on body weight, FSBM substitution adversely affected the height and width of the intestinal villi. The expression levels of Hepcidin-1, Hepcidin-2, TGF-β1, and IL-10 in 30% and 50% FSBM treatments were lower than those in the control, whereas the expression levels of SOD1 and SOD2 in 30% FSBM treatment were higher than those in the control group, and the expression level of SOD3a in 30% FSBM treatment was equal to the control. In addition, FSBM substitution could affect the composition of intestinal microorganisms and thus influence metabolic pathways, especially the biosynthesis of amino acids and plant secondary metabolites.

Abstract:An 8-week feeding trial was conducted to determine the effects of imbalanced levels of arginine and lysine in the diets of black sea bream, Acanthopagrus schlegelii, fingerlings. Eight isonitrogenous diets were formulated using a combination of intact protein and crystalline amino acids to contain different arginine/lysine levels: 2.83/3.25, 3.46/4.06, 2.27/4.09, 3.51/2.70, 2.31/2.74, 2.87/4.03, 2.91/4.68 and 2.85/5.24, % dry diet (analysis values), respectively. Each diet was fed to triplicate groups of 25 fish (mean weight, 10.0±0.1g) twice a day to apparent satiation. Fish fed diets with disproportionate levels of arginine and lysine showed significant (P<0.05) growth reduction compared to fish fed the control diet. The lower growth rates observed were due in part to lower feed intake, reduced nutrient availability (arginine and lysine), and increased catabolism of absorbed amino acids. Supplementing additional arginine to the diet was able to partially ameliorate the reduced growth, feed utilization and hepatic arginase activity observed in fish fed with excess dietary lysine, suggesting the possibility that there exists an antagonistic relationship between dietary lysine and arginine in black sea bream. Fish fed diets with excess lysine had lower whole-body protein content compared to fish fed the control diet. The significant increase in plasma ammonia levels as well as enzymes related to amino acid deamination and transamination reactions were found in the present study. To ensure optimal growth of black sea bream, the dietary levels of both arginine and lysine should be considered together when formulating feeds for this commercially farmed marine fish.
Assessment of Left Ventricular Global Twist in Essential Hypertensive Heart by Speckle Tracking Imaging

Abstract:The left ventricular twist was evaluated by 2-dimensional ultrasound speckle-tracking imaging (STI) in 50 patients with hypertension with normal geometric left ventricle (LV) and 45 normal subjects as control group. The mean value of LV rotation was obtained at each plane using STI. LV twist and twist velocity were defined as apical rotation/rotation rate relative to the base respectively. To adjust the intersubject differences in heart rates, the time sequence were normalized. The results showed that peak twist developed near the end of systole. Peak LV twist was significantly higher in patients with hypertension than normal controls (P<0.001). The diastolic untwisting mainly occurred in early diastole (≈38%). Compared with normal controls, untwisting rate (Untw R) in patients with hypertension was significantly reduced (P<0.001), and untwisting half-time (UHT) was significantly delayed (P<0.05). This study demonstrated that STI has a potential ability to evaluate the early change of heart function in patients with hypertension by measuring the twist of LV.

Abstract:The present study was conducted to investigate the effects of dietary inclusion of protein hydrolysates on growth performance, digestive enzyme activities, protein metabolism, and intestinal health in larval largemouth bass (Micropterus salmoides). The experimental feeding trial presented in this study was based on five isonitrogenous and isolipidic diets formulated with graded inclusion levels of protein hydrolysates, and it showed that protein hydrolysates improved growth performance, reduced larval deformity rate, and increased the activity of digestive enzymes, including pepsin and trypsin. Gene expression results revealed that the supplementation of protein hydrolysates upregulated the expression of intestinal amino acid transporters LAT2 and peptide transporter 2 (PepT2), as well as the amino acid transporters LAT1 in muscle. Dietary provision of protein hydrolysates activated the target of rapamycin (TOR) pathway including the up-regulation of TOR and AKT1, and down-regulation of 4EBP1. Additionally, the expression of genes involved in the amino acids response (AAR) pathway, ATF4 and REDD1, were inhibited. Protein hydrolysates inhibited the transcription of some pro-inflammatory cytokines, including IL-8 and 5-LOX, but promoted the expression of anti-inflammatory cytokines TGF-beta and IL-10. The 16S rRNA analysis, using V3-V4 region, indicated that dietary protein hydrolysates supplementation reduced the diversity of the intestine microbial community, increased the enrichment of Plesiomonas and reduced the enrichment of Staphylococcus at the genus level. In summary, protein hydrolysates have been shown to be an active and useful supplement to positively complement other protein sources in the diets for largemouth bass larvae, and this study provided novel insights on the beneficial roles and possible mechanisms of action of dietary protein hydrolysates in improving the overall performance of fish larvae.

Abstract:In order to evaluate the effects of the interaction between different proteins and feeding frequency on largemouth bass (Micropterus salmoides) and to provide scientific guidance for the application of novel proteins and the corresponding optimal feeding strategy, a two-factorial design (5 × 3) with five protein feeds (fishmeal (FM), Clostridium autoethanogenum protein (CAP), Tenebrio molitor (TM), Chlorella meal (ChM), cottonseed protein concentrate (CPC)), and three feeding frequency (1, 2, and 3 times/d; FF1, FF2, FF3) was designed in culturing largemouth bass (initial weight, 2.98 ± 0.22 g/fish) for 8 weeks. Z-score combined with cluster analysis was used to analyze and compare the effects of different treatments on different indicators, such as growth performance, feed utilization, antioxidant capacity, and immune response to draw a general picture of the relationship among all these massive biomarkers. The results showed that different protein sources and feeding frequencies had significant interactive effects on growth performance, feed utilization efficiency, body lipid, and health status of largemouth bass. Fish fed with ChM feed showed similar performance to that in FM group, implying its potential for complete replacement of fishmeal in largemouth bass. Fish fed with CAP, TM, and CPC feeds showed worse performance compared to FM and ChM groups, characterized by poor growth and feed utilization, enhanced stress, chronic inflammation, and varying symptoms of histological changes in the liver and intestine, which demonstrated the adverse effects of the complete replacement of fishmeal by these three proteins. In terms of feeding frequency, fish fed with FM feed in FF3 group led to liver hypertrophy, fat accumulation, and the risk of fatty liver, while inducing liver inflammation. In addition, the TM and CAP group had the higher expression levels of inflammatory factors at FF3 group, which displayed that the interactions between FM, CAP, TM feeds and feeding frequency at FF3 might aggravate the occurrence of liver inflammation and oxidative damage of hepatocytes. Overall, FF2 had higher feed efficiency, protein efficiency, antioxidant enzyme and lysozyme activities, lower MDA content, and lower gene expression of inflammatory cytokines and could be considered as the optimum feeding frequency for largemouth bass fed with different protein feeds.
Excessive Replacement of Fish Meal by Soy Protein Concentrate Resulted in Inhibition of Growth, Nutrient Metabolism, Antioxidant Capacity, Immune Capacity, and Intestinal Development in Juvenile Largemouth Bass (Micropterus salmoides)

Abstract:This study investigated the effects of replacing 0% (SPC0), 25% (SPC25), 50% (SPC50), 75% (SPC75), and 100% (SPC100) of fish meal (FM) with soy protein concentrate (SPC) on the growth, nutritional metabolism, antioxidant capacity, and inflammatory factors in juvenile largemouth bass (Micropterus salmoides) (17.03 ± 0.01 g). After 56 days of culturing, various growth parameters including FW, WGR, and SGR were not significantly different among SPC0, SPC25, and SPC50 groups; however, they were significantly higher than those in SPC75 and SPC100 groups. Conversely, significantly lower FCR were determined for the SPC0, SPC25, and SPC50 groups compared with that for the SPC100 group; specifically, no significant difference among SPC0, SPC25, and SPC50 groups was found. Moreover, compared with SPC75 and SPC100 groups, a significantly higher FI was observed in the SPC0 group, whereas a significantly lower SR was observed in SPC100 compared with that in SPC0 and SPC25 groups. Compared with the SPC0 group, significantly lower mRNA levels of tor, rps6, 4ebp1, pparγ, and fas were found in SPC75 and SPC100. Additionally, the mRNA levels of cpt were significantly higher in SPC0, SPC25, and SPC50 groups than in SPC75 and SPC100 groups. Moreover, the mRNA levels of scd and acc remained unchanged for all the groups. Replacement of FM with SPC did not significantly affect the mRNA levels of gk, pk, and pepck. Compared with the SPC0 group, significantly decreased activities of CAT were observed in the SPC50, SPC75, and SPC100 groups, and significantly decreased activities of GSH-Px were observed in the SPC75 and SPC100 groups. In addition, significantly lower activity of SOD was observed in SPC100 compared with the other groups. Moreover, compared with the other groups, the SPC75 and SPC100 groups had significantly decreased and increased contents of GSH and MDA, respectively, while significantly lower mRNA levels of nrf2, cat, sod, and gsh-px were found in SPC50, SPC75, and SPC100; however, significantly higher mRNA levels of keap1 were observed in SPC75 and SPC100 groups. Additionally, significantly higher mRNA levels of il-8 and nf-κb were found in the SPC50, SPC75, and SPC100 groups compared with the SPC0 group. Conversely, significantly lower mRNA levels of il-10 and significantly higher mRNA levels of tnf-α were found in the SPC75 and SPC100 groups compared with the other groups. Compared with the SPC0 group, mucosal thickness and villus height were significantly decreased in the SPC75 and SPC100 groups. Collectively, SPC replacing 50% FM did not affect its growth of juvenile largemouth bass. However, SPC replacing 50% or more FM might inhibit antioxidant capacity and immune capacity to even threaten the SR, resulting in impaired intestinal development in replacing FM level of 75% or more.
High replacement of fishmeal by Chlorella meal affects intestinal microbiota and the potential metabolic function in largemouth bass (Micropterus salmoides)

Abstract:Microalgae have beneficial effects on the performance of fish as additives and they are becoming a promising alternative to fishmeal as macronutrient ingredients. However, the impact on the fish intestinal microbiome and the function, caused by microalgae as protein sources in diets, remains unclear. This study aimed to determine the composition and potential function of the intestinal microbial community of largemouth bass ( Micropterus salmoides ) fed diets at five replacement levels (0, 25, 50, 75 and 100%) of fishmeal by Chlorella meal in a basal diet (400 g kg −1 ) after 8 weeks. The results showed significant decreases in unique amplicon sequence variants in the intestine at the higher levels of fishmeal replacement. At 50% of fishmeal replacement, dietary inclusions of Chlorella meal had no impact on species richness and Shannon diversity and the community structure of the intestinal microbiota. However, high levels of fishmeal replacement (75 and 100%) significantly induced intestinal community disturbance and diversity loss in largemouth bass. Responding to the high fishmeal replacement level, the dominant genus Cetobacterium and Pleslomonas sharply increased and several taxa from Lactobacillus decreased significantly. Functional data predicted by PICRUSt revealed that nutrition-related metabolism was dominant in the intestinal microbiota of fish fed all the five diets, although some potential functions, particularly amino acid and lipid metabolisms, and energy metabolism, were upregulated firstly, and then downregulated in fish fed diets with the increase of dietary Chlorella meal. Meanwhile, certain pathways were not enriched in intestinal microbiome until up to 75% of fishmeal replacement, such as carbohydrate metabolism, and cofactors and vitamins metabolism. To conclude, this study reveals that fishmeal replacement (50%) by Chlorella meal at the level of 237 g kg −1 in diets is feasible for largemouth bass without impairing the microbiome structure and the metabolism function, providing an alternative strategy for evaluating the possibility of fishmeal replacement by microalgae in aquafeeds.

Abstract:Single-cell proteins are attracting growing attention as viable alternatives for fishmeal (FM) in aquatic feed. Methanotroph (Methylococcus capsulatus, Bath) bacteria meal FeedKind(R) (FK) is a type of single cell protein with high protein content (75.14%) and desirable amino acids profile, produced by Methylococcus capsulatus (Bath) living on methane consumption. The present study evaluated the potential of replacing FM with FK in the diet of black sea bream (Acanthopagrus schlegelii). Five iso-energetic and iso-nitrogenous diets were designed with FK replacing 0, 4.13, 8.27, 16.53, and 24.80% FM protein in the basal diet (40% FM content), respectively. All the diets were fed to three replicates of fish (initial weight 6.56 +/- 0.02 g) for 70 days. After the feeding trial, replacing dietary 8.27% FM protein with FK significantly improved the weight gain and specific growth rate of fish (P < 0.05), while other groups showed no significant difference in the growth performance (P > 0.05). The fish fed diets with 8.27 and 16.53% replacement levels exhibited significantly increased feeding rates. The 8.27% FK diet significantly increased the whole-body and muscle crude protein contents, apparent digestibility of crude lipid, foregut, and midgut amylase activities. The microvillus density in the midgut of fish fed the 24.80% FK diet significantly increased. The diet with 8.27% FK increased the serum triglyceride content of the fish, while the 24.80% FK diet reduced the serum triglyceride, total cholesterol, and low-density lipoprotein cholesterol contents of the fish. In conclusion, the results indicated that replacing dietary FM protein with up to 24.80% FK had no adverse effects on the growth of black sea bream, whilst replacing 8.27% FM protein with FK enhanced its growth performance and feed utilization.

Abstract:The effects of diets fermented with compound probiotics, namely Lactobacillus acidophilus, Limosilactobacillus reuteri and Lactiplantibacillus plantarum, on the growth performance, physiological and biochemical indexes, fatty acid composition and intestinal health of juvenile largemouth bass (Micropterus salmoides) were investigated. Three hundred healthy juvenile M. salmoides (5.29 ± 0.02 g) were selected and randomly divided into two groups with triplicates for each. The basic diet was set as the control group (CON), and fermentation of the basic diet with a mixed bacterial solution (1.8 × 109 cfu/mL, L. acidophilus:L. reuteri:L. plantarum = 1:1:1) was set as the fermentation group (FER). Fish were hand fed to satiation for 56 days and two-thirds of the culture water was renewed every 3 days. The results showed that feed intake of fish in the FER group was significantly lowered, thereby increasing feed efficiency (FE) and protein efficiency (PER) (p < 0.05). Serum alanine aminotransferase (ALT) activity was significantly decreased, and catalase (CAT) activity was significantly increased in the FER group (p < 0.05). The liver superoxide dismutase activity (SOD) was significantly enhanced, and intestinal trypsin was significantly increased in the FER group (p < 0.05). Being fed with the fermentation diet significantly increased the content of n-3 polyunsaturated fatty acids (PUFA), docosahexaenoic acid (DHA) and the n-3/n-6 PUFAs ratios in the liver (p < 0.05). Intestinal histology showed that villus height and width of the intestine and the number of goblet cells were significantly increased in the FER group (p < 0.05). Those fed with fermentation diets had limited diversity of gut microbiota. Compared to the CON group, the relative abundance of Aeromonas decreased significantly (p < 0.05), while the relative abundance of Fusobacteria, Cetobacteria and Lactobacillusis in FER increased greatly in the gut microbiota of the FER group. In conclusion, fermented feed with the three probiotics effectively improved the feed utilization and antioxidant capacity, promoted digestion and absorption of dietary protein, improved the ability of synthesize DHA and n-3 PUFAs in the liver and reduced the abundance of pathogenic bacteria in the gut. Therefore, the present research provided a new way of co-fermented feed with three probiotics for the aquaculture of M. salmoides.

Abstract:This study appraised the impact of enzymatic cottonseed protein concentrate (ECP) as a fish meal (FM) substitute on the growth and health of largemouth bass (Micropterus salmoides) (initial weight 14.99 ± 0.03 g). Five diets with equal nitrogen, fat, and energy were designed to replace 0%, 7.78%, 15.56%, 23.33%, and 31.11% FM by adding 0%, 3.6%, 7.2%, 10.8%, and 14.4% ECP, named ECP0, ECP3.6, ECP7.2, ECP10.8, and ECP14.4, respectively. We fed 300 fish with five experimental diets for 60 days. The results revealed that weight gain rate (WGR) and specific growth rate (SGR) did not notably reduce until the addition of ECP exceeded 7.2%. The proximate composition of fish was not affected by the amount of ECP added in diets. Plasma total protein (TP), albumin (ALB), and high-density lipoprotein (HDL) concentrations increased with the increase of ECP dosage, while the triglyceride (TG) and low-density lipoprotein (LDL) concentrations and alkaline phosphatase (ALP) activity showed an opposite trend. For hepatic antioxidant capacity, the hepatic total superoxide dismutase (T-SOD) and catalase (CAT) activities, glutathione (GSH) content, and the expression of nuclear factor erythroid 2-related factor 2 (Nrf2), superoxide dismutase (SOD), and CAT were increased by ECP, while the hepatic malondialdehyde (MDA) content and the expression of kelch-like-ECH-associated protein 1 (Keap1) were decreased. With regard to inflammation, the expression of nuclear factor-kappa B (NF-κB), interleukin-8 (IL-8), and tumor necrosis factor-α (TNF-α) were inhibited by ECP. In summary, the amount of ECP added to diet can reach 7.2% to replace 15.56% FM without hampering the growth of largemouth bass, and ECP can improve the antioxidant and immune capacity.
