Modeling and validation of oviposition by a polyphagous insect pest as a function of temperature and host plant species

Abstract:Modeling oviposition as a function of female insect age, temperature, and host plant suitability may provide valuable insight into insect population growth of polyphagous insect pests at a landscape level. In this study, we quantified oviposition by beet leafhoppers, Circulifer (= Neoaliturus) tenellus (Baker) (Hemiptera: Cicadellidae), on four common non-agricultural host plant species [Erodium cicutarium (L.) L'Hér. (Geraniaceae), Kochia scoparia (L.) Schrader (Amaranthaceae), Plantago ovata Forsskál (Plantaginaceae), and Salsola tragus L. (Amaranthaceae)] at two constant temperature conditions. Additionally, temperature-based oviposition models for each host plant species were validated, under semi-field and greenhouse conditions. We found that K. scoparia was the most suitable host plant, and optimal temperature for oviposition was estimated to be 30.6°C. Accordingly, beet leafhoppers appear to be well-adapted to high-temperature conditions, so increasing temperatures due to climate change may favor population growth in non-agricultural areas. Maximum total fecundity (Rm) was used as an indicator of relative suitability of host plants. S. tragus has been considered an important non-agricultural host plant, however, we found that S. tragus and E. cicutarium have lower Rm compared to K. scoparia and P. ovata. The combination of detailed experimental oviposition bioassays, modeling, and model validation is considered widely relevant and applicable to host plant assessments and modeling of population dynamics of other polyphagous insect pests.
Evidence for morph-specific substrate choice in a green-brown polymorphic grasshopper

Abstract:Orthopteran insects are characterized by high variability in body coloration, in particular featuring a widespread green-brown color polymorphism. The mechanisms that contribute to the maintenance of this apparently balanced polymorphism are not yet understood. To investigate whether morph-dependent microhabitat choice might contribute to the continued coexistence of multiple morphs, we studied substrate choice in the meadow grasshopper Pseudochorthippus parallelus. The meadow grasshopper occurs in multiple discrete, genetically determined color morphs that range from uniform brown to uniform green. We tested whether three common morphs preferentially choose differently colored backgrounds in an experimental arena. We found that a preference for green backgrounds was most pronounced in uniform green morphs. If differential choices improve morph-specific performance in natural habitats via crypsis and/or thermoregulatory benefits, they could help to equalize fitness differences among color morphs and potentially produce frequency-dependent microhabitat competition, though difference appear too small to serve as the only explanation. We also measured the reflectance of the grasshoppers and backgrounds and used visual modeling to quantify the detectability of the different morphs to a range of potential predators. Multiple potential predators, including birds and spiders, are predicted to distinguish between morphs chromatically, while other species, possibly including grasshoppers themselves, will perceive only differences in brightness. Our study provides the first evidence that morph-specific microhabitat choice might be relevant to the maintenance of the green-brown polymorphisms in grasshoppers and shows that visual distinctness of color morphs varies between perceivers.
Altitudinal variation in behavioural thermoregulation: local adaptation vs. plasticity in California grasshoppers

Abstract:We investigated the adaptive significance of behavioural thermoregulation in univoltine populations of the grasshopper Melanoplus sanguinipes along an altitudinal gradient in California using laboratory tests of animals raised under different temperatures. Trials consisted of continuous body temperature measurements with semi-implanted microprobes in a test arena, and observation and simultaneous recording of behavioural responses. These responses included mobility, basking and orientation of the body axes (aspect angle) towards a radiation source. Mobility and basking are determined by the altitudinal origin of the parental generation and not by the temperature treatments. With increasing altitude, individuals tend increasingly to raise body temperatures via mobility and increased basking. In contrast, body orientation towards the radiation source is influenced by the temperature treatments but not by the altitude of origin. Individuals experiencing higher temperatures during rearing show a lower tendency to lateral flanking. We conclude that body orientation responses are not adapted locally. In contrast other components of the behavioural syndrome that increase body temperature, such as mobility and basking, are adaptive in response to local selection pressure. The thermoregulatory syndrome of these grasshoppers is an important contribution to life-history adaptations that appropriately match season lengths.

Summer egg diapause in a matchstick grasshopper synchronizes the life cycle and buffers thermal extremes

Abstract:The phenological response is among the most important traits affecting a species' sensitivity to climate. In insects, strongly seasonal environments often select for a univoltine life cycle such that one seasonal extreme is avoided as an inactive stage. Through understanding the underlying mechanisms for univoltinism, and the consequences of its failure, we can better predict insect responses to climate change. Here we combine empirical data and simulation studies to investigate the consequences of an unusual diapause mechanism in a parthenogenetic matchstick grasshopper, Warramaba virgo, from arid southern Australia. Our field body temperature measurements indicate that this species is a thermoconformer and our laboratory studies of the thermal response of feeding rate imply strong constraints on winter activity. However, the species exhibits no obligate winter diapause, and eggs can develop in 1 month under constant temperatures approximating the mean soil temperature at the time of oviposition (summer). We show that diurnal temperature cycles exceeding a peak of 36 °C inhibit egg development in summer, and that this is sufficient to prevent autumnal hatching of eggs. Development is also strongly retarded below 24 °C. Microclimate-driven simulation studies of egg development show that these thermal responses provide robust maintenance of a univoltine life cycle, thereby resulting in survival of heat stress as an egg (due to limited developmental state) and avoidance of cold stress as a nymph and adult (due to overwintering in the soil as an egg).

Abstract:In a world grappling with climate change, understanding the enduring impact of changes in temperatures on insect adult traits is crucial. It is proposed that cold- and warm-adapted species exhibit specialized behavioural and physiological responses to their respective temperature ranges. In contrast, generalist species maintain more stable metabolic and developmental rates across a broader range of temperatures, reflecting their ability to exploit diverse thermal niches. Here, we explored this intricate response to temperature exposure in three Drosophila species: Drosophila ezoana originating in Arctic regions, D. novamexicana in arid, hot environments, and in the cosmopolitan species D. virilis. Rearing these flies at 15, 20, 25, and 30 °C revealed striking variations in their cuticular hydrocarbon (CHC) profiles, known to mediate mate recognition and prevent water loss in insects. The cold-adapted D. ezoana consistently exhibited reduced CHC levels with increasing temperatures, while the warm-adapted D. novamexicana and the cosmopolitan D. virilis displayed more nuanced responses. Additionally, we observed a significant influence of rearing temperature on the mating behaviour of these flies, where those reared at the extreme temperatures, 15 and 30 °C, exhibiting reduced mating success. Consequently, this led to a decrease in the production of adult offspring. Also, these adult offspring underwent notable alterations in life history traits, reaching adulthood more rapidly at 25 and 30 °C but with lower weight and reduced longevity. Furthermore, among these offspring, those produced by the cold-adapted D. ezoana were more vulnerable to desiccation and starvation than those from the warm-adapted D. novamexicana and the cosmopolitan D. virilis. In summary, our research demonstrates that Drosophila species from diverse ecological regions exhibit distinct responses to temperature changes, as evidenced by variations in CHC profiles, mating behaviours, fertility, and life history traits. This provides valuable insights into how environmental conditions shape the biology and ecology of insects.
Plasticity in reproductive output and development in response to thermal variation in ladybird beetle, Menochilus sexmaculatus

Abstract:All organisms are exposed to daily as well as seasonal variation in temperature in nature. However, ectotherms are strongly affected by temperature and its variations. Most laboratory studies on effect of temperature on life history traits employ constant instead of fluctuating temperature regimes, though the latter is more reflective of nature. We conducted investigations on the effect of constant, fluctuating and alternate temperature regimes on reproductive and developmental attributes of a biocontrol agent ladybird, Menochilus sexmaculatus (Fabricius) (Coleoptera: Coccinellidae). We used three constant, viz. cold, optimum and hot (15°C, 27°C and 35°C); two fluctuating with variation in mean temperature of the day (15°C/27°C, 35°C /27°C; 10/14h) and three alternating (15°C/27°C, 35°C/27°C, 15°C/35°C; 24/24h) temperatures to evaluate their effects on these life attributes. Oviposition and percent egg viability along with offspring development (immature life stages developmental time and developmental rate) and elytral colouration pattern of adults were observed. The best response in terms of oviposition and percent egg viability was recorded at the constant temperature of 27°C followed by the fluctuating regime with a mean temperature of 30.3°C. The fastest development was found at a constant temperature of 35°C, followed by a fluctuating temperature of 35°C/27°C for 10/14h. Among the different temperature cycles, the most uncustomary trend of oviposition and percent egg viability was recorded on an alternate temperature of 15°C/35°C for 24/24h. The constant (27°C) and fluctuating (35°C/27°C for 10/14h) temperatures caused an incremental effect on reproductive output and developmental rate. This study demonstrates that M. sexmaculatus is able to adjust its physiology according to surrounding environmental conditions and acclimatizes well with the fluctuating/alternate temperatures suggesting its wide range of tolerance. The findings of present work would be helpful in augmenting population dynamics of this biocontrol agent to suppress insect pest.
Micro‐ and macroclimate interactively shape diversity, niches and traits of Orthoptera communities along elevational gradients

Abstract:Aim Temperature is one of the main drivers shaping species diversity and assembly processes. Yet, site‐specific effects of the local microclimate on species and trait compositions of insect communities have rarely been assessed along macroclimatic temperature clines. Location Bavarian Alps, Germany. Methods Bayesian joint species distribution models were applied to investigate how ecological and morphological traits drive variation in the climatic niches of 32 Orthoptera species on 93 grassland sites with contrasting microclimatic conditions along a steep elevational macroclimatic gradient in an Alpine region in Central Europe. Results Species richness and abundance decreased along the elevational macroclimatic gradient, and both benefitted from warm microclimate. Interactive effects of elevation and microclimate on the abundance were, however, species‐specific, and partly mediated by traits: Warm microclimatic conditions facilitated the occurrence of demanding xerophilic and late‐hatching species, resulting in marked community dissimilarities at mid‐elevations where colder sites harboured only a subset of the species. The latter mainly occurred at low elevations together with long‐winged species. Abundance peaks of non‐xerophilic species were further upslope when microclimate was warm. Intraspecifically, the body sizes and wing lengths of the larger females, but not the males, decreased with elevation akin the community mean, and brown colour morphs were more frequent at sites with warm microclimate. Main Conclusions Our nuanced results reveal that trait‐dependent responses of species to microclimate play a key role in the assembly and structuring of insect communities along macroclimatic gradients. Since microclimate preferences changed with elevation, we conclude that species temperature niches are narrower than the elevational range suggests and both macro‐ and microclimatic conditions must be considered when predicting species responses to climate change. Microclimatic contrasts among sites at similar elevations enhanced species turnover mediated by moisture preferences and phenology, highlighting the importance of mountains for conservation as climatic refugia where species with diverging niches can persist in proximity.

Abstract:Trichogramma parasitoids are effective biocontrol agents and a reliable component of integrated strategies against lepidopterous pests. The success of these parasitoids in pest management relies not only on their ability to parasitize their hosts but also on their adaptation to the climatic conditions of the release area, particularly temperature. The expression of life history traits of Trichogramma spp. can vary significantly with temperature, depending on the species or strains being tested. Trichogramma cacoeciae (Marchal), T. euproctidis (Girault), T. minutum (Riley), and T. brassicae (Bezdenko) (Hymenoptera: Trichogrammatidae) are currently used in biocontrol programs against important lepidopteran pests. We aimed to assess the temperature sensitivity of these parasitoids during oviposition and preadult development, and to identify the most tolerant species to high temperatures conditions commonly encountered in Mediterranean Basin countries during the growing seasons. The biological characteristics of the four species were determined at seven temperature regimes expressed as temperatures during oviposition and preadult development (25/25, 25/30, 25/35, 25/40, 30/30, 35/35 and 40/40 °C), using Ephestia kuehniella Zeller (Lepidoptera: Pyralidae) eggs. Trichogramma cacoeciae showed the highest level of parasitism at 30/30 °C, while the other species exhibited the highest levels at 25/25 °C and 25/30 °C. All Trichogramma species were able to develop and survive from 25 °C to 35 °C, but not at 40 °C. Temperature significantly affected the longevity and fecundity of female progeny, with both decreasing when the temperature increased from 25 °C to 35 °C. When exposed to 35/35 °C, T. cacoeciae demonstrated the most optimal performance in terms of parasitization efficiency, developmental capacity, progeny longevity, and fecundity.

Abstract:Heat tolerance is a trait of paramount ecological importance and may determine a species' ability to cope with ongoing climate change. Although critical thermal limits have consequently received substantial attention in recent years, their potential variation throughout ontogeny remained largely neglected. We investigate whether such neglect may bias conclusions regarding a species' sensitivity to climate change. Using a tropical butterfly, we found that developmental stages clearly differed in heat tolerance. It was highest in pupae followed by larvae, adults and finally eggs and hatchlings. Strikingly, most of the variation found in thermal tolerance was explained by differences in body mass, which may thus impose a severe constraint on adaptive variation in stress tolerance. Furthermore, temperature acclimation was beneficial by increasing heat knock-down time and therefore immediate survival under heat stress, but it affected reproduction negatively. Extreme temperatures strongly reduced survival and subsequent reproductive success even in our highly plastic model organism, exemplifying the potentially dramatic impact of extreme weather events on biodiversity. We argue that predictions regarding a species' fate under changing environmental conditions should consider variation in thermal tolerance throughout ontogeny, variation in body mass and acclimation responses as important predictors of stress tolerance.

Abstract:The combined effect of the temperature and relative humidity on the survival and development time of the eggs of Spanish populations of the weevil Hypera postica, a key pest of alfalfa around the world, was evaluated under laboratory conditions. The experimental temperatures ranged from 8 to 36 °C, in 4 °C increments. Three relative humidity ranges were defined: high, medium, and low. Eggs of the alfalfa weevil successfully developed until larval emergence at all of the 24 conditions tested. However, the temperature and relative humidity affected the survival of the eggs. The egg developmental time decreased as the temperature increased from 8 to 32 °C, and the longest time was recorded at a low relative humidity (RH). The relationship between the development rate and temperature fit well to the lineal model for relative humidity. The minimum development threshold (T0) and the thermal requirement for egg development (K) ranged between 3 and 4 °C and 209 and 246 degree-days, respectively. According to these values and the occurrence of eggs and larvae (in winter) and adults (in autumn) in field samplings, the period of oviposition was determined. The results of the study contribute to better understanding the annual cycle and phenology of H. postica in the Iberian Peninsula and southern Europe.

Abstract:Otopheidomenidae include 30 species of parasites of insects. These mites occur in areas with warm and hot climates. The only exception is Hemipteroseius adleri , the distribution of which reaches the central European region, which is characterised by a moderate climate with below-zero temperatures in winter. In Europe, H. adleri only parasitises the red firebug ( Pyrrhocoris apterus ). Field observations indicate that only fertilised females of H. adleri survive winter conditions. The aim of the study therefore was to evaluate the impact of air temperature on the development and survival of H. adleri . A research model was established involving mites parasitising red firebugs in laboratory conditions at 15, 20, 25 and 30 °C. After oviposition, no egg development was possible at 15 °C. Egg development began at 20 °C, however the egg-to-adult period lasted for 13 days and larval mortality was significantly higher at this temperature than at 25 and 30 °C. At the latter tem temperatures, mites matured fastest (five and four days, respectively). The net reproductive rate ( R 0 ) was 10.8, the mean generation time was ( T ) 14.4 (SD = 0.273), the intrinsic rate of population increase ( r m ) was 0.165 (SD = 0.003), and the finite rate of population increase ( λ ) was 1.12. At 25 °C, H. adleri required 4.2 days to double its population. The results indicate that the thermal conditions prevailing in early spring in central Europe are highly unfavourable for the effective reproduction of H. adleri . The presence of northern populations of this mite is discussed in the context of behavioural thermoregulation mechanisms adapted by its host, P. apterus .

Warm and thermally variable incubation conditions reduce embryonic performance and carry over to influence hatchling tradeoffs

Abstract:Animals' thermal sensitivities have long been characterized by thermal performance curves (TPCs) or reaction norms, and TPCs may predict animals' responses to climate change. Typically, TPCs are parameterized by measuring performance at a range of constant temperatures. Yet, animals encounter a range of thermal environments, and temperature variability is an aspect of climate change that may affect animals more than gradual warming. Daily temperature variability is particularly important for eggs in most taxa because they are highly sensitive to temperature and cannot behaviorally avoid stressful temperatures. Thus, the legacy of thermal conditions experienced during incubation may carryover to subsequent life stages. Here, I factorially manipulated mean temperature (20, 25, or 30 °C) and daily temperature range (DTR; ±0, 5, or 10 °C) during incubation for eggs of the variable field cricket (Gryllus lineaticeps) to integrate the role of DTR into the established paradigm of TPCs. Low DTR (±5 °C) was not generally costly, and it even improved hatchling starvation resistance (sensu hormesis). However, high DTR (±10 °C) reduced and delayed hatching at a warm mean temperature (30 °C). The effects of high DTR carried over to accelerate hatchling development at an expense to hatchling starvation resistance-therefore, thermal conditions during incubation can shape tradeoffs among important traits related to life history and stress tolerance later in life. In sum, animals may exhibit complex responses to their increasingly warmer, more thermally variable environments.
Ecotypic differentiation between urban and rural populations of the grasshopper Chorthippus brunneus relative to climate and habitat fragmentation

Abstract:Urbanization alters environmental conditions in multiple ways and offers an ecological or evolutionary challenge for organisms to cope with. Urban areas typically have a warmer climate and strongly fragmented herbaceous vegetation; the urban landscape matrix is often assumed to be hostile for many organisms. Here, we addressed the issue of evolutionary differentiation between urban and rural populations of an ectotherm insect, the grasshopper Chorthippus brunneus. We compared mobility-related morphology and climate-related life history traits measured on the first generation offspring of grasshoppers from urban and rural populations reared in a common garden laboratory experiment. We predicted (1) the urban phenotype to be more mobile (i.e., lower mass allocation to the abdomen, longer relative femur and wing lengths) than the rural phenotype; (2) the urban phenotype to be more warm adapted (e.g., higher female body mass); and (3) further evidence of local adaptation in the form of significant interaction effects between landscape of origin and breeding temperature. Both males and females of urban origin had significantly longer relative femur and wing lengths and lower mass allocation to the abdomen (i.e., higher investment in thorax and flight muscles) relative to individuals of rural origin. The results were overall significant but small (2-4%). Body mass and larval growth rate were much higher (+10%) in females of urban origin. For the life history traits, we did not find evidence for significant interaction effects between the landscape of origin and the two breeding temperatures. Our results point to ecotypic differentiation with urbanization for mobility-related morphology and climate-related life history traits. We argue that the warmer urban environment has an indirect effect through longer growth season rather than direct effects on the development.

Abstract:The 9-(Z)-configuration was exclusively obtained in the carotenoid polyene chain irrespective of olefination and disconnection methods for terminal ortho-unsubstituted benzene rings. The 2,6-dimethyl substituents in the terminal rings secure an all-(E)-polyene structure. The single molecular conductance of the pure 9-(Z)-carotene was measured for the first time to be 1.53 × 10-4 ± 6.37 × 10-5G0, whose value was 47% that of the all-(E)-carotene ((3.23 × 10-4) ± (1.23 × 10-4) G0).
Magnetic resonance imaging of thymic epithelial tumors.

Abstract:The authors review their experience with magnetic resonance imaging (MRI) of the thymus and discuss the appearance of thymic epithelial tumors where MRI is clinically useful. Detailed descriptions of MRI findings in benign thymomas, invasive thymomas, and thymic carcinomas are provided. Most benign (noninvasive) thymomas appear with a slightly higher signal intensity than that of muscle on T1-weighted images. On T2-weighted images, thymomas have an increase in signal intensity on both T1- and T2-weighted images. Neither fibrous septa nor lobulated internal architecture can be detected on MRI. Gd-DTPA-enhanced MR images show homogeneous enhancement. On the other hand, invasive thymomas show the same signal intensity as benign thymomas, both on T1- and T2-weighted images. However, invasive thymomas appear inhomogeneous in signal intensity on T2-weighted images. T2-weighted images also show a lobulated border, fibrous septa, and lobulated internal architecture, characteristic of most invasive thymomas. Irregularity of tumor margins indicating invasion into surrounding structures is noted in some cases of invasive thymomas. Exceptionally minute thymomas (< 1 cm in diameter) show a different signal intensity on MRI as compared to those of usual thymomas: both T1- and T2-weighted MR images show a low signal intensity mass with irregular or unclear borders. Histopathologically, these minute thymomas contain numerous tiny cysts and/or abundant collagenous tissues. Generally, thymic carcinomas, except carcinoid tumors, appear with a relatively low signal intensity on T1- and T2-weighted MR images in comparison to those of thymomas. In particular, well-differentiated squamous cell carcinomas appear with a low signal intensity on both T1- and T2-weighted images. Abundant collagenous tissue may be a causative factor for the low signal intensity on T2-weighted MR images. Thymic carcinomas appear slightly inhomogeneous on both T1- and T2-weighted images. Neither fibrous septa nor lobulated internal architecture can be detected in any thymic carcinoma. If MRI is performed on a patient with anterior mediastinal tumors, thymic carcinoma may be precisely diagnosed when characteristic MR findings are demonstrated.
DNA Replication Dynamics of the GGGGCC Repeat of the C9orf72 Gene*

Abstract:Background: The (GGGGCC)n hexanucleotide repeat expansion of C9orf72 is the most common genetic cause of ALS-FTD. Results: C9orf72 repeat expansion increases instability and decreases replication efficiency by disrupting replication fork progression. Conclusion: C9orf72 repeat length and replication direction contribute to repeat instability in human cells. Significance: DNA replication-induced instability at the C9orf72 GGGGCC repeat can lead to further expansion and more severe disease. DNA has the ability to form a variety of secondary structures in addition to the normal B-form DNA, including hairpins and quadruplexes. These structures are implicated in a number of neurological diseases and cancer. Expansion of a GGGGCC repeat located at C9orf72 is associated with familial amyotrophic lateral sclerosis and frontotemporal dementia. This repeat expands from two to 24 copies in normal individuals to several hundreds or thousands of repeats in individuals with the disease. Biochemical studies have demonstrated that as little as four repeats have the ability to form a stable DNA secondary structure known as a G-quadruplex. Quadruplex structures have the ability to disrupt normal DNA processes such as DNA replication and transcription. Here we examine the role of GGGGCC repeat length and orientation on DNA replication using an SV40 replication system in human cells. Replication through GGGGCC repeats leads to a decrease in overall replication efficiency and an increase in instability in a length-dependent manner. Both repeat expansions and contractions are observed, and replication orientation is found to influence the propensity for expansions or contractions. The presence of replication stress, such as low-dose aphidicolin, diminishes replication efficiency but has no effect on instability. Two-dimensional gel electrophoresis analysis demonstrates a replication stall with as few as 20 GGGGCC repeats. These results suggest that replication of the GGGGCC repeat at C9orf72 is perturbed by the presence of expanded repeats, which has the potential to result in further expansion, leading to disease.
