Abstract:Further exploitation of the residual oil underground in post-polymer flooded reservoirs is attractive and challenging. Microbial-enhanced oil recovery (MEOR) is a promising strategy to enhance the recovery of residual oil in post-polymer flooded reservoirs. Identifying and selectively activating indigenous microorganisms with oil displacement capabilities is an urgent requirement in the current design of efficient microbial-enhanced oil recovery technologies. This study combines high-throughput sequencing with functional network analysis to identify the core functional microbes within the reservoirs. Concurrently, it devises targeted activation strategies tailored to oligotrophic conditions through an analysis of environmental factor influences. The feasibility of these strategies is then validated through physical simulation experiments. With nutrient stimulation, the overall diversity of microorganisms decreases while the abundance of functional microorganisms increases. The core displacement results showed that the oil recovery factor increased by 3.82% on the basis of polymer flooding. In summary, this research has established a system for the efficient activation of functional microorganisms under oligotrophic conditions by utilizing bioinformatics, network analysis, and indoor simulation systems. This achievement will undoubtedly lay a solid foundation for the practical implementation of microbial enhancement techniques in the field.

Abstract:Microbial enhanced oil recovery (MEOR) is a technology that could potentially increase the tertiary recovery of oil from mature oil formations. However, the efficacy of this technology in fractional-wet systems is unknown, and the mechanisms involved in oil mobilization therefore need further investigation. Our MEOR strategy consists of the injection of ex situ produced metabolic byproducts produced by Bacillus mojavensis JF-2 (which lower interfacial tension (IFT) via biosurfactant production) into fractional-wet cores containing residual oil. Two different MEOR flooding solutions were tested; one solution contained both microbes and metabolic byproducts while the other contained only the metabolic byproducts. The columns were imaged with X-ray computed microtomography (CMT) after water flooding, and after MEOR, which allowed for the evaluation of the pore-scale processes taking place during MEOR. Results indicate that the larger residual oil blobs and residual oil held under relatively low capillary pressures were the main fractions recovered during MEOR. Residual oil saturation, interfacial curvatures, and oil blob sizes were measured from the CMT images and used to develop a conceptual model for MEOR in fractional-wet systems. Overall, results indicate that MEOR was effective at recovering oil from fractional-wet systems with reported additional oil recovered (AOR) values between 44 and 80%; the highest AOR values were observed in the most oil-wet system.

Abstract:The microbial enhanced oil recovery (MEOR) process has been identified as a promising alternative to conventional enhanced oil recovery methods because it is eco-friendly and economically advantageous. However, the knowledge about the composition and diversity of microbial communities in artificially regulated reservoirs, especially after activating petroleum hydrocarbon-degrading bacteria (PHDB) by injecting exogenous nutrients, is still insufficient. This study utilized a combination of high-throughput sequencing and metagenomics technology to reveal the structural evolution characteristics of the indigenous microbial community in the reservoir during the PHDB activated for enhanced oil recovery, as well as the response relationship between the expression of its oil production functional genes and crude oil biodegradation. Results showed that Pseudomonas (>75%) gradually evolves into a stable dominant microbial community in the reservoir during the activation of PHDB. Besides, the gene expression and KEGG pathways after crude oil undergoes biodegradation by PHDB show that the number of genes related to petroleum hydrocarbon metabolism dominates the metabolism (21.98%). Meanwhile, a preliminary schematic diagram was drawn to illustrate the evolution mechanism of the EOR metabolic pathway after the targeted activation of PHDB. Additionally, it was found that the abundance of hydrocarbon-degrading enzymes increased significantly, and the activity of alcohol dehydrogenase was higher than that of aldehyde dehydrogenase and monooxygenase after PHDB activation. These research results not only filled in and expanded the theoretical knowledge of MEOR based on artificial interference or regulation of reservoir oil-recovery functional microbial community structure but also provided guidance for the future application of MEOR technology in oil field operations.

Abstract:Due to the inefficient reproduction of microorganisms in oxygen-deprived environments of the reservoir, the applications of microbial enhanced oil recovery (MEOR) are restricted. To overcome this problem, a new type of air-assisted MEOR process was investigated. Three compounding oil degradation strains were screened using biochemical experiments. Their performances in bacterial suspensions with different amounts of dissolved oxygen were evaluated. Water flooding, microbial flooding and air-assisted microbial flooding core flow experiments were carried out. Carbon distribution curve of biodegraded oil with different oxygen concentration was determined by chromatographic analysis. The long-chain alkanes are degraded by microorganisms. A simulation model was established to take into account the change in oxygen concentration in the reservoir. The results showed that the optimal dissolved oxygen concentration for microbial growth was 4.5~5.5mg/L. The main oxygen consumption in the reservoir happened in the stationary and declining phases of the microbial growth systems. In order to reduce the oxygen concentration to a safe level, the minimum radius of oxygen consumption was found to be about 145m. These results demonstrate that the air-assisted MEOR process can overcome the shortcomings of traditional microbial flooding techniques. The findings of this study can help for better understanding of microbial enhanced oil recovery and improving the efficiency of microbial oil displacement.

Abstract:<p>After conventional oil recovery operations, abundant residual oil is still trapped in pore space of complex capillary network in the reservoir. As a promising method of tertiary oil recovery, microbial enhanced oil recovery (MEOR) involves the use of indigenous or specially screened microorganisms to produce specific metabolites by injecting formation or ground fermentation to enhance crude oil recovery. This paper discussed the mechanism of mobilizing trapped crude oil by microorganisms and their metabolites in detail, including interfacial tension reduction, wettability alteration, selective plugging, and degradation. Furthermore, numerical simulation for MEOR and novel technologies, such as genetically engineered MEOR (GEMEOR) and enzyme enhanced oil recovery (EEOR) were also introduced. Finally, screening criteria of reservoirs and 19 field trials cases in 9 countries were summarised. Currently, the field application of MEOR is still restricted due to the inconsistent and unstable effects and the possible negative impact of nutrient and exogenous microbial injection. However, considering the economic feasibility and environmental benefit of MEOR, it has promising application prospects in the further development of the oil fields.</p>

Abstract:Primary and secondary recovery methods are usually not sufficient to maximize the oil recovery. In many cases, more than 40% of the Oil Initially in Place is left in the reservoir after implementing these recovery methods. To resolve the issue at hand, petroleum engineers have at their disposition a plethora of tertiary recovery methods such as chemical flooding, thermal recovery, and microbial enhanced oil recovery (MEOR). In this study, we propose the use of MEOR: a technique that uses naturally occurring microbes in the reservoir or injected microbes to enhance the oil recovery. The MEOR process is governed by several mechanisms such as viscosity reduction, relative permeability alteration, capillary pressure alteration, and other reaction-induced system changes such as pressurization. This study investigates the impact of these mechanisms on the hydrocarbon production by conducting a numerical simulation using our in-house MEOR numerical simulator. In the base case study, the case with MEOR shows the 20% of oil production improvement than the case without MEOR. We conduct a sensitivity analysis to evaluate the impact of microbial factors affecting MEOR to the system responses and production behavior, where the maximum growth rate of microbes is found to be the most influential factor with the scaled sensitivity coefficient of 1.384 × 10 5 . The findings provide the relative impacts of factors affecting the performance of MEOR, which subsequently suggest the factors to be investigated with emphasis for oil production improvement in the MEOR applications.

Abstract:Performance evaluation of microorganisms that have emulsifying and degrading effects on crude oil has been extensively conducted in the laboratory. However, the ultimate goal of microbial enhanced oil recovery is field application, so the pilot simulation experiments are crucial. In this study, a micro-visualization model and the real cores were chosen to investigate the actual recovery efficiency and the mechanism of the consortium bacteria B-ALL, which has been proven to have good emulsification and degradation effects in lab studies in porous media. At the same time, the cast thin sections and rate-controlled porosimetry were combined to analyze the pore throat structure of the displacement core. It was found that the recovery efficiency was positively correlated with the microbial injection volume as well as the incubation time. For the microscopic model with high pores and high permeability, the efficiency of secondary water flooding can be increased by 44.77% after six days of incubation with two pore volume microbes. For the real tight cores, the maximum secondary water flooding efficiency under the same condition was 6.98%. Through visual modeling, microorganisms increase the oil washing efficiency mainly by emulsification and changing the wettability. The generated oil droplets will play a role in plugging and improving the wave efficiency. However, tight reservoirs have the characteristics of large pores and small throats, and curved and necking throats are developed, greatly reducing permeability. The microbial recovery efficiency was lower under shorter cultivation times. This study provides a practical basis for the application of consortium bacteria in tight oil fields to enhance recovery.

Abstract:Based on preliminary investigation of microbial populations in a high pour-point oil reservoir, an indigenous microbial enhanced oil recovery (MEOR) field trial was carried out. The purpose of the study is to reveal the impact of the indigenous MEOR process on microbial community structure in the oil reservoir using 16Sr DNA clone library technique. The detailed monitoring results showed significant response of microbial communities during the field trial and large discrepancies of stimulated microorganisms in the laboratory and in the natural oil reservoir. More specifically, after nutrients injection, the original dominant populations of Petrobacter and Alishewanella in the production wells almost disappeared. The expected desirable population of Pseudomonas aeruginosa, determined by enrichment experiments in laboratory, was stimulated successfully in two wells of the five monitored wells. Unexpectedly, another potential population of Pseudomonas pseudoalcaligenes which were not detected in the enrichment culture in laboratory was stimulated in the other three monitored production wells. In this study, monitoring of microbial community displayed a comprehensive alteration of microbial populations during the field trial to remedy the deficiency of culture-dependent monitoring methods. The results would help to develop and apply more MEOR processes.

Abstract:The focus of this paper is the derivation of a nonstandard model for microbial enhanced oil recovery (MEOR) that includes the interfacial area (IFA) between oil and water. We consider the continuity equations for water and oil, a balance equation for the oil–water interfacial area, and advective–dispersive transport equations for bacteria, nutrients, and biosurfactants. Surfactants lower the interfacial tension (IFT), which improves oil recovery. Therefore, the parametrizations of the IFT reduction and residual oil saturation are included as a function of the surfactant concentration in the model. We consider for the first time in context of MEOR, the role of IFA in enhanced oil recovery. The motivation to include the IFA is to model the hysteresis in the capillary pressure–saturation relationship in a physically based manner, to include the effects of observed bacteria migration toward the oil–water interface and the production of biosurfactants at the oil–water interface. A comprehensive 2D implementation based on two-point flux approximation and backward Euler is proposed. An efficient and robust linearization scheme is used to solve the nonlinear systems at each time step. Illustrative numerical simulations are presented. The differences in the oil recovery profiles obtained with and without IFA are discussed. The presented model can also be used to design new experiments toward a better understanding and eventually optimization of MEOR.

Abstract:Microbial enhanced oil recovery (MEOR) is an important tertiary recovery technology to recover oil, especially heavy oil, from reservoirs. However, MEOR often fails to achieve satisfactory oil recovery in the field applications because of utilization of unsuitable microorganisms and nutrients. Recent studies proved the existence of microbes inside oil ("oil-phase" microbes) and their potential for application in MEOR. To validate this concept, we isolated microbes from the "oil-phase" of crude oil samples. With them, a synthetic four-member microbial consortium K, was selected to use in an experiment of core flooding, which showed a beneficial capacity for improving oil recovery. Furthermore, the consortium K and nutrients were applied in a field trial of microbial huff and puff in an extra heavy oil recovery. Results demonstrated the incremental cumulative oil production of two high water-cut (99%) shut-in wells reached 1078.8 t and 791.6 t, respectively, with the effective period of 14 months, while the viscosity of heavy oil had significantly declined decreased by 50%. In conclusion, our findings clearly demonstrate that the great potential of a novel methodology to use indigenous "oil-phase" microbes as seedbanks for improving oil recovery in extremely viscous oil reservoirs with high water-cut.

Abstract:After conventional oil recovery operations, more than half of the crude oil still remains in a form, which is difficult to extract. Therefore, exploring and developing new enhanced oil recovery (EOR) technologies have always been priority research in oilfield development. Microbial enhanced oil recovery (MEOR) is a promising tertiary oil recovery technology that has received widespread attention from the global oil industry in recent years due to its environmental friendliness, simplicity of operation, and cost-effectiveness. This review presents the: principle, characteristics, classification, recent development, and applications of MEOR technology. Based on hundreds of field trials conducted worldwide, the microbial strains, nutrient systems, and actual effects used in these technologies are summarized, with an emphasis on the achievements made in the development and application of MEOR in China in recent years. These technical classifications involve: microbial huff and puff recovery (MHPR), microbial flooding recovery (MFR), microbial selective plugging recovery (MSPR), and microbial wax removal and control (MWRC). Most of them have achieved good results, with a success rate of approximately 80%. These successful cases have accumulated into rich experiential indications for the popularization and application of MEOR technology, but there are still important yet uncertain factors that hinder the industrialization of this technology. Finally, based on the extensive research and development of MEOR by the authors, especially in both laboratory and industrial large scales, the main challenges and future perspectives of the industrial application for MEOR are presented.

Enrichment of microbial consortia for MEOR in crude oil phase of reservoir-produced liquid and their response to environmental disturbance

Abstract:Developing microbial consortiums is necessary for microbial enhanced oil recovery (MEOR) in heavy crude oil production. The aqueous phase of produced fluid has long been considered an ideal source of microorganisms for MEOR. However, it is recently found that rich microorganisms (including hydrocarbon-degrading bacteria) are present in the crude oil phase, which is completely different from the aqueous phase of produced fluid. So, in this study, the microbial consortia from the crude oil phase of produced fluids derived from four wells were enriched, respectively. The microbial community structure during passage was dynamically tracked, and the response of enriched consortia to successive disturbance of environmental factors was investigated. The results showed the crude oil phase had high microbial diversity, and the original microbial community structure from four wells was significantly different. After ten generations of consecutive enrichment, different genera were observed in the four enriched microbial consortia, namely, Geobacillus, Bacillus, Brevibacillus, Chelativorans, Ureibacillus, and Ornithinicoccus. In addition, two enriched consortia (eG1614 and eP30) exhibited robustness to temperature and oxygen perturbations. These results further suggested that the crude oil phase of produced fluids can serve as a potential microbial source for MEOR.

Abstract:For the last 20 years Shengli oilfield has done research into the application of microbial enhanced oil recovery (MEOR). The authors summarize and analyze MEOR progress in mechanism research and field application in Shengli oilfield. The results indicate MEOR could improve oil recovery after water flooding and polymer flooding with multiple mechanisms. Cumulative oil increment of MEOR application on seven blocks was 212,366 t. The research and application of MEOR should concentrate on air assisted microbial flooding and indigenous microbial flooding in the future.

Abstract:Component transportation is one of the main mechanisms for numerical simulation in microbial oil recovery. However, the research on the component transportation considering the inhibition of metabolites is very limited. A mathematical model of oil displacement in a microorganism system including microbial growth and metabolism equation, component transport equation, and porous media physical property variation equation was established in this paper. The equation was discretized and solved by implicit pressure and explicit saturation. The MATLAB simulation results showed that the chromatographic separation between microorganisms and nutrients happened because of the adsorption of porous media and the activity of microorganisms during the transportation, and the separation degree of the chromatography became higher as the permeability became lower and the injection speed became slower. The multislug alternative injection mode could reduce the degree of chromatographic separation, and the recovery rate can be increased to 50.82%. The results of this study could provide theoretical guidance for the popularization and application of microbial enhanced oil recovery (MEOR).This article has not yet been cited by other publications.

Abstract:Understanding microbial community in oil reservoirs is crucial to application of microbial enhanced oil recovery (MEOR) technology. Much effort has been directed toward more insights into the microbial community in oil field, resulting in the development of culture dependent and independent techniques. Culture-dependent methods for enhancement of microbial culturability and culture-independent methods for analyzing microbial communities are reviewed in an attempt to better understand the recent progress in methodological researches on the oil field microbial communities. Microbial community is analyzed conventionally by culture-dependent approaches which cultivate, enrich and isolate microbial cells, identify and count them, then figure out the microbial structure from the data of the cultivated microbial cells. However, the cultivating nutrients and conditions afforded in laboratory are too simplified and somehow far different from those in the environments where microorganisms live. Consequently, majority of microorganisms can not grow and be culturable. In addition, microorganisms grow at different speeds on the same media and under the same conditions, leading to different enrichment and increase of the microbial population. The relative amounts of the cultivated microorganisms are therefore different from the real ratio of the microorganisms in the environments. As a consequence, the pattern of microbial community structure obtained by the conventional culture-dependent methods is not true and not helpful for directing the development and application of MEOR. Two strategies, the increasing microbial culturability and culture-independent methods, are therefore developed and applied for getting more culturable microbial isolates and more insights into the real microbial community structure and functions. Increasing microbial culturability is to cultivate microorganisms with novel methods and under novel conditions. These novel cultivating conditions include: "oligotrophic" media instead of "rich" nutrients, adding signal molecules, novel electronic donating and accepting chemicals to media, etc.. The novel cultivating methods include dilution culture, high-through culturing, diffusion-growth chamber, cell encapsulation, sequence-guiding isolation techniques, etc. which can mimic environmental conditions and lead to more microorganisms cultivated. Although the novel cultivating techniques and conditions can make much more microorganisms culturable, they can not figure out the overall pattern of microbial community structure, the culture-independent approaches are therefore absolutely necessary for analyzing oil reservoir microbial community. Culture-independent approaches are based on the analyses of the functional genes, such as mcrA gene, and 16S rRNA and its gene (16S rDNA). The analyzing techniques contain FISH, T-RFLP, DGGE, clone library and sequencing etc.. Many researches with the culture-independent methods have showed the power of these methods and got many new ideas on the structure and functions of the microbial community in oil reservoirs. Moreover, it is a trend to apply both culture-dependent and culture-independent approaches to understand the functions of microorganisms for MEOR and structure of microbial community. With the microbial strains obtained from oil reservoirs and the knowledge of oil reservoir microbial community, MEOR can be developed and applied feasibly and reliably by manipulating the microbial community in the oil reservoirs.

The remaining ~70% of the world’s conventional oil (one-third of the available total petroleum) is trapped in depleted and marginal reservoirs, and could thus be potentially recovered and used. The only means of extracting this oil is via microbial enhanced oil recovery (MEOR). This tertiary oil recovery method employs indigenous microorganisms and their metabolic products to enhance oil mobilization. Although a significant amount of research has been undertaken on MEOR, the absence of convincing evidence has contributed to the petroleum industry’s low interest, as evidenced by the issuance of 400+ patents on MEOR that have not been accepted by this sector. The majority of the world’s MEOR field trials are briefly described in this review. However, the presented research fails to provide valid verification that the microbial system has the potential to address the identified constraints. Rather than promising certainty, MEOR will persist as an unverified concept unless further research and investigations are carried out.

Microbial Functioning on Crude Oil in a Gas-Permeable Single Microfluidic Channel

Abstract:Although various studies have been conducted in screening oil-degrading microorganisms and the macroscale degrading efficiency, critical processes of microorganisms in the microscale porous space are not clear, which are important for microbial enhanced oil recovery (MEOR) and bioremediation of oil contaminated environments. In this work a gas-permeable microchannel filled with crude oil was employed to study the microbial processes in a microscale environment. Cells of a petroleum-degrading Dietzia strain were suspended with two different media and the resulting suspensions were ejected into the microchannel. The degradation processes in the oil–suspension system were then analyzed. The changes in the behavior of the cells and the variation of the oil–suspension interface as well as the evolution of the suspension patterns and structures were observed. ‘Aggregates’, ‘clustered cells’, and ‘oil droplets’ were characteristically formed in comparison with the control experiments. Oil droplets were formed at the oil–suspension interface and dispersed into the suspension, and this process could be drastic that a plume of oil droplets was issuing into the suspension. All the changes could be attributed to the degradation of crude oil by the cells and the production of biosurfactants which could reduce the interface tension and increase the dissolution of the crude oil. The present work is a preliminary step toward a comprehensive understanding of microbial process in microscale porous space.
Enhanced Oil Recovery in a Co-Culture System of Pseudomonas Aeruginosa and Bacillus Subtilis

Abstract:Microbial enhanced oil recovery (MEOR) is a promising technology for oil field extraction. This study investigated a co-culture system of Pseudomonas aeruginosa and Bacillus subtilis to increase MEOR efficacy. We analyzed bacterial growth, biosurfactant production, and crude oil emulsified performance under different inoculation ratios. Compared to single cultures, the co-culture system showed superior growth and functional expression, with an optimal inoculation ratio of 1:1. Quantitative assessments of the cell numbers and biosurfactant production during the co-culture revealed that rapid B. subtilis proliferation in early stages significantly stimulated P. aeruginosa growth. This interaction increased cell density and rhamnolipid production by 208.05% and 216.25%, respectively. The microscopic etching model displacement results demonstrated enhanced emulsification and mobilization of crude oil by the co-culture system, resulting in 94.48% recovery. A successful field application in a block-scale reservoir increased cumulative oil production by 3.25 × 103 t. An analysis of microbial community structure and function in different phases revealed that after co-culture system injection, Pseudomonas became the dominant genus in the reservoir community, with an average abundance of 24.80%. Additionally, the abundance of biosurfactant-producing and hydrocarbon-degrading bacteria increased significantly. This research and the application of the P. aeruginosa and B. subtilis co-culture system provide novel insights and strategies for MEOR.

Abstract:Microbial Enhanced Oil Recovery (MEOR) is a pivotal technology for enhancing tertiary oil recovery. The production of partly extracellular polymeric substances (EPS) by Enterobacter cloacae exhibits excellent control over mobility and holds promise for applications in heterogeneous reservoir exploration. To investigate the impact of Ba2+ on microbial EPS production, a series of tests were conducted, including EPS production tests, polysaccharide and protein content analyses, Inductively Coupled Plasma Optical Emission Spectrometer (ICP-OES) measurements, X-ray diffraction (XRD), laser particle size measurements, and transcriptome analysis.The results demonstrated the stimulating effect of Ba2+ on EPS membrane production. Ba2+ stimulation led to increased secretion of proteins (5.82 g/L) and polysaccharides (9.0 g/L). Moreover, the binding of proteins with Ba2+ resulted in an enlargement of the particle size of the EPS membrane and an increase in grain size, reaching a maximum crystallinity of 77.77 %. Transcriptome analysis revealed the up-regulation of 790 genes and down-regulation of 728 genes in response to Ba2+ addition. Enrichment analysis of the up-regulated genes highlighted the significant enrichment of gene ontology number GO: 00160120, which directly influenced microbial EPS membrane production. Furthermore, microscopic displacement and core flooding experiments were conducted to evaluate the plugging ability of microbial-produced EPS membranes. Remarkably, under the influence of Ba2+, microorganisms exhibited significantly enhanced sealing capacity on glass slides etched with rock cores. The results from simulated flow experiments demonstrated a maximum sealing rate of 81.5 % on rock cores, with an RF (Resistance Factor) value of 2.5 and an RRF (Residual Resistance Factor) value of 5.45. This study innovatively explores oil-producing microbial genes, combining simulation experiments and rock core slicing to reveal the microscale mechanism of Ba2+ stimulating microbial EPS secretion for reservoir plugging. It provides theoretical support for the application of MEOR technology in improving oil recovery rates.
