Abstract:The hybrid oxidation effect of ultrasonics (US) and ultraviolet light (UV-A) on Cyanidin-3-O-glucoside (C3G) stability were examined. For comparison, sonolysis and photolysis experiments were also conducted. The results showed that under hybrid sonophotolysis and sonolysis treatment, C3G degradation undergoes zero-order kinetics, while under photolysis, first-order degradation kinetic prevailed. The degradation rate increases with the increase in US power, with the lowest, was recorded as 0.70 mu g/ml/h (14 W/L) and 0.77 mu g/ml/h (28 W/L), and highest as 0.80 mu g/ml/h (42 W/L). Similarly, the degradation ability of UV photolysis at 400 mu W/cm(2) was weak, which increased with increasing UV power (600 mu W/cm(2)). Overall the sonophotolysis degradation rate was significantly higher than that of the individual effect. With the addition of gallic acid (GA), the degradation of C3G was found lower under sonophotolysis; thus, it could be used as a natural protective agent for C3G during food processing.
Chemical stability of carboxylpyranocyanidin-3-O-glucoside under β-glucosidase treatment and description of their interaction

Abstract:Anthocyanins are susceptible to degradation by β-glycosidase, resulting in color loss. This study analyzed the impact of β-glycosidase on carboxylpyranocyanidin-3-O-glucoside (Carboxyl-pycy-3-gluc) and its precursor cyanidin-3-O-glucoside (Cy-3-gluc). Carboxyl-pycy-3-gluc exhibited enhanced stability upon treatment with β-glucosidase. Ultraviolet-visible and circular dichroism spectroscopy revealed slight changes in the microenvironment and secondary structure of β-glycosidase when carboxyl-pycy-3-gluc was present. The fluorescence experiment indicated that anthocyanins quench the fluorescence of β-glycosidase through static quenching via hydrophobic interactions. Molecular docking of six types of carboxylpyranoanthocyanins and their precursors with β-glycosidase revealed that carboxylpyranoanthocyanins exhibited lower binding affinity than their precursors, consistent with the enzyme kinetic experiment results. The incorporation carboxyl-pycy-3-gluc into Sanhua Plum Juice and Wine endowed them with vivid and stable coloration. The study illustrated that carboxyl-pycy-3-gluc exhibits low binding affinity with β-glycosidase, thereby maintaining stability and confirming its potential as a colorant.
Enzymatic hydrolysates of soy protein promote the physicochemical stability of mulberry anthocyanin extracts in food processing

Abstract:Soy protein papain hydrolysate (SPAH) and soy protein pepsin hydrolysate (SPEH) were used as protective agents for mulberry anthocyanin extracts (MAEs) to inhibit its color fading and enhance the anthocyanin stability at pH 6.3. Both SPAH and SPEH showed a significant protective effect on total anthocyanins in MAEs solutions. 1.0 mg/mL of SPEH presented the best protective effect on MAEs by increasing its half-life from 1.8 to 5.7 days. SPAH/SPEH-cyaniding-3-O-glucoside (C3G) interactions were investigated at pH 6.3 by fluorescence, Fourier-transform infrared spectroscopy (FT-IR), and Circular Dichroism (CD). Their association was mainly driven by hydrophobic interactions, and SPEH showed a higher binding affinity for C3G than SPAH, with a KA value of 2.62 × 105 M-1 at 300 K. The second structures of SPAH and SPEH were altered by C3G, with a decrease in the β-sheets and an increase in the turns and random coils.
Improving effects of three selected co-pigments on fermentation, color stability, and anthocyanins content of blueberry wine

Abstract:The effects of caffeic acid, syringic acid, and quercetin co-pigments on fermentation, color changes, and anthocyanins stability of blueberry wine were evaluated. The co-pigmented blueberry wines exhibited a higher alcohol and titratable acidity, and a lower sugar content. This was accompanied by a lower L* and a* and a greater b*, a significant shift in the λ max and an increase in absorbance value at λ max, altogether, indicating a darker and vivid bluish red color of blueberry wines. Total anthocyanins content in the wine samples with co-pigments were significantly higher with a greater stability after 20 days of post-fermentation. The malvidin glucoside and cyanidin glucoside exhibited the most stability in co-pigmented blueberry wines whilst none of the used co-pigments were able to protect cyanidin galactoside. We speculate that co-pigmentation with caffeic acid, syringic acid, or quercetin is an effective way to produce blueberry wine with a higher nutritional and sensory quality.
Protective effects of selenium on cadmium toxicity in rats: role of altered toxicokinetics and metallothionein.

Abstract:Selenium prevents the toxicity of the carcinogenic metal cadmium through undefined mechanisms. In this study, we determined the effects of selenium on cadmium toxicokinetics and on the ability of cadmium to induce metallothionein, a metal-binding protein that is thought to confer tolerance to cadmium toxicity. To assess the acute protective effects of selenium, male Wistar (WF/NCr) rats were given selenium (as SeO2; 10 mumol/kg, sc) at -24, 0, and +24 h relative to cadmium (as CdCl2; 45 mumol/kg, sc). Over a 14-d period this dose of cadmium killed 6 out of 10 rats, while 100% of the cadmium-treated rats given concurrent selenium treatments survived. The acute increases in testicular weight that were seen with cadmium, indicative of edematous damage, were also prevented by concurrent selenium treatments. Further studies assessed the distribution and excretion of cadmium and its ability to induce metallothionein in rats given 40 mumol Cd/kg, sc, at time 0 and selenium (10 mumol/kg, sc) at -24 and 0 h. Selenium treatments enhanced cadmium accumulation at 24 h in the liver (23%), testes (145%), and epididymis (35%) but reduced renal accumulation by more than half. Urine samples, collected at 0-3, 3-6, and 6-24 h following cadmium administration, indicated a markedly reduced excretion of cadmium in selenium treated rats during all time periods. The synthesis of metallothionein was stimulated to a much lesser extent by cadmium in selenium-treated rat kidney (41% decrease) but was unaffected in liver. The levels of cadmium-binding proteins within the testes were markedly reduced by cadmium treatment, an effect unmodified by selenium treatments. These results suggest selenium prevents acute cadmium toxicity through a mechanism that does not involve induction of metallothionein and in spite of a markedly enhanced retention of cadmium.
Pectin forms polymeric pigments by complexing anthocyanins during red winemaking and ageing

Abstract:The long-term stability of red wine color depends on the formation of polymeric pigments from anthocyanins. Although there is still a lot of uncertainty about the specific structure of this diverse group of pigments, there is consensus that they are reaction products of anthocyanins and other polyphenols. Interactions between anthocyanins and pectic polysaccharides have been suggested to stabilize anthocyanins. This study explores the impact of such interactions by adding pectin during red winemaking. The results demonstrate that these interactions induce the formation of additional polymeric pigments which enhance the pigment stability during fermentation and aging. While initial pigment formation is higher in wines with added pectin, a notable proportion of the complexes degrades in the later stages of fermentation. Presumably, tannins form insoluble complexes with pectin, reducing tannin concentration by more than 300 mg/L. Anthocyanin concentrations decrease by over 400 mg/L, and polymeric pigments double. Anthocyanins that form polymeric pigments with pectic polysaccharides expand the range of pigments in red wines with possible consequences for the sensory properties of the wine. These findings highlight the complex interactions between pectin, anthocyanins, and tannins, and their influence on pigment formation and wine composition during fermentation and aging.

Abstract:This work aims to evaluate the structure-stability relationship of anthocyanins in cell culture. An early degradation time (T-C(10)) and half-degradation time (T-C(50)) were used to characterise the stability of 10 of the most common anthocyanins, incubated with DMEM at 37 degrees C, pH = 7.4, 5% CO2 for different time periods. According to the glycosylation, the glycosylated forms were more stable than the not glycosylated forms. The methylation at 3'' or 5' position at ring B enhanced their stability; contrarily, the hydroxylation at 3' or 5' position at ring B weakened their stability. Glycosylated forms were much more stable in water than in the culture medium. Although not glycosylated forms were also instable in water, their stability was improved compared with culture medium. Together with the cell culture experiments and, in order to avoid artefacts, stability tests of polyphenols should be performed in parallel experiments with DMEM.
Effects of sucrose degradation product furfural on cyanidin-3-O-glucoside: Mechanism of action, stability, and identification of products in sugar solutions

Abstract:Anthocyanins are often affected by the sweetener sucrose during processing, which is closely related to its typical degradation product, furfural (Ff). However, the specific mechanism is unclear. In this study, Ff and cyanidin-3-O-glucoside (C3G) were used to explain the mechanism of the effect. The results showed that Ff destabilized anthocyanins by chemically reacting with C3G to generate three new adducts. Meanwhile, the color of the C3G solution changed from bright red to dark purple, and the value of the color difference (Delta E) increased significantly by 2.69. Furthermore, the new adducts were less stable than C3G and continued to promote the degradation of C3G when they coexisted with it. The above adducts were also detected in sugar solutions supplemented with C3G, and these adducts were more likely to accumulate under light storage conditions. These results provide a theoretical basis for reducing anthocyanin loss in food processing.
Monoglucoside versus Diglucoside Anthocyanin Evolution of Red Wine Produced Using a Fungus-Resistant Grape Cultivar (Downy Mildew and Powdery Mildew) under Oxidative Conditions

Abstract:The need to reduce the use of pesticides in viticulture is increasing the interest in wines produced using fungal-resistant grapevine varieties, which are characterized by relevant contents of both monoglucoside and diglucoside anthocyanins. Aging in wooden barrels induces oxygen permeation into wine, but little is known about diglucoside anthocyanin evolution. Cabernet cortis wine was subjected to addition of oxygen and oak chips, and the anthocyanin changes were followed for 1 month. Decreases of 90% total monoglucosides, 80% acylated monoglucosides, 65% diglucosides, and 90% acylated diglucosides were observed. Monoglucosides formed pyranoanthocyanins, and the lower steric hindrance favored their polymerization with flavanols. Instead, the decrease in diglucosides was correlated to the number of hydroxyl groups of ring B, indicating the predominant oxidation of aglycones. However, three flavonol-anthocyanin-diglucoside derivatives named (epi)catechin-ethyl-Mv-dihexoside, (epi)catechin-ethyl-Pn-dihexoside, and (epi)catechin-Mv-dihexoside A-type were identified in wine for the first time. These research findings are useful for tuning suitable oenological practices to stabilize the color of these wines (type of barrel, aging times, oxygenation practices) and lower the malvin content, which currently is recommended by the OIV at a maximum of 15 mg/L and is a critical issue for their commercialization.

Abstract:The color of red wine will change constantly during fermentation and storage, which is related to the yeast introduced. Wild yeast has a strong adaptability to the environment, and effect of which on the color change of red wine is more obvious. In order to explore the molecular mechanism of wild yeast on color change of red wine, this paper summarized the sources and species of wild yeast including influence on production capacity of anthocyanin. The variation of wine color regulated by yeast was related to the genetic material of parent of hybrid strain producing pigment, chromogenic enzyme and storage temperature. Proanthocyanins and yeasts could promote each other. Different yeasts could produce anthocyanins with similar species but greater differences. Therefore, wine color was controlled by various yeasts. Isotopic labeling technology could detect the obvious symbiotic relationship between yeast and anthocyanin, and track accurately and timely the change of wine color. Metabolism of wild yeast and instability of anthocyanin could co-lead to discoloration of wine, however the molecular mechanism of colour fading was not clear nowadays. Therefore, there was an urgent need to screen the wild yeast with the safety and stability to wine color.
Polyphenolic Compositions and Chromatic Characteristics of Bog Bilberry Syrup Wines

Abstract:Phenolic compounds determine the color quality of fruit wines. In this study, the phenolic compound content and composition, color characteristics and changes during 6 months of bottle aging were studied in wines fermented with bog bilberry syrup under three different pHs. The total anthocyanins and total phenols were around 15.12-16.23 mg/L and 475.82 to 486.50 mg GAE/L in fresh wines and declined 22%-31% and about 11% in bottle aged wines, respectively. In fresh wines, eight anthocyanins, six phenolic aids and 14 flavonols, but no flavon-3-ols were identified; Malvidin-3-O-glucoside, petunidin-3-O-glucoside and delphinium-3-O-glucoside were the predominant pigments; Chlorogentic acid was the most abundant phenolic acid, and quercetin-3-O-galactoside and myricetin-3-O-galactoside accounted for nearly 90% of the total flavonols. During 6 months of bottle storage, the amounts of all the monomeric anthocyanins and phenolic acids were reduced dramatically, while the glycosidyl flavonols remained constant or were less reduced and their corresponding aglycones increased a lot. The effects of aging on blueberry wine color were described as the loss of color intensity with a dramatic change in color hue, from initial red-purple up to final red-brick nuances, while the pH of the fermentation matrix was negatively related to the color stability of aged wine.

Abstract:In this study, the dynamic changes of different anthocyanins in the processing of 'Ziyan' tea wine were investigated quantitatively. Results showed that six types of anthocyanins, namely petunidin, malvidin, pelargonidin, delphinidin, cyanidin and peonidin, as well as two co-pigmented substances, procyanidins and flavonoids, were detected in 'Ziyan' tea wine. As fermentation proceeded, the contents of petunidin, pelargonidin, delphinidin, cyanidin and peonidin decreased. Among them, petunidin, peonidin and pelargonidin showed a tendency of decreasing first, then increasing and finally decreasing, whereas delphinidin and cyanidin continued to decrease during fermentation. Variation trend of procyanidins and flavonoids was consistent with those of petunidin. Furthermore, metabolism of delphinidin, cyanidin and pelargonidin were main pathways responsible for the anthocyanin changes during 'Ziyan' tea wine processing. These findings suggested that the color of 'Ziyan' tea wine was achieved by the combination of various anthocyanins in different ratios and the co-pigmentation of procyanidins and flavonoids.
Non-acylated and acylated anthocynins in red wines of different ages: Color contribution and evaluation

Abstract:This study investigated the non-acylated and acylated anthocyanins and chromatic characteristics of 108 red wines with a range of ages. The K-means cluster analysis showed that the evolution pattern of cyanidin-3-O-glucoside (Cy) differed among non-acylated anthocyanins and the pattern of malvidin-3-O-(6-O-acetyl)-glucoside (Mv-ac) differed among acylated anthocyanins. Partial least squares regression (PLS) showed that non-acylated anthocyanins, especially Cy, contributed to the red-violet color. Wine age, tawny characteristics, and color density strongly negatively correlated with anthocyanins. Wine age played an essential role in chromatic characteristics and anthocyanins, especially more than 4 years. A prediction model of wine age was established using the chosen variables of color parameters and anthocyanins employing PLS. This model showed good performance in predicting the wine age. Besides malvidin-3-O-glucoside (Mv), other anthocyanins and acylated forms with low content were demonstrated to be promising and practical chemical parameters for predicting the age of dry red wine.

Abstract:Anthocyanins are bioactive compounds responsible for various physiological processes in plants and provide characteristic colors to fruits and flowers. Their biosynthetic pathway is well understood; however, the enzymatic degradation mechanism is less explored. Anthocyanase (β-glucosidase (BGL)), peroxidase (POD), and polyphenol oxidase (PPO) are enzymes involved in degrading anthocyanins in plants such as petunias, eggplants, and Sicilian oranges. The aim of this work was to investigate the physicochemical interactions between these enzymes and the identified anthocyanins (via UPLC-MS/MS) in cranberry (Vaccinium macrocarpon) through molecular docking to identify the residues likely involved in anthocyanin degradation. Three-dimensional models were constructed using the AlphaFold2 server based on consensus sequences specific to each enzyme. The models with the highest confidence scores (pLDDT) were selected, with BGL, POD, and PPO achieving scores of 87.6, 94.8, and 84.1, respectively. These models were then refined using molecular dynamics for 100 ns. Additionally, UPLC-MS/MS analysis identified various flavonoids in cranberries, including cyanidin, delphinidin, procyanidin B2 and B4, petunidin, pelargonidin, peonidin, and malvidin, providing important experimental data to support the study. Molecular docking simulations revealed the most stable interactions between anthocyanase and the anthocyanins cyanidin 3-arabinoside and cyanidin 3-glucoside, with a favorable ΔG of interaction between −9.3 and −9.2 kcal/mol. This study contributes to proposing a degradation mechanism and seeking inhibitors to prevent fruit discoloration.

Abstract:The development of blueberry wine provides an alternative method for maintaining the nutritional value and extending the shelf life of blueberries. However, anthocyanin loss and off-flavor compound generation during fermentation impair blueberry wine color and quality. Hydroxycinnamate decarboxylase from yeast can catalyze the conversion of hydroxycinnamic acids to vinylphenols, which later may condense with anthocyanins to form more stable vinylphenolic pyranoanthocyanins. In this study, 10 non-Saccharomyces yeasts from Daqu that showed hydroxycinnamate decarboxylase activity were screened. Among the 10 strains, Wickerhamomyces anomalus Y5 showed the highest consumption (34.59%) of the total tested phenolic acids and almost no H2S production. Furthermore, Y5 seemed to produce four vinylphenol pyranoanthocyanins (cyanidin-3-O-galactoside/glucoside-4-vinylcatechol, cyanidin-3-O-galactoside/glucoside-4-vinylsyringol, malvidin-4-vinylguaiacol, and malvidin-4-vinylcatechol) during blueberry wine fermentation, which may improve the color stability of blueberry wine. These findings provide new insights for improving the quality of blueberry wine using non-Saccharomyces yeasts.
Influence of Saccharomyces and non-Saccharomyces Yeasts in the Formation of Pyranoanthocyanins and Polymeric Pigments during Red Wine Making

Abstract:Yeast are able to modulate many sensory parameters of wines during red must fermentation. The effect on color and on the formation of derived pigments during fermentation has been studied thoroughly since the 90s. Yeast can increase grape anthocyanin’s color by acidification by hyperchromic effect (increase of flavylium molecules). Recent studies with non-Saccharomyces species, as Lachancea thermotolerans, described the intense effect of some strains on anthocyanin’s color, and subsequent, stability, by strongly reducing wine’s pH during fermentation. Moreover, selected yeast strains of Saccharomyces have been shown to release metabolites such as pyruvic acid or acetaldehyde that promote the formation of vitisin A and B pyranoanthocyanins during must fermentation. Schizosaccharomyces pombe, because of its specific metabolism, can produce higher concentrations of pyruvate, which enhances the formation of vitisin A-type derivatives. The hydroxycinnamate decarboxylase activity that some Saccharomyces strains express during fermentation also promotes the formation of vinylphenolic derivatives. Some non-Saccharomyces species, such as S. pombe or P. guilliermondii can also improve the production of these derivatives compared to selected strains of Saccharomyces cerevisiae. Lastly, some yeasts are also able to modulate the formations of polymeric pigments between grape anthocyanins and flavonoids, such as catechins and procyanidins.

Abstract:Yeast, lactic acid bacteria and acetic acid bacteria were used to ferment Chinese bayberry pomace, and then the effects of mixed fermentation on the quality (color, antioxidant capacity and phenolic compounds) were studied. We found that mixed fermentation could protect the color of beverage and slow down the degradation of anthocyanins. Anthocyanins interact with acetaldehyde, acetic acid and lactic acid produced by lactic acid bacteria and acetic acid bacteria could improve the color of the beverage. However, the action of enzymes would seriously damage the color and aggravate the degradation of anthocyanins. Mixed fermentation promoted the conversion of flavonoids into flavonol aglycones. After mixed fermentation and enzymes action, the type of phenolic acid did not change, but the contents increased. According to the results of DPPH-HPLC, myricetin did not have the ability to scavenging DPPH free radical. Therefore, mixed fermentation could improve the quality of beverage.

Abstract:Acetaldehyde is a key carbonyl by-product during red wine alcoholic fermentation; it is reactive and takes part in certain reactions involving anthocyanin. The aim of this study was to investigate the influence of SO2 and condensed tannin on the acetaldehyde accumulation of Saccharomyces cerevisiae (S. cerevisiae) during alcoholic fermentation and the ripple effect on wine anthocyanin. In this study, six sets of Cabernet Sauvignon alcoholic fermentation with two different sulfur levels (HS and LS) were carried out by adding exogenous condensed tannins before fermentation (T0) in the acetaldehyde rise period (TA) of S. cerevisiae and at the end of fermentation (TE), separately. The acetaldehyde evolution was identified during fermentation and anthocyanin was analyzed comparatively. The results showed that HS treatment slowed down the degradation of acetaldehyde, while tannins accelerated the degradation of acetaldehyde during alcoholic fermentation, especially TA wines. Furthermore, TA wines possessed a unique anthocyanin profile after fermentation regardless of SO2 level compared with other wines. These results suggest that acetaldehyde-mediated anthocyanin polymerization most likely occurs timely at the acetaldehyde production phase of S. cerevisiae during alcoholic fermentation, and managing tannin addition time during production could be used to regulate the anthocyanin profile.

Abstract:This study evaluated the degradation kinetics of selected anthocyanins and the change in polymeric color, browning index, and haze development of grape juices from "Merlot" and "Ruby" grape cultivars stored at 5, 25, and 35°C for up to 360 days. Five major anthocyanins namely malvidin-3-O-glucoside (M3G), delphinidin-3-O-glucoside (D3G), petunidin-3-O-glucoside (Pt3G), peonidin-3-O-glucoside (Pn3G), and cyanidin-3-O-glucoside (C3G) were identified. Juice from "Merlot" had significantly higher (p < 0.05) content of all individual anthocyanins as compared to "Ruby." During the long-term storage, total, and individual anthocyanins from both cultivars degraded following first-order reaction kinetics at the rate strongly dependent on temperature. At the end of the storage, noticeably higher loss of anthocyanins (95-99.9%) was observed at 25 and 35°C as compared to storage at 5°C [50-60% ("Merlot"); 74-81% ("Ruby")]. Considerably lower rate of decay was observed at 5°C (k = 0.01-0.04) as compared to 25 (k = 0.04-0.14) and 35°C (k = 0.05-0.14) storage temperatures. The most temperature sensitive anthocyanin compounds were C3G (Ea = 66.5 kJ/mol) and D3G (Ea = 63.3 kJ/mol). At higher storage temperatures, significant (p < 0.05) and strong negative correlations were observed between anthocyanin concentrations and the levels of haze, polymeric and brown color development during storage. Storing grape juice, at lower temperature conditions could reduce the continuous loss of biologically active anthocyanins as well as the development of haze and brown color.

Abstract:Non-enzymatic browning occurs widely in both white and red wines, and it has a huge impact on the color evolution and aging potential. Previous studies have proved that phenolic compounds, especially those with catechol groups, are the most important substrates involved in browning reactions of wine. This review focus on the current knowledge of non-enzymatic browning in wine resulting from monomeric flavan-3-ols. First, some relevant aspects of monomeric flavan-3-ols are introduced, including their structures, origins, chemical reactivities, as well as potential impacts on the organoleptic properties of wine. Second, the mechanism for non-enzymatic browning induced by monomeric flavan-3-ols is discussed, with an emphasis on the formation of yellow xanthylium derivatives, followed by their spectral properties and effects on the color change of wine. Finally, attentions are also be given to the factors that influence non-enzymatic browning, such as metal ions, light exposure, additives in winemaking, etc.
