Abstract:The objective of the present study was to improve the stability of (water-in-oil-in-water) double emulsions by dual gelation, including the gelation of lipid phase through oleogels and the gelation of continuous aqueous phase through hydrogels. Glycerol monostearate (GMS) could be used to form the oleogels and improve the stability of primary emulsions and double emulsions through reinforcing oil film. However, the redundant GMS crystals in both the oil phase and the continuous phase could cause the destabilization of double emulsions. The double emulsions reinforced by oleogels were successfully encapsulated in Ca-alginate hydrogel beads, and this encapsulation would not influence the integrity of double emulsions. The redundant GMS crystals might cause the leakage of the inner aqueous phase in the encapsulation process, and this leakage was more likely to happen during the mixing step, rather than the crosslinking step. Ca-alginate hydrogel beads were conducive to the confinement of the encapsulated oil globules.
Modulating in vitro digestion of whey protein cold-set emulsion gels via gel properties modification with gallic acid and EGCG

Abstract:Gallic acid (GA) and epigallocatechin gallate (EGCG), cooperated at varied ratios (1:0, 3:1, 1:1, 1:3, and 0:1), were employed to modify gel properties of calcium induced-whey protein emulsion gel. The effects of GA/EGCG on emulsion morphology, as well as gel properties and in vitro digestive behavior of the emulsion gels were investigated. Compared with emulsions without phenolics, GA/EGCG induced slightly smaller particle size and stronger electrostatic repulsion between emulsion droplets. Moreover, GA/EGCG, notably at a ratio of 3:1, promoted electrostatic and hydrophobic interactions between protein molecules and the formation of a compact and filamentous gel microstructure, resulting in a remarkable increment in the gel strength (up to 106 %). Furthermore, in vitro oral digestion, dynamic gastric digestion (using an artificial gastric digestive system, AGDS), and intestinal digestion of the emulsion gels were simulated. Particle size and protein hydrolysis results revealed that GA/EGCG was prone to weaken the physical disintegration of gels, reduce protein hydrolysis, and enhance the stability of emulsified oil droplets during dynamic gastric digestion. As a consequence, delayed release of oil droplets was observed in the gels and more free fatty acids were released in the intestinal digestion, particularly in the gel with GA/EGCG (3:1). These findings would provide novel strategies for application of phenolic compounds in developing protein gel-based delivery systems.

Abstract:Based on the single factor experiment,uniform design and orthogonal experimental design were adopted to select the optimal formula and optimize the preparation technics.The stability of microemulsion was evaluated via particle diameter,the conductivity at different temperatures,the range of dilution in water and electrolyte solution and the centrifugal stability.After these trials,oil/water(O/W) diacylglycerol microemulsion was prepared successfully,with the content of DAG at 25%,and the particle diameter at 95.8 nm.Besides,the microemulsion has a good stability at 80℃,a wide water dilution range and its physical structure insusceptible to the common electrolyte in food systems.
In vitro digestion behavior of water-in-oil-in-water emulsions with gelled oil-water inner phases

Abstract:Double emulsions may be able to protect and release in a controlled manner bioactive compounds during digestion of food matrices. It was hypothesized that the physical state and solid content in the inner phases of water-in-oil-in-water (W1/O/W2) emulsions may affect the overall stability and the release behavior of bioactives during in vitro digestion. Therefore, hydrophobic (phytosterols or Vitamin D3) and hydrophilic (Vitamin B12) molecules were incorporated in double emulsions prepared either with a liquid (soybean oil - SO) or oil-fat gel (soybean oil+trimyristin - STO) lipid phase and liquid internal aqueous phase. In addition, the impact of a gelled inner aqueous phase was studied, using high methoxyl pectin. W1/O/W2 emulsions were prepared with polyglycerol polyricinoleate (PGPR) and sodium caseinate as emulsifiers. After the 30min in vitro gastric stage, all double emulsions showed no significant change in size. Lipid crystals were visible in the STO emulsions. Fat crystallization, and the formation of an oil fat gel, led to coalescence of the inner aqueous droplets. The inner aqueous droplets were no longer visible by confocal microscopy after the initial stages of 2h in vitro duodenal digestion. Fat crystals and droplets of non-spherical shape were also noted in the STO double emulsions up to 25min of in vitro duodenal stage. Overall, the STO emulsions had a higher extent of free fatty acid release and consequent bioactive transfer compared to the SO emulsions. The presence of the medium chain fatty acids (from trimyristin), in addition to the surface-to-core distribution of the hydrophobic bioactives within the oil droplet were key factors in lipid digestibility and bioactive release. The STO and SO samples did not differ in terms of the release of the hydrophilic molecule, vitamin B12, over time. On the other hand, there was a significant increase in the stability of the inner water phase, after gastric digestion, when this phase was gelled with high methoxyl pectin. This work demonstrated that the physical properties of the different internal phases of W1/O/W2 influenced lipid digestion and bioactive transfer kinetics during in vitro digestion.

Abstract:Distearin (DS) can be used as an emulsifier, due to its surface activity derived from the amphiphilic nature of the molecule, moreover, it can also crystallize and form a 3D crystal network that can induce oil gelation. This research aimed to examine the ability to combine both emulsifying and oil gelation properties to structure and stabilize water-in-oil emulsion gel systems. Different water contents and DS concentrations produce emulsion gels with different textural attributes while incorporating up to 30% of water in a 15% wt. DS-based oleogel resulted in stable white gels. Microscopy imaging confirmed the formation of a water-in-oleogel type emulsion gel characterized by DS crystallization in the continuous phase and at the interface through Pickering mechanism. A positive relation was observed between the G' and hardness values and water content, suggesting gel strengthening resulted from interactions between the DS crystals at the interface and the continuous phase, as suggested by the active filler theory. Thermal analysis revealed two broad melting events at the temperature range of 42.2-44.9 °C and 55.9-58.6 °C for emulsion gels with 10-30% water content, suggesting initial melting of β' polymorph and transition to β during melting, which was confirmed by XRD. The results showed that homogenization significantly improved the oil retention of the gels due to increased crystal surface area, while water addition slightly reduced it. Compared with traditional emulsions or oleogels, this water-in-oil gel system demonstrated prolonged stability and enhanced mechanical properties due to the dual functionality of DS at the water/oil interface and bulk.

Abstract:Double emulsions have very promising application prospects in the pharmaceutical, cosmetic, and food industries. Nevertheless, its usage in the food industry is restricted mainly due to its immense stability problems. Here, W1/O/W2 double emulsion was prepared by two-step emulsification method using ultrasonic emulsification technology. The physicochemical stability, rheological properties, thermal stability, and microstructure of the double emulsion were investigated. The results revealed that W1/O/W2 double emulsion with optimum stability was prepared on the parameters that the ratio of W1 (gelatin)/O primary emulsion to the external aqueous phase (gelatin-EGCG-high methoxyl pectin ternary complex) was 6:4, ultrasonic power was 80 W, ultrasonic time was 6 min, and ternary complex concentration was 0.75%. The double emulsion produced under these conditions had the smallest particle size (1,143 nm) and the maximum absolute zeta potential value (39.17 ± 0.50 mV). The results of full-featured stability analyzer manifested that the emulsion system no longer underwent phase separation. Confocal laser scanning microscopy (CLSM) successfully observed a uniformly dispersed double emulsion. Furthermore, dynamic rheological properties demonstrated that the cohesive strength of W1/O/W2 double emulsion was strength than that of W1/O primary emulsion. Differential scanning calorimetry (DSC) analysis suggested that both remained stable throughout the temperature change, while the W1/O/W2 double emulsion was more thermally stable. Hence, the gelatin-EGCG-high methoxyl pectin ternary complex can act as an effective emulsion stabilizer to stabilize the W1/O/W2 double emulsion.

Abstract:Emulsion micro-gels exhibit significant potential as functional ingredients for modifying food texture, replacing saturated fats, or serving as templates for the controlled release of bioactive compounds. Structural design principles are being applied more frequently to develop innovative emulsion micro-gels. In this paper, whey protein concentrate (WPC), κ-carrageenan and sodium alginate (SA) were utilized for preparing emulsion micro-gels. To reveal the regulation mechanism of the structural and physicochemical properties of emulsion micro-gels on lipid digestion, the influence of SA additions on the structural, physicochemical properties and in vitro digestion behavior of κ-carrageenan/WPC-based emulsion micro-gel were explored. The FTIR results suggest that the emulsion micro-gels are formed through non-covalent interactions. With the increase of SA addition (from 0.7 g/100 mL to 1.0 g/100 mL), the decreased mean droplet size, the increased hardness, elasticity indexes, and water holding capacity, the reduced the related peak times all indicated that the emulsion micro-gels exhibit enhanced rheological, stability, and mechanical properties. It can be concluded from the microstructure, particle size distribution of the emulsion micro-gels during simulated digestion and free fatty acid release that both κ -carrageenan/WPC-based emulsion micro-gel and κ -carrageenan/WPC/SA-based emulsion micro-gel can inhibit lipid digestion due to the ability to maintain structural stability and hindering the penetration of bile salts and lipase through the hydrogel networks. And the ability is regulated by the binding properties the gel matrix and oil droplets, which determine the structure and physicochemical properties of emulsion micro-gels. The research suggested that the structure of emulsion micro-gels can be modified to produce various lipid digestion profiles. It may be significant for certain practical application in the design of low-fat food and controlled release of bioactive agents.

Endothelial cells exhibiting angiogenesis in vitro proliferate in response to TGF‐β1

Abstract:Transforming growth factor‐β1 (TGF‐β1) has been implicated in the positive regulation of angiogenesis in vivo, whereas it inhibits the proliferation of endothelial cells in vitro. To reconcile these apparently contradictory effects, we have investigated the effect of TGF‐β1 on bovine aortic endothelial cells that exhibit spontaneous angiogenesis in vitro. We show that concentrations of TGF‐β1 which stimulate proliferation of cells that form endothelial cords and/or tubes inhibit proliferation of the same cells grown at subconfluent densities. An increase in cell number of 35% over control cultures was achieved with 0.5 ng TGF‐β1/ml. The proliferative effect was blocked by antibodies against TGF‐β. Immunological detection of BrdU‐labeled nuclei revealed an increase greater than 220% in cells treated with TGF‐β1. Moreover, a population of cells within the cords appeared to be a selective target for this cytokine. The stimulatory effect was not restricted to bovine aortic endothelial cells, as similar results were obtained with endothelial cells derived from rat microvessels. Significant levels of active TGF‐β1 were detected in cultures containing cords/tubes, whereas only latent TGF‐β1 was detected in subconfluent cultures. We show further that endothelial cells exhibiting angiogenesis in vitro secrete plasminogen activator, an enzyme that regulates activation of TGF‐β. The major increases in mRNA transcripts for extracellular matrix proteins that are typically associated with TGF‐β1 were not seen in cells exhibiting angiogenesis in vitro. Since the formation of tubular networks requires both invasion and proliferation, we propose that TGF‐β1 is a major morphoregulatory factor in angiogenesis that specifically controls endothelial cell proliferation and extracellular matrix turnover.

Abstract:W/O/W emulsions can be used to encapsulate both hydrophobic and hydrophilic bioactive as nutritional products. However, studies on protein stabilized gel-like W/O/W emulsions have rarely been reported, compared to the liquid state multiple emulsions. The purpose of this study was to investigate the effect of different oil–water ratios on the stability of W/O/W emulgels fabricated with salt-soluble proteins (SSPs) of Sipunculus nudus. The physical stability, structural characteristics, rheological properties, and encapsulation stability of vitamin C and β-carotene of double emulgels were investigated. The addition of W/O primary emulsion was determined to be 10% after the characterization of the morphology of double emulsion. The results of microstructure and rheological properties showed that the stability of W/O/W emulgels increased with the increasing concentration of SSPs. Additionally, the encapsulation efficiency of vitamin C and β-carotene were more than 87%, and 99%, respectively, and still could maintain around 50% retention of the antioxidant capacity after storage for 28 days at 4 °C. The aforementioned findings demonstrate that stable W/O/W emulgels are a viable option for active ingredients with an improvement in shelf stability and protection of functional activity.

Abstract:In this study, the effect of Torreya grandis protein enzymatic hydrolysates (TGPH)/alginate dialdehyde (ADA) complexes in the internal aqueous phase on the physical stability of the water-in-oil-in-water (W 1 /O/W 2 ) emulsions was studied. In the case of TGPH/ADA emulsions, the presence of ADA decreased the apparent viscosity of the emulsions and changed the flow behavior from shear thinning to Newtonian, leading to a decrease in volume-weighted average droplet diameter (D 43 ) of the emulsions. Additionally, the emulsions at the TGPH/ADA ratios of 1:1 showed a lower turbiscan stability index (TSI) value, and smaller change in delta backscattering signal, compared to the emulsions. The enhanced pH stability and storage stability of the emulsions at the TGPH/ADA ratios of 1:1 was due to the formation of Schiff bases between TGPH and ADA. These results suggested that the covalent cross-linking of TGPH with ADA could significantly improve the stability of the emulsions, which provided an effective means for the development of new food-grade protein-polysaccharide complexes stabilized emulsions.

Effect of diacylglycerol on partial coalescence of aerated emulsions: Fat crystal-membrane interaction and air-liquid Interface interaction insights

Abstract:Currently, the poor whipping capabilities of anhydrous milk fat (AMF) in aerated emulsion products are a major obstacle for their use in beverages like tea and coffee, as well as in cakes and desserts, presenting fresh hurdles for the food industry. In this study, the mechanism of action of diacylglycerols (DAGs) with different carbon chain lengths and degrees of saturation on the partial coalescence of aerated emulsions was systematically investigated from three fundamental perspectives: fat crystallization, air-liquid interface rheology, and fat globule interface properties. The optimized crystallization of long carbon chain length diacylglycerol (LCD) based on stearate enhances interactions between fat globules at the air-liquid interface (with an elastic modulus E' reaching 246.42 mN/m), leading to a significantly reduced interface membrane strength. This promotes fat crystal-membrane interactions during whipping, resulting in a thermally stable foam structure with excellent shaping capability due to enhanced partial coalescence of fat globules. Although Laurate based medium carbon chain length diacylglycerol (MCD) promoted fat crystallization and optimized interface properties, it showed weaker foam properties because it did not adequately encapsulate air bubbles during whipping. Conversely, oleate long carbon chain length diacylglycerol (OCD) proved to be ineffective in facilitating fat crystal-membrane interaction, causing foam to have a subpar appearance. Hence, drawing from the carefully examined fat crystal-membrane interaction findings, a proposed mechanism sheds light on how DAGs impact the whipping abilities of aerated emulsions. This mechanism serves as a blueprint for creating aerated emulsions with superior whipping capabilities and foam systems that are resistant to heat.

Abstract:Despite the success of transplantation, many transplant candidates and transplant recipients die each year. Some die awaiting transplants and some die months or years after receiving an organ. Quality end-of-life care can play a valuable role in easing the impact of death and dying in transplantation, as it focuses on enhancing patients' quality of life near death. Quality end-of-life care recognizes the values and preferences of patients and their families, and involves a process of shared decision making about patients' healthcare treatment in collaboration with healthcare practitioners. Advance care planning involves discussions with patients about their wishes and values about care, in the event that the patient becomes incapable of making such decisions. This article focuses on the application to transplantation of quality end-of-life care and advance care planning and identifies the effects that death and dying of transplant patients have on others. The information herein encourages healthcare practitioners to view and deliver quality end-of-life care as part of transplant patients' overall treatment management.

Abstract:Water-in-oil-in-water (W/O/W) Pickering double emulsions are promising materials for the construction of carriers for water-soluble and oil-soluble molecules or drug delivery systems if the contradictive trade-off between their extreme stability and controlled release properties can be resolved. In this study, biodegradable and biocompatible poly(ethylene glycol)-b-poly(ε-caprolactone-co-δ-valerolactone) (PEG-b-PCVL) diblock copolymers with predesigned hydrophilic to hydrophobic block length ratios and nearly identical ε-caprolactone/δ-valerolactone molar ratio (8/2), were synthesized by ring-opening copolymerization. Then, they self-assembled to create semicrystalline micelles. The melting points of PEG-b-PCVL copolymers and their lyophilized micelles were within a physiological range of temperatures, as determined by differential scanning calorimetry. Water contact angle measurements provided evidence that the surface wettability of PEG-b-PCVL micelles could be tuned by the PCVL block mass fractions or temperature stimulus. Such PEG-b-PCVL micelles were employed as a single particulate stabilizer to develop Pickering double emulsions through a one-step emulsification technique. W/O/W Pickering double emulsions could be generated using relatively hydrophobic PEG-b-PCVL micelles with high mass fractions (exceeding about 89%) of PCVL blocks, and they displayed excellent long-term physical stabilities at room temperature. However, the Pickering double emulsions underwent a rapid microstructural transition into simple oil-in-water Pickering emulsions instead of complete demulsification at elevated temperature (37 °C), which was attributed to the hydrophilicity of micelles enhanced when the core-forming PCVL melted realized by temperature stimulus. Consequently, such W/O/W Pickering double emulsions stabilized solely with semicrystalline PEG-b-PCVL micelles exhibit thermal responsiveness, enabling them to release vitamin B12 encapsulated within the internal aqueous phase rapidly.
Effect of surfactants on the interfacial behaviors of diacylglycerol-based solid lipid nanoparticles and physical stability of W/O emulsion

Abstract:The influence of surfactants (Span 80, Span 60, PGPR and sodium caseinate) on the stability of W/O emulsions stabilized by diacylglycerol (DAG)-based solid Lipid nanoparticles (SLN) was investigated. DAG-based SLN showed phase separation at high NaCl concentration and acidic pH although the SLN-stabilized W/O emulsion was minimally affected. Span 80 increased the hydrophobicity whereas Span 60 increased the hydrophilicity of SLN. With concentration close to CMC , both Span surfactants caused detachment of SLN from interface and reduced emulsion stability. Nonetheless, addition of a small amount (0.1 wt%) of Span surfactants increased the interfacial modulus. The co-adsorption of SLN and PGPR at the oil-water interface promoted the formation of denser network structure and higher interfacial elasticity, which enhanced the emulsion rigidity and freeze-thaw stability. Sodium caseinate (NaCAS) showed intense competition with particles for adsorption at the interface and caused emulsion destabilization. Consequently, the NaCl release rate was directly influenced by the emulsion stability and the interaction between surfactants with SLN. The results have important implications for applying DAG nanoparticles as stabilizers in W/O emulsions with tailoring interfacial rigidity and water-soluble component release behaviors.

Abstract:Insulin (INS, hydrophilic) and quercetin (Q, hydrophobic) have broad biological benefits; however, their application is limited because of their instability and poor bioaccessibility. Thus, in this study, water-in-oil-in-water (W/O/W) double emulsion gels based on black bean protein-sodium alginate Maillard conjugate-stabilized double emulsions were fabricated using three different inducers. The water holding capacity (WHC), intermolecular interaction forces, INS and Q encapsulation efficiencies (EEs), rheological and textural properties, microstructures, and structures were influenced by both inducer type [CaCl2, glucolactone (GDL), and glutamine transaminase (TGase)] and concentration. The addition of each inducer significantly increased the WHC, INS, and Q EEs, and textural properties of the emulsion gel. Moreover, the rheological behavior analysis showed that all the emulsion gels exhibited marked elastic behavior. These results will help develop an emulsion gel functional carrier based on a black soybean protein-carbohydrate-stabilized W/O/W double emulsion for the targeted and sustained release of drugs.

Abstract:Products such as cosmetics, pharmaceuticals, and crude oil often exist as water-in-oil (W/O) emulsions during their processing or in final form. In many cases, their dispersed aqueous phase is encased in a crystal network and/or by interfacially-adsorbed (‘Pickering’) particles [paraffins, triacylglycerols, polymers, etc.] that promote emulsion kinetic stability by hindering droplet–droplet contact, coalescence and macroscopic phase separation. In processed foods, important questions remain regarding whether a continuous phase fat crystal network or Pickering crystal provides better stabilization. This review explores the following factors related to crystal-stabilized W/O emulsions: i) the key properties dictating fat crystal spatial distribution (at the interface or in the continuous phase); ii) how temperature and freeze–thaw emulsion destabilization are intimately linked with fat crystal spatial distribution, and; iii) why oil-soluble surfactant interactions with the continuous oil phase influence fat crystal wettability and emulsifier efficacy. It is shown that these parameters strongly govern W/O emulsion formation and stability.

Abstract:Double emulsions are normally considered as metastable systems and this limit in stability restricts their applications. To enhance their stability, the outer shell can be converted into a mechanically strong layer, for example, a polymeric layer, thus allowing improved performance. This conversion can be problematic for food and drug applications, as a toxic solvent is needed to dissolve the polymer in the middle phase and a high temperature is required to remove the solvent. This process can also be highly complex, for example, involving UV initiation of polymeric monomer crosslinking. In this study, we report the formation of biocompatible, water-in-oil-in-water (W/O/W) double emulsions with an ultrathin layer of fish oil. We demonstrate their application for the encapsulation and controlled release of small hydrophilic molecules. Without a trigger, the double emulsions remained stable for months, and the release of small molecules was extremely slow. In contrast, rapid release was achieved by osmolarity shock, leading to complete release within 2 h. This work demonstrates the significant potential of double emulsions, and provides new in-sights into their stability and practical applications.
Fabrication and characterization of gel-in-oil-water (G/O/W) double emulsion stabilized by flaxseed gum/whey protein isolate complexes

Abstract:The aim of this study was to fabricate stable water-in-oil-in-water double emulsions with gelled internal water phase (G/O/W emulsion). The polyglycerol polyricinoleate (PGPR) was used as the lipophilic emulsifier and flaxseed gum (FG) /whey protein isolate (WPI) electrostatic complexes were used as the hydrophilic emulsifier. The rheological results showed that W1 became gelled when the FG concentration reached 0.4 wt%. Increased concentrations of FG in W1 significantly enhanced the stability of W1/O/W2 emulsions, accompanied with the diminution in droplet size and increased viscosity. It could be attributed to the strong interfacial activity and thickening effect of FG. Optical and confocal microscopy confirmed the formation of W/O/W emulsion structure and supported the particle size results. The maximum complex formation between FG and WPI occurred at pH 3.8, with the mass ratio of 1:1. In comparison to WPI, the complex of FG/WPI exhibited a significant improvement in the stability of G/O/W double emulsions. The storage stability test showed that the G/O/W emulsion fabricated with 0.8 wt% FG and 0.5 wt% FG/WPI complexes showed the highest stability without phase separation or oil-off phenomenon within 28 days. Consequently, the incorporation of FG (≥0.4 wt%) in W1 and utilization of FG/WPI complexes could be considered as a new strategy for obtaining W/O/W emulsions with long-term storage stability.
