Abstract:A C‐type lectin‐like protein (Ec‐ CTLP ) was cloned from the grouper Epinephelus coioides . The full‐length cDNA of Ec‐ CTLP was composed of 905 bp with a 522 bp open reading frame that encodes a 174‐residue protein. The putative amino acid sequence of Ec‐ CTLP contains a signal peptide of 19 residues at the N‐terminus and a CLECT domain from Cys43 to Arg169 and a conserved imperfect WND (Trp‐Asn‐Asp) motif. The homologous identity of deduced amino acid sequences is from 32 to 42% with other fishes. The expression of Ec‐ CTLP was differently upregulated in E. coioides spleen (germline stem) cells after being challenged at 16 and 4° C. Intracellular localization revealed that Ec‐ CTLP was distributed only in the cytoplasm. Recombinant Ec‐ CTLP ( rEc‐CTLP ) was expressed in Escherichia coli BL21 ( DE3 ) and purified for mouse Mus musculus anti‐Ec‐ CTLP serum preparation. The rEc‐CTLP fusion protein does not possess haemagglutinating activity, but improves survival from frozen bacteria. The survival of bacteria (including gram‐negative E. coli and gram‐positive Staphylococcus aureus ) was positively correlated with the concentration of the rEc‐CTLP . These findings can provide clues to help understand the probable C‐type lectin in marine fish innate immunity.

Abstract:Interleukin-15 (IL-15) is a crucial cytokine involved in immune system regulation, which is produced by various cell types, including dendritic cells, monocytes, and macrophages. IL-15 plays a key role in the proliferation and activation of natural killer (NK) cells, CD8 + T cells, and memory CD8 + T cells, supporting their survival and enhancing their effector functions. Although IL-15 homologues in fish have been identified, their functions remain poorly understood. In this study, we cloned and investigated the bioactivities of an IL-15 homologue, referred to as IL-15 like ( Cc IL-15L), in common carp ( Cyprinus carpio L.). An expression pattern analysis revealed that CcIL-15L was constitutively expressed in all examined tissues of healthy common carp, with the highest expression level observed in the intestine. Additionally, CcIL-15L expression was significantly up-regulated in the head kidney, spleen, gills, and intestine following Aeromonas hydrophila infection. In vitro , the recombinant protein Cc IL-15L can significantly up-regulated the gene expression levels of pro-inflammatory cytokines ( IL-1β , IL-6 , IFN-γ , and TNF-α ) and NK cell activation ( perforin and Eomesa ). We constructed a 3×FLAG eukaryotic expression vector and successfully expressed it in common carp by intramuscular injection. Additionally, the heterologous Cc IL-15L protein was successfully overexpressed in vivo , and immune-related genes including CD4-1 , CD8β2 , TNF-α , and IgM showed significant induction in the head kidney and spleen. Furthermore, Cc IL-15L overexpression reduced the bacterial loads after 24 h post- A. hydrophila infection in the liver, spleen, and kidney. Phagocytic and chemotaxis assays showed that r Cc IL-15L could promoted the phagocytosis and chemotactic abilities of common carp HKLs. Our study provides a new perspective on the role for Cc IL-15L in immunological functions in common carp.

[A clinical prediction model for N2 lymph node metastasis in clinical stage I non-small cell lung cancer].

Abstract:OBJECTIVE To estimate the probability of N2 lymph node metastasis and to assist physicians in making diagnosis and treatment decisions. METHODS We reviewed the medical records of 739 patients with computed tomography-defined stage I non-small cell lung cancer (NSCLC) that had an exact tumor-node-metastasis stage after surgery. A random subset of three fourths of the patients (n=554) were selected to develop the prediction model. Logistic regression analysis of the clinical characteristics was used to estimate the independent predictors of N2 lymph node metastasis. A prediction model was then built and externally validated by the remaining one fourth (n=185) patients which made up the validation data set. The model was also compared with 2 previously described models. RESULTS We identified 4 independent predictors of N2 disease: a younger age, larger tumor size, central tumor location, and adenocarcinoma or adenosquamous carcinoma pathology. The model showed good calibration (Hosmer-Lemeshow test: P=0.923) with an area under the receiver operating characteristic curve (AUC) of 0.748 (95% confidence interval, 0.710-0.784). When validated with all the patients of group B, the AUC of our model was 0.781 (95% CI: 0.715-0.839) and the VA model was 0.677 (95% CI: 0.604-0.744) (P =0.04). When validated with T1 patients of group B, the AUC of our model was 0.837 (95% CI: 0.760-0.897) and Fudan model was 0.766 (95% CI: 0.681-0.837) (P < 0.01). CONCLUSION Our prediction model estimated the pretest probability of N2 disease in computed tomography-defined stage I NSCLC and was more accurate than the existing models. Use of our model can be of assistance when making clinical decisions about invasive or expensive mediastinal staging procedures.

Abstract:The alpha-chain of IL-15 receptor complex serves as a high-affinity, specific subunit for IL-15 binding. It shares functional and structural features with IL-2 receptor alpha-chain. Here we report for the first time that the molecular cloning, characterization and sequence analysis of the teleostean IL-15R alpha from Oncorhynchus rnykiss. The full-length cDNA comprises of a 5' untranslated region (UTR) of 167 bp, an open reading frame of 732 bp and a large 3'UTR of 630 bp, constitutively expressed in all the tissues examined. Another two variants are found derived from alternative splicing. Two copies of rainbow trout IL-15Ra (rtIL-15R alpha) were detected in the genome by Southern blot hybridization. Moreover, EST evidence is also traced from other fishes. The deduced rtIL-15R alpha of 243 amino acids contains a 17aa signal peptide at N-terminus and a transmembrane region at C-terminus, as well as a typical sushi domain included in the extracellular region. Phylogenetic tree analysis groups rtIL-15R alpha with other IL-1R alpha s but separates it from IL-2R alpha s. In addition, a putative sequence was found in the sushi domain which is conserved and versatile among all species and this might relate to cytokine recognition. (c) 2006 Elsevier Ltd. Abstract:Immunoglobulin M (IgM) is the primary antibody in fish, which is transported from epithelial cells into the external secretion system by polymeric immunoglobulin receptor (pIgR). In this study, the full length of IgM and pIgR complementary DNA sequences from leopard coral grouper (Plectropomus leopardus) was characterized, and their expression levels under Vibrio harveyi infection were studied. cDNA sequence analysis showed that the pl-IgM heavy chain cDNA contained an open reading frame (ORF) of 1797 bp encoding a polypeptide of 597 amino acids consisting of a signal peptide, a variable region, and 4 constant regions. pl-pIgR (1041 bp) encodes 346 amino acids with 2 Ig-like domains (ILDs). The expression levels of pl-IgM and pl-pIgR in healthy fish were mainly in the intestine and spleen of fish, which indicates the relationship between the mucosal-associated lymphoid tissue and system lymphoid tissue. Messenger RNA (mRNA) expression profiles of pl-IgM and pl-pIgR were significantly increased in the intestine, head kidney, spleen, and liver after V. harveyi infection. The cDNA sequences of two ILDs were cloned and expressed in Escherichia coli BL21 (DE3). The recombinant pl-pIgR protein had antibacterial function against V. harveyi and Vibrio alginolyticus at 200 ng/μl. Our results indicate that pl-IgM and pl-pIgR are involved in the mucosal-associated immune and systemic immune response in the antibacterial immunity of leopard coral grouper.

Identification and Characterization of Pro-Interleukin-16 from Mud Crab Scylla Paramamosain: the First Evidence of Proinflammatory Cytokine in Crab Species

Abstract:IL-16 is a pro-inflammatory cytokine originally designated as a lymphocyte chemoattractant factor. In mammal and avian, it has been characterized as an essential regulator of various cellular processes including cell recruitment and activation against pathogen invasion. So far, neither of the full-length of IL-16 homologue nor the response mechanism against pathogen was reported in crab species. In the present study, the pro-IL-16 homologue was firstly cloned and characterized from mud crab Scylla paramamosain. The full-length Sp-pro-IL-16 consisted of 4107 bp with an opening reading frame encoding 1369 amino acids. Multiple alignment analysis showed the putative amino acid sequence of Sp-pro-IL-16 had about 73.86% identity with Litopenaeus vannamei pro-IL-16. Additionally, two conserved PDZ domains and protein binding sites were found in Sp-pro-IL-16 and showed high similarities about 94.19% and 51.14% with their Litopenaeus vannamei and Mus musculus counterparts. RT-PCR analysis indicated that Sp-pro-IL-16 transcripts were constitutively expressed in all tissues examined with an extreme high level in hepatopancreas. Moreover, Sp-pro-IL-16 transcripts in hepatopancreas were significantly up-regulated 15-fold at 72 h after Vibrio alginolyticus challenge and 3.5-fold at 12 h after virus-analog Poly (I:C) challenge. The Western blot analysis revealed that Sp-pro-IL-16 can be cleaved to its bioactive form, an approximately 35 kDa mature IL-16, and the protein levels of both pro-IL-16 and mature IL-16 increased after Vibrio alginolyticus challenge. It is the first experimental identification of pro-inflammatory cytokine IL-16 in arthropods. This study could shed new light on further understanding of the response mechanism of pro-inflammatory cytokine IL-16 in Scylla paramamosain against pathogens. Meanwhile, it brought new insight into the origin and evolution of IL-16 in crab species.

Abstract:Interleukin-21 (IL-21) is an important immune cytokine that was well characterized in human and mammals, but little is known in fish. In present study, an IL-21 homologue was cloned and well characterized from Tetraodon nigroviridis. The full-length Tetraodon IL-21 cDNA was 849bp in size, containing an open reading frame (ORF) of 438bp that translated a 145 amino-acid peptide, a 5’ untranslated region (UTR) of 69bp, and a 3’ UTR of 342bp. The deduced peptide shared identity of 20–49% with other known IL-21 sequences. The Tetraodon IL-21 gene had six exons while both human and Takifugu IL-21 gene contained only five exons. However, the level of synteny between human, Takifugu and Tetraodon genomes was well conserved during evolution. In vivo expression study showed that Tetraodon IL-21 mRNAs were constitutively expressed at a low level and only in limited tissues, including gut, gill and gonad in healthy fish, and stimulation with LPS increased the expression of IL-21 in these tissues and induced the expression of IL-21 in kidney, spleen and skin, indicating that IL-21 is an inflammatory stress inducible gene associated with the anti-bacterial defense in fish. Our study provided further evidence for the existence of IL-21 in fish, and gained further insight into the immunological functions of IL-21 gene in fish.

Abstract:Interleukin-15 (IL-15) plays an important role in adaptive immune systems in vertebrates with similar bioactivities to interleukin-2 (IL-2). Here we report molecular cloning, sequence analysis and distribution of an IL-15 homologue from a pufferfish (Tetraodon nigroviridis). It is located within a 3088 bp genomic fragment, transcribed into a 1056 bp mRNA including 158 bp 5′UTR (untranslated region), 519 bp ORF (open reading frame) and 379 bp 3′UTR. T. nigroviridis IL-15 is constitutively detectable in tissues and organs selected. Levels of transcripts were observed after various stimulations. Gene organization is similar to mammals and birds, and a high degree of conservation of chromosome synteny exists between them. Systematic genomics search against Takifugu rubripes genome supports our conclusions. The T. nigroviridis IL-15 precursor with 172aa (amino acids) contains a putative 53aa signal peptide, while the mature peptide has a calculated molecular mass of 13.36 kDa and a theoretical pI of 4.67. The protein sequence shares 13.3–62.1% identity with reported IL-15s. Phylogenetic analysis grouped Tetraodon with other fish on a separated branch, excluded from mammalian and avian IL-15s. In addition, our analysis on another annotated T. nigroviridis IL-15 demonstrated that it may be a paralogue of IL-15. To differentiate it from the known IL-15s, we described it as IL-15x.
Molecular and functional characterization of teleost-specific Interleukin-17N in yellow catfish (Pelteobagrus fulvidraco)

Abstract:In mammals, six interleukin-17 (IL-17) genes, as potent pro-inflammatory cytokines, all accelerate the inflammatory responses. In teleosts, seven IL-17 genes have been found in various species, but little is known about the function of teleost-specific IL-17N. In this study, teleost IL-17N and IL-17A/F2 genes all had six conserved cysteine residues forming three intrachain disulfide bridges, the length of three exons of teleost IL-17N gene was similar to that of teleost IL-17A/F2 gene, and the neighbor-joining (NJ) phylogenetic tree showed that teleost IL-17N was clustered with vertebrate IL-17A/F, implying that teleost IL-17N gene may be a paralog of teleost IL-17A/F gene. Pelteobagrus fulvidraco (Pf) IL-17N gene was highly expressed in the blood, brain and kidney of healthy yellow catfish. Pf_IL-17N transcript and protein were notably up-regulated in the spleen, head kidney, gill and kidney detected after Edwardsiella ictaluri infection. Lipopolysaccharides (LPS), polyinosinic-polycytidylic acid (Poly I:C) and peptidoglycan (PGN) also remarkably induced the expression of Pf_IL-17N in the isolated peripheral blood leucocytes (PBLs) of yellow catfish. These results reveal that Pf_IL-17N may play important roles in preventing the invasion of pathogens. Furthermore, the recombinant (r) Pf_IL-17N protein could significantly induce the mRNA expressions of inflammatory cytokines, chemokines and antimicrobial peptide genes in yellow catfish in vivo and in vitro, and it also notably promoted the phagocytosis of myeloid cells in the PBLs and the chemotaxis of the PBLs and gill leucocytes (GLs) in yellow catfish. Besides, though the rPf_IL-17N protein could induce and aggravate inflammation infiltration in the kidney of yellow catfish, it did not effectively and notably increase the survival rate of yellow catfish after E. ictaluri infection. Furthermore, the rPf_IL-17N protein could induce the mRNA expressions of nuclear factor kappa-B (NF-κB) and mitogen-activated protein kinase (MAPK) signal pathways related genes, and the inhibitor of NF-κB and MAPK signal pathways could restrain the rPf_IL-17N protein-induced inflammatory response. This study provides crucial evidence that the Pf_IL-17N may mediate inflammatory response to eliminate invasive pathogens.

Abstract:Toll-like receptors (TLRs) are major pattern recognition receptors (PRRs) that recognize multiple pathogen-associated molecular patterns (PAMPs) through the leucine-rich repeat (LRR) domain and mount effective immune responses. Vibrio parahaemolyticus is the main pathogen that causes vibriosis in aquatic animals, yet the mechanisms of its recognition by innate immune system in teleost fish remain unknown. Here, the results reveal that TLR13 in orange-spotted grouper (Epinephelus coioides) (EcTLR13) recognizes a conserved 23S ribosomal RNA (23S rRNA) sequence in V. parahaemolyticus, and the 13-nucleotide motif near the 23S rRNA ribozyme activation site (VP13) acts as a PAMP. After challenge with RNA and 23S rRNA from V. parahaemolyticus and with the synthetic oligoribonucleotide VP13, the expression of EcTLR13 in grouper spleen cells (GS cells) was significantly increased. EcTLR13-knockdowned GS cells were stimulated with the same stimulants as listed above, the expression of IL-6, IL-12, IL-1β and TNFα was significantly reduced. RNA-protein immunoprecipitation revealed that VP13 could directly bind to EcTLR13. The dual-luciferase reporter assay also showed that EcTLR13 enhanced the fluorescence activity of IFNβ rather than that of NF-κB when the cells were challenged with RNA from V. parahaemolyticus or with synthetic VP13. Our study established the mechanism of fish TLR13-mediated recognition of microbial products during V. parahaemolyticus infection.

Abstract:Fish interleukin (IL)-17A/F is homologous with mammalian IL-17A and IL-17F, which plays a key role in regulating inflammatory responses and autoimmune diseases. In fish, IL-17A/F1, 2, and 3 have been identified and described. However, IL-17A/F3 has received little attention in fish. In this study, a homolog of IL-17A/F3 was identified in common carp (Cyprinus carpio L.), which was termed as Cc_IL-17A/F3. The deduced amino acid sequence of Cc_IL-17A/F3 has four conserved cysteine residues, which could form two intrachain disulfide bonds. Homology comparison showed that the Cc_IL-17A/F3 was in the range of 31.7–71.9% of sequence similarity with these of other fishes. The Cc_IL-17A/F3 gene was constitutively expressed in various tissues, with higher expression levels in the skin and gills. After common carp were infected by Aeromonas. hydrophila, the mRNA expression levels of Cc_IL-17A/F3 were significantly up-regulated in the spleen, head kidney, gills, and intestine. Based on the indirect immunofluorescence assay, Cc_IL-17A/F3 proteins were found to be obviously increased in the intestine and spleen upon A. hydrophila infection at 24 h post-infection. The recombinant protein rCc_IL-17A/F3 could enhance the gene expression levels of pro-inflammatory cytokines (IL-1β, IL-6, IFN-γ, and TNF-α) as well as chemokines (CXCL8 and CXCL20) in primary head kidney leukocytes. In vivo and in vitro experiments have similar stimulatory effects. When Cc_IL-17A/F3 was overexpressed in common carp, the expressions of pro-inflammatory cytokines and chemokines were significantly up-regulated in head kidney and spleen. In summary, the results derived from the present study suggested that the Cc_IL-17A/F3 plays an important role in defending against bacterial infections, and probably participates in mucosal immunity of the host.

Abstract:Interleukins (ILs) play important roles in transmitting information, mediating proliferation and differentiation of immune cells, and regulating immune responses. However, due to the large variety, number, and complexity of IL genes, complete and systematic studies on ILs remain largely unknown. In the present study, we identified and characterized 39 IL genes in grass carp (Ctenopharyngodon idella) based on genomic and transcriptomic datasets. These IL genes were classified into six gene families and others. Gene structures, conserved domains, and motif analyses indicated that ILs in the same gene family were more conserved. Collinearity analysis showed that the segmental duplication events played a critical role in the expansion of IL gene families during evolution. RNA-seq data from different healthy tissues showed that many IL genes were widely expressed in various tissues. Based on RNA-seq data, we further analyzed the expression profiles of IL genes in tissues post grass carp reovirus (GCRV) or Aeromonas hydrophila challenge, and the results showed that the expressions of some genes were significantly upregulated or downregulated post-challenge. Finally, qRT-PCR verified mRNA expressions of the representative IL genes post GCRV or A. hydrophila challenge. Taken together, these studies provide a theoretical basis for a comprehensive understanding of ILs and further functional investigation.

Specific bioactivity of IL-22 in intestinal cells as revealed by the expression of IL-22RA1 in Mandarin fish, Siniperca chuatsi

Abstract:IL-22, a multifunctional cytokine, acts as an important regulator in host immunity in mammals. IL-22 homologues have been characterized in several species of fish, with its expression found in multiple tissues/cells in fish, but its target cells have not been fully analyzed. In the present research, different organ/tissue isolated cells were examined for the expression of IL-22 and the induced IL-22 responses in mandarin fish. The mandarin fish IL-22 was found to be expressed in all these tested cells with high basal expression in intestinal cells. The HKLs showed low basal expression but significant increase in expression of IL-22 after LPS treatment or bacterial infection. Only intestinal cells showed response to IL-22 by enhanced expression of hepcidin, LEAP2 and IL-22BP, with unresponsiveness observed in other tested cells, which indicated the cell-specificity of IL-22 bioactivity in mandarin fish. One of the heterodimeric receptor components for IL-22, the IL-22RA1, was cloned in mandarin fish, with four tandem fibronectin type III (FNIII) domains identified in its extracellular part. IL-22RA1 exhibited an intestinal cell-specific expression pattern, although another receptor component of IL-22, IL-10R2, displayed constitutive expressions in all these tested cells. The present study reveals that the mandarin fish IL-22 exhibits its bioactivity in a cell-specific manner in intestinal cells, which is reflected in the restrictive expression of its receptor unit, IL-22RA1.

Expression and Function Analysis of Interleukin-17A/F1, 2, and 3 Genes in Yellow Catfish (Pelteobagrus fulvidraco): Distinct Bioactivity of Recombinant IL-17A/F1, 2, and 3

Abstract:In mammals, Interleukin-17 cytokine family plays critical roles in both acute and chronic inflammatory responses. In fish species, three Interleukin-17A/F (IL-17A/F) genes have been identified to be homologous to mammalian IL-17A and IL-17F, but little is known about their functional activity. In this study, Pf _IL-17A/F1, 2 and 3 genes were cloned from yellow catfish ( Pelteobagrus fulvidraco ) and they differed in protein structure and exon length, implying that they may have divergent bioactivity. Real-time quantitative PCR analyses revealed that three Pf _IL-17A/F genes were highly expressed in blood and mucosal tissues (skin+mucus and gill) from healthy adult fish. The mRNA expressions of Pf _IL-17A/F1, 2 and 3 genes were significantly up-regulated in the gill, skin+mucus, head kidney and spleen after challenge with Edwardsiella ictaluri and in the isolated peripheral blood leucocytes (PBLs) of yellow catfish after stimulation with phytohaemagglutinin (PHA), lipopolysaccharides (LPS), peptidoglycan (PGN) and polyinosinic-polycytidylic acid (Poly I:C). These results indicate that Pf _IL-17A/F1, 2 and 3 genes may play a vital role in the regulation of immune against pathogens. Additionally, the recombinant (r) Pf _IL-17A/F1, 2 and 3 proteins significantly induced the mRNA expressions of proinflammatory cytokines, chemokines and antibacterial peptides genes, and the r Pf _IL-17A/F 2 and 3 proteins promoted phagocytosis of PBLs more powerfully than the r Pf _IL-17A/F1. Furthermore, the r Pf _IL-17A/F1, 2 and 3 proteins might activate the NF-κB and MAPK signal pathways by IL-17RA, ACT1, TRAF6, TRAF2, TRAF5 and TAK1, indicating that the three Pf _IL-17A/F proteins may play different roles in promoting inflammatory response.

Abstract:Background This study was to determine whether patients with significant nighttime heartburn had more disturbed sleep and more gastroesophageal acid reflux than those without significant nighttime heartburn. Methods Thirty-three reflux patients were stratified into 2 groups (nighttime heartburn, without nighttime heartburn). All patients completed questionnaires assessing daytime and nighttime heartburn and subjective sleep by Pittsburgh Sleep Quality Index. All participants underwent 24-hour esophageal pH monitoring and an overnight polysomnographic study. Results The number of reflux events longer than 5 minutes was significantly greater in patients with nighttime heartburn than in those without nighttime heartburn for the total (P=0.004) and upright (P=0.01) position periods. Acid contact time was significantly greater in patients with nighttime heartburn during the upright (P=0.003) period and during the total (P=0.001) period. Patients with nighttime heartburn reported significantly greater subjective sleep impairment (8.1±0.7) than those without nighttime heartburn (6.1±0.4; P=0.02), but no difference could be observed in any objective sleep parameter by an overnight polysomnographic study between the groups. Conclusions Patients with significant nighttime heartburn seem to have more acid reflux compared with those without nighttime heartburn. Nighttime heartburn together with sleep complaints is associated with excessive gastroesophageal reflux.
The bioactivity of teleost IL-6: IL-6 protein in orange-spotted grouper (Epinephelus coioides) induces Th2 cell differentiation pathway and antibody production

Abstract:Interleukin 6 (IL-6) is a protein secreted by T cells and macrophages and plays an important role in immune response. IL-6 regulates the proliferation and differentiation of T cells, and elicits immunoglobulin production in B cells. In this study, the cDNA il-6 (gil-6) sequence of the orange spotted grouper (Epinephelus coioides) was obtained. The deduced IL-6 (gIL-6) protein comprised 223 amino acids, the sequence shared approximately 30% similarity with mammalian IL-6, and between 47% and 69% similarity with other available teleost IL-6. The protein comprises the signal peptide, the IL-6 family signature, and conserved amino acid residues found in IL-6 sequences of other teleost. In order to understand the bioactivity and influence of gIL-6 on humoral immune response, recombinant gIL-6 (rgIL-6) synthesized by prokaryotes was injected into orange spotted groupers, and the immune-related gene expression at various times in various organs was observed. Our results revealed that the Th1 specific transcription factor t-bet was down-regulated and Th2 specific transcription factors gata3, and c-maf were up-regulated in immune organs, following IL-6 stimulation. Additionally, higher levels of igm mRNA and translated protein were detected in rgIL-6 stimulated fish. These results indicate that IL-6 in groupers regulates the differentiation of naїve T helper cells into Th2 cells and elicits the production of antibodies.

Abstract:Interleukin-8 (IL-8 or C-X-C motif chemokine ligand 8, CXCL8) is a cytokine secreted by numerous cell types and is best known for its functional roles in inflammatory response by binding to specific receptors (the interleukin-8 receptors, IL-8Rs). From the transcriptomic data of largemouth bass (Micropterus salmoides), we identified an IL-8R that is highly homologous to the functionally validated teleost IL-8Rs. The M. salmoides IL-8 receptor (MsCXCR2) was further compared with the C-X-C motif chemokine receptor 2 subfamily by phylogenetic analysis. Briefly, the full-length CDS sequence of MsCXCR2 was cloned into the pEGFP-N1 plasmid, and the membrane localization of fusion expressing MsCXCR2-EGFP was revealed in HEK293 cells. To determine the functional interaction between IL-8 and MsCXCR2, secretory expressed Larimichthys crocea IL-8 (LcIL-8) was used to stimulate MsCXCR2 expressing cells. MsCXCR2 was demonstrated to be activated by LcIL-8, leading to receptor internalization, which was further revealed by the detection of extracellular regulated protein kinase (ERK) phosphorylation. Quantitative real-time PCR was used to evaluate the expressional distribution and variation of MsCXCR2 in healthy and Nocardia seriolae infected fish. Based on our findings, MsCXCR2 was constitutively expressed in all examined tissues, despite at different levels. Furthermore, gene expression was found to be significantly upregulated in the liver and head kidney of diseased fish. Collectively, our findings reveal the molecular activity of MsCXCR2 and indicate the functional involvement of this IL-8R in the immune response induced by N. seriolae in M. salmoides.

Abstract:Interleukin-16 (IL-16), as a lymphocyte chemoattractant cytokine, plays a crucial role in regulating cellular activities and anti-pathogen immunity. In teleost, the information about the antibacterial effect of IL-16 is scarce. In our study, we examined the immune functions of an IL-16 homologue (CsIL-16) from tongue sole Cynoglossus semilaevis. The CsIL-16 precursor (proCsIL-16) is comprised of 1181 amino acid residues, sharing 21.1%–67.3% identities with IL-16 precursor from invertebrate and vertebrate. The C-terminal proCsIL-16 containing two PDZ domains was designated as mature CsIL-16 which was released into the supernatant of peripheral blood leukocytes (PBLs). CsIL-16 was expressed in various tissues and regulated by bacterial invasion. Recombinant CsIL-16 (rCsIL-16), as a homodimer, was able to bind to the membrane of PBLs and played essential roles in regulating chemotaxis and activation of PBLs, which in vitro inhibited intracellular survival of E. tarda. Under in vivo condition, rCsIL-16 could dramatically regulate the induction of inflammatory genes, and suppress the bacterial dissemination in fish tissues. Collectively, our results reveal that CsIL-16 plays positive roles in antibacterial immunity, and provide insights into the immune function of CsIL-16.

Abstract:Myeloid differentiation factor 88 (MyD88) is a universal adaptor protein involved in Toll-like receptors and in interleukin-1 receptor-induced nuclear factor-kappaB (NF-κB) activation. In this study, a new MyD88 gene (designated as Og-MyD88) was cloned from orange-spotted grouper, Epinephelus coioides, based on the expressed sequence tag (EST) obtained following Roche 454 GS-FLX™ sequencing. The full-length Og-MyD88 cDNA is composed of 1682 bp and encodes a deduced polypeptide of 289 amino acids with 86% homology to MyD88 of Siniperca chuatsi. The deduced amino acid sequence of Og-MyD88 contains a typical death domain at the amino terminus and a conserved Toll/IL-1R (TIR) domain at the carboxyl terminus, as well as three highly conserved motifs (Box1, Box2 and Box3) within the C-terminal TIR domain. In healthy fish, Og-MyD88 was found to be strongly expressed in immune-related tissues, including the spleen, head kidney, kidney, liver, skin and intestine, with lower expression in heart, stomach, brain and muscle. Transcripts of Og-MyD88 were found to be markedly up-regulated in fish spleen after challenge with Singapore grouper iridovirus (SGIV), a highly lethal viral pathogen to grouper fish. Furthermore, the full length Og-MyD88 and its N-terminal death domain were capable of inducing NF-κB activity in HEK-293 cells. Overexpressed Og-MyD88 showed the ability to inhibit replication of SGIV in grouper spleen (GS) cells. These results suggest that Og-MyD88 is involved in the grouper immune response to invasion of viral pathogens and may share similar functions to those observed in higher vertebrates.
