Abstract:Seaweed draws a lot of attention for its vital role in aquaculture as it contains beneficial biological compounds that undoubtedly might help in the development of this field. The current study sheds light on the potential efficiency of dietary supplements of Grateloupia acuminata and G. doryphore (Halymeniaceae) nanoparticles (GNS) at different levels with bionanocomposite cellulose acetate membranes (CA/bio-AgNps) on improved growth performance, digestive enzyme activity, immunity, antioxidative, resistance against infectious pathogens, and characterization of water quality treated with CA/bio-AgNps that is used in rearing Nile tilapia ( Oreochromis niloticus ). Four concentrations (0.1, 0.25, 0.5, and 1.0 ml/L) of GNS extract were tested as potential anti-bacterial and for the efficacy of being parasitic. Fish with an average weight (24.46 ± 0. 50 g) were apportioned into six experimental groups (T0, T1, T2, T3, T4, and T5) represented as 0.0%, 0.0%, 0.1%, 0.25%, 0.5%, and 1.0% GNS in diets with CA/bio-AgNps, respectively. Injection of fish with Aeromonas hydrophila was performed at the end of the trial. Chemical and bacteriological water indices significantly showed improvement after being treated with CA/bio-AgNps than the control group. Growth, carcass composition, digestive enzyme, and hematological and biochemical indices were significantly noticed positive ( p< 0.05), especially T4 and T5, than the control group. In parallel, a significant improvement was noticed in serum lysozyme, total immunoglobulin, complement C3, antioxidative enzyme, and the relative expression of hepatic and inflammatory genes with an increased level of GNS ( p< 0.05) are upregulated than the control group. Remarkably, GNS-supplemented diets and extracts provided positive efficacy against A. hydrophila with a decreased percentage of fish mortality, besides efficacy on antibacterial strains and Cichlidogyrus tilapiae , respectively. To sum up, the seaweed extract with CA/bio-AgNps resulted in better growth performance of fish, antipathogenic effect, and health status. Furthermore, CA/bio-AgNps were vital in improving water characteristics. They should be studied and applied more in the future.

Abstract:Background: Mannanoligosaccharides (MOS) usage in fish production has drawn more attention because of their positive benefits on disease resistance and fish performance. Aim: The ongoing research was executed to assess the potential advantages of Bio-Mos® dietary supplementation regarding the growth outcomes, physiological response, oxidative biomarkers, and immunity-linked gene expression in Nile tilapia (Oreochromis niloticus) fingerlings exposed to bacterial infection with Aeromonas hydrophila. Methods: Four experimental diets were developed using a 30% protein baseline diet, with Bio-Mos® added at variable levels; 0.0, 0.5, 1, and 2 g/kg, respectively. 240 healthy Nile tilapia fingerlings were split into 4 groups at random and assigned to 12 glass aquariums (three replicates of 20 fish/treatment). Diets were admitted at a 3% rate of fish biomass/aquarium for 8 weeks. Following the feeding trial, fish from every treatment were intraperitoneally injected with pathogenic A. hydrophila, and then observed for 15 days to record the survival rate percent (SR%) post challenge. Results: Results revealed significant improvement in growth performance, physiological response, immunological parameters (phagocytic index, phagocytic activity, and lysozyme), and antioxidant parameters [catalase, malondialdehyde, glutathione peroxidase (GSH-Px), and superoxide dismutase (SOD)] among Bio-Mos® treated groups. Moreover, Bio-Mos® increased the expression of tumor necrosis factor alpha and Interleukin 1β, genes linked to the liver immune system. Growth-related genes (GHr), antioxidant-related genes (SOD and GSH-Px). In fish subjected to pathogens, dietary MOS supplementation could significantly lower oxidative stress, showing promise as a preventative supplement for Nile tilapia in place of antibiotics. On the other hand, Bio-Mos® considerably improved each of the three intestinal morphological measures (villus width, villus length, and crypt depth), showing the best overall intestinal structure-improving impact. The challenge with A. hydrophila caused marked degenerative alterations in the intestine, hepatopancreas, spleen, and posterior kidney of Nile tilapia, in the control group. However, lesion severity was greatly decreased and showed marked amelioration with an increased concentration of Bio-Mos®. The A. hydrophila-challenged groups revealed a 100% SR% mainly among the Bio-Mos® supplemented groups. Conclusion: It is recommended to enrich the Nile tilapia fingerlings diets with 2 g.kg-1 of MOS for better results on the growth rate, physiological response, immunological response, and intestinal absorptive capacity.

Abstract:Immunosaccharides such as β-glucans and fructooligosaccharide (FOS) strengthen the host's immune responses. This study examined the influences of three levels of the β-1, 3 glucan-FOS combination on Red tilapia performance. Four diets were prepared: T 0 as a control, while T 0.5 , T 1 , and T 1.5 indicate the addition of β-1, 3 glucan-FOS mixture at 0.5%, 1%, and 1.5%, respectively. Then, 240 red Tilapia ( Oreochromis niloticus x O. mossambicus ) (weight 6.1 ± 0.07gm) were allocated into four groups, and every group was fed tested diets for 56 days. After the feeding trial, growth parameters, erythrogram profile, liver and kidney function testes, glucose, histopathological analysis, and gene expressions for antioxidants, catalase (CAT) and glutathione peroxidase ( GPX ) besides growth hormone (GH) and insulin-like growth factor 1 IGF1-related markers were assessed. Red tilapia fed T 0.5 , T 1 , and T 1.5 exhibited consistent growth, survival rate, and homeostasis compared with the control group. Different supplement concentrations displayed varying levels of responses. The hepatorenal biomarkers (alanine transaminase (ALT = SGPT) and aspartate transaminase (AST = SGOT), alkaline phosphatase (ALP = ALK), urea, and creatinine) and glucose showed a significant reduction in the supplemented groups compared with the control, especially in the T 1 and T 1.5 groups. The intestinal morphometric study revealed that fish group fed on T 1 represented the best result, whereas group T 1.5 , followed by group T 0.5, was moderately treated, compared to the control. The real-time quantitative reverse transcription PCR (qRT-PCR) analysis displayed up-regulated expression of antioxidant and growth-correlated genes in the T 1.5 groups. After 56 days, the β-1,3 glucan-FOS fed groups also exhibited an increase in survival rates compared to the control when challenged with a pathogenic Vibrio alginolyticus. Current findings suggest that inclusion of β-1,3 glucan-FOS in diets could enhance red tilapia biochemical parameters, growth, and protection against pathogenic V. alginolyticus infection.

Abstract:This study aimed to evaluate the effect of adding liquid extract of algae (Hypnea musciformis, Grateloupia acuminata, and Sargassum muticum) (HGS) and Magnesium oxide nanoparticles (MgO NPs) using this extract to rear water of Oreochromis niloticus, on improving culture water indices, growth performance, digestive enzyme, hemato-biochemical characters, immune, antioxidative responses, and resistance after challenged by Aeromonas hydrophila with specific refer to the potential role of the mixture in vitro as resistance against three strains bacteria (Aeromonas sobria, Pseudomonas fluorescens, P. aeruginosa) and one parasite (Cichlidogyrus tilapia). The first group represented control, HGS0, whereas the other group, HGS5, HGS10, and HGS15 mL-1 of liquid extract, as well as all groups with 7.5 μg mL-1 MgO-NPs added to culture water of O. niloticus, for 60 days. Data showed that increasing levels at HGS 10 and HGS15 mL-1 in to-culture water significantly enhanced growth-stimulating digestive enzyme activity and a significantly improved survival rate of O. niloticus after being challenged with A. hydrophila than in the control group. The total viability, coliform, fecal coliform count, and heavy metal in muscle partially decreased at HGS 10 and HGS15 mL-1 than in the control group. Correspondingly, the highest positive effect on hemato-biochemical indices was noticed at levels HGS 10 and HGS15 mL-1. Fish noticed an improvement in immune and antioxidant indices compared to control groups partially at HGS 10 and HGS15 mL-1. Interestingly, fish cultured in rearing water with the mixture provided downregulated the related inflammatory genes (HSP70, TNF, IL-1β, and IL-8) partially at HGS15 mL-1. In vitro, the mixture showed positive efficiency as an antibacterial and partially antiparasitic at HGS 10 and HGS15 mL-1. This study proposes utilizing a mixture of (HGS) and (MgO-NPs) with optimum levels of 10-15 mL-1 in cultured water to improve water indices, growth, health status, and increased resistance of O. niloticus against bacterial and parasitic infection.

Abstract:Backgroung: The aim of palliative care services is to ensure the best quality of life for patients and their carers. Depression is common amongst palliative care patients and has been shown to adversely affect quality of life. This study aimed to examine the independent contribution of depression to quality of life. Objective: To investigate the hypothesis that a) illness severity, pain, anxiety and depression are all associated with impaired health-related quality of life and b) once the effects of illness severity have been adjusted for, there is further independent contribution to quality of life from anxiety and depression. Method:Consecutive patients with advanced cancer under the care of palliative care services were screened. Sixty-eight patients were evaluated for levels of anxiety and depression, severity of illness, pain severity and health-related quality of life. Results: Thirty-three males and 35 females were recruited and had an age range of 41-93 years (median 71). Seventeen (25%) of patients were anxious [anxiety score] = 11 on the Hospital Anxiety and Depression Scale (HADS)], 15 (22%) were depressed (HADS depression score] = 11). After controlling for the effects of pain and illness severity, anxiety and depression were independently associated with global health status, emotional and cognitive functioning, and fatigue. Anxiety further contributed significantly towards social functioning, nausea and vomiting. Conclusions: This study has confirmed that pain, anxiety and depression were associated with impaired quality of life. Anxiety and depression contributed independently towards various dimensions of quality of life. Longitudinal studies are required to examine the direction of the causal association between pain and depression in patients receiving palliative care.
Dietary biogenic selenium nanoparticles improve growth and immune-antioxidant indices without inducing inflammatory responses in Nile tilapia

Abstract:The present study evaluated the use of green-synthesized selenium nanoparticles (SeNPs), using the microalgae Pediastrum boryanum as a diet additive in aquaculture to improve the growth performance, health, and immune response of Nile tilapia. Nile tilapia were fed different concentrations of green SeNPs (79.26 nm) as follows: 0, 0.75, and 1.5 mg/kg of SeNPs for 8 weeks. Following the trial, growth performance, biochemical indices, antioxidant and pro-inflammatory cytokine-related genes, and tissue histological examinations were performed. The study showed that SeNPs significantly improved ( P < 0.05) growth performance and innate immune parameters ( P < 0.001, IgM, and lysozyme) at both supplemented doses compared with the control. The protein profile and liver function enzymes were normal compared with those in the control group ( P > 0.05). Serum malondialdehyde and superoxide dismutase levels were not significantly changed, while reduced glutathione and catalase were significantly enhanced ( P < 0.01, P < 0.05) in the SeNPs 1.5 mg/kg compared to the control group. No inflammatory response was detected upon SeNP supplementation, as indicated by the absence of changes in the expression of pro-inflammatory cytokine genes. The earlier assays' results were histopathologically evidenced, where hepatic and splenic tissue architectures in SeNPs groups did not reveal any deviation from the control group. Our findings indicate that green selenium nanoparticles can potentially improve the growth and immunological response of Nile tilapia, offering opportunities for incorporating health benefits into functional foods and nutraceuticals, which corresponds to the increasing consumer interest in eco-friendly, environmentally sustainable dietary supplements.

Abstract:The objective of this study was to evaluate the effect of graded levels (0.0 , 2.5, 5.0 and 7.5g /kg diet ) of the dietary blue-green microalgae (Spirulina) Arthrospira platensis as feed supplement to assess growth performance , nutrient utilization, immune response , some hematological parameters, and proximate composition of the Nile tilapia (O. niloticus) . A completely randomized experimental design was developed with 4 treatments and two replicates . Fish (80 all-male Nile tilapia fingerlings) ,with an average initial weight of 6±0.20 g , were randomly distributed into 8 glass aquaria (112 liter) and each aquarium holding 10 fish and randomly assigned to one of two replicates of the diets and offered feed to satiation to fingerlings O. niloticus. Fish were fed a balanced diet of 30% protein along the period of the experiment. The treatments were Diet 1 (Control) without Spirulina, Diet 2 (2.5g Spirulina /kg diet), Diet 3 (5g Spirulina /kg diet) and Diet 4 (7.5 g Spirulina /kg diet ). The results indicate that, the fish groups received Spirulina supplemental diets revealed significant improvement in growth parameters (body weight gain, feed conversion ratio and protein efficiency ratio). Since the diets contained microalga Spirulina at a level (7.5 gram/kg diet) showed the highest values of growth parameters and protein utilization. Carcass composition of the experimental fish was relatively affected by different dietary treatments. This study suggested that Spirulina at a level of 7.5 gram/kg can be used in the feed of Nile tilapia without negative effect on growth parameters, feed utilization and blood parameters.
Effects of Sophora flavescens dietary supplementation on the growth performance, haematological indices, and intestinal bacterial community of Oreochromis niloticus

Abstract:Sophora flavescens is considered to be of various medicinal and economic importance. Previous studies have shown that it has antibacterial, antioxidant, and immune-enhancing functions. To explore the full potential of S. flavescens in fish feed development, we investigated the effects of different levels of S. flavescens added to the diet on the growth performance, haematological indices, and the intestinal bacterial community of Nile tilapia (Oreochromis niloticus). The feeding trial lasted for 56 days, and the initial weight of the fish was 40.00 ± 5.00 g. Nile tilapia were randomly divided into six groups: a control group (CN) and five S. flavescens supplementation groups (0.02%, 0.04%, 0.08%, 0.16%, and 0.3% [w/w] of S. flavescens in diet). The growth performance of fish increased first and then decreased with the increase of S. flavescens supplemental level. Compared with other experimental groups, the growth performance of fish supplemented with 0.08% S. flavescens was significantly improved (p < 0.05). Haematological indices exhibited that erythrocyte (RBC), along with leucocyte (WBC) indices, exhibited a secondary trend of increasing and then decreasing with the increase of S. flavescens, reaching the highest level at 0.08% (p < 0.05). However, mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH), and mean corpuscular haemoglobin concentration (MCHC) exhibited a secondary trend of first decreasing and then increasing with the increase of S. flavescens and reached the lowest value at 0.08% (p < 0.05). Compared with the CN, alkaline phosphatase (AKP) significantly decreased and changed when the additional amount of S. flavescens was 0.04% in each treatment group (p < 0.05). In another way, the gut microbial profiles revealed that Bacteroides dominated the gut communities, and compared with the control group, two uncultured bacteria were suppressed. In addition, when the supplemental level of S. flavescens was more significant than 0.04%, the proliferation of Arcobacter cryaerophilus, Solibacillus silvestris, and Escherichia sp. in the gut of Nile tilapia was promoted. The results revealed that S. flavescens, as an additive to the Nile tilapia diet with levels ranging from 0.04% to 0.08%, can enhance the growth performance and immunity and promote the proliferation of intestinal beneficial bacteria of Nile tilapia.

Abstract:Functional trace elements and vitamins can boost immunity and anti-oxidative response in aquatic animals with effects on nutritional physiology. Nano-selenium (nano-Se) and vitamin C (VC) have been used as immunomodulators and antioxidants in fish feed. The present work was performed to determine the protective effects of diets supplemented with different combinations of nano-Se and VC on Nile tilapia (Oreochromis niloticus). Triplicate groups of 20 fish/tank (13.87 ± 0.10 g) were reared and fed with basal diet (control-T1) (without supplementation of nano-Se and VC) and three experimental diets as T2, T3, and T4 (100, 200, and 300 mg/kg VC respectively) with a pre-determined dose of nano-Se (1.0 mg/kg) for 90 days. Different immune indices, haemato-biochemical, and antioxidant activities were measured at the end of the first, second, and third months of feeding. The findings depicted that significantly (p < 0.05) higher growth was observed in T4. Red blood cells, white blood cells, and haemoglobin were found significantly (p < 0.05) higher in T4 for the third month. Serum biochemical-immunological indices (alkaline phosphatase, glucose, cholesterol, lysozyme, myeloperoxidase, total protein, albumin and globulin) followed the same trend. Furthermore, antioxidant assays such as catalase, superoxide dismutase, glutathione peroxidase, glutathione S-transferase, and malondialdehyde were significantly (p < 0.05) improved in T4 for the third month. Significantly (p < 0.05) least cumulative mortality against Aeromonas hydrophila was obtained in the fish-fed diets incorporated with nano-Se and VC. Therefore, dietary supplementation with nano-Se and VC is noteworthy for improving growth, serum biochemical status, immune response, antioxidant status, and disease resistance.

Abstract:Background: Identifying alternative sustainable feed sources with high nutritional values is crucial for the future of environmentally and socially responsible aquaculture. In this regard, microalgae have been proven to have positive effects on fish health, which overwhelmed our interest in this study. Methods: Pediastrum boryanum (P. boryanum) was incorporated into Nile tilapia feed at concentrations of 0, 0.75, and 1.5 mg/kg, as control, PbExt0.75, and PbExt1.5 groups to assess its effects on growth and biochemical indices, oxidant/antioxidant activities, immune and stress-related gene expression, and intestinal morphology. Results: After 8 weeks, fish fed P. boryanum supplemented feed exhibited significant increases in final weight, length, condition factor, body weight gain, and specific growth rate, while the spleen-somatic index (SSI) and hepatosomatic index (HSI) showed no significant differences compared to the control group. Dietary P. boryanum supplementation also enhanced IgM levels and lysozyme activity, along with no marked effect on markers of liver function enzymes (alanine aminotransferase/ALT and aspartate aminotransferase/AST) or protein status (total protein and albumin). Furthermore, P. boryanum addition increased the activity of superoxide dismutase (SOD), catalase (CAT), and reduced glutathione (GSH) enzymes, highlighting its antioxidant potential, whereas malondialdehyde (MDA) concentrations showed no significant differences among the groups. Gene expression analysis revealed that tumor necrosis factor-α (TNF-α), interleukin-10 (IL-10), and transforming growth factor-β1 (TGF-β1) expression notably increased in groups fed P. boryanum containing feed, while no significant difference was observed in hepatic Heat Shock Protein 70 (HSP70) mRNA expression. Histopathological examination revealed no adverse effects of P. boryanum supplementation on the liver, spleen, or intestinal tissues. Villous height and villous surface area were notably increased in the high P. boryanum supplementation group, suggesting improved intestinal integrity and nutrient absorption. Conclusion: Dietary P. boryanum supplementation can potentially improve growth performance, immune response, antioxidant status, and intestinal health of Nile tilapia, making it a promising candidate for sustainable aquaculture.

Abstract:The present study was conducted to investigate the effects of nano-selenium (Nano Se) or/and vitamin E (VE) on growth performance, blood health, intestinal histomorphology, oxidative status, and immune-related gene expression of Nile tilapia. Nano Se or/and VE at a rate of 0, 1 mg Nano Se/kg, 100 mg VE/kg, and 1 mg Nano Se/kg + 100 mg VE diet were fed to fish for 8 weeks. FBW was significantly (P < 0.05) increased in fish fed with Nano Se and VE, while fish fed with Nano Se or Nano Se and VE diets displayed significantly (P < 0.05) higher WG and SGR than the other groups. The lowest FCR was significantly (P < 0.05) detected in fish fed with Nano Se and VE, while the highest value was observed in fish VE diet. The intestinal morphometry (villi length and width) of fish fed with Nano Se or/and VE reported significantly (P < 0.05) the highest values with high number of goblet cells. Blood hematology and biochemistry parameters of fish fed with Nano Se or/and VE showed normal values with insignificant differences except for the blood total protein increased in fish fed with Nano Se or/and VE (P < 0.05). Dietary Nano Se or Nano Se and VE significantly (P < 0.05) increased the GPX, SOD, CAT, NBT, lysozyme, and phagocytosis values with decreased MDA. Liver and spleen TNF-α and IL-1β expressions were significantly (P < 0.05) upregulated in fish fed on Nano Se or Nano Se and VE. Thus, Nano Se or/and VE can be used effectively in tilapia diets for improving the growth, intestinal health, blood health, oxidative status, and immune-related gene expression.

Abstract:Plant extracts are a phytochemically-rich alternative to antibiotic and synthetic feed additives, with high systemic bioactivity in animals. The present study aimed to evaluate the effect of a hydroalcoholic extract of custard apple (Annona squamosa) leaf (ASLE) on the growth, hematobiochemical parameters, digestive enzyme activities, redox status, nonspecific immune response, and cold and bacterial infection tolerance in Nile tilapia (Oreochromis niloticus). A total of 300 Nile tilapia fingerlings (11.87 ± 0.48 g) were fed ASLE-supplemented diets at increasing levels of 0, 5, 10, 15, and 20 g/kg for 60 days. At the end of the feeding period, the fish were experimentally challenged with cold water stress or Aeromonas sobria, and mortalities were recorded for 10 days. The results revealed that the growth performance and feed conversion ratio were significantly improved with an increasing level of ASLE supplementation. The hematologic profile and hepato-renal functions were retained within a healthy range in the various groups supplemented with an ASLE diet. Antioxidant status was significantly improved in the serum of fish fed ASLE-supplemented diets, in terms of superoxide dismutase (SOD), catalase (CAT) activities, reduced glutathione, and total antioxidant capacity. Meanwhile, the myeloperoxidase (MPO) and malondialdehyde (MDA) levels decreased significantly. Similarly, there was a noticeable improvement in the hepatic CAT and SOD activities and a reduction of hepatic MDA. Marked improvements in lysozyme activity, nitric oxide production, complement3 level, and phagocytic activity were recorded in groups fed ASLE-supplemented diets, which peaked with the 20 g ASLE/kg diet. Moreover, the serum glucose and cortisol levels significantly declined in groups fed ASLE at levels of 15–20 g/kg compared to the other groups. Supplementation with ASLE increased the activities of protease, lipase, and α-amylase. ASLE supplementation at a concentration of 10–20 g/kg diet enhanced the resistance of Nile tilapia to A. sobria infection. According to this study, ASLE supplementation enhanced the antioxidant balance, non-specific immune response, physiological status, resistance against infection, and growth performance of Nile tilapia at supplementation levels of 10–20 g/kg diet.

Abstract:This investigation attempts to evaluate the effect of diet additives via aqueous or ethanolic herbal extracts from Azadirachta indica leaves on Nile tilapia, Oreochromis niloticus. Five dietary categories were assigned to the fish: the first category (N1, with no extract) was kept under control conditions; two categories contained aqueous extract (N2 (1.0 g/kg) and N3 (2.0 g/kg); and two categories contained ethanolic extract, N4 (1.0 g/kg) and N5 (2.0 g/kg), with each group being fed for 60 days. After the feeding trial, Aeromonas hydrophila was injected intraperitoneally into fish for 14 days; fish mortality was recorded during this period. The results showed that the fish-fed dietary A. indica significantly improved growth performance and intestinal health (digestive enzymes and intestinal morphology), especially in the N4 and N5 categories. However, N4 and N5 categories demonstrated a significant decrease in AST and ALT activities and an increase in total protein, serum albumin, globulin, growth hormone (GH), leptin hormone (LEP), hemoglobin, white blood cells, and hematocrit (P < 0.05) in comparison with the control category (N1). Compared to the control category, the N4 and N5 categories have revealed a significant reduction in MDA activity and improvements in immunological activities (lysozyme, complement C3, and nitric oxide) and antioxidant enzymes (CAT, SOD, and GPX). Moreover, in tilapia-fed A. indica, the expression of IL-8, IL-1β, and Nf-κb genes was downregulated partially in the N4 and N5 categories than the control category. In contrast, the lysozyme, C3, GPX, and CAT genes were upregulated partially at N4 and N5 compared to the control category. Following the bacterial challenge, fish in the N4 and N5 categories also displayed the lowest fish mortality compared to the control category. The ethanolic extract displayed a more potent resistance against the parasite Cichlidogyrus tilapia in vitro than the aqueous and control categories, partially at 2 g/L. According to these findings, an ethanolic neem extract (2.0 g/kg feed) activates the immune system and antioxidant response in Nile tilapia fingerlings, improving growth and fish resistance to parasitic and bacterial infections.

Abstract:In recent times, nutraceuticals have been used extensively to identify promising feed additives for the improvement of the aquaculture industry through the enhancement of growth and survival rates, potentiation of the immune responses, and fortification of the resistance against infectious bacterial diseases. In this study, Nile tilapia (Oreochromis niloticus) were fed with diets supplemented with quinoa seeds (QU) or prickly pear fruit peel (PP) at the dose levels of 10% or 20% of the diet. After 45 days of the feeding trial, the fish were exposed to Aeromonas sobria (A. sobria) challenge. The pre-challenge indices indicated that both supplements mediated a significant improvement in most of the estimated parameters, including survival rate, antioxidant status, hematological and immunological indices, and hepatoprotective potential. These effects were recorded in the groups fed with high doses of the supplements (20%). The least changes were observed in the QU10-supplemented fish. In the spleen tissue, the TGF-β gene was upregulated in the PP10-, PP20- and QU20-supplemented groups, while the expression of the IFN-γ gene remained unaffected in all the supplemented groups, except for the PP20-supplemented group, which showed an upregulation. After the challenge with A. sobria, the relative survival percentage was improved by the supplementation of PP and QU, particularly in the PP20-supplemented group, possibly via the promotion of immunological responses, hepatoprotective potency, and modulation of the studied genes. Moreover, the morphological structure of the tissues showed marked recovery. The findings suggest that Nile tilapia fed with different levels of PP peel and QU seeds, particularly at the level of 20%, enhanced the immune response in fish and improved their resistance against A. sobria infection.

Abstract:A rice potassium transporter contributes to salt tolerance and is regulated by a transcription factor. Sodium transporters play key roles in plant tolerance to salt stress. Here, we report that a member of the High-Affinity K+ Transporter (HKT) family, OsHKT1;1, in rice (Oryza sativa ‘Nipponbare’) plays an important role in reducing Na+ accumulation in shoots to cope with salt stress. The oshkt1;1 mutant plants displayed hypersensitivity to salt stress. They contained less Na+ in the phloem sap and accumulated more Na+ in the shoots compared with the wild type. OsHKT1;1 was expressed mainly in the phloem of leaf blades and up-regulated in response to salt stress. Using a yeast one-hybrid approach, a novel MYB coiled-coil type transcription factor, OsMYBc, was found to bind to the OsHKT1;1 promoter. In vivo chromatin immunoprecipitation and in vitro electrophoresis mobility shift assays demonstrated that OsMYBc binds to AAANATNC(C/T) fragments within the OsHKT1;1 promoter. Mutation of the OsMYBc-binding nucleotides resulted in a decrease in promoter activity of OsHKT1;1. Knockout of OsMYBc resulted in a reduction in NaCl-induced expression of OsHKT1;1 and salt sensitivity. Taken together, these results suggest that OsHKT1;1 has a role in controlling Na+ concentration and preventing sodium toxicity in leaf blades and is regulated by the OsMYBc transcription factor.
Effects of dietary white mulberry leaves on hemato-biochemical alterations, immunosuppression and oxidative stress induced by Aeromonas hydrophila in Oreochromis niloticus

Abstract:The present work was designed to assess the potential hemato-biochemical protective action, immunemodulatory and antioxidant conclusions of varied concentration of white mulberry Morus alba leaves (MAL) extract supplementation on Nile tilapia (Oreochromis .niloticus). A total two hundred and forty of O. niloticus were haphazardly sorted into four groups. The control (CT) group was fed on basal diet. A group MAL1, MAL3 and MAL5 was fed on 1, 3 and 5 g/kg MAL respectively for thirty days. On day thirty one, half of replicates in each group were challenged by 0.5 ml × 108Aeromonas hydrophila where, the residual replicates were kept without challenge. A. hydrophila challenged tilapias revealed anemia that alleviated by supplementation with 5 g/kg MAL also, recovers the shift of leucogram prompted by the challenge. Elevation of alkaline phosphatase, aminotransferases, lactate dehydrogenase and malondialdehyde (ALP, ALT, AST, LDH and MDA) in CT, MAL1 and MAL3 in the challenged replicates respectively where within normal at MAL5. Supplementation with MAL5 showed more potent antioxidant and immune reaction than MAL1 and MAL3. There were a rapid increase of immunoglobulin M, lysozymes, nitric oxide, catalase and superoxide dismutase and their allied genes expression (IgM, CAT and SOD) in MAL groups with contrast in CT challenged groups. Where in challenged groups, there was suppression in genes expression of interleukins (8 and 1 beta) and interferon ɤ (IL8. IL-1β and INFɤ). Tilapias challenged by A. hydrophila unveiled plentiful surge in the percentage of mortality in CT challenged fish (80%), followed by the groups supplemented with MAL1 and MAL3 were (73.33%) where MAL5 was 20%. The mortalities have been halted from the 6th, 13th, 14th and 15th days in, MAL5, MAL3, MAL1, and CT correspondingly. These previous results could be fulfilled that using of MAL 5 g/kg protect tilapias from hemato-biochemical alterations and enhance its immune feedback, antioxidant defense and resistance against A. hydrophila.
Using Azadirachta indica protein hydrolysate as a plant protein in Nile tilapia (Oreochromis niloticus) diet: Effects on the growth, economic efficiency, antioxidant-immune response and resistance to Streptococcus agalactiae

Abstract:A feeding trial for 90 days was conducted on Nile tilapia (Oreochromis niloticus) (average weight: 25.50 ± 0.05 g) to evaluate the effect of dietary inclusion of Azadirachta indica seed protein hydrolysate (AIPH). The evaluation included the impact on the growth metrics, economic efficiency, antioxidant potential, hemato-biochemical indices, immune response, and histological architectures. A total of 250 fish were randomly distributed in five treatments (n = 50) and received diets included with five levels of AIPH (%): 0 (control diet, AIPH0), 2 (AIPH2), 4 (AIPH4), 6 (AIPH6) or 8 (AIPH8), where AIPH partially replace fish meal by 0, 8.7%, 17.4%, 26.1%, and 34.8%, respectively. After the feeding trial, a pathogenic bacterium (Streptococcus agalactiae, 1.5 × 108 CFU/mL) was intraperitoneally injected into the fish and the survival rate was recorded. The results elucidated that AIPH-included diets significantly (p < 0.05) enhanced the growth indices (final body weight, total feed intake, total body weight gain, and specific growth rate) and intestinal morpho-metrics (villous width, length, muscular coat thickness, and goblet cells count) in comparison to the control diet, with the AIPH8 diet recording the highest values. Dietary AIPH inclusion significantly improved (p < 0.05) the economic efficacy indicated by reduced feed cost/kg gain and increased performance index. The fish fed on the AIPH diets had noticeably significantly higher (p < 0.05) protein profile variables (total proteins and globulin) and antioxidant capabilities (superoxide dismutase and total antioxidant capacity) than the AIPH0 group. The dietary inclusion of AIPH significantly (p < 0.05) boosted the haematological parameters (haemoglobin, packed cell volume %, and counts of red blood cells and white blood cells) and immune indices (serum bactericidal activity %, antiprotease activity, and immunoglobulin M level) in a concentration-dependent manner. The blood glucose and malondialdehyde levels were significantly (p < 0.05) lowered by dietary AIPH (2%-8%). The albumin level and hepatorenal functioning parameters (aspartate aminotransferase, alanine aminotransferase, and creatinine) were not significantly (p > 0.05) altered by AIPH diets. Additionally, AIPH diets did not adversely alter the histology of the hepatic, renal or splenic tissues with moderately activated melano-macrophage centres. The mortality rate among S. agalactiae-infected fish declined as dietary AIPH levels rose, where the highest survival rate (86.67%) was found in the AIPH8 group (p < 0.05). Based on the broken line regression model, our study suggests using dietary AIPH at the optimal level of 6%. Overall, dietary AIPH inclusion enhanced the growth rate, economic efficiency, health status, and resistance of Nile tilapia to the S. agalactiae challenge. These beneficial impacts can help the aquaculture sector to be more sustainable.

Abstract:Protein kinase CK2, a protein serine/threonine kinase, plays a global role in activities related to cell growth, cell death, and cell survival. CK2 has a large number of potential substrates localized in diverse locations in the cell including, for example, NF‐κB as an important downstream target of the kinase. In addition to its involvement in cell growth and proliferation it is also a potent suppressor of apoptosis, raising its key importance in cancer cell phenotype. CK2 interacts with diverse pathways which illustrates the breadth of its impact on the cellular machinery of both cell growth and cell death giving it the status of a “master regulator” in the cell. With respect to cancer, CK2 has been found to be dysregulated in all cancers examined demonstrating increased protein expression levels and nuclear localization in cancer cells compared with their normal counterparts. We originally proposed CK2 as a potentially important target for cancer therapy. Given the ubiquitous and essential for cell survival nature of the kinase, an important consideration would be to target it specifically in cancer cells while sparing normal cells. Towards that end, our design of a tenascin based sub‐50 nm (i.e., less than 50 nm size) nanocapsule in which an anti‐CK2 therapeutic agent can be packaged is highly promising because this formulation can specifically deliver the cargo intracellularly to the cancer cells in vivo. Thus, appropriate strategies to target CK2 especially by molecular approaches may lead to a highly feasible and effective approach to eradication of a given cancer.

Abstract:Search for novel best-performing starter feed ingredients is one of the main challenges of modern aquaculture industry. The goal of this article is to evaluate the effect of microalgae supplementation enriched with omega-3 polyunsaturated eicosapentaenoic acid and the powerful antioxidant fucoxanthin on growth performance of Red tilapia fry ( Oreochromis mossambicus × Oreochromis niloticus ). We formulated three experimental diets, using two strains of microalgae from our culture collection – Vischeria magna SBV-108 (Eustigmatophyceae), rich in eicosapentaenoic acid and Mallomonas furtiva SBV-13 (Chrysophyceae), rich in fucoxanthin – and their combination. Vischeria magna SBV-108 is a new strain which biochemical properties and growth characteristics have not been previously studied. Coppens Supreme-15 was used as a control diet. The best growth performance was recorded in eicosapentainoic acid enriched Diet 1 (10% w/w V. magna ), that significantly (P<0.05) overperformed control feed on absolute growth, average growth, specific growth rate, feed conversion rate, protein efficiency ratio, protein productive value and energy productive value. Diet 3 (5% w/w V. magna and 5% w/w M. furtiva ) and Diet 2 (10% w/w M. furtiva ) were not significantly different from control at most parameters. Generally, supplementation of feed with microalgae rich in omega-3 eicosapentaenoic acid significantly improved Red tilapia fry growth parameters and overall feed performance, while supplementation of feed with microalgae rich in antioxidant fucoxanthin was not beneficial compared to the control.
