Abstract:The objective was to investigate the in vitro antioxidant activity of a novel polysaccharide AAP-3-1 from Auricularia auricula. AAP-3-1 was isolated from the fruiting bodies of A. auricula by hot water extraction and ethanol precipitation and was purified by DEAE FF ion exchange chromatography and Superdex 200 gel filtration chromatography. The molecular weight and monosaccharide composition were determined by high performance gel permeation chromatography and high-performance liquid chromatography, respectively. Ultraviolet visible spectroscopy, Fourier transform infrared spectroscopy and scanning electron microscopy were used for structural characterization. The results showed that AAP-3-1 is a heteropolysaccharide and is mainly composed of mannose and glucose in a molar ratio of 1.4:1 with a molecular weight of 320.9 kDa. AAP-3-1 exhibited antioxidant activity in a concentration-dependent manner and the scavenging rates at 1.6 mg/mL on superoxide anions and hydroxyl and 1,1-diphenyl-2-picrylhydrazyl radicals were 88.13%, 93.03% and 68.31%, respectively. AAP-3-1 effectively ameliorated 2,2'-azobis-2-methyl-propanimidamide-induced oxidative stress in HepG2 cells by inhibiting reactive oxygen species generation, decreasing the content of malondialdehyde, and increasing the activities of superoxide dismutase, glutathione peroxidase and catalase. The results indicated that the antioxidant mechanism of AAP-3-1 was associated with both non-enzymatic and enzymatic defense systems.

Abstract:An efficient extraction method of Auricularia auricula polysaccharides (AAPs) by neutral protease was developed and optimized by response surface methodology. AAPs were graded by stepwise ethanol precipitation, the fraction with high recovery rate and strong radical scavenging rate were obtained, then its antioxidant and lipid lowering effect were studied using Caenorhabditis elegans as model organism. The extract yield and ABTS+ scavenging rates of AAPs could reach 14.90% and 86.0% at 50 °C, 75 mL/g of liquid-to-material ratio and pH 9.0. AAP3 obtained by 15% ethanol was a heteropolysaccharide comprised of mannose, glucose, glucuronic acid, xylose, galactose and glucosamine. AAP3 could significantly prolong the lifespan of C. elegans and enhance the activity of antioxidant enzymes including superoxide dismutase (SOD), catalases (CAT) at 0.25 mg/mL (p < 0.05). The qRT-PCR results showed that AAP3 could up regulate mRNA expression levels of daf-16 and skn-1 (>1.6 fold) at 0.25 mg/mL. Besides, AAP3 could significantly reduce the level of body fat and triglyceride in C. elegans (p < 0.05). These studies demonstrated that A. auricula polysaccharides prepared by neutral protease had a prominent protective effect to the damage induced by the intracellular free radical generating agents.

Abstract:Auricularia auricula-judae polysaccharide (AAP)-based nanoparticles (NPs) prepared via an anti-solvent precipitation approach were studied. Response surface methodology (RSM) design was carried out on the basis of single factor experiments, using average size and polydispersity index (PDI) as indicators. The optimal preparation conditions were determined to include an AAP concentration of 1 mg/mL, a pH of 8, and an anti-solvent/solvent volume ratio of 6. The average particle sizes of the AAP-NPs, PDI and electrical characteristic (ζ-potential) were found to be 150.27 ± 3.21 nm, 0.135 ± 0.012 and −31.10 ± 0.52 mV, respectively. Furthermore, Fourier transform infrared spectroscopy (FTIR) was used to determine the chemical structure of the AAP-NPs. It was observed that the intensity of AAP-NPs in the wide spectral band of 3000–3750 cm−1 was significantly stronger than that of the AAP, as was the characteristic peak of carboxyl anion, and the characteristic band moved to shorter wavelengths. Subsequent thermogravimetric analysis showed that the antisolvent precipitation method improved the thermal stability of the AAP, while scanning electron microscopy (SEM) and X-ray diffraction (XRD) showed that the morphology of AAP-NPs was uniform and well-distributed, and that their single crystal structures had remained unaffected during the process. Moreover, the DPPH and ABTS scavenging activities of AAP-NPs were increased, and the IC50 values were 0.544 ± 0.241 mg/mL and 0.755 ± 0.226 mg/mL, respectively.

Abstract:Auricularia auricula is a well-known traditional edible fungus with high nutritional and pharmacological values. Polysaccharides are identified as one of the pivotal bioactive constituents of natural extracts of A. auricula. Accumulating evidence has revealed that polysaccharides from A. auricula (AAPs) exhibit diverse biological functions. Hence, a systematic review of research advances and future prospects of AAPs is very necessary for facilitating their better understanding. In this review, we have summarized current knowledge about extraction and purification methods, structural features, biological activities and potential molecular mechanisms of AAPs. Besides, the proposed structure-activity relationships of AAPs have also been highlighted and discussed. This review may provide some valuable insights for further researches regarding AAPs.

Abstract:Polysaccharides derived from Auricularia auricula exhibit diverse biological activities and hold significant potential for commercial utilization as functional food ingredients. In this investigation, polysaccharides from A. auricula were obtained using six extraction techniques (ammonium oxalate solution extraction, sodium hydroxide solution extraction, hot water extraction, pectinase and cellulase-assisted extraction, ultrasonic-assisted extraction, and microwave-assisted extraction). Subsequently, a comprehensive comparison was conducted to evaluate their physicochemical properties and biological functionalities. The ammonium oxalate solution extraction method yielded a higher extraction rate (11.76%) and polysaccharide content (84.12%), as well as a higher uronic acid content (10.13%). Although the six Auricularia polysaccharides had different molecular weight distributions, monosaccharide molar ratios, similar monosaccharide compositions, and characteristic functional groups of polysaccharides, they exhibited different surface morphology. In vitro assays showed that polysaccharides extracted by ammonium oxalate solution possessed good scavenging ability against DPPH free radical, hydroxyl free radical and superoxide anion free radical as well as reduction power of iron ion. At the same time, both polysaccharides extracted by ammonium oxalate solution and sodium hydroxide solution promoted NO production in mouse macrophages along with the secretion of cytokines TNF-α, IL-1β, and IL-6. These results indicated significant differences in the structure and characteristics among Auricularia polysaccharides prepared by various extraction methods, which may be related to the variety or origin of A. auricula; furthermore, their bioactivities varied accordingly in vitro assays where the ammonium oxalate solution extraction method was found more beneficial for obtaining high-quality bioactive Auricularia polysaccharides.

Abstract:This research enhanced the extraction procedure for Auricularia auricula crude polysaccharides by utilizing a modified Fenton reagent as a solvent, and obtained A . auricula polysaccharides (AAPs-VH) via alcohol precipitation and deproteinization. The HPLC profile revealed that the purified AAPs-VH using Sepharose 6FF was mainly a heteropolysaccharide, consisting primarily of mannose, glucuronic acid, glucose, and xylose. The M w and M n of the purified AAPs-VH were 87.646 kDa and 48.854 kDa, respectively. The FT-IR and NMR spectra revealed that the purified AAPs-VH belonged to pyranose and were mainly formed by (1 → 3)-linked-β-D glucan formation. In vivo experiments conducted with Caenorhabditis elegans , AAPs-VH was found to notably influence the lifespan, improve the antioxidant system, and decrease the level of cell apoptosis. This might be achieved by up-regulating the expression of genes in the IIS and TOR pathways. The study concludes that the modified Fenton reagent can increase Auricularia auricula polysaccharide solubleness and active sites, which may be an essential prompt for future studies.

Abstract:Pteridium aquilinum (L.) Kuhn (Pteridaceae family) has been widely used as a food and medicine in China and Korea. Previous studies indicate that P. aquilinum contains a variety of bioactive chemical components such as flavonoids, phenols, terpenoids, saponins, polysaccharides, and so on. In the present study, a novel polysaccharide (named as PAP-3) with average molecular weight of 2.14 Chi 10(5) Da was obtained from P. aquilinum. The structure was studied through physicochemical and spectroscopic analysis. The results indicated that PAP-3 consists of arabinose, rhamnose, fucose, galactose, mannose, and xylose in a molar ratio of 1.58:1.00:3.26:4.57:4.81:3.33. The polysaccharide is mainly composed of (1 -> 2)-linked xylose and (1 -> 3,6)-linked mannose on the main chain, with (1 -> 2)-linked xylose, (1 -> 6)-linked mannose, and (1 -> 6)- and (1 -> 3,6)-linked galactose as side chains. Galactose, fucose, and xylose are located at the end of the side chains. The in vitro immunomodulatory and antioxidant activities were assayed. PAP-3 has strong free-radical scavenging activity on DPPH and ABTS radicals and significant immunomodulatory activity on RAW264.7 cells. These data provide useful information for further study on the polysaccharides of P. aquilinum and their applications in the food and medical industries.
Potential inhibitory effect of Auricularia auricula polysaccharide on advanced glycation end-products (AGEs)

Abstract:In this study, a novel polysaccharide, AAP-2S, was extracted from Auricularia auricula, and the anti-glycosylation effect of AAP-2S and its underlying mechanisms were investigated using an in vitro BSA-fructose model and a cellular model. The results demonstrated the inhibiting formation of advanced glycation end products (AGEs) in vitro by AAP-2S. Concurrently, it attenuated oxidative damage to proteins in the model, preserved protein sulfhydryl groups from oxidation, reduced protein carbonylation, prevented structural alterations in proteins, and decreased the formation of β-crosslinked structures. Furthermore, AAP-2S demonstrated metal-chelating capabilities. GC-MS/MS-based metabolomics were employed to analyze changes in metabolic profiles induced by AAP-2S in a CML-induced HK-2 cell model. Mechanistic investigations revealed that AAP-2S could mitigate glycosylation and ameliorate cell fibrosis by modulating the RAGE/TGF-β/NOX4 pathway. This study provides a foundational framework for further exploration of Auricularia auricular polysaccharide as a natural anti-AGEs agent, paving the way for its potential development and application as a food additive.
Effect of deep eutectic solvent extraction on Auricularia auricula polysaccharide solubilization and antioxidant potential

Abstract:This study was conducted to assess the potential of a green and eco-friendly technique: ultrasound-assisted natural deep eutectic extraction (UAE-DES) for the preparation of Auricularia auricula polysaccharides (AAPs), and to evaluate the antioxidant capacity of the products. Firstly, the optimal parameters for AAPs by Box-Behnken using deionized water as the solvents, and the yield was 25.07% ± 0.43% at 50 °C, 70 mL/g of liquid-to-material ratio and 150 W of ultrasonic power. Next, the effects of five types of DES on the output of the AAPs under this parameter were investigated. DES (ethylene glycol-choline chloride, molar ratio of 2:1) containing 40% water increased the yield by 18.68%. The molecular weight of AAPs-UW (using deionized water as solvents) was 851.029 kDa, while the UAE-DES fractured AAPs-UD (using DES as solvents) into three polysaccharides, 811.895, 277.178 and 15.520 kDa, respectively. AAPs-UD showed significantly stronger ABTS + /OH·/DPPH· scavenging capacity than AAPs-UW. Compared with AAPs-UW, AAPs-UD treatment prolonged the maximum lifespan and the mean survival time of C. elegans at low concentration. In addition, qRT-PCR results indicated that AAPs-UD could up-regulate the expression levels of antioxidant related genes such as daf-16 , sir-2.1 and sod-3 in the insulin and IGF-1 signaling pathway (>1.50, p < 0.05), and the expression of skn-1 in the target of rapamycin pathway (>1.70, p < 0.05).

Abstract:The antioxidant activities of polysaccharides from the fruiting body (PFB), extracellular polysaccharides (EPS) and intracellular polysaccharides (IPS) from Agaricus bitorquis (QuéL.) Sacc. Chaidam ZJU-CDMA-12 in vitro were compared. IPS showed stronger antioxidant activities than PFB and EPS in vitro. Further purification and structure analyses indicated that IPS mainly consisted of three fractions (IPS-I, IPS-II and IPS-III). FT-IR and NMR data indicated that IPS was mainly composed of (1 → 6)-linked α-d-glucose. There are significant differences of antioxidant activities among IPS-I, IPS-II and IPS-III fractions in vitro, and IPS-III showed stronger antioxidant activity than IPS-I and IPS-II. IPS-III also possesses a potent antioxidant ability inside HepG2 cells, and it could protect HepG2 cells from H2O2-induced cytotoxicity by scavenging overproduced cellular ROS and inhibiting SOD, CAT and GSH depletion to weaken lipid peroxidation. These findings suggested that IPS-III could be a novel antioxidant and that it could afford protection against H2O2-induced cytotoxicity and oxidative stress.

Abstract:Assessments of molecular weight distribution and activity/efficacy of Auricularia heimuer polysaccharides (AAP) are of substantial significance for its extraction process optimisation. In the present study, single-factor orthogonal test and response surface methodology were employed to optimise extraction conditions of AAP. Furthermore, a rat hyperlipidaemia model was established to compare the lipid-lowering activity of polysaccharides obtained by three extraction methods. Conditions for enzymatic hydrolysis were optimised as pH 5.0, 1% cellulase, 2.5% substrate concentration and enzymolysis time of 1.5 h, leading to an up to 31.8% polysaccharide yield and 89.13% of polysaccharides within the molecular weight range of 5 000 Da to 10 000 Da. The results of animal experiments showed that the lipid-lowering activity of enzymolysis-extracted polysaccharides was significantly higher than that of water- and ultrasonic-extracted ones (P < 0.01). So the present study revealed that enzymatic hydrolysis-extracted polysaccharides showed the strongest hypolipidaemia activity, providing a basis for the development of A. heimuer-based functional foods and drugs.

Abstract:In the present study, polysaccharides named ALPS and ASPS were isolated from Ampelopsis grossedentata leaves and stems, respectively. Physicochemical properties, in vitro antioxidant activities and the inhibitory effects on alpha-glucosidase of ALPS and ASPS were investigated. It was found that both ALPS and ASPS were acid protein-bound heteropolysaccharides, although with considerably different chemical composition and molecular weight distribution. Meanwhile, in comparison with ALPS, ASPS exhibited stronger antioxidant activity and inhibitory potential against alpha-glucosidase according to the in vitro evaluation. Moreover, our results suggested that protein and uronic acid might, at least partly, contribute positively to the biological behavior of ALPS and ASPS.

Abstract:We extracted, purified, and characterized three neutral and three acidic polysaccharides from the roots, stems, and leaves of Aralia continentalis Kitigawa. The results of the analysis of monosaccharide composition indicated that the polysaccharides from the roots and stems were more similar to each other than they were to the polysaccharides from the leaves. The in vitro antioxidant results demonstrated that the acidic polysaccharides had stronger antioxidant activity than the neutral fractions. Therefore, we investigated the primary purified acidic polysaccharide fractions (WACP(R)-A-c, WACP(S)-A-c, and WACP(L)-A-d) by NMR and enzymatic analysis. The structural analytical results indicated that WACP(R)-A-c contained homogalacturonan (HG); WACP(S)-A-c contained HG and rhamnogalacturonan II (RG-II), and WACP(L)-A-d contained HG, RG-II, and rhamnogalacturonan I (RG-I) domains. Our findings offer insights into the screening of natural polysaccharide-based antioxidants and provide a theoretical basis for the application of A. continentalis.

Abstract:PAP1-A, a novel heteropolysaccharide with an average molecular mass of 1.35×105Da, was isolated from Pteridium aquilinum using a combination of chromatography by DEAE Sepharose Fast Flow and Sepharose 4B. The monosaccharide component of PAP1-A was L-rhamnose, L-arabinose, L-fucose, D-xylose, D-mannose, D-glucose and D-galactose in the molar ration of 1.82: 1.53: 1.42: 1.31: 5.24: 1: 12.35. The predicted structure of PAP1-A was established to be a complex polysaccharide with a main chain consisted of α-(1→4)-linked galactose partially substituted at O-6 position, with the (1→2)-linked xylose, (1→3)-linked arabinose, (1→3)-linked rhamnose, (1→3,6)-linked mannose, and (1→6)-linked mannose, as branches. Fucose, glucose, mannose, and rhamnose were located at the termini of the branches. The immunomodulatory activity assay showed that PAP1-A could significantly promote the RAW264.7 cells proliferation and induce the production of NO from RAW264.7 cells. Therefore, PAP1-A shows as a potent immunomodulator with potential applications in the medical and food industries.

Abstract:Asteris Radix et Rhizoma (AR) has been widely used as a herbal medicine for treating various symptoms and possesses a number of bioactivities. A neutral polysaccharide ARP-1 was isolated from AR with weight-average molecular weight of 214 kDa. The heteropolysaccharide ARP-1 was composed of fucose, arabinose, galactose, glucose and mannose with a molar ratio of 0.40:14.25:10.22:1.06:0.41. Linkage and NMR analysis showed that ARP-1 had a backbone containing →3,6)-β-d-Galp-(1→ and →6)-β-d-Galp-(1 → residues, and oligosaccharide side chains containing Araf and Galp units were attached to the backbone at C-3 of →3,6)-β-d-Galp-(1 → residues. Antioxidant activity assays showed that ARP-1 exhibited potent antioxidant activities, including ABTS, hydroxy and DPPH radicals scavenging and reducing power. Besides, ARP-1 decreased the production of ROS and MDA, and improved the activities of SOD, which resulted in the protection of PC12 cells against H2O2-induced oxidative stress. Abstract:A water-soluble protein-bound polysaccharide was extracted from the fruiting bodies of Ganoderma atrum and isolated by gel-filtration chromatography. Its primary structural features and molecular weight were characterized by infrared spectrometry, gas chromatography, size exclusion chromatography, amino acid analyzer and high-performance liquid chromatography (HPLC). The data obtained indicated that the glycoprotein contains 10.1% of protein and 17 general amino acids and it is rich in glutamic acid, asparagic acid, alanine, glycine, threonine, and serine. It was mainly composed of mannose, galactose and glucose in a molar ratio of 1:1.28:4.91, with an average molecular weight of about 1013kDa. The existence of an O-glycosidic linkage in PSG-1 (polysaccharide1) was demonstrated by a β-elimination reaction. The antioxidant activity of the purified polysaccharides was evaluated in vitro by 1,1-diphenyl-2-picryl-hydrazyl (DPPH) free radical scavenging assay, self-oxidation of 1,2,3-phentriol assay. Those various antioxidant activities were compared to standard antioxidants vitamin C and BHT. It was found that the scavenging effects of the purified polysaccharides increased with measuring concentration. The results indicated that the purified polysaccharides showed strong DPPH free radical and superoxide anion radical scavenging activities. This study suggested that the purified polysaccharides could potentially be used as natural antioxidants.

Abstract:In this study, Auricularia cornea var. Li. polysaccharides (ACP) were used as the research object to prepare liposome gel and determine its antioxidant activity in vitro. Phosphorylated Auricularia cornea var. Li. polysaccharides (P-ACP) were prepared via the phosphorylation of ACP by the phosphate method. Additionally, phosphorylated Auricularia cornea var. Li. polysaccharide liposomes (P-ACPL) were prepared using a reverse evaporation method. Finally, phosphorylated Auricularia cornea var. Li. polysaccharide liposome gel (P-ACPLG) was prepared by dispersing the P-ACPL in the gel matrix. The results show that the phosphorylation of the P-ACP was 15.51%, the containment rate of the P-ACPL was 84.50%, the average particle size was (192.2 ± 3.3) nm, and the particle size distribution map had a homogeneous peak, resulting in the particle dispersion being uniform and the polydispersion index (PDI) being 0.134 ± 0.021. The average Zeta potential was (-33.4 ± 0.57) mV. In addition, the in vitro antioxidant activity of the P-ACPL was slightly higher than that of the ACP and P-ACP. After the P-ACPL was emulsified into P-ACPLG, the DPPH, hydroxyl radical clearance, and reducing the ability of P-ACPL remained unchanged. In general, the P-ACPLG prepared in this study has good antioxidant activity in vitro and can retain the antioxidant activity of P-ACPL in vitro well.
Ultrasonic–assisted extraction of polysaccharides from Auricularia auricula and effects of its acid hydrolysate on the biological function of Caenorhabditis elegans

Abstract:The present study was designed to explore the in vivo-antioxidant capacity and the probable mechanism of AAPs-H, prepared from Auricularia auricula polysaccharides with the optimal extraction conditions by Box–Behnken design and acid hydrolysis, using Caenorhabditis elegans as a model organism. The effects of AAPs-H on the locomotion behavior, life span, antioxidant-related enzymes activities, and antioxidants levels in C. elegans were studied. Furthermore, the potentials of AAPs-H in up-regulating the expression of antioxidant-related genes in C. elegans, such as sknr-2, sod-3 and sir-2.1, were also discussed. AAPs-H demonstrated a highly significant protective effect against the damage caused by paraquat, could significantly increase U-Turn frequency of worms (p < 0.01), extend their lifespan, enhance antioxidant systems including GR by 63.96% (p < 0.05), GSH-Px by 71.16% (p < 0.01), SOD by 78.65% (p < 0.01) and CAT by 98.52% (p < 0.01), increase the level of GSH by 28.12% (p < 0.05), and decrease the level of MDA by 39.29% (p < 0.01). The qRT-PCR results showed that AAPs-H could up regulate mRNA expression levels of skn-1, sod-1, sod-2, sod-3 and sir-2.1 in wild-type C. elegans (>1.6 fold) when treated with the concentration of 0.4 mg/mL (p < 0.05 or p < 0.01). Our studies provide evidence that AAPs-H improves antioxidant defense system, and up-regulation of oxidative stress related genes for prevention of stress damage in C. elegans.
