Abstract:The accumulation of carcinogenic arsenic in rice, the world's main staple crop, represents a health threat to millions of people. The speciation of arsenic controls its mobility and bioavailability and therefore its entry into the food chain. Inorganic and methylated oxyarsenic species have been a focus of research, but arsenic characterization in the field has largely ignored thioarsenates, in which sulfur takes the place of oxygen. Here, on the basis of field, mesocosm and soil incubation studies across multiple paddy soils from rice cultivation areas in Italy, France and China, we find that thioarsenates are important arsenic species in paddy-soil pore waters. We observed thioarsenates throughout the cropping season, with concentrations comparable to the much-better-investigated methylated oxyarsenates. Anaerobic soil incubations confirmed a large potential for thiolation across a wide diversity of paddy soil types in different climate zones and with different parent materials. In these incubations, inorganic thioarsenates occurred predominantly where soil pH exceeded 6.5 and in the presence of zero-valent sulfur. Methylated thioarsenates occurred predominantly at soil pH below 7 and in the presence of their precursors, methylated oxyarsenates. High concentrations of dissolved iron limited arsenic thiolation. Sulfate fertilization increased thioarsenate formation. It is currently unclear whether thiolation is good or bad for rice consumption safety. Nevertheless, we highlight thiolation as an important factor to arsenic biogeochemistry in rice paddies. Thioarsenates are found in the pore waters of rice paddy fields, comparable in concentration to methylated oxyarsenates, according to field, mesocosm and soil incubation studies.

Abstract:Arsenic species transformation in paddy soils has important implications for arsenic accumulation in rice grains and its safety to the consumers. Methylated thioarsenates including highly toxic dimethylated monothioarsenate (DMMTA) have been detected in paddy soils, but their production and dynamics remain poorly understood. In the present study, we first optimized a HPLC-ICP-MS method to quantify methylated thioarsenate species. Using this method together with 10 mM diethylenetriamine pentaacetate (DTPA) to preserve As speciation, we investigated methylated thioarsenate species in porewaters of seven As-contaminated soils incubated under flooded conditions and of two paddy fields. DMMTA was the main methylated thioarsenate species in the porewaters in both incubated soils and paddy fields, with concentrations ranging from 0.2 to 36.2 μg/L and representing ca. 58% of its precursor dimethylarsenate (DMA). The temporal production and dynamics of DMMTA were linked with the DMA concentrations. When soils were drained, DMMTA was converted to DMA. In the two paddy fields, DMMTA concentrations in rice grains were 0.4–10.1 μg/kg. Addition of sulfur fertilizer and rice straw incorporation increased grain DMMTA by 9–28%. Abstract:Quite a few new thioarsenicals have recently been found in urine of arsenic-exposed humans and animals, and some of them have been shown to be highly toxic to cells. However, little is known about their toxic effects and metabolism in the body. In order to elucidate the toxic mechanism of thioarsenicals, we further focused on the distribution and metabolism of monomethylmonothioarsonic acid (MMMTA(v)) in rats. MMMTA(v) was synthesized chemically and injected intravenously into rats at the dose of 0.5 mg As/kg, followed by speciation analysis of selected organs and body fluids at 10 min and 12 h after the injection. MMMTA(v) was excreted into urine in its intact form, and approximately 35% of the dose was recovered in urine at 12 h after the injection, suggesting that MMMTA(v) was taken up more effectively by organs/tissues than non-thiolated, monomethylarsonous acid (MMA(v)) previously studied. On the other hand, the liver and kidneys contained arsenic that was in a protein-binding form with free forms of DMA(v) or DMDTA(v) at 10 min, and disappeared at 12 h after the injection. Moreover, these bound arsenic species in kidneys were converted back to MMA(v) after oxidation with H(2)O(2), suggesting that the arsenic bound to proteins had been reduced within the body and was in a trivalent oxidation state. In red blood cells (RBCs), most of the arsenic was in the form of DMA(III) bound to hemoglobin (Hb), and approximately 40% of the dose was recovered in RBCs at 12 h after injection. These results indicate that arsenic accumulated preferentially in RBCs after being transformed to DMA(III). In addition, we have also discussed the effect of MMMTA(v) on viability of human bladder cancer T24 cells in comparison with MMA(v). Abstract:Arsenic (As) accumulation in rice is of global concern for human health and international trade. Rice is typically reported to contain inorganic As (iAs) and dimethylated arsenate (DMA), with current food guidelines limiting toxic iAs but not less-toxic DMA. Here, we show that the highly toxic dimethylated monothioarsenate (DMMTA) is also found in rice worldwide and has been unknowingly determined as less-toxic DMA by previous routine analytical methods. Using enzymatic extraction followed by high-performance liquid chromatography-inductively coupled plasma-mass spectrometry (HPLC-ICP-MS) analysis with a C18 column, DMMTA was detected in rice grains (n = 103) from a field survey from China and in polished rice grains (n = 140) from a global market-basket survey. Concentration ranged from <0.20 to 34.8 μg/kg (median 10.3 μg/kg), accounting for 0 to 21% of total As. A strong linear correlation was observed in all rice samples between DMA and DMMTA (being 30 ± 8% of DMA) concentrations. This robust relationship allows an estimation of DMMTA in rice grains from the DMA data reported in previous market-basket surveys, showing a general global geographical pattern with DMMTA concentration increasing from the equator toward high-latitude regions. Based on the global occurrence and potential high toxicity, DMMTA in rice should be considered in health risk assessments and for setting food regulations.

Abstract:Cellular and animal studies involving MMA(III) (monomethyl arsonous acid) and DMA(III) (dimethyl arsinous acid) have indicated that their toxicities meet or exceed that of iAs. Thiolated arsenic metabolites were observed in urine after oral exposure of inorganic arsenic in some studies. For these species, the toxicological profile was not yet fully characterized in human cells. Some studies revealed that trivalent organoarsenic species are well absorbed in the intestine compared to iAs. However, other studies also indicated that a significant amount of rice-bound As reaches the colon, which may be attributed to the fibre-rich nature of the rice. Studies have revealed that microorganisms from the gut environment are important contributors to arsenic speciation changes. We aimed to study how the gut microbial metabolism affects As in different rice matrices. This was done in vitro using colon suspension from the Simulator of the Human Intestinal Microbial Ecosystem (SHIME system). Significant amounts of MMA(III), DMA(III) and MMMTA(V) were formed due to microbial metabolic processes like methylation and thiolation. These results suggested that presystemic metabolism by human gut micro biota should not be neglected in risk assessment studies. In this context, also toxicity and absorption of thiolated species by mammalian cells should be further investigated.

Abstract:Dimethylarsenate [DMAs(V)] can be produced by some soil microorganisms through methylation of inorganic arsenic (As), especially in anoxic paddy soils. DMAs(V) is more phytotoxic than inorganic As and can cause the physiological disorder straighthead disease in rice. Rice cultivars vary widely in the resistance to DMAs(V), but the mechanism remains elusive. Here, we investigated the differences in DMAs(V) uptake, translocation, and reduction to dimethylarsenite [DMAs(III)], as well as the effects on the metabolome, between two rice cultivars Mars and Zhe733. We found that Mars was 11-times more resistant to DMAs(V) than Zhe733. Mars accumulated more DMAs(V) in the roots, whereas Zhe733 translocated more DMAs(V) to the shoots and reduced more DMAs(V) to DMAs(III). DMAs(III) was more toxic than DMAs(V). Using heterologous expression and in vitro enzyme assays, we showed that the glutathione-S-transferases OsGSTU17 and OsGSTU50 were able to reduce DMAs(V) to DMAs(III). The expression levels of OsGSTU17 and OsGSTU50 were higher in the shoot of Zhe733 compared to Mars. Metabolomic analysis in rice shoots showed that glutathione (GSH) metabolism was perturbed by DMAs(V) toxicity in Zhe733. Application of exogenous GSH significantly alleviated the toxicity of DMAs(V) in Zhe733. Taken together, the results suggest that Mars is more resistant to DMAs(V) than Zhe733 because of a lower root-to-shoot translocation and a smaller capacity to reduce DMAs(V) to DMAs(III).

Abstract:Methylation of arsenic in soil influences its environmental behavior and accumulation by plants, but little is known about the factors affecting As methylation. As speciation was determined in the pore waters of six soils from diverse geographical locations over 54 days of incubation under flooded conditions. The concentration of methylated As (monomethylarsonic acid, MMA, and dimethylarsinic acid, DMA) varied from 0 to 85 μg L(-1) (0 - 69% of the total As in pore water). Two Bangladeshi paddy soils contaminated by irrigation of As-laden groundwater produced large concentrations of inorganic As but relatively little methylated As. Two contaminated paddy soils from China produced a transient peak of DMA during the early phase of incubation. Methylated As represented considerable proportions of the total soluble As in the two uncontaminated soils from the UK and U.S. The copy number of the microbial arsenite methyltransferase gene (arsM) correlated positively with soil pH. However, pore-water methylated As correlated negatively with pH or arsM copy number, and positively with dissolved organic C. GeoChip assay revealed considerable arsM diversity among the six soils, with 27-35 out of 66 sequences in the microarray being detected. As speciation in rice plants grown in the soils generally mirrored that in the pore water. The results suggest that methylated As species in plants originated from the soil and As methylation in soil was influenced strongly by the soil conditions.

Abstract:Arsenic's effect on rice plant health is a critical environmental issue. This study reveals that rice plants absorb inorganic arsenic and dimethylarsenic differently, with dimethylarsenic posing a greater threat to rice plant health. These findings contribute to our understanding of arsenic toxicity in plants, highlighting the need for further research into detoxification strategies for dimethylarsenic. Arsenic toxicity in plants, particularly the effects of different arsenic species, is not well understood. This study investigated the response of juvenile rice plants, grown hydroponically, to prolonged exposure to inorganic and dimethyl arsenic species. The hydroponic system removed complexity by eliminating soil processes. The accumulation of inorganic As (As i ) and dimethylarsenic (DMA) in hydroponically grown rice was monitored for plants exposed to different As concentrations (0–6.7 μmol L −1 ). Dose–response experiments were conducted to compare the effects of As species on plant health in terms of growth. Plants absorb As i and DMA linearly, with faster As i uptake than DMA. As i exposure leads to higher As concentrations in roots and shoots than DMA. Despite more As i in roots, its translocation to shoots is lower. As i and DMA accumulation in shoots remains relatively constant at lower As concentrations. At the highest As concentration, more As i and DMA accumulate in shoots. Exceeding 1.6 μmol L −1 , As i and DMA reduce plant height and biomass. As i -exposed plants show little health differences except at the highest concentrations. DMA-exposed plants show more unhealthy instances above 1.6 μmol L −1 . DMA's lower uptake rate aligns with other rice species results, as do lower shoot and root translocation factors. Near constant As concentrations in shoots at low As i concentrations suggest an As i exposure threshold before plants lose their As sequestration ability, resulting in reduced growth. DMA exposure increases the number of unhealthy plants, suggesting a greater potential effect on plant health and fitness, differing from As i -induced stress.

Abstract:Acid extraction is commonly used to analyze arsenic species in rice. During the extraction process, spiked monomethylarsonic acid (MMA) is often transformed into different compounds. A similar phenomenon is observed in the arsenic speciation analysis of seafood. To identify these compounds, we analyzed a previously prepared extract using liquid chromatography–time-of-flight/mass spectrometry in differential analysis and liquid chromatography–inductively coupled plasma-MS. The compound was identified as monomethylmonothioarsonic acid (MMMTA), a thioarsenical, which is estimated to be more cytotoxic than MMA. As MMMTA was readily produced by bubbling hydrogen sulfide through MMA, this suggests that MMA reacts with sulfur in rice during the extraction process. Our data also suggested that dimethylarsinic acid could be transformed into another compound, although the generation rate was low. For reliable arsenic speciation analyses, the transformation of arsenic compounds during extraction must be avoided. This study demonstrates that arsenic compounds can be transformed by dilute acid extraction.

Abstract:The role of the retinoblastoma gene product, RB, in transmitting the signals of apoptosis is unclear, but RB is considered to be antiapoptotic because RB mediates cell cycle arrest that also can interrupt intracellular signaling pathways leading to apoptosis. Gamma-radiation can cause apoptosis, the process of programmed cell death, via several mechanisms including DNA damage, ceramide production, and the generation of free radical oxygen species. We investigated the effect of RB on radiation-induced apoptosis by restoring normal RB expression in DU-145 prostate cancer cells that have one deleted and one truncated RB gene. DU-145 cells are highly resistant to apoptosis induced either by radiation or by the addition of ceramide. Two independently derived RB-positive DU-145 derivative cell lines underwent apoptosis after irradiation or exposure to the cell permeable C2-ceramide. Apoptosis in the RB-positive cell lines was not associated with major changes in the cell cycle response to irradiation. RB-mediated apoptosis occurred in the absence of expression of caspases 8, 6, 3, and 7 and without detectable cleavage of poly(ADP)ribose polymerase. However, a specific inhibitor of serine proteases, Na-p-Tosyl-L-lysyl-chloromethyl ketone, inhibited radiation-induced apoptosis in DU-145 cells expressing RB. Radiation-induced apoptosis was preceded by an increase in JUN protein expression and accompanied by activation of the stress-related JUN kinase. Our data show that RB is proapoptotic in DU-145 cells and acts upstream of JUN expression and JNK activation.
Geographical variations in arsenic contents in rice plants from Latin America and the Iberian Peninsula in relation to soil conditions

Abstract:Arsenic is a ubiquitous, toxic element that is efficiently accumulated by rice plants. This study assessed the spatial variability in the total As (tAs) contents and organic and inorganic forms in different types of rice, plant parts (husk, stem, leaves and phytoliths) and residues. Samples were collected in different countries in Latin America (Ecuador, Brazil and Peru) and the Iberian Peninsula (Spain and Portugal). The tAs content in commercial polished rice from the Latin American countries was similar (0.130-0.166 mg kg-1) and significantly lower than in the rice from the Iberian countries (0.191 ± 0.066 mg kg-1), and together, the tAs concentration in brown rice (236 ± 0.093 mg kg-1) was significantly higher than in polished and parboiled rice. The inorganic As (iAs) content in rice was similar in both geographical regions, and the aforementioned difference was attributed to dimethylarsinic acid (DMA). The relative abundance of organic species increased as the tAs content in rice grain increased. A meta-analysis of our and previously reported data confirmed the negative correlation between iAs/tAs and tAs. At low tAs concentrations, inorganic forms are dominant, while at higher values (tAs > 0.300 mg kg-1) the concentration of organic As increases substantially and DMA becomes the dominant form in rice grain. On the contrary, inorganic arsenic was always the dominant form, mainly as arsenate [As(V)], in leaves and stems. The presence in soils of high concentrations of amorphous Fe and Al oxides and hydroxides, which are capable of strongly adsorbing oxyanions (i.e. arsenate), was associated with low concentrations of As in rice plants. In addition, the presence of high concentrations of As(V) in stems and leaves, low concentration of As in phytoliths, and the As associated with organic matter in stems and husk, together suggest that rice plants take up more As(V) than As(III).

Abstract:Rice, as the most essential food grain, is frequently exposed to high concentrations of arsenic. Among the arsenic species, dimethylarsenate (DMAs(V)) is preferentially translocated from paddy soils to rice grains, posing serious threats to food safety and yield. Herein, we report an efficient strategy for DMAs(V) mitigation in paddy soils with nanoscale Zero-Valent Iron (nZVI). Species and concentrations of arsenic in paddy porewater were monitored during a 28-d soil-water incubation. Effects of nZVI dose towards microbial sulfate reduction and methane generation potential in paddy soils, which are crucial for arsenic methylation and demethylation, were analyzed via metagenomic sequencing. Results demonstrated that the maximal DMAs(V) concentration in paddy porewater decreased from 0.37 to 0.04 μM in arsenic-contaminated paddy soils with nZVI dose increasing from 0 to 5.0 g/kg. Accordingly, the maximal concentration of inorganic arsenite (iAs(III)), which is the precursor of DMAs(V), decreased from 1.39 to 0.23 μM. Furthermore, the application of nZVI reshaped the structure of microbial community in paddy soils. Specifically, the relative abundance of δ-proteobacteria involved in sulfate reduction, which is crucial for iAs(III) methylation, waned from 7.62 % to 3.17 %, while that of Methanomicrobia for DMAs(V) demethylation and methanogenesis proliferated from 7.03 % to 13.62 %, with nZVI dose increasing from 0 to 5.0 g/kg. Via simultaneous inhibition of DMAs(V) formation and acceleration of DMAs(V) transformation, nZVI efficiently controls the accumulation of DMAs(V) in paddy porewater. In conclusion, these findings prove the efficient performance for DMAs(V) mitigation with nZVI and uncover its underlying mechanisms.

Abstract:Rice consumption is a source of arsenic (As) exposure, which poses serious health risks. In this study, the accumulation of As in rice was studied. Research shows that As accumulation in rice in Taiwan and Bangladesh is higher than that in other countries. In addition, the critical factors influencing the uptake of As into rice crops are defined. Furthermore, determining the feasibility of using effective ways to reduce the accumulation of As in rice was studied. AsV and AsIII are transported to the root through phosphate transporters and nodulin 26-like intrinsic channels. The silicic acid transporter may have a vital role in the entry of methylated As, dimethylarsinic acid (DMA) and monomethylarsonic acid (MMA), into the root. Amongst As species, DMA(V) is particularly mobile in plants and can easily transfer from root to shoot. The OsPTR7 gene has a key role in moving DMA in the xylem or phloem. Soil properties can affect the uptake of As by plants. An increase in organic matter and in the concentrations of sulphur, iron, and manganese reduces the uptake of As by plants. Amongst the agronomic strategies in diminishing the uptake and accumulation of As in rice, using microalgae and bacteria is the most efficient.

Abstract:Widespread arsenic (As) contamination in paddy rice (Oryza sativa) from both geologic and anthropogenic origins is an increasing concern globally. Substantial efforts have been made to elucidate As transformation and uptake processes in rhizosphere and metabolism in rice plant, which provides an essential foundation for the development of mitigation strategies. However, a range of crucial mechanisms from As mobilization in rhizosphere to transport to grains remain poorly understood. To provide new insight into the underlying mechanisms of As accumulation in rice, a range of new perspectives on As bioavailability, transport pathways, and in situ speciation are reviewed here. Specifically, the prominent effects of water regime, Fe plaque, and biochar on As mobilization in rice rhizosphere are discussed critically. An updated understanding of arsenite (AsIII) and methylated As transport from root to vascular bundle and grain is integrated and discussed in detail. Special attention is given to As speciation and distribution in rice grain with potential coping strategies being provided and discussed. Future research priorities are also identified. The new insight into As bioavailability, transport and speciation in rice would lead to a better understanding of As contamination in rice. They would also provide useful strategies from agronomic measures to genetic engineering for more effective restriction of As transport and accumulation in food chain.
Speciation of Arsenic in Rice by High-Performance Liquid Chromatography–Hydride Generation-Atomic Fluorescence Spectrometry with Microwave-Assisted Extraction

Abstract:Arsenic speciation in paddy rice is of considerable interest due to its impact on the food safety and human health. In this study, a simple methodology was developed to simultaneously extract and analyze As species in rice from China. Arsenic species, including arsenite (As(III)), arsenate (As(V)), dimethylarsinic acid (DMA), and monomethylarsonic acid (MMA), were extracted by methanol-water (50:50, v/v) containing 0.02 mol L−1 nitric acid with a microwave-assisted procedure, and then determined by high performance liquid chromatography–hydride generation-atomic fluorescence spectrometry (HPLC–HG-AFS). The results showed that the method has good efficiency (>90%) for rice, indicating that there were no significant losses or transformations of arsenic during sample treatment and analysis. The limits of quantification (LOQ) of the method were 8.0, 20, 12, and 12 ng g−1 for As(III), As(V), DMA, and MMA, respectively. When this method was applied to the analysis of rice, As(III) had the highest concentration, followed by DMA, As(V), and MMA. The estimated weekly intake of inorganic As from rice by Chinese people accounted for 11.83% of the provisional tolerable weekly intake. The As speciation results in this study suggest that the risk associated with As in rice to human health may be negligible.

Abstract:To investigate how soil fertilization/amendments alters arsenic speciation grain off-take in paddy rice, rice was grown to maturity in growth chambers fertilized with standard mineral fertilizer, wood ash (rich in silica), pig slurry (rich in organic matter), and non-amended control. The soil was sourced from a Chinese paddy field. The primary fertilized elements (nitrogen, phosphorus, potassium) were kept constant across treatments. Porewater chemistry and soil microbiology were monitored throughout the experiments. Total grain arsenic, sum of inorganic arsenic and dimethylarsinic acid (DMA), was significantly different between treatments ( P = 0.024), with inorganic arsenic varying from 0.025 to 0.08 mg/kg and DMA from 0.08 to 0.16 mg/kg for control compared to fertilized, respectively. Fertilizer source made no difference to arsenic speciation concentration in grain. Porewater analysis found that as anaerobism set in, inorganic arsenic, phosphorus and manganese greatly lowered in concentration. Methylated arsenic species concentrations increased over time, concurrent with an increase in pH, decrease in Eh, and increase in total organic carbon and iron, with no strong treatment effects, except for pig slurry that enhanced pH and decreased Eh. Methanogenic archaea, sulfate-reducing bacteria and Acidobacteria increased with time and some Actinobacteria and Firmicutes increased due to slurry, but then decreased with time ( P < 0.01). Methanogenic archaea and sulphate-reducing bacteria correlated positively with porewater DMA and negatively with porewater inorganic arsenic ( P < 0.05). Genera within the Actinobacteria and Burkholderiaceae correlated negatively with DMA, while genera with iron-reducing capacity (Clostridiales) correlated positively with porewater inorganic arsenic and DMA ( P < 0.05).

Abstract:BACKGROUND: Arsenic (As) toxicity is primarily based on its chemical speciation. Although inorganic and methylated As species are well characterized in terms of metabolism and formation in the human body, the origin of thiolated methylarsenicals is still unclear.OBJECTIVES: We sought to determine whether sulfate-reducing bacteria (SRB) from the human gut are actively involved in the thiolation of monomethylarsonic acid (MMAV).METHODS: We incubated human fecal and colon microbiota in a batch incubator and in a dynamic gut simulator with a dose of 0.5 mg MMAV in the absence or presence of sodium molybdate, an SRB inhibitor. We monitored the conversion of MMAV into monomethyl monothioarsonate (MMMTAV) and other As species by high-performance liquid chromatography coupled with inductively coupled plasma mass spectrometry analysis. We monitored the sulfate-reducing activity of the SRB by measuring hydrogen sulfide (H2S) production. We used molecular analysis to determine the dominant species of SRB responsible for As thiolation.RESULTS: In the absence of sodium molybdate, the SRB activity-primarily derived from Desulfovibrio desulfuricans (piger)-was specifically and proportionally correlated (p < 0.01) to MMAV conversion into MMMTAV. Inactivating the SRB with molybdate did not result in MMAV thiolation; however, we observed that the microbiota from a dynamic gut simulator were capable of demethylating 4% of the incubated MMAV into arsenous acid (iAsIII), the trivalent and more toxic form of arsenic acid (iAsV).CONCLUSION: We found that SRB of human gastrointestinal origin, through their ability to produce H2S, were necessary and sufficient to induce As thiolation. The toxicological consequences of this microbial As speciation change are not yet clear. However, given the efficient epithelial absorption of thiolated methylarsenicals, we conclude that the gut microbiome-and SRB activity in particular-should be incorporated into toxicokinetic analysis carried out after As exposure.

Abstract:It is well known that arbuscular mycorrhizal (AM) fungi can enhance plant arsenic (As) resistance by influencing As uptake, translocation, and speciation; however, As transformation and volatilization by an entire plant inoculated with AM fungus remains uninvestigated. In the present study, AM symbiosis of Rhizophagus irregularis with unbroken Medicago sativa was successfully established in vitro. Afterwards, five concentrations of arsenate were applied to the culture media. The results showed that AM inoculation could methylate inorganic As into dimethylarsinic acid (DMA), dimethylarsine (DMAsH), and trimethylarsine (TMAs), which were detected in the plants, media, or air. Volatile As, accounting for a small proportion of total organic As, appeared under high arsenate exposure, accompanied by remarkable upregulation of root RiMT-11, an arsenite methyltransferase gene in R. irregularis. In addition, AM colonization significantly increased arsenite percentages in plant tissues and external media. Regardless of As species, AM inoculation tended to release the transformed As into the environment rather than transfer them to plant tissues. Our present study, for the first time, comprehensively verified As methylation, volatilization, and reduction by AM fungus associated with the entire plant under absolute axenic conditions and gained a deeper insight into As metabolism in AM symbionts.

Important Roles of Thiols in Methylmercury Uptake and Translocation by Rice Plants

Abstract:The bioaccumulation of the neurotoxin methylmercury (MeHg) in rice is a significant concern due to its potential risk to humans. Thiols have been known to affect MeHg bioavailability in microorganisms, but how thiols influence MeHg accumulation in rice plants remains unknown. Here, we investigated effects of common low-molecular-weight thiols, including cysteine (Cys), glutathione (GSH), and penicillamine (PEN), on MeHg uptake and translocation by rice plants. Results show that rice roots can rapidly take up MeHg, and this process is influenced by the types and concentrations of thiols in the system. The presence of Cys facilitated MeHg uptake by roots and translocation to shoots, while GSH could only promote MeHg uptake, but not translocation, by roots. Conversely, PEN significantly inhibited MeHg uptake and translocation to shoots. Using labeled 13Cys assays, we also found that MeHg uptake was coupled with Cys accumulation in rice roots. Moreover, analyses of comparative transcriptomics revealed that key genes associated with metallothionein and SULTR transporter families may be involved in MeHg uptake. These findings provide new insights into the uptake and translocation of MeHg in rice plants and suggest potential roles of thiol attributes in affecting MeHg bioavailability and bioaccumulation in rice.
