Abstract:In the early days of blood group specific seed agglutinins,14 it was hoped that a wide range of specificities would be discovered. These hopes have not been realized, and recent advances have been largely confined to finding what may be termed “new uses for old seed agglutinins,” chiefly the elucidation of erythrocyte polyagglutination, the study of erythrocytes with various membrane defects, and the investigation of tumor cell membranes. It is not really surprising that the range of specificities of seed agglutinins is limited, because these substances combine with a single sugar or even part of a single sugar, and the sugar components of the red cell membrane are restricted to the seven sugars galactose, mannose, fucose, glucose, N-acetylglucosamine, N-acetylgalactosamine, and N-acetylneuraminic acid. So far as is known at present, the specificities of plant agglutinins are confined to anti-A (for example, Phaseolus Zunatus), anti-A, (Dolichos bijlorus) , anti-B (Fomes fomenturius), anti-A + B (for example, Phlomis fruticosa), anti-H (for example, Ulex europueus), anti-M (some select strains of various Zberis species), anti-N (for example, Vicia gruminea) , anti-A + N (MoluccelZa laevis), anti-T (Arachis hypogaea) , and anti-Tn (Salvia sclarea). Some of these reagents are highly specific, such as Dolichos bijlorus, and some cross-react: for example, anti-A + B agglutinins, which are unable to distinguish a-galactose from a-N-acetylgalactosamine because of their chemical similarity. Although the capacity of monosaccharides to inhibit seed agglutinins has provided useful leads in establishing the chemical basis of blood group specificity: the value of isolated monosaccharides for this purpose is understandably limited, because serological activity is dependent on a number of factors, such as anomeric conformation, the nature of the next-to-terminal structure, the site of attachment to the next sugar or carrier proteidlipid, the accessibility of the receptor, both in terms of steric conformation and the degree of steric hindrance offered by neighboring structures, and perhaps the number and distribution of receptor sites. Many examples of the influence of these factors can be cited; some are briefly mentioned below. Anomeric Conformation. The red cell membrane abounds with &galactose residues, yet the Fomes fomentarius agglutinin reacts only with red cells that have terminal a-galactose residues, for example, B cells. Next-to-Terminal Structure. Human blood group A substance and the group C Sfreprococcus both have a-linked N-acetylgalactosamine in the terminal position.
