Abstract:This study investigated the effect of crude protein (CP) and non-essential amino acid (NEAA) supplementation on the growth performance, blood profile, intestinal morphology, mRNA relative abundance of inflammatory and antioxidant markers, and tight junction proteins in piglets over the first 2 weeks after weaning. Ninety 21-day-old piglets (7.55 ± 0.72 kg) were assigned in a randomized block design to one of three dietary treatments: (1) high CP, a diet with 24% CP; (2) low CP, a diet with 18% CP; and (3) low CP + NEAA, a diet with 18% CP supplemented with 5 g/kg Arg (L-arginine; purity >99%) and 10 g/kg Glu + Gln (minimum 10% L-glutamine and minimum 10% L-glutamate). Piglets were fed with corn-soybean meal basal diets in a 14-day trial. There was an improvement ( p < 0.05) in the feed conversion ratio of piglets fed the high-CP diet compared to treatments with low CP or low CP + NEAA. Serum urea nitrogen was higher ( p < 0.05) in piglets fed high CP compared to other dietary treatments. In the duodenum, the villus height of animals fed the low-CP + NEAA diets was greater ( p < 0.05) than those fed with the high- and low-CP diets. The goblet cell proportion of piglets fed low CP + NEAA or high CP was higher ( p < 0.05) compared to low CP. In the jejunum, the crypt depth of the piglets with the high-CP dietary treatment was greater ( p < 0.05) in comparison with low CP + NEAA. In the jejunum, IFN-γ mRNA expression was higher ( p < 0.05) in animals fed the high-CP diets compared to other dietary treatments. However, superoxide dismutase and occludin mRNA expression were higher ( p < 0.05) in animals fed low CP + NEAA than in piglets on the high-CP diets. In the ileum, the number of Peyer's patches in piglets fed high CP was higher ( p < 0.05) compared to other dietary treatments. In conclusion, the high-CP diet (24% CP) improves the feed conversion of piglets in the first 2 weeks after weaning compared to the low-CP diet (18% CP) supplemented or not with NEAA. However, the low-CP diet supplemented with NEAA (Arg, Gln, and Glu) improves intestinal health in piglets by promoting greater villus height and proportion of goblet cells in the duodenum, reducing jejunal crypt depth, and reducing Peyer's number patches in the ileum. In addition, piglets that received the low-CP + NEAA diet showed an increase in superoxide dismutase and occludin and a lower expression of IFN-γ mRNA.

Abstract:Forty-eight piglets(Duroc×Large white×Landrace) weaned at 21 days were used to study the effects of soybean antigen protein on the growth performance in a period of 15 days.The results showed that soybean antigen protein significantly decreased AFI by 25.88%(P0.01)and increased diarrhea rate by 58.77%(P0.01) compared with the control group.The serum D-xylose detection indicated that the serum D-xylose concentration was decreased by 62.39%(P0.01),68.54%(P0.01),67.27%(P0.01),67.98%(P0.01) and 69.95%(P0.01) after 1,2,3,4 h and 5 h respectively compared with the control group.The lengths of the villi within the jejunum and ileum tissues of the test group significantly decreased by 27.56% and 22.43%(P0.01) and the crypt depth significantly increased by 30.01% and 53.03%(P0.01) compared with the control group.There was no significant difference between duodenal villi length and crypt length.The activity of NF-κB in jejunum were increased 71.87%(P0.01).The results indicated that soybean antigen protein in diet exerted a negative influence on growth performance of weaning piglets.Soybean antigen protein could regulate the nuclear factor activity of intestinal mucosa tissue,and affect the morphology and structure of intestinal mucosa tissue,furthermore,affect feed intake,diarrhea rate and growth performance of weaning piglets.

Abstract:This experiment was conducted to investigate the effects of glutamine(Gln) and Clostridium butyricum(CB) on growth performance,immune function,small intestinal morphology and cecal microflora in weanling piglets.Two hundred and eighty-eight piglets weaned at the age of 21 d were randomly divided into four groups with three replicates in every group and twenty-four piglets in every replicate.The four groups were basal diet group(CT),basal diet+1% glutamine group(GT),basal diet+500 mg/kg Clostridium butyricum group(CBT),and basal diet+1% glutamine+500 mg/kg Clostridium butyricum group(GCT),respectively.The results were showed as follows: 1) during 14 to 21 d and 0 to 21 d after weaned,the average daily gain(ADG) of GT and GCT was significantly higher than that of CT,the ADG of GCT was significantly higher than that of CBT,and the feed to gain ratio of GCT was significantly lower than that of CT.2) On the 7th day after weaned,the concentration of complement 4(C4) in GCT and SOD activity in GT,CBT and GCT were significantly higher than those in CT,meanwhile,the T-AOC in CBT and GCT were significantly higher than that in CT.On the 14th day after weaned,the lysozyme activity,the concentration of complement 3(C3) and T-AOC in GT,CBT and GCT were significantly higher than those in CT;the concentration of C4 and SOD activity in GT and GCT were significantly higher than those in CT,respectively.3) On the 7th day after weaned,the villus height of duodenum in GT,CBT and GCT were significantly increased compared with that in CT(P0.05);the villus height of ileum in CBT was increased significantly compared with that in CT(P0.05);the crypt depth of jejunum and ileum in GCT were significantly reduced compared with those in CT(P0.05).On the 14th day after weaned,the villus height of duodenum and ileum in GCT were significantly increased compared with those in CT(P0.05),while the crypt depth of duodenum and ileum in GCT were significantly reduced compared with those in CT(P0.05).4) On the 14th day after weaned,the number of E.coli in GCT was significantly less than that in CT(P0.05),while the number of Bifidobacterium and Lactobacillus were significantly larger than those in CT(P0.05).

Abstract:A total of 64 Duroc×Large Yorkshire×Landrace 21-day weaned piglets were allocated to two groups,each of which had 4 replicates,8 piglets per replicate.The control group and experimental group were fed with basal diet and basal diet + 0.3% peptides produced by enzymatic hydrolysis from silkworm protein,respectively.Results show as follows:(1) Average daily gain is higher significantly,whereas F/G and diarrhea ratio are lower dramatically in the experimental group relative to the control group.(2) The alkaline phosphatase activity and iron level in the serum of piglets are higher in the experimental group than those of control.(3) The diet supplemented with the peptides increases the villus height and decreases the crypt depth of duodenum,jejunum and ileum on day 14 postweaning.(4) The diet supplemented with the peptides increases significantly the activities of sucrase and maltase in jejunum mucosa during 1-14d of postweaning.

Abstract:One hundred and eight piglets weaned at 28 d were divided into six groups(0%, 0.15%, 0.3%, 0.45%, 0.6% GLY-L-GLN and 1% GLN supplementation) for 21 d trial. The results of growth performance showed that 0.3% GLY-L-GLN and 1% GLN in diets could improve growth performance and reduce the percentage of diarrhea significantly(P0.05). The serum D-xylose concentrations in piglets fed 0.15%, 0.3%, 0.45% GLY-L-GLN and 1% GLN diets were significantly higher than that in piglets fed control diet(P0.05).

Abstract:Seventy-two piglets weaned at 21d were allocated to two treatments,each of which had 3 replicates,12 piglets per replicate.The control treatment was fed basal diet,while the experimental group was fed basal diet supplemented with 1% glutamine(Gln).The experiment lasted for 21days.The results showed that 1% Gln in diets could significantly improve growth performance and relative intestinal weight at 7d post weaning.Piglets fed with glutamine had significantly higher intestinal villus height and lower crypt depth at 7d post weaning,but at 14d post weaning,the effects of Gln on growth performance and intestinal mucous development were not significant.

Abstract:Glutamine has an essential role with a beneficial function in improving the nutrition status of young mammals. The influence of L-Glutamine (GM) on the Coefficient of Total Tract Apparent Digestibility (CTTAD) and Apparent Ileal Digestibility (AID), the jejunal enzyme activity associated with nutrient absorption and the energy production in weaned piglets has not been sufficiently studied. The aim of the present study is to provide a profile of the effects of Gln on CTTAD, AID, the activities of jejunal enzymes in connection with nutrient digestion and absorption and energy production. The piglets were weaned at 21 days of age. There were two groups in Experiment 1 and 2 representing supplementation with 0 or 1% Gln to the basal diet. In Experiment 1, the CTTAD of the dietary components and energy was assessed at 3, 5, 10, 15 and 30 days after weaning. In Experiment 2, productive performance, AID, jejunal enzyme activities and expression of Peroxisome Proliferator-Activated Receptor gamma (PPAR gamma) were measured at 10 and 30 days post-weaning. Results showed that dietary Gln supplementation significantly improved the CTTAD of DM, OM, GE and AA and the CTTAD increased significantly with the extension of days after weaning. For the entire experiment, the average daily gain increased by 12.40% (p = 0.049) in the Gin group. Dietary Gln supplementation increased the AID of GE, Leu, Lys, Cys and Pro by 12.50 (p = 0.047), 7.03% (p = 0.041), 5.95% (p = 0.036), 9.30 (p = 0.025), 11.17% (p = 0.009), respectively at 10 days post-weaning; Pro by 6.11% (p = 0.044) at 30 days post-weaning. Jejunal brush border membrane-bound alkaline phosphatase activity increased in the Gln-supplemented pigs by 30.36% (p = 0.048) and 6.21% (p = 0.30) at 10 and 30 days post-weaning, respectively. Compared with the control pigs, the mRNA level of PPAR gamma decreased by 10.85% (p = 0.14) and 41.88% (p = 0.023) after the administration of 1% Gln for 10 and 30 days, respectively. The activity of glutamine synthetase decreased by 48.89% (p = 0.044) at 10 days post-weaning and pyruvate kinase by 13.13% (p = 0.036) at 30 days post-weaning in the Gln-supplemented pigs. In conclusion, 1% Gln supplementation to the post-weaned piglet diet enhanced the CTTAD and AID of diet, improved intestinal absorption and modified jejunal enzyme activities related to Gin metabolism and energy production.

Abstract:To the Editor: In March 2008, nicotine patches (15 mg per 16 hours) were authority-listed for Aboriginal and Torres Strait Islander people by the Pharmaceutical Benefits Advisory Committee (PBAC) as part of efforts to improve access to medications.1,2 Although timely and welcome, the decision to list only 15 mg patches should be revisited, as it is not consistent with current smoking cessation clinical guidelines.3-5 Neither is it consistent with evidence from a 2008 tobacco survey that we conducted in remote Aboriginal communities in Arnhem Land, Northern Territory.

Abstract:We propose a family of entanglement witnesses and corresponding positive maps that are not completely positive based on local orthogonal observables. As applications the entanglement witness of a 3x3 bound entangled state [P. Horodecki, Phys. Lett. A 232, 333 (1997)] is explicitly constructed and a family of dxd bound entangled states is introduced, whose entanglement can be detected by permuting local orthogonal observables. The proposed criterion of separability can be physically realized by measuring a Hermitian correlation matrix of local orthogonal observables.
Effects of Glutamine on Growth Performance of Weanling Piglets

Abstract:An experiment was conducted to investigate the effects of glutamine on growth performance of weanling piglets. Sixty piglets weaned at 21 days of age were randomly assigned to two groups (10 piglets per pen, 3 pens per group). The control group received a maize-soybean meal-based diet. The treatment group received a maize-soybean meal-based diet supplemented with 1.0% free -glutamine. Piglets were fed the diets for 20 days. Results showed that piglets fed the glutamine diet had lower diarrhoea ratio and shorter diarrhoea duration than those fed the control diet during 20 days after weaning. During the first ten days after weaning, pigs supplemented with glutamine had a 12.05% lower feed:gain ratio than those fed the control diet ( P 0.05) compared to the control. During the second ten days after weaning, serum urea nitrogen was reduced by 4.27% and serum concentrations of total protein increased by 18.70% in pigs supplemented with glutamine compared to the control (P > 0.05). There were no differences in albumin, T 3, T 4 and growth hormone.
The effect of dietary supplementation of low crude protein on intestinal morphology in pigs

Abstract:To explore the effects of reducing the Cp levels on intestinal barrier function, low Cp (LP) and NRC standard Cp (NP) diets were fed to pigs from 45 to 160 days, and in vitro experiments were performed using monolayers of IPEC-J2 cells. The number of goblet cells, expression of proteins related to cell junction, amino acid transport, glucose transport, transepithelial electrical resistance (TEER), dextran permeability, and IL-6 secretion level were detected in pigs. The results demonstrated that a moderate reduction of Cp levels did not affect intestinal morphology, as demonstrated by a normal villi height, crypt depth and normal numbers of goblet cells. The maintenance of the intestinal structure obtained with LP was also confirmed by stable mRNA expression levels of muc2 and E-cadherin in the jejunum. We also found that LP did not affect the protein expression of cationic amino acid transporter 1 (CAT-1) and alanine serine cysteine transporter 1 (ASCT1) from 45 to 160 days. Moreover, the excitatory amino acid transporter 3 (EAAT3), sodium-glucose cotransporter 1 (SGLT1) and glucose transporter (GLUT2) protein expression levels in the jejunum were significantly increased at a certain age during the rearing period. Furthermore, we also demonstrated that a reduction in protein concentration up to 15% in the cultural medium of IPEC-J2 cells did not impact the mucosal barrier function. This study demonstrated that a moderate reduction of the protein level did not affect intestinal mucosal barrier function and morphology in the jejunum.
Response to Dietary L-glutamine Supplementation in Weaned Piglets: a Serum Metabolomic Comparison and Hepatic Metabolic Regulation Analysis.

Abstract:A novel metabolomic method based on gas chromatography-mass spectrometry was applied to investigate serum metabolites in response to dietary Gln supplementation in piglets. Sixteen, 21-d-old pigs were weaned and assigned randomly to 2 isonitrogenous diets: 1) Gln diet, which contained 1% L-Gln (as-fed basis), and 2) control diet, which contained L-Ala to make this diet isonitrogenous with the Gln diet. Serum samples were collected to characterize metabolites after a 30-d treatment. in addition, 4 liver samples per treatment were collected to examine enzyme activity and gene expression involved in metabolic regulation. Results indicated that 12 metabolites were altered (P < 0.05) by Gln treatment, including carbohydrates, AA, and fatty acids. A leave-one-out cross validation of random forest analysis indicated that Pro was most important among the 12 metabolites. Thus, these data demonstrate that the control and Gln-supplemented pigs differed (P < 0.05) in terms of metabolism of carbohydrates, Pro, Tyr, and glycerophospholipids. Principal component analysis yielded separate clusters of profiles between the Gln and control groups. Metabolic enzyme activities of Ala aminotransferase and hexokinase increased by 26.8% (P = 0.026) and 26.2% (P = 0.004) in the liver of Gln-supplemented pigs vs. control, respectively, whereas pyruvate kinase (PK) activity decreased by 29.1% (P = 0.001). The gene expression of PK in the liver decreased by 66.1% (P = 0.034) by Gln treatment for 30 d. No differences were observed for the mRNA abundance of mammalian target of rapamycin and PPARγ. On the basis of these data, Gln treatment affected carbohydrate, lipid, and AA metabolism in the whole body of the early weaned piglets. These findings provide insight into specific metabolic pathways and lay the groundwork for the complex metabolic alteration in response to dietary Gln supplementation of pigs.

Abstract:The traditionally classified nutritionally non-essential amino acids are now known to be insufficiently synthesized for maximal growth and optimal health in piglets. This study determined the effects of dietary supplementation with an amino acid blend (AAB; glutamate:glutamine:glycine:arginine:N-acetylcysteine = 5:2:2:1:0.5) on piglet growth performance and intestinal functions. Sixteen piglets (24-day-old) were randomly assigned to a corn and soybean meal-based diet supplemented with 0.99% alanine (isonitrogenous control) or 1% AAB. On day 20 of the trial, blood and intestinal tissue samples were obtained from piglets. Compared with the control, AAB supplementation reduced (P < 0.05) diarrhoea incidence; plasma alanine aminotransferase and diamine oxidase activities; intestinal concentrations of hydrogen peroxide, malondialdehyde, and heat shock protein-70, and intestinal mRNA levels for interleukin-1β, interferon-γ, and chemokine (C–X–C motif) ligand-9; and the numbers of Enterobacterium family, Enterococcus genus and Clostridium coccoides in the colon digesta. Furthermore, AAB supplementation enhanced (P < 0.05): the plasma concentrations of serine, aspartate, glutamate, cysteine, tyrosine, phenylalanine, tryptophan, lysine, arginine, citrulline, ornithine, taurine, and γ-aminobutyric acid; intestinal villus height and surface area, villus height/crypt depth ratio, antioxidative enzyme activities, and mRNA levels for porcine β-defensin-1, sodium-independent amino acid transporters (b0,+AT and y+LAT1), aquaporin (AQP) 3, AQP8, AQP10, nuclear factor erythroid 2-related factor 2 and glutathione S-transferase omega-2, and protein abundances of AQP3, AQP4, claudin-1, occludin and myxovirus resistance 1; and the numbers of Bifidobacterium genus and Lactobacillus genus in the colon digesta. Collectively, these comprehensive results indicate that dietary AAB supplementation plays an important role in improving piglet growth and intestinal function.

Abstract:This study investigated the effects of diet with different amylose–amylopectin ratios (AAR) on the growth performance, intestinal morphology, digestive enzyme activities and mRNA expression of nutrients transporters in piglets with short-term lipopolysaccharide (LPS) intraperitoneal injections. Sixty 21 days-old piglets (Landrace × Yorkshire; 6.504 ± 0.079) were randomly assigned based on their body weight (BW) and litters of origins to five groups with experimental diets with an AAR of 0.00, 0.20, 0.40, 0.60, or 0.80 (namely, the 0.00, 0.20, 0.40, and 0.80 groups), respectively. Each treatment included 12 piglets (one piglet per pen). This experiment lasted for 28 days. On the 28th day, six piglets in each treatment were randomly selected for an LPS intraperitoneal injection (100 μg/kg BW), and other piglets were injected with normal saline. Twelve hours after LPS injection, all piglets were sacrificed to collect small intestinal mucosa for analysis. Although different AAR did not influence the final BW in piglets, the piglets in the 0.40 group represented the poorest feed-to-gain ratio (F/G) in the first, second and fourth week (p < 0.05) and the lowest average daily gain (ADG) in the fourth week (p < 0.05) compared with other groups. In terms of the small intestinal morphology, piglets in the 0.20 and 0.60 groups showed better ileal villous width (p < 0.05). Piglets in the 0.60 group presented greater activities of jejunal maltase, sucrase and alkaline phosphatase (p < 0.05) than those of 0.20 and 0.40. However, a low amylose diet increased the mRNA expression of jejunal glucose and amino acid transporters (p < 0.05). In addition, compared to saline injection, the LPS challenge significantly lessened jejunal digestive enzyme activities (p < 0.01) and, ileal villous width and downregulated the gene expression of glucose and amino acid transporters (p < 0.05) in piglets. Interestingly, certain diet -LPS interactions on duodenal VH/CD, jejunal maltase activity (p < 0.05) and the expression of glucose transporters (p < 0.05) were observed. Taken together, in terms of small intestinal digestion and absorption capacity, these results demonstrated that a diet with an AAR of 0.60 diets could improve the intestinal digestive and absorptive capability by affecting small intestinal morphology, digestive enzymes, and nutrients absorptions in piglets. In addition, the diets containing an AAR of 0.40–0.60 were more likely to resist the damage of LPS stress to intestinal morphology and nutrient absorption.

Abstract:Abstract Feeding low protein (LP) diets to nursery pigs has been reported to improve intestinal health, reduce incidence of post-weaning diarrhea (PWD) and nitrogen excretion. However, these benefits are often undermined by significant reduction in growth performance. A total of 210 piglets (Duroc × Landrace × Yorkshire) were weaned at 21 d of age and allowed a 3-d adaptation to a common solid post weaning diet. At 24 d old, pigs were reweighed (6.02 ± 0.05 kg) and allocated to 5 dietary treatments in a completely randomized block design to give 7 replicates per treatment (n = 6 piglets per replicate). The 5 dietary treatments included i) a high protein positive control diet (PC) with 24% crude protein (CP); ii) a low protein negative control (NC) with 16% CP; iii) NC with 0.1% dietary nucleotide inclusion (NC01); iv) NC with 0.3% dietary nucleotide inclusion (NC03); and v) NC with 0.9% dietary nucleotide inclusion (NC09). All diets were corn-soybean meal based, supplemented with lysine, methionine, tryptophan, and threonine, with all low protein diets (LP) additionally fortified with crystalline isoleucine, and valine to meet the ideal amino acid requirements. Diets were provided ad libitum for 35 d and weekly feed intake (FI) and body weight (BW) were measured. On d 35, blood and fecal samples were collected to determine serum metabolites and nutrient digestibility respectively. Data were analyzed by PROC GLM and orthogonal polynomial contrast of SAS. Comparison between positive and negative control diets and nucleotide levels were performed with orthogonal contrast analysis. Relative to PC, NC diet had decreased overall average daily gain (343.5 vs. 305.5 g/d), incidence of PWD (2.5 vs. 1.2 fecal consistency score), blood urea nitrogen (BUN; 11.3 vs. 3.4 mg/dL) and total tract digestibility of energy (81.3 vs. 80.1 %; P < 0.05, P < 0.05, P < 0.0001 and P < 0.1, respectively). Relative to PC, the NC01, NC03 and NC09 treatments had comparable average daily gain (P > 0.05) while maintaining reduced (P < 0.05) BUN and incidence of PWD. Total tract digestibility of dry matter (83.1 vs. 82.1%) and energy (81.3 vs. 80.1%) were similar between PC and NC09 treatments (P > 0.05). Crude protein reduction decreased serum glutathione (P < 0.05), but without an effect of nucleotide supplementation. Serum glucose and insulin concentrations were not different across treatments (P > 0.05). Results suggest that feeding LP diets supplemented with dietary nucleotide after weaning can increase protein utilization efficiency, reduce incidence of PWD while partially ameliorating the negative effects of LP diets on growth performance.

Abstract:Breeding for higher fertility has resulted in a higher number of low birthweight ( LBW ) piglets. It has been shown that LBW piglets grow slower than normal birthweight (NBW) littermates. Differences in growth performance have been associated with impaired small intestinal development. In suckling and weaning piglets, glutamine (Gln) supplementation has been associated with improved growth and intestinal development. This study was designed to examine the effects of oral Gln supplementation on growth and small intestinal parameters in LBW and NBW suckling piglets. At birth (day 0), a total of 72 LBW (1.10 ± 0.06 kg) and 72 NBW (1.51 ± 0.06) male piglets were selected. At day 1, litters were standardized to 12 piglets, and experimental piglets supplemented daily with either Gln (1 g/kg BW) or isonitrogenous amounts of Alanine (Ala) as control (1.22 g/kg BW) until day 12. Creep feed was offered from day 14 onward. Subgroups of piglets were euthanized at days 5, 12, and 26 for the analyses of jejunal morphometry, cellular proliferation, glutathione concentration and transcript abundance of tight junction proteins. From age day 11 to 21, Gln supplemented LBW ( LBW-Gln ) piglets were heavier than Ala supplemented LBW ( LBW-Ala ) littermates ( P = 0.034), while NBW piglets were heavier until age day 26 compared to LBW littermates. Villus height was higher in LBW-Gln compared to LBW-Ala on age day 12 ( P = 0.031). Sporadic differences among supplementation and birthweight groups were detected for jejunal cellular proliferation, cellular population and glutathione concentration, whereas age was the most dominant factor. These results show that Gln supplementation improved the growth of LBW piglets compared to LBW-Ala beyond the termination of Gln supplementation, but this was not associated with consistent effects on selected parameters of jejunal development.

Abstract:This experiment was conducted to investigate the effects of immobilized antimicrobial peptides on growth performance, serum biochemical index, inflammatory factors, intestinal morphology, and microbial community of weaning piglets. A total of 21 weaning piglets [Duroc × (Landrace × Yorkshire)] with initial body weight (7.64 ± 0.65 kg) were randomly allocated to one of three treatments with seven replicates (one pig per replicate) per treatment according to sex and weight in randomized complete block design. Pigs in the three treatments were fed corn–soybean meal-based diet (CON), corn–soybean meal based diet + flavomycin (25 mg/kg) + quinone (50 mg/kg) (AB), and corn–soybean meal based diet + 1,000 mg/kg immobilized antimicrobial peptides (IAMPs), respectively. The experiment lasted for 28 days, including early stage (0–14 days) and late stage (15–28 days). The results showed the following: (1) compared with the CON group, the average daily gain in the whole experimental time ( p < 0.05) was significantly increased, and the diarrhea rate of weaning piglets was decreased ( p < 0.01) in the IAMPs group; (2) compared with the CON group, the concentrations of serum IgM and superoxide dismutase (SOD) in the IAMPs group were significantly higher than the CON and AB groups ( p < 0.01); (3) compared with CON group, the concentrations of serum interleukin (IL)-10 and transforming growth factor (TGF-β) were significantly increased ( p < 0.05), and the concentration of IL-12 was significantly decreased ( p < 0.05) in the IAMPs group; (4) compared with CON group, the concentrations of serum endotoxin and D-lactate of piglets were significantly reduced ( p < 0.05), and the relative expression of ZO-1 and occludin in the jejunum of piglets were significantly increased ( p < 0.05) in the IAMPs group; (5) compared with the CON group, the villus height of the duodenum and jejunum of weaning piglets in IAMPs and AB groups was significantly increased ( p < 0.05); and (6) compared with CON group, the relative abundance of Escherichia–Shigella in the colon and cecal digesta was decreased. In summary, the addition of 1,000 mg/kg immobilized antimicrobial peptides in the diet effectively relieved weaning stress by showing improved growth performance, antioxidant and immune capacity, intestinal morphology, and microorganisms.

Abstract:Abstract This study aimed to investigate the effects of increased NSP from alternative feedstuffs in feeds with or without NSPase supplementation on growth performance, nutrient digestibility, intestinal health, immune status, and modulation of the intestinal microbiome of nursery pigs and the carryover effect from dietary nursery intervention on the subsequent phases of production. One hundred and sixty newly weaned pigs at 7.0 ± 0.3 kg BW were allotted in a 2 x 2 factorial arrangement with NSP level and NSPase supplementation serving as factors and sex as block. Pigs were assigned to one of 4 dietary treatments with the control having corn DDGS as the main source of NSP and HNSP having soy hulls and wheat bran replacing corn DDGS as the source of NSP. Pigs were fed for 37 d in 2 phases (19 d and 18 d, respectively) and housed in a pen of 4. Growth performance was measured at the end of each phase. At d 37, a pig representing the median BW within each pen from each treatment was euthanized for sampling jejunal mucosa, jejunal tissue, and ileal digesta to analyze intestinal health variables, oxidative stress products, and digesta viscosity. The remaining pigs were then fed a common diet for the subsequent grower and finisher phases to evaluate the carryover effect. At the end of the finisher phase, all pigs were euthanized to obtain carcass data. Data were analyzed using Mixed procedure of SAS 9.4. Overall, NSP level and type nor NSPase supplementation had an effect on growth performance; however, HNSP tended to have improved (P = 0.087) feed efficiency (0.52 to 0.53) in the grower period immediately following the nursery phase. Additionally, HNSP increased (P < 0.05) IgG (4.84 to 5.83 μg/mg of protein) in the jejunal mucosa and tended to increase (P = 0.096) the villus height in the jejunum (482.8 to 506.6 μm). The body weight of HNSP pigs was more uniform (P < 0.05; 8.7 to 11.6%) than the control at the end of the 120-day trial. No differences were observed in carcass traits however, the hot carcass weight of pigs in HNSP tended to be more uniform (P = 0.089; 6.4 to 10.00%) than the control. In conclusion, increased amounts of NSP from alternative feedstuffs does not further impair or damage the intestinal health and subsequent growth performance of nursery pigs and may aid the pig through generated metabolites and alterations to the intestinal microbiota as they are transitioned to the ensuing phases of production as evidenced by the increased uniformity at the end of the trial.
