Abstract:The mammalian microbial communities in the gastrointestinal tract (GIT) play important roles in host nutrition and health. However, we still lack an understanding of how these communities are organized across GIT in natural environments. Here, using 16S rRNA gene sequencing, we analyzed the bacterial community diversity, network interactions and ecosystem stability across five gut regions (mouth, stomach, small intestine, cecum and colon) emanating from two common pika species in China, including Plateau pikas (Ochotona curzoniae) inhabiting high-altitude regions, as well as Daurian pikas (O. daurica) occupying low-altitude areas. The relative abundances of dominant Bacteroidetes and Firmicutes exhibited an increasing trend from mouth to colon. Cecum and colon harbored higher bacterial diversity compared with other anatomical regions. Gut region significantly influenced the structure of bacterial communities in the GIT. Network analysis indicated that topological features showed marked variations among gut regions. Interestingly, the ecosystem stability of bacterial communities increased gradually from mouth to colon. Our results suggest that gut region influences the diversity, structure and network interactions of bacterial communities in pikas. For hindgut-fermenting herbivorous mammals, relatively higher bacterial diversity and ecosystem stability in the cecum may provide a favorable condition for the fermentation of indigestible plant polysaccharides.

Abstract:The gut microbiota in mammals plays a key role in host metabolism and adaptation. However, relatively little is known regarding to how the animals adapts to extreme environments through regulating gut microbial diversity and function. Here, we investigated the diet, gut microbiota, short-chain fatty acid (SCFA) profiles, and cellulolytic activity from two common pika (Ochotona spp.) species in China, including Plateau pika (Ochotona curzoniae) from the Qinghai-Tibet Plateau and Daurian pika (Ochotona daurica) from the Inner Mongolia Grassland. Despite a partial diet overlap, Plateau pikas harbored lower diet diversity than Daurian pikas. Some bacteria (e.g., Prevotella and Ruminococcus) associated with fiber degradation were enriched in Plateau pikas. They harbored higher gut microbial diversity, total SCFA concentration, and cellulolytic activity than Daurian pikas. Interestingly, cellulolytic activity was positively correlated with the gut microbial diversity and SCFAs. Gut microbial communities and SCFA profiles were segregated structurally between host species. PICRUSt metagenome predictions demonstrated that microbial genes involved in carbohydrate metabolism and energy metabolism were overrepresented in the gut microbiota of Plateau pikas. Our results demonstrate that Plateau pikas harbor a stronger fermenting ability for the plant-based diet than Daurian pikas via gut microbial fermentation. The enhanced ability for utilization of plant-based diets in Plateau pikas may be partly a kind of microbiota adaptation for more energy requirements in cold and hypoxic high-altitude environments.
Host Species Shapes the Co-Occurrence Patterns Rather Than Diversity of Stomach Bacterial Communities in Pikas.

Abstract:The mammalian stomach acts as an important barrier against ingested pathogens into the entire gastrointestinal tract, thereby playing a key role in host health. However, little is known regarding to the stomach microbial compositions in wild mammals and the factors that may influence the community compositions. Using high-throughput sequencing of the 16S rRNA gene, we characterized the stomach bacterial community compositions, diversity, and interactions in two common pika (Ochotona sp.) species in China, including Plateau pikas (Ochotona curzoniae) and Daurian pikas (Ochotona daurica) living in the Qinghai-Tibet Plateau and the Inner Mongolia Grassland, respectively. The bacterial communities can be divided into two distinct phylogenetic clusters. The most dominant bacteria in cluster I were unclassified bacteria. Cluster II was more diverse, predominantly consisting of Bacteroidetes, followed by unclassified bacteria, Firmicutes and Proteobacteria. Three dominant genera (Prevotella, Oscillospira, and Ruminococcus) in pika stomachs were significantly enriched in cluster II. In addition, seasons, host species, and sampling sites as well as body weight and sex had no significant impacts on the composition and diversity of pika stomach communities. Interestingly, Plateau pikas harbored a more complex bacterial network than Daurian pikas, and these two pika species showed different co-occurrence patterns. These results suggested that the pika stomach harbors a diverse but relatively stable and unique bacterial community, which is independent on host (host species, body weight, and sex) and measured environmental factors (sampling sites and seasons). Interestingly, host species shapes the microbial interactions rather than diversity of stomach bacterial communities in pikas, reflecting specific niche adaptation of stomach bacterial communities through species interactions.

Abstract:The composition of the mammalian gut bacterial communities can be influenced by the introduction of environmental bacteria in their respective habitats. However, there are no extensive studies examining the interactions between environmental bacteriome and gut bacteriome in wild mammals. Here, we explored the relationship between the gut bacterial communities of pika (Ochotona spp.) and the related environmental bacteria across host species and altitudinal sites using 16S rRNA gene sequencing. Plateau pikas (O. curzoniae) and Daurian pikas (O. daurica) were sampled at five different sites, and plant and soil samples were collected at each site as well. Our data indicated that Plateau pikas and Daurian pikas had distinct bacterial communities. The pika, plant and soil bacterial communities were also distinct. Very little overlap occurred in the pika core bacteria and the most abundant environmental bacteria. The shared OTUs between pikas and environments were present in the environment at relatively low abundance, whereas they were affiliated with diverse bacterial taxa. These results suggested that the pika gut may mainly select for low-abundance but diverse environmental bacteria in a host species-specific manner.

Abstract:Gut microbiome plays crucial roles in animal adaptation and evolution. However, research on adaptation and evolution of small wild high-altitude mammals from the perspective of gut microbiome is still limited. In this study, we compared differences in intestinal microbiota composition and function in Plateau pikas ( Ochotona curzoniae ) and Daurian pikas ( O. daurica ) using metagenomic sequencing. Our results showed that microbial community structure had distinct differences in different pika species. Prevotella, Methanosarcina, Rhizophagus , and Podoviridae were abundant bacteria, archaea, eukaryotes, and viruses in Plateau pikas, respectively. However, Prevotella, Methanosarcina, Ustilago , and Retroviridae were dominated in Daurian pikas. Functional pathways related to carbohydrate metabolism that refer to the utilization of pectin, hemicellulose, and debranching enzymes were abundant in Plateau pikas, while the function for degradation of chitin, lignin, and cellulose was more concentrated in Daurian pikas. Pika gut had abundant multidrug resistance genes, followed by glycopeptide and beta-lactamase resistance genes, as well as high-risk antibiotic resistance genes, such as mepA, tetM , and bacA. Escherichia coli and Klebsiella pneumoniae may be potential hosts of mepA . This research provided new insights for adaptation and evolution of wild animals from perspective of gut microbiome and broadened our understanding of high-risk antibiotic resistance genes and potential pathogens of wild animals.

Abstract:Wild mammals often consume different food sources as they become geographical available. This change in diet composition is likely to influence the gut microbial community, yet it remains unclear what the relationship looks like-particularly in small herbivores-under natural conditions. We used DNA sequencing approaches to characterize the diet composition and gut microbial community of wild plateau pikas (Ochotona curzoniae) collected from three altitudes. We tested if diet and gut microbiota composition changes across altitudes, and the relationship between diet diversity and gut microbiota diversity. Our results showed that altitude significantly influences the composition of diet and gut microbial communities. Notably, the alpha diversity (Shannon diversity and observed OTUs) of individual diet was not significantly correlated with that of gut microbiota, whereas the beta diversity (Jaccard and Bray-Curtis dissimilarity) of diet was positively correlated with that of gut microbiota. Our study is the first time to highlight the relationship between diet and gut microbiota composition in wild pikas on the Qinghai-Tibet Plateau. It suggests that the species richness within individual gut microbiota does not linearly increase with diet diversity, whereas those individuals that are more similar in diet composition harbor more similar gut microbiota.

Abstract:ABSTRACT Differentiation of the gut microbiota between individuals within a species can be identified by enterotypes. However, whether different enterotypes are associated with host-related physiological status and adaptation to specific habitats is still poorly understood. In this study, we collected 308 cecal samples from plateau pikas during varying seasons and altitudes. We identified three bacterial and two fungal enterotypes. The proportion of Rikenellaceae_RC9_gut_group - and Prevotella -predominated enterotype 3 was higher than that of the other two enterotypes in all samples. Enterotype 1, distinguished by norank Muribaculaceae , was mainly detected during the warm season and at high altitudes, whereas enterotype 2, which is unclassified Lachnospiraceae , was mainly detected during the cold season. Plateau pikas with bacterial enterotypes 2 and 3 showed significantly higher alpha diversity than those with enterotype 1, while pikas with fungal enterotype 1 displayed higher diversity than those with enterotype 2. Functional pathways of lipid metabolism were enriched in bacterial enterotype 1, carbohydrate and amino acid metabolism were enriched in enterotype 2, and energy metabolism was enriched in enterotype 3. The neutral process was more important in bacterial enterotype 3 than in enterotypes 1 and 2. The body temperature and resting metabolic rate of pikas with bacterial enterotypes 1 and 3 were significantly higher than those with enterotype 2. Pikas with unclassified Sporormiaceae - and Sporormiella -dominated fungal enterotypes may engage more frequently in coprophagy. Overall, this study revealed that both gut bacterial and fungal communities in pikas can be clustered into enterotypes, and individuals with different enterotypes showed significant differences in thermogenesis. IMPORTANCE The gut microbiotas of small mammals play an important role in host energy homeostasis. However, it is still unknown whether small mammals with different enterotypes show differences in thermogenesis characteristics. Our study confirmed that plateau pikas with different bacterial enterotypes harbored distinct thermogenesis capabilities and employed various strategies against cold environments. Additionally, we also found that pikas with different fungal enterotypes may display differences in coprophagy.

Empirical Correlates for the Minnesota Multiphasic Personality Inventory-2-Restructured Form in a German Inpatient Sample

Abstract:We identified empirical correlates for the 42 substantive scales of the German language version of the Minnesota Multiphasic Personality Inventory (MMPI)-2-Restructured Form (MMPI-2-RF): Higher Order, Restructured Clinical, Specific Problem, Interest, and revised Personality Psychopathology Five scales. We collected external validity data by means of a 177-item chart review form in a sample of 488 psychiatric inpatients of a German university hospital. We structured our findings along the interpretational guidelines for the MMPI-2-RF and compared them with the validity data published in the tables of the MMPI-2-RF Technical Manual. Our results show significant correlations between MMPI-2-RF scales and conceptually relevant criteria. Most of the results were in line with U.S. validation studies. Some of the differences could be attributed to sample compositions. For most of the scales, construct validity coefficients were acceptable. Taken together, this study amplifies the enlarging body of research on empirical correlates of the MMPI-2-RF scales in a new sample. The study suggests that the interpretations given in the MMPI-2-RF manual may be generalizable to the German language MMPI-2-RF.

Abstract:Hypertrophic herpes simplex genitalis is an atypical presentation of genital herpes described in the context of immunosuppression, particularly HIV‐positive patients. This situation can become a diagnostic and therapeutic challenge. For this reason, alternative therapies are currently being discussed in the literature. We report a case of hypertrophic genital herpes in a HIV‐positive patient who was successfully treated with topical 5% imiquimod after treatment failures with oral and i.v. antivirals.

Abstract:Intestinal parasites, such as Eimeria , are common among plateau pika ( Ochotona curzoniae ). The gut microbiome is an essential driver of the host response to gastrointestinal parasites. However, the effects of intestinal protozoal parasites on the temporal variations in the gut microbiome and behavioral and physiological activities remain unknown. Our study conducted treatments involving experimental feeding of pika with Eimeria oocysts or anticoccidia under laboratory conditions to focus on the parasite-associated alterations in gut bacterial communities, host behavioral activity, physiology, and host–bacteria relationships. The results showed insignificant differences in bacterial community structures among treatments on the basis of Bray–Curtis distance metrics, whereas the patterns of temporal alterations in the bacterial communities were changed by the treatments. Bacterial alpha diversities did not vary with the treatments, and experimental feeding with Eimeria slowed down the decrement rate of alpha diversity. Furthermore, few bacterial members were significantly changed by the treatments—only the genus Ruminococcus and the species Ruminococcus flavefaciens , which were associated with energy metabolism. Experimental feeding with Eimeria modified the temporal variations in the bacterial members, including a lower loss rate of the relative abundance of the dominant families Muribaculaceae and Ruminococcaceae in the group with Eimeria experimental feeding. Moreover, a shifting energy trade-off was suggested by the parasite-induced increments in thyroid hormones (triiodothyronine and tetraiodothyronine) and decrements in exploration behavior in the group with Eimeria feeding. However, we did not detect specific connections between gut bacterial communities and pika behaviors and physiology in terms of energy trade-offs. Further in-depth research is needed to examine the role of Eimeria -modified differences in the gut bacteria of plateau pika.

Abstract:Burrowing animals are a critical driver of terrestrial ecosystem functioning, but we know little about their effects on soil microbiomes. Here, we evaluated the effect of burrowing animals on microbial assembly processes and co-occurrence patterns using soil microbiota from a group of habitats disturbed by Plateau pikas (Ochtona curzoniae). Pika disturbance had different impacts on bacterial and fungal communities. Fungal diversity generally increased with patch area, whereas bacterial diversity decreased. These strikingly different species–area relationships were closely associated with their community assembly mechanisms. The loss of bacterial diversity on larger patches was largely driven by deterministic processes, mainly due to the decline of nutrient supply (e.g., organic C, inorganic N). In contrast, fungal distribution was driven primarily by stochastic processes that dispersal limitation contributed to their higher fungal diversity on lager patches. A bacterial co-occurrence network exhibited a positive relationship of nodes and linkage numbers with patch area, and the fungal network presented a positive modularity–area relationship, suggesting that bacteria tended to form a closer association community under pika disturbance, while fungi tended to construct a higher modularity network. Our results suggest that pikas affects the microbial assembly process and co-occurrence patterns in alpine environments, thereby enhancing the current understanding of microbial biogeography under natural disturbances.

Abstract:Antibiotic residues from veterinary medicines have increased the dissemination of antibiotic resistance genes (ARGs), leading to the enrichment of ARGs in wildlife. We found livestock grazing significantly altered the gut microbiota and resistome profile of plateau pika, with the enrichment of colistin (MCR) and streptogramin (vat) resistance genes. Our results hint that plateau pika is a potential reservoir of new antibiotic resistance genes. With the overuse of antibiotics in health care and animal husbandry, antibiotic resistance becomes a serious threat to public health. Antibiotic residues from veterinary medicine have increased the dissemination of antibiotic resistance genes (ARGs) by horizontal gene transfer globally, leading to the enrichment of ARGs in wildlife. Plateau pika (Ochotona curzoniae) is a small herbivore endemic to the Qinghai–Tibetan Plateau. Previous studies reveal that pika evolves a coprophagy behavior toward cohabitated yak, which makes the pika population a potential reservoir of ARGs. Yet, little is known about the resistome of pika under different grazing intensities. Here, we sampled the cecum content of pika from three different grazing intensity areas in the Qinghai–Tibetan Plateau to evaluate the effect of grazing on its gut microbiota and resistome. By using the 16S full‐length amplicon and metagenomic sequencing, our study revealed that livestock grazing significantly altered the gut microbial community of plateau pika as compared to prohibited grazing areas. We found bacterial lineage Prevotellaceae, Lachnospirales, and RF39 increased in grazing areas. Analysis of the resistome revealed that pika from continuous grazing areas enriched a higher abundance of colistin (MCR) and streptogramin (vat) resistance genes. Moreover, we observed significant correlations between the gut microbial community, ARGs, and mobile genetic element profiles, hinting that pika gut microbiota was an important shaping force of the resistome. In future studies, the continuous monitoring of wildlife gut resistome and environmental antibiotic residues is imperative for a better understanding and for tackling the horizontal gene transfer of ARGs across the wildlife–livestock interface.

Abstract:This study investigated the relationship between the gut bacterial community and the phylogeny of five wild ungulates (Pseudois nayaur, Pantholops hodgsonii, Gazella subgutturosa, Bos mutus, and Equus kiang) inhabiting the Qinghai‐Tibetan Plateau using 16S rRNA gene data. The results of UPGMA and HCA were inconsistent with those of IQ‐TREE, indicating that host phylogeny is not the only factor influencing the gut microbiota, and other factors, such as diet and habitat, may also have an impact. There were significant differences in gut bacterial composition among the five wild ungulates, and each species had distinctive bacterial features, primarily associated with fiber and cellulose breakdown, contributing to gastrointestinal homeostasis. Several studies have investigated the gut bacterial composition of wild ungulates in the Qinghai–Xizang Plateau. However, the relationship between their gut microbiome dendrograms and their phylogenetic tree remains unclear. In this study, we analyzed 45 amplicons (V3–V4 region of the 16S rRNA gene) from five wild ungulates—Pseudois nayaur, Pantholops hodgsonii, Gazella subgutturosa, Bos grunniens, and Equus kiang—from the Qinghai–Xizang Plateau to clarify the relationship between their phylogenies and gut microbiome dendrograms. The unweighted pair group method with arithmetic mean analysis and hierarchical clustering analysis indicated that G. subgutturosa is closely related to P. nayaur; however, these results were inconsistent with their phylogenetic trees. Additionally, the indicator genera in the microbiome of each wild ungulate showed strong associations with the diets and habitats of their host. Thus, diet and space niche differentiation may primarily account for the differences between the gut microbiome characteristics of these wild ungulates and their phylogeny. In summary, our research provides insights into the evolutionary factors influencing the gut microbiome of wild ungulates in the Qinghai–Xizang Plateau.

Abstract:Plateau pikas ( Ochotona curzoniae ) are high-altitude model animals and famous “ecosystem engineers” on the Qinghai-Tibet Plateau. Pika activities may accelerate the degradation of alpine meadows. Nevertheless, little is known about the responses of bacterial, fungal, and archaeal communities, and ecosystem multifunctionality to pika perturbations. To address this question, we studied the impacts of only pika disturbance and combined disturbance (pika disturbance and grazing) on ecological networks of soil microbial communities and ecosystem multifunctionality. Our results demonstrated that Proteobacteria, Ascomycota, and Crenarchaeota were dominant in bacteria, fungi, and archaea, respectively. Bacteria, fungi, and archaea were all influenced by the combined disturbance of grazing and pika. Most fungal communities became convergent, while bacterial and archaeal communities became differentiated during the succession of surface types. In particular, the bacterial and fungal networks were less stable than archaeal networks. In response to the interference, cross-domain cooperation between bacterial and fungal communities increased, while competitive interactions between bacterial and archaeal communities increased. Pika disturbance at high intensity significantly reduced the ecosystem multifunctionality. However, the mixed effects of grazing and pika weakened such influences. This study revealed how pika activities affected microbial networks and ecosystem multifunctionality. These results provide insights to designing reasonable ecological management strategies for alpine grassland ecosystems.

Abstract:Besides overgrazing by livestock, browsing by small native mammals, such as plateau pikas, is considered an important cause of alpine grassland degradation on the Qinghai-Tibetan Plateau (QTP). Preference of pikas for degraded grasslands, resulting in increased population density, has been proposed as the most accepted hypothesis. However, few studies have explored the mechanisms underlying the habitat preferences of pikas. Herein, we analyzed aboveground biomass, community composition, forage quality of lightly and severely degraded alpine grasslands, and the diet and nutritional intake of pikas living there to test whether the plant community structure and associated nutrient composition of severely degraded grasslands are more suitable for pikas in terms of dietary and nutritional requirements. Our results showed that the plant community structure differed between lightly and severely degraded grasslands, with lightly degraded grasslands containing more food plants preferred by pikas. The nutritional composition of plants in severely degraded grasslands was less suitable for pikas than that in lightly degraded grasslands. Our results reject the nutritional hypothesis that severely degraded grasslands meet the dietary and nutritional needs of pikas better. We conclude that food and nutrient selection are not drivers of population increase in pikas in the severely degraded grasslands of the QTP.
