Abstract:The effect of treatment with 50 mg·kg~(-1) GA_3 solution for 30 min on lignification and endogenous hormone levels in postharvest bamboo(Phyllostachys praecox f. preveynalis)shoots were investigated. Results showed that the activities of phenylalanine ammonia lyase(PAL),cinnamate 4-hydroxylase(C4H) and peroxidase(POD) increased initially and declined later during the 30 days storage at 2 ℃. The lignin content and firmness continuously increased, but edible rate decreased gradually. The contents of GA_3 and IAA declined continuously, whereas ABA content increased firstly and then declined. The activities of PAL, C4H and POD were correlated with the rate of ABA/GA_3. Exogenous GA_3 treatment increased endogenous level of GA_3 content and decreased the rate of ABA/GA_3 as well as inhibited the increase of PAL, C4H and POD activities in bamboo shoots. The firmness and lignin content in GA_3 treated bamboo shoot were lower than those of control, while the edible rate was higher than that of control. It is suggested that GA_3 treatment retarded lignification through decreasing the rate of ABA/GA_3 in postharvest bamboo shoots.

Abstract:(1) The phytohormones gibberellins (GAs) play a crucial role in plant growth and development, such as seed germination, flowering, fruiting, and stem elongation. Although many biological roles of GAs have been studied intensively, the molecular mechanisms of GAs in woody plants are still unclear. (2) In this study, we investigated the effects of exogenous application of GAs on Neolamarckia cadamba. (3) The height and biomass of N. cadamba increased after 7 days of GA treatment, especially on the second internode. Transcriptome analysis showed that although the majority of genes involved in the GA signaling pathway were up-regulated, the expression of GA20 oxidase (GA20ox) and GA3 oxidase (GA3ox) was down-regulated in the 3 days GA-treated group compared to the CK group. The expression of the cell elongation-related basic helix-loop-helix genes bHLH74 and bHLH49 was up-regulated in the GA-treated group compared with the CK group. Transcriptional expression levels of transcription factors involved in hormone signaling were changed, mainly including bHLH, ethylene response factor (ERF), and WRKY families. In addition, the transcriptional expression level of the key enzymes engaged in the phenylalanine pathway was downregulated after GA treatment. (4) In brief, our findings reveal the physiological and molecular mechanisms of exogenous GA treatment stimulation in N. cadamba.

Effects of Exogenous GA3 and DPC Treatments on Levels of Endogenous Hormone and Expression of Key Gibberellin Biosynthesis Pathway Genes During Stem Elongation in Sugarcane

Abstract:Internode elongation determines plant height, which in turn affects the cane yield in sugarcane. This study aimed to explore the mechanisms of sugarcane internode elongation regulation by plant growth regulators, gibberellic acid (GA3) and its inhibitor 1, 1-dimethylpiperidinium chloride (DPC, also called mepiquat chloride). We sprayed GA3 or DPC on the leaves at the early shoot elongation stage with water as the control. The contents of endogenous hormones and the expression of GA20 ox1, GID1, and GAI in sugarcane leaves were measured at days 3, 6, 12, and 24 after treatment. It was found that the height of sugarcane plants treated with GA3 showed a significant increase compared to the control, whereas an opposite trend was observed in the DPC-treated plants. Compared to the control, the content of gibberellins (GAs) in leaves was increased significantly, while the indole acetic acid (IAA) content showed an initial increase, then a decline, and eventually remained stable in the GA3-treated plants. The contents of cytokinins (CTK), abscisic acid (ABA), and ethylene (ETH) in leaves exhibited a significant downward trend in the GA3-treated plants compared to the control. In the DCP-treated plants, however, the levels of ABA and ETH in leaves were significantly increased, IAA and GAs significantly decreased, and CTK showed a trend of an initial decline followed by an increase. After GA3 treatment, the gene expression levels of GA20 ox1 and GID1 in sugarcane increased first then decreased, whereas the gene expression level of GAI first decreased and then stabilized. After DPC treatment, the expression of GA20 ox1 in sugarcane showed an initial decrease followed by an increase, while the expression of GID1 decreased gradually and then stabilized, and the expression of GAI showed a trend of an initial increase followed by stabilization. The results of the present study indicate that exogenous GA3 treatment stimulated the internode elongation in sugarcane mainly by increasing the endogenous levels of GAs, decreasing the levels of ABA and ETH, and regulating the expression of GA20 ox1, GID1 and GAI genes; the DPC treatment mainly increased the contents of ABA and ETH and reduced the contents of GAs and IAA, thereby inhibiting the expression of GID1, promoting the expression of GAI, and ultimately inhibiting the internode elongation in sugarcane.

Abstract:Although phytohormones are known to be important signal molecules involved in wood formation, their roles are still largely unclear. Here, Populus simonii × P. nigra seedlings were treated with different concentrations of exogenous phytohormones, indole-3-acetic acid (IAA), gibberellin (GA3), and brassinosteroid (BR), and the effects of phytohormones on growth were investigated. Next, 27 genes with known roles in wood formation were selected for qPCR analysis to determine tissue-specificity and timing of responses to phytohormone treatments. Compared to the control, most IAA, GA3, and BR concentrations significantly increased seedling height. Meanwhile, IAA induced significant seedling stem diameter and cellulose content increases that peaked at 3 and 30 mg·L−1, respectively. Significant increase in cellulose content was also observed in seedlings treated with 100 mg·L−1 GA3. Neither stem diameter nor cellulose content of seedlings were affected by BR treatment significantly, although slight effects were observed. Anatomical measurements demonstrated improved xylem, but not phloem, development in IAA- and BR-treated seedlings. Most gene expression patterns induced by IAA, GA3, and BR differed among tissues. Many IAA response genes were also regulated by GA3, while BR-induced transcription was weaker and slower in Populus than for IAA and GA3. These results reveal the roles played by phytohormones in plant growth and lay the foundation for exploring molecular regulatory mechanisms of wood formation in Populus.

Abstract:'Duli' (Pyrus betulifolia Bunge) is one of the main rootstocks of pear trees in China. Gibberellin (GA) is a key plant hormone and the roles of GA in nitrate (NO3-) uptake and metabolism in plants remain unclear. In this study, we investigated the effects of exogenous GA3 on the N metabolism of 'Duli' seedlings under NO3- deficiency. The results showed that exogenous GA3 significantly improves 'Duli' growth under NO3- deficiency. On the one hand, GA3 altered the root architecture, increased the content of endogenous hormones (GA3, IAA, and ZR), and enhanced photosynthesis; on the other hand, it enhanced the activities of N-metabolizing enzymes and the accumulation of N, and increased the expression levels of N absorption (PbNRT2) and the metabolism genes (PbNR, PbGILE, PbGS, and PbGOGAT). However, GA3 did not delay the degradation of chlorophyll. Paclobutrazol had the opposite effect on growth. Overall, GA3 can increase NO3- uptake and metabolism and relieve the growth inhibition of 'Duli' seedlings under NO3- deficiency.

Abstract:In this study, we investigated the roles of the plant hormones indole-3-acetic acid (IAA) and gibberellin (GA) in regulating Pinus massoniana Lamb. plantlet regeneration during long-term in vitro subculture. Six generations (1st, 5th, 10th, 20th, 30th and 40th with 40 days for each transfer cycle) of subcultured shoots derived from mature and juvenile explants were characterised for their different rooting capacities. In the present experiment, shoots were subcultured on rooting medium containing indole-3-acetic acid (IAA), naphthaleneacetic acid (NAA) or gibberellin (GA) inhibitor (paclobutrazol, PBZ). In addition, high-performance liquid chromatography (HPLC) was used to analyse endogenous hormone levels in shoots developed on medium with low concentrations of NAA. The concentration of endogenous IAA was highest in the 20th generation shoots, which also exhibited the strongest rooting ability. However, the highest GA concentration was observed in the 40th generation shoots, which exhibited a poor rooting capacity. For shoots subcultured for 40 generations, incubation on rooting medium containing IAA caused an enhanced root number, shoot length and survival rate. This suggested that the effects of IAA on shoot and root formation were positive. PBZ promoted plantlet regeneration from in vitro cultures but impaired shoot elongation at high levels. Furthermore, PBZ improved performance of in vitro cultures after long-term (3 to 4 y) subculture. These results suggest the inhibitory role of a high endogenous GA level in plant regeneration and the use of appropriate levels of PBZ for plant regeneration of P. massoniana.

Abstract:Sweetness is one of the key factors determining peach fruit quality. To better understand the molecular basis of gibberellic acid (GA) and 1-naphthaleneacetic acid (NAA) interference with sugar biosynthesis, a middle-late maturing commercial cultivar, ‘Jinxiu’ yellow peach fruit, was treated with three different concentrations of GA 4+7 and four of NAA. Fruit weight, firmness, total soluble solids, different sugar contents and the expression level of sugar-related genes were evaluated. The results showed that maximum increase in cv. ‘Jinxiu’ peach fruit size and sucrose content was with 1.25 mM GA 4+7 , compared to control fruits and the other treatments during the ripening stages. The sucrose-phosphate synthase gene ( PpSPS 2 ) which had a high level of expression and positive correlation with sucrose content was significantly regulated by 1.25 mM GA 4+7 in the final ripening stages. 0.5 mM NAA treatments significantly reduced the sucrose content and fruit size. Ninety percent of the fruits were deformed or dropped from the trees with treatments of 1 mM NAA and 2 mM NAA in the early development period. The crosstalk of different phytohormones and the related genes will be further investigated to get an insight into the inherent association between hormone control and sugar accumulation.

Abstract:Gibberellin (GA) is an important phytohormone that can participate in various developmental processes of plants. The study found that application of GA 3 can induce parthenocarpy fruit and improve fruit set. However, the use of GA 3 affects endogenous hormones in fruits, thereby affecting fruit quality. This study mainly investigates the effect of exogenous GA 3 on endogenous hormones in sweet cherries. The anabolic pathways of each hormone were analyzed by metabolome and transcriptome to identify key metabolites and genes that affect endogenous hormones in response to exogenous GA 3 application. Results showed that exogenous GA 3 led to a significant increase in the content of abscisic acid (ABA) and GA and affected jasmonic acid (JA) and auxin (IAA). At the same time, the key structural genes affecting the synthesis of various hormones were preliminarily determined. Combined with transcription factor family analysis, WRKY genes were found to be more sensitive to the use of exogenous GA 3 , especially the genes belonging to Group III ( PaWRKY16 , PaWRKY21 , PaWRKY38 , PaWRKY52 , and PaWRKY53 ). These transcription factors can combine with the promoters of NCED , YUCCA , and other genes to regulate the content of endogenous hormones. These findings lay the foundation for the preliminary determination of the mechanism of GA 3 's effect on endogenous hormones in sweet cherry and the biological function of WRKY transcription factors.

Abstract:Elevating the yield and altering the composition of seed tocopherols (Toc's) are important to rapeseed breeding and production. However, little is known about the biosynthesis of Toc's in response to environmental signals. In this study, we investigated the effects of exogenous gibberellin (GA(3)) and paclobutrazol (PAC) on Toc biosynthesis. We also explored the interactive effects between the two plant growth regulators (PGRs) and other factors, such as PGR treatment duration, genotype, and growing location on the total Toc yield and composition in oilseed rape seed. GA(3) significantly enhanced the production of Toc's and elevated the alpha-/gamma-Toc ratio in a time- and genotype-dependent manner. By contrast, PAC significantly reduced Toc yield. Genotypic differences were observed in the effects of GA(3) on Toc yield and composition in the seeds. GA(3) significantly increased the Toc yield and alpha-/gamma-Toc ratio in Zheyou-50, a genotype with a low proportion of very long chain fatty acids (VLCFAs). However, GA(3) did not significantly influence these parameters in Jiu-Er-13Xi, a genotype with a high VLCFA proportion. The increased Toc yield induced by GA(3) was mediated by the upregulation of genes (BnPDS1 and BnVTE1) that catalyze the production of Toc precursors. Therefore, applying GA(3) can improve rapeseed quality by increasing Toc yield and improving Toc composition.

Abstract:Light intensity and hormones (gibberellins; GAs) alter plant growth and development. A fine regulation triggered by light and GAs induces changes in stem cell walls (CW). Cross-talk between light-stimulated and GAs-induced processes as well as the phenolic compounds metabolism leads to modifications in lignin formation and deposition on cell walls. How these factors (light and GAs) promote changes in lignin content and composition. In addition, structural changes were evaluated in the stem anatomy of tobacco plants. GA3 was sprayed onto the leaves and paclobutrazol (PAC), a GA biosynthesis inhibitor, via soil, at different irradiance levels. Fluorescence microscopy techniques were applied to detect lignin, and electron microscopy (SEM and TEM) was used to obtain details on cell wall structure. Furthermore, determination of total lignin and monomer contents were analyzed. Both light and GAs induces increased lignin content and CW thickening as well as greater number of fiber-like cells but not tracheary elements. The assays demonstrate that light exerts a role in lignification under GA3 supplementation. In addition, the existence of an exclusive response mechanism to light was detected, that GAs are not able to replace.

Abstract:Background: Moso bamboo (Phyllostachys edulis) is a well-known bamboo species of high economic value in the textile industry due to its rapid growth. Phytohormones, which are master regulators of growth and development, serve as important endogenous signals. However, the mechanisms through which phytohormones regulate growth in moso bamboo remain unknown to date. Results: Here, we reported that exogenous gibberellins (GA) applications resulted in a significantly increased internode length and lignin condensation. Transcriptome sequencing revealed that photosynthesis-related genes were enriched in the GA-repressed gene class, which was consistent with the decrease in leaf chlorophyll concentrations and the lower rate of photosynthesis following GA treatment. Exogenous GA applications on seedlings are relatively easy to perform, thus we used 4-week-old whole seedlings of bamboo for GA- treatment followed by high throughput sequencing. In this study, we identified 932 cis-nature antisense transcripts (cis-NATs), and 22,196 alternative splicing (AS) events in total. Among them, 42 cis-nature antisense transcripts (cis-NATs) and 442 AS events were differentially expressed upon exposure to exogenous GA3, suggesting that post-transcriptional regulation might be also involved in the GA3 response. Targets of differential expression of cis-NATs included genes involved in hormone receptor, photosynthesis and cell wall biogenesis. For example, LAC4 and its corresponding cis-NATs were GA3-induced, and may be involved in the accumulation of lignin, thus affecting cell wall composition. Conclusions: This study provides novel insights illustrating how GA alters post-transcriptional regulation and will shed light on the underlying mechanism of growth modulated by GA in moso bamboo.

Abstract:Secondary vascular tissue (SVT) development and regeneration are regulated by phytohormones. In this study, we used an in vitro SVT regeneration system to demonstrate that gibberellin (GA) treatment significantly promotes auxin-induced cambium reestablishment. Altering GA content by overexpressing or knocking down ent-kaurene synthase (KS) affected secondary growth and SVT regeneration in poplar. The poplar DELLA gene GIBBERELLIC ACID INSENSITIVE (PtoGAI) is expressed in a specific pattern during secondary growth and cambium regeneration after girdling. Overexpression of PtoGAI disrupted poplar growth and inhibited cambium regeneration, and the inhibition of cambium regeneration could be partially restored by GA application. Further analysis of the PtaDR5:GUS transgenic plants, the localization of PIN-FORMED 1 (PIN1) and the expression of auxin-related genes found that an additional GA treatment could enhance the auxin response as well as the expression of PIN1, which mediates auxin transport during SVT regeneration. Taken together, these findings suggest that GA promotes cambium regeneration by stimulating auxin signal transduction.
Effects of exogenously applied hormones on seedling elongation and content of endogenous GA3 in a dwarf mutatant (Brassica napus L. )

Abstract:Effects of exogenous gibberellin(GA3),auxin(IAA)and brassinosteroids(BRs) application on early seedling growth of a Brassica napus dwarf mutant,namely 'DDF-1',was studied.The results showed that proper exogenous GA3(7 mg/L)applied could partially rescued the hypocotyl elongation of the dwarf,but no significant differences made by IAA and BRs at any levels of application.Furthermore,no interaction effects of GA3,IAA and BR on the elongation of the seedling hypocotyl were detected by orthogonal test of these hormones.Vertical section of hypocotyls of the dwarf indicated a reduction for the cell length in comparison with the wild type,which may lead to the shorter hypocotyl of the mutant.Endogenous GA3 content, determined by ELISA(enzyme-linked immune sorbent assay),showed higher in the dwarf than that in the wild type,and lower than that in the exogenous GA3 treated dwarf hypocotyl.The hypothesis,suggesting that the dwarf mutant was GA insensitive and controlled by mutated gene(s) in gibberellins biosynthesis pathway,was discussed.
Regulatory mechanism of GA3 application on grape (Vitis vinifera L.) berry size

Abstract:Gibberellin A3 (GA3) is often used as a principal growth regulator to increase plant size. Here, we applied Tween-20 (2%)-formulated GA3 (T1:40 mg/L; T2:70 mg/L) by dipping the clusters at the initial expansion phase of 'Red Globe' grape (Vitis vinifera L.) in 2018 and 2019. Tween-20 (2%) was used as a control. The results showed that GA3 significantly increased fruit cell length, cell size, diameter, and volume. The hormone levels of auxin (IAA) and zeatin (ZT) were significantly increased at 2 h (0 d) -1 d after application (DAA0-1) and remained significantly higher at DAA1 until maturity. Conversely, ABA exhibited an opposite trend. The mRNA and non-coding sequencing results yielded 436 differentially expressed mRNA (DE_mRNAs), 79 DE_lncRNAs and 17 DE_miRNAs. These genes are linked to hormone pathways like cysteine and methionine metabolism (ko00270), glutathione metabolism (ko00480) and plant hormone signal transduction (ko04075). GA3 application reduced expression of insensitive dwarf 2 (GID2, VIT_07s0129g01000), small auxin-upregulated RNA (SAUR, VIT_08s0007g03120) and 1-aminocyclopropane-1-carboxylate synthase (ACS, VIT_18s0001g08520), but increased SAUR (VIT_04s0023g00560) expression. These four genes were predicted to be negatively regulated by vvi-miR156, vvi-miR172, vvi-miR396, and vvi-miR159, corresponding to specific lncRNAs. Therefore, miRNAs could affect grape size by regulating key genes GID2, ACS and SAUR. The R2R3 MYB family member VvRAX2 (VIT_08s0007g05030) was upregulated in response to GA3 application. Overexpression of VvRAX2 in tomato transgenic lines increased fruit size in contrast to the wild type. This study provides a basis and genetic resources for elucidating the novel role of ncRNAs in fruit development.

Abstract:Gibberellins (GAs), a class of phytohormones, act as an essential natural regulator of plant growth and development. Many studies have shown that GA is related to rhizobial infection and nodule organogenesis in legume species. However, thus far, GA metabolism and signaling components are largely unknown in the model legume Medicago truncatula. In this study, a genome-wide analysis of GA metabolism and signaling genes was carried out. In total 29 components, including 8 MtGA20ox genes, 2 MtGA3ox genes, 13 MtGA2ox genes, 3 MtGID1 genes, and 3 MtDELLA genes were identified in M. truncatula genome. Expression profiles revealed that most members of MtGAox, MtGID1, and MtDELLA showed tissue-specific expression patterns. In addition, the GA biosynthesis and deactivation genes displayed a feedback regulation on GA treatment, respectively. Yeast two-hybrid assays showed that all the three MtGID1s interacted with MtDELLA1 and MtDELLA2, suggesting that the MtGID1s are functional GA receptors. More importantly, M. truncatula exhibited increased plant height and biomass by ectopic expression of the MtGA20ox1, suggesting that enhanced GA response has the potential for forage improvement.

Abstract:We have cloned and expressed a mouse brain cDNA, mShal, that encodes a transient, A-type K+ current. mShal, the vertebrate homolog of the Drosophila Shal gene, defines a distinct subfamily of voltage-gated K+ channels. The Shal deduced proteins are more highly conserved between mouse and Drosophila than other presently known K+ channels. mShal carries a "low-threshold" A-type current with a hyperpolarized steady-state inactivation midpoint. Marked similarity was observed between mShal and its Drosophila homolog, fShal, with regard to voltage sensitivity of activation, macroscopic inactivation, steady-state inactivation, and 4-aminopyridine sensitivity. Sequence conservation for Shal proteins is unusually high at the amino terminus, an area considered important for inactivation. Removal of conserved amino-terminal residues from mShal modifies macroscopic inactivation but the transient nature of the current is preserved. Underlying the very high conservation of mShal and fShal may be a role in the nervous system that is conserved in widely divergent species.
Analysis of the Metabolic Pathways Affected by Poly(γ-glutamic Acid) in Arabidopsis Thaliana Based on GeneChip Microarray.

Abstract:Plant growth is promoted by poly(γ-glutamic acid) (γ-PGA). However, the molecular mechanism underlying such promotion is not yet well understood. Therefore, we used GeneChip microarrays to explore the effects of γ-PGA on gene transcription in Arabidopsis thaliana. Our results revealed 299 genes significantly regulated by γ-PGA. These differently expressed genes participate mainly in metabolic and cellular processes and in stimuli responses. The metabolic pathways linked to these differently expressed genes were also investigated. A total of 64 of the 299 differently expressed genes were shown to be directly involved in 24 pathways such as brassinosteroid biosynthesis, α-linolenic acid metabolism, phenylpropanoid biosynthesis, and nitrogen metabolism, all of which were influenced by γ-PGA. The analysis demonstrated that γ-PGA promoted nitrogen assimilation and biosynthesis of brassinosteroids, jasmonic acid, and lignins, providing a better explanation for why γ-PGA promotes growth and enhances stress tolerance in plants.
Gibberellins Play an Essential Role in the Bud Growth of Petunia hybrida

Abstract:This study delves into the role of gibberellin (GA) in governing plant branch development, a process that remains incompletely understood. Through a combination of exogenous hormone treatment, gene expression analysis, and transgenic phenotype investigations, the impact of GA on petunia's branch development was explored. The results showed that GA3 alone did not directly induce axillary bud germination. However, paclobutrazol (PAC), an inhibitor of GA synthesis, effectively inhibited bud growth. Interestingly, the simultaneous application of GA3 and 6-BA significantly promoted bud growth in both intact and decapitated plants compared to using 6-BA alone. Moreover, this study observed a significant downregulation of GA synthesis genes, including GA20ox1, GA20ox2, GA20ox3, GA3ox1, and CPS1, alongside an upregulation of GA degradation genes such as GA2ox2, GA2ox4, and GA2ox8. The expression of GA signal transduction gene GID1 and GA response factor RGA was found to be upregulated. Notably, the PhGID1 gene, spanning 1029 bp and encoding 342 amino acids, exhibited higher expression in buds and the lowest expression in leaves. The overexpression of PhGID1 in Arabidopsis resulted in a noteworthy rise in the number of branches. This study highlights the crucial role of GA in bud germination and growth and the positive regulatory function of GA signaling in shoot branching processes.
