Abstract:Chitosan was modified with thioctic acid and used to prepare soft films. As confirmed by FTIR and XPS measurements, a condensation reaction occurred between the amino groups in the chitosan and the carboxyl groups in the lipoic acid to form amide bonds in the modified chitosan. Films were then prepared by casting at ambient conditions, and the effects of the chemical modification on the physical-mechanical, antibacterial, and thermal properties of the films were investigated. The results showed that the tensile strength, flexibility and recovery performance of the modified films were significantly different from those of the unmodified films. For example, the Young's modulus of a pure chitosan film was 2600 MPa, while the modified films were much more flexible with a Young's modulus as low as 32.5 MPa. Moreover, the modified chitosan films were not dissolved or damaged by common organic solvents or in highly acidic (pH 1) or highly basic (pH 13) conditions. The modified films also showed good antibacterial activity against both E coli and S aureus with inhibition rates of almost 100 %. These desirable properties suggest that the modified chitosan films prepared here have possible application prospects in flexible devices and packaging.

Abstract:Disulfide-crosslinked hollow polyelectrolyte microcapsules composed of thiolated chitosan (CS-SH) and hyaluronic acid (HA-SH) were prepared by combining the layer-by-layer (LBL) technique and horseradish peroxidase (HRP)-mediated oxidative cross-linking reaction in mild conditions. FITC-dextran-doped CaCO3 microspheres were used as template core and removed after LBL depositing CS-SH and HA-SH on the surface. The disulfide-crosslinked (CS/HA) microcapsules were readily fabricated by HRP-mediated oxidative coupling of the thiol groups in CS/HA shell layer in the presence of HRP (10 units/mL) and Tyramine hydrochloride (Tyr, 35 mmol/L). The kinetics of enzymatic disulfide-crosslinking reaction was investigated through the real-time monitoring of the consumption of thiol groups by UV absorption spectra. It found that the formation of disulfide linkages by the enzymatic thiol oxidation reaction showed a gradual acceleration. The disulfide-crosslinked CS/HA hydrogel were rapidly formed in gelation time between approximately 17 and 30 min, which were dependent on the concentrations of HRP and Tyr. The disulfide linkages endowed the microcapsule-enhanced physical stability and low permeability under physiological conditions and redox-responsive degradability in reducing environments. The structural stability of disulfide-crosslinked (CS/HA) microcapsules was visualized by confocal laser scanning microscopy in phosphate-buffered saline containing 5.0 mmol/L dithiothreitol (DTT) to evaluate the redox-responsive disassembly process. Redox-responsive controlled release of encapsulated FITC-dextran from the disulfide-crosslinked (CS/HA) microcapsules were obtained. The release profiles of FITC-dextran could be manipulated by controlling the shell thickness and the concentration of DTT. The conformational stability analyses and more than 94% esterase activity of released bovine serum albumin (BSA) from (CS/HA) microcapsules conformed that the structural integrity and bioactivity were well preserved during the encapsulation and release process. The microcapsules exhibited excellent cytocompatibility for HEK 293 cells up to a concentration of 1.0 mg/mL. The microcapsules efficiently delivered loaded FITC-BSA into HeLa cells and released the protein in the reducing cytosol. This study proposed a novel approach for producing disulfide-crosslinked microcarriers for intracellular delivery and redox-responsive controlled release of protein.

Abstract:A chitosan-based nanocomposite film with tannic acid (TA) as a cross-linker and titanium dioxide nanoparticles (TiO2) as a reinforcing agent was developed with a solution casting technique. TA and TiO2 are biocompatible with chitosan, and this paper studied the synergistic effect of the cross-linker and the reinforcing agent. The addition of TA enhanced the ultraviolet blocking and mechanical properties of the chitosan-based nanocomposite film. The reinforcement of TiO2 in chitosan/TA further improved the nanocomposite film’s mechanical properties compared to the neat chitosan or chitosan/TA film. The thermal stability of the chitosan-based nanocomposite film was slightly enhanced, whereas the swelling ratio decreased. Interestingly, its water vapor barrier property was also significantly increased. The developed chitosan-based nanocomposite film showed potent antioxidant activity, and it is promising for active food packaging.

Abstract:Chitosan-based materials have attracted considerable attention owing to their inherent biodegradability, biocompatibility, and antimicrobial properties. However, the establishment of a scalable technology for the fabrication of high-strength and high-toughness chitosan films remains a major challenge. Herein, a new route for the fabrication of high-strength and high-toughness chitosan films mediated by a unique hydrated chitosan crystal structure is reported, using an aqueous KOH/urea solution as a solvent and an aqueous KCl solution as a neutralization bath. The KCl concentration, neutralization temperature, and neutralization time significantly affect the self-assembly and lateral aggregation of chitosan chains and the microstructure and morphology of the chitosan hydrogels and films. Furthermore, stretching orientation endows the chitosan films with a high tensile strength, Young’s modulus, and work of fracture of 579 ± 52 MPa, 18.8 ± 1.5 GPa, and 26.7 ± 6.9 MJ m−3, respectively. To the best of our knowledge, the chitosan films developed in this study are the first example of extremely high-strength and high-toughness chitosan films. We believe that our findings will be useful for the fabrication of polysaccharide-based films with excellent mechanical properties and in broader applications in the fields of biomedicine, flexible bioelectronics, water treatment, and food packaging.

Abstract:Chitosan materials perform a great potential for applications in sustainable and flexible electronics, owing to their abundance, biocompatibility, and biodegradability. While limited by the inherent shortcoming of having a melting temperature higher than its degradation temperature, chitosan is hard to be manufactured by traditional thermoforming or solvent-free methods. Herein, we demonstrated a tractable roll-forming method to process chitosan films under the plasticizing effect of ionic liquids. In particular, the additional ionic liquids could be removed by Soxhlet extraction and then recycled toward sustainability. The final regenerated chitosan films exhibited outstanding strength (53.1 MPa), high elongation (3.5%) and hardness (0.4 GPa). Simultaneously, the successful fabrication of interdigital electrode sensors on the chitosan film indicated the feasibility of manufacturing flexible electronics using chitosan substrates. Thus, our innovative strategy enables the sustainable formation of high-performance chitosan films, offering a broader prospect for the new generation of chitosan-based biodegradable electronics.

Abstract:Crystallization of phytochromes and other photochromic proteins is hampered by the conformational changes that they undergo on exposure to light. As a canonical phytochrome, cyanobacterial Cph1 switches between two stable states upon absorption of red/far-red light. Consequently, it is mandatory to work in darkness from protein purification to crystal cryoprotection in order to ensure complete occupancy of one state or the other. With the simple and inexpensive methods that have been developed, phytochromes and other photochromic molecules can effectively be handled and crystallized, as has been demonstrated by the solution of the three-dimensional structure of the Cph1 sensory module.
Physical, Mechanical and Degradation Properties, and Schwann Cell Affinity of Cross-Linked Chitosan Films.

Abstract:Three kinds of cross-linked chitosan films were prepared with hexamethylene diisocyanate (HDI), epichlorohydrin (ECH) and glutaraldehyde (GA) as cross-linking agents, respectively. The physical and mechanical properties, biodegradability and Schwann cell affinity of the cross-linked films were investigated. A significant decrease in the degradation rate in lysozyme solution and a large change in the mechanical properties were observed compared with non-cross-linked chitosan films. The protein adsorption on chitosan films was determined by means of enzyme-linked immunosorbent assay (ELISA). In comparison with the non-cross-linked films, the chitosan films cross-linked with HDI showed a significant increase (up to 40-50%) in both fibronectin and laminin adsorption, while the protein adsorption on the other two kinds of cross-linked films was similar to that on non-crosslinked films. In addition, cell culture revealed that the HDI cross-linked chitosan films enhanced the spread and proliferation of Schwann cells while the other cross-linked films delayed the cell proliferation. These results suggest that HDI cross-linking of chitosan films provides a combination of physical properties, biodegradability and Schwann cell affinity suitable for peripheral nerve regeneration.

Abstract:Natural polymer-based films, due to their favorable biological and mechanical properties, have demonstrated great potential as coatings for biomedical applications. Among them, chitosan films have been widely studied both as coating materials and as controlled drug release systems. Crosslinkers are often used to tune chitosan’s crosslinking degree and thus to control the drug release kinetics. For this purpose, quercetin, a plant-derived natural polyphenol, has gained attention as a crosslinker, mainly for its intrinsic anti-inflammatory, antioxidant, and antibacterial features. In this study, chitosan films crosslinked with three different concentrations of quercetin (10, 20, and 30% w/w) have been used as controlled release systems for the delivery of the antibacterial drug trimethoprim (TMP, 10% w/w). Physicochemical and antimicrobial properties were investigated. Surface wettability and composition of the films were assessed by contact angle measurements, X-ray photoelectron spectroscopy (XPS), and Fourier-transform infrared spectroscopy (FTIR), respectively. The release kinetic of TMP in phosphate-buffered saline (PBS) and 2-(N-morpholino) ethanesulfonic acid (MES) was studied over time. Finally, antibacterial properties were assessed on E. coli and S. aureus through Kirby–Bauer disc diffusion and micro-dilution broth assays. Results show that quercetin, at the tested concentrations, clearly increases the crosslinking degree in a dose-dependent manner, thus influencing the release kinetic of the loaded TMP while maintaining its bactericidal effects. In conclusion, this work demonstrates that quercetin-crosslinked chitosan films represent a promising strategy for the design of antibiotic-releasing coatings for biomedical applications.

Abstract:The aim of this work to find out the influence of salicylic acid-grafted-chitosan (SA-g-CS) and salicylic acid incorporated chitosan on the properties of chitosan film. The SA-g-CS conjugated films (SA-g-CS Ⅰ, SA-g-CS Ⅱ, and SA-g-CS Ⅲ) and the SA-CS incorporated films (SA-CS Ⅰ, SA-CS Ⅱ, and SA-CS Ⅲ) with equivalent salicylic acid content were prepared, respectively. The physical, mechanical, and biological properties of SA-g-CS conjugated and SA-CS incorporated films were evaluated. The opacity and water solubility were enhanced, thickness and water vapor permeability changed, and moisture content, tensile strength and elongation at break of chitosan film decreased or increased after conjugation or incorporation of SA. Compared with CS film, SA-g-CS conjugated and SA-CS incorporated films showed enhanced antioxidant and antibacterial activities, and these activities increased with the increasing of SA content. SA-g-CS conjugated films possessed better water vapor barrier property, antioxidant and antibacterial properties than SA-CS incorporated films. Therefore, SA-g-CS conjugated films exhibited better functional properties than SA-CS incorporated films. Compared with incorporation, conjugation of SA with CS is a more efficient way to improve properties of CS film.

Abstract:In this study, the active food packaging film were prepared using hydroxypropyltrimethyl ammonium chloride chitosan with different substitution sites (O-HACC & N-HACC) and dialdehyde chitosan (DCS) grafted with protocatechuic acid (PA). To explore the effect of chitosan quaternization positions and crosslinking approaches on the slow-release and antibacterial properties, the double-crosslinked film were fabricated through the self-coupling reaction of PA and Schiff base reaction between amino groups on HACC and aldehyde groups on DCS. The HACC/DCS-based film exhibited stable porous three-dimensional networks with high nisin loading ratios (>90 %). With the participation of the catechol-catechol structure, the dense double-crosslinked film effectively restricted the diffusion of the water molecules, resulting in excellent slow-release properties fitting with the Korsmeyer-Peppas kinetic model. Especially, O-HACC/PA-g-DCS film, which had more reaction sites for Schiff base crosslinking than N-HACC, exhibited the equilibrium swelling ratio of 800 % at 60 h and could sustainably release nisin via non-Fickian diffusion behavior until 48 h. Moreover, the HACC/DCS-based double-crosslinked film performed good long-time antibacterial activity and preservation effects on salmon. On the 10th day of storage, the TVBN of N-HACC/PA-g-DCS and O-HACC/PA-g-DCS groups were only 28.26 ± 1.93 and 29.06 ± 1.68 mg/100 g and still lower than the thresholds.

Abstract:Chitosan hydrogels fabricated by covalent crosslinking exhibit tough mechanical properties and chemical stability. In this paper, debranched starch (DBS) is oxidized to dialdehyde debranched starch (DADBS), which is used as a new type of a crosslinking agent to prepare hydrogels. Chitosan hydrogels with excellent properties are prepared by dynamic Schiff-base crosslinking between the aldehyde groups in DADBS and the amino groups in chitosan. Hence, chitosan hydrogels exhibit a rapid gelation ability, with a gelation time of less than 30 s, and their storage modulus increases with the gelation time. By adjusting the molar ratio of the amino group of chitosan to the aldehyde group of DADBS and the reaction temperature, the hydrogels exhibit tunable elasticity and mechanical properties. Notably, scanning electron microscopy revealed the presence of 100–200 nm microgels in the hydrogel network, which could exert a strengthening effect on the mechanical properties of the hydrogels. In addition, chitosan hydrogels exhibit a rapid self-healing ability and remarkable fluorescence properties; also, they can be 3D printed in different shapes. Overall, the DADBS cross-linked chitosan hydrogels demonstrate potential applications in food, medicine, agriculture, and materials.

Abstract:Active films were developed based on chitosan, esterified chitin nanofibers and rose essential oil (REO). The joint effects of chitin nanofibers and REO on structure and physicochemical properties of chitosan film were investigated. Scanning electron microscopy and Fourier transform infrared spectroscopy showed that the chitin nanofibers and REO had significant effects on the morphology and chemical structure of chitosan composite films. The negatively charged esterified chitin nanofibers formed a compact network structure through intermolecular hydrogen bonding and electrostatic interactions with the positively charged chitosan matrix. Chitin nanofibers and REO synergistically enhanced the water resistance, mechanical properties and UV resistance of chitosan-based films, but the addition of REO increased the oxygen permeability. Furthermore, the addition of REO enhanced the inhibition of ABTS and DPPH free radicals and microorganisms by chitosan-based film. Therefore, chitosan/chitin nanofiber-based active films containing REO as food packaging materials can potentially provide protection to extend food shelf life.

Abstract:The aim of the current study is to develop chitosan-based biomaterials which can sustainably release acetylsalicylic acid while presenting significant biological activity. Herein, an innovative ionic bonding strategy between hydroxypropyl trimethyl ammonium chloride chitosan (HACC) and acetylsalicylic acid (AA) was proposed, skillfully utilizing the electrostatic attraction of the ionic bond to achieve the controlled release of drugs. Based on this point, six crosslinked N-[(2-hydroxy-3-trimethylammonium)propyl]chitosan acetylsalicylic acid salt (CHACAA) hydrogel films with varying acetylsalicylic acid contents were prepared by a crosslinking reaction. The results of 1H nuclear magnetic resonance spectroscopy (1H NMR) and scanning electron morphology (SEM) confirmed the crosslinked structure, while the obtained hydrogel films possessed favorable thermal stability, mechanical properties, and swelling ability. In addition, the drug release behavior of the hydrogel films was also investigated. As expected, the prepared hydrogel films demonstrated the capability for the sustainable release of acetylsalicylic acid due to ion pair attraction dynamics. Furthermore, the bioactivities of CHACAA-3 and CHACAA-4 hydrogel films with acetylsalicylic acid molar equivalents of 1.25 and 1.5 times those of HACC were particularly pronounced, which not only exhibited an excellent drug sustained-release ability and antibacterial effect, but also had a higher potential for binding and scavenging inflammatory factors, including NO and TNF-α. These findings suggest that CHACAA-3 and CHACAA-4 hydrogel films hold great potential for applications in wound dressing, tissue engineering scaffolds, and drug carriers.

Abstract:Chitosan (CS)/cellulose (BC) blend films were successfully prepared using ZnCl2•3H2O as a solvent. Homogeneous structures without obvious phase separation between CS and BC for all blend films were observed by scanning electron microscope (SEM) analysis. The tensile strengths of CS/BC blend films decreased with the increase of chitosan content. The results of X-ray diffraction (XRD) analysis indicated that the crystal structures of BC and CS were disrupted during the processes of dissolving and regeneration. Also, the reformation of hydrogen bonds between CS and BC during dissolution and regeneration processes resulted in the shift of diffraction peaks. Fourier transforms infrared spectroscopy (FT-IR) and thermogravimetric analysis (TGA) analysis results confirmed this observation. Moreover, obvious antimicrobial capability of CS/BC blend films against E. coli has been observed, indicating that antibacterial activity of chitosan has not been significantly inactivated while using ZnCl2•3H2O as a solvent. Therefore, ZnCl2•3H2O can be regarded as a promising solvent to prepare degradable films with antibacterial properties.
Preparation and Properties of Self-Cross-Linking Hydrogels Based on Chitosan Derivatives and Oxidized Sodium Alginate

Abstract: A self-cross-linking and biocompatible hydrogel has wideapplicationpotential in the field of tissue engineering. In this work, an easilyavailable, biodegradable, and resilient hydrogel was prepared usinga self-cross-linking method. This hydrogel was composed of N-2-hydroxypropyl trimethyl ammonium chloride chitosan (HACC)and oxidized sodium alginate (OSA). A stable and reversible cross-linkingnetwork was formed by the Schiff base self-cross-linked and hydrogenbonding. The addition of a shielding agent (NaCl) may weaken the intenseelectrostatic effect between HACC and OSA and solve the problem offlocculation caused by the rapid formation of ionic bonds, which providedan extended time for the Schiff base self-cross-linked reaction forforming a homogeneous hydrogel. Interestingly, the shortest time forthe formation of the HACC/OSA hydrogel was within 74 s and the hydrogelhad a uniform porous structure and enhanced mechanical properties.The HACC/OSA hydrogel withstood large compression deformation dueto improved elasticity. What’s more, this hydrogel possessedfavorable swelling property, biodegradation, and water retention.The HACC/OSA hydrogels have great antibacterial properties against Staphylococcus aureus and Escherichiacoli and demonstrated good cytocompatibility as well.The HACC/OSA hydrogels have a good sustained release effect on rhodamine(model drug). Thus, the obtained self-cross-linked HACC/OSA hydrogelsin this study have potential applications in the field of biomedicalcarriers.

Abstract:Highly substituted perfluoroacylated chitosan can alter the physicochemical properties of chitosan; however, the currently synthesized perfluoroacylated chitosan has a low degree of substitution. In this study, we present a simple method for the homogeneous preparation of highly substituted N-perfluoroacylated chitosan, conducted at room temperature without requiring strict anhydrous or oxygen-free conditions. Various perfluorocarbon chains were successfully attached to chitosan through a reaction between perfluorinated acid esters and amines, catalyzed by DBU. The synthesized N-perfluoroacylated chitosan, with high degree of substitution, demonstrated excellent solubility in common organic solvents. Comprehensive characterization was performed using elemental analysis, nuclear magnetic resonance (including two-dimensional NMR), gel permeation chromatography, infrared spectroscopy, X-ray diffraction, and thermal analysis. The resulting films exhibited high water contact angles. Notably, as the fluorocarbon chain length increased, tensile strength gradually decreased, while elongation at break improved. Additionally, water uptake, water vapor transmission rate, and oxygen transmission rate all exhibited a declining trend. The films exhibited good biocompatibility, and in the grape preservation experiment, HFBC treatment effectively delayed grape aging and deterioration while enhancing quality preservation. These results suggest that HFBC film holds promising potential for food packaging applications.
An Ultra‐Strong, Water Stable and Antimicrobial Chitosan Film with Interdigitated Bouligand Structure

Abstract:Natural polymeric materials are an attractive potential replacement for non-biodegradable plastics. However, one of the main challenges for high-end applications of these materials, is to maintain their high mechanical properties in a watery environment. Here, inspired by the reinforcement principle of multiscale architecture in natural crab exoskeletons, an ultra-strong and hydrostable film is developed through a top-down method directly from the crab shell. The resulting chitosan film exhibits an outstanding isotropic tensile strength of approximate to 220 MPa and a superior wet strength of approximate to 70 MPa, which is significantly higher than that of conventional polysaccharide-based materials and many commercially used plastics. Its excellent mechanical properties are due to the realization of strong interactions between chitosan fibrils even in wet conditions through interdigitation and densification processes without other additives. Additionally, it also possesses outstanding transparency, flexibility, antimicrobial ability, and biodegradability, making it a promising high-performance and eco-friendly material for many potential applications.
Formation chitosan-based hydrogel film containing silicon for hops β-acids release as potential food packaging material

Abstract:Hydrogel film composed of chitosan (CS) as raw material was prepared by free radical polymerization. Silicon was introduced into the hydrogel film in different ways (covalent/non-covalent) to improve the physical properties of the film, and β-acids were loaded to enhance the antibacterial activity of the film. Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), and X-ray diffraction (XRD) analysis were used to characterize the structure of films. The mechanical results indicated that when nano-silica (0.3%) was introduced into film (containing 0.2% β-acids) by non-covalently bond, the tensile strength increased to 8.59 MPa. Meanwhile silicon (0.3%) entered the film by covalent bonding, the tensile strength increased to 7.99 MPa. The films loaded with β-acids had well ability to blocks ultraviolet rays and exhibited inhibitory effect on E. coli and S. aureus. In the PBS (37 °C, pH = 7.4) simulant solution, the release mechanism of most films to release the β-acids followed non-Fick diffusion (n > 0.5). It could be concluded that the prepared hydrogel films loading with β-acids had broad application prospects in food packaging material with antibacterial property and controlled release.

Abstract:We prepared silk fibroin (SF)-mediated porous chitosan films using the ionic gelation approach. Citric acid was employed as an eco-friendly crosslinker and poly(ethylene glycol) (PEG) as a porogen. The prepared films were characterized for various physicochemical, mechanical and antibacterial properties, and subsequently loaded with gentamicin sulfate as a model drug. The water contact angle indicated that the pristine chitosan film was hydrophobic but became hydrophilic after the release of PEG. This could facilitate the uptake of the hydrophilic gentamicin sulfate. The infrared spectroscopy expressed strong intermolecular interactions across the amino groups of silk protein and the hydroxyl groups of the chitosan matrices. The XRD indicated molecular interaction between SF and chitosan caused a crystalline trend in the blend film. The thermal analysis revealed that the prepared films were stable up to 330oC. The silk reinforcement significantly improved the tensile strength (from 25.53 ± 2.5–45.24 ± 4.5 MPa) and elongation at break (from 22.24 ± 2.20–36.12 ± 3.60%) with an appropriate swelling ratio. The prepared SF-mediated porous chitosan films exhibited broad-spectrum antibacterial activity, appropriate biocompatibility and desirable polar interaction with the gentamicin sulfate for controlled drug release applications.
