Abstract:Phytochemical investigation on the aerial parts of Corydalis impatiens (pall.) Fisch (Papaveraceae) resulted in the identification of four previous undescribed benzylisoquinoline alkaloids, impatienines A-D ( 1 – 4 ), together with 14 known analogues ( 5 – 18 ). The structures of these compounds were elucidated by extensive spectroscopic analysis (IR, HR-ESIMS, 1D- and 2D-NMR) as well as ECD calculations. All the compounds obtained were investigated for their inhibitory effect on the growth of A549, H1299 and HepG2 cancer cells. Compounds 7 and 15 exhibited pronounced inhibition against the A549 cancer cells with IC 50 values of 6.81 μM and 3.17 μM, while the positive control cisplatin was 1.83 μM. Compounds 1 – 3 showed moderate inhibitory on the H1299 cancer cells. Compounds 4 , 10 – 12 , and 16 showed signiffcant activity against HepG2 cancer cells with IC 50 values range of 4.41–8.75 μM.

Abstract:A series of 4-(1 H-indol-3-yl-methylene)-1 H-pyrazol-5(4 H)-one derivatives have been synthesised. The Z structure of 4-[(1-methyl-1 H-indol-3-yl)methylene]-3-phenyl-1 -p-tolyl-1 H-pyrazol-5-one was determined by X-ray crystallography. The antitumour activity was evaluated against five cancer cells by MTT assay. [(1 H-Indol-3-yl)methylene]-1-(2,4-dini-trophenyl)-3-methyl-1 H-pyrazole-5-one and 4-{4-[(1-benzyl-1 H-indol-3-yl)methylene]-3-methyl-5-oxo-4,5-dihydro-1 H-pyrazol-1-yl}-benzoic acid have similar anticancer activity with 5-UF on the test cancer cells (exception of A375). Almost all the target compounds displayed antitumour activity against A549 and PC-9, and those with benzyl at 1-position of indole had higher activity against PC-9 (IC50 value lower than 30 μM). Those with benzyl at the indole and carboxyl at the phenyl part of of pyrazole were more active against PC-9 and A549 cells, providing a good indication for subsequent optimisation as lung cancer inhibitory agents.

Abstract:The chemical investigation on Corydalis balansae resulted in the isolation of three previous undescribed compounds (1, 10, and 11) and 17 known compounds. Compound 1 and 2 were obtained as two lignanamide dimers, and compound 11 had a spiro [benzofuranone-benzazepine] skeleton, which was found in Corydalis for the first time. The structures of new compound were determined by the detailed analysis of 1D/2D NMR, UV, and IR data. Absolute configurations of compounds 10 and 11 were defined by their crystal X-ray diffraction data and calculations of electronic circular dichroism (ECD). The CCK-8 method was used to assay the inhibition effect of all the compounds on the growth of Hela, MGC-803, A549, and HepG2 cancer cells. Compound 2, 13, and 14 showed moderate inhibitory activity against the tested cell lines. Compound 2 exhibited potential antitumor activity against MGC-803 cells with an IC50 value of 20.8 μM, while the positive control etoposide was 17.3 μM. Furthermore, results from the cellular-mechanism investigation indicated that compound 2 could induce S-phase cell-cycle arrest and MGC-803 cells apoptosis, which was triggered by the up-regulation of PARP1, caspase-3 and -9, Bax, and down-regulation of Bcl-2. The 2-induced strong apoptosis indicated that compound 2 had good potential as an antitumor lead compound.

Abstract:Objective:To study the chemical constitutes from the roots of Lindera glauca and the alkaloids influence on proliferation of HT-29,SGC-7901,SMMC-7721 and A549 cell lines.Methods:The constituents were isolated by column chromatography such as RP-18,Sephadex LH-20 and silica gel,and their structures were elucidated by spectroscopic data analysis and compared with literature data. The antitumor activity was determined by MTT assay.Results:Ten compounds had been isolated and identified as(-)-magnocurarine( 1),N-methyl-laurotetanine( 2),laurotetanine( 3),( +)-boldine( 4),(-)-norisoboldine( 5),( +)-norisocorydine( 6),pmethane-3,8-trans-diol( 7),p-methane-3,8-cis-diol( 8),eudesm-4( 15)-ene-7,11-diol( 9) and 4β,6β-dihydroxy-1α,5β( H)-guai-9-ene( 10). Compounds 2 ~ 4 showed significant inhibitory activities against HT-29,SGC-7901,SMMC-7721 and A549 cells.Conclusion:Compound 1,9 and 10 are isolated from this plant for the first time. The IC50 value of compound 2 against HT-29 and SGC-7901 cell lines is even lower than VP-16.
Isolation, synthesis and bioactivity evaluation of isoquinoline alkaloids from Corydalis hendersonii Hemsl. against gastric cancer in vitro and in vivo

Abstract:Isoquinoline alkaloid displays significant anti-gastric cancer effects due to its unique structure, which is attracting more and more attention for the development of anti-gastric cancer drugs. In this study, we explore the active components against gastric cancer from the Tibetan Medicine Corydalis hendersonii Hemsl, which is rich in isoquinoline alkaloids. 14 compounds including 2 previously undescribed natural products were obtained. Interestingly, an new active compound displays potent anti-gastric cancer activity. After accomplishing the total syntheses of the active compound and its derivatives, the anti-gastric cancer activity of the active compound was further investigated. In vitro experiments revealed that the active compound significantly attenuated the proliferative capacity, caused G2/M phase arrest, inhibited the cell migration and invasion, and induced cell apoptosis. Mechanistically, the active compound could increase the Bax/Bcl-2 ratio, elevate cytochrome c in the cytosol, and activate caspase-9/3, along with inactivating the upstream PI3K/Akt/mTOR signaling pathway. In addition, the active compound could also cause gastric cancer cell death by inhibiting topoisomerase I activity. More importantly, the anti-gastric cancer activity of the active compound was confirmed in MGC-803 xenograft nude mice in vivo. This work not only promotes the exploitation of Corydalis hendersonii Hemsl., but also provides some experience for discovering new entities from natural sources.

Synthesis and Antitumor Activity Study of Nitrogen-Containing Curcumin Derivatives

Abstract:Research on new antitumor drugs is of great significance since most of the currently available drugs had the disadvantages of high toxicity,low effection and multidrug resistance against subset of solid tumors.Structure modification of natural products may still deliver new hope for antitumor drug discovery.Curcumin and flavonoids are two kinds of important natural products with antitumor activity for recent years.Based on SAR of curcuminoids and our previous work,15 new novel curcumin analogues were designed and synthesized according to pharmacophore and scaffold hopping principle.Structurally,the new analogues are 1,5-diarylpentadienones which possess one nitrogen-containing substitution at the position 5 on one of the aromatic ring.The target compounds 6a-6n and 6o were obtained using 4-hydroxy-3-methoxybenzaldehyde and 2,4-dihydroxybenzaldehyde as starting materials,respectively.After a series of reactions,and the new compounds were finally obtained via Mannich reaction.The structure of new compounds were confirmed by MS(ESI) and 1H-NMR.All of the new compounds were screened for antiproliferation activity against two human tumor cell lines in vitro.Most of the new analogues showed more potent antiproliferation activitities against both tested cells,than that of curcumin,with IC50 values below 10 μmol · L-1.The substituted groups introduced in the aromatic ring didn't make significant effect on their activity.The IC50 value of compound 6c and 6b was lower than that of curcumin and chalcone.The results indicated that compounds with alkaline groups,especially morpholine group,resulted in stronger potent activities.Compounds with cyclohexone(6m,6n) showed decreased activities than that with straight carbon chain(6a).Also,compounds with the second alkaline side chain(6h,6k) did not enhance the activities.The compound(6l) introduced a methy group at 4-position obviously resulted in the decreased antitumor activity.In summary,new compounds with five-carbon chain exhibited more potent activities than those with seven-carbon or three-carbon chain.However,substituted groups were introduced at the 4-positon and 2-position resulted in the decreased bioactivities of compounds.And compounds with a chlorine at the 2-position or trifluoromethyl at the 4-position of ring B enhanced antitumor activities.And the'drug-like' properties predicted by Discovery Studio 2.0 suggested that most of them may be well absorbed by intestine and have good bioavailability.

Abstract:A series of novel podophyllotoxin derivatives were designed and synthesized. The cytotoxic activities of these compounds were tested against three tumor cell lines (HeLa, K562, and K562/A02). Most of the derivatives (IC50 = 1-20 mu M) were found to have stronger cell growth inhibitory activity than positive control etoposide. Among them, 4 beta-N-[(E)-(5-((4-(4-nitrophenyl)-piperazin-1-yl)methyl)furan-2-yl) prop-2-en-1-amine]-4-desoxy-podophyllotoxin (9l) demonstrated significant inhibitory activity against HeLa, K562, and K562/A02 cell lines with IC50 values of 7.93, 6.42, 6.89 mu M, respectively.

Isovaleryl Sucrose Esters from Atractylodes japonica and Their Cytotoxic Activity

Abstract:Cancer represents one of the most significant health challenges currently facing humanity, and plant-derived antitumour drugs represent a prominent class of anticancer medications in clinical practice. Isovaleryl sucrose esters, which are natural constituents, have been identified as having potential antitumour effects. However, the mechanism of action remains unclear. In this study, 12 isovaleryl sucrose ester components, including five new (1–5) and seven known compounds (6–12), were isolated from the roots of Atractylodes japonica. The structures of the compounds were elucidated using 1D and 2D-NMR spectroscopy, complemented by HR-ESI-MS mass spectrometry. The cytotoxic activities of all the compounds against human colon cancer cells (HCT-116) and human lung adenocarcinoma cells (A549) were also evaluated using the CCK8 assay. The results demonstrated that compounds 2, 4, and 6 were moderately inhibitory to HCT-116 cells, with IC50 values of 7.49 ± 0.48, 9.03 ± 0.21, and 13.49 ± 1.45 μM, respectively. Compounds 1 and 6 were moderately inhibitory to A549, with IC50 values of 8.36 ± 0.77 and 7.10 ± 0.52 μM, respectively. Molecular docking revealed that compounds 1–9 exhibited a stronger affinity for FGFR3 and BRAF, with binding energies below −7 kcal/mol. Compound 2 exhibited the lowest binding energy of −10.63 kcal/mol to FGFR3. We screened the compounds with lower binding energies, and the protein-ligand complexes already obtained after molecular docking were subjected to exhaustive molecular dynamics simulation experiments, which simulated the dynamic behaviour of the molecules in close proximity to the actual biological environment, thus providing a deeper understanding of their functions and interaction mechanisms. The present study provides a reference for the development and use of iso-valeryl sucrose esters in the antitumour field.

Abstract:OBJECTIVE:To study the alkaloids from Corydalis impatiens.METHODS:The alkaloids were isolated and purified by chromatography and their structures were identified by spectral data and others methods.RESULTS:Seven alkaloids were isolated and identified as bicuculline(1), ochotensine(2), ochotensimine(3), ochrobirine(4), tetrahydrothalifendine(5), norochotensimine(6), N-methylactinodaphnine(7).CONCLUSION:All these compounds are isolated from this plant for the first time.

Abstract:In the present study, HSQC-based Small Molecule Accurate Recognition Technology (SMART) strategy discovery of diverse cytotoxic isoquinolines. Two new isoquinoline alkaloids, corydalisines A and B ( 1 and 14 ), along with 17 known isoquinoline alkaloids ( 2 - 13, 15 - 19 ) were obtained from Corydalis saxicola . The structures were characterized using spectroscopic data analysis (NMR, HR-ESI-MS, IR, UV and X-ray diffraction), and the absolute configurations of new compounds were established by ECD data analysis. Biologically, Compounds 3, 7 , and 17 - 19 exhibited moderate cytotoxicity against six human cancer cell lines, with IC 50 values of 4.16-48.66 μM.

Abstract:This paper aimed to study the chemical constituents from Clitocybe clavipes. Silica gel, ODS, Sephadex LH-20, and semi-p reparative HPLC were employed to separate the ethanol extract of C. clavipes. Six compounds were identified by ~1H-NMR, ~(13)CNMR,and ESI-MS as clavilactone L(1), clavilactone A(2), clavilactone B(3), clavilactone E(4), clavilactone H(5), and clav ilactone I(6). Among them, compound 1 was a new meroterpenoid with a 10-membered carbocycle connected to a hydroquinone. Theantitumor activities of compounds 1-6 were determined by the methyl thiazolyl tetrazolium(MTT) ass ay. The results showed that compounds 1-6 exerted inhibitory effects on the proliferation of human gastric cancer cells(MGC-803),human non-small cell lung cancer cells(A549), and cervical cancer cells(HeLa). Compound 1 exhibited significant inhibitory activity against MGC-803 cells, with the half maximal inhibitory concentration(IC_(50)) of 11. 76 μmol·L~(-1).

Abstract:Nine alkaloids and two phenolic glycosides were isolated from EtOH extract of the whole plants of Corydalis hendersonii by various chromatographic techniques including silica gel, ODS, Sephadex LH-20, and semi-preparative HPLC. Their structures were identified as groenlandicine (1), berberine (2), protopine (3), cryptopine (4), N-trans-feruloyloctopamine(5), 3-(4-hydroxy-3-methoxyphenyl)-N-[2-(4-hydroxyphenyl)-2-methoxyethyl] acrylamide (6), N-cis-p-coumaroyloctopamine (7), N-trans-p-coumaroylnoradrenline (8),N-cis-feruloyloctopamine (9), apocynin (10), and glucoacetosyringone (11) by the spectroscopic data analysis and comparison with those in the literature. Among them, compounds 10 and 11 were isolated from this genus for the first time, and 1, 2, and 5-9 were isolated from the species for the first time. All isolates were tested for their protection for in vitro PC12 cell line and antiplatelet aggregation activity. The results showed that compounds 5 and 7 displayed protective effects at a concentration of 10 μmol·L⁻¹, and compound 2 showed antiplatelet aggregation activity induced by THR, ADP, and AA, and compound 3 exhibted inhibitory effect induced by THR.

Abstract:Ten phenylpropanoid amides were isolated from the whole plants of Corydalis edulis Maxim. by various of column chromatographies including silica gel, Sephadex LH-20, and ODS. Their structures were identified on the basis of physicochemical properties, MS, NMR, and IR spectroscopic data. These compounds were identified as N-trans-sinapoyl-3-methoxytyramine-4'-O-β-glucoside(1), N-trans-sinapoyl-3-methoxytyramine(2), N-trans-sinapoyltyramine(3), N-trans-p-coumaroyltyramine(4), N-trans-sinapoyl-7-hydroxytyramine(5), N-cis-feruloyltyramine(6), N-cis-p-coumaroyltyramine(7), N-trans-feruloyltyramine(8), N-trans-feruloyl-3-methoxytyramine(9), and N-trans-feruloyl-7-hydroxytyramine(10). Compound 1 is a new compound. Compounds 2-7 are obtained from the plants of Papaveraceae for the first time, while compounds 8-10 are firstly isolated from C. edulis.
Two New Indole Alkaloids from Nauclea Officinalis and Their Evaluation for Cytotoxic Activities

Abstract:Two new indole alkaloids, naucleamide H (1) and (+/-)-19-O-butylangustoline (8), along with seven known alkaloids, 3,14-dihydroangustine (2), (-)-naucleofficine D (3a), (+)-naucleofficine D (3b), nauclefine (4), angustidine (5),19-O-ethylangustoline (6) and angustine (7) were isolated from the water extract of Nauclea officinalis. The structures of these compounds were established by spectroscopic analysis. Among them, the cytotoxicity of 1, 2, 6 and 8 were evaluated against six human cancer cell lines (HepG-2, SKOV3, HeLa, SGC 7901, MCF-7 and KB) in vitro for the first time with 5-fluorouracil as a positive control drug. The new compound 1 had a strong inhibitory effect on the proliferation of HepG-2 with an IC50 value of 19.59 mu g/mL. The new compound 8 had a strong inhibitory effect on HepG-2, SKOV3, HeLa, MCF-7 and KB, IC50 value was 5.530, 23.11, 31.30, 32.42 and 37.26 mu g/mL, respectively. [GRAPHICS] .

Abstract:Two series of diphyllin 4‐C derivatives were designed and synthesized, including six ester derivatives and eight 1, 2, 3‐triazole derivatives. They showed potent cytotoxicity and V‐ATPase inhibitory. The natural lignan diphyllin has shown promising antitumor activity, although its clinical advancement has been impeded by challenges such as low solubility, poor metabolic stability, and limited potency. In response, we developed and synthesized two sets of diphyllin 4‐C derivatives, comprising six ester derivatives and eight 1, 2, 3‐triazole derivatives. Notably, among these derivatives, 1, 2, 3‐triazole derivatives 7c and 7e demonstrated the most potent cytotoxic effects, with IC50 values ranging from 0.003 to 0.01 μM. Treatment with 0.2 μM of 7c and 7e resulted in a reduction of V‐ATPase activity in HGC‐27 cells to 23% and 29%, respectively.

Abstract:In our continuing search for more potent anticancer agent, eight novel nitrates couple with C-10 colchicine nitric oxide-releasing derivatives (5a-f and 7a-b) were prepared. These target compounds were assayed for their anti-tumor activity in vitro against four cancer cell lines, including human hepatocellular carcinoma cells (BEL7402), human ovary carcinoma cells (A2780), human lung adenocarcinoma cells (A549) and human breast carcinoma cells (MCF7). Preliminary results indicated that most of them exhibited significant cytotoxic effect toward cancer cells. Compound 7a was investigated further for its more potent cytotoxicity than colchicine.

The structures of the new isolates including absolute configurations were elucidated by the analysis of spectroscopic data and comparison of the experimental and calculated electronic circular dichroism (ECD) data. Compound 1 inhibited the NO production in lipopolysaccharide (LPS)-induced RAW 264.7 cells with an IC50 value of 70.3 mu M, and increased the cell viabilities with 40.0 +/- 3.9% against the oxygen glucose deprivation injury in H9c2 cells at 40 mu M.

Abstract:Corydalis hendersonii(CH) is a Tibetan folk medicine with the functions of clearing heat, detoxifying, cooling blood, checking diarrhea, and lowering blood pressure. It is often used to treat high altitude polycythemia, vasculitis, peptic ulcer, and diarrhea. Nine compounds were separated from the ethanol extract of CH by silica gel, ODS, Sephadex LH-20 chromatography and semi-preparative HPLC. Their structures were identified as hendersine H(1),hendersine I(2), dehydrocheilanthifoline(3), protopine(4), izmirine(5), 6,7-methylenedioxy-1(2H)-isoquinolinone(6), icariside D_2(7), ethyl 4-(β-D-glucopyranosyloxy)-3-methoxybenzoate(8), 3-hydroxy-4-methoxybenzoic acid(9), respectively, by the spectroscopic data analysis and comparison with those in the literature. Among them, compounds 1 and 2 are new isoquinoline alkaloids, and compounds 7-9 are reported the first time for Corydalis. The hypoglycemic model of H9c2 cardiomyocytes and the inflammatory model of H9c2 cardiomyocytes induced by conditional supernatant were employed to determine the activities of the above compounds. The results showed that 20 μmol·L~(-1) compound 1 had a protective effect on H9c2 cardiomyocytes and 10 μmol·L~(-1) compounds 4 and 5 inhibited H9c2 cardiomyocyte inflammation induced by conditional supernatant.

Abstract:A series of glycyrrhetinic acid, oleanolic acid and ursolic acid derivatives were designed and synthesized. Structures of all novel compounds were determined by 1H NMR, 13C NMR, and HRMS methods. In vitro anti-proliferative effects of all compounds were evaluated against A549, Hela and HepG2 cancer cell lines. The three compounds G2, G3 and G4 showed better anti-proliferative effects than the positive control doxorubicin on the three kinds of cancer cells. The two compounds O1 and U11 showed better anti-proliferative effects than doxorubicin on Hela cells. The four compounds O2, O3, O8 and U8 showed better anti-proliferative effects than doxorubicin on HepG2 cells. Notably, compound G3 demonstrated the strongest growth inhibitory effects with IC50 values of 0.16 ± 0.23 µM and 0.33 ± 0.41 µM on Hela and HepG2 cells, respectively, and compound G2 demonstrated the strongest growth inhibitory effect with IC50 values of 0.80 ± 1.03 µM on A549 cells.
