This review discusses aspects of cell culture, media optimization, scaffold design, and the nutrition, taste, and safety considerations in cultivated meat production. It also examines the socio‐political and economic impacts and offers strategies for efficient large‐scale production. As the global population grows, the demand for protein‐enriched foods like meats is rising rapidly. Traditional farming practices face challenges including animal welfare, waste management, and carbon emissions, harming the environment. Consequently, meat substitutes have emerged as a promising area of research and as an alternative to traditional livestock‐sourced meats. Cultivated meat, produced through cell culture techniques, is a key area in cellular agriculture and offers a safe and well‐controlled production process for such future foods. The manufacturing of cultivated meat involves several disciplines, including cell culture, media optimization, scaffold design, and advanced manufacturing, along with nutrition, taste and safety topics to meet consumer acceptance and regulatory approvals. Although several review articles have addressed various aspects of cultivated meat, they tend to focus on specific domains rather than a comprehensive analysis of this transformative technology. This review highlights innovative and applied research findings in the field of cultivated meat, with a focus on critical aspects such as nutrition, cells, materials, and scaffold manufacturing technologies. Furthermore, the socio‐political and economic impacts of cultivated meat are explored, and practical recommendations for low‐cost and large‐scale production. Finally, the review also addresses existing challenges and outlines future directions for the development of cultivated meat.

Abstract:Cellular agriculture is an emerging branch of biotechnology that aims to address issues associated with the environmental impact, animal welfare and sustainability challenges of conventional animal farming for meat production. Cultured meat can be produced by applying current cell culture practices and biomanufacturing methods and utilizing mammalian cell lines and cell and gene therapy products to generate tissue or nutritional proteins for human consumption. However, significant improvements and modifications are needed for the process to be cost efficient and robust enough to be brought to production at scale for food supply. Here, we review the scientific and social challenges in transforming cultured meat into a viable commercial option, covering aspects from cell selection and medium optimization to biomaterials, tissue engineering, regulation and consumer acceptance.

Abstract:Background: Cultured meat is a promising alternative to animal-derived meat production, as it has the potential to offer a sustainable and ethical source of meat for human consumption. Despite the many advantages of cultured meat, scaling up its production remains a major challenge. Among the key factors that must be addressed for efficient production is high-density cell culture, which poses several obstacles, such as the accumulation of metabolic by-products, including toxic ammonia and the inhibitory effects of lactate. Additionally, the use of expensive growth factors and recombinant proteins limits the cost-effectiveness of cultured meat production. Scope and approach: This review provides an overview of methods for reducing the accumulation of metabolic byproducts (ammonia and lactate), including genetic engineering, culture strategies, physicochemical adsorption, biocatalytic conversion, and electrochemistry. Immobilization of growth factors by separation and concentration enhances their biological activity, moderate release, and recovery. Additionally, we discuss the advantages and disadvantages of these methods in the field of cultured meat. Key findings and conclusions: The study concludes that adsorption is a promising pathway for the removal of metabolic by-products. However, the development of adsorbents with favorable biocompatibility, high adsorption efficiency, and reusability remains a challenge. The development of cost-effective alternatives to animal derived sources and media cycling technology to optimize the usage of media components are essential for the progress of cultured meat production.

Abstract:BACKGROUND: Cultured meat forms part of the emerging field of cellular agriculture. Still an early stage field it seeks to deliver products traditionally made through livestock rearing in novel forms that require no, or significantly reduced, animal involvement. Key examples include cultured meat, milk, egg white and leather. Here, we focus upon cultured meat and its technical, socio-political and regulatory challenges and opportunities.SCOPE AND APPROACH: The paper reports the thinking of an interdisciplinary team, all of whom have been active in the field for a number of years. It draws heavily upon the published literature, as well as our own professional experience. This includes ongoing laboratory work to produce cultured meat and over seventy interviews with experts in the area conducted in the social science work.KEY FINDINGS AND CONCLUSIONS: Cultured meat is a promising, but early stage, technology with key technical challenges including cell source, culture media, mimicking the in-vivo myogenesis environment, animal-derived and synthetic materials, and bioprocessing for commercial-scale production. Analysis of the social context has too readily been reduced to ethics and consumer acceptance, and whilst these are key issues, the importance of the political and institutional forms a cultured meat industry might take must also be recognised, and how ambiguities shape any emergent regulatory system.

Abstract:Great importance is being given to the impact our food supply chain and consumers' food habits are having on the environment, human health, and animal welfare. One of the latest developments aiming at positively changing the food ecosystem is represented by cultured meat. This form of cellular agriculture has the objective to generate slaughter-free meat products starting from the cultivation of few cells harvested from the animal tissue of interest. As a consequence, a large number of cells has to be generated at a reasonable cost. Just to give an idea of the scale, there were billions of cells just in a bite of the first cultured-meat burger. Thus, one of the major challenges faced by the scientists involved in this new ambitious and fascinating field, is how to efficiently scale-up cell manufacture. Considering the great potential presented by cultured meat, audiences from different backgrounds are very interested in this topic and eager to be informed of the challenges and possible solutions in this area. In light of this, we will provide an overview of the main existing bioprocessing technologies used to scale-up adherent cells at a small and large scale. Thus, giving a brief technical description of these bioprocesses, with the main associated advantages and disadvantages. Moreover, we will introduce an alternative solution we believe has the potential to revolutionize the way adherent cells are grown, helping cultured meat become a reality.

Abstract:Global pressure from consumers to improve animal welfare, and reduce microbiological risks or the use of antibiotics pose new challenges for the meat industry. Today's livestock production, despite many undertaken measures, is still far from being sustainable. This forced the need to work on alternative protein types that come from plants, insects, fungi, or cell culture processes. Due to some technical and legal barriers, cultivated meat is not present on the European market, however, in 2020 it was approved in Singapore and in 2022 in the USA. While the technology of obtaining cell cultures from animal muscles has been known and successfully practiced for years, the production of a stable piece of meat with appropriate texture, taste, and smell, is still a problem for several scientific groups related to subsequent companies trying to obtain the highest quality product, in line with the expectations of customers. Although the work on optimal cell meat production has been going on for years, it is still in an early stage, mainly due to several limitations that represent milestones for industrial production. The most important are: the culture media (without animal serum), which will provide an environment for optimal muscle development, natural or close to natural (but still safe for the consumer) stable scaffolds for growing cells. Here, we review the actual knowledge about the above-mentioned challenges which make the production of cellular meat not yet developed on an industrial scale.

Abstract:Biotechnology and food science have pioneered the notion of cultured meat. Conventional meat production face issues related to butchering, dietary inadequacy, foodborne disease, and the emanation of methane, which cultured meat evades while promising the texture and feel of real meat. Mass production techniques for plant-based meat analogs have been developed, whose products have hit the market. In vitro production on scaffolding and self-organizing techniques have manufactured small-scale meat products offering tunable nutrition, although more specialized contrivances are needed to build a cultured meat framework on a large scale. Prospective techniques like 3D/4D bio-printing, biophotonics, and cloning are current research subjects. Cultured meat needs to overcome societal and regulatory hurdles prior to commercialization, and, in any event, is a long-term necessity for humankind, although the high production cost and affirmation among people is the principal impediment.

Abstract:The concept behind cellular agriculture is to produce new meat and plant product with almost little or zero meat or plant input by using their tissue or cell. It is a new approach that allows meat to be produced in a bioreactor rather than from farm animals. This concept also can be used by biotechnologists to make meat or skeletal tissue outside of an organism. The purpose of this paper is to review the types of production of cultured meat, potential benefits, and potential challenges in producing cultured meat. Two types of cultured meat production are highlighted in this review, as well as the reduction of environmental burden in food production, nutritional value, and animal welfare, as well as technical and ethical limitations in cultured meat production, public perception, and consumer acceptance of cultured meat. As a result, the concerns regarding the food supply in the face of expanding population and climate change have prompted more research on the importance of cultured meat. Cultured meat can be produced in several ways, and each has the potential to contribute to agricultural technology in ensuring food security, nutritional and hygienic requirements, and reduction of carbon. Cultured meat is a vital source of nutrition, particularly in developing nations where hunger is an issue. This review paper will assist researchers in identifying relevant information about cultured meat and the various perspective including religious perception in consuming and accepting this cultured meat.

Choices for cultured meat are being explored using innovative techniques like stem cells, Embryonic Stem Cells, Induced pluri potent stem cells [(iPSC) derived from skin or blood cells of an animal that have been reprogrammed back into an embryonic-like pluripotent], Totipotent stem cells [immature cells capable of giving rise to any cell type or (of a blastomere) a complete embryo], pluripotent stem cells (immature cells capable of giving rise to several different cell types), satellite cells or myoblasts, etc. In these methods, animal tissue is grown in a controlled surroundings using culture cell technology. Several attempts are made to develop different types of media and techniques like the self-organizing techniques and scaffold-based techniques to advance the growth of the cell. Cultured meat is also called in various names like ‘clean meat’, ‘in-vitro meat’, or ‘artificial meat’. The concept is in the budding stage of commercialization. Abstract:Food systems of the future will need to face an increasingly clear reality - that a protein-rich diet is essential for good health, but traditional meat products will not suffice to ensure safety, sustainability, and equity of food supply chains at a global scale. This paper provides an in-depth analysis of bioprocess technologies needed for cell-based meat production and challenges in reaching commercial scale. Specifically, it reviews state-of-the-art bioprocess technologies, current limitations, and opportunities for research across four domains: cell line development, cell culture media, scaffolding, and bioreactors. This also includes exploring innovations to make cultured meat a viable protein alternative across numerous key performance indicators and for specific applications where traditional livestock is not an option (e.g., local production, space exploration). The paper explores tradeoffs between production scale, product quality, production cost, and footprint over different time horizons. Finally, a discussion explores various factors that may impact the ability to successfully scale and market cultured meat products: social acceptance, environmental tradeoffs, regulatory guidance, and public health benefits. While the exact nature of the transition from traditional livestock to alternative protein products is uncertain, it has already started and will likely continue to build momentum in the next decade.

Abstract:Environment, food, and disease have a selective force on the present and future as well as our genome. Adaptation of livestock and the environmental nexus, including forest encroachment for anthropological needs, has been proven to cause emerging infectious diseases. Further, these demand changes in meat production and market systems. Meat is a reliable source of protein, with a majority of the world population consumes meat. To meet the increasing demands of meat production as well as address issues, such as current environmental pollution, animal welfare, and outbreaks, cellular agriculture has emerged as one of the next industrial revolutions. Lab grown meat or cell cultured meat is a promising way to pursue this; however, it still needs to resemble traditional meat and be assured safety for human consumption. Further, to mimic the palatability of traditional meat, the process of cultured meat production starts from skeletal muscle progenitor cells isolated from animals that proliferate and differentiate into skeletal muscle using cell culture techniques. Due to several lacunae in the current approaches, production of muscle replicas is not possible yet. Our review shows that constant research in this field will resolve the existing constraints and enable successful cultured meat production in the near future. Therefore, production of cultured meat is a better solution that looks after environmental issues, spread of outbreaks, antibiotic resistance through the zoonotic spread, food and economic crises.

Abstract:It is estimated that around 8% of the entire world population identifies themselves as vegetarians. That leaves us with statistics of more than 90% of the world's population prefers a meat-based diet. Now with the alarmingly increasing population of over 7.8 billion, the demand for meat is skyrocketing. Due to the high demand, factors like land, water and greenhouse emission are becoming unsustainable. Even after taking all these concerns into account, why is it so difficult for us to eliminate/reduce meat from our diet? How is the future generation going to satisfy their meat cravings? However, through recent research, it seems there is an alternative, a solution to all these concerns, cultured/ lab-grown meat. Cultured meat is meat produced by in vitro cell culture of animal cells, instead of from slaughtered animals using various tissue engineering techniques. This invitro meat technology is still at its elementary stages but shows great potential in curbing the problem of livestock overcrowding and its impact on the environment. Cultured meat seems to promise a sustainable and reliable alternative for its consumers. This review aimed to provide an elaborate comprehension of the entire process of the production, processing and commercialization of cultured meat, and also provides an insight on the possible obstacles it encounters, especially overcoming ethical conundrums and social prejudice.

Abstract:Cultured meat production technology suggested that can solve the problems of traditional meat production such as inadequate breeding environment, wastewater, methane gas generation, and animal ethics issues. Complementing cultured meat production methods, sales and safety concerns will make the use of cultured meat technology easier. This review contextualizes the commercialization status of cultured meat and the latest technologies and challenges associated with its production. Investigation was conducted on materials and basic cell culture technique for cultured meat culture is presented. The development of optimal cultured meat technology through these studies will be an innovative leap in food technology. The process of obtaining cells from animal muscle, culturing cells, and growing cells into meat are the basic processes of cultured meat production. The substances needed to production of cultured meat were antibiotics, digestive enzymes, basal media, serum or growth factors. Although muscle cells have been produced closer to meat due to the application of scaffolds materials and 3 D printing technology, still a limit to reducing production costs enough to be used as foods. In addition, developing edible materials is also a challenge because the materials used to produce cultured meat are still not suitable for food sources.

Abstract:Cultivated meat (CM) technology has the potential to disrupt the food industry—indeed, it is already an inevitable reality. This new technology is an alternative to solve the environmental, health and ethical issues associated with the demand for meat products. The global market longs for biotechnological improvements for the CM production chain. CM, also known as cultured, cell-based, lab-grown, in vitro or clean meat, is obtained through cellular agriculture, which is based on applying tissue engineering principles. In practice, it is first necessary to choose the best cell source and type, and then to furnish the necessary nutrients, growth factors and signalling molecules via cultivation media. This procedure occurs in a controlled environment that provides the surfaces necessary for anchor-dependent cells and offers microcarriers and scaffolds that favour the three-dimensional (3D) organisation of multiple cell types. In this review, we discuss relevant information to CM production, including the cultivation process, cell sources, medium requirements, the main obstacles to CM production (consumer acceptance, scalability, safety and reproducibility), the technological aspects of 3D models (biomaterials, microcarriers and scaffolds) and assembly methods (cell layering, spinning and 3D bioprinting). We also provide an outlook on the global CM market. Our review brings a broad overview of the CM field, providing an update for everyone interested in the topic, which is especially important because CM is a multidisciplinary technology.

Abstract:As the global population grows, we need a stable protein supply to meet the demands. Although plant-derived protein sources are widely available, animal meat maintains its popularity as a high-quality and savory protein source. Recently, cultured meat, also known as <i>in vitro</i> meat, has been suggested as a meat analog produced through <i>in vitro</i> cell culture technology. Cultured meat has several advantages over conventional meat, such as environmental protection, disease prevention, and animal welfare. However, cultured meat manufacturing is an emerging technology; thus, its further and dynamic development would be pivotal. Commercialization of cultured meat to the public will take a long time but cultured meat undoubtedly will come to our table someday. Here, we discuss the social and economic aspects of cultured meat production as well as the recent technical advances in cultured meat technology.

Abstract:To satisfy the increasing demand for food by the growing human population, cultured meat (also called in vitro, artificial or lab-grown meat) is presented by its advocates as a good alternative for consumers who want to be more responsible but do not wish to change their diet. This review aims to update the current knowledge on this subject by focusing on recent publications and issues not well described previously. The main conclusion is that no major advances were observed despite many new publications. Indeed, in terms of technical issues, research is still required to optimize cell culture methodology. It is also almost impossible to reproduce the diversity of meats derived from various species, breeds and cuts. Although these are not yet known, we speculated on the potential health benefits and drawbacks of cultured meat. Unlike conventional meat, cultured muscle cells may be safer, without any adjacent digestive organs. On the other hand, with this high level of cell multiplication, some dysregulation is likely as happens in cancer cells. Likewise, the control of its nutritional composition is still unclear, especially for micronutrients and iron. Regarding environmental issues, the potential advantages of cultured meat for greenhouse gas emissions are a matter of controversy, although less land will be used compared to livestock, ruminants in particular. However, more criteria need to be taken into account for a comparison with current meat production. Cultured meat will have to compete with other meat substitutes, especially plant-based alternatives. Consumer acceptance will be strongly influenced by many factors and consumers seem to dislike unnatural food. Ethically, cultured meat aims to use considerably fewer animals than conventional livestock farming. However, some animals will still have to be reared to harvest cells for the production of in vitro meat. Finally, we discussed in this review the nebulous status of cultured meat from a religious point of view. Indeed, religious authorities are still debating the question of whether in vitro meat is Kosher or Halal (e.g., compliant with Jewish or Islamic dietary laws).

Abstract:The current food system, which is responsible for about one third of all global gas emissions, is considered one of the main causes of resource depletion. For this reason, scientific research is investigating new alternatives capable of feeding an ever-growing population that is set to reach 9-11 billion by 2050. Among these, cell-based meat, also called cultured meat, is one possible solution. It is part of a larger branch of science called cellular agriculture, whose goal is to produce food from individual cells rather than whole organisms, tracing their molecular profile. To date, however, cultured meat aroused conflicting opinions. For this reason, the aim of this review was to take an in-depth look at the current European legislative framework, which reflects a 'precautionary approach' based on the assumption that these innovative foods require careful risk assessment to safeguard consumer health. In this context, the assessment of possible risks made it possible not only to identify the main critical points during each stage of the production chain (proliferation, differentiation, scaffolding, maturation and marketing), but also to identify solutions in accordance with the recommendations of the European Food Safety Authority (EFSA). Further, the main challenges related to organoleptic and nutritional properties have been reviewed.. Finally, possible future markets were studied, which would complement that of traditional meat, implementing the offer for the consumer, who is still sceptical about the acceptance of this new product. Although further investigation is needed, the growing demand for market diversification and the food security opportunities associated with food shortages, as well as justifying the commercialisation of cultured meat, would present an opportunity to position cultured meat as beneficial.
Stem cell-based strategies and challenges for production of cultivated meat

Abstract:Cultivated meat scale-up and industrial production will require multiple stable cell lines from different species to recreate the organoleptic and nutritional properties of meat from livestock. In this Review, we explore the potential of stem cells to create the major cellular components of cultivated meat. By using developments in the fields of tissue engineering and biomedicine, we explore the advantages and disadvantages of strategies involving primary adult and pluripotent stem cells for generating cell sources that can be grown at scale. These myogenic, adipogenic or extracellular matrix-producing adult stem cells as well as embryonic or inducible pluripotent stem cells are discussed for their proliferative and differentiation capacity, necessary for cultivated meat. We examine the challenges for industrial scale-up, including differentiation and culture protocols, as well as genetic modification options for stem cell immortalization and controlled differentiation. Finally, we discuss stem cell-related safety and regulatory challenges for bringing cultivated meat to the marketplace.
