Abstract:In current work, three N-acetyl pyrazoline derivatives ( 1 - 3 ) have been prepared via 1,4-addition reaction of hydrazine on chalcones, followed by cyclization of the adduct in the presence of acetic acid. The structural interpretation of synthesized compounds was accomplished using various characterization techniques like FT-IR, Ultraviolet/visible spectroscopy, 1 HNMR and 13 C NMR. The synthesized compounds were evaluated for antioxidant, Urease inhibitory and Lipoxygenase inhibitory activities and results revealed that compound 2 showed promising antioxidant, Urease inhibitory and Lipoxygenase inhibitory activity with IC 50 values 33.2± 0.45, 32.7 ± 0.11 and 42.2 ±0.77, respectively. Moreover, DNA binding interactions of the N-acetyl pyrazoline derivatives have also been carried out with SS-DNA (Salmon Sperm DNA) using absorption spectroscopy. Furthermore, molecular docking was conducted for a ready comparison with the experimental data and to determine the interactions involved. ADMET profiling of pyrazoline derivatives was also done using Admet SAR. Besides that, quantum chemical calculations using DFT (density functional theory) were carried out to explore optimized geometries, FMOs (Frontier molecular orbitals), NBOs (Nonbonding orbitals) of synthesized compounds and to predict their NLO (Nonlinear optical) properties. The synthesized compounds showed much better NLO behavior than standard Urea along with a strong agreement between computed and empirically recorded spectral data.
Synthesis, Crystal Structures, Fluorescence and Xanthine Oxidase Inhibitory Activity of Pyrazole-Based 1,3,4-Oxadiazole Derivatives

Abstract:A series of pyrazole-based 1,3,4-oxadiazole derivatives were rationally designed and synthesized in good yields by following a convenient route. All the newly synthesized molecules were fully characterized by IR, H-1 NMR and elemental analysis. Eight compounds were structurally determined by single crystal Xray diffraction analysis. The fluorescence properties of all the compounds were investigated in dimethyl sulfoxide media. In addition, these newly synthesized compounds were evaluated for in vitro inhibitory activity against commercial enzyme xanthine oxidase (XO) by measuring the formation of uric acid from xanthine. Among the compounds synthesized and tested, 3d and 3e were found to be moderate inhibitory activity against commercial XO with IC50 = 72.4 mu M and 75.6 mu M. The studies gave a new insight in further optimization of pyrazole-based 1,3,4-oxadiazole derivatives with excellent fluorescence properties and XO inhibitory activity. (C) 2015 Elsevier B.V. Design and Synthesis of Carbothioamide/Carboxamide-Based Pyrazoline Analogs as Potential Anticancer Agents: Apoptosis, Molecular Docking, ADME Assay, and DNA Binding Studies.

Abstract:To discover anticancer drugs with novel structures and expand our research scope, pyrazoline derivatives (-) were designed and synthesized through cyclization of chalcones with thiosemicarbazide/semicarbazide in CHCOOH as a solvent. All newly synthesized pyrazoline derivatives were fully characterized using several spectroscopic experiments such as H, C NMR, FT-IR spectroscopy, and mass analysis. By , the purity of all analogs was found above and both lead compounds ( and ) were also validated by . Anticancer activity of synthesized pyrazoline derivatives (-) was investigated by the MTT assay against the human lung cancer cell (A549), human cervical cancer cell (HeLa), and human primary normal lung cells (HFL-1). Staurosporine (STS) was used as a standard drug. The anticancer results showed that two potent analogs and exhibit excellent activity against (IC = 13.49 ± 0.17 and 22.54 ± 0.25 μM) and cells (IC = 17.52 ± 0.09 and 24.14 ± 0.86 μM) and low toxicity against the HFL-1 (IC = 114.50 ± 0.01 and 173.20 ± 10 μM). The flow cytometry was further used to confirm the anticancer activity of potent derivatives against the A549 cancer cell line. DNA binding interaction of anticancer agents and with Ct-DNA has been carried out by absorption, fluorescence, EtBr (dye displacement assay), circular dichroism, cyclic voltammetry and time-resolved fluorescence, which showed noncovalent binding mode of interaction. Anticancer activity of both lead compounds ( and ) may be attributed to DNA binding. The evaluation of the antioxidant potential of pyrazoline analogs 3a and 3h by 2,2-diphenyl-1-picrylhydrazyl free radical showed promising antioxidant activity with IC values of 0.132 ± 0.012 and 0.215 ± 0.025 μg/mL, respectively. In silico molecular docking of pyrazoline derivatives was also performed using autodock vina software against the DNA hexamer with PDB ID: 1Z3F and ADMET properties to explore their best hits.

Abstract:s The pyrimidine scaffold is often considered as one of the most important pharmaceutically active chemicals. Pyrimidine functionalized with a halogen group becomes an appropriate precursor for several structural variations in the synthesis of pyrimidine-based compounds. Herein, an effort was made to design and synthesize a series of 4, 6-dichloropyrimidine chalcone derivatives of biological and pharmaceutical interest. These compounds showed anticancer potential against A549 cells under In vitro conditions observed through MTT assay. Among all the tested compounds 1c-thiourea and 1 d-urea displayed highest cytotoxic potential with IC 50 value of 91.36 μM and 128.14 μM. Further, we studied the interaction of these compounds (1c-thiourea and 1d-urea) with the human serum albumin (HSA) using UV–Vis., fluorescence emission, and far-UV-CD spectroscopic techniques. The steady state fluorescence studies show quenching in the fluorescence emission intensity of HSA in the presence of 1c-thiourea and 1d-urea compounds, which infers the binding affinity of HSA with our synthesized compounds. The observed thermodynamic parameter, i.e., Gibbs free energy (∆ G o ) of the system suggests that the binding process is spontaneous in nature. In addition, structural changes in the secondary structure of HSA induced by 1c-thiourea and 1d-urea were studied using far UV-CD. The α-helical content of HSA is increased in the presence of both the ligands indicating alterations in the secondary structure of protein. Molecular docking was performed to assess the binding affinity of all these compounds with HSA, and the results suggest strong binding with HSA through H-bonding and hydrophobic interactions. The current study provides vital information regarding the interaction of HSA with the synthesized compounds, which may provide a new path for designing novel potent 4, 6-dichloropyrimidine chalcone derivatives for use in the pharmaceutical and biomedical fields.

Abstract:Dissolution of biologically important sparingly soluble salts, such as calcium carbonate and calcium oxalate, is possible by use of carboxyl- and carboxyl/phosphonate-bearing, anionic additives, citrate, malate, carboxyphosphonate, and butane tetracarboxylate. Calcium-containing dissolution products have been identified, characterized, and independently synthesized. These are polymeric materials composed of calcium and the additive as the ligand. Their full characterization was carried out by single-crystal X-ray crystallography and other techniques.

Abstract:A series of novel 5-alkyl-2-ferrocenyl-6,7-dihydropyrazolo[1,5-a]pyrazin-4(5H)-one derivatives were synthesized by the reaction of ethyl 1-(2-bromoethyl)-3-ferrocenyl-1H-pyrazole-5-carboxylate with non-aromatic primary amines in one-pot procedure and characterized by IR, 1H NMR, 13C NMR, HRMS and X-ray diffraction analysis. The effects of all the compounds on A549 cell growth were investigated. The results showed that all compounds had almost inhibitory effects on the growth of A549 cells.

Abstract:Four new pyrazolo[3,4-d]pyrimidines (P1–P4) were successfully synthesized in good relative yields by reacting 3-methyl-1-phenyl-1H-pyrazolo[3,4-d]pyrimidin-4-ol with various alkylating agents (methyl iodide, propargyl bromide, and phenacyl bromide) at room temperature in DMF solvent, employing liquid–solid phase transfer catalysis. The P1–P4 structures were elucidated using NMR spectroscopy and X-ray diffraction. Intermolecular interactions in P1–P4 were analyzed via Hirshfeld surface analysis and 2D fingerprint plots. Geometrical parameters were accurately modeled by DFT calculations using the B3LYP hybrid functional combined with a 6–311++G(d,p) basis set. The antiproliferative activity of P1–P4 towards colorectal carcinoma (HCT 116), human hepatocellular carcinoma (HepG2), and human breast cancer (MCF-7) cell lines, along with one normal cell line (WI38) was investigated using the MTT assay and sunitinib as a reference. Compounds P1 and P2 exhibited antiproliferative activities comparable to the reference drug towards all tested cells, with an IC50 range of 22.7–40.75 μM. Both compounds also showed high selectivity indices and minimal cytotoxic effects on the normal cell line. Molecular docking revealed that the significant antiproliferative activity may attributed to the number and type of intermolecular hydrogen bonding established between pyrazolo[3,4-d]pyrimidines and DNA topoisomerase, a common target for various anticancer agents.

Abstract:The current work describes the design, synthesis and molecular mocking studies of a series of new 1,4-disubstituted-1,2,3-triazole linked 2-pyrimidinone derivatives. Firstly, 4-(4-acetyl-5-methyl-1H-1,2,3-triazol-1-yl)benzene sulfonic acid 1 was synthesized as a key starting material. This compound was reacted with a series of aromatic aldehydes under-investigated conditions to give a new series of chalcones 2a-f. Reaction of compounds 2a-f with urea in the presence of an aqueous solution of sodium hydroxide led to the construct 2-pyrimidinone ring system to obtain a series of new compounds containing 1,2,3-triaozle ring and pyrimidinone ring 3a-f. The newly synthesized compounds 2a-f and 3a-f were characterized by FT-IR, 1H-NMR and 13C-NMR spectra. In-silico molecular docking simulations, compounds 3a-f and their precursors 2a-f were conducted on two selected proteins: 7dpp and 8cx9. The results revealed that all of the newly synthesized compounds 2a-f and 3a-f displayed have a good binding affinity with the target proteins and higher than values recorded for the selected three standard antiviral drugs.

Polycyclic Aromatic Compounds

Abstract:In this work, three N' -arylidene-5-phenyl- 1H -pyrazole-3-carbohydrazide ( 3a - 3c ), derivatives have been synthesized and fully characterized by IR, 1 H & 13 C NMR and ESI-MS using experimental and theoretical methods. Theoretical calculations were carried out by DFT/B3LYP method using 6-311++G(d,p) basis set. For theoretical IR, 1 H & 13 C NMR (with GIAO method), MEP, HOMO-LUMO energies analyses of compounds 3a-3c were performed over the optimized structures. All newly synthesized compounds were screened for their cytotoxicity, proylenpeptidase and α-glucosidase inhibitory activities. Compounds 3c showed significant anticancer activity against lung cancer cell line (H460) with the IC 50 value 0.15 ± 0.01 μM. The other compounds exhibited prolyl endopeptidase (PEP) activity. Compounds 3a and 3c have demonstrated a potent α-glucosidase inhibitory activity with the IC 50 value 360.4 ± 0.7 μM and 370.3 ± 1.17 μM, respectively. Furthermore, in-silico based molecular docking study was performed between the title ligands and lung cancer cell line-H460 protein: 1M17. The best affinity bond was observed between compound 3c and the protein 1M17 with an energy of −8.6 (kcal/mol), an inhibition constant of 0.496769 μM and an active hydrogen bond to three.

Abstract:The three component reaction between 5‑chloro-3-methyl-1-phenyl-1H-pyrazole-4- carbaldehyde, thiosemicarbazide and 4-nitrophenacyl bromide furnished (E)-2-(2-((5‑chloro-3-methyl-1-phenyl-1H-pyrazol-4-yl)methylene)hydrazineyl)-4-(4 nitrophenyl)thiazole which was characterized by FT-IR and NMR spectral methods. These spectroscopic methods confirmed the structural framework of the titled compound. The density functional theory (DFT) approach with a 6–311++G(d,p) method was used and electronic and spectroscopic parameters were analyzed. Frontier molecular orbital analysis of the titled compound found a 3.13 eV energy gap with HOMO primarily populated on pyrazole and thiazole rings whereas LUMO populated on benzene carrying nitro group. According to the Non-Linear Optical (NLO) study, the titled compound might be significant in the fabrication of nonlinear optical materials. The results of the in-silico molecular docking study against cytochrome P450 14-sterol demethylase (CYP51) showed that hydrogen bonds, aromatic and hydrophobic interactions, and van Der Waals forces all contributed to strong binding affinity with the ligand. The experimental and computational spectral comparison led assignments of IR and NMR signals. The ADME predictions suggested a good pharmacological profile of the titled compound.

Abstract:BackgroundChalcones have a place with the flavonoid family and show a few very important pharmacological activities. They can used as initial compounds for synthesis of several heterocyclic compounds. The compounds with the backbone of chalcones have been reported to possess various biological activities.ResultsPyridine and thioamide derivatives were obtained from the reaction of 3-(furan-2-yl)-1-(p-tolyl)prop-2-en-1-one with the appropriate amount of malononitrile, benzoylacetonitrile, ethyl cyanoacetate and thiosemicarbazide in the presence of ammonium acetate. The reaction of 3,5-di(furan-2-yl)-4,5-dihydro-1H-pyrazole-1-carbothioamide with ethyl 2-chloro-3-oxobutanoate, 3-chloropentane-2,4-dione or ethyl chloroacetate produced thiazole derivatives. Pyrano[2,3-d]thiazole derivatives were obtained as well from thiazolone to arylidene malononitrile. The structures of the title compounds were clarified by elemental analyses, and FTIR, MS and NMR spectra. Some compounds were screened against various microorganisms (i.e., bacteria +ve, bacteria −ve and fungi). We observed that compounds (3a), (4a), (4d), (5), (7) and compound (8) exhibited high cytotoxicity against the MCF-7 cell line, with IC50 values of 23.6, 13.5, 15.1, 9.56, 14.2 and 23.5 μmol mL−1, respectively, while compound (9) was displayed the lowest values against MCF-7 cell lines.ConclusionsEfficient synthetic routes for some prepared pyridines, pyrazoline, thioamide, thiazoles and pyrano[2,3-d]thiazole were created. Moreover, selected newly-synthesized products were evaluated for their antitumor activity against two carcinoma cell lines: breast MCF-7 and colon HCT-116 human cancer cell lines.

Abstract:A series of new pyrazolines (4a–4g) and isoxazolines (5a–5g) have been synthesized in good-to-excellent yield from the corresponding chalcones (3a–3g) under ultrasound irradiation. The newly synthesized compounds were characterized and screened for their in vitro antimicrobial activity against different bacterial and fungal strains by standard agar dilution method and in vitro antioxidant activity by using DPPH method. Some of the synthesized compounds displayed significant antimicrobial and antioxidant activity. Docking study of the synthesized compounds has been performed, and results showed good binding mode in the active site of fungal enzyme P450 cytochrome lanosterol 14 α-demethylase. ADMET properties of synthesized compounds showed good drug-like properties.

Abstract:In this study, one-pot multicomponent reactions of novel chromeno[2,3- c ]pyrazole derivatives (1–14) were performed using an AlCl 3 catalyst via cyclisation. Various spectral and chromatographic techniques were used to elucidate the structure of the synthesised derivatives (1–14) . The synthesised compounds were then inspected for their antibacterial, antioxidant, and tyrosinase inhibition activities. An in silico screening approach was also employed to identify highly potent derivatives. Besides, we utilised density functional theory (DFT) with the B3LYP/6-31G + (d, p) basis set to optimise the newly modified derivatives. This approach was used to calculate various properties, including electron density, electrostatic potential map, interaction strength, frontier molecular orbital energy, and reactivity characteristics. To examine the binding affinity, modes, and stability of the protein-drug complex, molecular docking with the 2Y9X protein structure were employed. The findings from DFT computations, along with physicochemical information and molecular docking binding affinity, showed promising results than standard and low active compound 1 . The absorption, metabolism, and cytotoxic characteristics of all the novel derivatives were investigated in the ADMET prediction. Our findings could prove valuable in developing novel drugs for medicinal and pharmaceutical fields.

Abstract:Pyrimidine which is an important constituent of the genetic material of deoxyribonucleic acid, is identified with a large number of biological activities. Based on this, pyrimidine-derived Schiff bases (1-6) of hydroxy-1-naphthaldehyde were synthesized by using the condensation method. In addition, the molecular docking studies against topoisomerase II DNA gyrase, human hematopoietic cell kinase, urate oxidase from Aspergillus flavus, and cyclin-dependent kinase 8 to explore the antibacterial, antioxidant, antifungal, and anticancer properties respectively and binding affinities through bioinformatics approaches to determine the interaction among active molecules with the receptor. Hence, the computational docking analyses identified that all synthesized pyrimidine Schiff bases (1-6) are active and exhibited better binding affinities as compared to the standard drugs. Furthermore, all the prepared materials were characterized by using nuclear magnetic resonance, infrared, and elemental analysis. Additionally, the phase-transition and thermal decomposition temperatures were determined by differential scanning calorimetry and thermo-gravimetric analysis measurements. Moreover, the structures of pyrimidine-derived Schiff bases 1, 2, 3, 4, and 5 were also confirmed by the X-ray single-crystal diffraction technique. The pyrimidine-derived Schiff bases 5 possess significant antibacterial, antioxidant, antifungal, and anticancer agent properties which confirms its promising biological activities over standard drugs.

Abstract:A series of novel pyridine and fused pyridine derivatives have been prepared starting from 6-(3,4-dimethylphenyl)-2-hydrazinyl-4-(thiophen-2-yl)-pyridine-3-carbonitrile 1 which on treatment with appropriate formic acid, acetic acid/ acetic anhydride, benzoyl chloride and/or carbon disulfide afforded the corresponding triazolopyridine derivatives 2–5. Also, treatment of hydrazide 1 with diethyloxalate, chloroacetyl chloride, chloroacetic acid and/or 1,2-dichloroethane yielded the corresponding pyridotriazine derivatives 7–10. Further transformation of compound 1 with a different active methylene group, namely acetyl acetone, diethylmalonate, ethyl cyanoacetate, ethyl benzoylacetate and/or ethyl acetoacetate, produced the pyridine–pyrazole hybrid derivatives 11–15. These newly synthesized compounds (1–15) were subjected to in silico molecular docking screenings towards GlcN-6-P synthase as the target protein. The results revealed moderate to good binding energies of the ligands on the target protein. All the newly prepared products exhibited antimicrobial and antioxidant activity.

Abstract:INTRODUCTION Hydrofluoric acid is a fluoride-substituted compound used in the chemical industry. Burns and hypocalcaemia result from ingestion or contact with the skin or mucosal membranes. We observed burns and skin necrosis on the hands after home use of low-concentration hydrofluoric acid. CASE REPORT A 57-year-old woman consulted in February 1994 for oedema, erythema and very painful burns of the palms of both hands. The day before, she had used a home-made furniture cleanser containing 5 p. 100 hydrofluoric acid. At admission, calcium and radiography of the two hands were normal. She was given a topical application of 5 p. 100 calcium chloride. The clinical course was favourable with squamation of both palms then necrotic lesions of the pulp on the 1st, 2nd and 3rd fingers. DISCUSSION Such exposure in a household situation is unusual. Hydrofluoric acid has two dangerous mechanisms of action. First it is a caustic substance producing late-onset burns and secondly hypocalcaemia results from precipitation of insoluble calcium fluoride. The risk of hypocalcaemia is greatest when a large area of the skin is exposed. Prognosis depends on early treatment based on prevention of hypocalcaemia by abundant washing of the teguments and permanent application of a 5 p. 100 calcium gluconate solution associated with local skin treatments. Careful follow-up is required with regular calcium chemistries.

Abstract:We have investigated the photophysical properties of chalcone derivatives by UV-visible absorption spectra. The HOMO–LUMO energies were calculated. Electrochemical properties of the compounds ( 5a – 5d ) via cyclic voltammetry showed that compounds have oxidation peaks. In addition, chalcone derivatives were analyzed for DNA binding study. It revealed that chalcone derivatives had a promising affinity towards DNA double helix. DNA binding constants of the complexes indicate the intercalative binding mode involving a strong aromatic chromophore and DNA base pairs.

Abstract:Molecules bearing pyrazole nucleus present diverse biological properties such as antitumor and anti-inflammatory activities that can be associated with DNA interactions. This study aimed to the synthesis of new pyrazol derivatives and evaluated their ability to interact with the DNA and antitumor and topoisomerase inhibition activities. All derivatives were successfully synthesized, and their structures were elucidated by H-1-NMR and high resolution (HR)-MS (electrospray ionization positive mode (ESI+)). Antiproliferative inhibition assays, UV titration assays, fluorescence titration assays, circular dichroism (CD) assays, KI quenching studies, topoisomerase inhibitory activity assays and molecular docking were evaluated for these compounds. Especially, compounds 5e and 5q showed higher antitumor activity with IC50 values <13 mu M for the tested cell lines. However, compounds 5e and 5q did not inhibit the topoisomerase activity evaluated by relaxation assay. These results show that the pyrazole nucleus contributes to the incorporation of molecules into the DNA. Moreover, it was highlighted that positive charges are relevant for the design of promising antitumor and DNA binding compounds.

Abstract:In this work, we present the synthesis of a newer series of 15 chalcone-based pyrimidine compounds 4a-o. All the compounds were characterized by elemental analysis, melting point determination, mass, FTIR, and NMR analysis. We have evaluated the antimicrobial activity of these compounds. The compounds showed good inhibition activity towards different bacterial and fungal species such as S. aureus, S. pneumonia, E. coli, P. aeruginosa, Candida albicans, Aspergillus niger, and Alternaria alternata. Compounds 4c, 4h, 4k, and 4g showed comparable activities to those of commercially available drugs. Molecular docking study showed good interaction between each of the compounds and DNA gyrase enzyme. The docking score of the compounds ranges between -8.0 to -8.9 kcal/mol. Further, the ADMET analysis indicated the potential of the compounds as a drug candidate

Abstract:Fused pyrimidines play an imperative role in our life due to their biological importance in the struggle of microorganisms. A series of 4-aryl-3-methyl-1-phenyl-1,4,6,7-tetrahydropyrazolo[3,4-d] thiazolo[3,2-a]pyrimidine (4a-j) were synthesized by cyclization of 3-methyl-1-phenyl-4-aryl-1,4,5,7-tetrahydro-6H-pyrazolo[3,4-d]pyrimidine-6-thiones (3a-j) with 1,2-dibromoethane in presence of anhydrous potassium carbonate. Earlier, compounds (3a-j) were synthesized by the condensation of 5-methyl-2-phenyl-2,4-dihydro-3H-pyrazol-3-one (2), different substituted benzaldehyde and thiourea with catalytic amount of con. HCl in methanol. Compound 2 was synthesized by condensation of ethyl acetoacetate (1) and phenylhydrezine in presence of a catalytic amount of acetic acid at reflux temperature. The constitution of the synthesized products has been characterized by using elemental analysis, Infrared, 1H-NMR spectroscopy and further supported by Mass spectroscopy. All the products have been screened for their in-vitro biological assay like antibacterial and antifungal activity at concentration of 500 μg/ml. It was exposed that most of the compounds displayed inspiring antibacterial and antifungal activity compared to the used reference standard.
