Abstract:Most of the existing electrochemical systems failed to achieve satisfactory results in early diagnosis of Alzheimer's disease (AD) owing to a deficiency of effective signal transduction . A new method for the electrochemical detection of AD biomarkers (amyloid-beta oligomers, Aβ1-42 oligomers) was developed based on a double amplification strategy. Titanium carbide (Ti 3 C 2 ) MXene decorated by gold nanoparticles (Au-MXene) as the electrode substrate not only gave rise to the electrochemical response due to its paradoxical surface area and conspicuous charge mobility, but also provided vast numbers of binding sites for aptamers (Apt) of Aβ1-42 oligomers. Meanwhile, AuNPs were incorporated into covalent organic frameworks (COFs), which were further modified by Apt and electron mediator (toluidine blue, TB). The Apt/TB-Au@COFs composite was utilized as a label because of their improvement of the electron-hole separation efficiency and optimization of the charge-carrier utilization. The proposed electrochemical assay established highly efficient platform for the detection of Aβ1-42 oligomers with a linear range from 0.01 pg mL −1 to 180 pg mL −1 and an ultralow detection limit of 4.27 fg mL −1 (S/N = 3). This biosensing platform had potential applications in molecular diagnostics of AD serum samples.

Abstract:Accurate and sensitive detection of amyloid-β 42 oligomer (Aβ42O) is of great significance for early diagnosis of Alzheimer's disease (AD). Herein, a signal on-off ratiometric electrochemical immunosensor was developed for highly selective and quantitative determination of Aβ42O by using novel covalent organic frameworks (COFs) composites as the sensing platform. This immunosensor produced two independent electrochemical signals from the [Fe(CN)6]3-/4- and methylene blue (MB) probes at different potentials based on the electrocatalytic activity of gold nanoparticle-functionalized porphyrinyl COFs nanocomposites toward [Fe(CN)6]3-/4- and the signal probe of MB encapsulated in the aptamer-modified alkynyl COFs. Because the two signals of [Fe(CN)6]3-/4- and MB changed in opposite directions, a signal on-off mode was generated which can correct the results by introducing a reference signal and effectively eliminate background interference. Under optimal experimental conditions, the current ratio (IMB/I[Fe(CN)6]3-/4-) was well linearly related to the logarithmic value of Aβ42O concentrations in the range of 10 pM to 1 μM, and the detection limit was 5.1 pM (S/N = 3). Additionally, the immunosensor exhibited satisfactory performance in case of real cerebrospinal fluid samples. The designed ratiometric electrochemical immunosensor provides a valuable route for early diagnosis of AD and our results also pave the way for designing of sensing platforms using COF-based nanomaterials and extending their functions and applications to bioanalysis.
In-situ nanozyme catalytic amplification coupled with a universal antibody orientation strategy based electrochemical immunosensor for AD-related biomarker

Abstract:An in-situ nanozyme signal tag combined with a DNA-mediated universal antibody-oriented strategy was proposed to establish a high-performance immunosensing platform for Alzheimer's disease (AD)-related biomarker detection. Briefly, a Zr-based metal-organic framework (MOF) with peroxidase (POD)-like activity was synthesized to encapsulating the electroactive molecule methylene blue (MB), and subsequently modified with a layer of gold nanoparticles on its surface. This led to the creation of double POD-like activity nanozymes surrounding the MB molecule to form a nanozyme signal tag. A large number of hydroxyl radicals were generated by the nanozyme signal tag with the help of H2O2, which catalyzed MB molecules in situ to achieve efficient signal amplification. Subsequently, a DNA-aptamer-mediated universal antibody-oriented strategy was proposed to enhance the binding efficiency for the antigen (target). Meanwhile, a poly adenine was incorporated at the end of the aptamer, facilitating binding to the gold electrode and providing anti-fouling properties due to the hydrophilicity of the phosphate group. Under optimal conditions, this platform was successfully employed for highly sensitive detection of AD-associated tau protein and BACE1, achieving limits of detection with concentrations of 3.34 fg/mL and 1.67 fg/mL, respectively. It is worth mentioning that in the tau immunosensing mode, 20 clinical samples from volunteers of varying ages were analyzed, revealing significantly higher tau expression levels in the blood samples of elderly volunteers compared to young volunteers. This suggests that the developed strategy holds great promise for early AD diagnosis.

Abstract:Amyloid β-peptide oligomer (AβO) has received extensive attention from researchers because of its clinical therapeutic intervention targets and the value of reliable biological macromolecules markers for early diagnosis of Alzheimer's disease. We have developed a novel label-free electrochemical detection sensor for AβO based on hybridization chain reaction (HCR)-triggered poly adenine to absorb silver nanoparticles (AgNPs). In this method, we first use the "capture probe" to immobilize the aptamer 1 on the surface of the gold electrode (GE) via poly adenine-Au. Next, aptamer 2 and AβO were deposited on the electrode surface. The HCR process was initiated by the aptamer 2 fragment as a primer, producing a large number of long DNA sequences, which contained many adenines. Thereafter, the HCR product with long-repeated adenines could absorb many AgNPs on the surface of the electrode, which were used for subsequent electrochemical stripping of the AgNPs. The concentration range of the electrochemical signal of AβO was 1 pM-10 nM, and the detection limit was 430 fM, which indicated that that the detection system has high selectivity for the target protein.

Abstract:Amyloid beta‐protein (Aβ) is a main hallmark of Alzheimer's disease (AD), and a low amount of Aβ protein accumulation appears to be a potential marker for AD. Here, we developed an electrochemical DNA biosensor based on polyamide/polyaniline carbon nanotubes (PA/PANI‐CNTs) with the aim of diagnosing Alzheimer's disease early using a simple, low‐cost, and accessible method to rapidly detect Aβ42 in human blood. Electrospun PA nanofibers served as the skeleton for the successive in‐situ deposition of PANI and CNTs, which contributed both high conductivity and abundant binding sites for the Aβ42 aptamers. After the aptamers were immobilized, this aptasensor exhibited precise and specific detection of Aβ42 in human blood within only 4 min with an extremely fastest response rate, lower detection limit, and excellent linear detection range. These findings make a significant contribution to advancing the development of serum‐based detection techniques for Aβ42, thereby paving the way for improved diagnostic capabilities in the field of AD. Colorimetric determination of amyloid-β peptide using MOF-derived nanozyme based on porous ZnO-Co3O4 nanocages

Abstract:A sensitive and rapid colorimetric biosensor has been developed for determination of amyloid-β peptide (Aβ) and study of amyloidogenesis based on the high peroxidase-like activity of porous bimetallic ZnO-Co3O4 nanocages (NCs). Due to the high binding ability of Aβ monomer to ZnO-Co3O4 NCs, the catalytic activity of ZnO-Co3O4 NCs can be significantly suppressed by Aβ monomer. This finding forms the basis for a colorimetric assay for Aβ monomer detection. The detection limit for Aβ monomer is 3.5 nM with a linear range of 5 to 150 nM (R2 = 0.997). The system was successfully applied to the determination of Aβ monomer in rat cerebrospinal fluid. Critically, the different inhibition effects of monomeric and aggregated Aβ species on the catalytic activity of ZnO-Co3O4 NCs enabled the sensor to be used for tracking the dynamic progress of Aβ aggregation and screening Aβ inhibitors. Compared with the commonly used thioflavin T fluorescence assay, this method provided higher sensitivity to the formation of Aβ oligomer at the very early assembly stage. Our assay shows potential application in early diagnosis and therapy of Alzheimer's disease (AD).

Abstract:Beta-amyloid 42 (Aβ-42) peptide is an essential biomarker for Alzheimer's disease (AD) diagnosis. This study presents high sensitivity and stability sensors for Aβ-42 peptide detection, along with an integrated handheld analysis and display device. The sensors, with a wide response range and high accuracy, capable of measuring Aβ-42 peptide concentration at the femtogram level, could have enabled early screening for AD. Employing an in-situ grown three-dimensional sensor, the specific surface area of the sensor was increased by 200 % which, thereby, provided more antibody binding sites to enhance detection accuracy. This electrochemical sensor demonstrated a broad linear response range from 4.5 × 10 −8 pg/mL to 4.5 × 10 2 pg/mL and could accurately determine the concentration of spiked analytes (including artificial saliva, human plasma, and human blood). Furthermore, the sensor exhibited satisfactory stability, with only a 5 % sensitivity loss after 35 days of storage at 4 °C. Additionally, this handheld Aβ-42 peptide testing device incorporates a display for result delivery. Such innovative technology could fill the gap in the market for portable testing instruments and could provide an efficient and reliable solution for early AD screening.

It integrates fluorescence resonance energy transfer (FRET)-based fluorescence detection with a poly(dimethylsiloxane) (PDMS)-based microfluidic chip. The carboxyl fluorescein-modified AβO aptamers are employed as the fluorophore, and three-dimensional Fe3O4/MXene nanospheres are used as the fluorescence quencher for the first time. Moreover, the PDMS-based multichamber microfluidic chip serves as a fluorescence detection platform. The Fe3O4/MXene-based nanosphere-based biosensor shows an excellent linear relationship between the fluorescence intensity and the logarithm of the AβO concentration in the range of 0.10–200 nM. The detection limit is ∼0.05 nM at the ultralow sample volumes of ∼4.50 μL. These results strongly show that the fabricated high-performance biosensor can be widely used in smart healthcare including but not limited to medical diagnosis, health monitoring, and biological studies.

Abstract:Amyloid-β(1–42) [Aβ(1–42)] oligomer accumulations are associated with physiologic alterations in the brains of individuals with Alzheimer’s disease. In this study, we demonstrate that a nanostructured gold electrode with deposited gold nanoparticles, induced via electrochemical impedance spectroscopy (EIS), may be used as an Aβ(1–42) conformation biosensor for the detection of Alzheimer’s disease. Monoclonal antibodies (12F4) were immobilized on self-assembled monolayers of the electrochemical sandwich immunoassay biosensor to capture Aβ(1–42) monomers and oligomers. Western blot and fluorescence microscopy analyses were performed to confirm the presence of Aβ(1–42) monomers and oligomers. EIS analysis with an equivalent circuit model was used to determine the concentrations of different Aβ(1–42) conformations in this study. We identified conformations of Aβ(1–42) monomers and Aβ(1–42) oligomers using probe antibodies (12F4) by employing EIS. R Aβ ( 1 − 42 ) indicates the sum resistance of impedance measured during Aβ(1–42) immobilization. Δ R 12 F 4 refers to the concentration of probe antibody (12F4) binding with Aβ(1–42). The concentration of Aβ(1–42) oligomer was defined as the percentage of Aβ(1–42) aggregation R 12 F 4 / R Aβ ( 1 − 42 ) . The experimental results show that the biosensor has high selectivity to differentiate Aβ(1–40) and Aβ(1–42) monomers and Aβ(1–42) oligomers and that it can detect Aβ(1–42) oligomer accurately. The linear detection range for Aβ(1–42) oligomers was between 10 pg/ml and 100 ng/ml. The limit of detection was estimated to be 113 fg/ml.

Abstract:Fall in skin temperature during initial muscular work was investigated in ten healthy men. Bicycle exercise was performed at workloads of 50-150 W in a climatic chamber at ambient temperatures of 10-40 degrees C (relative humidity 45-55%). Skin temperatures at seven or eight points over the body surface were measured using thermography and thermocouple recording systems. Sweat rates were significantly higher at 40 degrees C than at 30 degrees C, whereas the fall in skin temperature was almost equal. The reduction of skin temperature during exercise was the same throughout the year, although sweat rate was significantly higher in summer than in winter. In coloured thermographics of the skin temperature distribution during exercise of both 50 and 150 W at 10 or 20 degrees C, the skin temperature began to decline immediately at the onset of the exercise. Increased work intensities reduced skin temperature. The results suggest that fall in skin temperature during initial exercise was not due to increased evaporative cooling but to vasoconstriction, probably caused by non-thermal factors.

Abstract:More than 55 million people live with dementia worldwide in 2021, and there are nearly 10 million new cases every year. Alzheimer's disease (AD) is the most common cause of dementia. Despite urgent need, early detection of AD and long-term monitoring of AD progression have been challenging. This is due to the limited availability of brain imaging facilities and the highly invasive procedure with the cerebrospinal fluid assay to assess the level of AD biomarkers, such as beta-amyloid (Aβ). Reliable measurements of AD biomarkers in blood samples are still difficult because of their very low abundance. Here, we develop a rapid, specific, and ultrasensitive immunoassay using plasmonic-gold nanoisland (pGOLD) chips with near-infrared fluorescence-enhanced detection for Aβ<sub>1-40</sub> and Aβ<sub>1-42</sub>. We show step-by-step processes and results during the platform establishment, including antibody specificity and sensitivity tests, antibody pair examination, condition optimization, and procedure refinement. Finally, we demonstrate the platform performance with detection sensitivity at the subpicogram per milliliter level. This platform, therefore, has a great application potential for early detection of AD using blood samples.

Nanomaterials are promising for constructing highly sensitive transistor-based biosensors due to their small size. However, such biosensors are difficult to fabricate on a large scale and suffer from the lack of combined optimization of reproducibility and sensitivity in complex physiological fluids. In this work, field-effect transistor (FET) biosensors based on highly uniform semiconducting carbon nanotube (CNT) thin films are mass produced to achieve highly sensitive and selective detection of the AD core blood biomarkers of β-amyloid (Aβ). The combination of the mass-produced CNT FET sensors and oligonucleotide aptamers as efficient bioreceptors enables reliable and reproducible sub-femtomolar detection in full human serum for Aβ42 and Aβ40 peptides and has outperformed other methods reported to date. The adsorption of biological substrates to the sensor was significantly reduced by multiple blocking steps, resulting in selectivity ratios of up to 730% (Aβ40) and 800% (Aβ42). The aptamer-functionalized CNT FET biosensor exhibits a large dynamic range (>104), rapid response time (several minutes), and low variation (<10%) and can be delivered as a low-cost and rapid clinical detection technology for the early diagnosis and mass screening of AD. This platform will help bring complex laboratory-based and expensive diagnostic tools to the point of care.

Abstract:Amyloid-β oligomer (AβO) is believed to cause neurotoxicity which is linked to Alzheimer's disease (AD). Therefore, the detection of AβO has been proposed as an effective method for the early diagnosis of AD. In this work, several three-dimensional layers using gold nanoparticles (AuNPs) embedded in different conducting polymer matrix, including poly (thiophene-3-acetic acid), poly (pyrrole-2-carboxylic acid), and poly (pyrrole-3-carboxylic acid), were harnessed as transducers for the large surface area and high electrical conductivity. The cellular prion protein (PrPC) was utilized as the biorecognition element for the specific detection of AβO. The prepared electrochemical sensors were fabricated and compared afterward, in terms of sensitivity and detection range for the highly sensitive detection of AβO. In comparison, the PrPC/AuNPs embedded in Ppy-3-COOH matrix (AuNPs-E-Ppy-3-COOH matrix) was demonstrated to have higher sensitivity and wider detection range (10−9 to 103 nM). Subsequently, through the ex vivo real sample analysis within cerebrospinal fluid and blood test, it is proved that PrPC/AuNPs-E-Ppy-3-COOH matrix can be used for the early diagnosis of AD.

Nanozyme sensor array based on manganese dioxide for the distinction between multiple amyloid β peptides and their dynamic aggregation process

Abstract:The determination of the amyloid β (Aβ) peptide and its aggregation intermediates helps to understand the pathological mechanism of Alzheimer's disease (AD) caused by toxic amyloid fragments. Because of the transient and heterogeneous properties of Aβ aggregates, it is very difficult to dynamically detect Aβ and its aggregation intermediates. Herein, we successfully constructed a two-dimensional manganese dioxide (MnO2) nanozyme sensor array by modulating the peroxidase-mimicking activity using various Aβ species and accurately distinguished among six types of Aβ within 1 h through linear discriminant analysis (LDA), with a dynamic detection range of 0.01-500 nmol/L and a detection limit of 0.44 pmol/L. Subsequently, 30 unknown blind samples were used to verify the practicability of the sensor array, and all unknown samples were identified with 100% accuracy. It is worth noting that the sensor array successfully distinguished healthy individuals from AD patients using clinical blood samples. This study provides a convenient and reliable nanozyme biosensing system for detecting Aβ species and their related aggregation processes.
Cobalt-based Metal-Organic Frameworks As Co-Reaction Accelerator for Enhancing Electrochemiluminescence Behavior of N-(aminobutyl)-N-(ethylisoluminol) and Ultrasensitive Immunosensing of Amyloid-Β Protein

Abstract:In this work, a "luminophore/coreactant/co-reaction accelerator" ternary electrochemiluminescence (ECL) system is proposed by using N-(aminobutyl)-N-(ethylisoluminol) (ABEI) functionalized cobalt-based MOFs (Co-MOFs/ABEI) with superior peroxidase-like activities as the luminophore. ABEI, the derivative of luminol, is used as the reductant and organic ligand to synthesize flower-like Co-MOFs/ABEI. The as-synthesized Co-MOFs/ABEI not only function as desirable nano-carriers for the enrichment of ABEI, but also act as co-reaction accelerators for the catalysis of H2O2 decomposition. Furthermore, Au nanoparticles are decorated on Co-MOFs/ABEI via electrostatics attraction, endowing it with favorable biocompatibility and electrocatalytic property to bond with detection antibodies and generate more excited state ABEI*. Besides, with the advantages of large specific surface area and easy preparation, Au nanoparticles functionalized Fe3O4@polypyrrole is utilized to anchor the capture antibodies for immunoaction with target analyze. Amyloid-beta protein (A beta(42)), a principal hallmark of Alzheimer's disease, is chosen as the target analyze. The proposed A beta(42) immunosensor performs with a broad linear range from 10 fg mL(-1) to 100 ng mL(-1) and a detection limit down to 3 fg mL(-1), opens up new prospects for clinical analysis of A beta(42) and provides promising approaches to synthesize luminophore-functionalized MOFs with high luminous efficiency for ECL bioanalysis.

Abstract:Alzheimer's disease (AD) is a neurodegenerative disorder associated with a severe loss in thinking, learning, and memory functions of the brain. To date, no specific treatment has been proven to cure AD, with the early diagnosis being vital for mitigating symptoms. A common pathological change found in AD-affected brains is the accumulation of a protein named amyloid-β (Aβ) into plaques. In this work, we developed a micron-scale organic electrochemical transistor (OECT) integrated with a microfluidic platform for the label-free detection of Aβ aggregates in human serum. The OECT channel-electrolyte interface was covered with a nanoporous membrane functionalized with Congo red (CR) molecules showing a strong affinity for Aβ aggregates. Each aggregate binding to the CR-membrane modulated the vertical ion flow toward the channel, changing the transistor characteristics. Thus, the device performance was not limited by the solution ionic strength nor did it rely on Faradaic reactions or conformational changes of bioreceptors. The high transconductance of the OECT, the precise porosity of the membrane, and the compactness endowed by the microfluidic enabled the Aβ aggregate detection over eight orders of magnitude wide concentration range (femtomolar-nanomolar) in 1 μL of human serum samples. We expanded the operation modes of our transistors using different channel materials and found that the accumulation-mode OECTs displayed the lowest power consumption and highest sensitivities. Ultimately, these robust, low-power, sensitive, and miniaturized microfluidic sensors helped to develop point-of-care tools for the early diagnosis of AD.

Abstract:Amyloid-β protein (Aβ) is an important biomarker and plays a key role in the early stage of Alzheimer’s disease (AD). Here, an ultrasensitive photoelectrochemical (PEC) sensor based on ZnO@polydopamine/Au nanocomposites was constructed for quantitative detection of Aβ. In this sensing system, the ZnO nanorod array decorated with PDA films and gold nanoparticles (Au NPs) have excellent visible-light activity. The PDA film was used as a sensitizer for charge separation, and it also was used for antibody binding. Moreover, Au NPs were loaded on the surface of PDA film by in situ deposition, which further improved the charge transfer efficiency and the PEC activity in visible light due to the localized surface plasmon resonance effect of Au NPs. Therefore, in ZnO@polydopamine/Au nanocomposites, a significantly enhanced photocurrent response was obtained on this photoelectrode, which provides a good and reliable signal for early detection of AD. Under the optimized conditions, the PEC immunosensor displayed a wide linear range from 1 pg/mL to 100 ng/mL and a low detection limit of 0.26 pg/mL. In addition, this PEC immunosensor also presented good selectivity, stability, and reproducibility. This work may provide a promising point-of-care testing method toward advanced PEC immunoassays for AD biomarkers.
