Abstract:One of the key transitions in early cognitive development is from participating in face-to-face interactions to engaging in joint attention exchanges. It is known that the ability to jointly attend with another person to an object is essential for the development of abilities such as language in later life. Strikingly, little is known about the function of joint attention in infants in the first year. We developed a novel interactive-live paradigm to assess the neural mechanisms of joint attention in 9-month-old infants. An adult interacted with each infant, and infants' electrical brain activity was measured in two contexts. In the joint attention context, a live adult gazed at the infants' face and then to a computer displayed novel object. In the non-joint attention context the adult gazed only to the novel object. We found that the negative component of the infant event-related potential (ERP), a neural correlate indexing attentional processes, was enhanced in amplitude during the processing of objects when infants were engaged in a joint attention interaction compared to a non-joint attention interaction. These results suggest that infants benefit from joint attention interactions by focusing their limited attentional resources to specific aspects of the surrounding environment.

The real-time effects of parent speech on infants' multimodal attention and dyadic coordination

Abstract:Parental responsiveness to infant behaviors is a strong predictor of infants' language and cognitive outcomes. The mechanisms underlying this effect, however, are relatively unknown. We examined the effects of parent speech on infants' visual attention, manual actions, hand-eye coordination, and dyadic joint attention during parent-infant free play. We report on two studies that used head-mounted eye trackers in increasingly naturalistic laboratory environments. In Study 1, 12-to-24-month-old infants and their parents played on the floor of a seminaturalistic environment with 24 toys. In Study 2, a different sample of dyads played in a home-like laboratory with 10 toys and no restrictions on their movement. In both studies, we present evidence that responsive parent speech extends the duration of infants' multimodal attention. This social "boost" of parent speech impacts multiple behaviors that have been linked to later outcomes-visual attention, manual actions, hand-eye coordination, and joint attention. Further, the amount that parents talked during the interaction was negatively related to the effects of parent speech on infant attention. Together, these results provide evidence of a trade-off between quantity of speech and its effects, suggesting multiple pathways through which parents impact infants' multimodal attention to shape the moment-by-moment dynamics of an interaction.

Abstract:Abstract Initiating joint attention (IJA), the behavioral instigation of coordinated focus of 2 people on an object, emerges over the first 2 years of life and supports social-communicative functioning related to the healthy development of aspects of language, empathy, and theory of mind. Deficits in IJA provide strong early indicators for autism spectrum disorder, and therapies targeting joint attention have shown tremendous promise. However, the brain systems underlying IJA in early childhood are poorly understood, due in part to significant methodological challenges in imaging localized brain function that supports social behaviors during the first 2 years of life. Herein, we show that the functional organization of the brain is intimately related to the emergence of IJA using functional connectivity magnetic resonance imaging and dimensional behavioral assessments in a large semilongitudinal cohort of infants and toddlers. In particular, though functional connections spanning the brain are involved in IJA, the strongest brain-behavior associations cluster within connections between a small subset of functional brain networks; namely between the visual network and dorsal attention network and between the visual network and posterior cingulate aspects of the default mode network. These observations mark the earliest known description of how functional brain systems underlie a burgeoning fundamental social behavior, may help improve the design of targeted therapies for neurodevelopmental disorders, and, more generally, elucidate physiological mechanisms essential to healthy social behavior development.

Abstract:Parents play a significant role in directing infant’s attention to environmental stimuli via joint attention. We hypothesized that infants whose parents provide more bids for joint attention will display a more complex neural response when viewing social scenes. Sixty-one 8-month-old infants underwent electroencephalography (EEG) while viewing videos of joint-and parallel-play and participated in a parent-infant free play interaction. EEG data was analyzed using multiscale entropy, which quantifies moment-to-moment neural variability. Free play interactions were coded for parent alternating gaze, a behavioral mechanism for directing attention to environmental cues. We found a significant positive association between parent alternating gaze and neural entropy in frontal and central brain regions. These results suggest a relationship between parent behavior and infant neural mechanisms that regulate social attention, underlying the importance of parent cues in the formation of neural networks in infancy.

Proactive or reactive? Neural oscillatory insight into the leader-follower dynamics of early infant-caregiver interaction

Abstract:We know that infants' ability to coordinate attention with others toward the end of the first year is fundamental to language acquisition and social cognition. Yet, we understand little about the neural and cognitive mechanisms driving infant attention in shared interaction: do infants play a proactive role in creating episodes of joint attention? Recording electroencephalography (EEG) from 12-mo-old infants while they engaged in table-top play with their caregiver, we examined the communicative behaviors and neural activity preceding and following infant- vs. adult-led joint attention. Infant-led episodes of joint attention appeared largely reactive: they were not associated with increased theta power, a neural marker of endogenously driven attention, and infants did not increase their ostensive signals before the initiation. Infants were, however, sensitive to whether their initiations were responded to. When caregivers joined their attentional focus, infants showed increased alpha suppression, a pattern of neural activity associated with predictive processing. Our results suggest that at 10 to 12 mo, infants are not routinely proactive in creating joint attention episodes yet. They do, however, anticipate behavioral contingency, a potentially foundational mechanism for the emergence of intentional communication.

Abstract:Among 11 subtypes of heterotrimeric G-protein gamma-subunit, gamma1 (rod), gamma8 (cone) and gamma11 are modified with farnesyl while the others are modified with geranylgeranyl at the C-terminus. To understand the role of specific isoprenylation (farnesylation) of retinal transducin, we examined how and to what extent the type of isoprenyl group affects transducin-beta gamma (beta1 gamma1) functions such as interactions with membranes, Galpha/receptor, and effectors. To this end, the C-terminal farnesylation signal sequence (CVIS) of gamma1 was replaced by a geranylgeranylation signal (CVIL), and the resultant mutant (S74L) or wild-type (WT) gamma1 was coexpressed with beta1 in the baculovirus-Tn5 insect cell system. Both gamma1WT and gamma1S74L expressed as a beta gamma complex were mixtures modified with farnesyl and geranylgeranyl groups. The ratio of farnesyl to geranylgeranyl in preparations of beta1 gamma1WT and beta1 gamma1S74L purified from the Tn5 cell membrane fraction was about 1:2 and 1:6, respectively. These two forms of recombinant beta1 gamma1 and retinal beta1 gamma1 were different in their abilities to associate with rod outer segment membranes with the following rank order: beta1 gamma1S74L > beta1 gamma1WT > retinal beta1 gamma1. Functionally, beta1 gamma1S74L was the most potent to promote pertussis toxin-catalyzed ADP ribosylation of transducin-alpha (Talpha), to stimulate metarhodopsin II-catalyzed GTPgammaS-binding reaction to Talpha and to modulate adenylyl cyclase and phospholipase C activities. All of the beta1 gamma1 functions absolutely required the isoprenylation of the gamma-subunit. As for the interaction with Goalpha and adenylyl cyclase, predominantly geranylgeranylated beta1 gamma1S74L was less effective than geranylgeranylated beta1 gamma2 purified from bovine brain. These results demonstrate that the properties of Gbeta gamma are strongly affected by the type of functionally indispensable isoprenylation in addition to the amino acid sequence of Ggamma. The relative contribution of the two factors depends on proteins with which Gbeta gamma interacts.

Abstract:In a complex social environment, stimuli from different sensory modalities need to be integrated to decode communicative meanings. From very early in life, infants have to combine a multitude of sensory features with social and affective attributes. Of all senses, touch constitutes a privileged channel to carry affective‐motivational meanings and foster social connection. In the present study, we investigate whether sharing sensory stimulation that varies for its affective value differentially affects infants' attention towards visual stimuli. 6 to 11‐month‐old infants (N = 42) were familiarized with two characters respectively matched with tactile (affective or non‐affective) and auditory stimulation; then repeatedly exposed to scenes where the two characters moved towards target objects. Our results showed a main effect of stimulation (sound vs. touch) on looking times during familiarization, with longer looking times when sound is provided. During scenes presentation, a main effect of the type of touch (affective vs. non affective) emerged, with longer looking times in infants that previously experienced affective touch, suggesting that this sensory experience may critically engage the self and modulate infant attention. Overall, these findings suggest that while sound acts as attention getter, affective touch supports sustained attention towards complex visual scenes beyond the stimulation period itself.

Abstract:Joint attention, the shared attentional focus of at least two people on a third significant object, is one of the earliest steps in social development and an essential aspect of reciprocal interaction. However, the neural basis of joint attention (JA) in the course of development is completely unknown. The present study made use of an interactive eye-tracking paradigm in order to examine the developmental trajectories of JA and the influence of a familiar interaction partner during the social encounter. Our results show that across children and adolescents JA elicits a similar network of "social brain" areas as well as attention and motor control associated areas as in adults. While other-initiated JA particularly recruited visual, attention and social processing areas, self-initiated JA specifically activated areas related to social cognition, decision-making, emotions and motivational/reward processes highlighting the rewarding character of self-initiated JA. Activation was further enhanced during self-initiated JA with a familiar interaction partner. With respect to developmental effects, activation of the precuneus declined from childhood to adolescence and additionally shifted from a general involvement in JA towards a more specific involvement for self-initiated JA. Similarly, the temporoparietal junction (TPJ) was broadly involved in JA in children and more specialized for self-initiated JA in adolescents. Taken together, this study provides first-time data on the developmental trajectories of JA and the effect of a familiar interaction partner incorporating the interactive character of JA, its reciprocity and motivational aspects.

Association between caregiver and infant visual neurocognition

Abstract:Previous work has shown that caregiver attention shapes visual cognition in infants through dyadic interactions. Is this association measurable when visual cognition is objectively measured in caregivers and infants using comparable experimental paradigms? In the current study, we presented infants (N = 86) and caregivers (N = 78) with age-specific variants of the same preferential looking visual cognition task to investigate whether caregiver visual cognition was associated with their infants' visual cognition. In each trial of the task, two side-by-side flashing displays of coloured shapes were presented. On the 'unchanging' side, the colours of the shapes remained the same. On the 'changing' side, the colour of one shape changed after each flash. Load was varied by changing the number of shapes across trials (low, medium, and high loads). We extracted looking dynamics using video recordings and brain function using functional near-infrared spectroscopy as both infants and caregivers engaged with the task. Change preference (CP) score, which represented the amount of time spent looking at the changing side divided by the total looking duration, showed a load-dependent modulation for both infants and caregivers. Both groups showed the highest CP scores at the low load. Further, higher caregiver CP scores was associated with higher infant CP scores at the low load. Both infants and caregivers engaged canonical regions of the fronto-parietal network involved in visual cognition. Critically, higher caregiver CP scores were associated with greater activation in the left superior parietal lobule in younger infants, a region involved in allocating visuo-spatial attention and working memory maintenance. Further, there was spatial overlap between performance-dependent regions in the right parietal cortex in caregivers and younger infants. Our findings provide first evidence of a heritability-related visual neurocognitive association between caregivers and their children in the first year of life.

Abstract:The emergence of joint attention is still a matter of vigorous debate. It involves diverse hypotheses ranging from innate modules dedicated to intention reading to more neuro-constructivist approaches. The aim of this study was to assess whether 12-month-old infants are able to recognize a "joint attention" situation when observing such a social interaction. Using a violation-of-expectation paradigm, we habituated infants to a "joint attention" video and then compared their looking time durations between "divergent attention" videos and "joint attention" ones using a 2 (familiar or novel perceptual component)×2 (familiar or novel conceptual component) factorial design. These results were enriched with measures of pupil dilation, which are considered to be reliable measures of cognitive load. Infants looked longer at test events that involved novel speaker and divergent attention but no changes in infants' pupil dilation were observed in any conditions. Although looking time data suggest that infants may appreciate discrepancies from expectations related to joint attention behavior, in the absence of clear evidence from pupillometry, the results show no demonstration of understanding of joint attention, even at a tacit level. Our results suggest that infants may be sensitive to relevant perceptual variables in joint attention situations, which would help scaffold social cognitive development. This study supports a gradual, learning interpretation of how infants come to recognize, understand, and participate in joint attention.

Abstract:Eye-to-eye contact is a spontaneous behavior between interacting partners that occurs naturally during social interactions. However, individuals differ with respect to eye gaze behaviors such as frequency of eye-to-eye contacts, and these variations may reflect underlying differences in social behavior in the population. While the use of eye signaling to indicate a shared object of attention in joint attention tasks has been well-studied, the effects of the natural variation in establishing eye contact during joint attention have not been isolated. Here, we investigate this question using a novel two-person joint attention task. Participants were not instructed regarding the use of eye contacts; thus all mutual eye contact events between interacting partners that occurred during the joint attention task were spontaneous and varied with respect to frequency. We predicted that joint attention systems would be modulated by differences in the social behavior across participant pairs, which could be measured by the frequency of eye contact behavior. We used functional near-infrared spectroscopy (fNIRS) hyperscanning and eye-tracking to measure the neural signals associated with joint attention in interacting dyads and to record the number of eye contact events between them. Participants engaged in a social joint attention task in which real partners used eye gaze to direct each other's attention to specific targets. Findings were compared to a non-social joint attention task in which an LED cue directed both partners' attention to the same target. The social joint attention condition showed greater activity in right temporoparietal junction than the non-social condition, replicating prior joint attention results. Eye-contact frequency modulated the joint attention activity, revealing bilateral activity in social and high level visual areas associated with partners who made more eye contact. Additionally, when the number of mutual eye contact events was used to classify each pair as either "high eye contact" or "low eye contact" dyads, cross-brain coherence analysis revealed greater coherence between high eye contact dyads than low eye contact dyads in these same areas. Together, findings suggest that variation in social behavior as measured by eye contact modulates activity in a subunit of the network associated with joint attention.

Abstract:In addition to an assimilatory sulfite reductase, studies of cultures of Clostridium pasteurianum supplemented with methionine, cysteine, and 35SO42- provides evidence for another reductase which is induced by SO32-. This inducible reductase appears to be dissimaltory because of the copious sulfide production arising when the cells are grown on SO32-. Cysteine can repress the assimilatory sulfite reductase but does not affect the inducible reductase. During late logarithmic growth on 1 mM SO42- + 10mM cysteine, depression of the inducible reductase occurred along with increased sulfide production. The presence of 1 mM cysteine and (or) 1 mM cysteine and (or) 1 mM methionine does not affect the inverse sulfur isotope effect for evolved H2S. However, 5 and 10 mM cysteine reduce the maximum delta34S value for released H2S from +40 to 10%. A small conversion of cysteine to H2S by C. pasteurianum occurs, but only in the stationary phase.
Real-time monitoring of infant theta power during naturalistic social experiences

Abstract:Infant-directed speech and direct gaze are important social cues that shape infant's attention to their parents. Traditional methods for probing their effect on infant attention involve a small number of pre-selected screen-based stimuli, which do not capture the complexity of real-world interactions. Here, we used neuroadaptive Bayesian Optimization (NBO) to search a large 'space' of different naturalistic social experiences that systematically varied in their visual (gaze direct to averted) and auditory properties (infant directed speech to nonvocal sounds). We measured oscillatory brain responses (relative theta power) during episodes of naturalistic social experiences in 57 typically developing 6- to 12-month-old infants. Relative theta power was used as input to the NBO algorithm to identify the naturalistic social context that maximally elicited attention in each individual infant. Results showed that individual infants were heterogeneous in the stimulus that elicited maximal theta with no overall stronger attention for direct gaze or infant-directed speech; however, individual differences in attention towards averted gaze were related to interpersonal skills and greater likelihood of preferring speech and direct gaze was observed in infants whose parents showed more positive affect. Our work indicates NBO may be a fruitful method for probing the role of distinct social cues in eliciting attention in naturalistic social contexts at the individual level.
Infant attention to rhythmic audiovisual synchrony is modulated by stimulus properties

Abstract:Musical interactions are a common and multimodal part of an infant's daily experiences. Infants hear their parents sing while watching their lips move and see their older siblings dance along to music playing over the radio. Here, we explore whether 8- to 12-month-old infants associate musical rhythms they hear with synchronous visual displays by tracking their dynamic visual attention to matched and mismatched displays. Visual attention was measured using eye-tracking while they attended to a screen displaying two videos of a finger tapping at different speeds. These videos were presented side by side while infants listened to an auditory rhythm (high or low pitch) synchronized with one of the two videos. Infants attended more to the low-pitch trials than to the high-pitch trials but did not display a preference for attending to the synchronous hand over the asynchronous hand within trials. Exploratory evidence, however, suggests that tempo, pitch, and rhythmic complexity interactively engage infants' visual attention to a tapping hand, especially when that hand is aligned with the auditory stimulus. For example, when the rhythm was complex and the auditory stimulus was low in pitch, infants attended to the fast hand more when it aligned with the auditory stream than to misaligned trials. These results suggest that the audiovisual integration in rhythmic non-speech contexts is influenced by stimulus properties.
Factors affecting production of mold mycelium and protein in synthetic media

Abstract:The effects of certain cultural conditions on the yield of dry mycelium, protein, and total amino acid content of Rhizopus oligosporus Saito (NRRL 2710), Rhizopus rhizopodiformis (Cohn apud Lichtheim) Zopf (NRRL 6246), and Absidia corymbifera (Cohn) Sacc. et Trotter (NRRL 6247) were studied. The yield of mycelium was found to significantly increase as the spore inoculum was increased from 187,500 to 2,250,000 spores. But the total amino acids (grams/liter) did not change significantly, whereas the percentage of crude protein decreased. An inoculum containing approximately 750,000 spores/ml was used in all of the other experiments. Mycelial production was highest at 37 degrees C for all three molds. However, the best temperature for percentage of crude protein and total amino acids varied with the organism. The mycelial yield and total crude protein of R. oligosporus showed some significant changes as the C/N ratio was increased in 3% glucose medium. In a synthetic medium having a 15:1 C/N ratio, the strains of R. oligosporus, R. rhizopodiformis, and A. corymbifera had better yields from falactose than glucose, not only in dry mycelium but also in total crude protein (grams/liter) and total amino acids (grams/liter). R. oligosporus grew very well on several ammonium salts. but the maximum yield of dry mycelium, total crude protein (grams/liter), and total amino acids (grams/liter) occurred with ammonium sulfate. The optimum pH for both Rhizopus species was 4.0, although R. oligosporus grew equally well at pH 3.0 and slightly less at pH 5.0. The highest yield of mycelium for A. corymbifera was obtained in a medium with an initial pH of 8.0. It was calculated that a fermenter chanrged with an adequate medium and 1,000 lb (about 450 kg) of R. oligosporus or A. corymbifera cells could produce 88 or 90 lb of protein (on a dry-weight basis) per h if the product was removed continuously.

Abstract:Attention-sensitive signalling is the pragmatic skill of signallers who adjust the modality of their communicative signals to their recipient's attention state. This study provides the first comprehensive evidence for its onset and development in 7-to 20-month-olds human infants, and underlines its significance for language acquisition and evolutionary history. Mother-infant dyads ( N = 30) were studied in naturalistic settings, sampled according to three developmental periods (in months); [7–10], [11–14], and [15–20]. Infant's signals were classified by dominant perceptible sensory modality and proportions compared according to their mother's visual attention , infant-directed speech and tactile contact . Maternal visual attention and infant-directed speech were influential on the onset and steepness of infants' communicative adjustments. The ability to inhibit silent-visual signals towards visually inattentive mothers ( unimodal adjustment) predated the ability to deploy audible-or-contact signals in this case ( cross-modal adjustment). Maternal scaffolding of infant's early pragmatic skills through her infant-directed speech operates on the facilitation of infant's unimodal adjustment, the preference for oral over gestural signals, and the audio-visual combinations of signals. Additionally, breakdowns in maternal visual attention are associated with increased use of the audible-oral modality/channel. The evolutionary role of the sharing of attentional resources between parents and infants into the emergence of modern language is discussed.

Abstract:The present study aimed at investigating the ability of 7‐ to 20‐month‐old infants to display attention‐sensitive communication using either canonical markers of language acquisition (e.g., pointing gestures, canonical babblings) or other signals based on the physical features actually perceived by the mother in everyday interaction (e.g., body movements, mouth sounds). We studied 30 French mother‐infant dyads in naturalistic settings. We assessed the infants' attention‐sensitive communication through unimodal and cross‐modal adjustment, defined as the capacity of infants to address visually inattentive mothers by avoiding visual communication mismatches and/or favoring communication matches through audible‐or‐contact signals. Unimodal and cross‐modal adjustments were tested for specific signals across spontaneous "conditions" of maternal visual attention (attentive/inattentive) from video footage filmed in the home. Both canonical markers of language development and signals belonging to an extended repertoire of communication were used by infants to adjust to their mother's visual attention. Gaze‐coordinated signals were overall not significantly better adjusted to maternal attention than non‐gaze‐coordinated signals, except for specific silent‐visual signals at certain ages. Overall, these results indicate that attention‐sensitive communication is relevant to the development of early pragmatic skills and that the intentional use of signals may be more reliably approximated by this capacity than by gaze‐coordination with signals.
