Abstract:Ion mobility spectrometry (IMS) opened a potential avenue for the rapid detection of organophosphorus pesticides (OPPs), though an improved selectivity of stand-alone IMS was still in high demand. In this study, a stand-alone positive photoionization ion mobility spectrometry (PP-IMS) apparatus was constructed for the rapid detection of OPPs with acetone as dopant. The photoionization of acetone molecules was induced by the ultraviolet irradiation to produce the reactant ions (Ac)2H+, which were employed to ionize the OPPs including fenthion, imidan, phosphamidon, dursban, dimethoate and isocarbophos via the proton transfer reaction. Due to the difference in proton affinity, the tested OPPs exhibited the different dopant-dependent manners. Based on this observation, the switching of dopant concentration was implemented to improve the selectivity of PP-IMS for OPPs detection. For instance, a mixture of fenthion, dursban and dimethoate was tested. By switching the concentration of doped acetone from 0.07 to 2.33 to 19.94mgL-1, the ion peaks of fenthion and dursban were inhibited in succession, achieving the selective detection of dimethoate at last. In addition, another mixture of imidan and phosphamidon was initially detected by PP-IMS with a dose of 0.07mgL-1 acetone, indicating that their ion peaks were severely overlapped; when the concentration of doped acetone was switched to 19.94mgL-1, the inhibition of imidan signals promised the accurate identification of phosphamidon in mixture. Finally, the PP-IMS in combination of switching dopant concentration was applied to detect the mixed fenthion, dursban and dimethoate in Chinese cabbage, demonstrating the applicability of proposed method to real samples.

Abstract:The performance of gas chromatography (GC) combined with the improved identification properties of ion mobility separation coupled to high-resolution mass spectrometry (IMS-HRMS) is presented as a promising approach for the monitoring of (semi)volatile compounds in complex matrices. The soft ionization promoted by an atmospheric pressure chemical ionization (APCI) source designed for GC preserves the molecular and/or quasi-molecular ion information enabling a rapid, sensitive, and efficient wide-scope screening. Additionally, ion mobility separation (IMS) separates species of interest from coeluting matrix interferences and/or resolves isomers based on their charge, shape, and size, making IMS-derived collision cross section (CCS) a robust and matrix-independent parameter comparable between instruments. In this way, GC-APCI-IMS-HRMS becomes a powerful approach for both target and suspect screening due to the improvements in (tentative) identifications. In this work, mobility data for 264 relevant multiclass organic pollutants in environmental and food-safety fields were collected by coupling GC-APCI with IMS-HRMS, generating CCS information for molecular ion and/or protonated molecules and some in-source fragments. The identification power of GC-APCI-IMS-HRMS for the studied compounds was assessed in complex-matrix samples, including fish feed extracts, surface waters, and different fruit and vegetable samples.

Abstract:Ion Mobility Spectrometry (IMS) has gained relevance in the field of analytical techniques over the past decades. If compared with well-established techniques like mass spectrometry or infrared spectroscopy, IMS is considerably less developed or employed in specific fields but presents promising results and a substantial margin for improvements. Its outstanding sensitivity and selectivity, analytical flexibility, instrumental versatility and almost real-time results capacity have contributed to elevate IMS among the main analytical techniques for the detection of volatile organic compounds. Due to its growth potential, it is mandatory to assess in which scientific fields IMS has played a relevant role in the past years of academic research and understand in which areas it can become equally important in the near future. For this purpose, hundreds of scientific works from the past ten years were addressed and the most relevant were reviewed in this work. Three main categories of IMS applications were defined to group the reviewed articles: Environmental and Safety Research, Health Research and Food Research. In addition, some original studies were specifically developed for this review paper, to act as elucidative examples. The working principle of the IMS is included for clarification purposes. A glossary of all the mentioned compounds was also included. Throughout the text, it is clear how relevant IMS has become and how diverse its applicability can be, ranging from simpler topics like fraud detection to more complex ones like pathologies diagnosis. It is safe to say that IMS has been, step by step, gaining relevance as an analytical technique and its potential for supporting many diverse scientific fields is evident.

Abstract:A screening method in the combination of ultrasonic extraction,gas chromatogra-phy-mass spectrometry detection and automated mass spectrometry deconvolution technique was developed for non-target screening of non-polar and weak polar pollutants in sediments and sludges. The samples were extracted by ultrasonication for 20 min using dichloromethane for three times. The extraction solutions were cleaned-up by gel permeation chromatography and a silica gel column,and then 3 g of copper powder was used to remove the sulfur by ultrasonica-tion for 10 min. Parallel experiments were carried out for 5 times and the RSDs were ranged from 5. 8% to 14. 9%. Automated mass spectral deconvolution &identification system( AMDIS) would improve the resolution of overlapping peaks,and identify the pure mass spectrum of the analytes in the cases of stronger background interference and co-extracted substances covering. Standard spectrum databases,such as NISTDRUG,NISTEPA,NISTFDA,Mass Spectral Library, etc,would qualitatively identify the organic pollutants in the samples. As a result,a total of 290 organic pollutants were identified,of which 190 and 153 pollutants were found in sediments and sludges,respectively. The identified pollutants included the Environmental Protection Agency (EPA)priority pollutants,pharmaceuticals,herbicides,antioxidants,intermediates,organic solvents and chemical raw materials. The proposed method is proved to be a promising one for non-target screening of complex matrix samples with the advantages of higher sensitivity and better repeatability.
Evaluation of Ion Mobility Spectrometry for Improving Constitutional Assignment in Natural Product Mixtures

Abstract:The comprehensive chemical characterization of biological samples remains a central challenge in the field of natural products. Conventional workflows using liquid chromatography (LC)-coupled high-resolution tandem mass spectrometry (MS/MS or MS2) allow the detection of relevant small molecules while providing diagnostic fragment ions for their structural assignment. Still, many natural product extracts are of a molecular complexity that challenges the resolving power of modern LC-MS2 pipelines. In this study, we examined the effect of integrating ion mobility spectrometry (IMS) to our LC-MS2 platform for the characterization of natural product mixtures. IMS provides an additional axis of separation in the gas phase as well as experimental collision cross-sectional (CCS) values. We analyzed a mixture of 20 commercial standards at 2 concentration ranges, either solubilized in solvent or spiked into an actinobacterial extract. Data were acquired in positive ion mode using both data-dependent acquisition (DDA) and data-independent acquisition (DIA) MS2 fragmentation approaches and assessed for both chemical coverage and spectral quality. IMS-DIA identified the largest number of standards in the spiked extract at the lower concentration of standards (17), followed by IMS-DDA (10), DDA (8), and DIA (6). In addition, we examined how these data sets performed in the Global Natural Products Social Molecular Networking (GNPS) platform. Overall, integrating IMS increased both metabolite detection and the quality of MS2 spectra, particularly for samples analyzed in DIA mode.

Abstract:High-resolution mass spectrometry (HRMS)-based suspect and nontarget screening techniques are powerful tools for the comprehensive identification of per- and polyfluoroalkyl substances (PFASs), but the interference of complex matrices (especially for wastewater) pose an analytical challenge. This study explored the potential of combining ion mobility spectrometry (IMS) with HRMS and machine learning techniques to achieve the rapid and accurate suspect and nontarget screening of PFAS in wastewater. There were fewer interfering peaks and a clearer spectrum in the data acquired by IMS-HRMS than conventional HRMS. The introduction of collision cross section (CCS) in PFAS homologous series search could filter out 63% of false positive results. Retention time and CCS prediction models were helpful in improving the confidence for PFAS qualitative identification and the random forest algorithm combined with RDKit descriptor performed best for CCS prediction. With the inclusion of extra dimensional information, this study also proposed a comprehensive and concise confidence assignment criterion to better convey the certainty of the qualitative identification of PFAS. Finally, a total of 56 potential PFASs were identified in the wastewater sample using the newly developed method and 45 of them were identified outside reference standards, emphasizing the importance of suspect and nontarget screening for PFAS.

Abstract:Ion mobility spectrometry‐mass spectrometry (IMS‐MS or IM‐MS) is a powerful analytical technique that combines the gas‐phase separation capabilities of IM with the identification and quantification capabilities of MS. IM‐MS can differentiate molecules with indistinguishable masses but different structures (e.g., isomers, isobars, molecular classes, and contaminant ions). The importance of this analytical technique is reflected by a staged increase in the number of applications for molecular characterization across a variety of fields, from different MS‐based omics (proteomics, metabolomics, lipidomics, etc.) to the structural characterization of glycans, organic matter, proteins, and macromolecular complexes. With the increasing application of IM‐MS there is a pressing need for effective and accessible computational tools. This article presents an overview of the most recent free and open‐source software tools specifically tailored for the analysis and interpretation of data derived from IM‐MS instrumentation. This review enumerates these tools and outlines their main algorithmic approaches, while highlighting representative applications across different fields. Finally, a discussion of current limitations and expectable improvements is presented.

Abstract:Ion mobility spectrometry (IMS) has been widely studied and applied as an effective analytical technology for the on-site detection of volatile organic compounds (VOCs). Despite its superior selectivity compared with most gas sensors, its limited dynamic range is regarded as a major drawback, limiting its further application in quantitative measurements. In this work, we proposed a novel sample introduction method based on pulsed membrane adsorption, which effectively enhanced IMS's ability to measure analytes at higher concentrations. Taking N-methyl-2-pyrrolidone (NMP) as an example, this new sampling method expanded the dynamic range from 1 ppm to 200 ppm. The working principle and measurement strategy of this sampling method were also discussed, providing new insights for the design and application of IMS-based instruments.

Abstract:Ion mobility spectrometry (IMS) is a rapid separation technique that has experienced exponential growth as a field of study. Interfacing IMS with mass spectrometry (IMS-MS) provides additional analytical power as complementary separations from each technique enable multidimensional characterization of detected analytes. IMS separations occur on a millisecond timescale, and therefore can be readily nested into traditional GC and LC/MS workflows. However, the continual development of novel IMS methods has generated some level of confusion regarding the advantages and disadvantages of each. In this critical insight, we aim to clarify some common misconceptions for new users in the community pertaining to the fundamental concepts of the various IMS instrumental platforms (i.e., DTIMS, TWIMS, TIMS, FAIMS, and DMA), while addressing the strengths and shortcomings associated with each. Common IMS-MS applications are also discussed in this review, such as separating isomeric species, performing signal filtering for MS, and incorporating collision cross-section (CCS) values into both targeted and untargeted omics-based workflows as additional ion descriptors for chemical annotation. Although many challenges must be addressed by the IMS community before mobility information is collected in a routine fashion, the future is bright with possibilities.

Exploiting Molecular Ions for Screening Hydrophobic Contaminants in Sediments using Gas Chromatography-Atmospheric Pressure Chemical Ionization-Ion Mobility-Mass Spectrometry

Abstract:Hydrophobic organic compounds (HOCs) are conventionally screened by matching experimental- to reference electron ionization (EI) mass spectra acquired using gas chromatography-mass spectrometry (GC-MS). However, for identification of novel substances that are absent from EI databases, de novo structure elucidation using EI is hampered by extensive in-source fragmentation. To address this problem, a new method based on GC-atmospheric pressure chemical ionization (APCI) coupled to ion mobility-high resolution mass spectrometry (IM-HRMS) was developed for simultaneous target, suspect, and non-target screening of HOCs. Of 102 target chemicals, 85.3% produced (quasi-)molecular ions as base peaks, with 71.6% displaying method detection limits lower than those of GC-EI-low resolution MS. The optimized method was applied to standard reference sediment and sediments from the Baltic Sea, an Arctic shelf, and a Norwegian lake. In total, we quantified 56 target chemicals with concentrations ranging from 4.86 pg g-1 dwt to 124 ng g-1 dwt. Further, using a combination of exact mass, collision cross section (CCS), retention time, and MS2 spectra, a total of 54 suspects were identified at Confidence Level (CL) 2. Among the remaining features, 169 were prioritized using a halogen-selective CCS cutoff (100 Å2 + 20% mass), leading to annotation 54 substances (CL ≤3). Notably, a suite of fluorotelomer thiols, disulfides, and alkyl sulfones were identified in sediment (CL 1-2) for the first time. Overall, this work demonstrates the potential of GC-APCI-IM-HRMS as a next-generation technique for resolving complex HOC mixtures in environmental samples through exploitation of molecular ions.

Abstract:In the last decade, ion mobility spectrometry (IMS) has reemerged as an analytical separation technique, especially due to the commercialization of ion mobility mass spectrometers. Its applicability has been extended beyond classical applications such as the determination of chemical warfare agents and nowadays it is widely used for the characterization of biomolecules (e.g., proteins, glycans, lipids, etc.) and, more recently, of small molecules (e.g., metabolites, xenobiotics, etc.). Following this trend, the interest in this technique is growing among researchers from different fields including food science. Several advantages are attributed to IMS when integrated in traditional liquid chromatography (LC) and gas chromatography (GC) mass spectrometry (MS) workflows: (1) it improves method selectivity by providing an additional separation dimension that allows the separation of isobaric and isomeric compounds; (2) it increases method sensitivity by isolating the compounds of interest from background noise; (3) and it provides complementary information to mass spectra and retention time, the so-called collision cross section (CCS), so compounds can be identified with more confidence, either in targeted or non-targeted approaches. In this context, the number of applications focused on food analysis has increased exponentially in the last few years. This review provides an overview of the current status of IMS technology and its applicability in different areas of food analysis (i.e., food composition, process control, authentication, adulteration and safety).

Abstract:Membrane-extraction ion mobility spectrometry (ME-IMS) has been developed for in situ sampling and analysis of trace chlorinated hydrocarbons in water in a single procedure. The sampling is configured so that aqueous contaminants permeate through a spiral hollow poly(dimethylsiloxane) (PDMS) membrane and are carried away by a vapor flow through the membrane tube. The extracted analyte flows into an atmospheric-pressure chemical-ionization (APCI) chamber and is analyzed in a specially made IMS analyzer. The PDMS membrane was found to effectively extract chlorinated hydrocarbon solvents from the liquid phase to vapor. The specialized IMS analyzer has measured resolutions of R = 33 and 41, respectively, for negative- and positive-modes and is capable of detecting aqueous tetrachloroethylene (PCE) and trichloroethylene (ICE) as low as 80 and 74 mu g/L in the negative ion mode, respectively. The time-dependent characteristics of sampling and detection of ICE are both experimentally and theoretically studied for various concentrations, membrane lengths, and flow rates. These characteristics demonstrate that membrane-extraction IMS is feasible for the continuous monitoring of chlorinated hydrocarbons in water.

Abstract:Chemical ionization mass spectrometry (CIMS) is a powerful tool for on-line characterization of atmospheric trace species and reveal of new atmospheric oxidation mechanisms. The applications of CIMS promoted understanding of new particle formation and the roles of trace species in air-quality issues. In the past decade or so, various new techniques in ion sources have been applied to improve sensitivity and replace radioactive sources. Moreover, researchers have developed multiple sampling inlets for CIMS to meet the measurement demand of reactive trace species. In this study, we discussed these developments and reviewed a limited number of representative applications of CIMS, including trace highly reactive gaseous pollutants, aerosols and highly oxygenated organic molecules. In particular, we summarized the choice of reagent ions, the influencing factors of the sensitivity, and the strategies in improving instrument sensitivity. This review summarizes CIMS instrument developments and its applications from an appreciation of instrument technology developing perspectives.

Abstract:Ion mobility spectrometry (IMS) is a compact and sensitive trace gas analysis instrument that ionizes the sample into ions for detection. Typically, an ion gate is used to cut the continuous ion beam into ion packets for separation and detection. However, commonly used ion gates suffer from complex structures or low ion transmission rates, making the gateless IMS a viable alternative. In this study, an IMS based on a pulsed photoelectric effect ionization source was designed. The photoelectrons were generated by irradiating a photoelectric material with a back-illuminated pulsed xenon lamp. This allows for low-energy photoelectron generation and the production of simple reactant ions (O2-(H2O)n) and thus negative product ions. The photoelectron current generated by this ionization source was analyzed, which can reach an intensity of a few microamperes and can be converted into an ion signal exceeding 10 nA. The introduction of the pulsed photoelectric effect ionization source makes it possible to generate separate ion packets and complete ion injection when a constant electric field is maintained in the ionization region. And with an assisted pulsed electric field in the ionization region, the resolving power of the system can be effectively improved to 1.85 times that of the constant electric field. The IMS developed in this study was used for the detection of common volatile hazardous chemicals, yielding effective results. The detection limit for phenol was below 1 ppb, and the dynamic response range exceeded 1 order of magnitude, which implies the potential applications of this IMS to detect substances with high electron affinity, such as explosives detection in public safety.
Real-time Monitoring of Dynamic Isomer Populations with CI-SLIM IMS-MS

Abstract:Ion mobility spectrometry (IMS) using Structures for Lossless Ion Manipulations (SLIM) is an emerging powerful tool for rapid isomer separations. This technology offers high mobility resolution due to prolonged ion mobility path lengths that are achieved on a small form factor separation device. In this work, we interface SLIM IMS separation with a chemical ionization source, which allows one to sample from the gas- and particle-phases directly. As such, one can monitor dynamic isomer populations in ambient air in real-time without prior sample preparation. This technology opens the door to new possibilities in atmospheric chemistry where isomer distribution is expected to play a key role in gas phase processes and in the formation of organic aerosols. The CI-IMS-TOF instrument built by TOFWERK produces ions via a two-step chemical ionization process, which involves 1) producing reagent ions and 2) ionizing neutral analyte molecules via the reagent ions through either charge transfer or adduct formation. Once generated, these secondary ions travel into the SLIM IMS region, where a series of DC- and AC-electrodes on printed circuit boards create a traveling wave driving force. As the ions travel through the helium buffer gas, they separate based on their rotationally averaged collision cross-sections. In the present work, various experiments were performed using an aerosol flow tube reactor and an atmospheric simulation chamber to recreate atmospheric conditions. Gas-phase oxidation of isoprene was used to explore the capabilities of the CI-SLIM IMS-MS under atmospheric relevant conditions. Firstly, the most important oxidation products produced from the OH-oxidation of isoprene were used including methacrolein, methyl vinyl ketone, isoprene epoxy diols (IEPOX), isoprene hydroxy hydroperoxide (1,2 and 4,3-ISOPOOH), and other C5H10O3 reactive uptake products as a single component or as a mixture to first evaluate the capabilities of the CI-SLIM IMS-MS at resolving the different isomers. Secondly, the reactive uptake of IEPOX onto acidic particles and OH-initiated oxidation (low and high NO regimes) of isoprene were studied to characterize the dynamic of the isomers generated within the simulation chamber under various environmental conditions. Results will be presented to demonstrate the capabilities of the newly developed CI-SLIM IMS-MS at resolving isomers in real time.
Accurate Collisional Cross Section Measurement by Multi-pass Cyclic Ion Mobility Spectrometry

Abstract:Ion mobility-mass spectrometry (IM-MS) is a powerful analytical tool for structural characterization. IM measurement provides collision cross section (CCS) values that facilitate analyte identification. While CCS values can be directly calculated from mobility measurements obtained using drift tube ion mobility spectrometry (DT-IMS), this method has limited mobility resolution due to the practical constraints on the length of the ion drift path. Consequently, DT-IMS cannot differentiate analytes with similar mobilities or resolve fine mobility features of individual ions. Cyclic IMS (cIMS) instruments leverage a cyclic path enabled by traveling wave ion mobility (TWIM) technology and offer increased mobility solution to address this challenge. While TWIM devices must first be calibrated to enable CCS measurements, current calibration strategies are primarily tailored for single-pass analyses. This preference is partly attributed to the challenges associated with multi-pass calibration methods, which require both calibrants and analytes to experience the same number of passes. Achieving this consistency can be complicated due to factors like peak splitting and diffusion, and may not be feasible for on-line IM-MS analyses. A recent report employed average ion velocities obtained from multiple measurements under different separation pathlengths as a pathlength-independent metric for CCS calibration. However, the ability to exploit this averaging approach is limited by observed variation in ion drift time/velocity in these measurements. In this study, we introduce a novel calibration strategy designed for multi-pass cIMS analyses, directly targeting the root cause for the pathlength- and mobility-dependent variations in ion drift time. With this method, we demonstrate that CCS values derived from multi-pass measurements closely align with those obtained from single-pass analyses, with an average deviation of 0.1%. We apply this method to characterize four isomeric trisaccharides. Our approach not only results in excellent agreement between our measured cIMSCCS values and the reported DTCCS values, with an average difference of only 0.5%, but also allows us to effectively identify subtle mobility characteristics of each compound and determine their respective CCS values. This level of detail and accuracy was previously unattainable using DT-IMS or single-pass cIMS measurements. We developed an algorithm for reconstructing arrival time distribution in cases where wrap-around has resulted in peak splitting. Collectively, the new calibration strategy and the reconstruction procedure maintain reproducibility and precision in CCS measurements while largely eliminating the need for meticulous selection of separation times. We expect that our method will empower researchers to harness the high mobility resolution offered by multi-pass cIMS analyses without compromising the accuracy of CCS measurement, making it appropriate for straightforward use across a wide range of applications.

Abstract:Chemical ionization mass spectrometry (CIMS) is a soft ionization technique and has become an important research tool for real-time atmospheric pollutants measurement, providing reliable datasets for elucidating the pollution formation in atmospheric environment. This review presented a summary of recent progress in development, equipment and application of CIMS in atmospheric environment, including origin of CIMS, development of ion chemistry theory in CIMS, and new hardware design of CIMS, as well as application of CIMS in monitoring of atmospheric pollutants. First, the origin and emergence of CIMS were presented through summarizing the detailed evolution history of mass spectrometry. Next, the ion chemistry theories of CIMS and their development were elaborated, including proton transfer, electron transfer, hydride abstraction, cluster or adduct ion formation and other reactions. Then, the progress of the hardware design of CIMS was focused on its ion source, drift tube and mass analyzer, and the latest equipment development was also discussed. Subsequently, on the basis of introducing the calibration method, the recent application of CIMS in the field observation and laboratory research toward atmospheric inorganic gas, volatile organics, free radicals, and aerosols were introduced. Finally, future directions and trends for CIMS were proposed to promote its efficacy in detecting atmospheric pollutants. The review provides a latest summary of CIMS development in the theory, hardware and application, facilitating a profound exploration of the potential impact of atmospheric pollutants on environment quality and human health.

Abstract:Many designs have been proposed to improve the sensitivity of ion mobility spectrometry (IMS) by modifying its electric field in recent years. However, most of them have shown severe differences between the theoretical predictions and the experimental results. In this work, the effect of electric field on the drifting process was studied from the perspective of spatial ion density. Theoretical derivation illustrated that the nonuniform electric field has no direct influence on the spatial ion density in free space, and the mean spatial ion density was difficult to increase but easy to decrease when crossing grids. Therefore, to improve the sensitivity, a denser ion swarm should be produced in the reaction zone and dilution should be prevented when crossing the ion shutter. Based on this principle, an optimized design was proposed to improve the sensitivity of IMS equipping a UV radiation ionization source. A denser ion swarm was produced by reducing the electric field strength of the reaction zone, and a tri-grid ion shutter was adopted to realize the anti-dilution function. Experimental results showed an impressive improvement, and the SNR of dimethyl methylphosphonate (DMMP) dimer peak increased by 22.6 times without any loss in the resolving power.
