Abstract:Abstract Enzyme‐driven micro/nanomotors (MNMs) have demonstrated potentials in the biomedical field because of their excellent biocompatibility, versatility, and fuel bioavailability. However, the fragility of enzymes limits their practical application, because of their susceptibility to denaturation and degradation in realistic scenarios. Herein, a simple yet versatile and effective approach is reported to preserve the enzymatic activity and propulsion capability of enzymatic MNMs under various harsh conditions using metal organic frameworks (MOFs) as a protective shell. Urease can be encapsulated within the exoskeleton of zeolitic imidazolate framework‐8 (ZIF‐8) via biomimetic mineralization to form ZIF‐8@urease (ZU‐I) nanomotors that exhibit self‐propulsion in the presence of urea. When exposed to harsh conditions, including high temperature, presence of proteases, and organic solvents, the ZU‐I nanomotors still maintained their activity and mobility, whereas ZIF‐8 with externally modified urease (ZU‐O) nanomotors with externally modified urease as a control rapidly lost their motion capabilities owing to the inactivation of urease. Furthermore, ZU‐I nanomotors exhibit effectively enhanced diffusion within the small intestine fluid, achieving a fourfold higher mucus penetration than the ZU‐O nanomotors. The results highlight the effectiveness of using MOFs as protective shells for enzyme nano‐engines, which can greatly advance the practical applications of enzymatic MNMs under realistic conditions, especially for biomedical purpose.

Chemically Powered Nanomotors with Magnetically Responsive Function for Targeted Delivery of Exosomes

Abstract:A self‐assembly technique is employed to modify the surfaces of magnetic silica (SiO2@Fe3O4) nanoparticles with platinum (Pt) nanoparticles, resulting in the formation of Janus‐structured chemically powered nanomotors with magnetically responsive function. The catalase‐like activity of nanomotors is more than twice that of conventional nanomotors using Pt film. Then, the nanomotors are utilized to deliver exosomes, greatly enhancing the targetability to tumor cells. Janus structure plays a crucial role in achieving chemically driven nanomotors with exceptional motion performance. However, Janus‐structured chemically driven nanomotors with magnetic responsiveness are commonly fabricated by sputtering metal films. In the study, a self‐assembly technique is employed to asymmetrically modify the surfaces of magnetic silica (SiO2@Fe3O4) nanoparticles with platinum nanoparticles, resulting in the formation of this kind nanomotors. Compared to platinum film, platinum nanoparticles exhibit a larger surface area and a higher catalytic activity. Hence, the nanomotors demonstrate improved diffusion capabilities at a significantly lower concentration (0.05%) of hydrogen peroxide (H2O2). Meanwhile, exosomes have gained attention as a potential tool for the efficient delivery of biological therapeutic drugs due to their biocompatibility. However, the clinical applications of exosomes are limited by their restricted tropism. The previously obtained nanomotors are utilized to deliver exosomes, greatly enhancing its targetability. The drug doxorubicin (DOX) is subsequently encapsulated within exosomes, acting as a representative drug model. Under the conditions of H2O2 concentration at the tumor site, the exosomes exhibited a significantly enhanced rate of entry into the breast cancer cells. The utilization of the nanomotors for exosomes presents a novel approach in the development of hybrid chemically and magnetically responsive nanomotors.

Abstract:Enzyme-powered micro- and nanomotors make use of biocatalysis to self-propel in aqueous media and hold immense promise for active and targeted drug delivery. Most (if not all) of these micro- and nanomotors described to date are fabricated using a commercially available enzyme, despite claims that some commercial preparations may not have a sufficiently high degree of purity for downstream applications. In this study, the purity of a commercial urease, an enzyme frequently used to power the motion of micro- and nanomotors, was evaluated and found to be impure. After separating the hexameric urease from the protein impurities by size-exclusion chromatography, the hexameric urease was subsequently characterized and used to functionalize hollow silica microcapsules. Micromotors loaded with purified urease were found to be 2.5 times more motile than the same micromotors loaded with unpurified urease, reaching average speeds of 5.5 μm/s. After comparing a number of parameters, such as enzyme distribution, protein loading, and motor reusability, between micromotors functionalized with purified vs unpurified urease, it was concluded that protein purification was essential for optimal performance of the enzyme-powered micromotor.

Abstract:Self-propelled micro/nanomotors (MNMs) have shown great application potential in biomedicine, sensing, environmental remediation, etc. In the past decade, various strategies or technologies have been used to prepare and functionalize MNMs. However, the current preparation strategies of the MNMs were mainly following the pre-designed methods based on specific tasks to introduce expected functional parts on the various micro/nanocarriers, which lacks a universal platform and common features, making it difficult to apply to different application scenarios. Here, we have developed a modular assembly strategy based on host-guest chemistry, which enables the on-demand construction of imaging-trackable nanomotors mounted with suitable driving and imaging modules using a universal nanocarrier platform, according to different application scenarios. These assembled nanomotors exhibit ed enhanced diffusion behavior driven by enzymatic reactions. The loaded imaging functions were used to dynamically trace the swarm motion behavior of assembled nanomotors with corresponding fuel conditions both in vitro and in vivo. The modular assembly strategy endowed with host-guest interaction provides a universal approach to producing multifunctional MNMs in a facile and controllable manner, which paves the way for the future development of MNMs systems with programmable functions.

Abstract:Micro/nanomotors (MNMs) are miniaturized machines that can convert many kinds of energy into mechanical motion. Over the past decades, a variety of driving mechanisms have been developed, which have greatly extended the application scenarios of MNMs. Enzymes exist in natural organisms which can convert chemical energy into mechanical force. It is an innovative attempt to utilize enzymes as biocatalyst providing driving force for MNMs. The fuels for enzymatic reactions are biofriendly as compared to traditional counterparts, which makes enzyme-powered micro/nanomotors (EMNMs) of great value in biomedical field for their nature of biocompatibility. Until now, EMNMs with various shapes can be propelled by catalase, urease and many others. Also, they can be endowed with multiple functionalities to accomplish on-demand tasks. Herein, combined with the development process of EMNMs, we are committed to present a comprehensive understanding of EMNMs, including their types, propelling principles, and potential applications. In this review, we will introduce single enzyme that can be used as motor, enzyme powered molecule motors and other micro/nano-architectures. The fundamental mechanism of energy conversion process of EMNMs and crucial factors that affect their movement behavior will be discussed. The current progress of proof-of-concept applications of EMNMs will also be elaborated in detail. At last, we will summarize and prospect the opportunities and challenges that EMNMs will face in their future development.

Abstract:Micro/nanomotors are newly developed micro/nanoscale autonomous materials, and have been gradually applied in many fields such as bioanalysis and drug delivery owing to the advantages of simple preparation and mass production. By template-based electrodeposition and mechanical etching, micro/nanomotors in different shapes and compositions have been prepared. The most developed micro/nanomotors are asymmetric nanowire and microsphere and conical tube based motors. The motor based micro/nano biosensors can be prepared easily by functionalizing the motors with specific bioreceptors. Due to the complexity of the biological sample composition, traditional detection methods are often time-consuming and complicated because of multi cleaning and separation steps. However, benefiting from the unique property of autonomic movement, the motor based micro/nano biosensors can recognize, isolate and enrich target biomolecules from untreated sample solutions autonomously, hence realize the rapid, sensitive and in situ detection of target biomolecules such as DNA, protein, cell and so on. Generally, the micro/nanomotors can be propelled by chemical and field energies, for example, chemical reactions, magnetic field and ultrasound. In this review, we briefly introduce the motion mechanism of different micro/nanomotors, summarize the recent advances of micro/nanomotor-based biosensing applications in protein, DNA and cell detection and drug delivery, and prospect the future development of artificial micro/nanomotors.
Surface Charge-Reversible Tubular Micromotors for Extraction of Nucleic Acids in Microsystems

Abstract:Extraction of nucleic acids in microsystems is of significance for biomedical applications, but the current extraction methods generally require sophisticated microchannels and external equipment, hindering their practical applications. In this work, we have demonstrated a simple, versatile and efficient approach to extract nucleic acids in microsystems by developing cationic branched polyethyleneimine (PEI)-functionalized tubular micromotors. The as-developed tubular micromotors are fabricated by a two-step process combining the template-assisted electrodeposition and carbodiimide chemistry, and contain an inner catalytic Pt layer, a middle magnetic Ni layer and an outer cationic PEI layer. They exhibit autonomous bubble-propelled motion in aqueous hydrogen peroxide solutions, which can be guided by an external magnetic field, and the surface charges can be reversibly modulated by changing the pH value of the solution. Consequently, the as-developed tubular micromotors can selectively absorb nucleic acids from acidic solutions and desorb them into alkaline solutions, leading to the extraction of nucleic acids with high efficiency without external stirring. Furthermore, they can be operated in a microchannel chip without the aid of a pumping system. Our results indicate that this PEI-functionalized tubular micromotor platform provides a novel, simple and versatile microsystem nucleic acid extraction technology, holding considerable promise for important practical applications.

Abstract:Efficient capture and transport of biological targets by functionalized micromotors in microfluidic chips have emerged as to be promising for bioanalysis and detection of targets. However, the crucial step-target capture-is still inefficient due to the low utilization of active spots on the functionalized motor surfaces. Herein, we designed a multichannel microchip for integrating confined space with the oscillatory movement of micromotors to increase the capture efficiency. Acoustically driven, magnetically guided Au/Ni/Au micromotors were employed as the target carriers, while E.coli bacteria were chosen as the targets. Under optimized conditions, a capture efficiency of 96% and an average loading number of 3-4 (targets per single motor) could be achieved. The possibility of simple separation of targets from micromotors has also been demonstrated. This microfluidic system could facilitate the integration of multiple steps for bioanalysis and detection of targets.

Abstract:Micro/nanomotors (MNMs) have evolved from single self-propelled entities to versatile systems capable of performing one or multiple biomedical tasks. When single MNMs self-assemble into coordinated swarms, either under external control or triggered by chemical reactions, they offer advantages that individual MNMs cannot achieve. These benefits include intelligent multitasking and adaptability to changes in the surrounding environment. Here, we provide our perspective on the evolution of MNMs, beginning with the development of enzymatic MNMs since the first theoretical model was proposed in 2005. These enzymatic MNMs hold immense promise in biomedicine due to their advantages in biocompatibility and fuel availability. Subsequently, we introduce the design and application of single motors in biomedicine, followed by the control of MNM swarms and their biomedical applications. In the end, we propose viable solutions for advancing the development of MNM swarms and anticipate valuable insights into the creation of more intelligent and controllable MNM swarms for biomedical applications.

Abstract:In the microscopic world, synthetic micro/nanomotors (MNMs) can convert a variety of energy sources into driving forces to help humans perform a number of complex tasks with greater ease and efficiency. These tiny machines have attracted tremendous attention in the field of drug delivery, minimally invasive surgery, in vivo sampling, and environmental management. By modifying their surface materials and functionalizing them with bioactive agents, these MNMs can also be transformed into dynamic micro/nano-biosensors that can detect biomolecules in real-time with high sensitivity. The extensive range of operations and uses combined with their minuscule size have opened up new avenues for tackling intricate analytical difficulties. Here, in this review, various driving methods are briefly introduced, followed by a focus on intelligent detection techniques based on MNMs. And we discuss the distinctive advantages, current issues, and challenges associated with MNM-based intelligent detection. It is believed that the future advancements of MNMs will greatly impact the diagnosis, treatment, and prevention of diseases.

Abstract:Micro/nanomotors (MNMs) are miniaturized machines that can perform assigned tasks at the micro/nanoscale. Over the past decade, significant progress has been made in the design, preparation, and applications of MNMs that are powered by converting different sources of energy into mechanical force, to realize active movement and fulfill on-demand tasks. MNMs can be navigated to desired locations with precise controllability based on different guidance mechanisms. A considerable research effort has gone into demonstrating that MNMs possess the potential of biomedical cargo loading, transportation, and targeted release to achieve therapeutic functions. Herein, the recent advances of self-propelled MNMs for on-demand biomedical cargo transportation, including their self-propulsion mechanisms, guidance strategies, as well as proof-of-concept studies for biological applications are presented. In addition, some of the major challenges and possible opportunities of MNMs are identified for future biomedical applications in the hope that it may inspire future research.

Abstract:Synthetic nano/micromotors are a burgeoning class of materials with vast promise for applications ranging from environmental remediation to nanomedicine. The motility of these motors is generally controlled by the concentration of accessible fuel, and therefore, engineering speed-regulation mechanisms, particularly using biological triggers, remains a continuing challenge. Here, control over the movement of superassembled porous framework micromotors via a reversible, biological-relevant pH-responsive regulatory mechanism is demonstrated. Succinylated -lactoglobulin and catalase are superassembled in porous framework particles, where the -lactoglobulin is permeable at neutral pH. This permeability allows the fuel (H2O2) to access catalase, leading to autonomous movement of the micromotors. However, at mild acidic pH, succinylated -lactoglobulin undergoes a reversible gelation process, preventing the access of fuel into the micromotors where the catalase resides. To one's knowledge, this study represents the first example of chemically driven motors with rapid, reversible pH-responsive motility. Furthermore, the porous framework significantly enhances the biocatalytic activity of catalase, allowing ultralow H2O2 concentrations to be exploited at physiological conditions. It is envisioned that the simultaneous exploitation of pH and chemical potential of such nanosystems could have potential applications as stimulus-responsive drug delivery vehicles that benefit from the complex biological environment.
Light-driven upconversion fluorescence micromotors

Abstract:Light-driven micromotors with high spatial resolution are powerful tools for targeted drug delivery and biomedical diagnosis. To combine the function of biosensing, light-driven micromotors have been modified with fluorescence materials such as quantum dots or dyes. However, these fluorescence micromotors are generally driven and excited by ultraviolet or visible lights, which may cause photo-damage to biological cells or tissues. Here we propose upconversion fluorescence micromotors (UCFMs) constructed by lanthanide (NaYF4: Yb3+, Er3+) doped microrods that were driven and excited by near-infrared lights. The UCFMs were moved to the surfaces of the targeted cancer cells using scanning optical tweezers (SOTs). The upconversion fluorescence spectra were measured to determine the temperatures of the cells, with an absolute sensitivity from 1.71 × 10-3 to 1.74 × 10-3 K-1 and a relative sensitivity from 0.53% to 0.68% K-1. The UCFMs were then optically driven to actuate the local flow to deliver the polystyrene (PS) microparticles and doxorubicin-loaded mesoporous silica nanoparticles to the vicinity of the cancer cells. By integrating the actuator and sensor into a single device, the UCFMs hold great potential for applications to precise biosensing, single-cell biomedical analysis, and targeted drug delivery.

Abstract:Miniaturization of synthetic micro/nanomotors (MNMs) brings great application prospects but limits their functionalization ability. Here, we report self-fueled metal organic framework (MOF) micromotors that are endowed with capabilities of self-propulsion and antibacterial therapy by their material's intrinsic properties. The spontaneous degradation of the MOF micromotors in water would release their own constituting components of ions which act as fuels to propel themselves by ionic diffusionphoresis with a high energy conversion efficiency. Meanwhile, the metal cations released from the MOF micromotors can also serve as antibacterial reagents to kill Escherichia coli (E. coli) with motion enhanced efficacy, which could significantly accelerate the wound closure in a bacterially infected wound model in vivo. Our work provides a general guidance for constructing functional MNMs by taking advantage of the motors' own materials to achieve self-propulsion and on-demand task assignments, which would promote future development of highly integrated micro/nanorobotic systems at micro/nanoscale.

Abstract:Biocatalytic micro/nanomotors are objects with micron or nanometer sizes that can transfer local chemical energy into mechanical energy through enzyme catalysis, and thus they hold great promise in the fields of biomedicine, sensors, and environmental science. Their combination with metal- organic frameworks endows biocatalytic motors with many characterizations, including facile synthesis, high and controllable porosity, good biocompatibility, and stimuli-responsive multifunctional behaviors. In this perspective, after illustrating the prerequisites of movement at the micro- and nanoscale, we highlight the recent success of biocatalytic MOFtors and how they resolved specific problems associated with applications in healthcare and water treatment. Also, fundamental principles about the design of biocatalytic MOFtors are elaborated, providing a general guideline to further improve their propulsion performance.

Abstract:"On-the-fly" molecule enrichment by micro/nanomotors obviously improves heterogeneous catalysis, trace detection, and environmental monitoring, yet faces challenges of the trade-off between collection range and interaction time. Inspired by the versatile foraging process of predators, this work demonstrates that micromotors doing spontaneous multipattern motion with microvortex can greatly enhance "on-the-fly" enrichment, demonstrated by highly sensitive surface-enhanced Raman scattering detection. It leverages an axis-asymmetric bowl-shaped structure and the nonlinear Ag-AgCl reaction, realizing alternating low-velocity swinging forward and accelerated steering motions for prolonged interaction and large work area. Moreover, the bowl-shaped microstructure bestows a micro-vortex above the Ag side due to the competition of electric potential and pressure gradient, also extending interaction time during the acceleration. Consequently, it exhibits at least an order of magnitude larger enhancement of detection signals than the counterparts. This proof-of-concept study highlights the significance of motion mode and structure design in guiding flow field, offering substantial benefits for applications. Leveraging an axis-asymmetric bowl-shaped structure and Ag-AgCl nonlinear chemical reaction can produce a micromotor spontaneously doing switchable multipattern motion between swinging forward and accelerated steering with a microvortex. Abstract:As a typical, classical, but powerful biochemical sensing technology in analytical chemistry, enzyme-linked immunosorbent assay (ELISA) shows excellence and wide practicability for quantifying analytes of ultralow concentration. However, long incubation time and burdensome laborious multistep washing processes make it inefficient and labor-intensive for conventional ELISA. Here, we propose rod-like magnetically driven nanorobots (MNRs) for use as maneuverable immunoassay probes that facilitate a strategy for an automated and highly efficient ELISA analysis, termed nanorobots enabled ELISA (nR-ELISA). To prepare the MNRs, the self-assembled chains of Fe3O4 magnetic particles are chemically coated with a thin layer of rigid silica oxide (SiO2), onto which capture antibody (Ab1) is grafted to further achieve magnetically maneuverable immunoassay probes (MNR-Ab1s). We investigate the fluid velocity distribution around the MNRs at microscale using numerical simulation and empirically identify the mixing efficiency of the actively rotating MNRs. To automate the analysis process, we design and fabricate by 3-D printing a detection unit consisting of three function wells. The MNR-Ab1s can be steered into different function wells for required reaction or wishing process. The actively rotating MNR-Ab1s can enhance the binding efficacy with target analytes at microscale and greatly decrease incubation time. The integrated nR-ELISA system can significantly reduce the assay time, more importantly during which process manpower input is greatly minimized. Our simulation of the magnetic field distribution generated by Helmholtz coils demonstrates that our approach can be scaled up, which proves the feasibility of using current strategy to construct high throughput nR-ELISA detection instrument. This work of taking magnetic micro/nanobots as active immunoassay probes for automatic and efficient ELISA not only holds great potential for point-of-care testing (POCT) in future but also extends the practical applications of self-propelled micro/nanorobots into the field of analytical chemistry.

Abstract:Catalytic micro-/nanomotors (MNMs) that use H 2 O 2 as fuel to generate O 2 bubbles for propulsion hold great promise for exciting applications in the biological and environmental fields. However, it is still a challenge to alleviate or avoid the negative impact of H 2 O 2 on the ecological environment of water body. Herein, a novel glucose-driven nanomotor with robust dual enzyme-like activities was prepared by precisely controlling the phase composition and enzyme immobilization to construct a 3D hierarchical structure integrating Fe-MOF, MnO 2 and glucose oxidase (GOx) on halloysite nanotube (HNTs) support. Embedding GOx into the framework of Fe-MOF by a simple one-step coprecipitation process allowed the in-situ generation of H 2 O 2 in the presence of glucose. Such H 2 O 2 could be further decomposed to O 2 bubbles to drive nanomotors. Meanwhile, O 2 bubbles and residual H 2 O 2 could convert to active species superoxide radical (O 2 • − ) and hydroxyl radical (•OH) due to the high peroxidase/oxidase-like activities of Fe-MOF/MnO 2 in nanomotors. As a proof of concept, the as-synthesized nanomotors could realize sensitively colorimetric detection of glutathione (GSH) with a detection limit of 9.3 × 10 −8 M. In particular, the nanomotors could also rapidly degrade tetracycline hydrochloride (TCH) under neutral conditions without additional H 2 O 2 . It is envisioned that the new strategy that combines strong enzyme-like activity with enhanced autonomous motion can promote the nanomotors efficiency for biomolecular sensing and environmental remediation.

Abstract:Micro/nanomotors are containers that pass through liquid media and carry cargo. Because they are tiny, micro/nanomotors exhibit excellent potential for biosensing and disease treatment applications. However, their size also makes overcoming random Brownian forces very challenging for micro/nanomotors moving on targets. Additionally, to achieve desired practical applications, the expensive materials, short lifetimes, poor biocompatibility, complex preparation methods, and side effects of micro/nanomotors must be addressed, and potential adverse effects must be evaluated both in vivo and in practical applications. This has led to the continuous development of key materials for driving micro/nanomotors. In this work, we review the working principles of micro/nanomotors. Metallic and nonmetallic nanocomplexes, enzymes, and living cells are explored as key materials for driving micro/nanomotors. We also consider the effects of exogenous stimulations and endogenous substance conditions on micro/nanomotor motions. The discussion focuses on micro/nanomotor applications in biosensing, treating cancer and gynecological diseases, and assisted fertilization. By addressing micro/nanomotor shortcomings, we propose directions for further developing and applying micro/nanomotors.

Abstract:Tumor-derived exosomes containing multiple proteins originating from parent cancer cells have emerged as biomarkers for cancer diagnosis. Herein, we propose a three-dimensional DNA motor-based exosome assay platform for the selective and sensitive detection of exosomes. The DNA motor used gold nanoparticle (GNP) tracks, consisting of fluorescein-labeled substrate strands and aptamer-locked motor strands. Recognition of the target protein on exosomes by its aptamer unlocked the motor strand and triggered the DNA motor process. Powered by restriction endonuclease, the motor strands autonomously walked along the GNP track. Each movement step cleaved one substrate strand and restored one fluorescein molecule. For exosome detection, the proposed method displayed a broad dynamic range acrossing 5 orders of magnitude with the detection limit as low as 8.2 particles/μL in PBS. The method also exhibited good selectivity among different tumor-derived exosomes and performed well in complex biological samples. The capability to profile exosomal surface proteins efficiently endowed our DNA motor great potential for developing a simple and cost-effective device for clinical diagnosis.
