Abstract:Anthurium is an important ornamental crop in the world market and its floral scent can enhance its ornamental value. To date, studies of the components and formation mechanism of the floral scent of Anthurium are relatively few. In this study, the scent profiles of two Anthurium varieties were measured by gas chromatograph-mass spectrometer (GC-MS). There were 32 volatile organic compounds (VOCs) identified in Anthurium 'Mystral', and the most abundant compound was eucalyptol (57.5%). Extremely small amounts of VOCs were detected in Anthurium 'Alabama'. Compared with A. 'Alabama', most genes related to floral scent synthesis exhibited a higher expression in A.'Mystral', including AaDXS, AaDXR, AaMDS, AaHDS, AaTPS, AaDAHPS, AaADT2, AaPAL1, and AaPAL2. In order to produce new varieties of Anthurium with fragrance, 454 progenies of two crossbred combinations of A. 'Mystral' and A. 'Alabama' were obtained. Four F1 generation plants with different floral scent intensities were selected for further study. The major components of floral scent in the progenies were similar to that of the parental A.'Mystral' plant. The expression patterns of genes related to floral scent synthesis were consistent with the relative contents of different types of VOCs. This study revealed the profiles of volatile compounds and associated gene expression in two Anthurium cultivars and their F1 hybrids, which provided a basis for the floral scent inheritance of Anthurium andraeanum.

Abstract:Plant improvement and resolving complex traits of plant need to understand the germplasm diversity and hereditary basis of traits.With the development of genomics and biostatistics software,especially the extensive application of molecular markers in plant genetics and breeding,association analysis based on the linkage disequi-librium (LD) offers a new method for genetic study of plants quantitative characters and a new idea for molecular design breeding.In this paper,the basic statistic principles,strategies,characteristics and current successful appli-cations of association analysis in plants were reviewed.The potential application of association mapping to molec-ular dissection of plant traits and molecular breeding were also discussed.
Genetic Dissection of Leaf-Related Traits Using 156 Chromosomal Segment Substitution Lines

Abstract:A two-line super-hybrid rice (Oryza sativa L.) variety [Liangyoupei9 (LYP9)] demonstrated superiority over its both parents, viz. elite inbred lines 93-11 and Pei-ai64S (PA64S), as well as other conventional hybrids, and had long been exploited in China. However, the genetic basis of its leaf-related traits, supposed to be an important component for yield potential, remains elusive. Here, initially a set of chromosome segment substitution lines (CSSLs) was constructed, in which the genome of Pei-ai64S has been introgressed into the background of 93-11. This set was developed by marker aided selection, based on 123 polymorphic SSR markers. The introgressed chromosomal segments presented in the 156 CSSLs covered 96.46% of Pei-ai64S genome. Afterwards, the CSSLs were deployed to assess the genetic basis of leaf size (length and width) and chlorophyll content of top three leaves across five different environments. The CSSLs showed transgressive segregation for all of the traits, and significant correlations were detected among most of the traits. A total of 27 quantitative trait loci (QTL) were identified on ten chromosomes, and three QTL cluster affecting related traits were found on chromosome 3, 6, and 8, respectively. Remarkably, two key QTLs, qALW3-1 and qALW3-2, both controlling the antepenultimate leaf width, were identified in all five environments, and their effect were further validated by CSSLs harboring the two QTL alleles. Our results indicate that developing CSSLs is a powerful tool for genetic dissection of quantitative traits. Meanwhile, the QTLs controlling leaf-related traits uncovered here provide useful information for marker-assisted selection in improving the performance of leaf morphology and photosynthetic ability.

Abstract:The anemone-type inflorescence of chrysanthemum features prominent, colored tubular florets. Here, the inheritance of a set of traits associated with floral form was investigated using a quantitative trait locus (QTL) mapping approach applied to a segregating population bred from a cross between the two varieties ‘Nannong Xuefeng’ and ‘Monalisa’. The coefficient of variation of the various traits ranged from 12.8 to 59.2%, with an average of 28.0%, and there was notable transgressive segregation in both directions. A set of 26 QTL was uncovered, explaining a portion of the variation in eight of the eleven traits; individually, the loci accounted for between 6.3 and 11.9% of the phenotypic variation. A further 16 pairs of epistatically acting QTL were identified, associated with a contribution of between 3.7 and 13.6% of the phenotypic variance; the majority of these interactions involved background loci. The conclusion was that both additive and epistatic effects were involved in the genetic determination of floral form in chrysanthemum.
SRAP-based Mapping and QTL Detection for Inflorescence-Related Traits in Chrysanthemum (dendranthema Morifolium)

Abstract:Chrysanthemum (Dendranthema morifolium) is an economically important ornamental species and comprises a large proportion of the flower industry in south-east Asian and European countries. In this study, a segregating population of 142 F-1 progeny of the cross between the two chrysanthemum cultivars 'Yuhualuoying' and 'Aoyunhanxiao' was used to construct two separate genetic linkage maps via a double pseudo-testcross mapping strategy. Genotyping was performed using 500 SRAP primer combinations, of which about 50% were informative. This allowed the definition of 896 SRAP loci, of which about 23% showed some segregation distortion. The 'Yuhualuoying' map consisted of 333 testcross markers arranged into 57 linkage groups (LGs). It covered > 1,900 cM with a mean inter-marker distance of 6.9 cM. The map constructed from 'Aoyunhanxiao' comprised 342 test cross markers arranged into 55 LGs. It spanned nearly 1,900 cM, with a mean inter-marker distance of 6.6 cM. The markers were distributed rather uniformly along both maps. A quantitative trait loci analysis was conducted to investigate the pattern of inheritance of three inflorescence traits. This led to the detection of 12 putative loci at a LOD score > 2.5, of which four each specified flower diameter, ray floret layer number, and ray floret length. This study provides molecular mapping information on marker-assisted selection programs for the improvement of multiple traits of interest.

Abstract:Anthurium amnicola Dressler possesses a number of desirable and novel ornamental traits such as a purple-colored upright spathe, profuse flowering, and floral scent, some of which have been introgressed into modern Anthurium cultivars. As a first step in identifying genes associated with these traits, the transcriptome from root, leaf, spathe, and spadix from an accession of A. amnicola was assembled, resulting in 28,019 putative transcripts representing 19,458 unigenes. Genes involved in pigmentation, including those for the metabolism of chlorophyll and the biosynthesis of carotenoids, phenylpropanoids, and flavonoids were identified. The expression levels of one MYB transcription factor was highly correlated with naringenin 3-dioxygenase (F3H) and dihydroflavonol-4-reductase (DFR) in leaves, whereas a bHLH transcription factor was highly correlated with flavonoid 3'-monooxygenase (F3'H) and a DFR in spathes, suggesting that these two transcription factors might regulate flavonoid and anthocyanin synthesis in A. amnicola. Gene sequence and expression data from four major organs of A. amnicola provide novel basal information for understanding the genetic bases of ornamental traits and the determinants and evolution of form and function in the Araceae.
High-density genetic map construction and identification of loci controlling flower-type traits in Chrysanthemum (Chrysanthemum × morifolium Ramat.)

Abstract:Flower type is an important and extremely complicated trait of chrysanthemum. The corolla tube merged degree (CTMD) and the relative number of ray florets (RNRF) are the two key factors affecting chrysanthemum flower type. However, few reports have clarified the inheritance of these two complex traits, which limits directed breeding for flower-type improvement. In this study, 305 F1 hybrids were obtained from two parents with obvious differences in CTMD and RNRF performance. Using specific-locus amplified fragment sequencing (SLAF-seq) technology, we constructed a high-density genetic linkage map with an average map distance of 0.76 cM. Three major QTLs controlling CTMD and four major QTLs underlying RNRF were repeatedly detected in the 2 years. Moreover, the synteny between the genetic map and other Compositae species was investigated, and weak collinearity was observed. In QTL regions with a high degree of genomic collinearity, eight annotated genes were probed in the Helianthus annuus L. and Lactuca sativa L. var. ramosa Hort. genomes. Furthermore, 20 and 11 unigenes were identified via BLAST searches between the SNP markers of the QTL regions and the C. vestitum and C. lavandulifolium transcriptomes, respectively. These results lay a foundation for molecular marker-assisted breeding and candidate gene exploration in chrysanthemum without a reference assembly.
Mapping Single-Locus and Epistatic Quantitative Trait Loci for Plant Architectural Traits in Chrysanthemum

Abstract:Plant architecture is important for chrysanthemum cultivation and breeding. To determine the genetic basis of plant architectural traits in chrysanthemum, a population of 142 F1 plants derived from a cross between the creeping ground-cover chrysanthemum cultivar Yuhualuoying and the erect potted cultivar Aoyunhanxiao was used to detect quantitative trait loci (QTL) associated with plant height, plant width, inter-node length and flower neck length. The broad-sense heritability h B 2 for the four plant architectural traits ranged from 0.33 to 0.83, and transgressive segregation was observed. Single-locus QTL analysis revealed a total of five QTL, accounting for 6.0–16.1% of the phenotypic variation. Additionally, 11 pairs of epistatic QTL were identified, explaining 3.5–14.5% of the phenotypic variations. The majority of the interactions detected occurred between background loci. These results indicate that both additive and epistatic effects contribute to phenotypic variation in the plant architecture of chrysanthemum. It is expected that the identified markers associated with the additive QTL and epistatic QTL detected in this study will be of importance in future breeding programs to develop chrysanthemum cultivars exhibiting desirable plant architecture.

Abstract:Anthurium andreanum is one of the popular flowers in the floriculture industries and the biggest cluster in the Araceae group. Genetic variabilities of Anthurium andreanum were obtained through hybridization and conventional breeding program. The study's objective was to evaluate the morphological character variation of anthurium in the F1 population. The F1 population of anthurium crossing was used for observation and evaluation in this research. The parameter observations were the morphological character of spadix and spathe, quantitative characters comprised of spathe width and length, spadix length, and the total number of flowers per plant in the first flowering stadia. There were 77 plants in the F1 population of crossing between anthurium cv. Sempre x Alvin. The longest spadix was 4.8 cm (SA.18), the longest spathe was 8 cm (SA.19), and the widest spathe was 8.7 cm (SA.19). Average of flowers per plant in the first blooming stadia was about 2-4 flowers. Variation of morphological characteristics was obtained in F1 population evaluation in some parameters. The evaluation showed that most offspring in the F1 population were dominantly inherited from A. Sempre.
Detection of Quantitative Trait Loci for Leaf Traits in Chrysanthemum

Abstract:Summary To identify quantitative trait loci (QTL) controlling leaf traits and to provide some reference for future molecular marker-assisted selection and breeding for leaf traits in chrysanthemum, QTL analyses were conducted for three leaf traits: leaf length (LL), leaf width (LW), and the ratio of leaf length to width (LL:LW). Phenotypic data were collected over 2 years in a segregating F1 population of 142 hybrids of ‘Yuhualuoying’ × ‘Aoyunhanxiao’ and analysed using previously published sequence-related amplified polymorphism (SRAP)-based maps. A total of 12 additive QTLs were identified for the three leaf traits. Each QTL accounted for between 5.8% and 13.4% of the observed phenotypic variation. Seven QTLs were detected at the same genomic regions in both years, indicating that they were stable over different environments. The tight causal linkages between the different QTLs controlling LL and LW on linkage group Y14 of the ‘Yuhualuoying’ map and on linkage group A13 of the ‘Aoyunhanxiao’ map may account for the significant phenotypic correlation between the two leaf traits. In addition, three epistatic QTLs for LL and LW accounted for 2.1 – 11.8% of the phenotypic variation in these traits. The results obtained in the present study add new knowledge and a deeper understanding of the genetic basis of leaf traits in chrysanthemum, and may be valuable in future breeding programmes to develop chrysanthemum cultivars that exhibit more desirable leaf traits.
Genetic characterization of anemone‐type chrysanthemum (Chrysanthemum morifolium) using floral morphology and SRAP markers

Abstract:An appreciation of the genetic diversity retained in the gene pool of anemone-type chrysanthemums is needed to drive further genetic improvement of this commercially important ornamental species. Here, a panel of 67 accessions was characterized with respect to both floral morphology, assessed using a set of 12 inflorescence-associated traits, and to genotype, using sequence-related amplified polymorphism (SRAP) fingerprinting. Abundant variation and a high level of broad-sense heritability (>88%) were revealed for all traits except number of tubular floret lobes. Of the possible 66 pairwise inter-trait combinations, 37 proved to be significantly correlated. A principal component analysis of the trait data showed that the first four principal components explained similar to 78% of the variance; meanwhile a hierarchical clustering analysis revealed a number of discrete groups reflecting the various cultivated types, but was not predictive of provenance. The SRAP genotyping employed 22 primer combinations and generated 238 informative fragments. A moderately high level of the Jaccard's similarity coefficient (mean 0.67) was calculated for the examined panel. A hierarchical clustering analysis based on the genotypic data revealed four groups, which were clearly reflective of cultivated type; moreover, a comparison of the within-cluster means for each of the two key anemone-related traits-tubular floret length and tubular floret length/style length-showed that the genotype-based clusterings were associated with the anemone levels, albeit imperfectively. A principal coordinate analysis showed that the first two coordinates accounted for about 14% of the total variation. In summary, the present study adds new understanding of the genetic diversity present in the anemone-type chrysanthemum gene pool, and the findings will lay the foundation for implementing a genotype/phenotype association strategy to reveal the genetic and molecular basis of floral form in this valuable ornamental species.
Improved Transformation of Anthurium

Abstract:Methods to increase transformation efficiency and yields of transgenic Anthurium andraeanum Linden ex. André hybrids were sought while effecting gene transfer for resistance to the two most important pests, bacterial blight ( Xanthomonas axonopodis pv. dieffenbachiae ) and nematodes ( Radopholus similis and Meloidogyne javanica ). Differentiated explant tissues, embryogenic calli, and comingled mixtures of the two were transformed with binary DNA plasmid constructs that contained a neomycin phosphotransferase II ( npt II) selection gene with a nos promoter and terminator. Explants included ≈1-cm long laminae, petioles, internodes, nodes, and root sections from light- and dark-grown in vitro plants. Bacterial blight resistance genes were NPR1 from Arabidopsis , attacin from Hyalophora cecropia , and T4 lysozyme from the T4 bacteriophage. For nematode resistance, rice cystatin and cowpea trypsin inhibitor genes were used. Cocultivation with Agrobacterium tumefaciens strains EHA105, AGLØ, and LBA4404 ranged from 2 to 14 days. Over 700 independent, putatively transformed lines were selected with 5 and 20 mg·L −1 geneticin (G418) for cultivars Midori and Marian Seefurth, respectively. Putative transgenic lines were selected 1 to 11.5 months, but on average 5.2 to 8.4 months, after cocultivation depending on the tissue type transformed. Significantly more embryogenic calli (one line per 5 mg calli) produced transgenic lines than did explants (one line per 143 mg explants) ( P < 0.004) from ≈30 mg of tissue. Calli grew selectively from all explant types, but the type of explant from which each selection was made was not recorded because root, internode, and petiole explants were difficult to discern by the time calli developed. Shoots formed 3 months after calli were transferred to light. Non-transgenic control and transgenic ‘Marian Seefurth’ formed flower buds in the greenhouse ≈28 months after cocultivation. The plants resembled commercially grown plants from a private nursery. No non-transformed escapes were detected among the selections screened for NPTII by enzyme-linked immunosorbent assay and polymerase chain reaction (PCR). The selections were positive for transgenes as assayed by PCR and Southern hybridizations. Southern blots showed single-copy insertions of the NPR1 regulatory gene. The ability to produce large quantities of independent transgenic lines from embryogenic calli in a relatively short time period should enable researchers to evaluate the effectiveness of any transgene by screening numerous anthurium lines for improved performance.

Abstract:Abstract Phalaenopsis spp. represent the most popular orchids worldwide. Both P. equestris and P. aphrodite are the two important breeding parents with the whole genome sequence available. However, marker–trait association is rarely used for floral traits in Phalaenopsis breeding. Here, we analyzed markers associated with aesthetic traits of Phalaenopsis orchids by using genome-wide association study (GWAS) with the F1 population P. Intermedia of 117 progenies derived from the cross between P. aphrodite and P. equestris . A total of 113,517 single nucleotide polymorphisms (SNPs) were identified in P. Intermedia by using genotyping-by-sequencing with the combination of two different restriction enzyme pairs, Hinp1 I/ Hae III and Apek I/ Hae III. The size-related traits from flowers were negatively related to the color-related traits. The 1191 SNPs from Hinp1 I/ Hae III and 23 simple sequence repeats were used to establish a high-density genetic map of 19 homolog groups for P. equestris . In addition, 10 quantitative trait loci were highly associated with four color-related traits on chromosomes 2, 5 and 9. According to the sequence within the linkage disequilibrium regions, 35 candidate genes were identified and related to anthocyanin biosynthesis. In conclusion, we performed marker-assisted gene identification of aesthetic traits with GWAS in Phalaenopsis orchids.

Abstract:Chrysanthemum is one of the most leading commercial flower crops well-known for its flower shape diversity. Anemone type chrysanthemums receive increasing market attention that are distinguished by their pigmented and elongated disk florets. However, there are few reports on quantitative trait loci (QTL) mapping and the identification genes for anemone in chrysanthemum. In this study, bulked segregant analysis coupled with whole-genome sequencing (BSA-seq) approach was employed to detect candidate genomic regions controlling anemone type flower. Two mixed DNA pools composed of 28 anemone type and 20 non-anemone type F1 lines derived from a cross between 'Nannong Xuefeng' and 'Monalisa' together with two parents were used for highthroughput sequencing. A total of 42 consistent QTLs covering 30.00 Mb were simultaneously identified by |o(SNP/InDel-index)| and Euclidean distance (ED) algorithms based on 15,783,313 SNPs (single-nucleotide polymorphisms) and 1,546,200 small InDels (insertions and deletions). Twenty-two key candidate genes were screened out by a combination of gene functional annotation and transcriptome analysis, in which evm.model. scaffold_915.436 (CBF5), evm.model.scaffold_24.208 (U2AF65A), and a uncharacterized gene evm.model.scaffold_260.201 were highlighted by qRT-PCR. Furtherly, four associations were successfully converted to KASP (kompetitive allele specific PCR) markers, among them, the KASP markers developed for SNP locus Chr6__33961007 and Chr6__179207697 were proven to efficiently discriminate anemone and non-anemone type chrysanthemums being a couple of diagnostic markers in two populations. The results presented herein provide insights for further research on elucidating the formation mechanism of anemone type flower, and the anemonespecific KASP marker will be highly useful for applying MAS to accelerate breeding improvement of chrysanthemum.

Abstract:Chrysanthemum (Chrysanthemum morifolium Ramat.), one of the four most popular cut flowers in the world, occupies a large share of global floriculture trade. Leaf angle (LA) is an important factor affecting plant architecture, photosynthesis and yields of cut chrysanthemum. However, the genetic basis and related genes underlying LA remain largely unknown. In this research, phenotypes of five LA-related traits in 200 cut chrysanthemum accessions were investigated across two consecutive years (Y2020 and Y2021), and their broadsense genetic heritabilities ranged from 68.91% to 84.70%. Leaf orientation value (LOV) exhibited the maximum phenotypic coefficient of variation of 35.66% in Y2020 and 32.34% in Y2021. A new methodological framework named 3VmrMLM was applied for multi-locus genome-wide association study (GWAS) based on a set of 330,710 single nucleotide polymorphisms (SNPs) to explore the genetic loci and candidate genes of LA. As a result, 303 quantitative trait nucleotides (QTNs) in single-environment, 120 QTNs and 75 QTN x environments interactions (QEIs) in multiple-environments were identified. Among the genes around these QTNs and QEIs, 26 known genes were considered to be associated with LA, including ARF16, IBH1, AN3, etc. Furthermore, the haplotype analysis of ARF16 revealed four accessions with elite LA. Our findings provide a fundamental understanding to the genetic architecture of LA and contribute to further gene cloning and molecular improvement of chrysanthemum plant architecture.
Identification of key genes responsible for green and white colored spathes in Anthurium andraeanum (Hort.)

Abstract:Modern anthuriums, Anthurium andraeanum (Hort.) are among the most popular flowering plants and widely used for interior decoration. Their popularity is largely attributed to the exotic spathes with different colors. Previous studies have reported color development in red spathe cultivars, but limited information is available on key genes regulating white and green colored spathes. This study analyzed anthocyanin, chlorophyll, and carotenoid contents as well as transcript differences in spathes of eight cultivars that differed in spathe colors ranging from red to white and green. Results showed that increased expression of a transcription factor AaMYB2 was associated with elevated levels of anthocyanin in spathes, but decreased expression of AaMYB2 and increased expression of AaLAR (leucoanthocyanidin reductase) and AaANR (anthocyanidin reductase) were accompanied with the accumulation of colorless proanthocyanidin, thus the white spathe. As to the green colored spathe, chlorophyll content in the green spathe cultivar was substantially higher than the other cultivars. Correspondingly, transcripts of chlorophyll biosynthesis-related genes AaHemB (porphobilinogen synthase) and AaPor (protochlorophyllide oxidoreductase) were highly upregulated but almost undetectable in white and red spathes. The increased expression of AaHemB and AaPor was correlated with the expression of transcription factor AaMYB124 . Subsequently, qRT-PCR analysis confirmed their expression levels in nine additional cultivars with red, white, and green spathes. A working model for the formation of white and green spathes was proposed. White colored spathes are likely due to the decreased expression of AaMYB2 which results in increased expression of AaLAR and AaANR , and the green spathes are attributed to AaMYB124 enhanced expression of AaHemB and AaPor . Further research is warranted to test this working model.

Abstract:A genetic linkage map comprised of 131 loci was constructed with a F 2 population derived from inter-subspecific cross between Brassica campestris L. ssp. chinensis variety 'aijiaohuang' and B. Campestris L. ssp. rapifera variety 'qisihai', which covered 1 810.9cM in 12 linkage groups. The interval mapping was used to determine QTLs associated with eight morphological traits in Chinese cabbage. 24 QTLs affecting eight traits of shoots were identified using Mapmaker/QTL1.1 software. The QTLs was associated with 8.9%- 86.5% of the phenotypic variation in different traits. Two QTLs were identified for leaf length and leaf number respectively; three QTLs for leaf width and plant height, four QTLs for number of leaf lobes and petiole width, five QTLs for petiole length.

Abstract:MicroRNAs (miRNAs) are known to play vital roles in coloration of leaves, flowers, and fruits in plants. However, their functions in spathe coloration are poorly known. Anthurium andraeanum is a popular ornamental plant with various spathe colors. In this study, small RNA and degradome libraries from three A. andraeanum cultivars with different-colored spathes were constructed and sequenced. Illumina sequencing resulted in 94 conserved miRNAs, and 34 novel miRNAs in total were then identified based on precursor sequences and hairpin structures. Differential expression analysis showed that 52, 51, and 49 miRNAs were differentially expressed in comparisons of orange- versus white-colored spathe, purple- versus white-colored spathe, and purple- versus orange-colored spathe, respectively. The expression patterns of miRNAs and their corresponding targets involved in spathe coloration were further analyzed, and displayed that miR156b and miR529 were highly abundant in the spathes with higher anthocyanin content. These two miRNAs co-targeted a gene encoding SPL17 , which may function as a negative regulator in anthocyanin accumulation. In addition, miR408 was also abundantly expressed in purple- and orange-colored spathes, and its typical targets were also identified. This comprehensive integrated analysis provides insight into the miRNA-mediated genetic regulation in spathe coloration of A. andraeanum .

Abstract:Flowering time is an important trait in chrysanthemum, but its genetic basis remains poorly understood. An intra-specific mapping population bred from the cross between the autumn-flowering cultivar 'Yuhualuoying' and the summer-flowering 'Aoyunhanxiao' was used to determine the number and relative effect of QTL segregating for five measures of flowering time. From flowering time data recorded over two consecutive seasons, 35 additive QTL were detected, each explaining between 5.8% and 22.7% of the overall phenotypic variance. Of these, 13 were detected in both years. Nine genomic regions harboring QTL for at least two of the five traits were identified. Ten pairs of loci epistatically determined the flowering time, but their contribution to the overall phenotypic variance was less than for the additive QTL. The results suggest that flowering time in chrysanthemum is principally governed by main effect QTL but that epistasis also contributes to the genetic architecture of the trait, and the major QTL identified herein are useful in our ongoing efforts to streamline the improvement of chrysanthemum via the use of molecular methodology.
A Preliminary Genetic Linkage Map of Chrysanthemum (chrysanthemum Morifolium) Cultivars Using Rapd, Issr and Aflp Markers

Abstract:A genetic linkage map of chrysanthemum (Chrysanthemum morifolium) was constructed by genotyping 142 F1 progeny of the bi-parental cross ‘Yuhualuoying’×‘Aoyunhanxiao’ with a combination of RAPD, ISSR and AFLP markers in a double pseudo-testcross mapping strategy. A total of 567 polymorphic markers, including 153 RAPDs, 61 ISSRs and 353 AFLPs, were used in linkage mapping. 336 of 567 (60%) markers were grouped on the two parental maps, leaving 231 (40%) markers unlinked. In the ‘Yuhualuoying’ linkage map, 210 markers including 116 testcross and 94 intercross markers were placed in 12 major and 32 minor (8 triplets and 24 doublets) linkage groups, covering 1034cM with an average map distance of 6.2cM between adjacent markers. In ‘Aoyunhanxiao’ linkage map, 190 markers consisting of 113 testcross and 77 intercross markers were resolved into 9 major and 24 minor linkage groups, with genome coverage of 1095cM and a mean inter-marker separation of 6.9cM between adjacent markers. Six pairs of homologous linkage groups were established on the basis of 64 intercross markers shared by the two parental maps. The maps lay a foundation for further quantitative traits loci (QTL) mapping and marker-assisted breeding of chrysanthemum.
