Abstract:The methods of detecting numerous prohibited components are not included in the Technical Specifications for Cosmetic Safety (2015 Edition). Recently, owing to its high speed, sensitivity, and anti-interference properties, ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) became the preferred method of detecting banned substances in cosmetics. In this study, a UPLC-MS/MS method was developed for use in determining 87 prohibited ingredients in cosmetics, including 33 sex hormones, 20 anti-infective drugs, 15 antihistamines, 7 coumarins, 4 sedative-hypnotic drugs, 4 antipyretic and analgesic drugs, 2 allergenic fragrances, and 2 drugs with vasoconstriction effects. The main factors affecting the response, recovery, and sensitivity of the method, such as the type of extraction solvent, extraction time, ratio of the mobile phases, and MS conditions, were optimized during sample pretreatment and instrumental analysis. Accordingly, approximately 0.2 g of the toner or cream sample was dispersed in 2 mL acetonitrile in a 10 mL colorimetric tube. After diluting to 10 mL with 50% acetonitrile aqueous solution, the sample was ultrasonically extracted for 20 min and centrifuged, and the mixture was then filtered through a 0.22 μm membrane. Approximately 0.2 g of the oil sample was dispersed in 2 mL n-hexane in a 15 mL polypropylene centrifuge tube and extracted twice with 3 mL 70% acetonitrile aqueous solution. The extracts were transferred into a 10 mL colorimetric tube and diluted to 10 mL with 50% acetonitrile aqueous solution, and the mixture was then filtered through a 0.22 μm membrane. The samples were separated using a CORTECS C18 column (150 mm×2.1 mm, 2.7 μm), employing a gradient elution program with acetonitrile and 0.1% formic acid aqueous solution as the mobile phases. The flow rate, column temperature, and injection volume were respectively set at 0.3 mL/min, 40 ℃, and 2 μL. The 87 compounds were monitored in multiple reaction monitoring (MRM) mode with electrospray ionization (ESI) under positive and negative conditions. Matrix-matched external standard calibration was used for quantification, and the analysis was completed within 33 min. The prohibited compounds exhibited good linear relationships, with r values of >0.99, and the limits of detection (LODs) and quantification (LOQs) for the 87 compounds were 0.07-0.38 and 0.21-1.15 μg/g, respectively. Three types of cosmetic matrices were selected to verify the recovery and precision of the method at LOQ, 2 LOQ, and 10 LOQ levels. The average recoveries of the 87 prohibited compounds were in the range of 81.7%-115.4%, and the relative standard deviations (RSDs, n=6) were 0.4%-9.9%. The reliability of the developed method was demonstrated by applying it to 349 commercial cosmetics obtained from the market, and 8 positive samples were identified. The positive components included trimethoprim, terbinafine, naphazoline, 7-methoxycoumarin, and 7-methylcoumarin. The established method displays the advantages of simple operation and rapidness and a high sensitivity and good recovery. And, this method provides technical support for rapid risk screening and the revision of national standards for cosmetics.
Simultaneous Determination of Phthalates and Parabens in Cosmetic Products by Gas Chromatography/Mass Spectrometry Coupled with Solid Phase Extraction

Abstract:Studies have been carried out on the simultaneous determination of 8 phthalates, i. e. di-ethyl phthalate (DEP) , di-propyl phthalate (DPP) , di-isobutyl phthalate (DIBP) , dibutyl phthalate (DBP) , benzyl butyl phthalate ( BBP) , di-cyclohexyl phthalate (DCHP) , di-(2-ethylhexyl) phthalate (DEHP), di-octyl phthalate (DOP) and 4 parabens, i. e. methylparaben (MPB), ethylparaben (EPB), propyl paraben (PPB), and butyl paraben (BPB) by gas chromatography in combination with mass spectrometry (GC/MS) in electron ionisation mode (EI) with selected-ion monitoring (SIM) acquisition method. The phthalates and parabens in 15 cosmetic products, including hair sprays, perfumes, deodorants, cream, lotion, etc. were determined. The determination of the samples were performed after sonication-assisted extraction with methanol, cleaned up with an LC-C18 column (3 mL) and analyzed by GC/MS method. The base peak (m/z 149) of the phthalates and the base peak (m/z 121) of the parabens were selected for the screening studies. The characteristic ions, m/z 121, 149, 177, 222 for DEP; m/z 149, 191, 209 for DPP; m/z 57, 149, 223 for DIBP; m/z 104, 149 for DBP; m/z 91, 132, 149, 206 for BBP; m/z 55, 149, 167 for DCHP; m/z 113, 149, 167, 279 for DEHP; m/z 149, 279 for DOP; m/z 65, 93, 121, 152 for MPB; m/z 93, 121, 138, 166 for EPB; m/z 93, 121, 138, 180 for PPB; and m/z 93, 121, 138, 194 for BPB were chosen for quantitative studies. These techniques are capable to detect phthalates and parabens at the level of 0. 1 -5. 0 microg/kg. Overall recoveries were 80% - 100% with relative standard deviations (RSDs) less than 10%. Only one of the 15 examined samples was free from phthalates and parabens. The rest 14 samples were found to contain at least 3 or more of these phthalates and/or parabens. The predominant phthalates detected in the studied samples were MPB, PPB, DPP, DCHP and DEHP. The residue levels were at 1. 42 -4 278 mg/kg.

Abstract:Prostate cancer is the most common malignancy among the male survivorship population in the United States, representing 44% of approximately 7 million survivors. In the era of modern medicine and value-based care, successfully treating only the cancer is not sufficient. The cancer survivor represents an individual in need of restoration and protection against future events. A well-designed and well-supported survivorship program not only meets a mandate for accreditation, it logically translates into better patient care. This review summarizes the history of the survivorship movement, outlines some key elements of a survivorship program, and highlights the opportunity to apply these principles to improve cancer-related care, develop relationships with colleagues that may allow increased identification of men at risk, and expand both the experience and outcomes of individual specialists within men's health.
Screening and quantification of 18 glucocorticoid adulterants from herbal pharmaceuticals and health foods by HPLC and confirmed by LC-Q-TOF-MS/MS

Abstract:A procedure for the screening and quantification of 18 glucocorticoids, i.e. hydrocortisone sodium succinate (HSS), prednisolone (PDL), prednisone (PDS), hydrocortisone (HCS), methylprednisolone (MPS), betamethasone (BTM), dexamethasone (DXM), triamcinolone acetonide (TA), prednisolone acetate (PLA), hydrocortisone acetate (HA), fludrocortisone acetate (FA), prednisone acetate (PA), cortisone acetate (CA), dexamethasone acetate (DA), hydrocortisone butyrate (HB), triamcinolone acetonide acetate (TAA), fluocinonide (FN) and halcinonide (HC) from herbal pharmaceuticals and health foods was established and fully validated. The samples were extracted by methanol and separated by HPLC. The retention times and ultraviolet spectra were used for the preliminary screening, and the suspected adulterants were then confirmed by liquid chromatography-quadrupole-time of flight mass spectrometry (LC-Q-TOF-MS/MS) and quantified by HPLC. The developed procedure was successfully applied to 14 herbal samples, and 316.3 µg g<sup>-1</sup> of DA and 13.6 µg mL<sup>-1</sup> of BTM were found in a tablet sample and a spray sample, respectively. To our best knowledge, this is the first report of the simultaneous screening and quantification of these 18 glucocorticoids from any matrix.

Abstract:RATIONALE: There is a strong demand to develop a method capable of rapid screening for the adulteration of glucocorticoids (GCs) in cosmetics. An ambient ion source-direct analysis in real time (DART), coupled with quadrupole time of flight mass spectrometry (QTOF MS) was used for the rapid screening of GCs in essential oils. METHODS: Liquid-liquid extraction was employed prior to the DART-QTOF MS analysis. Calibration curves for eight GCs were obtained using methyltestosterone as an internal standard. MS/MS experiments and accurate mass measurements were carried out to provide reliable and powerful evidence for the adulteration of targeted GCs. RESULTS: Quantification results were obtained in terms of linearity (R-2 for all GCs, 0.986-0.996), sensitivity (limit of detection, 2.0-50 ng/mL), and repeatability (RSD, 1.2-6.0%). The entire analytical process can be finished in 5 min, compared with the GC/MS or LC/MS methods for which typical analysis times range from tens of minutes to > 1 h. In addition, comparison of the performance of DART and ESI ion sources showed that DART possessed a relatively low matrix effect when handling complex samples. CONCLUSIONS: A new method for the rapid screening and quantification of GCs in essential oils was developed using DART-QTOF MS. Copyright (C) 2016 John Wiley & Sons, Ltd.
Stability-indicating HPLC-DAD assay for simultaneous quantification of hydrocortisone 21 acetate, dexamethasone, and fluocinolone acetonide in cosmetics

Abstract:Abstract A rapid and specific method was developed for simultaneous quantification of hydrocortisone 21 acetate (HCA), dexamethasone (DEX), and fluocinolone acetonide (FCA) in whitening cream formulations using reversed-phase high-performance liquid chromatography. The effect of the composition of the mobile phase, analysis temperature, and detection wavelength was investigated to optimize the separation of studied components. The analytes were finally well separated using ACE Excel 2, C18 AR column having 150 mm length, 3 mm internal diameter, and 2 μm particle size at 35°C using methanol with 1% formic acid and double-distilled deionized water in the ratio of 60:40 (v/v), respectively, as the mobile phase in isocratic mode. Ten microliters of sample were injected with a flow rate of 0.5 mL/min. The specificity, linearity, accuracy, precision, recovery, limit of detection (LOD), limit of quantification (LOQ), and robustness were determined to validate the method as per International Conference on Harmonization guidelines. All the analytes were simultaneously separated within 8 min, and observed retention times of HCA, DEX, and FCA were 4.5, 5.5, and 6.9 min, respectively. The proposed method showed good linearity with the correlation coefficient, R 2 = 0.999 over the range of 1–150 μg/mL for all standards. The linear regression equations were y = 12.7 x + 118.7 ( r = 0.999) for HCA, y = 12.9 x + 106.8 ( r = 0.999) for DEX, and y = 12.9 x + 96.8 ( r = 0.999) for FCA. The LOD was 0.25, 0.20, and 0.08 μg/mL for HCA, FCA, and DEX and LOQ was 2.06, 1.83, and 1.55 μg/mL for HCA, FCA, and DEX, respectively. The recovery values of HCA, DEX, and FCA ranged from 100.7–101.3, 102.0–102.6, and 100.2–102.0%, respectively, and the relative standard deviation for precision (intra- and interday) was less than 2, which indicated repeatability and reproducibility. The novelty of the method was described by forced degradation experimentation of all analytes in the combined form under acidic, basic, oxidative, and thermal stress. The proposed method was found to be simple, rapid, and reliable for the simultaneous determination of HCA, DEX, and FCA in cosmetics.

Abstract:Abstract Background: Measurements of serum or urine concentrations of synthetic glucocorticoids are useful for assessing suspected iatrogenic hypothalamic-pituitary-adrenal axis suppression and Cushing syndrome. We have developed a liquid chromatography–tandem mass spectrometry (LC-MS/MS) assay for the simultaneous quantitative analysis of beclomethasone dipropionate, betamethasone, budesonide, dexamethasone, fludrocortisone, flunisolide, fluorometholone, fluticasone propionate, megestrol acetate, methylprednisolone, prednisolone, prednisone, triamcinolone, and triamcinolone acetonide. Methods: Stable isotopes of cortisol-9,11,12,12-d4 and triamcinolone-d1 acetonide-d6 were added as internal standards to calibrators, controls, and unknown samples. After acetonitrile precipitation, these samples were extracted with methylene chloride, and the extracts were washed and dried. Reconstituted extract (15 μL) was injected on a reversed-phase column and analyzed by LC-MS/MS in positive-ion mode. Assay precision, accuracy, linearity, and sample stability were determined by use of enriched samples. Clinical validation included analysis of 8 serum and 20 urine samples from patients with undetectable cortisol concentrations and analysis of different types of tablets. Results: Functional assay sensitivity was as low as 0.6–1.6 nmol/L for all compounds except for triamcinolone (7.6 nmol/L). Interassay CVs were 3.0–20% for concentrations of 0.6–364 nmol/L for all analytes. Recoveries of all analytes (except triamcinolone in serum) were 82–138% at 19.2–693 nmol/L. All but one of the serum and urine samples from patients who were tested because of suppressed cortisol concentrations contained at least one synthetic steroid. Tablet analysis recovered 75% of the synthetic steroids in suspected drugs. Conclusions: LC-MS/MS allows simultaneous quantitative detection of various synthetic steroids in serum, plasma, urine, and tablets. This provides a valuable tool for evaluating the clinical effects of topical and systemic synthetic corticosteroids.

Abstract:A comprehensive analytical method has been developed and validated for the simultaneous determination of seventeen glucocorticoid residues in eggs and milk. The mass spectrometer parameters, the composition of the mobile phase and the sample preparation method were firstly optimized to obtain maximum sensitivity. The samples were deconjugated with beta-glucuronidase/arylsulfatase enzyme and concentrated using an Oasis HLB solid-phase extraction cartridge, followed by cleanup with a dual Sep-pak silica and aminopropyl cartridge. The analytes were quantified by ultra-performance liquid chromatography (using a C18 column)/electrospray ionization tandem mass spectrometry (UPLC/ESI-MS/MS) operating in the negative ion mode. The assay for the 17 glucocorticoids was linear over the range of 1-200 microg/L for milk and egg samples with a high correlation coefficient (>0.99). The limits of quantification (LOQs) for the target analytes were 0.04-1.27 microg/kg for the egg samples and 0.03-0.73 microg/kg for the milk samples. The average extraction recoveries of the glucocorticoids from eggs and milk at two concentration levels (spiked at 0.40 and 2.00 microg/kg) were 65.6-118.7% and 61.5-119.6%, respectively, with relative standard deviations between 1.8-17.0% and 2.4-18.4%, respectively. Because of its high sensitivity, good precision and specificity, the method was found to be suitable for trace analysis of synthetic and natural glucocorticoids in complex biosamples such as eggs and milk.

Abstract:Rationale Illegal addition of anti‐infective drugs to cosmetics at low concentrations has been found. The illicit addition of anti‐infective drugs encompasses a wide variety of medications. The current sample purification methods are inadequate to detect all these compounds. A sensitive, wide‐coverage, and weak‐matrix‐effect measurement method needs to be established to address this issue. Methods Samples were extracted using acetonitrile, diluted 25 times, and then analyzed using liquid chromatography–tandem mass spectrometry (LC–MS/MS) to detect 111 anti‐infective drugs. The method was validated and assessed for matrix effect before being applied to cosmetic products. Results The calibration curves for the analytes exhibited a strong correlation coefficient ( r > 0.995). The limit of detection ranged from 0.006 to 0.6 mg/kg. Matrix effects were significantly improved after a 25‐fold dilution. The method was successfully applied to various cosmetics. Two of 82 samples tested contained lincomycin and miconazole, respectively. Conclusions The developed method is quick and reliable to analyze anti‐infective drugs in cosmetics, with potential for both qualitative and quantitative analyses. It is a valuable tool for cosmetic research and development, contributing to safer and more effective cosmetic products.

Abstract:An analytical method has been developed and validated for the simultaneous determination of 18 pharmaceuticals and personal care products (PPCPs), including antibiotics (trimethoprim, erythromycin x 2H2O, norfloxacin, ofloxacin, pencilline G, penicillin V potassium salt, cephalexin and sulfamethoxazole), beta-bloker (atenolol), anophelifuge (N, N-diethyl-3-methylbenzoylamide, DEET), antiepileptics (carbamazepine), central nervous system stimulant (caffeine), lipid modifying agent (clofibric acid), non-steroidal anti-inflammatory drugs (ibuprofen, naproxen and diclofenac sodium salt) and antimicrobial agents (triclosan and triclocarban). The detection and qualification of the target compounds were performed by ultra-high performance liquid chromatography-tandem mass spectrometry. The optimized mobile phases were methanol as organic phase, 0. 3% (v/v) formic acid-5 mmol/L ammonium acetate for positive electrospray ionization (ESI+) and 5 mmol/L ammonium acetate for ESI- as inorganic phase. Water samples were concentrated by solid phase extraction at 2 mL/min, and all the target PPCPs were efficiently extracted at pH 7. The extracted PPCPs were eluted by the mixture of methanol and acetonitrile (1 : 1, v/v). The average recoveries of the target compounds in the spiked pure water samples ranged from 53.9% - 112%. The average recoveries of the target compounds ranged from 45.1% - 156.6% with the relative standard deviations ranged from 2.4% - 15.7%, in the surface water samples spiked at 100 ng/L. The surface water samples collected from Yu Hangtang River in Hangzhou were detected. The results showed that nine PPCPs were detected including caffeine that reached a maximum concentration of 550.7 ng/L. It proved that this analytical method is reliable and acceptable.
Determination Of Formaldehyde Residue In Cosmetics By Short-Column High Performance Liquid Chromatography With Mass Spectrometric Confirmation

Abstract:In this work, we report a rapid analytical method for the determination of trace formaldehyde in cosmetics on a 12.5 mm C-18 guard column. The shortest time (0.5 min) was obtained for one separation with 1.5 mL min(-1) of methanol-water 50 : 50 (v/v) as the mobile phase at a column temperature of 30 degrees C. A linear response between peak area and the concentration of formaldehyde was obtained over the range 0.1 to 20 mg L-1 with a relative standard deviation of 1.4% (n = 10) at the concentration of 1.0 mg L-1 and a detection limit of 5.1 mg L-1. The confirmation assay by liquid chromatography tandem mass spectrometry indicated that there was no interfering effect from other carbonyl compounds. The proposed method was applied for the determination of formaldehyde residue in cosmetics at the mg L-1 level. Free formaldehyde up to 27 mg kg(-1) was found in cosmetics and the spike recovery at the spiked levels of 1.0 and 10.0 mg L-1 varying from 92% to 105% was also obtained. The difference between the values determined by the proposed method and the standard spectrophotometric method was below 3.7%. The results proved the accuracy and precision of the method, showing potential for the routine analysis of formaldehyde residue.
An Eco-Friendly HPLC-DAD Determination of Corticosteroids Combined with Other Therapeutic Compounds in Bulk and Topical Formulations: Greenness and Whiteness Assessment

Abstract:Creams are among the most analytically challenging dosage forms both in product analysis and in cleaning validation. This is especially true when highly lipophilic drugs are the active ingredients. The case is getting more challenging with the greenness emerges as an evaluation criterion where solvents and wastes should be minimized. Green methods are difficult to achieve with lipophilic drugs and creams that may encounter high carry-over tendency. In this study, an ecofriendly RP-HPLC-DAD method was developed for the determination of hydrocortisone acetate ( HCA ), mometasone furoate ( MOM ), betamethasone valerate ( BTM ), clotrimazole ( CLT ) and oxytetracycline ( OXT ) in the presence of miconazole as examples of lipophilic drugs commonly formulated in creams. The separation was performed on a C 18 (5 μm, 150 × 4.6 mm ID) column using methanol–water (80 : 20, v/v) as a mobile phase and a photo-diode array detector. The method was validated according to ICH guidelines. Greenness and whiteness were assessed by NEMI, GAPI, AGREE, Eco-scale, and the RGB 12 model and compared to currently available methods. The developed method was found linear with r 2 > 0.99 within the range of 0.05–9.00 HCA, 0.1–9.0 MOM, 0.2–9.0 BTM, 0.6–9.0 CLT, and 5.0–9.0 μg/mL OXT. The accuracy (98.4 ± 1.7)–(102.3 ± 1.5) and precision (CV < 10%) were in acceptable ranges while retaining high sensitivity (LOD down to 15 ng). The proposed method proved to be environmentally friendly by applying different greenness and whiteness algorithms and was successfully applied to different cream formulations. In conclusion, the developed method outperforms currently available methods for the determination of tested lipophilic compounds in creams.

Abstract:The objective of this study was to establish a novel method for rapid detection of six glucocorticoids (prednisone, prednisone acetate, prednisolone, hydrocortisone, hydrocortisone acetate, and dexamethasone) added illegally in dietary supplements simultaneously by combining thin layer chromatography (TLC) with spot-concentrated Raman scattering (SCRS). The doping ingredients were separated by TLC, and viewed and located with UV light (254 nm), enriched by chromatography, then Raman spectra were directly detected by a Raman Imagine microscope with 780 nm laser source. This method had complementary advantages of TLC and Raman spectroscopy, which enhanced the specificity of the test results. The limit of detection (LOD) of the reference substances were 4 μg, 4 μg, 4 μg, 6 μg, 6 μg, and 4 μg, respectively. The method was used to study the six glucocorticoids added illegally in five dietary supplements. Fake drugs had been detected. The study showed that the TLC-SCRS method is simple, rapid, specific, sensitive, and reliable. The method could be used for effective separation and detection of six chemical components used in dietary supplement products, and would have good prospects for on-site qualitative screening of dietary supplement products for adulterants.

Abstract:Various types of oxidative dyes used in hair dye products possess poor stability and have varying frequency of use. Interference problems also frequently arise in actual sample measurements. Therefore, it is necessary to establish a simple, rapid, accurate, and specific method for the determination of common dyes in hair dye products for their effective regulation. In this study, dyes were grouped according to their frequency of use. Using a C18 column that minimizes the silanol effect and influence of metals, the quantitative high performance liquid chromatography (HPLC) method for 32 dyes listed in Safety and Technical Standards for Cosmetics (2015 edition) was optimized, and a high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) confirmatory method for the dyes was established. The samples were extracted using a mixed solution of ethanol-water (1∶1, v/v) with 10 g/L sodium bisulfite solution as an antioxidant, vortexed and mixed, and then extracted by ultrasonication in an ice bath for 10 min. Methanol, acetonitrile, and phosphate buffer were used as the mobile phases in the HPLC analysis. Additionally, two different elution conditions (chromatographic gradient) were used for the separation of 32 oxidative dyes, which were detected at a wavelength of 280 nm. The HPLC separations were compared using columns of particle sizes 5 μm and 2.7 μm; 5 μm C18 columns with better anti-interference and antiblocking ability were selected. Satisfactory separation was achieved for all three commercial C18 columns with a particle size of 5 μm, and the method had good general usability. In condition 1, 17 commonly used dyes and three less commonly used dyes were assigned to group Ⅰ and separated by HPLC; in condition 2, eight banned dyes and four other less commonly used dyes were assigned to group Ⅱ and separated by HPLC. The HPLC-MS/MS method used 5 mmol/L ammonium acetate aqueous solution-acetonitrile and 5 mmol/L acetic acid aqueous solution-acetonitrile as mobile phases in the positive and negative ion modes, respectively. Multiple reaction monitoring (MRM) was performed for qualitative and quantitative analyses in the electrospray ionization mode. Under the examined conditions, six pairs of isomers were well resolved. For the HPLC and HPLC-MS/MS methods, the relative standard deviations (RSDs) of the intra-day precision and 48 h stability tests were less than 10%. The recoveries were between 82.6% and 114.9% (RSD<10%). In the HPLC method, 32 dyes showed good linearity in an approximate range of 10-500 mg/L (r2>0.99), and the limits of detection (LODs) were 9.7-40.1 μg/g. The linear range of hydroquinone in the HPLC-MS/MS method was 2.0-79.7 mg/L, and the LOD was 8.0 μg/g; the linear ranges of the other components were approximately 0.1-4 mg/L, and the LODs were 0.01-0.4 μg/g. The actual samples were simultaneously measured by HPLC, HPLC-MS/MS, and the standard method. Finally, 16 of the 32 dyes were detected, and the detected contents ranged from 58 to 25160 μg/g. The RSDs of the results obtained from the three detection methods were between 1.9% and 10.1%. All detected components were within the limits of group Ⅰ of this method. In comparison with methods reported in the literature and the standard method, this method covers all components for the routine regulatory inspection of oxidative dyes in cosmetics. The method can separate the commonly used dyes under the same HPLC conditions and avoid interference from 15 other commonly used dyes during the analysis of actual samples. A suitable HPLC-MS/MS confirmatory method was also established for the identification of currently unknown substances in the statutory inspection of cosmetics. The method is simple, rapid, accurate, and specific with general usability and good operability.
Development and Validation of HPLC–Photodiode Array Method for Detecting Steroids in Skin Whitening Products Simultaneously

Abstract:Since steroids are prohibited to be present in cosmetic products, thus an identification of these outlawed compounds intentionally incorporated in skin whitening product is enough for market surveillance purpose. Simultaneous method for detecting prednisone (PRN), triamcinolone acetonide (TCA), hydrocortisone acetate (HCA), clobetasole propionate (CBP), and mometasone furoate (MTF) has not been developed until now. The goals of this study were to develop and to validate for screening these five steroid compounds in cosmetic product by using reverse phase (RP) high-performance liquid chromatography (HPLC) equipped with photo diode array (PDA) detector. Validation method was applied including the system suitability test, determination of specificity and selectivity, and determination of detection limit. . In optimization step, we succeeded distinguish and identify five steroid compounds with different retention time.The retention times of PRN, TCA, HCA, CBP and MTF were detected at 3.267, 4.260, 4.910, 15.360, and 16.707 min, respectively. The method performed good selectivity with resolution more than 1.5. While for determination of limit of detection (LOD) was carried out by calibration curve approach over the range 0.5-5 μg/ml and resulted in LOD value for PRN, TCA, HCA, CBP and MTF were 7.20, 8.85, 8.17, 9.30, and 8.42 μg/g, respectively. The developed method can be applied for routine analysis of PRN, TCA, HCA, CBP and MTF in various type of cosmetic products.
Simultaneous determination of eight alkaloids and oleandrin in herbal cosmetics by dispersive solid-phase extraction coupled with ultra high performance liquid chromatography and tandem mass spectrometry

Abstract:We utilized ultra-high performance liquid chromatography with tandem mass spectrometry and dispersive solid-phase extraction to develop a new method for the detection of nine analytes (scopolamine, cephaeline, strychnine, hyoscyamine, brucine, hydrastine, ajmalicine, colchicine, and oleandrin) in herbal cosmetics. Acetonitrile/water and 2-propylaminoethylamine were used to disperse and purify during the dispersive solid-phase extraction step. The analytes were separated by a Waters UPLC HSS T3 column and detected through electrospray ionization source in the positive mode with multi-reaction monitoring conditions. Under the optimal conditions, the calibration curves were linear in the range of 0.2-100.0 μg/L with the correlation coefficients higher than 0.995. The method limit of quantitation (S/N = 10) were 5.0 μg/kg for oleandrin and 1.0 μg/kg for the other eight alkaloids. The mean recoveries at three spiked concentration levels of 1.0-10.0 μg/kg were in the range of 86.9-116.5% with the intra-day relative standard deviations (n = 6) ranging from 2.4 to 8.8%, and inter-day relative standard deviations ranging from 2.7 to 5.7%. This method is accurate, simple and rapid, and has been applied to the quality supervision of herbal cosmetics in Guangzhou.
Multi-detection of corticosteroids in sports doping and veterinary control using high-resolution liquid chromatography/time-of-flight mass spectrometry

Abstract:A liquid chromatography/time-of-flight mass spectrometry (LC/TOFMS) method was developed using the latest high-resolution LC column technology, the ultra performance liquid chromatography (UPLC), and electrospray ionization (ESI) in the positive ion mode. Gradient UPLC separation conditions were optimized for a group of 22 analytes comprising 17 glucocorticosteroids, specific designer steroids such as tetrahydrogestrinone (THG) and specific beta2-agonists such as formoterol. The UPLC/TOFMS separation obtained required 5.5 min only for all the substances tested. Even the critical pair of dexamethasone and betamethasone isomers was almost completely resolved. Thanks to the over 10,000 full-width at half maximum (FWHM) mass resolution and high mass accuracy features of TOFMS 50 mDa window accurate mass chromatograms could be reconstructed for the individual analytes. Sensitive screening in human and calf urine samples fortified at the glucocorticosteroids minimum required performance limit (MRPL) of 30 microg L(-1) (human urine, sports doping) and 2 microg L(-1) (calf urine, veterinary control) could be obtained. The potential of UPLC/TOFMS for confirmatory analysis was shown by determining the accurate mass of all compounds and fragment ions upon in-source collision-induced dissociation (CID) at different energies. The exact mass measurement errors for all glucocorticosteroids were found to be within 6 ppm. Considering veterinary control, limits of detection (LOD) and limits of quantification (LOQ) were determined for most of the analytes in calf urine and found to range from 0.1 to 3.3 and from 0.4 to 4.4 microg L(-1), respectively. The method can be easily extended with other banned substances of interest, as demonstrated by the addition of 21 beta2-agonists to the original analyte mixture in urine, without causing any interferences.

Abstract:A comprehensive analytical method based on ultra-performance liquid chromatography coupled with tandem mass spectrometry (UPLC-MS/MS) has been developed for the simultaneous determination of 22 regulated coumarin derivatives in cosmetics. Various categories of cosmetic samples (creams, lotions, shampoos, and lipsticks) were subjected to optimized ultrasound-assisted extraction and solid-phase extraction procedures, which enabled exhaustive extraction of target analytes and efficient cleanup of sample matrices prior to instrumental analysis. The rapid chromatographic separation of the 22 coumarin derivatives was performed on a sub-2 µm Waters ACQUITY UPLC BEH C18 column (50 mm × 2.1 mm, 1.7 µm), preceded by a pre-column of the same packing material within 5 min. The entire column eluent from UPLC was directed into a triple quadrupole mass spectrometer with an electrospray ionization source. The MS/MS data acquisition was conducted under the mode of multiple reaction monitoring (MRM) with two precursor-product ion transitions recorded but only one used for quantitation. The proposed method was validated in terms of matrix effects, linearity, sensitivity, accuracy, precision, specificity, and stability, and could be applied for the screening and monitoring of these prohibited or restricted coumarin derivatives in cosmetics to ensure consumers’ health and protection.

Abstract:To objectively quantify changes in steroid hormones in organisms caused by adverse environmental loads, we developed a simple and sensitive UPLC-MS/MS (ultra-performance liquid chromatography triple quadrupole mass spectrometry) method for the simultaneous determination of 18 steroid hormones on the HPG axis. This analytical method was based on liquid extraction and a multimode electrospray and atmospheric pressure chemical ionization (ESCi) source, which was optimized by mass spectrometry, liquid phase and pretreatment for the quantification of cholesterol (CH), aldosterone (A), cortisone (E), hydrocortisone (F), 21-deoxycortisol (21-DF), corticosterone (B), 11-deoxycortisol (11-DF), androstenedione (A2), estradiol (E2), estrone (E1), 2-methoxyestradiol (2-MeE2), 21-hydroxyprogesterone (21-OHP), 17-α hydroxyprogesterone (17α-OHP), testosterone (T), dehydroepiandrosterone (DHEA), progesterone (P4), dihydrotestosterone (DHT), and pregnenolone (P5). The method exhibits linearity in the analyte-concentration range 0.03-1000 μg mL-1 (r2 > 0.99), the spiked recoveries for the concentration range tested are 76.22-113.66%, and the relevant parameters of precision are 7.52-1.14%. Compared to other methods, this new method not only uses a small amount of serum (only 100 μL), but also permits the analysis of the challenging steroid, cholesterol. Furthermore, the method was successfully applied to the determination of steroids in Mus musculus, Carassius auratus, Rana catesbeiana Shaw, and Rana nigromaculata serum samples from randomly selected individuals. Therefore, this method is efficient and a very useful tool for assessing changes in steroid hormones.

Abstract:Fragrances are among the most common ingredients in cosmetics products. Importantly, exposure to fragrances on a daily basis might pose a health risk, leading to serious effects, such as contact dermatitis or contact eczema. Annex III of the European Union Directive on Cosmetic Products and Gulf Cooperation Council standardization organization (GSO) introduced restrictions for 26 allergens, with their concentrations exceeding 0.001% and 0.01% in leave-on products and rinse-off products, respectively. In the current study, we aimed to expand the scope of the analytical method (EN16274, 2012) to include a broader range of matrices. The optimized method was validated by examining a statistical approach, including selectivity, linearity, accuracy, precision, and measurement of uncertainty. Successfully, the validated data demonstrated acceptable limits according to validation protocols, with linearity showing satisfactory regression of r > 0.995. During method performance assessment, samples were extracted using ultrasound-assisted extraction to extract allergens that yielded relatively high recoveries. Studies on matrices spiked with allergens at different levels showed insignificant bias as an average of 0.07 μg/g. Method performance was assessed by analyzing 140 cosmetics samples, including perfumes, deodorants, aftershave, baby wet wipes, shampoos, lotions, and lip care products. The new optimized analytical method is believed to be a valuable analytical tool to be used in surveillance studies covering a wide range of cosmetic matrices.
