Abstract:Benzo(α)pyrene (BaP) and lead (Pb) are common pollutants discharged greatly in ocean and causing detrimental impacts on marine organisms. Although mussels are one of the most prominent and frequently studied biological models, the research on their genomic alterations induced by the mixture of two totally different chemicals, is still rare. In present study, local marine mussels Mytilus coruscus were exposed in vivo to BaP (53.74 ± 19.79 μg/L), Pb (2.58 ± 0.11 mg/L) and their mixture for 6 days. The genotoxic damages were assessed by comet assay, micronucleus (MNi) test, and random amplified polymorphic DNA (RAPD) analysis. Significantly increased though transitory genomic damage was investigated after the exposure and showed consistency using various detecting methods. Additive genotoxicity was only found after 3 days combined exposure by means of MNi test, suggesting that BaP and Pb may play with alternative biological targets during metabolism and/or interaction with the genome. The geno-stability and the recovery capability were further detected both in vivo and in vitro after challenged by BaP. RAPD results showed coherence in BaP induced genotoxicity, together with time-specific alterations. The genomic instability was found to recover in both in vivo and in vitro exposure scenarios in present study. To our knowledge, this is the first study to focus on the genotoxicitiy induced by BaP, Pb and their mixture by multiple detecting techniques. The attempt to utilize model pollutants and marine organism to validate the potential value of RAPD analysis highlighted that it might be a useful tool in the research of genotoxicology, especially on the effect-mechanism interplay at genetic level.

Abstract:The aim of this study was to investigate the bioaccumulation and oxidative damage of Manila clam, Ruditapes philippinarum, exposed to four selected mixtures of polycyclic aromatic hydrocarbons (PAHs; benzo (a) pyrene (BaP), benzo (a) anthracene (BaA), benzo (b) fluoranthene (BbF), and chrysene (Chr) in equal proportion. For this purpose, clams were exposed to PAHs (BaP:BbF:BaA:Chr = 1:1:1:1) at different concentrations (0.05, 0.5, and 5 μg/L) for 21 days, followed by a 15-day depuration period. All four PAHs accumulated in the gill, digestive gland, adductor muscle, and soft tissue of Manila clams, and all PAH treatment groups showed clear time and dose dependence. The decreasing order of bioaccumulation for the four PAHs in the exposure experiment was Chr > BaA > BaP > BbF. Moreover, the order of PAH bioaccumulation for the four tissues during the whole experiment was digestive gland > gill > soft tissues > adductor muscles. Although the initial concentrations of the four PAHs were the same, the final accumulated contents were different. Therefore, we also determined the detoxification processes of the four PAH mixtures in gills and digestive glands. The bioaccumulation of Chr was higher than the other three PAHs, probably because clams have a lower metabolic capacity for Chr than for BaP, BbF, and BaA. Exposure to PAH mixtures can result in oxidative damage, as indicated by the fact that DNA strand breaks, lipid peroxidation (LPO), and protein carbonyl (PC) were induced significantly (P < 0.05), except in the low-dose groups of PAHs, and different trends were detected with time of exposure. According to the correlation analysis, aryl hydrocarbon hydroxylase, glutathione s-transferase, superoxide dismutase, DNA strand break, PC, and LPO in both the gill and digestive gland are potential early indicators of PAH mixtures. We investigated the accumulation rules of R. philippinarum exposed to the selected PAHs and screened the potential biomarkers. The results of our study provide important scientific information for the purpose of monitoring marine pollution.
BaP-metals Co-Exposure Induced Tissue-Specific Antioxidant Defense in Marine Mussels Mytilus Coruscus.

Abstract:Both benzo(α)pyrene (BaP) and metals are frequently found in marine ecosystem and can cause detrimental effects in marine organism, especially the filter feeder-marine mussels. Although the biological responses in mussels have been well-studied upon the single metal or BaP exposure, the information about antioxidant defense, especially in different tissues of mussels, are still limited. Considering the variety of contaminants existing in the actual marine environment, single BaP (56 μg/L) and the co-exposure with Cu, Cd and Pb (50 μg/L, 50 μg/L and 3 mg/L respectively) were applied in a 6 days exposure followed by 6 days depuration experiment. The alterations of superoxide dismutase (SOD), catalase (CAT) activities and total antioxidant capacity (TAC) level were assessed in haemolymph, gills and digestive glands of marine mussels, Mytilus coruscus. An unparalleled change in antioxidant biomarkers was observed in all cells/tissues, with the SOD activity showing higher sensitivity to exposure. A tissue-specific response showing unique alteration in gill was investigated, indicating the different function of tissues during stress responses. Depressed antioxidant effects were induced by BaP-metals co-exposure, indicating the interaction may alter the intact properties of BaP. To our knowledge, this is the first research to explore the antioxidant defense induced by combined exposure of BaP-metals regarding to tissue-specific responses in marine mussels. The results and experimental model will provide valuable information and can be utilized in the investigation of stress response mechanisms, especially in relation to tissue functions in marine organism in the future.

Abstract:Polycyclic aromatic hydrocarbons (PAHs) are known for their carcinogenic, teratogenic and mutagenic properties. Benzo(a)pyrene (BaP) possesses the greatest carcinogenic potential among the various PAHs. In this study, digital gene expression (DGE) was performed to investigate the digestive gland transcriptome profile of the clam Ruditapes philippinarum exposed to BaP. A total of 10,508,312 and 11,414,297 clean reads were generated respectively, from control and BaP exposure DGE libraries. One hundred and forty-five differentially expressed genes were detected after comparing two libraries with 58 up-regulated and 87 down-regulated genes. GO annotation and KEGG pathway analyses were performed on all genes to understand their biological functions and processes. The results showed that numerous enriched differentially expressed genes are related to growth and development, antioxidant metabolism, apoptosis and detoxification metabolism. Quantitative real-time PCR was performed to verify the expressed genes of DGE. Our results provide evidences that RNA-seq is a powerful tool for toxicology and capable of generating novel and valuable information at the transcriptome level for characterizing deleterious effects caused by BaP.

Abstract:OBJECTIVE The study was undertaken to compare infectious morbidity and trial of labor (TOL) success stratified by weight in women. STUDY DESIGN Vaginal birth after cesarean section (VBAC) candidates were divided into groups based on prepregnancy weight: group I, 70 (<200 pounds); group II, 70 (200-300 pounds); and group III, 69 (>300 pounds). RESULTS The TOL success rate was 81.8% in group I compared with 57.1% in group II and 13.3% in group III (P =.001). The overall infectious morbidity was significantly greater in the obese women 39% (P =.001) compared with the average women at 11.4% and the lean women at 5.7%. CONCLUSION Infectious morbidity is increased and VBAC success is reduced in patients who weigh more than 300 pounds.
Transcriptomic Changes in the Ovaries of Scallop Chlamys Farreri Exposed to Benzo[a]pyrene

Abstract:In this study, digital gene expression (DGE) sequencing technology was used to identify alterations in gene transcription of the ovarian tissue of Chlamys farreri (C. farreri) exposed to benzo[a]pyrene (BaP) pollutants. A total of 124 genes were detected differentially expressed with 42 up-regulated and 82 down-regulated genes in BaP DGE libraries. Gene ontology annotation and kyoto encyclopedia of genes and genomes pathway analyses were performed on all genes to elucidate their biological functions and processes. The results showed that numerous enriched differentially expressed genes related to "detoxification process", "damage effects to extracellular matrix and cell interactions", "endocrine disrupting effects" and "tissue damage responses". Quantitative RT-PCR procedure was executed to understand the expression patterns of several important reproductive-related genes. To supplement the reproductive toxicity of BaP, histological analysis of the ovaries illustrated that after 21 days higher dose of BaP exposure, the immature oocytes and degenerated oocytes increased. The study aims to provide reference data for future study of organic pollutions in aquatic mollusks.
Potential molecular mechanism underlying the harmed haemopoiesis upon Benzo[a]pyrene exposure in Chlamys farreri

Abstract:Benzo[a]pyrene (B[a]P), a ubiquitous contamination in the marine environments, has the potential to impact the immune response of bivalves by affecting the hemocyte parameters, especially total hemocyte count (THC). THC is mainly determined by haematopoietic mechanisms and apoptosis of hemocytes. Many studies have found that B[a]P can influence the proliferation and differentiation of hemocytes. However, the link between the toxic mechanisms of haematopoietic and environmental pollutants is not explicitly stated. This study is to investigate the toxic effects of B[a]P on haematopoietic mechanisms in C. farreri. Through the tissue expression distribution experiment and EDU assay, gill is identified as a potential haematopoietic tissue in C. farreri. Subsequently, the scallops were exposed to B[a]P (0.05, 0.5, 5 μg/L) for 1d, 3d, 6d, 10d and 15d. Then BPDE content, DNA damage, gene expression of haematopoietic factors and haematopoietic related pathways were determined in gill and hemocytes. The results showed that the expression of CDK2 was significantly decreased under B[a]P exposure through three pathways: RYR/IP3-calcium, BPDE-CHK1 and Notch pathway, resulting in cell cycle arrest. In addition, B[a]P also significantly reduced the number of proliferating hemocytes by affecting the Wnt pathway. Meanwhile, B[a]P can significantly increase the content of ROS, causing a downregulation of FOXO gene expression. The gene expression of Notch pathway and ERK pathway was also detected. The present study suggested that B[a]P disturbed differentiation by multiple pathways. Furthermore, the expression of SOX11 and CD9 were significantly decreased, which directly indicated that differentiation of hemocytes was disturbed. In addition, phagocytosis, phenoloxidase activity and THC were also significant decreased. In summary, the impairment of haematopoietic activity in C. farreri further causes immunotoxicity under B[a]P exposure. This study will improve our understanding of the immunotoxicity mechanism of bivalve under B[a]P exposure.

Abstract:The gut microbiota is essential for health and physiological functions in the host organism. However, the toxicological evaluation of environmental pollutants on the gut microbiota is still insufficient. In the present study, the juvenile sea cucumber Apostichopus japonicus was exposed for 14 days to Benzo[a]pyrene (BaP), which is a model polycyclic aromatic hydrocarbon (PAH), at four different concentrations (0, 0.5, 5, and 25 μg/L). We analyzed the intestinal microbial community of A. japonicas using 16S rRNA gene amplicon sequencing. Our results demonstrate that BaP exposure caused alterations to the microbiome community composition in sea cucumbers. At the phylum level, Planctomycetes were significantly more abundant in BaP exposure groups at 14 d compared with the control group, and the abundance of Proteobacteria and Bacteroidetes increased while the abundance of Firmicutes decreased following BaP exposure. At the genus level, multiple beneficial and autochthonous genera declined in the BaP treatment groups compared to the control, including Lactococcus, Bacillus, Lactobacillus, Enterococcus, Leuconostoc and Weissella; however, a bloom of alkane-degrading bacteria was found in BaP-exposed guts and included Lutibacter, Pseudoalteromonas, Polaribacter, Rhodopirellula and Blastopirellula. Furthermore, histological morphology, enzymatic activity and gene expression analysis revealed that BaP exposure also negatively impacted gut structure and function and presented as inflammation or atrophy, oxidative stress and immune suppression in sea cucumber intestines. Collectively, these findings provide insights into the toxic effects of BaP exposure on A. japonicas associated with intestinal microbiota and health.

Abstract:Gonadal development is a prerequisite for the reproductive success of an organism, and might be affected by environmental factors such as emergent pollutants. Although marine crustaceans are threatened by ubiquitous emergent pollutants such as microplastics (MPs) and bisphenol A (BPA) under realistic scenarios, studies about the impacts of these pollutants on the gonadal development of crustacean species are rare. In this study, the effects of MPs and BPA, alone or in combination, on gonadal development were investigated in whiteleg shrimp (Litopenaeus vannamei). The results obtained demonstrated that whiteleg shrimp exposed to MPs and BPA had significantly smaller gonad-somatic index (GSI) and an obvious delay in the gonad developmental stage. In addition, exposure of whiteleg shrimp to pollutants tested resulted in a reduction in the oxygen consumption rate, elevation in the ammonia excretion rate, decline in the O: N ratio, and downregulation in the expression of metabolism-related genes, indicating significant disruptions of shrimp metabolism by the pollutants. Furthermore, in addition to a few exceptions, both the in vivo contents of gonadal development-related hormones (GIH and MIH) and the expression of genes encoding regulatory hormones (GIH, MIH, and CHH) were upregulated by the exposure of whiteleg shrimp to the pollutants investigated, suggesting a significant obstruction of endocrine regulation. Moreover, MP-BPA coexposure was shown to be more toxic to whiteleg shrimp than the corresponding single exposures and significantly greater amount of BPA accumulated in the gonads (both testis and ovaries) of shrimp with the coexistence of MPs, which may be caused by the Trojan horse effect and summation of the toxic impacts on common targets. In general, the data obtained in this study demonstrated that MPs and BPA at environmentally realistic concentrations significantly inhibited the gonadal development of whiteleg shrimp probably by interfering with metabolism and disrupting endocrine regulation.
Effects of Tributyltin and Benzo[a]pyrene on the Immune-Associated Activities of Hemocytes and Recovery Responses in the Gastropod Abalone, Haliotis Diversicolor.

Abstract:Our previous study reports that short-term exposure to sublethal concentrations of benzo[a]pyrene (BaP) induces immunomodulation in the gastropod abalone, Haliotis diversicolor. In the present study, it was further observed that long-term chronic exposure to sublethal concentrations of BaP modulated the immunocompetence of abalones in terms of the change in activity of the antioxidant and immune associated parameters tested. In addition, the effect of tributyltin (TBT), another important genotoxicant in the aquatic environment, was investigated. Exposure of abalones to sublethal concentrations of TBT and BaP for 21 days resulted in significant decrease of total hemocyte count, phagocytosis, membrane stability and lysozyme activity. Conversely induction of extra and intra cellular superoxide generation, nitric oxide, nitric oxide synthase and myeloperoxidase activity was present when the abalones were exposed to TBT and BaP. Most of the immune associated parameters tested showed clear time dependent response to both toxicants. Within 14 days after the 21 day exposure to BaP, recovery was observed as evidenced by most of the parameters returning to their normal level. However, no recovery was observed within 14 days after the 21 day exposure to TBT as evidenced by continued elevation of intra cellular superoxide and nitrite production and decrease in THC, membrane stability and lysozyme activity. This suggested a prolonged TBT-induced impact on the immune reaction and possibly more damage than that caused by BaP. Overall the results suggest that chronic exposure to sublethal concentrations of TBT or BaP causes modulations in the immunocompetence of abalones with most of the immune associated parameters tested being stimulated, and this might be harmful to the host.

Abstract:This study aims to investigate potential toxic effects of chrysene (CHR) on mature scallop Chlamys farreri during the reproduction period, using indicators of antioxidant defences and oxidative stress. Scallops were exposed to 0.2, 0.8 and 3.2μg/L waterborne CHR for 21 days, at day 10 scallops were induced to spawn. At days 1, 3, 6, 10, 11, 15 and 21, aryl hydrocarbon hydroxylase (AHH), 7-ethoxyresorufin-O-deethylase (EROD), glutathione-s-transferase (GST), glutathione (GSH), superoxide dismutase (SOD), lipid peroxidation (LPO), protein carbonyl (PC) and DNA strand breaks in digestive glands were examined by separately analysing male and female scallops. During the pre-spawn period, Levels of enzymatic activities and oxidative stress were all induced by the exposure to CHR for females and males. GST activity presented a good time- and dose-dependent relationship only in males, and GSH content showed a dose-dependent manner in both sexes. During the post-spawn period, different trends were observed, while PC contents maintained growth in time- and dose-dependent manner. Overall, males were more sensitive than females to CHR exposure in enzyme activities, and correspondingly, females suffered from more serious oxidative damages. Both GSH and PC contents seemed to be potential biomarkers for PAH exposure. These results will offer the information on toxicity of CHR in this species, and ensure the influence of gender and reproductive status on PAH detoxification metabolism.

Abstract:The study is aimed at investigating the bioaccumulation and oxidative damage of juvenile scallops (Chlamys farreri) exposed to three selected PAHs: benzo[a]pyrene (BaP), benzo[b]fluoranthene (BbF) and chrysene (CHR). For this purpose, a study was performed on juvenile scallops exposed to BaP (0.01, 0.2 and 4μg/L), BbF (0.02, 0.2 and 2μg/L) and CHR (0.2, 0.8 and 3.2μg/L) for 21 days. Accumulations of these three PAHs in soft parts of scallops, except the 0.01μg/L BaP group and the 0.02μg/L BbF group, showed obvious time and dose dependence, and CHR accumulation was higher when compared to BaP and BbF. Oxidative damage indicators, including lipid peroxidation (LPO), protein carbonyl (PC) and DNA strand breaks, were also measured in soft parts to assess effects of the selected PAHs. The results showed that the LPO levels, PC contents and DNA damage were induced significantly (P<0.05 or P<0.01), except in the low level groups of BaP and BbF, and different trends were detected with time of exposure. According to the correlation analysis results, PC content in soft parts showed a good correlation with the target contaminant and seemed to be proposed as a potential early indicator of BaP, BbF and CHR. In addition the sequence of toxicity is BaP>BbF>CHR, judging by the level of induction of oxidative damage at 0.2μg/L levels. The results of this research are expected to contribute to the establishment of a good biochemical index of exposure to PAHs in laboratory experiments, which can be further useful in field studies.

Abstract:Tetrabromobisphenol A (TBBPA) pollution in marine environmental media poses great risks to marine organisms due to its potential endocrine-disrupting effects. However, limited attention of TBBPA's endocrine-disrupting effects has been paid on marine invertebrates. In this work, the reproductive endocrine-disrupting effects of TBBPA were evaluated by observing the gametes development, quantifying the gender-specific gene expression, and determining vertebrate sex hormones in mussels Mytilus galloprovincialis treated with TBBPA for 30 days. Additionally, transcriptomic profiling and enzymes activities were conducted to investigate the potential mechanisms of reproductive endocrine-disrupting effects. We found that promotion of gametogenesis and alterations of vertebrate sex hormones occurred in TBBPA-treated mussels of both sexes. Meanwhile, estrogen sulfotransferase (SULT1E1) and steroid sulfatase (STS) were up-regulated at transcript level as a result of TBBPA treatments, suggesting that TBBPA disrupted the steroidogenesis in mussels through promoting steroids sulfonation and hydrolysis of sulfate steroids. The induction of SULTs for TBBPA biotransformation might be responsible for the dysregulation of steroidogenesis and steroids metabolism. Overall, these findings provide a new insight into assessing impact of TBBPA as well as TBBPA biomonitoring in marine environment.

Abstract:Polycyclic aromatic hydrocarbons (PAHs) constitute a class of persistent organic pollutants with strong lipophilicity, which readily accumulate within organisms and have the effect to induce disorders in lipid metabolism. The present study aimed to investigate the accumulation localization and pattern of PAHs in Ruditapes philippinarum, and to reveal the association between PAHs and lipids metabolism. The 21-day exposure experiment was conducted using a mixture of phenanthrene, chrysene, and benzo[a]pyrene (the proportion is 1:1:1) at concentrations of 0.4 μg/L, 2 μg/L, and 10 μg/L. The tissue distribution of PAHs indicated that the digestive gland was the primary site of PAHs accumulation. Meanwhile, fluorescence colocalization suggested that PAHs primarily accumulated within the lipid droplets of digestive gland cells. This study further determined the transcriptomic and lipidomic profiles of the digestive gland to analyze the key genes involved in disrupted lipid metabolism and the major lipids affected. Lipidomic analysis identified the key differential metabolites as triglycerides (TGs). Furthermore, TGs were upregulated in the digestive gland had a total carbon atom number of 50-64 and a total number of 3-9 double bonds in the acyl side chains. Biochemical analysis experiments and oil red O stained frozen sections confirmed that the content of TGs steadily increased in various tissues during the experiment, leading to an elevated digestive gland index. Changes of lipid metabolism associated genes expression level also indicated that the synthesis of lipid in digestive gland were up-regulated while the decomposition was down-regulated. This study is the first to demonstrate the cellular localization of PAHs accumulation in bivalves and confirms the pattern of variation in TGs, providing new insights into the mechanisms of PAHs bioaccumulation and lipid metabolism disruption.
Multiple-biomarker approach in the assessment of bisphenol A effect on the grooved carpet clam Ruditapes decussatus (Linnaeus, 1758)

Abstract:Abstract Background Bisphenol A (BPA), a plastic additive monomer, is among the most highly produced chemicals worldwide, and is broadly used in many industries, such as food and beverage containers, milk bottles, and paper products. Previous studies demonstrated that BPA has potential toxicity to aquatic organisms, causing endocrine disturbance and behavioural disorders. The current work aimed to determine the toxic impacts of BPA on the edible marine clam Ruditapes decussatus considering a multi-biomarker approach (mortality, biochemical studies, DNA strand breaks using comet assay, and histopathological examinations with semi-quantitative and quantitative histopathological analyses). The clams were exposed under laboratory conditions to three concentrations of BPA (0 “control”, 1, and 5 μg/L) for a period of 21 days. After the exposure period, BPA impacts were assessed in the digestive gland as a versatile and environmentally relevant organ for ecotoxicological studies. Results In BPA-treated clams, mortality (10%) occurred only at the highest BPA concentration (5 μg/L). Biochemical impairments were detected in a concentration-dependent manner as a consequence of BPA exposure. There were significant increases in malondialdehyde (MDA) and glutathione (GSH) levels, while catalase (CAT) activity was significantly reduced. Our results revealed that BPA induced neurotoxicity in R. decussatus , as evidenced by the inhibition of acetylcholinesterase (AChE) activity in a dose-dependent manner. Furthermore, DNA damage was strongly induced as BPA levels increased. Additionally, our results have been affirmed by alterations in digestive gland tissues at BPA treatments, which consequently can impair the clam’s ability for food absorption; these alterations included mainly atrophic and necrotic digestive tubules, epithelial cell vacuolization, hemocyte infiltration, and intertubular fibrosis. Based on the data obtained from the semi-quantitative and quantitative histopathological analyses, the exposure of the clam’s digestive gland to BPA with concentrations of 1 and 5 μg/L for 21 days showed significant histopathological alterations compared with the control clams. Conclusion The multi-biomarker approach used in the current study proved to be a useful tool for assessing the impact of diphenylmethane compounds, such as BPA. Water-borne BPA causes oxidative stress, neurotoxicity, genotoxicity, and deleterious effects on the clam digestive gland; all of these could deteriorate clam performance and health, causing tissue dysfunction.

Abstract:Benzo(a)pyrene is an important polycyclic aromatic hydrocarbon (PAH) which causes carcinogenic, teratogenic and mutagenic effects in various species and the level of contamination of this toxic agent in the marine environment is of great concern. In this study, metabolic responses induced by two doses (0.02 and 0.2 mu M) of BaP were characterized in the gill tissues of Manila clam Ruditapes philippinarum after exposure for 24, 48 and 96h. The high dose (0.2 mu M) of BaP induced the disturbances in energy metabolism and osmotic regulation based on the metabolic biomarkers such as succinate, alanine, glucose, glycogen, branched chain amino acids, betaine, taurine, homarine, and dimethylamine in clam gills after 24 h of exposure. In addition, hormesis induced by BaP was found in clams exposed to both doses of BaP. Overall, our results demonstrated the applicability of metabolomics for the elucidation of toxicological effects of marine environmental contaminants in a selected bioindicator species such as the Manila clam. Abstract:This study aims to investigate the effects of chrysene (CHR) on biotransformation and detoxification responses of mature scallop Chlamys farreri during the reproduction period. Scallops were exposed to 0.2, 0.8 and 3.2μg/L CHR for 21days; at day 10 scallops were induced to spawn. At days 1, 3, 6, 10, 11, 15 and 21, enzymatic activities of 7-ethoxyresorufin O-deethylase (EROD) and glutathione-s-transferase (GST), related mRNA expression levels of CYP1A1, GST-pi and P-glycoprotein (P-gp) in digestive glands and CHR bioaccumulation in tissues were examined by separately analyzing male and female scallops. During the pre-spawn period, CHR concentrations of the treated groups in tissues except the hemolymph increased rapidly. Levels of enzymatic activities and related gene expressions were all induced by the exposure to CHR for females and males. GST activity and GST-pi mRNA expression showed a good time- and dose-dependent relationship only in males, and P-gp mRNA expression exhibited a dose-dependent manner in both sexes. During the post-spawn period, spawning caused significant reductions of bioaccumulation in tissues but the gill and hemolymph. Enzymatic activities and related gene expressions were for females significantly depressed at day 21 at 0.8 or 3.2μg/L CHR. Overall, females accumulated more CHR than males, while males were more sensitive than females to CHR exposure in gene expressions and enzyme activities. P-gp mRNA expression seemed to be a potential biomarker for PAH exposure. These results will offer the information on CHR biotransformation in this species, and ensure the influence of gender and reproductive status on PAH detoxification metabolism.
Assessment of organic contamination along the coast of Laizhou Bay, China: chemical analysis and integrated biomarker responses in the clam Ruditapes philippinarum

Abstract:An investigative biomonitoring study was conducted along the coastal area of Laizhou Bay (China) to evaluate the impact of organic pollution on the clam Ruditapes philippinarum using bioaccumulation and multi-biomarker measurements. In addition, the polychlorinated biphenyls (PCBs), total petroleum hydrocarbons (TPHs) and nonylphenol (NP) content in surface sediment at the study sites were also analyzed. Concentrations of PCBs, TPHs and NP in the sediments of the study area were 1.90 ± 0.10 μg kg−1, 39.55 ± 2.42 mg kg−1, 9.23 ± 0.41 μg kg−1 dry weight, respectively, while the organic contaminants in the soft tissues of R. philippinarum were 14.81 ± 0.96 μg kg−1 for PCBs, 165.87 ± 5.03 mg kg−1 for TPHs and 86.16 ± 5.29 μg kg−1 for NP. Linear regression analysis on the levels of organic pollutants accumulated in R. philippinarum and in sediments showed no significant correlation. Multi-biomarkers including superoxide dismutase, catalase, glutathione peroxidase, glutathione S-transferase, total glutathione and lipid peroxidation were assayed in gills and digestive glands of R. philippinarum. Finally, the biomarkers in gills were selected to calculate the Integrated Biomarker Response (IBR) index and to evaluate the impact of the three organic contaminants on R. philippinarum collected from different sites. According to IBR results, the western coast and eastern coast exhibited higher environmental stress than the sampling sites along the southern coast of Laizhou Bay. Significant correlation was found between the level of organic contaminants in the sediments and IBR whereas no dependence was found between pollutants’ concentrations in sediments and separate biomarker responses. The results showed that PCBs and NP were the main organic pollutants among the three studied which have caused pollution pressure on R. philippinarum in Laizhou Bay coastal area.
