Abstract:The emergence of orientation selectivity in the visual cortex is a well-known phenomenon in neuroscience, but the details of such emergence and the role of different cortical layers and cell types, particularly in rodents which lack a topographical organization of orientation-selectivity (OS) properties, are less clear. To tackle this question, we use an existing biologically detailed model of the mouse V1 cortical column, which is constrained by existing connectivity data across cortical layers and between pyramidal, PV, SST and VIP cell types. Using this model as a basis, we implemented activity-dependent structural plasticity induced by stimulation with orientated drifting gratings, leading to a good match of tuning properties of pyramidal cells with experimentally observed OS laminar distribution, their evoked firing rate and tuning width. We then employed a mean-field model to uncover the role of co-tuned subnetworks in laminar signal propagation and explain the effects of intra- and inter- laminar coupling distributions. Our plasticity-induced modified model and mean-field model were able to explain both the excitatory enhancement through co-tuned subnetworks and inter-laminar disynaptic inhibition. Overall, our work highlights the importance of the clustering of neural selectivity features for effective excitatory transmission in cortical circuits.

Superior colliculus projections drive dopamine neuron activity and movement but not value

Abstract:To navigate complex environments, animals must rapidly integrate sensory information and respond appropriately to gather rewards and avoid threats. It is well established that dopamine neurons in the ventral tegmental area (VTA) and substantia nigra pars compacta (SNc) are key for creating and maintaining associations between environmental stimuli (i.e., cues) and the outcomes they predict, through Pavlovian learning. However, it remains unclear how relevant sensory information is integrated into dopamine (DA) pathways to guide exploration and learning. The superior colliculus (SC) receives direct visual input and is anatomically positioned as a relay for rapid sensory augmentation of dopamine neurons, which could underlie the formation of Pavlovian associations. Here, we characterize the anatomical organization and functional impact of SC projections to the VTA and SNc in rats. First, using anatomical tracing techniques, we show that neurons in the intermediate and deep layers of SC synapse densely throughout the ventral midbrain, interfacing directly with neurons projecting to the striatum and ventral pallidum. Using fiber photometry, we find that these SC projections excite both dopamine and GABA neurons in the VTA and dopamine neurons in the SNc in vivo. Despite this, cues predicting SC terminal stimulation did not reliably evoke behavior on their own in an optogenetic Pavlovian conditioning paradigm. Further, optogenetic activation of SC terminals in the VTA/SNc did not support primary reinforcement or produce place preference or avoidance. Instead, we find that stimulation of SC terminals in the VTA and SNc reliably evoked head turning behavior. This body reorientation increased in intensity with repeated stimulations, suggesting that strengthening this circuit could underlie sensorimotor learning related to exploration and attentional bias. Together our results show that collicular neurons contribute to cue-guided learning by controlling pose adjustments through interaction with dopamine systems.

Excitatory and inhibitory intracortical circuits for orientation and direction selectivity

Abstract:The computations performed by a neuron arise from the functional properties of the circuits providing its synaptic inputs. A prime example of these computations is the selectivity of primary visual cortex (V1) for orientation and motion direction. V1 neurons in layer 2/3 (L2/3) receive input mostly from intracortical circuits 1 , which involve excitation 2-9 and inhibition 10-12 . To understand how an L2/3 neuron achieves its selectivity, therefore, one must characterize the functional organization of both its excitatory and inhibitory presynaptic ensembles. Here we establish this organization, and show how it predicts orientation selectivity and reveals a new cortical circuit for direction selectivity. We identified the presynaptic partners of pyramidal neurons in mouse V1 through rabies monosynaptic tracing 1,13 , and imaged the functional properties of the postsynaptic neuron and of its presynaptic ensemble. Excitatory presynaptic neurons were predominantly tuned to the postsynaptic neuron’s preferred orientation. Excitation and inhibition described an inverted Mexican hat, with inhibitory presynaptic neurons densest near the postsynaptic neuron and excitatory ones distributed more distally. Excitation and inhibition also differed in laminar origin: inhibitory presynaptic neurons concentrated in L2/3 while excitatory ones dominated in L4. The distribution of excitatory neurons in visual space was coaxial with the postsynaptic neuron’s preferred orientation and lay upstream of the neuron’s preferred direction. Inhibitory presynaptic neurons, instead, clustered more symmetrically around the postsynaptic neuron and favoured locations downstream of its preferred direction. These results demonstrate that L2/3 neurons obtain orientation selectivity from co-tuned neurons in L4 and beyond, and enhance it by contrasting an elongated excitatory input with a concentric inhibitory input. Moreover, L2/3 neurons can obtain direction selectivity through visually offset 14 excitation and inhibition. These circuit motifs resemble those seen in the thalamocortical pathway 15-20 and in direction selective cells in the retina 21,22 , suggesting that they are canonical across brain regions.
Orientation-selective Responses in the Mouse Lateral Geniculate Nucleus

Abstract:The dorsal lateral geniculate nucleus (dLGN) receives visual information from the retina and transmits it to the cortex. In this study, we made extracellular recordings in the dLGN of both anesthetized and awake mice, and found that a surprisingly high proportion of cells were selective for stimulus orientation. The orientation selectivity of dLGN cells was unchanged after silencing the visual cortex pharmacologically, indicating that it is not due to cortical feedback. The orientation tuning of some dLGN cells correlated with their elongated receptive fields, while in others orientation selectivity was observed despite the fact that their receptive fields were circular, suggesting that their retinal input might already be orientation selective. Consistently, we revealed orientation/axis-selective ganglion cells in the mouse retina using multielectrode arrays in an in vitro preparation. Furthermore, the orientation tuning of dLGN cells was largely maintained at different stimulus contrasts, which could be sufficiently explained by a simple linear feedforward model. We also compared the degree of orientation selectivity in different visual structures under the same recording condition. Compared with the dLGN, orientation selectivity is greatly improved in the visual cortex, but is similar in the superior colliculus, another major retinal target. Together, our results demonstrate prominent orientation selectivity in the mouse dLGN, which may potentially contribute to visual processing in the cortex.

Abstract:The brain monitors the sensory environment via signals from the sensory periphery, such as the olfactory epithelium, the inner ear, and the retina. Understanding how sensory stimuli are processed throughout the sensory hierarchy, and how this relates to behavior, is a central outstanding question in the field of neuroscience. The processing of visual motion in mice offers unique opportunities for addressing these questions thanks to a rich literature on the anatomical and physiological properties of motion-sensitive neurons across the visual system, paired with recent developments of cutting-edge genetic and imaging approaches. A visual scene typically contains motion originating from either moving objects or optic flow caused by self-generated movements. Neurons encoding the direction of visual motion are said to be 'direction-selective'. It was historically believed the circuits responsible for creating direction selectivity <em>de novo</em> exist within the visual cortex. Yet, in mice, direction-selective responses can be found already in the retina, suggesting in this model organism visual motion analysis starts at the earliest stage of the visual hierarchy. This minireview presents emerging literature demonstrating how retinal direction-selective cells make causal contributions to central visual motion processing and visually guided behaviors in mice, and their potential clinical relevance, and outlines experiments for testing remaining questions. Research in this field will undoubtedly continue to advance our understanding of the basic principles of the visual system and how sensory neurons extract fundamental features of the world.

Neurotensin-specific corticothalamic circuit regulates innate response conflict

Abstract:Animals must simultaneously select and balance multiple action contingencies in ambiguous situations: for instance, evading danger during feeding. This has rarely been examined in the context of information selection; despite corticothalamic pathways that mediate sensory attention being relatively well characterized, neural mechanisms filtering conflicting actions remain unclear. Here, we develop a new loom/feed test to observe conflict between naturally induced fear and feeding and identify a novel anterior cingulate cortex (ACC) output to the ventral anterior and ventral lateral thalamus (VA/VL) that adjusts selectivity between these innate actions. Using micro-endoscopy and fiber photometry, we reveal that activity in corticofugal outputs was lowered during unbalanced/singularly occupied periods, as were the resulting decreased thalamic initiation-related signals for less-favored actions, suggesting that the integration of ACC-thalamic firing may directly regulate the output of behavior choices. Accordingly, the optoinhibition of ACC-VA/VL circuits induced high bias toward feeding at the expense of defense. To identify upstream "commander" cortical cells gating this output, we established dual-order tracing (DOT)-translating ribosome affinity purification (TRAP)-a scheme to label upstream neurons with transcriptome analysis-and found a novel population of neurotensin-positive interneurons (ACCNts). The photoexcitation of ACCNts cells indeed caused similarly hyper-selective behaviors. Collectively, this new "corticofugal action filter" scheme suggests that communication in multi-step cingulate circuits may critically influence the summation of motor signals in thalamic outputs, regulating bias between innate action types.

Abstract:Classic ON-OFF direction-selective ganglion cells (DSGCs) that encode the four cardinal directions were recently shown to also be orientation-selective. To clarify the mechanisms underlying orientation selectivity, we employed a variety of electrophysiological, optogenetic, and gene knock-out strategies to test the relative contributions of glutamate, GABA, and acetylcholine (ACh) input that are known to drive DSGCs, in male and female mouse retinas. Extracellular spike recordings revealed that DSGCs respond preferentially to either vertical or horizontal bars, those that are perpendicular to their preferred-null motion axes. By contrast, the glutamate input to all four DSGC types measured using whole-cell patch-clamp techniques was found to be tuned along the vertical axis. Tuned glutamatergic excitation was heavily reliant on type 5A bipolar cells, which appear to be electrically coupled via connexin 36 containing gap junctions to the vertically oriented processes of wide-field amacrine cells. Vertically tuned inputs are transformed by the GABAergic/cholinergic "starburst" amacrine cells (SACs), which are critical components of the direction-selective circuit, into distinct patterns of inhibition and excitation. Feed-forward SAC inhibition appears to "veto" preferred orientation glutamate excitation in dorsal/ventral (but not nasal/temporal) coding DSGCs "flipping" their orientation tuning by 90° and accounts for the apparent mismatch between glutamate input tuning and the DSGC's spiking response. Together, these results reveal how two distinct synaptic motifs interact to generate complex feature selectivity, shedding light on the intricate circuitry that underlies visual processing in the retina.
Target dependence of orientation and direction selectivity of corticocortical projection neurons in the mouse V1

Abstract:Higher order visual areas that receive input from the primary visual cortex (V1) are specialized for the processing of distinct features of visual information. However, it is still incompletely understood how this functional specialization is acquired. Here we used in vivo two photon calcium imaging in the mouse visual cortex to investigate whether this functional distinction exists at as early as the level of projections from V1 to two higher order visual areas, AL and LM. Specifically, we examined whether sharpness of orientation and direction selectivity and optimal spatial and temporal frequency of projection neurons from V1 to higher order visual areas match with that of target areas. We found that the V1 input to higher order visual areas were indeed functionally distinct: AL preferentially received inputs from V1 that were more orientation and direction selective and tuned for lower spatial frequency compared to projection of V1 to LM, consistent with functional differences between AL and LM. The present findings suggest that selective projections from V1 to higher order visual areas initiates parallel processing of sensory information in the visual cortical network.

Abstract:In contrast to neurons of the lateral geniculate nucleus (LGN), neurons in the primary visual cortex (V1) are selective for the direction of visual motion. Cortical direction selectivity could emerge from the spatiotemporal configuration of inputs from thalamic cells, from intracortical inhibitory interactions, or from a combination of thalamic and intracortical interactions. To distinguish between these possibilities, we studied the effect of adaptation (prolonged visual stimulation) on the direction selectivity of intracellularly recorded cortical neurons. It is known that adaptation selectively reduces the responses of cortical neurons, while largely sparing the afferent LGN input. Adaptation can therefore be used as a tool to dissect the relative contribution of afferent and intracortical interactions to the generation of direction selectivity. In both simple and complex cells, adaptation caused a hyperpolarization of the resting membrane potential (−2.5 mV, simple cells, −0.95 mV complex cells). In simple cells, adaptation in either direction only slightly reduced the visually evoked depolarization; this reduction was similar for preferred and null directions. In complex cells, adaptation strongly reduced visual responses in a direction-dependent manner: the reduction was largest when the stimulus direction matched that of the adapting motion. As a result, adaptation caused changes in the direction selectivity of complex cells: direction selectivity was reduced after preferred direction adaptation and increased after null direction adaptation. Because adaptation in the null direction enhanced direction selectivity rather than reduced it, it seems unlikely that inhibition from the null direction is the primary mechanism for creating direction selectivity.

Abstract:Primary visual cortex (V1) provides crucial insights into the selectivity and emergence of specific output features such as orientation tuning. Tuning and selectivity of cortical neurons in mouse visual cortex is not equivocally resolved so far. While many in-vivo experimental studies found inhibitory neurons of all subtypes to be broadly tuned for orientation other studies report inhibitory neurons that are as sharply tuned as excitatory neurons. These diverging findings about the selectivity of excitatory and inhibitory cortical neurons prompted us to ask the following questions: (1) How different or similar is the cortical computation with that in previously described species that relies on map? (2) What is the network mechanism underlying the sharpening of orientation selectivity in the mouse primary visual cortex? Here, we investigate the above questions in a computational framework with a recurrent network composed of Hodgkin-Huxley (HH) point neurons. Our cortical network with random connectivity alone could not account for all the experimental observations, which led us to hypothesize, (a) Orientation dependent connectivity (b) Feedforward afferent specificity to understand orientation selectivity of V1 neurons in mouse. Using population (orientation selectivity index) OSI as a measure of neuronal selectivity to stimulus orientation we test each hypothesis separately and in combination against experimental data. Based on our analysis of orientation selectivity (OS) data we find a good fit of network parameters in a model based on afferent specificity and connectivity that scales with feature similarity. We conclude that this particular model class best supports data sets of orientation selectivity of excitatory and inhibitory neurons in layer 2/3 of primary visual cortex of mouse.

Abstract:Appetitive locomotion is essential for animals to approach rewards, such as food and prey. The neuronal circuitry controlling appetitive locomotion is unclear. In a goal-directed behavior-predatory hunting, we show an excitatory brain circuit from the superior colliculus (SC) to the substantia nigra pars compacta (SNc) to enhance appetitive locomotion in mice. This tectonigral pathway transmits locomotion-speed signals to dopamine neurons and triggers dopamine release in the dorsal striatum. Synaptic inactivation of this pathway impairs appetitive locomotion but not defensive locomotion. Conversely, activation of this pathway increases the speed and frequency of approach during predatory hunting, an effect that depends on the activities of SNc dopamine neurons. Together, these data reveal that the SC regulates locomotion-speed signals to SNc dopamine neurons to enhance appetitive locomotion in mice.
Defined cell types in superior colliculus make distinct contributions to prey capture behavior in the mouse

Abstract:Summary The superior colliculus (SC) mediates rapid orienting to visual stimuli across species. To determine the specific circuits within the SC that drive orienting and approach behavior toward appetitive stimuli, we explored the role of three genetically defined cell types in mediating prey capture in mice. Chemogenetic inactivation of two classically defined cell types, the wide-field (WF) and narrow-field (NF) vertical neurons, revealed that they are involved in distinct aspects of prey capture. WF neurons were required for rapid prey detection and distant approach initiation, whereas NF neurons were required for continuous and accurate orienting during pursuit. In contrast, prey capture did not require parvalbumin-expressing (PV) neurons that have previously been implicated in fear responses. The visual coding of WF and NF cells in the awake mouse and their projection targets were consistent with their roles in prey detection versus pursuit. Thus, our studies link specific neural circuit connectivity and function with stimulus detection and orienting behavior, providing insight into visuomotor and attentional mechanisms mediated by superior colliculus. Highlights This study provides the first demonstration of the role of specific cell populations in the superior colliculus in orienting and approach behavior. A genetically targeted population of wide-field vertical neurons in the superior colliculus is required for rapid prey detection and initiation of long-distance approaches. A genetically targeted population of narrow-field vertical neurons is required for approach initiation, accurate targeting, and approach continuity. Visual response properties and projection targets of these cells are consistent with their role in prey capture, linking neural circuit connectivity and function with behavior.
Cortical Feedback Modulates Distinct Critical Period Development in Mouse Visual Thalamus

Abstract:In primary visual cortex (V1), critical period for ocular dominance (OD) plasticity is a well-defined developmental stage to shape neuronal circuits based on visual experience. Recent studies showed that V1-like OD plasticity existed in mouse dorsal lateral geniculate nucleus (dLGN). It is still unclear what the exact time win-dow is and how neural circuits contribute to OD plasticity in dLGN. Using in vivo electrophysiology, we defined a critical period for OD plasticity in dLGN from eye opening to puberty. There also existed an innate process of OD formation from contralateral to equal bias in dLGN binocular neurons. Instant V1 inactivation with muscimol had no effect on OD bias or plasticity. Short-term V1 inactivation with N-methyl-d-aspartate reversed the formation of equal OD bias, while long-term V1 inactivation retained dLGN development to an immature stage.
Functional Dynamics and Selectivity of Two Parallel Corticocortical Pathways from Motor Cortex to Layer 5 Circuits in Somatosensory Cortex

Abstract:In the rodent whisker system, active sensing and sensorimotor integration are mediated in part by the dynamic interactions between the motor cortex (M1) and somatosensory cortex (S1). However, understanding these dynamic interactions requires knowledge about the synapses and how specific neurons respond to their input. Here, we combined optogenetics, retrograde labeling, and electrophysiology to characterize the synaptic connections between M1 and layer 5 (L5) intratelencephalic (IT) and pyramidal tract (PT) neurons in S1 of mice (both sexes). We found that M1 synapses onto IT cells displayed modest short-term depression, whereas synapses onto PT neurons showed robust short-term facilitation. Despite M1 inputs to IT cells depressing, their slower kinetics resulted in summation and a response that increased during short trains. In contrast, summation was minimal in PT neurons due to the fast time course of their M1 responses. The functional consequences of this reduced summation, however, were outweighed by the strong facilitation at these M1 synapses, resulting in larger response amplitudes in PT neurons than IT cells during repetitive stimulation. To understand the impact of facilitating M1 inputs on PT output, we paired trains of inputs with single backpropagating action potentials, finding that repetitive M1 activation increased the probability of bursts in PT cells without impacting the time dependence of this coupling. Thus, there are two parallel but dynamically distinct systems of M1 synaptic excitation in L5 of S1, each defined by the short-term dynamics of its synapses, the class of postsynaptic neurons, and how the neurons respond to those inputs.

Dynamic control of visually guided locomotion through corticosubthalamic projections

Abstract:Goal-directed locomotion requires control signals that propagate from higher order areas to regulate spinal mechanisms. The corticosubthalamic hyperdirect pathway offers a short route for cortical information to reach locomotor centers in the brainstem. We developed a task in which head-fixed mice run to a visual landmark and then stop and wait to collect the reward and examined the role of secondary motor cortex (M2) projections to the subthalamic nucleus (STN) in controlling locomotion. Our behavioral modeling, calcium imaging, and optogenetics manipulation results suggest that the M2-STN pathway can be recruited during visually guided locomotion to rapidly and precisely control the pedunculopontine nucleus (PPN) of the mesencephalic locomotor region through the basal ganglia. By capturing the physiological dynamics through a feedback control model and analyzing neuronal signals in M2, PPN, and STN, we find that the corticosubthalamic projections potentially control PPN activity by differentiating an M2 error signal to ensure fast input-output dynamics.
The Emergence of Directional Selectivity in the Visual Motion Pathway of Drosophila

Abstract:The perception of visual motion is critical for animal navigation, and flies are a prominent model system for exploring this neural computation. In Drosophila, the T4 cells of the medulla are directionally selective and necessary for ON motion behavioral responses. To examine the emergence of directional selectivity, we developed genetic driver lines for the neuron types with the most synapses onto T4 cells. Using calcium imaging, we found that these neuron types are not directionally selective and that selectivity arises in the T4 dendrites. By silencing each input neuron type, we identified which neurons are necessary for T4 directional selectivity and ON motion behavioral responses. We then determined the sign of the connections between these neurons and T4 cells using neuronal photoactivation. Our results indicate a computational architecture for motion detection that is a hybrid of classic theoretical models.

Abstract:Sensory neurons are modulated by context. For example, in mouse primary visual cortex (V1), neuronal responses to the preferred orientation are modulated by the presence of superimposed orientations ("plaids"). The effects of this modulation are diverse; some neurons are suppressed, while others have larger responses to a plaid than its components. We investigated whether this diversity could be explained by a unified circuit mechanism. We report that this masking is maintained during suppression of cortical activity, arguing against cortical mechanisms. Instead, the heterogeneity of plaid responses is explained by an interaction between stimulus geometry and orientation tuning. Highly selective neurons are uniformly suppressed by plaids, whereas the effects in weakly selective neurons depend on the spatial configuration of the stimulus, transitioning systematically between suppression and facilitation. Thus, the diverse responses emerge as a consequence of the spatial structure of feedforward inputs, with no need to invoke cortical interactions.
